Kinds of Tolerant Terms modified by Tolerant Selected AbstractsThree novel thiopurine S-methyltransferase allelic variants (TPMT*20, *21, *22) , association with decreased enzyme function,,HUMAN MUTATION, Issue 9 2006Elke Schaeffeler Abstract The genetic polymorphism of the thiopurine S-methyltransferase, TPMT, comprises at least 21 alleles causing three distinct drug metabolism phenotypes termed normal/high, intermediate, and deficient methylators. In consequence, adverse drug reactions may occur if standard doses of thiopurines are applied routinely. Genetic prediction of the methylator phenotype as a basis for dose selection requires the extensive knowledge of single nucleotide polymorphisms occurring naturally in the population. Here we describe three novel missense variants in the TPMT gene which were associated with an intermediate red blood cell TPMT activity in three Caucasians. The following alleles were designated: TPMT*20 (c.712A>G), *21 (c.205C>G), and *22 (c.488G>C). No further genetic variations in remaining coding regions as well as the 5,flanking region of TPMT were identified. These sequence variants are present in highly conserved nucleotide positions of the TPMT gene throughout various mammalian species and in zebra fish, and are predicted to be intolerant when the functional consequences of variations were analyzed using SIFT (Sorting Intolerant From Tolerant) algorithm. In Caucasians the occurrence of these genetic variants appears to be extremely rare since none of these alleles were identified in a randomly selected control population of 1048 individuals. © 2006 Wiley-Liss, Inc. [source] Drought Tolerance in Cotton: Involvement of Non-enzymatic ROS-Scavenging CompoundsJOURNAL OF AGRONOMY AND CROP SCIENCE, Issue 4 2009L. Yildiz-Aktas Abstract Compounds with reactive oxygen species (ROS)-scavenging ability were studied. High-performance liquid chromatography (HPLC) pattern of polyphenols, contents of proline and carotenoids, and antiradical (AR) capacity were determined. The malonyldialdehyde (MDA) level was also assessed. Tolerant and sensitive cotton genotypes were compared, grown in the Aegean region of Turkey at normal (field capacity) and limited (1/3 field capacity) water supply. Chlorogenic acid isomers and flavonoids were identified in HPLC pattern of polyphenols. At normal water supply, the tolerant genotype was distinguished by a higher content of all polyphenol types, higher proline, carotenoids and AR capacity and lower MDA level compared with the sensitive genotype. In plants subjected to water deficit, a decline of all polyphenol compounds, carotenoids and AR capacity was observed. However, this response was less pronounced in the tolerant than in the sensitive genotype, i.e. despite the stress conditions imposed, the tolerant plants maintained a more effective defence system. The data are corroborated by the weaker structural membrane damage in the drought-exposed tolerant vs. sensitive genotype, according to the MDA test. Hence, diverse chemical types are involved in the non-enzymatic ROS-scavenging system of cotton plants and can be related to the drought tolerance of this important crop. [source] Assessment of Growth, Physiological and Biochemical Parameters and Activities of Antioxidative Enzymes in Salinity Tolerant and Sensitive Basmati Rice VarietiesJOURNAL OF AGRONOMY AND CROP SCIENCE, Issue 6 2007M. Singh Abstract This investigation was undertaken to compare the level of salinity tolerance of the newly bred CSR-30 basmati rice variety with that of the salinity sensitive HBC-19 and Pokkali rice varieties. Twenty-one-day-old hydroponically raised seedlings at 6 and 12 dS m,1 were investigated for growth, photosynthetic rate, chlorophyll content, ribulose-1,5-bisphosphate carboxylase-oxygenase (Rubisco) activity, relative water content (RWC), membrane stability index (MSI), lipid peroxidation, Na/K ratio and activities and gene expression of various isoforms of antioxidative enzymes. Salinity stress led to reduction in shoot length, leaf area, dry weight, RWC, MSI, rate of photosynthesis, chlorophyll content and Rubisco activity in all the three rice varieties. The levels of reduction in these parameters were maximal in HBC-19 followed by those in CSR-30 and Pokkali respectively. Cumulative superoxide dismutase (SOD) activity increased in Pokkali and CSR-30 in consonance with increase in salinity stress while it decreased in HBC-19. The Mn-SOD activity however, was enhanced in all three varieties in the presence of salinity stress while the activities of Fe-SOD, Cu/Zn-SOD and ascorbate peroxidase were decreased in HBC-19 when compared with CSR-30 and Pokkali. The activity of catalase (CAT) was higher in HBC-19 when compared with its activity in CSR-30 and Pokkali. The levels of gene expressions of the three isoforms of SOD ascertained by reverse transcriptase polymerase chain reaction were not necessarily indicative of the activities of the corresponding enzymes. Thus, despite the maximal enhancement in gene expression of Fe-SOD in HBC-19 in response to salinity stress, the activity of this enzyme in HBC-19 remained low. Similarly, despite a marginal increase in gene expression of Cu-Zn SOD in the three varieties, its activity was significantly higher in Pokkali and CSR-30 when compared with that in HBC-19. A significant enhancement in the activity of CAT at 12 dS m,1 in HBC-19 when compared with CSR-30 and Pokkali might confer a degree of tolerance to H2O2 stress in this variety in the presence of higher levels of NaCl at the seedling stage. [source] Differential response of antioxidant compounds to salinity stress in salt-tolerant and salt-sensitive seedlings of foxtail millet (Setaria italica)PHYSIOLOGIA PLANTARUM, Issue 4 2000N. Sreenivasulu The modulation of antioxidant components was comparatively analysed in a salt-tolerant (cv. Prasad) and salt-sensitive (cv. Lepakshi) cultivar of foxtail millet (Setaria italica L.) under different NaCl concentrations. Under conditions of salt stress, the salt-tolerant cultivar exhibited increased total superoxide dismutase (SOD) and ascorbate peroxidase (APX) activity, whereas both enzyme activities decreased in acutely salt-stressed seedlings of the sensitive cultivar. At 200 mM NaCl, the tolerant foxtail millet cultivar responded with induction of cytosolic Cu/Zn-SOD and the Mn-SOD isoform at the protein level. The induced accumulation of the cytosolic Cu/Zn-SOD protein/activity is positively correlated with an elevated level of the cytosolic APX gene activity. The elevated cytosolic Cu/Zn-SOD and cytosolic APX activity correlates with an induced accumulation of their transcripts. Tolerant 5-day-old seedlings grown during high salinity treatment (200 mM NaCl) contained a lower amount of Na+ ions and showed a lower electrolyte leakage than sensitive seedlings. In conclusion, our comparative studies indicate that salt-induced oxidative tolerance is conferred by an enhanced compartment-specific activity of the antioxidant enzymes in response to compartment-specific signals. [source] Evaluation of Juglans species for resistance to Phytophthora cinnamomi:,differences in isolate virulence and response to fosetyl-AlFOREST PATHOLOGY, Issue 3 2009A. Belisario Summary Phytophthora is considered as an important pathogen on walnut, and severe losses are reported in European as well as in American walnut stands. Though several Phytophthora spp. are known to attack walnut, P. cinnamomi is considered the most virulent and widespread in southern Europe. Up to now, no walnut species or hybrid is known to have a high resistance level towards P. cinnamomi. Efforts are addressed in finding rootstock material graft compatible with English walnut and resistant/tolerant to P. cinnamomi. The extension of P. cinnamomi lesions on five Juglans species was studied to find out sources of resistance/tolerance to this pathogen. Walnut species clustered into two main groups, J. hindsii, J. nigra, and J. mandshurica were the less susceptible to the colonization of P. cinnamomi, while J. regia and J. sieboldiana were the most susceptible. On this account, J. mandshurica represents the best alternative as rootstock because its employment overcomes the risk of the occurrence of black line disease, it has good level of resistance to Agrobacterium temefaciens and Brenneria nigrifluens, and it is tolerant to Xanthomonas arboricola pv. juglandis. J. mandshurica is also compatible in cross-pollinations with J. regia and J. nigra. Differences in virulence of P. cinnamomi isolates was assessed and a marked interaction between species and isolate emerged. Treatment with fosetyl-Al by dipping was mainly efficient in reducing the length P. cinnamomi lesions, and an interaction between species and treatment was evident with the highest efficacy on J. regia and J. sieboldiana. The mother's immune system reacts against the fetus and there is therefore a risk of destruction of or damage to the fetus. We are now beginning to understand some of the mechanisms that protect the fetus, but, when these are defective, intrauterine growth retardation or abortions may ensue. However, the cytokines of this maternal immune response to the fetus also monitor different phases of pregnancy, starting with effects on the ovarium and involving preparation of the decidua for the implantation of the fertilized egg, the growth of the trophoblasts, the production of hormones important for the pregnancy and finally of the prostaglandins that induce delivery. B. The mother and the child. Human milk contains anti-idiotypic antibodies which after transfer to the offspring are capable of enhancing antibody responses. Human milk contains numerous leucocytes especially during early lactation. There is increasing evidence that milk lymphocytes are taken up by the breastfed infant, which seems to have become tolerant to maternal HLA. Breastfeeding mothers are therefore good donors of renal transplants to their breastfed offspring in adult age, too. Conclusion: It is suggested that the milk lymphocytes may be taken up by the offspring and that immunological information is thereby carried over from the mother. This mechanism may explain why breastfeeding seems to confer enhanced protection against infections also some years after the termination of breastfeeding. Due to the decentralized nature of such a p2p system the carrier does not know anything about the current size, performance, and stability of its application. In this paper, we present an entirely distributed and scalable algorithm to monitor a running p2p network. The snapshot of the system enables a telecommunication carrier to gather information about the current performance parameters of the running system as well as to react to discovered errors. Bentley We examined the abundance of small mammal species in forests, corridors, remnants of araucarian vine forest, and Araucaria cunninghamii plantations and pastures. None of the forest mammal species persisted following conversion of forest to pasture. Plantations supported lowered abundances of a subset of forest species that were mainly habitat generalists with respect to their occurrence in different floristic types of undisturbed native forest. Within plantations, an increased subcanopy cover was associated with a more forest-like small mammal assemblage. Species' responses to habitat fragmentation varied. The floristic habitat generalists were largely tolerant of habitat fragmentation, their abundance being similar in forests, corridors, and remnants, and were capable of persisting in remnants a few hectares in area. Floristic habitat specialists were vulnerable to habitat fragmentation and thus were abundant in continuous forest, were less abundant in corridors, and were generally absent from remnants. Species that avoid the corridor matrix and are therefore constrained to the corridor may be disadvantaged by the linearity of the habitat, consistent with the predictions of central-place foraging theory. Although small remnants and corridors provide habitat for some species, those that are more specialized in their use of undisturbed habitat types require the retention or reestablishment of large intact areas. Resumen: La pérdida de hábitat y la fragmentación son amenazas importantes para la sobrevivencia de la fauna que depende del bosque. Examinamos la abundancia de especies de mamíferos pequeños en bosques, corredores, y en remanentes de bosques de vid araucarios y en plantaciones de Araucaria cunninghamii y pastizales. Ninguna de las especies de mamíferos del bosque persistió después de la conversión del bosque a pastizal. Las plantaciones favorecieron abundancias menores de un conjunto de especies del bosque integrado principalmente por generalistas de hábitat con respecto a su presencia en diferentes tipos florísticos de bosque nativo sin perturbar. Dentro de las plantaciones, cuanto mayor era la cobertura por debajo del dosel más se parecía el ensamblaje de mamíferos pequeños al del bosque. Las respuestas de las especies a la fragmentación del hábitat fueron variadas. Los generalistas del hábitat florístico por lo general toleraban la fragmentación del hábitat ( la abundancia en bosques, corredores y remanentes era similar) y fueron capaces de persistir en remanentes de unas pocas hectáreas de extensión. Sundkvist Abstracts Aims Sorbitol and myo -inositol levels and morphology of sural nerve were compared with nerve function and clinical neuropathy in men with diabetic, impaired (IGT), and normal glucose tolerance. Methods After neurography of sural nerve and determinations of sensory thresholds for vibration, warm and cold on the foot, whole nerve sural nerve biopsy was performed in 10 men with Type 1 diabetes mellitus, 10 with IGT, and 10 with normal glucose tolerance. Polyol levels were assessed by gas,liquid chromatography/mass spectrometry. Results Sural nerve amplitudes were significantly lower and sorbitol levels significantly higher in diabetic patients (median (interquartile range)) (3.7 (3.5) ,V and 643 (412) pmol/mg protein, respectively) both compared with IGT (11.3 (10.6) ,V; P = 0.04 and 286 (83) pmol/mg protein; P = 0.0032, respectively) and normally glucose tolerant (10.0 (11.6); P = 0.0142 and 296 (250) pmol/mg protein; P = 0.0191, respectively) subjects. There were no differences in nerve morphology between the three groups. Nerve myo -inositol levels correlated, however, positively with cluster density (rs = 0.56; P = 0.0054). In diabetic and IGT subjects, sural nerve amplitudes (2.6 (3.8) vs. 12.1 (10.6) ,V; P = 0.0246) and myelinated nerve fibre density (MNFD; 4076 (1091) vs. 5219 (668) nerve fibres/mm2; P = 0.0021) were significantly lower in nine subjects with clinical neuropathy than in 10 without. Conclusions Nerve degeneration (i.e. MNFD) correlated with clinical neuropathy but not with glucose tolerance status whereas nerve myo -inositol levels positively correlated with signs of nerve regeneration (i.e. increased cluster density). [source] Selection of preadapted populations allowed Senecio inaequidens to invade Central EuropeDIVERSITY AND DISTRIBUTIONS, Issue 4 2008Oliver Bossdorf ABSTRACT Invasive species often evolve rapidly in response to the novel biotic and abiotic conditions in their introduced range. Such adaptive evolutionary changes might play an important role in the success of some invasive species. Here, we investigated whether introduced European populations of the South African ragwort Senecio inaequidens (Asteraceae) have genetically diverged from native populations. We carried out a greenhouse experiment where 12 South African and 11 European populations were for several months grown at two levels of nutrient availability, as well as in the presence or absence of a generalist insect herbivore. We found that, in contrast to a current hypothesis, plants from introduced populations had a significantly lower reproductive output, but higher allocation to root biomass, and they were more tolerant to insect herbivory. Moreover, introduced populations were less genetically variable, but displayed greater plasticity in response to fertilization. Finally, introduced populations were phenotypically most similar to a subset of native populations from mountainous regions in southern Africa. Taking into account the species' likely history of introduction, our data support the idea that the invasion success of Senecio inaequidens in Central Europe is based on selective introduction of specific preadapted and plastic genotypes rather than on adaptive evolution in the introduced range. Ashton ABSTRACT Numerous hypotheses suggest that natural enemies can influence the dynamics of biological invasions. Here, we use a group of 12 related native, invasive, and naturalized vines to test the relative importance of resistance and tolerance to herbivory in promoting biological invasions. In a field experiment in Long Island, New York, we excluded mammal and insect herbivores and examined plant growth and foliar damage over two growing seasons. This novel approach allowed us to compare the relative damage from mammal and insect herbivores and whether damage rates were related to invasion. In a greenhouse experiment, we simulated herbivory through clipping and measured growth response. After two seasons of excluding herbivores, there was no difference in relative growth rates among invasive, naturalized, and native woody vines, and all vines were susceptible to damage from mammal and insect herbivores. Thus, differential attack by herbivores and plant resistance to herbivory did not explain invasion success of these species. In the field, where damage rates were high, none of the vines were able to fully compensate for damage from mammals. However, in the greenhouse, we found that invasive vines were more tolerant of simulated herbivory than native and naturalized relatives. Our results indicate that invasive vines are not escaping herbivory in the novel range, rather they are persisting despite high rates of herbivore damage in the field. While most studies of invasive plants and natural enemies have focused on resistance, this work suggests that tolerance may also play a large role in facilitating invasions. Using meta-analysis, we examined whether invasive species: (1) incur less damage, (2) exhibit better performance in the presence of enemies, and (3) tolerate damage more than native species. Invasive alien species did not incur less damage than native species overall. The performance of invasive alien species was reduced compared to natives in the presence of enemies, indicating the invasive alien species were less tolerant to damage than native species. However, there was no overall difference in performance of invasive alien and native species with enemies present. The damage and degree of reduction in performance of invasive alien relative to native species did not depend on relatedness to natives. Our results suggest aliens may not always experience enemy release, and enemy release may not always result in greater plant performance. [source] Ontogenetic switches from plant resistance to tolerance: minimizing costs with age?ECOLOGY LETTERS, Issue 3 2007Karina Boege Abstract Changes in herbivory and resource availability during a plant's development should promote ontogenetic shifts in resistance and tolerance, if the costs and benefits of these basic strategies also change as plants develop. We proposed and tested a general model to detect the expression of ontogenetic tradeoffs for these two alternative anti-herbivory strategies in Raphanus sativus. We found that ontogenetic trajectories occur in both resistance and tolerance but in opposite directions. The juvenile stage was more resistant but less tolerant than the reproductive stage. The ontogenetic switch from resistance to tolerance was consistent with the greater vulnerability of young plants to leaf damage and with the costs of resistance and tolerance found at each stage. We posit that the ontogenetic perspective presented here will be helpful in resolving the current debate on the existence and detection of a general resistance,tolerance tradeoff. [source] Effect of temperature on development, overwintering and establishment potential of Franklinothrips vespiformis in the UKENTOMOLOGIA EXPERIMENTALIS ET APPLICATA, Issue 2 2007Eleni Larentzaki Abstract This study investigated the effect of temperature on the development and overwintering potential of the predatory thrips Franklinothrips vespiformis (Crawford) (Thysanoptera: Aeolothripidae), a biological control agent used against glasshouse pests in continental Europe and Israel. Developmental rates increased linearly with rearing temperatures. It was estimated that 304.9 degree days, above a lower threshold temperature of 11.9 °C, were required for F. vespiformis to complete development from egg to adult eclosion. The effect of low temperatures (,5, 0, and 5 °C) was examined on adult female and larval survival. Subsequent reproductive and developmental attributes of survivors were also investigated. Lethal time experiments indicated that larval stages are more cold tolerant than adult F. vespiformis females. Surviving larvae increased their developmental times to adults with decreasing temperature and increasing exposure periods and second instars were significantly more successful than first instars in reaching adulthood. Surviving adult females decreased their oviposition rate with decreasing temperature and increasing exposure periods, and exposures to low temperatures affected the number of viable eggs produced. The results are discussed in the context of overwintering and establishment potential of F. vespiformis in the UK in the event of introducing the predatory thrips as a biological control agent against glasshouse pests. GOTO Abstract Cold and desiccation tolerance was investigated in the eggs of the band-legged ground cricket Dianemobius nigrofasciatus in relation to embryonic diapause. Diapause eggs were more tolerant to both desiccation and cold than non-diapause eggs. In addition, diapause-destined eggs on day zero (0,12 h after being laid) already showed high tolerance to these stresses before entering diapause. This clearly indicates that stress tolerance, like diapause, is controlled by photoperiod, but is not directly associated with diapause itself. Because the acquisition of stress tolerance predates the onset of diapause, it is plausible that diapause programming during some period before the onset of diapause is involved in the acquisition of stress tolerance. Weights and sizes were nearly identical in short-day and long-day eggs until day five. Sorbitol, a major sugar alcohol in eggs of D. nigrofasciatus, was accumulated at the same level in short-day and long-day eggs on days zero and five. These results indicate that the surface-to-volume ratio as well as the accumulation of sugar alcohol is not involved in the acquisition of stress tolerance. Maternal factors are clearly involved in the acquisition of stress tolerance in D. nigrofasciatus eggs, but the physiological mechanisms underlying the tolerance are still unclear. Durou Abstract Estuaries are subject to anthropogenic activities. Because the intrasedimentary worm Nereis diversicolor has ecological characteristics and bioindicator abilities, its use was pertinent in investigating the concepts and cost of tolerance to heavy metals (Cd, Cu, Zn). In this context, two approaches were carried out, performing toxicity tests and estimating energy reserves (glycogen and lipids), in populations originating from a contaminated site (Seine estuary) compared with those from a clean site (Authie estuary). Mean lethal times (LT50s) of organisms exposed to zinc from the Seine estuary were higher than those from the Authie estuary, but not of organisms exposed to Cd or Cu. The influence of animal weight and salinity on the sensitivity of worms also was studied. The biggest worms were more tolerant to zinc than the smallest ones, and worms survived longer at a reduced salinity (15,). Concentrations of glycogen and lipids in each sampling season were higher in specimens from the Authie estuary than in worms from the Seine estuary. No influence of salinity on glycogen and lipid levels was observed. Glycogen concentrations were not influenced by the weight of specimens, whereas lipid concentrations were significantly and positively correlated with weight. In conclusion, worms from the Seine estuary exhibited tolerance to Zn, and the depletion of energy reserves observed in this population could be interpreted as a cost of tolerance. © 2005 Wiley Periodicals, Inc. [source] Effects of the organophosphorus insecticide fenitrothion on growth in five freshwater species of phytoplanktonENVIRONMENTAL TOXICOLOGY, Issue 4 2001Consuelo Sabater Abstract The acute toxicity of the insecticide fenitrothion was measured using four freshwater algae (Chlorella saccharophila, Chlorella vulgaris, Scenedesmus acutus, and Scenedesmus subspicatus) and one cyanobacteria (Pseudanabaena galeata). Insecticide concentrations eliciting 50% growth reduction over 96 hr (EC50) ranged from 0.84 to 11.9 mg/L. Fenitrothion was more toxic than other pesticides studied with the same algal species such as chlorsulfuron, molinate, and pyridaphenthion. The transformation of effective concentrations of fenitrothion and other pesticides obtained from toxicity measurements into percent of the saturation level in water is used as a first evaluation of potential hazard to aquatic systems. The insecticides fenitrothion and pyridaphenthion were less hazardous than the herbicides atrazine, benthiocarb, cinosulfuron, chlorsulfuron, methyl-bensulfuron, and molinate. The two species of Chlorella and the cyanobacterium Pseudanabaena were more tolerant to fenitrothion than the two species of Scenedesmus. © 2001 John Wiley & Sons, Inc. Hill Abstract Disturbances such as dredging, storms, and bioturbation result in the resuspension of sediments. This may affect sessile organisms that live on hard substrates directly above the sediment. Localized sediment contamination exists around many Antarctic research stations, often resulting in elevated contamination loads in marine sediments. To our knowledge, the potential impact of resuspended contaminated sediments on sessile fauna has not been considered, so in the present study, we assessed the sensitivity of Antarctic spirorbid polychaetes to aqueous metals and to metal-contaminated sediments that had been experimentally resuspended. Worms were first exposed to aqueous metals, both singly and in combination, over 10 d. Spirorbid mortality was tolerant to copper (median lethal concentration [LC50], 570 ,g/L), zinc (LC50, >4,910 ,g/L), and lead (LC50, >2,905 ,g/L); however, spirorbid behavior responded to copper concentrations as low as 20,g/L. When in combination, zinc significantly reduced mortality caused by copper. A novel technique was used to resuspend sediments spiked with four concentrations of three metals (up to 450 ,g/g dry wt of copper, 525 ,g/g dry wt of lead, and 2,035 ,g/g dry wt of zinc). The response of spirorbids to unfiltered suspended sediment solutions and filtered solutions (aqueous metal exposure) was measured. Suspended sediments were toxic to filter-feeding spirorbids at concentrations approximating those found in contaminated Antarctica areas. Toxicity resulted both from aqueous metals and from metals associated with the suspended sediments, although suspended clean sediments had no impact. To our knowledge, the present study is the first to show that resuspension of contaminated sediments can be an important pathway for toxicity to Antarctic hard substrate organisms. Based on the present results, current sediment-quality guidelines used in the evaluation of Australian sediments may be applicable to Antarctic ecosystems. [source] Comparative toxicity of cadmium, zinc, and mixtures of cadmium and zinc to daphnidsENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 1 2006Joseph R. Shaw Abstract Investigations were conducted to determine acute (48-h) effects of cadmium and zinc presented individually and in combination on Ceriodaphnia dubia, Daphnia magna, Daphnia ambigua, and Daphnia pulex. Toxicity tests were conducted with single metals to determine lethal effects concentrations (lethal concentrations predicted for a given percent [x] of a population, LCx value). These were used to derive metal combinations that spanned a range of effects and included mixtures of LC15, LC50, and LC85 values calculated for each metal and species. In single-metal tests, 48-h LC50 values ranged from 0.09 to 0.9 ,mol/L and 4 to 12.54 ,mol/L for cadmium and zinc, respectively. For each metal, D. magna was most tolerant and showed a different pattern of response from all others as determined by slope of concentration,response curves. In the combined metal treatments, all daphnids showed a similar pattern of response when LC15 concentrations were combined. This trend continued with few exceptions when LC15 concentrations of cadmium were combined with LC50 or LC85 values for zinc. However, when this treatment was reversed (LC15, zinc + LC50 or LC85, cadmium), responses of all species except D. magna indicated less-than-additive effects. For C. dubia, a near complete reduction in toxicity was observed when the LC15 for zinc was combined with LC85 for cadmium. Multimetal tests with D. magna did not differ from additive. Collectively, these studies suggest that D. magna may not be representative of other cladocerans. [source] Copper toxicity thresholds for important restoration grass species of the western United States,ENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 12 2002Mark W. Abstract Copper toxicity thresholds for plant species that are used in restoration activities in western North America have not been established. As a result, ecological risk assessments must rely on toxicity thresholds established for agronomic species, which usually differ from those of species used in restoration. Thus, risk assessors have the potential for classifying sites as phytotoxic to perennial, nonagronomic species and calling for intensive remediation activities that may not be necessary. The objective of this study was to provide a better estimate of Cu toxicity thresholds for five grass species that are commonly used in restoration efforts in the western United States. We used a greenhouse screening study where seedlings of introduced redtop (Agrostis gigantea Roth.), the native species slender wheatgrass (Elymus trachycaulus [Link] Gould ex Shinners var. Pryor), tufted hairgrass (Deschampsia caespitosa [L.] Beauvois), big bluegrass (Poa secunda J. Presl var. Sherman), and basin wildrye (Leymus cinereus [Scribner&Merrill] A. Löve var. Magnar) and the agricultural species common wheat (Triticum aestivum L.) were grown in sand culture and exposed to supplemental concentrations of soluble Cu of 0 (control), 50, 100, 150, 200, 250, and 300 mg/L. We determined six measures of toxicity: the 60-d mean lethal concentration (LC50), 60-d mean effective concentration (EC50)-plant, 60-d EC50-shoot, 60-d EC50-root, phytotoxicity threshold (PT50)-shoot, and the PT50-root. Results suggest that these restoration grass species generally have higher Cu tolerance than agronomic species reported in the past. Of the species tested, redtop appeared to be especially tolerant of high levels of substrate and tissue Cu. Values of EC50-plant for restoration grasses were between 283 and 710 mg Cu/L compared to 120 mg Cu/L for common wheat. Measured PT50-shoot values were between 737 and 10,792 mg Cu/ L. These reported thresholds should be more useful for risk assessors than those currently used, which are based largely on agronomic crops. [source] Family members of relatives with alcohol, drug and gambling problems: a set of standardized questionnaires for assessing stress, coping and strainADDICTION, Issue 11 2005Jim Orford ABSTRACT Aims To describe a set of standard questionnaire measures for the assessment of the needs of family members of relatives with alcohol, drug or gambling problems, and to present evidence of their reliability and validity from a series of related studies. Design Includes cross-sectional and repeated-measurement studies. Setting and participants Family members affected by and concerned about the problem drinking or drug-taking of close relatives in treatment and non-treatment samples in the United Kingdom (white and Sikh) and Mexico City; family members of untreated heavy drinkers; and family members of problem gamblers. Measurements Four measures derived from a stress,strain,coping,support model of alcohol, drugs and gambling problems and the family: Family Member Impact scale (FMI), Symptom Rating Test (SRT), Coping Questionnaire (CQ), and Hopefulness,Hopelessness scale (HOPE). FMI, SRT and CQ assess stress, strain and coping, respectively. The exact role of HOPE in the model remains to be determined. The support component remains unmeasured. Findings Results from a number of studies support the internal reliability, discriminant and construct validity and sensitivity to change of the SRT and its two constituent scales (psychological and physical symptoms) and at least two subscales of the CQ (engaged and tolerant,inactive coping). Although showing evidence of satisfactory reliability and some evidence of discriminant validity, further work may be required on the CQ withdrawal coping subscale. Evidence suggests that the FMI is reliable and valid and may have a factor structure that will support future research (distinguishing worrying behaviour from active disturbance). HOPE is a new measure showing promising characteristics. Conclusions A set of standard measures is available for helping to assess the needs of concerned and affected family members, derived from an explicit model of the family in relation to excessive drinking, drug taking or gambling. It may have a role to play in correcting the current neglect of the needs of such family members, estimated to be in the region of nearly a million adults in Britain alone. [source] Reduced ethanol response in the alcohol-preferring RHA rats and neuropeptide mRNAs in relevant structuresEUROPEAN JOURNAL OF NEUROSCIENCE, Issue 2 2006Marc Guitart-Masip Abstract Roman rat strains, genetically selected for high (RHA) or low (RLA) active avoidance acquisition in the two-way shuttle box, differ in dopaminergic activity. These two strains appear to be a valid laboratory model of divergent sensation/novelty and substance-seeking profiles. RHA rats show higher ethanol intake and preference than do RLA rats, and it was suggested that RHA rats are more tolerant than RLA to the effects of alcohol. In the hole-board test, we found that the non-alcohol-preferring RLA rats showed enhanced responsiveness to the stimulatory effects of intraperitoneal administration of 0.25 g/kg ethanol when compared with RHA rats. In situ hybridization analysis showed higher levels of preprodynorphin in the accumbens shell and higher levels of preproenkephalin in the cingulate cortex in RHA rats. RLA rats showed higher levels of enkephalin gene transcripts in restricted areas of the dorsal striatum. Finally, differences in cholecystokinin gene transcript, suggestive of a different arrangement of certain interneurons, were found in different cortical areas. The differences in peptide gene expression found between the two strains might reflect the differences in alcohol preference and sensitivity. RHA rats may have more predictive value than other rodent alcoholism models, as high initial tolerance to ethanol is a risk factor for alcoholism in humans. When using 3-formylthiophene, the 2-arylated thiophenes were obtained with regioselectivities of 76,86,%, whereas the arylation of 3-formylthiophene diethyl acetal gave the 5-arylated thiophenes with regioselectivities of 64,88,%. These reactions were performed by using only 0.1 mol-% of the catalyst. Moreover, this procedure has been found to be tolerant to a variety of functional groups on the aryl bromide such as formyl, propionyl, benzoyl, nitrile, or nitro. Wise As the evolutionary importance of plant tolerance of herbivory is increasingly appreciated, more and more studies are not just measuring a plant's tolerance, but are comparing tolerance among plant genotypes, populations, species, and environments. Here, we suggest that caution must be taken in such comparative studies in the choice of measurement scales (and data transformations) for damage levels and plant performance. We demonstrate with a simple scenario of two plant groups of equal tolerance how the choice of scales can lead one to infer that the first group is more tolerant, the second group is more tolerant, or the two groups are equally tolerant,using the identical dataset. We conclude that to make reliable, logically consistent inferences when comparing tolerances among groups of plants, damage and performance should both be on an additive scale or both on a multiplicative scale. Lu Abstract:, Epidermal gene therapy may benefit a variety of inherited skin disorders and certain systemic diseases. Both in vivo and ex vivo approaches of gene transfer have been used to target human epidermal stem cells and achieve long-term transgene expression in immunodeficient mouse/human chimera models. Immunological responses however, especially in situations where a neoantigen is expressed, are likely to curtail expression and thereby limit the therapy. In vivo gene transfer to skin has been shown to induce transgene-specific immune responses. Ex vivo gene transfer approaches, where keratinocytes are transduced in culture and transplanted back to patient, however, may avoid signals provided to the immune system by in vivo administration of vectors. In the current study, we have developed a stable epidermal graft platform in immunocompetent mice to analyze host responses in ex vivo epidermal gene therapy. Using green fluorescent protein (GFP) as a neoantigen and an ex vivo retrovirus-mediated gene transfer to mouse primary epidermal cultures depleted of antigen-presenting cells (APCs), we show induction of GFP-specific immune responses leading to the clearance of transduced cells. Similar approach in immunocompetent mice tolerant to GFP resulted in permanent engraftment of transduced cells and continued GFP expression. Activation of transgene-specific immune responses in ex vivo gene transfer targeted to keratinocytes require cross-presentation of transgene product to APCs, a process that is most amenable to immune modulation. This model may be used to explore strategies to divert transgene-specific immune responses to less destructive or tolerogenic ones. [source] Tolerance to low O2: lessons from invertebrate genetic modelsEXPERIMENTAL PHYSIOLOGY, Issue 2 2006Gabriel G. Haddad There have been extensive studies and experiments on cells, tissues and animals that are susceptible to low O2, and many pathways have been discovered that can lead to injury in mammalian tissues. But other pathways that can help in the survival of low O2 have also been discovered in these same tissues. It should be noted, however, that the mechanisms that can lead to better survival in susceptible mammalian tissues have quantitatively a ,narrow range' for recovery, since these tissues are inherently at risk. Another strategy for understanding the susceptibility of organisms is to learn about pathways used by anoxia-resistant animals. Approximately a decade ago, I and my co-workers discovered that one such animal, Drosophila melanogaster, is very tolerant of low O2. Here, I detail some of the studies that we performed and the strategies that we developed to understand the mechanisms that underlie the fascinating resistance of Drosophila to measured partial pressure of O2 of zero. We employed three ideas to try to address our questions: (1) mutagenesis screens to identify loss-of-function mutants; (2) microarrays on adapted versus naïve flies; and (3) studying cell biology and physiology of genes that seem important in flies and mammals. The hope is to learn from these studies about the fundamental basis of tolerance to the lack of O2, and with this knowledge be able to develop better therapies for the future. CAMANHO ABSTRACT Delamination is one of the predominant forms of failure in laminated composite structures, especially when there is no reinforcement in the thickness direction. To develop composite structures that are more damage tolerant, it is necessary to understand how delamination develops, and how it can affect the residual performance. A number of factors such as residual thermal stresses, matrix-curing shrinkage and manufacturing defects affect how damage will grow in a composite structure. It is important to develop computationally efficient analysis methods that can account for all such factors. The objective of the current work is to apply a newly developed decohesion element to investigate the debond strength of skin-stiffener composite specimens. The process of initiation of delaminations and the propagation of delamination fronts is investigated. The numerical predictions are compared with published experimental results. [source] Identification and classification of genes required for tolerance to high-sucrose stress revealed by genome-wide screening of Saccharomyces cerevisiaeFEMS YEAST RESEARCH, Issue 2 2006Akira Ando Abstract Yeasts used in bread making are exposed to high concentrations of sucrose during sweet dough fermentation. Despite its importance, tolerance to high-sucrose stress is poorly understood at the gene level. To clarify the genes required for tolerance to high-sucrose stress, genome-wide screening was undertaken using the complete deletion strain collection of diploid Saccharomyces cerevisiae. The screening identified 273 deletions that yielded high sucrose sensitivity, approximately 20 of which were previously uncharacterized. These 273 deleted genes were classified based on their cellular function and localization of their gene products. Cross-sensitivity of the high-sucrose-sensitive mutants to high concentrations of NaCl and sorbitol was studied. Among the 273 sucrose-sensitive deletion mutants, 269 showed cross-sensitivities to sorbitol or NaCl, and four (i.e. ade5,7, ade6, ade8, and pde2) were specifically sensitive to high sucrose. The general stress response pathways via high-osmolarity glycerol and stress response element pathways and the function of the invertase in the ade mutants were similar to those in the wild-type strain. In the presence of high-sucrose stress, intracellular contents of ATP in ade mutants were at least twofold lower than that of the wild-type cells, suggesting that depletion of ATP is a factor in sensitivity to high-sucrose stress. The genes identified in this study might be important for tolerance to high-sucrose stress, and therefore should be target genes in future research into molecular modification for breeding of yeast tolerant to high-sucrose stress. [source] White pines, Ribes, and blister rust: integration and actionFOREST PATHOLOGY, Issue 3-4 2010R. S. Hunt Summary The preceding articles in this series review the history, biology and management of white pine blister rust in North America, Europe and eastern Asia. In this integration, we connect and discuss seven recurring themes important for understanding and managing epidemics of Cronartium ribicola in the white pines (five-needle pines in subgenus Strobus). Information and action priorities for research and management of the pathogen, telial and aecial hosts, and their interactions are listed in a detailed Appendix. Syntheses focused on genetics, plant disease, invasive species or forest management have provided alternative but knowledgeable lessons on the white pine blister rust pathosystem. Two critical issues for the conservation of white pines are to sustain ecosystems affected by blister rust and to maintain genetic diversity for adaptive traits such as disease resistance. Forest genetics includes tree improvement and molecular techniques for research; their application can increase rust resistance by artificial and natural selection. Silviculture augments genetics with methods to deploy and enhance resistance as well as to regenerate and tend white pine stands. Although cultivated or wild Ribes might serve as inoculum sources, silviculture and horticulture can reduce the risk of serious impacts from blister rust using genetics for breeding and epidemiology for hazard assessment and disease control. Climate change threatens to cause major alterations in temperature and precipitation regimes, resulting in maladapted conifers succumbing to various diseases and insect outbreaks. In contrast, many white pine species have broad ecological ranges and are tolerant of harsh environments,traits that permit successful establishment and growth over wide geographic and altitudinal zones. Given appropriate management, white pines could thrive as valuable commercial and ecologically important keystone species. In an uncertain environment, adaptive management provides a learning and participatory approach for sustaining resilient ecosystems. [source] Evaluation of Juglans species for resistance to Phytophthora cinnamomi:,differences in isolate virulence and response to fosetyl-AlFOREST PATHOLOGY, Issue 3 2009A. Belisario Summary Phytophthora is considered as an important pathogen on walnut, and severe losses are reported in European as well as in American walnut stands. Though several Phytophthora spp. are known to attack walnut, P. cinnamomi is considered the most virulent and widespread in southern Europe. Up to now, no walnut species or hybrid is known to have a high resistance level towards P. cinnamomi. Efforts are addressed in finding rootstock material graft compatible with English walnut and resistant/tolerant to P. cinnamomi. The extension of P. cinnamomi lesions on five Juglans species was studied to find out sources of resistance/tolerance to this pathogen. Walnut species clustered into two main groups, J. hindsii, J. nigra, and J. mandshurica were the less susceptible to the colonization of P. cinnamomi, while J. regia and J. sieboldiana were the most susceptible. On this account, J. mandshurica represents the best alternative as rootstock because its employment overcomes the risk of the occurrence of black line disease, it has good level of resistance to Agrobacterium temefaciens and Brenneria nigrifluens, and it is tolerant to Xanthomonas arboricola pv. juglandis. J. mandshurica is also compatible in cross-pollinations with J. regia and J. nigra. Differences in virulence of P. cinnamomi isolates was assessed and a marked interaction between species and isolate emerged. Treatment with fosetyl-Al by dipping was mainly efficient in reducing the length P. cinnamomi lesions, and an interaction between species and treatment was evident with the highest efficacy on J. regia and J. sieboldiana.
