KINETICS of QUALITY CHANGE DURING COOKING and FRYING of POTATOES: PART I. TEXTURE

ABSTRACT Kinetics of texture change during cooking and frying of potatoes were evaluated in this study. Potatoes were cut into cylinders (diameter × height: 20 mm × 20 mm for cooking, and 10 mm × 20 mm for frying) and cooked in a temperature controlled water bath at 80,100C or fried in a commercial fryer at 160,190C for selected times. the cooked samples were water cooled while the fried samples were air cooled immediately after the treatment. Test samples were then subjected to a single cycle compression test in a computer interfaced Universal Testing Machine and three textural properties (hardness, stiffness and firmness) were derived from the resulting force-deformation curves. Texture parameters of cooked potatoes decreased with progress of cooking time and the rate of texture changes associated with each temperature was found to be consistent with two pseudo first-order kinetic mechanisms, one more rapid than the other. Textural values of fried potatoes were found to increase with frying time and also followed a first order kinetic model. Temperature sensitivity of rate constants was adequately described by Arrhenius and z-value models. [source]


Is the circumferential tensile modulus within a human medial meniscus affected by the test sample location and cross-sectional area?

Quantifying the material properties of the human menisci is paramount to understanding their biomechanical functions within the knee. One important intrinsic material property governing the biomechanical functions of the meniscus is the circumferential tensile modulus. The purpose of this study was to determine if the circumferential tensile modulus of the human medial meniscus depends on the location and thickness of the sample tested. The following three hypotheses were tested: (a) the circumferential location (anterior, central, and posterior) does not significantly affect the tensile modulus, (b) the radial location (inner to outer) significantly affects the tensile modulus, and (c) the thicknes (cross-sectional area) significantly affects the tensile modulus. Test samples, whose length was oriented in parallel with the circumferential collagen fibers, were collected from different circumferential and radial locations throughout 30 human medial menisci. Samples of three different thicknesses (0.5, 1.5, and 3.0 mm) were taken from three equal groups of 10 menisci (i.e., one thickness per group). The circumferential tensile modulus was measured under quasi-statc loading. Statistical analysis showed no significant effect of the circumferential or radial location of the sample on the circumferential tensile modulus. This indicates that an overall circumferential tensile modulus may be calculated for the human medial meniscus by averaging the values determined at the various locations. However, the thickness of the test sample had a significant effect on the measured circumferential tensile modulus: the modulus varied inversely with the thickness. Thus, moduli determined from test samples that are too small in cross-sectional area overestimate the effective modulus of the tissue on the whole, and the cross-sectional area of the sample must be considered when determining a representative circumferential tensile modulus for the medial meniscus in a human knee. Polymer Bonded Ferrite Materials as EMC Components

Test samples and cases were prepared by hot or injection molding. Permeability spectra show loss contributions in the frequency range 100-1000,MHz. Absorption measurements of injection molded polymer-ferrite cases display a 3-5,dB better attenuation characteristics compared to graphite-loaded polyamide housings. Polymer-gebundene Ferritwerkstoffe für EMV-Anwendungen Polymer-gebundene Ferritwerkstoffe auf der Basis von Ferrit (Mn-Zn oder Ni-Zn Ferrite) , Thermoplast Mischungen wurden präpariert und hinsichtlich ihrer Eignung zur Absorption elektromagnetischer Strahlung untersucht. Es wurden sowohl Testproben wie auch komplette Gehäusekomponenten durch Heißpressen oder Spritzgießen hergestellt. Die Permeabilitätsspektren weisen verlustbehaftete Komponenten im Frequenzbereich von 100-1000,MHz auf. Schirmdämpfungsmessungen an spritzgegossenen Gehäuseteilen zeigen eine um 3-5,dB bessere Dämpfung im Vergleich zu Graphit-gefüllten Polyamid-Gehäusen. [source]


Simulations of IEF in microchannel with variable cross-sectional area

Abstract This study develops a 1-D mass transport model to describe the electrophoresis transport behavior within a microchannel with a variable cross-sectional area. Utilizing three different numerical schemes, simulations are performed to investigate the IEF of proteins in ampholyte-based pH gradients within both a planar microchannel and a contraction,expansion microchannel, respectively. The simulation results obtained using the modified 1-D mass transport model and the finite-volume method (FVM) for the IEF separation of a single protein sample in a ten-ampholyte-based pH gradient within a planar microchannel are consistent with those presented by Shim et al. [Electrophoresis 2007, 28, 572,586] using a 2-D FVM scheme. In addition, the Courant,Friedrichs,Lewy number insensitive conservation element and solution element (CNI-CESE) method is found to be both more robust and more computationally efficient than the conventional CESE scheme when modeling IEF phenomena within a contraction,expansion microchannel. In modeling the IEF separation of four sample ampholytes in a 20-ampholtye-based pH gradient within a contraction,expansion microchannel, the results obtained using the CNI-CESE scheme are in good agreement with those published in literature. Moreover, the simulations can be performed significantly faster with the new 1-D model and the CNI-CESE scheme. Finally, the results obtained using the modified 1-D mass transport model and the CNI-CESE scheme demonstrate that the concentration of the focused test sample and the resolution of the pH gradient within the microchannel increase as the number of ampholytes used to accomplish the IEF separation process is increased. Automatic analysis of multiplex ligation-dependent probe amplification products (exemplified by a commercial kit for prenatal aneuploidy detection)

Abstract For use in routine prenatal diagnostics, we developed software and methods for automatic aneuploidy detection based on a commercial multiplex ligation-dependent probe amplification (MLPA) kit. Software and methods ensure a reliable, objective, and fast workflow, and may be applied to other types of MLPA kits. Following CE of MLPA amplification products, the software automatically identified the peak area for each probe, normalized it in relation to the neighboring peak areas of the test sample, computed the ratio relative to a reference created from normal samples, and compensated the ratio for a side effect of the normalization procedure that scaled all chromosomally normal DNA peak areas slightly up or down depending on the kind of aneuploidy present. For the chromosomes 13, 18, 21, X, and Y, probe reliability weighted mean ratio values and corresponding SDs were calculated, and the significance for being outside a reference interval around ratio 1.0 was tested. p,,,1% suggested aneuploidy and 1,<,p,,,5% suggested potential aneuploidy. Individual peaks, where the normalized area was situated more than 4 SD from the corresponding reference, suggested possible partial deletion or gain. Sample quality was automatically assessed. Control probes were not required. Having used the software and methods for two years, we conclude that a reliable, objective, and fast workflow is obtained. [source]


A new Cosserat-like constitutive model for bedded salt rocks

Abstract Salt rocks are commonly used as geologic host rocks for storage of gas and crude oil, and are being considered for the disposal of radioactive waste. Different from the salt rock domes in many countries, the salt rock formations in China are usually laminar with many alternating layers, i.e. rock salt, anhydrite, and/or mudstone. Considering the unique stratigraphic characteristics of these salt rocks, a new Cosserat-like medium constitutive model is proposed in order to facilitate efficient modeling of the mechanical behavior of these formations. In this model, a new representative volume element, containing two different layers, is employed to simulate the compatibility of the meso-displacement between two different layers and also the bending effect. A new method for the deformation and failure analysis of bedded salt rocks is derived therefrom. Having the macro-average stresses, the conventional stresses in the different layers can be obtained in sequence. The conventional stresses can then be utilized in a routine way for the strength and failure analysis. For the initial numerical modeling, the new Cosserat-like medium is reduced to a transversely isotropic one. The simplified constitutive model for layered media is then implemented into FLAC3D codes. A test sample validates that the results by using the numerical model are in good agreement with that by using the built-in model, and the mesh size for the new model is reduced greatly. Finally, an application for the stability of oil storage caverns in deep thinly bedded salt rocks is carried out. The effects on convergence of storage caverns and on the failure of surrounding rock due to the presence of the mudstone interlayers (hard phase) are discussed in detail. Genetic variability of seven dog breeds based on microsatellite markers

Summary The present study, compared the genetic variability of seven dog breeds and a test sample from Switzerland by means of 26 microsatellite markers. Five loci were excluded from further analyses because one was monomorphic, one not in Hardy,Weinberg equilibrium in all breeds and three in linkage disequilibrium with linked loci. The proportion of shared alleles at the individual level of the remaining 21 microsatellite markers combined with the neighbour-joining method allowed for the clustering of the large majority of the individuals in accordance to their breed. The results were confirmed by analyses using a Bayesian approach for clustering and a Monte Carlo re-sampling method for individual assignment or exclusion to a given population. [source]


Multislice Computed Tomography of the First Rib: A Useful Technique for Bone Age Assessment

Abstract:, Macroscopic study of the first rib has been described by Kunos et al. as an efficient method of age estimation. We retrospectively reviewed a test sample of 160 first right ribs obtained from multislice computed tomography (MSCT) clinical investigations of living individuals aged 15,30 years old. Based on the descriptions of Kunos et al., we analyzed the morphological appearance of the sternal end of the first rib on two- and three-dimensional MSCT reconstructions and defined changes in appearance in stages (from 2 to 5). We also studied the calcification and ossification of the costal cartilage as an independent feature. By statistical analysis (ANOVA), we determined for each gender the correlation between the estimated stage of the costal face, the appearance of the costal cartilage, and the documented age of the subjects. We demonstrated that MSCT of the first rib appears to be an efficient noninvasive modality for bone age estimation. [source]

A new technique for differentiating between 2J(C,H) and 3/4J(C,H) connectivities

Abstract We present a new pulse sequence that yields two simultaneously detected types of long-range correlation spectra. The one spectrum is to show all nJ(C,H) connectivities and the other is to show exclusively 2J(C,H) connectivities. The method is demonstrated by using strychnine as a test sample. A comparison with HMBC shows that the 2J(C,H)/nJ(C,H) experiment supplies a nJ(C,H) spectrum that is of equal standard with regard to sensitivity and spectral information. The additional 2J(C,H) spectrum allows the disentanglement of 2J(C,H) and nJ(C,H) signals (n > 2) in HMBC type spectra, which greatly simplifies signal assignment and structure elucidation in general. Wide-range length metrology by dual-imaging-unit atomic force microscope based on porous alumina

Abstract A new dual-imaging-unit atomic force microscope (DIU-AFM) was developed for wide-range length metrology. In the DIU-AFM, two AFM units were combined, one as a reference unit, and the other a test one. Their probes with Z piezo elements and tips were horizontally set in parallel at the same height to reduce errors due to geometric asymmetry. An XY scanner was attached to an XY block that was able to move in the X direction with a step of about 500 nm. A standard porous alumina film was employed as the reference sample. Both reference sample and test sample were installed at the center of the XY scanner on the same surface and were simultaneously imaged. The two images had the same lateral size, and thus the length of the test sample image could be accurately measured by counting the number of periodic features of the reference one. The XY block together with the XY scanner were next moved in the X direction for about 1.5 ,m and a second pair of reference and test images were obtained by activating the scanner. In this way, a series of pairs of images were acquired and could be spliced into two wide-range reference and test images, respectively. Again, the two spliced images were of the same size and the length of test image was measured based on the reference one. This article presents a discussion about the structure and control of the DIU-AFM system. Some experiments were carried out on the system to demonstrate the method of length calculation and measurement. Experiments show a satisfactory result of wide-range length metrology based on the hexagonal features of the porous alumina with a periodic length of several tens of nanometers. Using this method the DIU-AFM is capable of realizing nanometer-order accuracy length metrology when covering a wide range from micron to several hundreds of microns, or even up to millimeter order. Isolation and characterization of microsatellites in the bryozoan Crisia denticulata

Abstract Cyclostomata bryozoa are thought to reproduce via polyembryony, a clonal replication of a fertilized egg. To test this hypothesis and to assess the impact of their reproductive strategy on the structure of populations, we isolated microsatellite markers in Crisia denticulata (Cyclostomata, Stenolemata), using an initial enrichment step with random amplified polymorphic DNA (RAPD) primers. A total of nine microsatellites, one tetra- and eight dinucleotides repeats were isolated; seven were found to be polymorphic in a test sample of 30 individuals, with allele numbers/locus varying from 2 to 6. The tetranucleotide locus showed heterozygote deficiency. These primers did not amplify the DNA of Crisia eburnea. [source]


Herpesviruses in periodontal pocket and gingival tissue specimens

Human cytomegalovirus (HCMV) and Epstein-Barr virus type 1 (EBV-1) are frequently detected in crevicular fluid of deep periodontal pockets, but little or no information is available on occurence of herpesviruses in gingival tissue. This investigation studied the presence of herpesviruses in periodontal pockets and the corresponding gingival tissues from 11 periodontally healthy and 14 periodontitis sites. A nested-polymerase chain reaction was employed to identify the presence of HCMV, EBV-1, EBV-2, herpes simplex virus, human herpesvirus (HHV)-6, HHV-7 and HHV-8 in each test sample. In healthy periodontal sites, HCMV was detected in 1 (9%) and EBV-1 in 2 (18%) pocket samples, and HCMV was detected in 2 (18%) and EBV-1 in 3 (27%) gingival tissue samples. In periodontitis lesions, HCMV was detected in 9 (64%) pocket samples and in 12 (86%) gingival tissue samples, and EBV-1 was detected in 6 (43%) pocket samples and in 11 (79%) gingival tissue samples. HHV-6 and HHV-8 were detected exclusively in gingival tissue samples. The present findings confirm the frequent presence of HCMV and EBV-1 in periodontitis lesions and suggest using gingival tissue specimens for detecting periodontal HHV-6, HHV-7 and HHV-8. [source]


Prospective determination of the specificity of a commercial snake venom detection kit in urine samples from dogs and cats

Objective To determine the specificity of a snake venom detection kit in urine samples from dogs and cats presenting to a referral centre for diseases unrelated to snake envenomation. Design Urine was collected from 50 dog and 25 cats presented for investigation and treatment of diseases unrelated to snake envenomation. Urine was collected as a voided sample, by cystocentesis or by catheterisation, and routine urinanalysis was performed. Snake venom testing was performed within 2 h of collection using a commercially available snake venom detection kit, which was observed continuously during the 10-min colour reaction phase for evidence of a visible colour indicating a positive test. Results No false-positive reactions occurred in any sample analysed. Conclusion The snake venom detection kit appears to have 100% specificity for using urine as a test sample. [source]


Identifying Combinations of Cancer Markers for Further Study as Triggers of Early Intervention

Summary. In many long-term clinical trials or cohort studies, investigators repeatedly collect and store tissue or serum specimens and later test specimens from cancer cases and a random sample of controls for potential markers for cancer. An important question is what combination, if any, of the molecular markers should be studied in a future trial as a trigger for early intervention. To answer this question, we summarized the performance of various combinations using Receiver Operating Characteristic (ROC) curves, which plot true versus false positive rates. To construct the ROC curves, we proposed a new class of nonparametric algorithms which extends the ROC paradigm to multiple tests. We fit various combinations of markers to a training sample and evaluated the performance in a test sample using a target region based on a utility function. We applied the methodology to the following markers for prostate cancer, the last value of total prostate-specific antigen (PSA), the last ratio of total to free PSA, the last slope of total PSA, and the last slope of the ratio. In the test sample, the ROC curve for last total PSA was slightly closer to the target region than the ROC curve for a combination of four markers. In a separate validation sample, the ROC curve for last total PSA intersected the target region in 77% of bootstrap replications, indicating some promise for further study. The influence of impact object characteristics on impact force and force absorption by mouthguard material

Abstract,,, Most impact force and impact energy absorption tests for mouthguards have used a steel ball in a drop-ball or the pendulum device. However, in reality most sports-related trauma is caused by objects other than the steel ball, e.g. various sized balls, hockey puck, or bat or stick. Also, the elasticity, the velocity and the mass of the object could change the degree and the extent of injuries. In this study, we attempted to measure the impact force from actual sports equipment in order to clarify the exact mechanism of dental-related sports injuries and the protective effects of mouthguards. The present study was conducted using the pendulum impact device and load cell. Impact objects were removable. Seven mobile impact objects were selected for testing: a steel ball, baseball, softball, field hockey ball, ice hockey puck, cricket ball, and wooden baseball bat. The mouthguard material used in this study was a 3-mm-thick Drufosoft (Dreve-Dentamid GmbH, Unna, Germany), and test samples were made of the one-layer type. The peak transmitted forces without mouthguard ranged from the smallest (ice hockey stick, 46.9 kgf) to the biggest (steel ball, 481.6 kgf). The peak transmitted forces were smaller when the mouthguard was attached than without it for all impact materials but the effect was significantly influenced by the object type. The steel ball showed the biggest (62.1%) absorption ability while the wooden bat showed the second biggest (38.3%). The other balls or the puck showed from 0.6 to 6.0% absorbency. These results show that it is important to test the effectiveness of mouthguards on specific types of sports equipment. In future, we may select different materials and mouthguard designs suitable for specific sports. [source]


Papanicolaou tests associated with cervical mucosal endometriosis: An analysis of cellular features and comparison to endocervical adenocarcinoma in situ

Abstract Endometrium directly sampled from endocervical mucosal endometriosis can mimic endocervical adenocarcinoma in situ (AIS) in Papanicolaou (Pap) tests. We analyzed a series of Pap tests to investigate the cellular features of mucosal endometriosis and to assess the utility of stroma and apoptotic bodies in the differential diagnosis with AIS. Pap test samples from patients known to have endocervical mucosal endometriosis were compared with samples containing AIS. Pap tests from patients with mucosal endometriosis had lesional cells in 13 (62%) cases which includes glandular and stromal cells (10 cases), stroma only (two cases), and glandular cells only (one case). Three (23%) cases had gland-stromal aggregates. Three (23%) cases had mitotic figures and two (15%) had apoptotic bodies. By comparison, only one (8%) AIS case had endometrial-type stroma. Seven (58%) AIS cases had apoptotic bodies and three (25%) had mitotic figures. We conclude that Pap tests from patients with mucosal endometriosis usually (62%) have lesional cells. These lesional cells almost always include stroma, which is useful in the differential diagnosis with AIS. We identified stroma significantly more often in endometriosis cases (92%) than in AIS cases (8%). Pathologists should look for endometrial stroma when considering an interpretation of directly sampled endometrium. In the absence of stroma, AIS should be considered. Diagn. A robust cross-linked polyacrylamide coating for microchip electrophoresis of dsDNA fragments

Abstract Surface derivatization plays an important role in microchip electrophoresis. It not only enhances the resolution, but also improves the reproducibility. So far, the most popularly used derivatization method for glass microchannels is to covalently attach a layer of linear polyacrylamide,(LPA) to the channel surfaces. However, LPA coating has two problems: incomplete coverage and limited lifetime. To address these issues, we have recently developed a cross-linked polyacrylamide,(CPA) derivatization protocol and demonstrated it for high-resolution protein separations by CIEF, CGE, and CZE. In this report, we used this protocol to coat microchip channels and exhibited the reliability and robustness of CPA coating for microchip electrophoresis of DNA molecules. dsDNA fragments were used as our test samples. High resolutions were obtained for fragments ranging from 100,bp to 10,kpb. After more than 800,runs, the CPA-coated microchannels still performed well and comparable resolutions were maintained throughout these runs. [source]


Interlaboratory comparison of a reduced volume marine sediment toxicity test method using the amphipod Ampelisca abdita

Abstract The U.S. Environmental Protection Agency has standardized methods for performing acute marine amphipod sediment toxicity tests. A test design reducing sediment volume from 200 to 50 ml and overlying water from 600 to 150 ml was recently proposed. An interlaboratory comparison was conducted to evaluate the precision of this reduced sediment volume toxicity test method using the marine amphipod Ampelisca abdita. A negative control and three sediment samples of varying degrees of toxicity ranging from low to high were tested by six laboratories. Complete agreement was reached in rank of relative toxicity for all samples tested by five out of six laboratories. Test acceptability for control survival was achieved by all laboratories, and 69% agreement in classification of the sediments as toxic or nontoxic was documented. Coefficients of variation in all test samples were similar to those reported in other interlaboratory studies using marine amphipods. Results of this study indicate that the reduced sediment volume test using A. abdita is a reliable and precise measure of acute toxicity in marine sediment samples. [source]


Structural characteristics of clay-dominated soils of a marsh and a palaeosol in a crossed diagram

Summary Although the shrink-swell phenomenon of clays has been thoroughly studied, the in situ relation of the shrinkage curve to the structure profile is rarely presented from the shrinkage limit to the liquid limit. We studied the consolidated structure of clay-dominated (<2 ,m) soils formed on ,pseudo-liquid' marsh sediments in the ,Marais de l'Ouest' (France). The profiles were studied in a grassland field and in a sunflower field from unsaturated surface soils down to deeper, saturated, levels characterized by a very large water content (100% by weight). The consolidation states were quantified recording cone resistance (Qd) profiles using a dynamic penetrometer in successive seasons. These Qd profiles were compared with the associated wet density and gravimetric water-content profiles. Two consolidation depths were evident, the surface soil and a 130-cm deep palaeosol. The seasonal Qd profiles demonstrate the partial irreversibility of the consolidation peaks associated with the surface soil and with the palaeosol. The shrinkage properties were established through drying curves of undisturbed test samples. In the void ratio (e) - water content (W) and water content (W) - saturation index (Sr) diagrams, the profiles as a whole exhibit only one clay soil behaviour from their pseudo-liquid to plastic to solid states. Each Qd profile is represented by a hyperbolic curve in the e/Qd diagram. Represented in a (e , W , Qd ,Sr) crossed diagram, the vertical evolution of the successive profiles shows the soil structure behaviour from the initial pseudo-liquid sediment to the consolidated soil. A simple cone resistance recording associated with gravimetric water-content profiles, characterizes the evolution of structural layers of soils for the seasonal drying-wetting cycles, for the over-consolidation associated with the palaeosol, and also for the effect of ploughing. Determination of Mechanical Properties of Copper at the Micron Scale,

Using a focused ion beam workstation, micron-sized bending and compression samples were fabricated from a pure copper single crystal. The bending and compression experiments exhibited a strong size effect on the flow stress of copper, reaching values in the order of 1,GPa for the smallest test structures. Conventional strain gradient plasticity approaches are not capable of explaining this behaviour. The surface damage introduced by Ga+ ion implantation during focused ion beam preparation was investigated using Auger electron spectroscopy and its consequence on the mechanical response of the miniaturized test samples is addressed. Spatial prediction of nitrate pollution in groundwaters using neural networks and GIS: an application to South Rhodope aquifer (Thrace, Greece)

Abstract Neural network techniques combined with Geographical Information Systems (GIS), are used in the spatial prediction of nitrate pollution in groundwaters. Initially, the most important parameters controlling groundwater pollution by nitrates are determined. These include hydraulic conductivity of the aquifer, depth to the aquifer, land uses, soil permeability, and fine to coarse grain ratio in the unsaturated zone. All these parameters were quantified in a GIS environment, and were standardized in a common scale. Subsequently, a neural network classification was applied, using a multi-layer perceptron classifier with the back propagation (BP) algorithm, in order to categorize the examined area into categories of groundwater nitrate pollution potential. The methodology was applied to South Rhodope aquifer (Thrace, Greece). The calculation was based on information from 214 training sites, which correspond to monitored nitrate concentrations in groundwaters in the area. The predictive accuracy of the model developed reached 86% in the training samples, 74% in the overall sample and 71% in the test samples. Anomalous Defects and Dynamic Failure of Armor Ceramics

The ballistic performance of state-of-the-art silicon carbide armor material can exhibit a fairly wide variability in certain test configurations, which, it is proposed, may be due to the presence of large (>0.1 mm), rare defects, termed, herein, "anomalous" defects. SiC rubble resulting from ballistic tests was examined, as were quasi-static test samples. Ballistic fragment fracture surfaces revealed large carbonaceous defects that seemed to affect fracture path and mode. Low-strength biaxial flexure samples demonstrated similar defects (>0.1 mm) as failure origins. Carbonaceous defects similar in appearance but smaller in size were also found at the fracture origins of SiC bend bars. Frequently, alumina inclusions were found within the carbonaceous discontinuities. These alumina inclusions may cause the graphitic regions to form during sintering. The random distribution of such large, rare carbonaceous discontinuities from sample-to-sample, as well as batch-to-batch variability, may explain high ballistic variability for SiC armor ceramics. Impact of impregnation chemical on surface glossiness of synthetic, acrylic, polyurethane, and water-based varnishes

Abstract This study was performed to determine the effects of impregnation chemical on surface glossiness of some varnishes. For this purpose, the test samples prepared from Oriental beech, European oak, Scotch pine, Oriental spruce, and Uludag fir woods according to ASTM D 358 were impregnated with Imersol-Aqua according to ASTM D 1413 and producer's definition by short-term (S), medium-term (M), and long-term (L) of dipping methods and coated by synthetic (Sv), acrylic (Ac), water-based (Wb), and polyurethane (Pu) varnishes according to ASTM D 3023. After the varnishing process, the surface glossiness parallel to fibers was determined according to TS 4318. Consequently, among the nonimpregnated wood samples, surface glossiness parallel to fibers was found to be the highest in Oriental spruce and the lowest in European oak. As for the period of dipping, the highest surface glossiness was obtained in medium-term dipping and the lowest in long-term dipping. As for the varnish types, the surface glossiness was found to be the highest in polyurathane varnish and the lowest in water-based varnish. Considering the interaction of wood type, period of impregnation, and type of varnish, surface glossiness was the highest in Uludag fir, long-term dipping, and polyurathane varnish (103.9 gloss) and the lowest in Oriental beech, long-term dipping, and water-based varnish (67.78 gloss). In consequence, in the massive constructions and furniture elements the surface glossiness parallel to fibers after the impregnation with Imersol-Aqua is of great concern, long-term impregnation of Uludag fir materials could be recommended. © 2008 Wiley Periodicals, Inc. A comparison of EDI with solvent-free MALDI and LDI for the analysis of organic pigments

Abstract To evaluate the applicability of EDI to material analysis as a new ionization method, a comparison of EDI with solvent-free matrix-assisted laser desorption ionization (MALDI) and laser desorption ionization (LDI) was made for the analysis of organic pigments, e.g. Pigment Yellow 93, Pigment Yellow 180, and Pigment Green 36, as test samples, which are poorly soluble in standard solvents. In EDI, the samples were prepared in two ways: deposition of suspended samples in appropriate solvents and dried on the substrate, and the direct deposition of the powder samples on the substrate. No matrices were used. Both sample preparation methods gave similar mass spectra. Equally strong signals of [M + H]+ and [M , H], ions were observed with some fragment ions for azo pigments in the respective positive or negative mode of operation. For the powder sample of the phthalocyanine pigment PG36, M+, and [M + H]+ in the positive mode and M,, in the negative mode of operation were observed as major ions. Positive-mode, solvent-free MALDI gave M+, [M + H]+ and [M + Na]+ and negative mode gave [M , H], depending on the sample preparation. Exact mass measurement on an electrospray ionization time-of-flight mass spectrometer: error distribution and selective averaging

Abstract An automated, accurate and reliable way of acquiring and processing flow injection data for exact mass measurement using a bench-top electrospray ionization time-of-flight (ESI-TOF) mass spectrometer is described. Using Visual Basic programs, individual scans were selected objectively with restrictions on ion counts per second for both the compound of interest and the mass reference peaks. The selected ,good scans' were then subjected to two different data-processing schemes (,combine-then-center' and ,center-then-average'), and the results were compared at various ion count limit settings. It was found that, in general, the average of mass values from individual scans is more accurate than the centroid mass value of the combined (same) scans. In order to acquire a large number of good scans in one injection (to increase the sampling size for statistically valid averaging), an on-line dilution chamber was added to slow down the typically rapid mass chromatographic peak decay in flow-injection analysis. This simple addition worked well in automation without the need for manual sample dilution. In addition, by dissolving the reference compound directly into the mobile phase, manual syringe filling can be eliminated. Twenty-seven samples were analyzed with the new acquisition and process routines in positive electrospray ionization mode. For the best method found, the percentage of samples with RMS error less than 5 ppm was 100% with repetitive injection data (6 injections per sample), and 95% with single injection data. Afterwards, 31 other test samples were run (with MW ranging from 310 to 3493 Da, 21 samples in ESI+ and 10 in ESI, mode) and processed with similar parameters and 100% of them were mass-calculated to RMS error less than 5 ppm also. Influence of fibre position on the flexural properties and strain energy of a fibre-reinforced composite

summary, The introduction of laboratory-processed composite systems and fibre reinforcement techniques have increased the possibilities for the prosthetic replacement of missing tooth tissues. Laboratory fabrication variables may significantly influence the properties of the final prosthesis. During the construction of a fibre-reinforced bridge it is necessary to place the fibre at some distance from the fitting surface of the restoration in the pontic region. No guidelines are available for optimal fibre placement in this respect. The purpose of this study was to assess the influence of placing ultra high molecular weight polyethylene (UHMWPE) fibre at five different distances from the tensile side of test samples on flexural properties and the strain energy stored within the dental composite. The results of this investigation showed that whilst moving the fibre reinforcement away from the tensile side by up to 1·5 mm led to a significant reduction in flexural strength, there was no significant decline in the increase in strain energy stored within the tested composite until this distance was exceeded. Is the circumferential tensile modulus within a human medial meniscus affected by the test sample location and cross-sectional area?

Quantifying the material properties of the human menisci is paramount to understanding their biomechanical functions within the knee. One important intrinsic material property governing the biomechanical functions of the meniscus is the circumferential tensile modulus. The purpose of this study was to determine if the circumferential tensile modulus of the human medial meniscus depends on the location and thickness of the sample tested. The following three hypotheses were tested: (a) the circumferential location (anterior, central, and posterior) does not significantly affect the tensile modulus, (b) the radial location (inner to outer) significantly affects the tensile modulus, and (c) the thicknes (cross-sectional area) significantly affects the tensile modulus. Test samples, whose length was oriented in parallel with the circumferential collagen fibers, were collected from different circumferential and radial locations throughout 30 human medial menisci. Samples of three different thicknesses (0.5, 1.5, and 3.0 mm) were taken from three equal groups of 10 menisci (i.e., one thickness per group). The circumferential tensile modulus was measured under quasi-statc loading. Statistical analysis showed no significant effect of the circumferential or radial location of the sample on the circumferential tensile modulus. This indicates that an overall circumferential tensile modulus may be calculated for the human medial meniscus by averaging the values determined at the various locations. However, the thickness of the test sample had a significant effect on the measured circumferential tensile modulus: the modulus varied inversely with the thickness. Thus, moduli determined from test samples that are too small in cross-sectional area overestimate the effective modulus of the tissue on the whole, and the cross-sectional area of the sample must be considered when determining a representative circumferential tensile modulus for the medial meniscus in a human knee. The Anxiolytic Effect of Two Oriental Herbal Drugs in Japan Attributed to Honokiol from Magnolia Bark

An improved elevated plus-maze test in mice revealed that seven daily treatments with two differnt traditional Chinese medicines, known as Kampo medicines in Japan, Hange-koboku-to (composed of extracts of 5 plants) and Saiboku-to (composed of extracts of 10 plants), produced an anxiolytic effect, and the effect was mainly due to the presence of honokiol derived from magnolia. This study was carried out to evaluate the anxiolytic potential of honokiol, Hange-koboku-to and Saiboku-to, which were prescribed with two different magnolia samples: Kara-koboku (Magnoliae officinalis) (KA) or Wa-koboku (Magnoliae obovata) (WA). The doses of test samples were adjusted to ensure a constant dose of honokiol at 0.2 mg kg,1. Although the doses of magnolol (an isomer of honokiol), as well as those of undetermined chemicals, varied among samples, the seven daily treatments with 9 out of 10 test samples produced an anxiolytic effect almost equivalent to that produced by 0.2 mg kg,1 honokiol. The only exception was the sample containing the lowest amount of honokiol. Magnolia-free preparations of Hange-koboku-to or Saiboku-to did not have any anxiolytic effect. These results confirm that honokiol derived from magnolia is the causal chemical of the anxiolytic effect of Hange-koboku-to and Saiboku-to. [source]


Ethanol-Induced Malfunction of Neutrophils Respiratory Burst on Patients Suffering From Alcohol Dependence

Background:, Polymorphonuclear, neutrophil granulocytes (PMN) play a major role in the control of infections, and people who abuse alcohol are susceptible to infections. Resistance against infections ensues intracellularly following initial phagocytosis of microorganisms with the oxygen-dependent respiratory burst, the key enzyme of which is the respiratory burst oxidase, whereby oxygen radicals are produced for microbial destruction. To date there is insufficient information available in connection with the process of impaired defence against infection in patients suffering from alcohol dependence. Therefore, our investigation was carried out to determine the influence of alcohol exposition on the formation of oxygen radicals and the respiratory burst. Methods:, 4.5 ml of whole blood was taken from 10 healthy adults and 10 patients suffering from alcohol dependence. An additional 3.5 ml of whole blood was taken from the alcoholic patients for determination of the blood alcohol concentration. The respiratory burst of PMN was tested using the Four-Colour-Continuous Flow Cytometer. Each experimental procedure consisted of 4 test samples [negative controls, Escherichia coli, FMLP-supplement (N-formyl-l-methionyl-l-leucyl-l-phenylalanin), PMA-supplement (phorbol-12-myristate-13-acetate)]. Differing concentrations of ethanol were also introduced to each of the tests performed (0.20 to 4.00 g/l). Results:, Ethanol revealed a marked decrease of burst activity in those patients suffering from alcoholism with increased alcohol concentration. A dependence between the burst activity and the ethanol concentration was seen to be statistically significant. This effect was only evident after stimulation with E. coli and FMLP in those patients with alcohol dependence. Conclusion:, The results presented in this study show an impairment in the function of PMN in those patients addicted to alcohol due to the decrease in burst activity. In view of the results of the different stimuli, the second-messenger effects were not evident. A clarification of this phenomenon could well be assumed as an allosteric receptor effect on the burst oxidase, namely, a direct effect on the phagocytosis interaction between circulating granulocytes and causative organisms.
