Gumudavelli ABSTRACT:,Salmonella Enteritidis (SE) contamination of poultry eggs is a major human health concern worldwide. The risk of SE from shell eggs can be significantly reduced through rapid cooling of eggs after they are laid and their storage under safe temperature conditions. Predictive models for the growth of SE in egg yolk under varying ambient temperature conditions (dynamic) were developed. The growth of SE in egg yolk under several isothermal conditions (10, 15, 20, 25, 30, 35, 37, 39, 41, and 43 °C) was determined. The Baranyi model, a primary model, was fitted with growth data for each temperature and corresponding maximum specific growth rates were estimated. Root mean squared error (RMSE) values were less than 0.44 log10 CFU/g and pseudo- R2 values were greater than 0.98 for the primary model fitting. For developing the secondary model, the estimated maximum specific growth rates were then modeled as a function of temperature using the modified Ratkowsky's equation. The RMSE and pseudo- R2 were 0.05/h and 0.99, respectively. A dynamic model was developed by integrating the primary and secondary models and solving it numerically using the 4th-order Runge,Kutta method to predict the growth of SE in egg yolk under varying temperature conditions. The integrated dynamic model was then validated with 4 temperature profiles (varying) such as linear heating, exponential heating, exponential cooling, and sinusoidal temperatures. The predicted values agreed well with the observed growth data with RMSE values less than 0.29 log10 CFU/g. The developed dynamic model can predict the growth SE in egg yolk under varying temperature profiles. [source] A Lipoprotein-derived Antimicrobial Factor from Hen-egg Yolk is Active Against Streptococcus SpeciesJOURNAL OF FOOD SCIENCE, Issue 8 2002D. Brady ABSTRACT: Oral administration of hen-egg yolk provides protection against specific pathogens. We examined the antibacterial activity of fractionated egg yolk against 2 pathogenic Streptococcus strains, using an in vitro assay. A water-soluble protein fraction (WSPF) of egg yolk consistently inhibited the growth of S. mutans by 25%. The WSPF treated with pancreatin demonstrated > 80% inhibition of bacterial growth. Growth of S. sanguis was completely inhibited. Gel filtration and ion exchange chromatography established that anti-Streptococcal activity resided with lipoproteins. Antibacterial activity was released by crude lipase or a combination of lipase and protease treatment of egg lipoproteins. Thus, hen-egg yolk lipoproteins are important molecules for lipid-mediated antimicrobial activity. Chang ABSTRACT: Hens were intramuscularly (im) immunized on thighs by using urease (E.C. 3.5.1.5) from Helicobactor pylori as antigen. The specificity of IgY against urease of H. pylori increased gradually after initial immunization. The collected yolk was microencapsulated with 10% or 20%,-cyclodextrin (,-CD) and gum arabic by a spray-drier. Microencapsulation was effective in protecting the IgY activity against pepsin. Liposome prepared at the lecithin/ cholesterol ratio of 1/0.25 (mole/mole) displayed satisfactory encapsulation efficiency (69%) of IgY. Increase in cholesterol content in the liposomal structure exhibited a stronger protection effect of IgY against pepsin and acid. [source] Hen Egg Yolk Prevents Bacterial Adherence: A Novel Function for a Familiar FoodJOURNAL OF FOOD SCIENCE, Issue 1 2001T. Deignan ABSTRACT: The aim of this study was to evaluate the ability of unfractionated hen egg yolk to prevent the attachment of Salmonella typhimurium to murine intestinal epithelial cells in vitro. Inhibition of adherence was examined microscopically and by an ELISA. Both methods showed that egg yolk from immunized and unimmunized hens significantly reduced bacterial adherence. Optical density (OD) readings ± SD of 0.825 ± 0.016 for untreated bacteria were reduced to 0.335 ± 0.024 and 0.267 ± 0.021 for bacteria pretreated with egg yolk from immunized hens and unimmunized hens, respectively. Microscopic analysis showed that egg yolk from either immunized or unimmunized hens reduced bacterial adherence from 17 ± 2.7 bacteria per epithelial cell to less than 4 bacteria per epithelial cell. These results show that hen egg yolk significantly inhibits adherence of S. typhimurium to intestinal epithelial cells in vitro. Boosting of specific antibody levels does not enhance this antiadhesive effect, suggesting that egg yolk contains novel antiadhesive factors. [source] Post-thaw Survival and Longevity of Bull Spermatozoa Frozen with an Egg Yolk-based or Two Egg Yolk-free Extenders after an Equilibration Period of 18 hREPRODUCTION IN DOMESTIC ANIMALS, Issue 3 2007R Muiño Contents The aim of the present study was to determine the suitability of using two egg yolk-free commercial extenders, Andromed® and Biociphos Plus®, as compared with the Tris-egg yolk based diluent Biladyl®, for the cryopreservation of bull spermatozoa when the freezing protocol involved holding the extended semen at 4°C for 18 h before the freezing. Six ejaculates from each of 10 Holstein bulls were collected by using artificial vagina. The ejaculates were evaluated for volume, sperm concentration and motility, divided in to three equal volumes, and diluted, respectively, with the three extenders as specified above. Extended semen was equilibrated for 18 h at 4°C and frozen in 0.25-ml straws. After thawing, 100- ,l aliquots of semen were labelled with SYBR-14, PI and PE-PNA (Phycoerythrin-conjugated Peanut agglutinin) and analysed by flow cytometry at 0, 3, 6 and 9 h after incubation at 37°C. A General Lineal Model procedure for repeated measures was used to determine the effects of extender, bull, replicate and the interaction between them, on sperm viability and acrosomal integrity. Semen samples frozen with Biladyl® showed higher (p < 0.001) sperm survival after 0 h (47.9%) and 9 h (30.3%) of incubation than those frozen with Andromed® (38.5% and 17.3%, after 0 and 9 h respectively) or Biociphos Plus® (34.9% and 21.6%, after 0 and 9 h respectively). The bull and replicate had significant effects (p < 0.001) on both sperm viability and acrosomal integrity, but the interactions between bull and extender and between replicate and extender were not significant. It was concluded that, when holding the semen overnight before freezing, the use of Biladyl® results in higher sperm survival and longevity than the use of Andromed® or Biociphos Plus®. Thus late-hatching nestlings in asynchronously hatching clutches somehow overcome any age- and size-related disadvantages of hatching after their nest-mates. One possible way for them to do this is for female House Wrens to add maternal androgens to the yolk of late-hatching eggs. We tested this hypothesis in a wild population of House Wrens that produces both asynchronously and synchronously hatching clutches. Yolks of eggs from both types of clutches were biopsied and the eggs returned to their nests to hatch. Radioimmunoassays revealed that total androgen levels in the yolk varied within and among clutches. However, total androgen levels in yolks did not vary predictably with egg position in either synchronously or asynchronously hatching clutches. Thus, deposition of androgens in yolk did not follow the expected pattern based on the potential for sibling competition in House Wrens. [source] Midblastula transition (MBT) of the cell cycles in the yolk and pigment granule-free translucent blastomeres obtained from centrifuged Xenopus embryosDEVELOPMENT GROWTH & DIFFERENTIATION, Issue 5 2005Yasuhiro Iwao We obtained translucent blastomeres free of yolk and pigment granules from Xenopus embryos which had been centrifuged at the beginning of the 8-cell stage with cellular integrity. They divided synchronously regardless of their cell size until they had decreased to 37.5 µm in radius; those smaller than this critical size, however, divided asynchronously with cell cycle times inversely proportional to the square of the cell radius after midblastula transition (MBT). The length of the S phase was determined as the time during which nuclear DNA fluorescence increased in Hoechst-stained blastomeres. When the cell cycle time exceeded 45 min, S and M phases were lengthened; when the cell cycle times exceeded 70 min, the G2 phase appeared; and after cell cycle times became longer than 150 min, the G1 phase appeared. Lengths of G1, S and M phases increased linearly with increasing cell cycle time. Enhanced green fluorescent protein (EGFP)-tagged proliferating cell nuclear antigen (PCNA) expressed in the blastomeres appeared in the S phase nucleus, but suddenly dispersed into the cytoplasm at the M phase. The system developed in this study is useful for examining the cell cycle behavior of the cell cycle-regulating molecules in living Xenopus blastomeres by fluorescence microscopy in real time. The applied NOM was a reverse osmosis isolate from Lake Schwarzer See (i.e., Black Lake, BL-NOM). Teratogenic effects were evaluated through changes in embryonic development within 48 h of exposure. Detoxication activities were assessed by the activities of phase II biotransformation enzymes, soluble and microsomal glutathione S -transferase (s, mGST). Oxidative stress was assessed by determining both the production of hydrogen peroxide and by analyzing the activities of the antioxidative enzymes, guajacol peroxidase (POD), catalase (CAT), glutathione peroxidase (GPx), and the glutathione restoring enzyme glutathione reductase (GR). Energetic costs were evaluated by determining contents of fat, carbohydrates, and proteins in both exposed and control embryos. BL-NOM attenuated toxic effects of MC-LF and MC-RR verified by less pronounced teratological effects within 24 h, in particular, as well as less rise in the activity of s-GST, when compared with embryos exposed to either pure toxins or in combination with organic matter. BL-NOM also diminished oxidative effects caused by MC-LF; however, it failed to attenuate oxidative stress caused by MC-RR. Content of lipids was significantly reduced in exposed embryos following a trend similar to that obtained with teratological and enzymatic assays confirming the attenuating effect of BL-NOM. Physiological responses to microcystins and NOM required energetic costs, which were compensated to the expense of the energy resources of the yolk, which in turn might affect the normal development of embryos. © 2006 Wiley Periodicals, Inc. Heinz Abstract Female mallards (Anas platyrhynchos) were fed diets containing 5, 10, or 20 ppm mercury as methylmercury chloride. One egg was collected from each bird before the start of the mercury diets and 15 eggs were collected from each bird while it was being fed mercury. The mercury diets were then replaced by uncontaminated diets, and each female was allowed to lay 29 more eggs. Mercury levels in eggs rose to about 7, 18, and 35 ppm wet-weight in females fed 5, 10, or 20 ppm mercury, respectively. Mercury levels fell to about 0.16, 0.80, and 1.7 ppm in the last egg laid by birds that had earlier been fed 5, 10, or 20 ppm mercury, respectively. Higher concentrations of mercury were found in egg albumen than in yolk, and between 95 and 100% of the mercury in the eggs was in the form of methylmercury. The synthetic estrogen diethylstilbestrol (DES) was used as a positive control. The test compounds were injected into the yolk of quail and chicken eggs early during incubation and the embryos were examined 2 d before anticipated hatching. At 200 ,g/g egg, BPA induced Müllerian duct (embryonic oviduct) malformation in female quail embryos and feminization of the left testis (ovotestis) in male chicken embryos. The estrogenic potency of BPA compared with DES was species and endpoint specific. Müllerian duct malformation was the most sensitive endpoint in quail embryos, whereas ovotestis formation was the most sensitive response in chicken embryos. Tetrabromobisphenol A caused high embryo mortality at 45 ,g/g egg in both species, but no estrogen-like effects were observed. Bisphenol A caused mortality only in chicken embryos at 67 and 200 ,g/g egg. To our knowledge, this is the first report on estrogen-like or embryolethal effects of BPA and TBBPA in birds. Guadagnolo Abstract Rainbow trout embryos were chronically exposed to silver (as AgNO3) in moderately hard water (120 mg CaCO3/L, 0.70 mM Cl,, 1.3 mg/L dissolved organic matter, 12.3 ± 0.1 °C) at nominal concentrations of 0.1, 1, and 10 ,g/L (measured = 0.117 ± 0.008, 1.22 ± 0.16, and 13.51 ± 1.58 ,g/L, respectively) to investigate the effects on mortality, ionoregulation, and silver uptake and distribution of the embryo. Mortalities in the low concentrations (0.1 and 1.2 ,g/L) were not significantly different from controls throughout embryonic development (days 1,32 postfertilization). Mortalities of embryos in the 13.5-,g/L treatment reached 56% by day 32 postfertilization (33% when accounting for control mortality), by which time more than 50% of surviving embryos had hatched. Accumulation of silver in whole embryos of 1.2- and 13.5-,g/L treatments reached the highest concentrations of 0.13 and 0.24 ,g/g total silver, respectively, by day 32, but whole embryo silver burden was not correlated with mortality. Silver concentrations in different compartments of the whole embryo (chorion, dissected embryo, and yolk) were greatest just before hatch and were higher in the chorion for all experimental treatments. Up to 85% of total whole embryo silver content was bound to the chorion, which acts as a protective barrier during silver exposure. Whole embryo Na+ concentration in the 13.5-,g/L treatment was significantly reduced relative to controls from days 23 to 32 postfertilization, and levels in the embryo were reduced by 40% at day 32 postfertilization, indicating that silver toxicity in the whole embryo is associated with an ion regulatory disturbance that is similar to the acute effect of AgNO3 in juvenile and adult trout. [source] Novel regulation of yolk utilization by thyroid hormone in embryos of the direct developing frog Eleutherodactylus coquiEVOLUTION AND DEVELOPMENT, Issue 5 2010Srikanth Singamsetty SUMMARY Thyroid hormone (TH) is required for metamorphosis of the long, coiled tadpole gut into the short frog gut. Eleutherodactylus coqui, a direct developing frog, lacks a tadpole. Its embryonic gut is a miniature adult form with a mass of yolky cells, called nutritional endoderm, attached to the small intestine. We tested the TH requirement for gut development in E. coqui. Inhibition of TH synthesis with methimazole arrested gut development in its embryonic form. Embryos treated with methimazole failed to utilize the yolk in their nutritional endoderm, and survived for weeks without further development. Conversely, methimazole and 3,3,,5-tri-iodo- l -thyronine, the active form of TH, stimulated gut development and utilization and disappearance of the nutritional endoderm. In Xenopus laevis, the receptor for TH, TR,, is upregulated in response to TH. Similarly, EcTR,, the E. coqui ortholog, was upregulated by TH in the gut. EcTR, expression was high in the nutritional endoderm, suggesting a direct role for TH in yolk utilization by these cells. An initial step in the breakdown of yolk in X. laevis is acidification of the yolk platelet. E. coqui embryos in methimazole failed to acidify their yolk platelets, but acidification was stimulated by TH indicating its role in an early step of yolk utilization. In addition to a conserved TH role in gut development, a novel regulatory role for TH in yolk utilization has evolved in these direct developers. [source] Origin of planktotrophy,evidence from early molluscsEVOLUTION AND DEVELOPMENT, Issue 4 2006Alexander Nützel SUMMARY The size of early ontogenetic shells (protoconchs) of ancient benthic molluscs suggests that feeding larvae occurred at about 490 myr (approximately, transition from Cambrian to Ordovician). Most studied Ordovician protoconchs were smaller than Cambrian ones, indicating smaller Ordovician eggs and hatchlings. This suggests substitution of nutritious reserve matter such as yolk by plankton as an energy source for larvae. The observed size change represents the first direct empiric evidence for a late Cambrian to Ordovician switch to planktotrophy in invertebrate larvae. It corroborates previous hypotheses about a possible polyphyly of planktotrophy. These hypotheses were primarily based on molecular clock data of extant clades with different types of larva, change in the overall body size, as well as increasing predation pressure on Early Paleozoic sea floors. The Early Ordovician is characterized by an explosive radiation of benthic suspension feeders and it was suggested that planktotrophy would prolongate escape from benthic predation on hatchlings. This biological escalation hypothesis does not fully explain why planktotrophy and suspension feeding became important at the same time, during a major biodiversification. An additional factor that probably included availability of nutrients must have played a role. We speculate that an increasing nutrient supply and availability of photoautotrophic plankton in world oceans have facilitated both planktotrophy and suspension feeding, which does not exclude a contemporaneous predation-driven escalation. It is very likely that the evolution of planktotrophy as well as increasing predation contributed to the Ordovician radiation. [source] Early development and segment formation in the centipede, Strigamia maritima (Geophilomorpha)EVOLUTION AND DEVELOPMENT, Issue 2 2004Ariel D. Chipman Summary Geophilomorph centipedes exhibit a number of unique characteristics that make them of particular developmental and evolutionary interest. Segment numbers in geophilomorphs are higher than in any other centipedes, ranging from 27 to 191. They may be constant within a species, presenting in extreme form the "counting" problem in development, or they may vary,a situation that provides us with the opportunity to study naturally occurring variation in segment numbers. All their segments are generated during embryogenesis, a situation unlike that in the more basal centipede orders, which generate only a fraction of their 15 trunk segments in the embryo and develop the rest postembryonically. Here we provide a foundation for further developmental studies of the Geophilomorpha, building on the one study that has been conducted to date, on the coastal species Strigamia maritima. Development begins with the migration of nuclei to the surface of the egg, which then condense to form an embryonic rudiment of more than 20,000 cells, covering an entire hemisphere. During early development, the embryo can be divided into two distinct areas: a large terminal disc of apparently undifferentiated tissue and the germ-band, which has a clear anteroposterior axis and differentiated segments. The germ-band forms from the anterior of the terminal disc and extends anteriorly as the disc contracts. New segments are formed at the posterior margin of the germ-band. Once the process of segmentation ends, the germ-band folds and sinks into the yolk. We note that the classic description of centipede development, by Heymons more than a century ago, contains a fundamental error in the identification of the axes and hence in the interpretation of early segmentation. [source] Localization of RNAs in oocytes of Eleutherodactylus coqui, a direct developing frog, differs from Xenopus laevisEVOLUTION AND DEVELOPMENT, Issue 6 2003Yvonne M. Beckham SummaryEleutherodactylus coqui develops directly on land to a frog. The large 3.5-mm oocyte of E. coqui has enough yolk to allow development without a feeding tadpole. In the smaller Xenopus laevis oocyte, 1.3 mm in diameter, mRNAs involved in germ layer formation, such as VegT and Vg1, are localized to the vegetal cortex of the oocyte. We hypothesized that an animal shift has occurred in the localization of the E. coqui Orthologs of VegT and Vg1 due to the large egg size. Through a combination of degenerate reverse transcriptase polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE), we cloned 1634 bp of EcVegT and 1377 bp of EcVg1. Northern blot analysis shows that the lengths of these transcripts are 2.5 kb and 1.3 kb, respectively. This result suggests that we have obtained the complete Vg1 transcript, although this transcript has an extremely short 3, untranslated region compared with X. laevis, 256 bp and 1268 bp, respectively. Zygotic expression of EcVegT closely resembles that of VegT, supporting their orthology. Radioactive RT-PCR and in situ hybridization demonstrated the presence of EcVegT and EcVg1 predominantly near the animal pole of the oocyte. RT-PCR showed that the animal blastomeres, formed from the first horizontal cleavage, inherit half of the EcVegT and EcVg1 transcripts, although they contain only about 1% of the embryo volume. Our results indicate major differences between the molecular organization of the eggs of X. laevis and E. coqui. [source] Surface-Protected Etching of Mesoporous Oxide Shells for the Stabilization of Metal NanocatalystsADVANCED FUNCTIONAL MATERIALS, Issue 14 2010Qiao Zhang Abstract Nanoparticles of transition metals, particularly noble metals, are widely used in catalysis. However, enhancing their stability during catalytic reactions has been a challenge that has limited the full use of the benefits associated with their small size. In this Feature Article, a general "encapsulation and etching" strategy for the fabrication of nanocatalyst systems is introduced in which catalyst nanoparticles are protected within porous shells. The novelty of this approach lies in the use of chemical etching to assist the creation of mesopores in a protective oxide shell to promote efficient mass transfer to encapsulated metal nanoparticles. The etching process allows for the direct transformation of dense silica coatings into porous shells so that chemical species can reach the catalyst surface to participate in reactions while the shells act as physical barriers against aggregation of the catalyst particles. By using the surface-protected etching process, both yolk,shell and core,satellite type nanoreactors are synthesized and their utilization in liquid- and gas-phase catalysis is demonstrated. The thermal and chemical stability of the metallic cores during catalytic reactions is also investigated, and further work is carried out to enhance recyclability via the introduction of superparamagnetic components into the nanoreactor framework. [source] Mechanisms of egg contamination by Salmonella EnteritidisFEMS MICROBIOLOGY REVIEWS, Issue 4 2009Inne Gantois Abstract Salmonella Enteritidis (SE) has been the major cause of the food-borne salmonellosis pandemic in humans over the last 20 years, during which contaminated hen's eggs were the most important vehicle of the infection. Eggs can be contaminated on the outer shell surface and internally. Internal contamination can be the result of penetration through the eggshell or by direct contamination of egg contents before oviposition, originating from infection of the reproductive organs. Once inside the egg, the bacteria need to cope with antimicrobial factors in the albumen and vitelline membrane before migration to the yolk can occur. It would seem that serotype Enteritidis has intrinsic characteristics that allow an epidemiological association with hen eggs that are still undefined. There are indications that SE survives the attacks with the help of antimicrobial molecules during the formation of the egg in the hen's oviduct and inside the egg. This appears to require a unique combination of genes encoding for improved cell wall protection and repairing cellular and molecular damage, among others. The distribution of fry leaving the redd was strongly peaked, 80% being caught within a 2-week period. Early in the dispersal period, all fry leaving the redd had remnants of yolk sac and had not fed; by half way through the dispersal period, no fry had any visible yolk, but 35% still had empty stomachs. Fry leaving the redd during the first half of the dispersal period tended to settle in different first feeding sites than those dispersing later. Predation on fry by larger salmonids was frequent, especially during dispersal. Coefficients of variation for length, weight and condition factor increased significantly over the study period and for individually recognised fry, growth rates varied markedly. Thus, individual salmon fry differ in physical status on emergence from the redd and these differences are amplified during the first few weeks after emergence. [source] Spatial distribution and feeding habits of Pacific cod (Gadus macrocephalus) larvae in Mutsu Bay, JapanFISHERIES OCEANOGRAPHY, Issue 2 2002Tetsuya Takatsu The spatial distributions and feeding habits of Pacific cod (Gadus macrocephalus) larvae, and the spatial distributions of copepod nauplii and copepodites, their main prey, were examined in Mutsu Bay from February to March during 1989,92. Yolk-sac larvae were caught at 30,45 m depth at the bay mouth. Larvae without yolk were collected at 8,45 m depth at the bay mouth and the inner part of the bay, and large larvae were chiefly found in the bay. This geographical pattern in larval size may have been because of transport to the inner part of Mutsu Bay by the Tsugaru Warm Current. The dominant taxa of copepod nauplii and copepodites in the diet and the environment changed each year. Larvae fed mainly on abundant taxa in the environment, suggesting that larvae are opportunistic feeders. Nauplii and copepodites were abundant in the bay, especially in 1992. Copepodites were slightly more abundant in the diet of cod larvae in 1992 than in 1991, but this difference was smaller than in the environment. In addition, larvae with empty digestive tracts were scarce in 1991 and 1992. Prey concentrations in the bay in 1991 and 1992 seem to have been high enough to sustain most Pacific cod larvae. Radder Summary 1One of the primary axes of life-history variation involves the proportion of embryonic development for which the offspring is retained within its parent's body; understanding trade-offs associated with prolonging that period thus is a critical challenge for evolutionary ecology. 2Prior to oviposition, most oviparous squamate reptiles retain developing eggs in utero for about one-third of embryogenesis; the strong conservatism in this trait is a major puzzle in reptilian reproduction. To clarify fitness consequences of this prolonged uterine retention, we need to experimentally modify the trait and examine the effects of our manipulation. 3We used transdermal application of corticosterone to induce gravid scincid lizards (Bassiana duperreyi) to lay their eggs ,prematurely', with relatively undeveloped embryos. Corticosterone application induced females to oviposit sooner (mean of 5·41 ± 0·51 days post-treatment) at earlier embryonic developmental stage (27 ± 0·21) than did controls (13·2 ± 1·22 days; embryonic stage 30·4 ± 0·16). 4Corticosterone levels in the egg yolk were unaffected by maternal treatment, so effects of earlier oviposition should not be confounded by endocrine disruption of embryogenesis. Nonetheless, early oviposition reduced hatchling fitness. Hatching success was lower, incubation periods post-laying were increased, and neonates from eggs laid at earlier embryonic stages were smaller and slower. 5These results suggest that retention of developing eggs in utero by oviparous squamates enhances maternal fitness, and does so via modifications to offspring phenotypes rather than (for example) due to accelerated developmental rates of eggs in utero compared to in the nest. 6More generally, our data support optimality models that interpret interspecific variation in the duration of maternal,offspring contact in terms of the selective forces that result from earlier vs. later termination of that maternal investment. JAWOR Summary 1Despite considerable recent interest in plasma and yolk testosterone (T) in female birds, relatively little is known about environmental regulation of female T, individual variation in female T or the relationship between plasma and yolk T. 2In breeding females of a wild population of dark-eyed junco (Junco hyemalis), we assessed variation in the responsiveness of the hypothalamo-pituitary-gonadal (HPG) axis to a challenge with gonadotropin-releasing hormone (GnRH) by measuring circulating T before and 30 min after a standardized injection of GnRH. We asked whether response to challenge varied seasonally or with stage of reproduction and whether it was repeatable within individuals or related to T deposited in eggs. 3Initial and post-challenge levels of T were measured using enzyme immunoassay. In a subset of these females, luteinising hormone (LH) was measured using radioimmunoassay (RIA). In addition, eggs were collected from nests of 15 females that had received a GnRH challenge, and yolk T was measured using RIA. 4During most of the breeding season, plasma T did not increase in response to GnRH. GnRH consistently caused increases in plasma T only during the 7 days before oviposition, when females were rapidly depositing yolk in eggs but had not yet begun to lay them. Among a small subset of females we found a positive correlation between the magnitude of this increase in plasma T in response to GnRH during egg development and the amount of T deposited in the yolk of eggs collected at a later time. 5These results suggest that ovarian response to GnRH-induced increases in LH is greatest when females are actively depositing yolk into eggs. Factors that stimulate the release of GnRH during egg formation may result in higher levels of plasma T which could influence adult female behaviour. Further, because plasma T was correlated with later yolk T, factors that stimulate GnRH release may also lead to higher levels of yolk T potentially influencing offspring development or behaviour. CASSEY Summary 1Carotenoids are a diverse group of organic compounds that function as important antioxidants and immunostimulants and are of particular importance to developing embryos and young birds. In addition, they constitute the majority of yellow to red hues in the integumentary pigments of birds. 2We investigated the maternally derived carotenoid concentrations and balances in the yolks of eight European passerine species successfully introduced and abundant in New Zealand. Specifically, we addressed whether variation in maternally derived yolk carotenoids were related to phylogeny, integument coloration, native vs introduced distribution, breeding habitat and laying sequence. 3Across species, the concentration and balance of carotenoids deposited in yolks varied significantly. Egg carotenoid concentration was positively associated with the occurrence of male carotenoid pigmented body regions. Carotenoid concentrations differed between agricultural habitat types within New Zealand but not between samples from New Zealand and Europe. Controlling for the differences among species, and among clutches within species, increased egg carotenoid concentrations were significantly associated with decreasing fresh egg mass and eggs laid earlier in the laying sequence. 4We conclude that variability in the concentration and balance of carotenoids deposited in the egg yolk imply different relative roles for ecological and phylogenetic factors that warrant further investigation, both within and across species. Gorman Abstract Innate immunity protects juvenile and adult vertebrates and invertebrates against potential pathogens; however, it is unknown when developing embryos become immune competent and just how they are guarded from infection. To address these questions, we studied the effect of immune challenge on early stage eggs of the tobacco hornworm, Manduca sexta. We detected many immune-related proteins and mRNAs in naive eggs. Upon immune challenge, antimicrobial protein genes were up-regulated, and antibacterial activity increased. Antimicrobial protein mRNAs and lysozyme were present in the extra-embryonic tissues of immune-challenged eggs; in addition, melanization in response to bacteria occurred in the yolk but not embryonic tissues. We conclude that the extra-embryonic tissues of early stage M. sexta eggs are immune competent and likely protect the developing embryo from infection. We suggest that innate immune responses of extra-embryonic tissues may be a common mechanism for protecting early embryos. Kalaikannan Summary The effects of whole egg powder (WEP), dried albumen (DA), dried yolk (DY) at a 1% level and liquid egg (LE) at a 3.7% level on the quality characteristics and refrigerated storage (4 ± 1 °C) stability of chicken patties made with meat and by-products of broiler-spent hens were evaluated. The addition of egg powders significantly enhanced the emulsion stability (ES) and product yield compared with control and LE. There was a marked increase in moisture content and sensory attributes by the addition of egg powders. During the refrigerated storage (4 ± 1 °C), thio-barbituric acid number (TBARS), aerobic plate count (APC) and psychrophilic count increased in all samples. Incorporation of DA and DY significantly reduced the TBARS number and APC during the storage over control. The addition of DA, DY and WEP significantly lowered the psychrophilic counts. Treated and control patties packaged in LDPE (low-density polyethylene) pouches were acceptable for 20 days and 15 days, respectively. We report on a study of reproduction and development in the Mediterranean vermetid gastropod Vermetus triquetrus from the SE coast of Spain. It is a gonochoristic species. The egg capsules are attached to the inside of the shell, and females brood up to 22 capsules simultaneously (more often 4,10). The capsules hold 10,61 eggs or embryos; the uncleaved eggs are yolk-rich, with a mean diameter of 377.3 ,m. A distinct polar lobe occurs during the first cleavage, and blastomere D has discernible qualities after the 4-cell stage. The formation of the mesentoblast 4d occurs at the transition from the 24-cell stage to the 25-cell stage. Gastrulation begins after the 36-cell stage. Internal yolk is the major source of nutrition for the encapsulated embryos, but some nurse eggs (, 12%) and some sibling larvae are also ingested by the developing embryos. Hatching occurs during the swimming/crawling pediveliger stage, and metamorphosis is completed outside the capsules soon after hatching. Hence, larval development in Vermetus triquetrus is lecithotrophic intracapsular, with a short free-swimming/crawling phase. Depending on availability, mothers may thus influence offspring quality by depositing variable amounts of carotenoids into the eggs. Sex allocation theory predicts that in good quality environments, females should invest into offspring of the sex that will provide larger fitness return, generally males. 2In a field experiment we tested whether female great tits bias their investment towards males when carotenoid availability is increased, and whether male offspring of carotenoid-supplemented mothers show higher body condition. We partially cross-fostered hatchlings to disentangle maternal effects from post-hatching effects, and manipulated hen flea Ceratophyllus gallinae infestation to investigate the relationship between carotenoid availability and resistance to ectoparasites. 3As predicted, we found that carotenoid-supplemented mothers produced males that were heavier than their sisters at hatching, while the reverse was true for control mothers. This suggests that carotenoid availability during egg production affects male and female hatchlings differentially, possibly via a differential allocation to male and female eggs. 4A main effect of maternal supplementation became visible 14 days after hatching when nestlings hatched from eggs laid by carotenoid-supplemented mothers had gained significantly more mass than control nestlings. Independently of the carotenoid treatment, fleas impaired mass gain of nestlings during the first 9 days in large broods only and reduced tarsus length of male nestlings at an age of 14 days, suggesting a cost to mount a defence against parasites. 5Overall, our results suggest that pre-laying availability of carotenoids affects nestling condition in a sex-specific way with potentially longer-lasting effects on offspring fitness. Sahin Abstract Aims:Campylobacter jejuni, a food-borne human pathogen, is widespread in poultry; however, the sources of infection and modes of transmission of this organism on chicken farms are not well understood. The objective of this study was to determine if vertical transmission of C. jejuni occurs via eggs. Methods and Results: Using a temperature differential method, it was shown that Campylobacter had limited ability to penetrate the eggshell. When C. jejuni was directly inoculated into the egg yolk and the eggs were stored at 18°C, the organism was able to survive for up to 14 days. However, viability of C. jejuni was dramatically shortened when injected into the albumen or the air sac. When freshly laid eggs from Campylobacter -inoculated specific pathogen-free (SPF) layers were tested, C. jejuni -contamination was detected in three of 65 pooled whole eggs (5,10 eggs in each pool) via culture and PCR. However, the organism was not detected from any of the 800 eggs (80 pools), collected from the same SPF flock, but kept at 18°C for 7 days before testing. Likewise, Campylobacter was not recovered from any of 500 fresh eggs obtained from commercial broiler-breeder flocks that were actively shedding Campylobacter in faeces. Also, none of the 1000 eggs from broiler breeders obtained from a commercial hatchery were positive for Campylobacter. Conclusions: These results suggest that vertical transmission of C. jejuni through the egg is probably a rare event and does not play a major role in the introduction of Campylobacter to chicken flocks. Significance and Impact of the Study: Control of Campylobacter transmission to chicken flocks should focus on sources of infection that are not related to eggs. Thus late-hatching nestlings in asynchronously hatching clutches somehow overcome any age- and size-related disadvantages of hatching after their nest-mates. One possible way for them to do this is for female House Wrens to add maternal androgens to the yolk of late-hatching eggs. We tested this hypothesis in a wild population of House Wrens that produces both asynchronously and synchronously hatching clutches. Yolks of eggs from both types of clutches were biopsied and the eggs returned to their nests to hatch. Radioimmunoassays revealed that total androgen levels in the yolk varied within and among clutches. However, total androgen levels in yolks did not vary predictably with egg position in either synchronously or asynchronously hatching clutches. Thus, deposition of androgens in yolk did not follow the expected pattern based on the potential for sibling competition in House Wrens. [source] |
