Methodological issues in the epidemiological study of the teratogenicity of drugs

ABSTRACT The review presented here discusses and exemplifies problems in epidemiological studies of drug teratogenesis according to methodology: case,control studies, cohort studies, or total population studies. Sources of errors and the possibility of confounding are underlined. The review stresses the caution with which conclusions have to be drawn when exposure data are retrospective or other possible bias exists. It also stresses the problem with the multiple testing situation that is usually present in the studies. It is therefore difficult to draw any firm conclusion from single studies and still more difficult to draw conclusions on causality. As randomized studies are in most cases out of the question, one has to rely on the type of studies which can be made, but the interpretation of the results should be cautious. The ideal study, next to a randomized one, is a large prospective study with detailed exposure information and detailed and unbiased outcome data. Even so, such a study can mainly be used for identifying possible associations which have to be verified or rejected in new studies. Nearly every finding of a risk increase, if not extremely strong, should only be regarded as a tentative signal to be tested in independent studies. [source]


Olfactory preference for own mother and litter in 1-day-old rabbits and its impairment by thermotaxis

Abstract We investigated the ability of rabbit pups to display preferences towards various elements of their postnatal environment during the stage of confinement in the nest. Subjects were submitted to a two-choice test during the first week after birth to assess if they could detect and discriminate between does, litters of pups, or nesting materials of the same developmental stage. On D1 and D7, pups were attracted to any lactating doe, litter, or nest when compared to an empty compartment. When two stimuli were opposed, pups preferred their own nest to an alien one on D1 and D7 but not their mother nor their siblings when opposed to alien does or pups. However, additional tests indicated that this lack of preference for kin conspecifics resulted from a predominant attraction to thermal cues over individual odors. Indeed, pups were strongly attracted to a warm compartment (37°C) than to a cooler one (20°C) and once thermal cues were controlled for in the testing situation, the pups were specifically attracted to odors of their own mother's hair and of their siblings. No preference was observed towards the mother's uterine secretions. In conclusion, pups can recognize olfactory cues emanating from their mother and their siblings the day after birth. The preference for nesting materials would reflect in major part the combined attraction to maternal and sibling odors present in the nest. © 2008 Wiley Periodicals, Inc. Acute Effects of Ethanol on Behavior of Adolescent Rats: Role of Social Context

Background: First experiences with alcohol in humans occur predominantly in adolescence, and to a large extent the attractiveness of alcohol at this age is based on its ability to facilitate certain forms of social behavior (social facilitation). Adolescence is strongly marked by a focus on peer relationships, and the social nature of the situation plays an important role in responsiveness to alcohol. Peer-directed social activity of adolescent rats may be a valuable experimental model for the study of ethanol-induced changes in social behavior and assessment of the role of the social context in responsiveness to ethanol. Method: In the present study we used a modified dyad social interaction test to characterize acute effects of ethanol on different forms of social behavior (social investigation, contact behavior, and play) and social motivation (preference/avoidance of a peer) in adolescent rats. Ethanol effects on behavior directed toward a peer were compared with those induced by exposure to an inanimate novel object. Results: In the social context, the effects of ethanol were dose-dependent and biphasic. Low doses of ethanol (0.25,0.75 g/kg) produced apparent social facilitation (increased social activity and enhanced social preference), whereas higher doses (3 and 4 g/kg) caused social inhibition (decreased social activity and avoidance of a peer). This pattern was not observed for a nonsocial stimulus: Although overall activity in the nonsocial context was suppressed by 2 and 3 g/kg of ethanol, 0.5 g/kg of ethanol did not activate overall activity directed to the inanimate object. Conclusions: These findings demonstrate that the social nature of the testing situation plays an important role in responsiveness to alcohol in adolescence, especially to its activating effects. The results suggest also that the study of ethanol effects on social behavior of adolescent rats may be an effective tool for the study of adolescent alcohol use and abuse. [source]


Faking on Personality Measures: Implications for selection involving multiple predictors

The potential for faking on noncognitive measures in high stakes testing situations remains a concern for many selection researchers and practitioners. However, the majority of previous research examining the practical effects of faking on noncognitive assessments has focused on these measures in isolation, rather than the more common situation in which they are used in combination with other predictors. The present simulation examined the effects of faking on a conscientiousness measure on criterion-related validity, mean performance of those selected, and selection decision consistency when hiring decisions were based on this measure alone vs in combination with two other predictors across a range of likely selection scenarios. Overall, results indicated that including additional predictors substantially reduced , but did not eliminate , the negative effects of faking. Faking effects varied across outcomes and selection scenarios, with effects ranging from trivial to noteworthy even for multiple-predictor selection. Implications for future research and practice are discussed. [source]


Guidelines for the laboratory diagnosis of trichomoniasis in East European countries

Abstract The laboratory diagnosis of sexually transmitted infections in many Eastern European countries remains suboptimal. The main objective of the present evidence-based guidelines is to provide comprehensive information regarding the laboratory diagnosis of infections caused by Trichomonas vaginalis in East European countries. In particular, the present guidelines recommend: (i) to encourage examination of the wet mounts of vaginal exudates, instead of stained smears, at all clinical settings; (ii) nucleic acid amplification tests (NAATs) or culture could be employed if no trichomonads are detected on microscopic examination of the wet preparation and there is a strong indication of infection and (iii) the use of NAATs is encouraged in screening, using non-invasive specimens, or high volume testing situations. In the absence of internationally recognized commercial NAAT systems, tests developed in-house should be validated using obtainable international standards and quality assured strictly. Individual East European countries may be required to make minor national adjustments to these guidelines as a result of lack of accessibility to some reagents or equipment, or laws in a specific country.
