The notion of a,market,where traffickers prey on the ,supply' of migrants that flows across international borders to meet the ,demand' for labour has become a central trope among anti-trafficking development organizations. This article problematizes such,economism,by drawing attention to the oscillating cross-border migration of Lao sex workers within a border zone between Laos and Thailand. It illuminates the incongruity between the recruitment of women into the sex industry along the Lao,Thai border and the market models that are employed by the anti-trafficking sector. It discusses the ways in which these cross-border markets are conceived in a context where aid programming is taking on an increasingly important role in the politics of borders. The author concludes that allusions to ideal forms of knowledge (in the guise of classic economic theory) and an emphasis on borders become necessary for anti-trafficking programmes in order to make their object of intervention legible as well as providing post-hoc rationalizations for their continuing operation. [source] Trafficking of neurokinin receptors: regulation, mechanism and functionEXPERIMENTAL DERMATOLOGY, Issue 9 2004N. Bunnett Cellular responses to agonists of G-protein-coupled receptors (GPCRs) depend in large part on the trafficking of receptors between the plasma membrane and intracellular locations. Receptor activation usually triggers rapid endocytosis of receptors, which either recycle to the cell surface or are targeted for degradation, depending on the receptor in question and the nature of the stimulation. Activation of neurokinin receptors (NKRs) induces membrane translocation of G-protein receptor kinases, which phosphorylate the receptors and ,-arrestins, which interact with phosphorylated receptors. ,-arrestins: 1) uncouple receptors from G-proteins to mediate desensitization; 2) are adaptors for clathrin and AP-2 and mediate clathrin and dynamin-dependent endocytosis of receptors; and 3) interact with components of the MAP kinase pathway such as src, and thereby determine the subcellular location and function of activated MAP kinases. The fate of endocytosed NKRs depends on the receptor and the nature of the stimulus. Transient stimulation with low concentrations of SP (1 nm, 10 min) induces rapid recycling of the NK1R from superficially located endosomes by a mechanism that is mediated by rab4a and rab11a. Higher concentrations of SP (10 nm) induce rab5a-dependent trafficking of the NK1R to perinuclear sorting endosomes and a gradual recycling to the plasma membrane. Continuous stimulation with high concentrations of SP (100 nm, 180 min) induces NK1R ubiquitination and trafficking for degradation. The fate of endocytosed receptors also depends on their interaction with ,-arrestins. The NK1R forms stable high-affinity interactions with both ,-arrestins 1 and 2 at the plasma membrane and in endosomes, whereas the NK3R interacts transiently only with ,-arrestin 2 at the cell surface. The nature of these interactions is specified by domains in the intracellular loop 3 and the carboxyl terminus and determine the rate of recycling and resensitization of these receptors. [source] AATYK1A phosphorylation by Cdk5 regulates the recycling endosome pathwayGENES TO CELLS, Issue 7 2010Tetsuya Takano Trafficking of recycling endosomes (REs) is regulated by the small GTPase, Rab11A; however, the regulatory mechanism remains elusive. Apoptosis-associated tyrosine kinase 1A (AATYK1A) is a Ser/Thr kinase expressed highly in brain. We have recently shown that AATYK1A localizes to Rab11A-positive RE and is phosphorylated at Ser34 by cyclin-dependent kinase 5 (Cdk5). Here, we have investigated a role of AATYK1A and its phosphorylation in recycling endosomal trafficking using Chinese hamster ovary-K1 (CHO-K1) cells. AATYK1A localizes predominantly to Rab11A-positive pericentrosomal endocytic recycling compartment (ERC). Phosphorylation at Ser34 of AATYK1A disrupts its accumulation in the pericentrosomal ERC. Consistently, phosphorylation-mimic mutant (AATYK1A-S34D) did not accumulate in the ERC and additionally attenuated ERC formation. ERC formation suppression can be reversed by constitutively active Rab11A-Q70L, suggesting a functional link between AATYK1A phosphorylation and Rab11A activity. Although no direct interaction between AATYK1A and Rab11A could be detected, the exchange of guanine nucleotides bound to Rab11A was significantly reduced in the presence of the phosphorylation-mimic AATYK1A-S34D. Together, our results reveal a regulatory role for AATYK1A in the formation of pericentrosomal ERC. They furthermore indicate that Cdk5 can disrupt ERC formation via Ser34 phosphorylation of AATYK1A. Finally, our data suggest a mechanism by which AATYK1A signaling couples Cdk5 to Rab11A activity. [source] Trafficking in Persons and Development: Towards Greater Policy CoherenceINTERNATIONAL MIGRATION, Issue 4 2010Gergana Danailova-Trainor Poverty is often regarded as the "root cause" of trafficking, but the linkages between poverty, a lack of development and trafficking are complex. For example, there is some evidence to suggest that victims of cross-border trafficking are more likely to originate from middle-income rather than lower-income countries. Trafficking and development have tended to be treated as very separate policy areas and the assessment of the development impact of counter-trafficking programmes is still at an early stage. This paper outlines a possible framework for a more evidence-based approach to understanding the linkages between trafficking, trafficking policy and human development. The paper argues that the human development gains from greater mobility could be significantly enhanced if there was greater coherence between policies to combat trafficking and policies to promote development. [source] Canadian Policy on Human Trafficking: A Four-year Analysis1INTERNATIONAL MIGRATION, Issue 4 2005Jacqueline Oxman-Martinez ABSTRACT This article introduces readers to Canadian Government policy and practice surrounding human trafficking since the adoption of the United Nations (UN) Protocol on Trafficking in 2000. After offering an overview of the UN Protocol, the article reviews and critically analyses Canada's efforts in the three key areas of the Protocol: prevention of human trafficking, protection of trafficking victims, and the prosecution of traffickers. Since the beginning of our research, progress has been made in Canadian policy responses. The Government began by developing and implementing its tools for the prosecution of traffickers, thereby responding to most of the prosecution recommendations of the UN Protocol. Different government agencies are also coordinating their efforts to implement prevention projects, both in source countries and at home, including awareness-raising campaigns, education campaigns, and policy development collaborations. However, the more structural elements of prevention have yet to be adequately addressed. Finally, without shifting their basic border control framework, Canadian government agencies are in the process of improving the protection of trafficking victims who are intercepted in law enforcement operations or who come forward for help. These protection measures would be strengthened further if migrants' rights were explicitly protected by law, something that has failed to occur given recent prioritization of crime and security. The formal protection of victims, as implemented to some degree in several European and American policies, is introduced for comparison. The article concludes with the remaining challenges that face Canadian policy makers, particularly in terms of shifting away from current focus on crime and security to the protection and promotion of the human rights of trafficking victims. LA POLITIQUE CANADIENNE EN MATIÈRE DE TRAITE DES ÊTRES HUMAINS : UNE ANALYSE SUR QUATRE ANS Cet article présente au lecteur la politique et la pratique du gouvernement canadien en matière de traite des êtres humains depuis l'adoption, en 2000, du Protocole additionnel à la Convention des Nations unies contre la criminalité transnationale organisée visant à prévenir, réprimer et punir la traite des personnes, en particulier des femmes et des enfants. Après avoir présenté le Protocole dans son ensemble, l'article examine et analyse de façon critique l'action du Canada dans trois domaines essentiels du Protocole : la prévention de la traite, la protection des victimes et la punition des trafiquants. Depuis le début de notre recherche, le Canada a fait un pas en avant dans les mesures politiques adoptées. Le gouvernement a commencé par élaborer et mettre en oeuvre des mécanismes pour punir les trafiquants, donnant de ce fait suite à la plupart des recommandations du protocole des Nations unies en la matière. Par ailleurs, différents organes de l'État coordonnent leurs efforts pour mettre en place des projets de prévention, tant dans les pays d'origine que sur le territoire canadien, avec des campagnes de sensibilisation et d'éducation, et des collaborations en matière d'élaboration des politiques. Il reste néanmoins à trouver une solution adéquate aux aspects les plus structurels de la prévention. Enfin, sans s'éloigner du cadre fondamental du contrôle des frontières, les services gouvernementaux canadiens améliorent actuellement la protection des victimes de la traite interceptées lors d'opérations de police ou qui viennent demander de l'aide. Ces mesures de protection seraient davantage renforcées si les droits des migrants étaient explicitement inscrits dans la loi, ce qui n'est pas le cas en raison de la priorité accordée à la répression de la criminalité et à la sécurité. La protection formelle des victimes, telle que mise en oeuvre jusqu'à un certain point par des mesures appliquées en Europe et aux États-Unis, est présentée à titre de comparaison. Tras hacer un repaso del Protocolo de las Naciones Unidas, este artículo examina y analiza críticamente los empeños del Canadá en tres esferas clave del Protocolo: prevención de la trata de personas, protección de las víctimas de la trata, y enjuiciamiento de los traficantes. Desde que se iniciara este estudio se han observado progresos en las respuestas políticas canadienses. El Gobierno comenzó desarrollando y llevando a la práctica sus instrumentos para la sanción y enjuiciamiento de los traficantes, respondiendo así a la mayoría de las recomendaciones de enjuiciamiento que contiene el Protocolo de las Naciones Unidas. Varias instituciones gubernamentales también coordinan sus esfuerzos con miras a la puesta en práctica de proyectos de prevención, tanto en los países de origen como en el Canadá, incluyendo campañas de concienciación, campañas educativas y colaboraciones con miras al desarrollo de políticas. [source] On the Way to a Better Future: Belgium as Transit Country for Trafficking and Smuggling of Unaccompanied Minors1INTERNATIONAL MIGRATION, Issue 4 2005Ilse Derluyn ABSTRACT During the last decade, irregular border crossings emerged as a new element in international migratory flows, with smuggling and trafficking networks being an essential part. Many people are compelled to use these networks to realize their dream of a better living, and for many this "promised land" is the United Kingdom (UK). Belgium has important sea connections with the UK, and is, therefore, an important migration transit zone, although many migrants are intercepted on Belgian territory during their tempt to reach the UK. Some are unaccompanied or separated children and adolescents, minors travelling without parent(s) or a legal caregiver. This study aims to gain insight about this population of unaccompanied minors travelling to the UK. We use the situation in Zeebrugge, one of Belgium's main ports, as a case study. We analysed 1,093 data files of unaccompanied minors intercepted in Zeebrugge, and carried out participatory observation at the shipping police station. The intercepted unaccompanied minors are mainly male, between 15 and 18 years of age, and from an Asian or Eastern European country. Of the 899 unique persons found in the data files, 113 were intercepted several times. After the interception, the Aliens Office gives the majority (82.9%) an identity document without a requirement to leave Belgium, while 15.3 per cent must leave Belgium immediately or within five days. In 82.9 per cent of the cases, a child protection officer is contacted to make a decision about the situation. In 67.2 per cent of these cases, no child protection measure is taken, and the minor may leave the police station; in 32 per cent of the cases, the minor is transferred to a centre, mostly crisis reception. Almost all unaccompanied minors are convinced they want to reach the UK to create a better livelihood, join a family member, or escape a difficult political situation. Nevertheless, most travel in difficult circumstances; are scared; and lack essential information about life in the UK, their possibilities in Belgium, what will happen if they are transferred to a centre, and so forth. Most minors also do not want to be transferred to a centre, and many , although not all , disappear again from the centres. This study has several implications concerning the kind of decisions taken by the legal authorities, the necessary physical and psycho-social care and the availability of an interpreter and social worker during the interception, the number of reception places and the care in these centres, and the tasks of the legal guardian. Finally, some limitations of the study are mentioned. VERS UN AVENIR MEILLEUR : LA BELGIQUE COMME PAYS DE TRANSIT POUR LA TRAITE ET L'INTRODUCTION CLANDESTINE DE MINEURS NON ACCOMPAGNÉS Au cours de la dernière décennie, le franchissement irrégulier des frontières est apparu comme un nouvel élément des flux migratoires internationaux, dont les réseaux de traite et d'introduction clandestine sont un aspect essentiel. Beaucoup de gens sont forcés d'utiliser ces réseaux pour réaliser leur rêve d'une vie meilleure et pour beaucoup, cette « terre promise », c'est le Royaume-Uni. La Belgique ayant d'importantes liaisons maritimes avec le Royaume-Uni con-stitue de ce fait une importante zone de transit pour les migrations, bien que de nombreux migrants soient interceptés sur le territoire belge alors qu'ils tentent d'atteindre le Royaume-Uni. Certains d'entre eux sont des enfants et des adoles-cents non accompagnés, séparés, des mineurs qui voyagent sans parent(s), sans personne qui en ait la garde juridique. Cette étude vise à mieux connaître cette population de mineurs non accompagnés voyageant en direction du Royaume-Uni. Nous prenons comme cas concret la situation à Zeebrugge, l'un des principaux ports belges. Nous avons analysé 1 093 fichiers de données concernant des mineurs non accompagnés interceptés à Zeebrugge, et nous sommes livrés à une observation participative au poste de police du port. Cette étude a diverses implications concernant le genre de décisions que pren-nent les autorités juridiques, les nécessaires soins physiques et psychosociaux pendant l'interception ainsi que la présence d'un interprète et d'un travailleur social, le nombre de places et les soins dans les centres d'accueil, les tâches de la personne qui a la responsabilité légale du mineur. Enfin, certaines limites de cette étude sont évoquées. Son muchas las personas que se ven obligadas a recurrir a estas redes para hacer realidad su sueño de una vida mejor y para muchos "la tierra prometida" es el Reino Unido. Bélgica tiene importantes conexiones marítimas con el Reino Unido y, por consi-guiente, es una zona de tránsito de la migración sumamente importante, aunque muchos migrantes son interceptados en territorio belga en su intento por llegar al Reino Unido. Entre las personas interceptadas se encuentran niños y adoles-centes, menores de edad que viajan solos, sin sus padres o tutor legal. Este estudio tiene por objeto comprender cómo esta población de menores no acom-pañados viaja al Reino Unido. Long Contemporary trafficking operations transform traditional bride wealth and marriage exchanges (prestations) by treating women's sexuality and bodies as commodities to be bought and sold (and exchanged again) in various Western capitals and Internet spaces. Such operations are also global with respect to scale, range, speed, diversity, and flexibility. Propelling many trafficking exchanges are political economic processes, which increase the trafficking of women in times of stress, such as famine, unemployment, economic transition, and so forth. However, the disparity between the global market operations, which organize trafficking, and the late nineteenth century social/public welfare system of counter-trafficking suggests why the latter do not effectively address women's risks and may even expose them to increased levels of violence and stress. Drawing on historical accounts, anthropological theory, and ethnographic work in Viet Nam and Bosnia and Herzegovina, this essay examines how specific cultural practices embedded in family and kinship relations encourage and rationalize sexual trafficking of girls and young women in times of stress and dislocation. The essay also analyses how technologies of power inform both trafficking and counter-trafficking operations in terms of controlling women's bodies, sexuality, health, labour, and migration. By analysing sexual trafficking as a cultural phenomenon in its own right, such an analysis seeks to inform and address the specific situations of girls and young women, who suffer greatly from the current migration regimes. Taran This review summarizes main trends, issues, debates, actors and initiatives regarding recognition and extension of protection of the human rights of migrants. Its premise is that the rule of law and universal notions of human rights are essential foundations for democratic society and social peace. Evidence demonstrates that violations of migrants' human rights are so widespread and commonplace that they are a defining feature of international migration today. About 150 million persons live outside their countries; in many States, legal application of human rights norms to non-citizens is inadequate or seriously deficient, especially regarding irregular migrants. Extensive hostility against, abuse of and violence towards migrants and other non-nationals has become much more visible worldwide in recent years. Research, documentation and analysis of the character and extent of problems and of effective remedies remain minimal. Resistance to recognition of migrants' rights is bound up in exploitation of migrants in marginal, low status, inadequately regulated or illegal sectors of economic activity. Unauthorized migrants are often treated as a reserve of flexible labour, outside the protection of labour safety, health, minimum wage and other standards, and easily deportable. Evidence on globalization points to worsening migration pressures in many parts of the world. Processes integral to globalization have intensified disruptive effects of modernization and capitalist development, contributing to economic insecurity and displacement for many. Extension of principles in the Universal Declaration of Human Rights culminated in the 1990 International Convention on the Protection of the Rights of All Migrant Workers and Members of Their Families. With little attention, progress in ratifications was very slow until two years ago. A global campaign revived attention; entry into force is likely in 2001. Comparative analysis notes that ILO migrant worker Conventions have generally achieved objectives but States have resisted adoption of any standards on treatment of non-nationals. A counter-offensive against human rights as universal, indivisible and inalienable underlies resistance to extension of human rights protection to migrants. A parallel trend is deliberate association of migration and migrants with criminality. Trafficking has emerged as a global theme contextualizing migration in a framework of combatting organized crime and criminality, subordinating human rights protections to control and anti-crime measures. Intergovernmental cooperation on migration "management" is expanding rapidly, with functioning regional intergovernmental consultative processes in all regions, generally focused on strengthening inter-state cooperation in controlling and preventing irregular migration through improved border controls, information sharing, return agreements and other measures. Efforts to defend human rights of migrants and combat xenophobia remain fragmented, limited in impact and starved of resources. Nonetheless, NGOs in all regions provide orientation, services and assistance to migrants, public education and advocating respect for migrants rights and dignity. Several international initiatives now highlight migrant protection concerns, notably the UN Special Rapporteur on Human Rights of Migrants, the Global Campaign promoting the 1990 UN Convention, UN General Assembly proclamation of International Migrants Day, the 2001 World Conference Against Racism and Xenophobia, anti-discrimination activity by ILO, and training by IOM. Suggestions to governments emphasize the need to define comprehensive, coordinated migration policy and practice based on economic, social and development concerns rather than reactive control measures to ensure beneficial migration, social harmony, and dignified treatment of nationals and non-nationals. NGOs, businesses, trade unions, and religious groups are urged to advocate respect for international standards, professionalize services and capacities, take leadership in opposing xenophobic behaviour, and join international initiatives. Need for increased attention to migrants rights initiatives and inter-agency cooperation by international organizations is also noted. [source] Trafficking: A Perspective from AsiaINTERNATIONAL MIGRATION, Issue 3 2000Ronald Skeldon The main theme of this article is market development and trafficking as a business. It touches upon most of the aspects of the phenomenon, which have been encountered elsewhere, and translates them into the relatively unfamiliar context of many of the Asian and South-East Asian economies. Equally, the literature cited is also probably unfamiliar. Themes touched upon include democratization, inter-state relations, human rights, and scale and perspectives, together with the problems of definitions, theory, and the reliability of data. The directions and characteristics of trafficking flows together with routes and border control are also considered. Coordinated official responses to criminality and criminal organizations, as well as to trafficked individuals, are beginning to emerge. There is a note of caution sounded that contextual and cultural perspectives, particularly on sex workers, must be viewed somewhat differently to those in Western societies. The article concludes that as long as countries in Asia maintain their policies of restrictive immigration, trafficking can be expected to continue and almost certainly increase. This is because accelerating development creates demand for labour at various skill levels and because even in times of recession migrants and brokers will seek to side-step attempts to expel immigrants and restrict access to labour markets. The elimination of trafficking is unlikely to be realistically achieved through legislation and declarations of intent but by improvements in the socio-economic status of the population. [source] Trafficking and Human Smuggling: A European PerspectiveINTERNATIONAL MIGRATION, Issue 3 2000John Salt The article reviews the empirical evidence for trafficking and human smuggling in Europe. It argues that a market for irregular migration services has emerged, in which the mechanisms and forms of organization are still relatively unknown. Irregular migrants using these services are exposed both to unscrupulous service providers and to the immigration and policing authorities, thereby generating a dependence on safeguards provided by the trafficking networks. Thus a symbiosis has developed between trafficker and trafficked. The enormous interest and concern for trafficking and human smuggling in governmental, inter-governmental and non-governmental organizations, in the media and popular opinion, is running ahead of theoretical understanding and factual evidence. This has implications for policy measures designed to combat trafficking and human smuggling, which may not work and also have unintended side effects. The article begins with a discussion of the main conceptual and definitional issues confronting researchers and politicians. This is followed by an assessment of the main theoretical approaches that have been developed and an evaluation of current statistical knowledge. Information on the organizational structure of trafficking organizations is then reviewed, followed by a summary of the characteristics of migrants involved, based on empirical studies that have been carried out. Its implications differ, however, in the northern and southern parts of the state. In the latter, especially in the capital, Acre, the development of a local market of urban consumers is closely connected to police corruption and the illegal use of violence by law enforcement agencies. In the former, where machinery for the social redistribution of illegal income seems to be more effective, the cocaine trade is contributing to a degree of prosperity, thanks in particular to recent growth in the service sector. While violence is, comparatively speaking, less necessary as a guarantee of social control in that region, the control exercised by drug barons and business people over the executive branches of the state means that political life as a whole is criss-crossed by relationships forged in the criminal world. FOP lymphocytes, a model system for exploring the BMP pathway in these patients, exhibit a defect in BMPRIA internalization and increased activation of downstream signaling, suggesting that altered BMP receptor trafficking underlies ectopic bone formation in this disease. Introduction: Fibrodysplasia ossificans progressiva (FOP) is a severely disabling disorder characterized by progressive heterotopic ossification of connective tissues. Whereas the genetic defect and pathophysiology of this condition remain enigmatic, BMP4 mRNA and protein are overexpressed, and mRNAs for a subset of secreted BMP antagonists are not synthesized at appropriate levels in cultured lymphocytes from FOP patients. These data suggest involvement of altered BMP signaling in the disease. In this study, we investigate whether the abnormality is associated with defective BMP receptor function in lymphocytes. Materials and Methods: Cell surface proteins were quantified by fluorescence-activated cell sorting (FACS). Protein phosphorylation was assayed by immunoprecipitation and immunoblotting. Protein synthesis and degradation were examined by [35S]methionine labeling and pulse-chase assays. mRNA was detected by RT-PCR. Results: FOP lymphocytes expressed 6-fold higher levels of BMP receptor type IA (BMPRIA) on the cell surface compared with control cells and displayed a marked reduction in ligand-stimulated internalization and degradation of BMPRIA. Moreover, in control cells, BMP4 treatment increased BMPRIA phosphorylation, whereas BMPRIA showed ligand-insensitive constitutive phosphorylation in FOP cells. Our data additionally support that the p38 mitogen-activated protein kinase (MAPK) signaling pathway is a major BMP signaling pathway in these cell lines and that expression of inhibitor of DNA binding and differentiation 1 (ID-1), a transcriptional target of BMP signaling, is enhanced in FOP cells. Conclusions: These data extend our previous observations of misregulated BMP4 signaling in FOP lymphocytes and show that cell surface overabundance and constitutive phosphorylation of BMPRIA are associated with a defect in receptor internalization. Resistance to cisplatin represents a major obstacle to effective cancer therapy because clinically significant levels of resistance quickly emerge after treatment. Based on previous studies indicating abnormal plasma membrane protein trafficking in cisplatin-resistant (CP-r) cells, Fluorescence (Alexa Fluor)-labeled cisplatin was used to determine whether this defect altered the trafficking and localization of cisplatin by comparing drug sensitive KB-3-1 and KB-CP-r cells. Alexa Fluor,cisplatin was readily internalized and localized throughout the KB-3-1 cells, but overall fluorescence decreased in KB-CP-r cells, as detected by flow cytometry (FACS) and confocal microscopy. Only punctate cytoplasmic staining was observed in KB-CP-r cells with less fluorescence observed in the nucleus. Colocalization experiments with a Golgi-selective stain indicate the involvement of Golgi-like vesicles in initial intracellular processing of Alexa Fluor conjugated cisplatin complexes. As detected using an antibody to Alexa Fluor,cisplatin, cisplatin complex-binding proteins (CCBPs) were reduced in membrane fractions of single-step cisplatin-resistant KB-CP.5 cells, and increased in the cytoplasm of KB-CP.5 cells compared to KB-3-1 cells. CCBPs localized to lower density fractions in KB-CP.5 cells than in KB-3-1 cells as determined by iodixanol gradient centrifugation. Previous studies indicate that the thermal element of fever augments lymphocyte migration across high endothelial venules (HEVs) of lymphoid organs by increasing the intravascular display of a gatekeeper trafficking molecule, intercellular adhesion molecule-1 (ICAM-1). Here, we evaluated the spatio-temporal relationship between the thermal induction of intravascular ICAM-1 and lymphocyte trafficking. Methods: Intravascular ICAM-1 density was quantified by immunofluorescence staining in mice exposed to fever-range whole-body hyperthermia (39.5±0.5°C). ICAM-1,dependent lymphocyte trafficking was measured in short-term homing assays. Results: A linear relationship was observed between the duration of heat treatment and intravascular ICAM-1 density in HEVs with maximal responses requiring sustained (i.e., five hours) thermal stress. Circulating lymphocytes were found to sense incremental changes in ICAM-1 on HEVs, such that trafficking is proportional to the intravascular density of ICAM-1. We further identified a hydroxamate-sensitive shedding mechanism that restores ICAM-1 expression to homeostatic levels following the cessation of thermal stress. Conclusions: The time-dependent response to thermal stress indicates that ICAM-1 density governs the efficiency of lymphocyte interactions with HEVs in vivo. These studies highlight the dynamic role of the microcirculation in promoting immune surveillance during febrile inflammatory responses. These lead, in turn, to hypopigmentation, prolonged bleeding and ceroid deposition. Positional cloning strategies have identified five mouse HPS genes. Two orthologous human diseases (HPS1 and HPS2) have likewise been identified. At least four of the five mouse genes encode proteins involved in the regulation of intracellular vesicle trafficking. The pearl (HPS2) and mocha genes encode the beta3A and delta subunits, respectively, of the AP-3 adaptor complex, which captures organelle membrane proteins at the trans-Golgi apparatus. The protein products of the pallid and gunmetal genes are also important components of the vesicular trafficking machinery. The former interacts with a t-SNARE, syntaxin13, and the latter is the alpha subunit of Rab geranylgeranyltransferase, which renders Rab proteins sufficiently lipophilic to function at their target membranes. The gene responsible for this disorder, dystonin/Bpag1, encodes several alternatively spliced forms of a cytoskeletal linker protein. Neural isoforms of dystonin/Bpag1 are predicted to link actin filaments to microtubules. Consistent with this, previous observations have demonstrated that the cytoskeleton within sensory neurites of dt mice is perturbed. Also, recent results have indicated that a neural isoform of dystonin/Bpag1 interacts with the dynein motor complex. Because microtubule organization and dynein motor function are essential for trafficking, we hypothesized that this process would be perturbed in dt sensory neurons. Here, we demonstrate that cultured primary dorsal root ganglion (DRG) neurons express dystonin/Bpag1 and that loss of this expression causes an increase in apoptosis and a decrease in average neurite length. In contrast, detailed examination showed that the organization of microtubules is indistinguishable in DRG neuronal cultures from neonatal dt and wild-type mice. In addition, the steady-state distribution of several molecules and organelles is unchanged in these cultures. Furthermore, the speeds of mitochondrial movement in both anterograde and retrograde directions were comparable in dt and wild-type sensory neurons cultured from neonatal mice. Thus, dystonin/Bpag1 is not essential for microtubule network assembly since the microtubule network is intact in short-term cultures of sensory neurons from neonatal mice lacking this protein. [source] Trafficking of the human transferrin receptor in plant cells: effects of tyrphostin A23 and brefeldin ATHE PLANT JOURNAL, Issue 5 2006Elena Ortiz-Zapater Summary Plant cells possess much of the molecular machinery necessary for receptor-mediated endocytosis (RME), but this process still awaits detailed characterization. In order to identify a reliable and well-characterized marker to investigate RME in plant cells, we have expressed the human transferrin receptor (hTfR) in Arabidopsis protoplasts. We have found that hTfR is mainly found in endosomal (Ara7- and FM4-64-positive) compartments, but also at the plasma membrane, where it mediates binding and internalization of its natural ligand transferrin (Tfn). Cell surface expression of hTfR increases upon treatment with tyrphostin A23, which inhibits the interaction between the YTRF endocytosis signal in the hTfR cytosolic tail and the ,2-subunit of the AP2 complex. Indeed, tyrphostin A23 inhibits Tfn internalization and redistributes most of hTfR to the plasma membrane, suggesting that the endocytosis signal of hTfR is functional in Arabidopsis protoplasts. Co-immunoprecipitation experiments show that hTfR is able to interact with a , -adaptin subunit from Arabidopsis cytosol, a process that is blocked by tyrphostin A23. In contrast, treatment with brefeldin A, which inhibits recycling from endosomes back to the plasma membrane in plant cells, leads to the accumulation of Tfn and hTfR in larger patches inside the cell, reminiscent of BFA compartments. Therefore, hTfR has the same trafficking properties in Arabidopsis protoplasts as in animal cells, and cycles between the plasma membrane and endosomal compartments. The purpose of this study was to clarify whether abnormalities other than exonic mutations, such as splicing disorders, decreased mRNA expression levels, or membrane transport abnormalities of SCN5A, play a role in the pathogenesis of Brugada syndrome. We analyzed all SCN5A exons and splice sites using genomic DNA from 23 Brugada syndrome patients. We also analyzed the mRNA obtained from RV cardiomyocytes using real time PCR and sequencing, to study the expression levels and splicing patterns of SCN5A. The localization of SCN5A was examined by immunofluorescence analysis. A de novo heterozygous G to A transversion in a 5, splice junction of the intron between exons 21 and 22 was detected in 1 patient. In the mRNA analysis of Brugada syndrome patients without a mutation of SCN5A no splicing abnormalities were detected, and the SCN5A mRNA levels were similar to those of normal controls. Immunofluorescence analyses revealed that SCN5A is located on the surface membrane not only in the RV cardiomyocytes of normal controls but also in those with Brugada syndrome. We can confirm that some Brugada syndrome patients without exonic mutations in SCN5A had no other SCN5A abnormalities, including any involving the location of the SCN5A protein. These results suggest the involvement of other proteins in the pathogenesis in Brugada syndrome. [source] |
