Odontogenic keratocyst expresses vascular endothelial growth factor: an immunohistochemical study

Background:, Vascular endothelial growth factor (VEGF) expression may act as a sensitive measure of the angiogenic potential of a lesion. Furthermore, VEGF has been implicated in the pathogenesis of cystic tumors and inflammatory odontogenic cysts. Thus, we studied the expression of VEGF in the epithelium of odontogenic keratocyst (OK) in association with cell proliferation and apoptosis. Methods:, Forty-two cases of OK, 26 cases of dentigerous cyst (DC), and 15 cases of residual cyst (RC) were retrospectively examined by immunohistochemistry for VEGF, Ki67/Mib-1 and anti-caspase-3. For VEGF and caspase-3, the intensity of immunostaining was qualitatively assessed, while for the evaluation of Ki67 the average number of positively stained nuclei in 10 high-power microscopic fields (×400) was calculated. Results:, The VEGF expression was stronger in OK when compared with DC (P < 0.007). The rate of nuclear Ki67 expression in OK was significantly higher than that in DC (P < 0.001) and RC (P < 0.001). Cytoplasmic caspase-3 expression was statistically more intense in RC than in OK (P = 0.001) or DC (P < 0.001). A statistically significant correlation was seen in OK for Ki67 (P < 0.001) and VEGF (P = 0.023), but not for caspase-3. Multiple regression analysis revealed a linear relationship between VEGF and Ki67. Conclusions:, The VEGF was expressed in the epithelium of OK, DC, and RC with a variable intensity, and in OK VEGF expression was related to Ki67. It is suggested that VEGF expression by the odontogenic epithelium is not induced solely by inflammation. [source]


Organic-walled dinoflagellate cyst production in relation to upwelling intensity and lithogenic influx in the Cape Blanc region (off north-west Africa)

SUMMARY Fossil dinoflagellate cyst assemblages are increasingly used in paleoclimatic research to establish paleoenvi-ronmental reconstructions. To obtain reliable reconstructions, it is essential to know which physical factors influence the cyst production. Most information about the relationship between variations in physical parameters and cyst production is known from middle and higher latitudes. Information from the (sub)tropics is rare. To increase this information, the temporal variation in cyst assemblages from the upwelling area off north-west Africa (off Mauritania) has been compared to environmental conditions of the upper water column. Samples were collected by the sediment trap CB9, off north-west Africa (Cape Blanc, 21°15,2,N, 20°42,2,W) between 11 June 1998 and 7 November 1999 at 27.5-day intervals. Off Cape Blanc, upwelling occurs throughout the year with variable intensity. This region is also characterized by frequently occurring Saharan dust storms. Seasonal variations in dust input, upwelling intensity and sea surface temperature are reflected by the production of organic-walled dinoflagellate cyst assemblages. Several cyst taxa are produced throughout the sampling interval, with the highest fluxes at times of strongest upwelling relaxation and/or dust input (Echinidinium aculeatum Zonneveld, Echini-din ium delicatum Zonneveld, Echinidinium granulaturn Zonneveld, Echinidinium spp., Impagidinium aculeatum (Wall) Lentin et Williams, Impagidinium sphaeri-cum (Wall) Lentin et Williams, Protoperidinium americanum (Gran et Braarud) Balech, Protoperidinium stellatum (Wall in Wall et Dale) Rochon etal., Protoperidinium spp., Selenopemphix nephroides (Benedek) Benedek et Sarjeant and Selenopemphix quanta (Bradford) Matsuoka). Species such as, for example, Bitectatodinium spongium (Zonneveld) Zonneveld et Jurkschat and Impagidinium patulum (Wall) Stover et Evitt do not show any production pattern related to a particular season of the year or to specific environmental conditions in the upper water column. The production of cysts of Protoperidinium monospinum (Paulsen) Zonneveld et Dale is restricted to intervals with increased nutrient concentrations in upper waters when sea surface temperatures at the sampling site is below approximately 24°C. Oxidative/nitrosative stress and peroxiredoxin 2 are associated with grade and prognosis of human renal carcinoma,

Peroxiredoxins (Prxs) 1,6 were assessed in 138 renal cell carcinomas (RCC) using immunohistochemistry and selected samples by Western blotting analysis. Oxidative/nitrosative damage was evaluated using nitrotyrosine immunoreactivity. The expressions of Prxs were correlated with tumor grade and survival and nitrotyrosine reactivity. Non-malignant kidney tubular cells showed positivity with variable intensity for all six Prxs. In RCCs, most cases were positive for Prxs 1 and 2, while only 15,20% of tumors showed expression for Prxs 3 and 4. Prx 2 was associated with tumors of a lower grade (p=0.009) and with a lower frequency of distant metastases (p=0.046). Patients with tumors expressing Prx2 had better prognosis (p=0.027). Instead, nitrotyrosine was significantly associated with high grade tumors (p=0.001). Compared with the non-malignant kidney tubular cells, low Prx expression in the tumor cells can make them more susceptible to oxidative damage. Prx 2 was more abundantly expressed in low grade tumors, suggesting that this protein could play a role in preventing the development of oxidative damage, which in turn can lead to the activation of pathways leading to aggressive tumors. Role of Environmental Factors on the Structure and Spectroscopic Response of 5,-DNA,Porphyrin Conjugates Caused by Changes in the Porphyrin,Porphyrin Interactions

Abstract We have explored the utility, strength, and limitation of through-space exciton-coupled circular dichroism in determination of the secondary structure of optically active chromophoric nanoarrays using the example of end-capped porphyrin, and metalloporphyrin,oligodeoxynucleotide conjugates. We put special emphasis on the explanation of the origin and significance of the distinctive multiple bands in the CD spectra (trisignate and tetrasignate CD bands). Such CD profiles are often observed in chiral aggregates or multichromophoric arrays but have never before been studied in detail. We found that variation of temperature and ionic strength has a profound effect on the geometry of the porphyrin,DNA conjugates and thus the nature of electronic interactions. At lower temperatures and in the absence of NaCl all three 5,-DNA,porphyrin conjugates display negative bisignate CD exciton couplets of variable intensity in the Soret region resulting from through-space interaction between the electric transition dipole moments of the two end-capped porphyrins. As the temperature is raised these exciton couplets are transformed into single positive bands originating from the porphyrin,single-strand DNA interactions. At higher ionic strengths and low temperatures, multisignate CD bands are observed in the porphyrin Soret region. These CD signature bands originate from a combination of intermolecular, end-to-end porphyrin,porphyrin stacking between duplexes and porphyrin,DNA interactions. The intermolecular aggregation was confirmed by fluorescence and absorption spectroscopy and resonance light scattering. DeVoe theoretical CD calculations, in conjunction with molecular dynamics simulations and Monte Carlo conformational searches, were used to mimic the observed bisignate exciton-coupled CD spectra as well as multiple CD bands. Calculations correctly predicted the sign and shape of the experimentally observed CD spectra. These studies reveal that the exciton-coupled circular dichroism is a very useful technique for the determination of the structure of optically active arrays. [source]


SMS pain diary: a method for real-time data capture of recurrent pain in childhood

Abstract Objectives:, To capture recurrent pain in children aged 9,15 years reported by short message service (SMS) and to test the compliance of such reporting in a pilot study. Methods:, After instructions, 15 children reported their pain six times a day on SMS for a week and the compatibility of the reporting was evaluated. The pain was expressed and reported on three variables: intensity captured using a numeric rating scale (NRS-11), duration in minutes and a verbal pain-related disability scale with six alternatives (0,5). The validity of this scale was tested in 37 children, and the reliability in a test,retest procedure in 20 children. Results:, Good compliance reporting the three variables intensity and duration of pain as well as pain-related disability on SMS was indicated. Support for construct validity and reliability of the verbal instrument for pain-related disability was achieved. Conclusion:, The study supports the hypothesis that pain experience expressed as intensity, duration and pain-related disability can be captured in real time by SMS in an inexpensive and compliant way in children aged 9,15 years. Validity and reliability was indicated for the constructed verbal pain-related disability scale. Further studies are needed to further confirm these findings. [source]
