Incorporating power system security into market-clearing of day-ahead joint energy and reserves auctions

Abstract This paper is intended to introduce a technique for incorporating system security into the clearing of day-ahead joint electricity markets, with particular emphasis on the voltage stability. A Multiobjective Mathematical Programming (MMP) formulation is implemented for provision of ancillary services (Automatic Generation Control or AGC, spinning, non-spinning, and operating reserves) as well as energy in simultaneous auctions by pool-based aggregated market scheme. In the proposed market-clearing structure, the security problem, as an important responsibility of ISO, is addressed and a nonlinear model is formulated and used as the extra objective functions of the optimization problem. Thus, in the MMP formulation of the market-clearing process, the objective functions (including augmented generation offer cost, overload index, voltage drop index, and loading margin) are optimized while meeting AC power flow constraints, system reserve requirements, and lost opportunity cost (LOC) considerations. Comparison of activity coefficient models for electrolyte systems

Abstract Three activity coefficient models for electrolyte solutions were evaluated and compared. The activity coefficient models are: The electrolyte NRTL model (ElecNRTL) by Aspentech, the mixed solvent electrolyte model (MSE) by OLI Systems, and the Extended UNIQUAC model from the Technical University of Denmark (DTU). Test systems containing a single salt (NaCl), multiple salts, and mixed solvent aqueous electrolyte solutions were chosen. In vitro organogenesis of pancreas in Xenopus laevis dorsal lips treated with retinoic acid

Dorsal lips of Xenopus laevis may differentiate into pancreas after treatment with retinoic acid in vitro. The dorsal lip region is fated to be dorsal mesoderm and anterior endoderm. Dorsal lip cells isolated from stage 10 early gastrula differentiate into tissues such as notochord, muscle and pharynx. However, in the present study, dorsal lips treated with 10,4M retinoic acid for 3 h differentiated into pancreas-like structures accompanied by notochord and thick endodermal epithelium. Sections of the explants showed that some cells gathered and formed an acinus-like structure as observed under microscopes. In addition to the morphological changes, expressions of the pancreas-specific molecular markers, XlHbox8 and insulin, were induced in retinoic acid-treated dorsal lip explants. Therefore, it is suggested that retinoic acid may induce the dorsal lip cells to differentiate into a functional pancreas. However, continuous treatment with retinoic acid did not induce pancreas differentiation at any concentration. Dorsal lips treated with retinoic acid within 5 h after isolation differentiated into pancreas-like cells, while those treated after 15 h or more did not. The present study provided a suitable test system for analyzing pancreas differentiation in early vertebrate development. [source]


On-line hybrid test combining with general-purpose finite element software

Abstract A new on-line hybrid test system incorporated with the substructuring technique is developed. In this system, a general-purpose finite element software is employed to obtain the restoring forces of the numerical substructure accurately. The restart option is repeatedly used to accommodate the software with alternating loading and analysis characteristic of the on-line test but without touching the source code. An eight-storey base-isolated structure is tested to evaluate the feasibility and effectiveness of the proposed test system. The overall structure is divided into two substructures, i.e. a superstructure to be analysed by the software and a base-isolation layer to be tested physically. Collisions between the base-isolation layer and the surrounding walls are considered in the test. Use of a high-throughput umu -microplate test system for rapid detection of genotoxicity produced by mutagenic carcinogens and airborne particulate matter

Abstract In the present study, we developed a rapid umu -microplate test system that uses the nitroreductase- and O -acetyltransferase-overproducing Salmonella typhimurium strain NM3009 and the O -acetyltransferase-overproducing S. typhimurium strain NM2009 to detect genotoxic activity in small volume samples. The assay was used to test the genotoxicity of several standard mutagens and environmental samples. Exponentially growing cultures of NM3009, NM2009, and the parental strain TA1535/pSK1002 were incubated in 96-well microplates with test chemicals both in the presence and in the absence of rat liver S9. The relative ,-galactosidase activities were then determined colorimetrically using either chlorophenol red-,- D -galactopyranoside (CPRG) or O -nitrophenyl-,- D -galactopyranoside (ONPG) as a measure of umuC gene induction activity. The sensitivities of NM3009 without S9 mix and NM2009 with S9 mix to nitroarenes and aromatic amines were up to 24- to 75-fold higher than those of the parent strain. Induction of umuC gene expression was detected more readily with CPRG than ONPG. The umu -microplate assay also detected genotoxicity in organic extracts of particulate matter from air samples collected in Osaka City, Japan. The pattern of the responses suggested that the genotoxic activity of the particulate extract was due primarily to nitrated polycyclic aromatic hydrocarbons. Our results indicate that the umu -microplate assay may be a useful way of carrying out rapid screens for genotoxicity in small-volume environmental samples. Environ. Mol. Development of a bacterial challenge test for gnotobiotic sea bass (Dicentrarchus labrax) larvae

Summary The use of probiotic microorganisms in aquaculture is gaining a lot of interest. Gnotobiotic model systems are required in order to fully understand the effects and modes-of-action of these microorganisms, as the native microbial communities present in non-sterile animals can lead to false conclusions. In this study, a gnotobiotic sea bass larvae (Dicentrarchus labrax) test system was developed. In order to obtain bacteria-free animals, the eggs were disinfected with glutaraldehyde and subsequently incubated in a solution of rifampicin and ampicillin. Axenity was confirmed using culture-dependent and -independent techniques. The gnotobiotic larvae were fed axenic Artemia sp. from 7 days after hatching onwards. In the challenge test, one of the three opportunistic pathogens, Aeromonas hydrophila, Listonella anguillarum serovar O1 and O2a, was added to the model system via the water and encapsulated in Artemia sp. Only serovar O2a led to increased mortality in the sea bass larvae. The presented gnotobiotic model can be used for research on, among others, reciprocal metabolic effects between microorganisms and the host (e.g. as measured by gene expression), immunostimulants, pharmacological research and the histological development of the gastrointestinal tract and growth of larvae. Single- and two-species tests to study effects of the anthelmintics ivermectin and morantel and the coccidiostatic monensin on soil invertebrates

Abstract Soil invertebrates in arable land are potentially exposed to veterinary medicines excreted by husbandry. The toxicity of three widely used pharmaceuticals was therefore investigated with the use of common soil invertebrates exposed in the laboratory in single- or two-species test system. The anthelmintic morantel did not cause significant mortality to either Folsomia fimetaria or Enchytraeus crypticus even at the highest tested concentration of 900 mg kg,1 dry soil. The coccidiostatic monensin affected the reproduction of F. fimetaria and E. crypticus with soil concentrations estimated to cause a 10% effect at values of approximately 109 and 71.8 mg kg,1 dry soil, respectively, but caused no mortality to adult. The anthelmintic ivermectin did not affect the survival of adult Hypoaspis aculeifer. Reproduction of H. aculeifer declined approximately 45% in response to ivermectin exposure of 5 mg kg,1 dry soil. Ivermectin was highly toxic to F. fimetaria and affected the survival of adults with soil concentrations estimated to cause a 50% mortality at values of 5.3 mg kg,1 dry soil in the single-species test system and 0.14 mg kg,1 dry soil in the two-species test system. Reproduction of F. fimetaria was reduced by ivermectin with 10% effective concentration at 0.19 mg kg,1 dry soil in the single-species test system and 0.02 mg kg,1 dry soil in two-species test system. It was shown that species interactions may influence the response of test organisms to toxic substances. The data from this study and previously published data showed that, whereas ivermectin is likely to pose a risk to soil-dwelling invertebrates, adverse effects of morantel and monensin are unlikely to occur as a result of residue excretion from treated farm animals. Uptake and accumulation of sediment-associated 4-nonylphenol in a benthic invertebrate (Lumbriculus variegatus, freshwater oligochaete)

Abstract In the present work, the oligochaete Lumbriculus variegatus was exposed for 56 d to lake sediment spiked with 4-nonylphenol (4-NP), which is a breakdown product of alkylphenol polyethoxylates, an important class of nonionic surfactants. During the exposure period, the content of 4-NP was determined in the oligochaetes, sediment, overlying water, and pore water in order to monitor the distribution of the 4-NP in the compartments of the test system. Concentration of 4-NP in L. variegatus increased linearly over the course of the test, with an uptake rate coefficient of 1.9 × 10,2 (± 0.2 × 10,2; [g carbon/(g lipid-h)]). No steady state was reached at the end of the exposure period, suggesting that the elimination of 4-NP by the organism was negligible. Ingested sediments played an important role in the accumulation of 4-NP in L. variegatus, which may achieve very high 4-NP body concentrations. The 56-d biota sediment accumulation factor (BSAF) was 24 ± 7 g carbon/g lipid. L. variegatus also was exposed to 4-NP-contaminated field sediment, and field oligochaetes and sediments were collected for 4-NP pollution assessment in aquatic ecosystem. The 4-NP uptake with natural sediment was in accordance with that measured with spiked sediments, suggesting that the bioavailability of sediment-associated 4-NP for L. variegatus was not affected by 4-NP sediment concentration and abiotic sediment characteristics. The BSAFs measured in field oligochaetes, ranging from 39 to 55 g carbon/g lipid, was relatively higher than the bioaccumulation factor measured in laboratory tests. The results suggest that 4-NP concentration can reach high levels in benthic oligochaetes; this can be an important way of exposure for their pelagic predators. [source]


Mutagenicity of nitroaromatic degradation compounds

Abstract The mutagenicity of 2,4-dinitrotoluene (24DNT), and 2,6-dinitrotoluene (26DNT), and their related transformation products such as hydroxylamine and amine derivatives, which are formed by Clostridium acetobutylicum, were tested in crude cell extracts using Salmonella typhimurium TA100. A previous publication already reported the mutagenic activities of 2,4,6-trinitrotoluene (TNT) and its related hydroxylamine derivatives in this test system. A time course of the mutagenicity during the anaerobic transformation of TNT, 24DNT, and 26DNT was also investigated under the same conditions to compare with the results from the pure compounds. The monohydroxylamino intermediates 2-hydroxylamino-4-nitrotoluene (2HA4NT), 4-hydroxylamino-2-nitrotoluene (4HA2NT) and 2-hydroxylamino-6-nitrotoluene (2HA6NT) formed during anaerobic transformation of dinitrotoluenes were proven to be mutagenic in the Ames test using Salmonella typhimurium TA100. This study reports that 4HA2NT is the most stable derivative, whereas 2HA4NT and 2HA6NT are less stable and these intermediates are mutagenic in the Ames test. Both 24DNT and 26DNT and their final metabolites 2,4-diaminotoluene (24DAT) and 2,6-aminotoluene (26DAT) appeared nonmutagenic. In a time-course study of TNT degradation, the temporal sample containing 85% of 2,4-dihydroxylamino-6-nitrotoluene (24HA6NT) is most mutagenic. These observations suggest that the bioremediation approach for treatment of 24DNT and 26DNT should be carried past the hydroxylamino intermediate. [source]


Studies on the effect of soil aging on the toxicity of pyrene and phenanthrene to a soil-dwelling springtail

Abstract Soil samples spiked with five concentrations of pyrene and phenanthrene were aged for 0, 10, 40, and 120 d before toxicity was investigated using a standardized bioassay with the soil-dwelling collembolan Folsomia fimetaria L. Toxic effects were measured as reductions in survival and reproductive output after 3 weeks of exposure. Both pyrene and phenanthrene were degraded in the test system during storage, phenanthrene to a higher degree than pyrene. However, when toxic effects of the tested substances were calculated on the basis of measured concentrations of the parent compounds, toxicity was unaffected by storage for up to 120 d. Many studies have shown a negative correlation between aging and the biodegradation of polycyclic aromatic hydrocarbons (PAHs), indicating a reduction in the bioavailability of PAHs to micro-organisms with time. Our results indicate that a reduction in toxicity, as related to the measured concentrations of the parent compounds in the soil, may not always be expected for aged soils. Provided that metabolites of pyrene and phenanthrene did not significantly contribute to the toxicity in aged soil samples, a possible explanation for the absence of aging effects is the high test concentrations used in combination with the low content of organic carbon (1.6%) in the test soil. Modeling pentachlorophenol bioavailability and bioaccumulation by the freshwater fingernail clam Sphaerium corneum using artificial particles and humic acids

Abstract The uptake of anthropogenic chemicals by benthic bivalves may occur through the water phase and also by the ingestion of particles from both the suspended matter and bottom sediments. Many chemicals sorb to sediments and, subsequently, are released in the digestive tract of animals. The assessment of sediment-bound chemicals has been difficult because of the complexity of the association between these chemicals and natural particles. To simplify this complexity, we previously devised a test system using artificial particles with known chemical structures. In the present work, we improved this experimental design by adding humic materials as a source of organic matter. Bioassays were conducted by exposing the fingernail clam Sphaerium corneum to sublethal levels of pentachlorophenol (PCP) in the presence or absence of the artificial particles, treated with or without a commercial preparation of humic acids. The results showed that the bioavailability and bioaccumulation could be explained on the basis of the interactions of PCP with the active groups and/or the backbone of the resins, both in systems with or without humic acids. This model may constitute a useful approach to modeling and predicting the uptake and accumulation of chemicals bound to natural sediments. [source]


A test system to evaluate the susceptibility of Oregon, USA, native stream invertebrates to triclopyr and carbaryl

Abstract The susceptibility of six indigenous macroinvertebrate species representative of U.S. Pacific Northwest streams (Ameletus sp., Brachycentrus americanus, Calineuria californica, Cinygma sp., Lepidostoma unicolor, Psychoglypha sp. early and late instar) to formulated triclopyr ester (herbicide) and carbaryl (insecticide) was determined using laboratory bioassays. Acute toxicity was expressed as the lethal concentration to 50% (LC50) and 1% (LC1) of the test population based on a 96-h exposure duration. Carbaryl was found to be 1,000 times more toxic than triclopyr for all the organisms tested. The LC1 values (7.5, 28.8, 9.0, 3.0, 9.5, 14.8, 33.8 ,g/L, respectively, for carbaryl and 1.8, 3.9, 4.0, 4.2, 29.0, 16.1 mg/L, respectively, for triclopyr) were used in the calculation of hazardous concentration to 5% of the stream macroinvertebrate community (HC5) based on the lower 95% confidence limit (HC5/95). The hazardous concentration (HC5/95) for triclopyr was 0.11 mg/L and for carbaryl ranged from 0.43 to 0.66 ,g/L, respectively. Triclopyr and carbaryl symptomology were analyzed for two organisms, C. californica and Cinygma sp. Carbaryl symptomology included knockdown and moribund states with severity and time of appearance being a function of dose. In triclopyr poisoning, death occurred suddenly with little or no symptomology. Time to 50% mortality (LT50) values were consistently higher for C. californica than for Cinygma sp. exposed to both chemicals at similar concentrations. [source]


The Relative Influence of Epileptic EEG Discharges, Short Nonconvulsive Seizures, and Type of Epilepsy on Cognitive Function

Summary: Purpose: This study addressed whether cognitive impairment in children with epilepsy is caused by disease-related stable factors, such as the type of epilepsy, or by acute effects of paroxysmal epileptic activity such as epileptic EEG discharges. We studied a nonselected group with short nonconvulsive seizures, as these seizures may elude detection and may therefore persist over a longer period. In this group, the diagnostic issue is to differentiate between the combined effects of several epilepsy-related factors on cognition. Methods: All children were assessed with 32-channel EEG, synchronized with a computerized cognitive test system and a video-monitoring system. Recording time was 2 h. The primary inclusion criteria were unclear seizures and fluctuations in cognitive performance and/or frequent epileptic EEG discharges in a recent EEG. Results: One hundred fifty-two patients met the inclusion criteria; 31 patients appeared not to have a diagnosis of epilepsy and were used as a nonepilepsy control group. Our results show that type of epilepsy has an impact on stable cognitive functions, such as educational achievement. Paroxysmal epileptic activity (acute effects of seizures and epileptic EEG discharges) affects primarily transient mechanistic cognitive processes (alertness, mental speed). Conclusions: These results suggest that the effects of paroxysmal epileptic activity on transient cognitive mechanisms may accumulate over time and consequently affect the more stable aspects of cognitive function such as educational achievement. The clinical relevance is that early detection of the cognitive impact of seizure-related activity and subsequent treatment may prevent its detrimental impact on cognitive and educational development. Testing of fine motor skills in dental students

Manual skills form only a part of the capabilities required of future dentists, but they are a very important component, which should be tested. With regard to the dental specialities, the present study tested speciality-independent fine motor skills. No objective, practical solution has been found up to now. 88 dental students and, as a control group, 23 medical students were examined in the longitudinal study. In the course of the analysis, 4 fine motor tests were carried out at the beginning of the 2nd and 6th semesters. The tests comprised the tremometer test, the tremometer test with a mirror, the 2-hand sinusoid test and archery using the Game Gear by SEGA. The test devices facilitate primarily the testing of components of accuracy of movements, indirect working methods, and eye-hand coordination. In the comparison of performances on test day A, the medical students' performance was noticeably better. As testing progressed, results showed stagnation in the performance of the medical students and a significant improvement in the performance of the dental students. That means that the test system can be used for a test over the course of study, but not as an initial test. [source]


Analyzing dynamic performance of stressed power systems in vicinity of instability by modal series method

Abstract Highly stressed power systems exhibit complex dynamic behaviors such as inter-area oscillations when subjected to large disturbances. In such conditions, nonlinear effects have dominant role in determining dynamic response of the systems. In this paper by using modal series method, dynamic behaviors of the stressed power systems in severe conditions and near instability have been studied. Also two measures, mode dominance measure (MDM) and most perturbed machine factor (MPF) have been introduced. They determine the most dominant modes and identify the most perturbed generators when the system is subjected to a given fault. Contribution factors have been used to show the links between identified modes and machines from the analysis. Time domain simulation has been helped for validation of the results. By using similarity transformation, state variables have been represented in modal space and utilized to check the results. The studies are carried out on the IEEE 50-generator test system which demonstrates a wide range of dynamic characteristics at different loading levels and fault scenarios. Local control of photovoltaic distributed generation for voltage regulation in LV distribution networks and simulation tools

Abstract The increasing connection of distributed generation (DG) in distribution networks may affect the quality of power offered to customers. One of the most relevant issues is the possibility to have unacceptable voltage rise at the point of common coupling (PCC). This work focuses on the problem of voltage control in LV distribution networks in the presence of photovoltaic (PV) DG. The paper presents a local voltage control method based on PV generation curtailment as an alternative to ,on/off' operation, typically required by distribution operators to prevent overvoltage at the PCCs by means of overvoltage protections embedded in the PV unit. To show the effect of the proposed local voltage control, a simulation tool, developed in MATLAB®,Simulink® environment, is presented. Appropriate numerical models for network components and PV generators are provided in order to describe computer simulation implementation of the test system. The final results show that the control system is able to adjust the active power output of local generators to keep the voltage profile of the feeder, in which DG is installed, within the range allowed by Norm EN 50160. The assessment of the impacts of the security management in a competitive environment

Abstract Security management is of the utmost importance in power system and many efforts have been made on this issue since a reliable supply of electricity is crucial to the modern societies. The traditional approach adopted in Europe is based on the "n,,,1" steady-state security that is efficient, although restrictive, and can continue to play an important role also in the new competitive context. In this paper, three alternative market-based models for security consideration are introduced, according to preventive and corrective control approaches. The models are presented and analyzed in terms of their economic impacts on the market throughout a set of useful indexes that allows for a ranking of the advantages got from the re-enforcement of the lines. Moreover, the positive impacts of the phase-shifter (PS) installation for the security and congestion management are quantified and indexes are derived to assess their optimal location. A new algorithm for reactive power management and pricing in an open access environment

Abstract In deregulated electricity markets, reactive power is one of the ancillary services that is provided by the Independent System Operator (ISO). In this paper we propose a new algorithm to ,tune' reactive power resources with the objectives of minimizing active power losses and optimizing system voltage profile and stability. In our solution, we assume that reactive power resources are either owned by or are under long-term contracts with the ISO. The optimization problem is solved as an Extended Multi-objective Optimal Power Flow (EMOPF) problem using Lexico Graphic Method (LGM). Thereafter, using the concept of Fair Resource Allocation (FRA), the cost of reactive power is allocated to existing transactions. The application of the algorithm to a test system is also presented. A new transmission pricing approach for the electricity cross-border trade in the ASEAN Power Grid

Abstract The electricity cross-border trade is presently introduced among the member countries of the Association of South East Asian Nations (ASEAN). The ASEAN Power Grid (APG) is a plan to interconnect transmission networks among the ASEAN countries to optimize the use of energy resources; to operate the power network in an efficient, economical, and reliable manner; and to provide a close relation among the member countries by electric power interconnection. Transmission pricing is one of the controversial tasks to achieve the APG objectives. In this paper, a transmission pricing method for the electricity cross-border trade based on a combination of postage stamp method and sensitivity indices is proposed. The postage stamp pricing is a uniform tariff expected to recover the project investments, and the operation and maintenance costs. With the combination of the postage stamp method and sensitivity indices, the proposed pricing method sends proper incentive signals to power traders, which are based on system usage and congestion management. To demonstrate its effectiveness, the proposed method is applied to a 12-bus test system. The nodal tariffs at the particular injecting points, payments of the users, and revenues of transmission owners are computed. The simulation results indicate that the proposed method ensures a recovery of the investment costs and the concurrent costs of operation and maintenance in an efficient, fair, and simple manner. In contrast to the time-domain approach, direct methods determine system stability based on energy functions. The basis of direct methods for the stability assessment of a system is knowledge of the stability region. During the last decade, many researches have thoroughly analysed the use of energy functions for the direct stability assessment of networks. Energy function analysis offers a different geometric view of voltage collapse. The Transient Energy Function, a technique based on Lyapunov stability theory and originally developed for direct stability analysis of power systems, has been successfully used as a voltage stability index for collapse studies. Stability improvement in power systems with non-linear TCSC control strategies

In this paper, a non-linear control scheme for the TCSC (thyristor-controlled series compensator) to dampen power oscillations and to improve the transient stability of power systems is presented. Based on an one-machine-infinite-bus system, a non-linear mathematical model is established which is proven as an affine nonlinear system. With the help of the feedback linearization technique, the affine non-linear model is exactly transferred to a linear model, and then the control scheme is designed for the TCSC based on the global linearization, where the input signal uses local measurements only. The effectiveness and robustness of the proposed non-linear control scheme are demonstrated with an one-machine test system, where the TCSC modelling and power system simulations are performed by using the program system NETOMAC. In comparison with a conventional control scheme, significant improvements of dynamical performance in the test power' system are achieved by the proposed non-linear control strategy for the TCSC. Establishment of a mouse xenograft model for mycosis fungoides

Abstract:, Mycosis fungoides (MF) is the most frequent variant of cutaneous T-cell lymphomas (CTCLs). MF primarily involves the skin initially with patches and plaques. In later stages, cutaneous tumors develop and tumor cells may spread to lymph nodes and finally to visceral sites. Here, we describe an animal model for MF in immune-deficient nude mice, using the CTCL cell line MyLa. Subcutaneous transplantation of MyLa cells leads to the formation of cutaneous tumors in 80% of the mice (50/60 total). Spread of tumor cells to visceral sites was detected by immunohistochemistry and polymerase chain reaction (PCR)-based detection of specific T-cell receptor-, rearrangement. MyLa cells were found circulating in the blood, lymph nodes, and in blood vessels of heart, kidney, lung, and liver. In lung and liver tissue, tumor cells presented perivascular invasion, but no large secondary tumors developed. The nude mouse model described here will be a valuable test system for new therapeutic approaches for the treatment of MF and opens the unique opportunity to study the disease in vivo. [source]


Covalently crosslinked complexes of bovine adrenodoxin with adrenodoxin reductase and cytochrome P450scc

NADPH-dependent adrenodoxin reductase, adrenodoxin and several diverse cytochromes P450 constitute the mitochondrial steroid hydroxylase system of vertebrates. During the reaction cycle, adrenodoxin transfers electrons from the FAD of adrenodoxin reductase to the heme iron of the catalytically active cytochrome P450 (P450scc). A shuttle model for adrenodoxin or an organized cluster model of all three components has been discussed to explain electron transfer from adrenodoxin reductase to P450. Here, we characterize new covalent, zero-length crosslinks mediated by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide between bovine adrenodoxin and adrenodoxin reductase, and between adrenodoxin and P450scc, respectively, which allow to discriminate between the electron transfer models. Using Edman degradation, mass spectrometry and X-ray crystallography a crosslink between adrenodoxin reductase Lys27 and adrenodoxin Asp39 was detected, establishing a secondary polar interaction site between both molecules. No crosslink exists in the primary polar interaction site around the acidic residues Asp76 to Asp79 of adrenodoxin. However, in a covalent complex of adrenodoxin and P450scc, adrenodoxin Asp79 is involved in a crosslink to Lys403 of P450scc. No steroidogenic hydroxylase activity could be detected in an adrenodoxin ,P450scc complex/adrenodoxin reductase test system. Because the acidic residues Asp76 and Asp79 belong to the binding site of adrenodoxin to adrenodoxin reductase, as well as to the P450scc, the covalent bond within the adrenodoxin,P450scc complex prevents electron transfer by a putative shuttle mechanism. Thus, chemical crosslinking provides evidence favoring the shuttle model over the cluster model for the steroid hydroxylase system. [source]


A fluorescence quenching test for the detection of flavonoid transformation

Abstract A novel fluorescence quenching test for the detection of flavonoid degradation by microorganisms was developed. The test is based on the ability of the flavonoids to quench the fluorescence of 1,6-diphenyl-1,3,5-hexatriene (DPH). Several members of the anthocyanidins, flavones, isoflavones, flavonols, flavanones, dihydroflavanones, chalcones, dihydrochalcones and catechins were tested with regard to their quenching properties. The anthocyanidins were the most potent quenchers of DPH fluorescence, while the flavanones, dihydroflavanones and dihydrochalcones, quenched the fluorescence only weakly. The catechins had no visible impact on DPH fluorescence. The developed test allows a quick and easy differentiation between flavonoid-degrading and flavonoid-non-degrading bacteria. The investigation of individual reactions of flavonoid transformation with the developed test system is also possible. [source]


Proper Splitting of Interconnected Power Systems

Abstract Power system islanding is the last defense line to protect power grids from incidence of wide-area blackout. As a wide-area control action, power system splitting is a comprehensive decision making problem that includes different subproblems. This paper introduces a novel approach for separation of the entire power system into several stable islands in different loading levels. The proposed method combines both the dynamic and the static characteristics of interconnected power network and determines the proper splitting schemes. The presented algorithm searches for proper islanding strategy in the boundary of primary determined coherent machines using Krylov subspace method and finds the proper splitting points by transferring some of the buses in one island to another island such that total load shedding is minimized. A spanning tree-based depth first search algorithm is used to find all possible combination of transferred buses. The presented method reduces the huge initial search space of islanding strategy considering dynamic characteristics of integrated power system and reduction of search space to only boundary network. The speed of the proposed algorithm is remarkably high and can be applied for islanding the power system in real-time. The presented algorithm is applied to IEEE 118 BUS test system. Results show the robustness, effectiveness, and capability of the algorithm to determine fast and accurate proper islanding strategy. Efficient IP-multicast via Inmarsat BGAN, a 3GPP satellite network

Abstract This paper outlines a number of challenges associated with supporting IP-multicast services efficiently across the Inmarsat Broadband Global Area Network (BGAN) 3GPP-based satellite network operating over the Imarsat-4 satellite constellation. The paper presents a network architecture that extends the 3GPP reference architecture to allow IP-multicast to be delivered when the Core Network is in a 3GPP Release-4 (non-MBMS compliant) configuration. This paper further extends the service and system concepts defined in 3GPP MBMS to provide improved flexibility and accountability, and improved scalability and efficiency when operating with the Inmarsat-4 BGAN TDM/TDMA air interface. This paper describes a number of radio resource management techniques that were deployed in a test system and the validation testing that was undertaken to support multimedia distribution and VoIP-based netted communications applications. Wheat Cellular Membrane Thermotolerance Under Heat Stress

Abstract Four genotypes of Triticum aestivum L. and Triticum turgidum subsp. durum chosen according to their genetic background diversity were subjected to heat stress after anthesis. Membrane permeability, lipid peroxidation and fatty acids (C14:0, C16:0, C16:1c, C16:1t, C18:0, C18:1, C18:2 and C18:3) were quantified. The estimation of the quantum yield of non-cyclic photosynthetic electron transport was used as well as a test system to further evaluate the implications on thylakoid functioning. It was found differences within bread and durum wheat species concerning the capability to cope with high temperatures at the stage of grain filling. The genotype Sever showed high thermal sensitivity concerning membrane lipid peroxidation and membrane permeability, as evaluated by the increased production of ethylene and MDA, as well as by the impact on TFA (at the middle term of grain filling). In the durum wheat genotypes, differences were also found, with TE 9306 displaying high membrane stability, with no increases on membrane permeability, MDA and ethylene content. In this way, the observed changes on TFA in this genotype might have constituted a mechanism to allow qualitative lipid changes, reflected in lower unsaturation level of membrane FAs which is a positive trait under high temperatures. The joint effects of larval density and 14C-cypermethrin on the life history and population growth rate of the midge Chironomus riparius

Summary 1Chemical effects on organisms are typically assessed using individual-level endpoints or sometimes population growth rate (PGR), but such measurements are generally made at low population densities. In contrast most natural populations are subject to density dependence and fluctuate around the environmental carrying capacity as a result of individual competition for resources. As ecotoxicology aims to make reliable population projections of chemical impacts in the field, an understanding of how high-density or resource-limited populations respond to environmental chemicals is essential. 2Our objective was to determine the joint effects of population density and chemical stress on the life history and PGR of an important ecotoxicological indicator species, Chironomus riparius, under controlled laboratory conditions. Populations were fed the same ration but initiated at different densities and exposed to a solvent control and three concentrations of 14C-cypermethrin in a sediment,water test system for 67 days at 20 ± 1 °C. 3Density had a negative effect on all the measured life-history traits, and PGR declined with increasing density in the controls. Exposure to 14C-cypermethrin had a direct negative effect on juvenile survival, presumably within the first 24 h because the chemical rapidly dissipated from the water column. Reductions in the initial larval densities resulted in an increase in the available resources for the survivors. Subsequently, exposed populations emerged sooner and started producing offspring earlier than the controls. 14C-cypermethrin had no effect on estimated fecundity and adult body weight but interacted with density to reduce the time to first emergence and first reproduction. As a result, PGR increased with cypermethrin concentration when populations were initiated at high densities. 4Synthesis and applications. The results showed that the effects of 14C-cypermethrin were buffered at high density, so that the joint effects of density and chemical stress on PGR were less than additive. Low levels of chemical stressors may increase carrying capacity by reducing juvenile competition for resources. More and perhaps fitter adults may be produced, similar to the effects of predators and culling; however, toxicant exposure may result in survivors that are less tolerant to changing conditions. If less than additive effects are typical in the field, standard regulatory tests carried out at low density may overestimate the effects of environmental chemicals. Further studies over a wide range of chemical stressors and organisms with contrasting life histories are needed to make general recommendations. [source]


Atrazine-induced changes in aromatase activity in estrogen sensitive target tissues

Abstract Atrazine (ATR) is a pesticide used widely throughout North America. Although not directly estrogenic, ATR treatment has been shown to increase aromatase activity in tumor cell lines. Thus, it is suggested that ATR can increase local tissue estrogen levels in estrogen sensitive target tissues through increased aromatase activity. Therefore the effect of ATR on aromatase activity was measured in human granulosa-lutein cell cultures, cells that abundantly express aromatase, and endometrial stromal cell (ESC) cultures, cells that do not express aromatase. Aromatase activity was quantified by the tritiated water method and the specificity of the assay was confirmed by co-incubation with 4-hydroxyandrostenedione, an irreversible inhibitor of the catalytic activity of aromatase. Aromatase activity in ATR treated (1,10 µm) granulosa-lutein cells was increased more than 2-fold compared with control cultures. There were no treatment related changes in cellular protein and thus it is suggested that the ATR-induced change in aromatase activity was not due to an increase in cell number. ATR-treatment had no effect on ESC aromatase activity at any concentration tested. Similarly, there was no effect of ATR treatment on human recombinant aromatase activity in our cell-free test system. Therefore it is concluded that µm concentrations of ATR can increase aromatase activity of human granulosa cells but not ESC and this effect is not elicited at the enzyme level. Free radical,scavenging activity and DNA damaging potential of auxins IAA and 2-methyl-IAA evaluated in human neutrophils by the alkaline comet assay

Auxins, of which indole-3-acetic acid (IAA) is the most widespread representative, are plant hormones. In addition to plants, IAA also naturally occurs in humans in micromolar concentrations. In the presence of peroxidase, indolic auxins are converted to cytotoxic oxidation products and have thus been proposed for use in gene-directed enzyme/prodrug tumor therapy. Since data on the genotoxicity of IAA and its derivatives are not consistent, here we investigate the early DNA damaging effects (2-h treatment) of the auxins, IAA, and 2-methyl-indole-3-acetic acid (2-Me-IAA) by the alkaline comet assay and compare them with their free radical,scavenging activity measured by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Human neutrophils are chosen as the test system since they possess inherent peroxidase activity. The results of the comet assay indicate an increase in DNA damage in a dose-dependent manner up to 1.00 mM of both auxins. Generally, IAA applied in the same concentration had greater potential to damage DNA in human neutrophils than did 2-Me-IAA. The genotoxicities of the two examined auxins are negatively correlated with their antioxidant activities, as measured by the DPPH assay; 2-Me-IAA showed a higher antioxidant capacity than did IAA. We assume that differences in the molecular structure of the tested auxins contributed to differences in their metabolism, in particular, with respect to interactions with peroxidases and other oxidative enzymes in neutrophils.
