Thirty-year durability of a 20-mil PVC geomembrane

In 1971, twenty circular aquaculture ponds were constructed for the W. K. Kellogg Biological Research Station in Hickory Corners, Michigan. The 30.5-m-diameter research ponds were lined using a 0.51-mm-thick fish-grade PVC geomembrane. Over the years the ponds became congested with dense, persistent stands of cattails, trees, and other vegetation, which required the ponds to be cleared and relined in September 2000 in order to allow the initiation of new experiments. The lack of holes in the exhumed geomembrane suggests that it resisted biological attack from microorganisms and also root penetration. Laboratory testing shows that the tensile behavior of the nearly 30-year-old PVC geomembrane is within current specifications for new 0.51-mm-thick PVC geomembranes. Test results also indicate that performing laboratory tests at in-situ moisture conditions provides a better estimate of the field properties of PVC geomembranes than desiccating the material prior to testing, as is required by ASTM Standard Test Methods. Information derived from sensitization test methods: test sensitivity, false positives and false negatives

Predictive toxicology tests for the prospective identification of skin-sensitizing chemicals are well known and have been used for many years. However, of these, only the local lymph node assay (LLNA) has actually undergone formal independent assessment to determine the accuracy of the predictions, particularly with respect to the likelihood of false positives and false negatives. Often, efforts to increase the sensitivity of a test (reducing false negatives) tend to increase the number of false positives. In this short review, these issues are discussed in particular relation to the 3 predictive tests available in regulatory toxicology, the guinea-pig maximization test, the occluded patch test of Buehler and the LLNA. A key perspective is that no predictive test is without limitations; having a good appreciation of these limitations is necessary for making the best use of the information derived from these methods. Are we reaching the limits or our ability to detect skin effects with our current testing and measuring methods for consumer products?

Testing for potential adverse skin effects is a key part of both the overall safety assessment for many consumer products and the evaluation of potential product improvements in mildness. Whilst modern tissue and paper products (i.e. facial tissues, catamenial products, baby wipes and baby and adult diapers) are inherently very mild to skin, current test methodology may not be robust enough to evaluate future improvements in such products. This article provides a commentary on several technologies we have been exploring to improve the sensitivity of test methods for tissue and paper products. The focus has been on three approaches: (i) further exaggerating exposure conditions using novel approaches to sample application, (ii) increasing the sensitivity of the manner in which we score for irritant effects, either visually or via instrumentation and (iii) quantitatively measuring additional endpoints, i.e. subjective sensory effects. A chemical dataset for evaluation of alternative approaches to skin-sensitization testing

Allergic contact dermatitis resulting from skin sensitization is a common occupational and environmental health problem. In recent years, the local lymph node assay (LLNA) has emerged as a practical option for assessing the skin-sensitization potential of chemicals. In addition to accurate identification of skin sensitizers, the LLNA can also provide a reliable measure of relative sensitization potency, information that is pivotal in successful management of human health risks. However, even with the significant animal welfare benefits provided by the LLNA, there is interest still in the development of non-animal test methods for skin sensitization. Here, we provide a dataset of chemicals that have been tested in the LLNA and the activity of which correspond with what is known of their potential to cause skin sensitization in humans. It is anticipated that this will be of value to other investigators in the evaluation and calibration of novel approaches to skin-sensitization testing. The materials that comprise this dataset encompass both the chemical and biological diversity of known chemical allergens and provide also examples of negative controls. It is hoped that this dataset will accelerate the development, evaluation and eventual validation of new approaches to skin-sensitization testing. [source]


Stability analysis for real-time pseudodynamic and hybrid pseudodynamic testing with multiple sources of delay

Abstract Real-time pseudodynamic (PSD) and hybrid PSD test methods are experimental techniques to obtain the response of structures, where restoring force feedback is used by an integration algorithm to generate command displacements. Time delays in the restoring force feedback from the physical test structure and/or the analytical substructure cause inaccuracies and can potentially destabilize the system. In this paper a method for investigating the stability of structural systems involved in real-time PSD and hybrid PSD tests with multiple sources of delay is presented. The method involves the use of the pseudodelay technique to perform an exact mapping of fixed delay terms to determine the stability boundary. The approach described here is intended to be a practical one that enables the requirements for a real-time testing system to be established in terms of system parameters when multiple sources of delay exist. Several real-time testing scenarios with delay that include single degree of freedom (SDOF) and multi-degree of freedom (MDOF) real-time PSD/hybrid PSD tests are analyzed to illustrate the method. From the stability analysis of the real-time hybrid testing of an SDOF test structure, delay-independent stability with respect to either experimental or analytical substructure delay is shown to exist. The conditions that the structural properties must satisfy in order for delay-independent stability to exist are derived. Real-time hybrid PSD testing of an MDOF structure equipped with a passive damper is also investigated, where observations from six different cases related to the stability plane behavior are summarized. Software framework for distributed experimental,computational simulation of structural systems

Abstract Supported by the recent advancement of experimental test methods, numerical simulation, and high-speed communication networks, it is possible to distribute geographically the testing of structural systems using hybrid experimental,computational simulation. One of the barriers for this advanced testing is the lack of flexible software for hybrid simulation using heterogeneous experimental equipment. To address this need, an object-oriented software framework is designed, developed, implemented, and demonstrated for distributed experimental,computational simulation of structural systems. The software computes the imposed displacements for a range of test methods and co-ordinates the control of local and distributed configurations of experimental equipment. The object-oriented design of the software promotes the sharing of modules for experimental equipment, test set-ups, simulation models, and test methods. The communication model for distributed hybrid testing is similar to that used for parallel computing to solve structural simulation problems. As a demonstration, a distributed pseudodynamic test was conducted using a client,server approach, in which the server program controlled the test equipment in Japan and the client program performed the computational simulation in the United States. Applications and statistical properties of minimum significant difference-based criterion testing in a toxicity testing program,

Abstract As a follow up to the recommendations of the September 1995 SETAC Pellston Workshop on Whole Effluent Toxicity (WET) on test methods and appropriate endpoints, this paper will discuss the applications and statistical properties of using a statistical criterion of minimum significant difference (MSD). We examined the upper limits of acceptable MSDs as acceptance criterion in the case of normally distributed data. The implications of this approach are examined in terms of false negative rate as well as false positive rate. Results indicated that the proposed approach has reasonable statistical properties. Reproductive data from short-term chronic WET test with Ceriodaphnia dubia tests were used to demonstrate the applications of the proposed approach. The data were collected by the North Carolina Department of Environment, Health, and Natural Resources (Raleigh, NC, USA) as part of their National Pollutant Discharge Elimination System program. Identification of pathotypes of Synchytrium endobioticum found in infested fields in Turkey

Pathotypes of four isolates of Synchytrium endobioticum collected from the infested areas of Turkey were determined using Glynne,Lemmerzahl and Spieckermann test methods. Two isolates (Ordu 1 and Nev,ehir 1) were tested by the Glynne,Lemmerzahl method in Germany. Ordu 1 was assigned as pathotype 1(D1) whereas the other isolate gave different results in 2 years of testing [pathotype 6(O1) or 18(T1)]. Two other isolates (Nev,ehir 2 and Nev,ehir 3) were tested using the Spieckermann method in Netherlands. Neither of these last two isolates tested were found to belong to the pathotypes known from Western Europe. The isolates Nev,ehir 2 and Nev,ehir 3 were given a new, unique code: 38 (Nev,ehir). "PHYBAL" a Short-Time Procedure for a Reliable Fatigue Life Calculation

Abstract The reliable calculation of the fatigue life of high-strength steels and components requires the systematic investigation of the cyclic deformation behaviour and the comprehensive evaluation of proceeding fatigue damage. Besides mechanical stress-strain hysteresis measurements, temperature and electrical resistance measurements were used for the detailed characterisation of the fatigue behaviour of the steel SAE 4140 in one quenched and tempered, one normalised, one bainitic and one martensitic condition. To guarantee optimal operation conditions the new fatigue life calculation method "PHYBAL" on the basis of generalised Morrow and Basquin equations was developed. It is a short-time procedure which requires the data of only three fatigue tests for a rapid and nevertheless precise determination of S-N (Woehler) curves. Consequently, "PHYBAL" provides the opportunity to reduce significantly experimental time and costs compared to conventional test methods. [source]

The Cover shows a hybrid joint for multi-material lightweight components realized by ultrasonic metal welding at the Institute of Materials Science and Engineering at the University of Kaiserslautern. Ultrasonic welding is one innovative technology for joining carbon fibre reinforced polymers (CFRP) with sheet metals like aluminium alloys or aluminium plated steels. The achievable mechanical properties of the ultrasonic welded joints were carried out by using statistical test methods. One example for the evaluation of the welding results is presented on the left hand side of the Cover. Additionally a scanning electron micrograph of the bonding zone of an aluminium/CFRP-joint is shown in the background. Carbon dioxide generation calorimetry,Errors induced by the simplifying assumptions in the standard test methods

Abstract Carbon dioxide generation (CDG) calorimetry is commonly used for measuring heat release rates of materials. Calorimetric equation provided in the ASTM E 2058 and the NFPA code 287 is a simplified equation since the water content in the ambient air and the fumes as well as the expansion factor of the combustion gases are neglected. This paper provides general equation for CDG calorimetry based on the Tewarson formulation. A comparison is proposed between the Standard test methods (simplified equation) and the general one. It is shown that the errors induced by the simplifying assumptions are negligible for oxygen depletion factor values commonly encountered in the Fire Propagation Apparatus (where large dilution factors of the combustion gases are used before the measurements of species concentrations). Animal use replacement, reduction, and refinement: Development of an integrated testing strategy for bioconcentration of chemicals in fish,

Abstract When addressing the use of fish for the environmental safety of chemicals and effluents, there are many opportunities for applying the principles of the 3Rs: Reduce, Refine, and Replace. The current environmental regulatory testing strategy for bioconcentration and secondary poisoning has been reviewed, and alternative approaches that provide useful information are described. Several approaches can be used to reduce the number of fish used in the Organization for Economic Cooperation and Development (OECD) Test Guideline 305, including alternative in vivo test methods such as the dietary accumulation test and the static exposure approach. The best replacement approach would seem to use read-across, chemical grouping, and quantitative structure-activity relationships with an assessment of the key processes in bioconcentration: Adsorption, distribution, metabolism, and excretion. Biomimetic extraction has particular usefulness in addressing bioavailable chemicals and is in some circumstances capable of predicting uptake. Use of alternative organisms such as invertebrates should also be considered. A single cut-off value for molecular weight and size beyond which no absorption will take place cannot be identified. Recommendations for their use in bioaccumulative (B) categorization schemes are provided. Assessment of biotransformation with in vitro assays and in silico approaches holds significant promise. Further research is needed to identify their variability and confidence limits and the ways to use this as a basis to estimate bioconcentration factors. A tiered bioconcentration testing strategy has been developed taking account of the alternatives discussed. [source]


SOFCo Planar Solid Oxide Fuel Cell

SOFCo-EFS Holdings LLC has developed a multi-layer, planar solid oxide fuel cell (SOFC) stack that has the potential to provide superior performance and reliability at reduced costs. Our approach combines state-of-the-art SOFC materials with the manufacturing technology and infrastructure established for multi-layer ceramic (MLC) packages for the microelectronics industry. With the proper selection of SOFC materials, implementation of MLC fabrication methods offers unique designs for stacks. Over the past two years, substantial progress has been made in the design and manufacturing development of our second-generation stack. Effective stack and manifold seals have been developed. Cell performance has been improved and relatively low non-cell contributions to stack resistance have been achieved. Stack development has been facilitated through the implementation of two key test methods: (1) a 10-cm single-cell test to bridge the gap in performance data obtained from button cell tests (used for cell R&D) and stack tests; and (2) a novel instrumented short stack (<5 cells) that allows for effective isolation of individual contributions to stack resistance. As a result of progress made to date, a clear pathway for improving stack performance has been established, thereby building confidence that commercial stack performance targets will be reached. Conditioning polymers in today's shampoo formulations , efficacy, mechanism and test methods

Synopsis Today's shampoo formulations are beyond the stage of pure cleansing of the hair. Additional benefits are expected, e.g. conditioning, smoothing of the hair surface, improvement of combability and lather creaminess. Cationic polymers play an important role in providing many of those features. Therefore, within the last few years their use in shampoos has increased greatly. In the only last two decades, shampoo designation has gradually changed from ,2-in-1' to ,3-in-1' and then to ,multifunctional', as at present. The consumer demands products which live up to their promises. Modern shampoos contain a wide variety of ingredients such as co-surfactants, vitamins and pro-vitamins, protein derivatives, silicones, natural-based plant extracts and other ,active ingredients', but there is still a need for conditioning polymers. The specific objective of this study is to assess the conditioning efficacy of cationic polymers and to investigate their mechanisms in a shampoo system. The investigations were carried out on formulations that contained sodium lauryl ether sulphate and different cationic polymers, e.g. Polyquaternium 7, 10, 11, cationic guar gum and Luviquat Care (Polyquaternium 44), a new branched copolymer of vinylpyrrolidone (VP) and quaternized vinylimidazolium salts (QVI). We used test methods relevant to the applications in question, such as combing force measurements, the feel of the hair and the creaminess of the lather, to assess the efficacy. Atomic force microscopy and electrokinetics (streaming potential) were used to detect polymer residues on treated hair. All the polymers under investigation improved the overall performance of the shampoo formulations. This was demonstrated by means of combing force measurements, sensorial tests and analytical methods, namely zeta potential measurement and atomic force microscopy. Polyquaternium 44 exhibited the best conditioning properties on wet hair without sacrificing removability or absence of build-up. The latter are the most striking weaknesses of cationic Guar Gum-based polymers. Polyquaternium 10 can also be removed from the hair after rinsing with anionic surfactant but it does not perform as well as Polyquaternium 44 in the fields of wet combability and sensorial criteria such as lather creaminess and feel of the hair. We postulate that the outstanding properties of Polyquaternium 44 as a conditioning agent for shampoos are due to its tailor-made ,branched' structure. There is a clear correlation between the molecular weight and the efficacy of the new copolymers of VP and QVI. Only cationic polymers with a very high molecular weight are effective as conditioners in shampoos based on anionic surfactants. Surprisingly, they do not have to have a high cationic charge. On the basis of all our results, our postulation is that the polymer residue which is responsible for conditioning does not form a flat layer on the hair. Rather, the polymer residue adsorbs with the few cationic moieties, while the uncharged part of the polymer forms loops, which are orientated away from the hair and which are responsible for the reduced friction between hairs. Résumé Les formulations actuelles de shampoing font plus qu'un simple nettoyage des cheveux. On en attend un intérêt supplémentaire, par exemple après-shampoing, lissage de la surface du cheveu, amélioration de la coiffabilité et aspect crémeux du savon. Les polymères cationiques jouent un rôle important dans l'apport de nombre de ces caractéristiques. Par conséquent, ces quelques dernières années leur utilisation a considérablement augmenté dans les shampoings. Dans les seules deux dernières décades, l'appellation du shampoing est progressivement passée de "2 en 1"à"3 en 1" puis ensuite à"multifonctionnel", comme actuellement. Le consommateur recherche des produits qui tiennent leurs promesses. Les shampoings modernes contiennent une grande diversité d'ingrédients tels que des co-tensioactifs, des vitamines et des provitamines, des dérivés de protéines, des silicones, des extraits à base de plantes naturelles et autres "ingrédients actifs", mais il existe toujours un besoin pour des polymères d'après shampoing. L'objectif spécifique de cette étude est d'évaluer l'efficacité comme après-shampoing de polymères cationiques et de rechercher leurs mécanismes dans le système de shampoing. Les recherches ont été menées sur des formulations qui contiennent du sulfate de lauryl éther sodium et différents polymères cationiques, par exemple du Polyquaternium 7, 10, 11, de la gomme de guar cationique et du Luviquat Care (Polyquaternium 44), un nouveau copolymère ramifié de vinylpyrrolidone (VP) et de sels quaternaires de vinylimidazolium (QVI). Nous avons utilisé les procédés de contrôle appropriées aux applications en question, tels que les mesures de force de coiffage, le toucher du cheveu et l'aspect crémeux du savon, pour évaluer l'efficacité. La microscopie atomique et l'électrocinétique (potentiel d'écoulement) ont été utilisées pour détecter les résidus de polymère sur le cheveu traité. Tous les polymères étudiés améliorent le comportement global des formulations de shampoing. Ceci est démontré au moyen des mesures de force de coiffage, des tests sensoriels et des méthodes analytiques, en l'occurrence la mesure du potentiel zêta et la microscopie atomique. Le Polyquaternium 44 présente les meilleures propriétés d'après-shampoing sur cheveu mouillé sans diminuer sa capacité d'élimination ou l'absence d'accumulation. Ces dernières sont les faiblesses les plus frappantes des polymères à base de gomme de guar cationique. Le Polyquaternium 10 peut aussi être éliminé du cheveu après rinçage avec un tensioactif anionique mais il ne se comporte pas aussi bien que le Polyquaternium 44 dans les domaines de la coiffabilitéà l'état mouillé et des critères sensoriels tels que l'aspect crémeux du savon et du toucher du cheveu. Nous supposons que les propriétés exceptionnelles du Polyquaternium 44 comme agent après-shampoing pour shampoings sont dues à sa structure "ramifiée" conçue sur mesure. Il existe une corrélation claire entre le poids moléculaire et l'efficacité des nouveaux copolymères de VP et QVI. Seuls les polymères cationiques avec un poids moléculaire très élevé sont efficaces comme après shampoings dans des shampoings à base de tensioactifs anioniques. Etonnamment, ils n'ont pas besoin d'avoir une charge cationique élevée. Sur la base de tous nos résultats, notre hypothèse est que le fragment de polymère qui est responsable du traitement ne forme pas une couche plate sur le cheveu. Le fragment de polymère adsorbe plutôt les quelques fragments cationiques, tandis que la partie non chargée du polymère forme des boucles, qui sont orientées à l'extérieur du cheveu et qui sont responsables de la friction réduite entre les cheveux. Right or wrong sample received for coagulation testing?

Tentative algorithms for detection of an incorrect type of sample
Summary Inappropriate blood collection potentially comprises the major pre-analytical problem for coagulation testing. Inappropriate samples are most difficult to detect when received as secondary aliquots, common for referred tests. This study aimed to identify a simple, quick and inexpensive process to help laboratories distinguish the type of sample, should there be suspicion of inappropriate collection. Samples from 15 patients [selected on the basis that four different primary tubes were available: serum, citrated plasma, ethylene diamine tetraacetic acid (EDTA) plasma, lithium-heparin plasma], were tested for common electrolytes that might substantially differ according to the type of sample. In citrated plasma, potassium, chloride, calcium and magnesium were significantly decreased compared with serum and lithium-heparin plasma, while sodium was markedly increased. In EDTA plasma, sodium and chloride were significantly decreased compared with both serum and lithium-heparin plasma, potassium was always >14 mmol/l, whereas magnesium and calcium were virtually undetectable. These data allowed development of two algorithms for differential identification of citrated plasma vs. other samples with 100% sensitivity and specificity, the former based on the sequential measurement of potassium, calcium and sodium, the latter on potassium and sodium. These simple assays can supplement classical coagulation test methods to identify most inappropriate blood collections and validate sample rejection. [source]

Safety, effectiveness and comparability of professional skin cleansers

Summary Background: There are no widely-accepted methodical specifications with which to objectify cleansing effectiveness and skin compatibility of occuptional skin cleansing products in Europe. Therefore the German Social Insurance Agency (DGUV) initiated a study with the goal to evaluate such products in view of the potency and the safety of hand cleansers. A market analysis was a part of the project. Methodology: The product descriptions and safety data sheets of 120 products (5,20/manufacturer) of 11 manufacturers were evaluated between 02/2008 and 04/2008. Results: The manufacturers used mainly ingredients of low irritancy. The declaration of the applied ingredients was in the majority of the cases correctly labeled according to the INCI Declaration. Although there was documentation of skin tolerability for most products, the manufacturers used widely differing tests of skin compatibility. Evidences for cleansing effectiveness were not declared or have not been provided. One manufacturer even promised medical effects of its products. There was no uniform general classification of products making it difficult to identify characteristics of cleansers and choose between them. Conclusions: Presently, there are no commonly accepted criteria to classify products in view of cleansing effectiveness und skin compatibility. Generally accepted criteria and test methods are needed for the evaluation of hand cleansers in order to provide the possibility of transparency and comparability. [source]


Relevance of the developmental toxicity of ethanol in the occupational setting: a review,

Abstract Numerous studies have been conducted investigating the reproductive toxicology of ethanol, the overwhelming majority concerning the adverse effects of consuming alcohol in beverages during pregnancy. Because many of the in vivo studies were designed to model alcoholism, they used comparatively high doses and assessed relatively few endpoints. Outcomes may have been affected by disturbances of metabolism at such high exposures, giving rise to secondary effects on development. The available data on ethanol from ,conventional' developmental toxicity study test methods of the type used for regulatory hazard assessment of chemicals are limited. It is in this context, however, i.e. the use of ethanol as an industrial chemical rather than as a component of beverages, that this review is based. Using the usual criteria applied for the purpose of hazard assessment of industrial chemicals, it is concluded that there is no evidence that industrial exposure to ethanol is a developmental toxicity hazard. Developmental toxicity may result from drinking alcoholic beverages, the threshold level for all aspects of which has yet to be de,ned. This is not, however, considered relevant to the low blood alcohol concentrations resulting from any conceivable inhalation or dermal exposure in the workplace or through the directed use of any consumer product containing ethanol. Methods for the identi,cation of chemical respiratory allergens in rodents: comparisons of cytokine pro,ling with induced changes in serum IgE

Abstract No validated or widely recognized test methods are currently available for the prospective identi,cation of chemicals with the potential to cause respiratory allergy. The cellular and molecular mechanisms that result in the induction of chemical sensitization of the respiratory tract are unclear, although there is evidence for the selective development of T helper 2 (Th2)-type responses and, in some cases, the production of IgE antibody. We have therefore examined the utility of cytokine pro,ling using BALB/c mice, together with the measurement of induced increases in the total serum concentration of IgE in the Brown Norway (BN) rat, as markers for the prospective identi,cation of chemical respiratory allergens. Responses provoked by the reference respiratory allergen trimellitic anhydride (TMA) have been compared with those stimulated by the respiratory sensitizing diisocyanates toluene diisocyanate (TDI) and hexamethylene diisocyanate (HDI) and by the acid anhydride hexahydrophthalic anhydride (HHPA). Topical exposure of BN rats to TMA, TDI and HHPA each provoked marked immune activation (increases in lymph node cellularity and proliferation). However, only treatment with TMA stimulated vigorous increases in the total serum concentration of IgE. In contrast, exposure to HHPA, TDI or HDI failed to provoke signi,cant changes in serum IgE concentration or induced only transient and relatively weak increases in serum IgE levels. In parallel experiments using BALB/c strain mice, however, topical application of all four chemical respiratory allergens provoked a marked Th2-type cytokine secretion pro,le in draining lymph node cells. These data suggest that the measurement of induced changes in serum IgE is not suf,ciently sensitive for the robust identi,cation of chemical respiratory allergens. Furthermore, irrespective of the reasons for variations in TMA-induced IgE production among BN rats, doubts remain regarding the utility of these animals for the characterization of immune responses to chemical allergens. Sonography versus nerve conduction studies in patients referred with a clinical diagnosis of carpal tunnel syndrome

Abstract Purpose. To compare the diagnostic value of high-resolution ultrasound (US) with nerve conduction studies (NCS) in patients with clinically defined carpal tunnel syndrome (CTS). Methods. A prospective study was conducted on 66 consecutive patients investigated for sensory hand symptoms. The gold standard was the clinical diagnosis of CTS. Results. NCS showed greater diagnostic sensitivity (82%) than US (62%) in supporting a diagnosis of CTS. With increasing neurophysiologic severity of median neuropathy, there was increasing convergence of the two test methods. Abnormal US as the only diagnostic supportive evidence of CTS was rare. However, the positive predictive value of US for CTS was 100%. Conclusion. NCS show better sensitivity than US in supporting a diagnosis of CTS. However, because of its high positive predictive value, one may consider using US as a screening test, eliminating the need for NCS in the majority of clinical suspicion of CTS and reserving NCS for cases in which US is negative. © 2009 Wiley Periodicals, Inc. Validation of LAB Color Mode as a Nondestructive Method to Differentiate Black Ballpoint Pen Inks,

Abstract:, Nondestructive digital processing methods such as lab color mode (available in Adobe® Photoshop®) are emerging as alternative methods for forensic document examiners to use when attempting to differentiate writing instrument inks. Although these techniques appear to be viable, little data currently exists regarding the known or potential error rates associated with these techniques. Without adequate data, the validity and reliability of these techniques, including lab color, can not be established. In an attempt to begin to address these issues, 44 black ballpoint ink pens were obtained and used to create 990 pen-pair samples for analysis using established lab color mode techniques. No erroneous findings of "different" were reported following the examination of the known pen-pair combinations in which the same pen was used to create the samples (n = 44). Of the remaining 946 samples, 737 pen-pair samples were differentiated using the lab color mode method, while 209 samples were unable to be differentiated and were recorded as either being "similar" (n = 153) or "unsure" (n = 56). Comparison of the lab color mode results with the results obtained through additional testing using traditional infrared reflectance and infrared luminescence test methods showed that lab color differentiated 102 pen-pair samples (11%; 102/946) that were not differentiated using a VSC-4C. Pharmaceutical antibiotic compounds in soils , a review

Antibiotics are highly effective, bioactive substances. As a result of their consumption, excretion, and persistence, they are disseminated mostly via excrements and enter the soils and other environmental compartments. Resulting residual concentrations in soils range from a few ,g upto g kg,1 and correspond to those found for pesticides. Numerous antibiotic molecules comprise of a non-polar core combined with polar functional moieties. Many antibiotics are amphiphilic or amphoteric and ionize. However, physicochemical properties vary widely among compounds from the various structural classes. Existing analytical methods for environmental samples often combine an extraction with acidic buffered solvents and the use of LC-MS for determination. In soils, adsorption of antibiotics to the organic and mineral exchange sites is mostly due to charge transfer and ion interactions and not to hydrophobic partitioning. Sorption is strongly influenced by the pH of the medium and governs the mobility and transport of the antibiotics. In particular for the strongly adsorbed antibiotics, fast leaching through soils by macropore or preferential transport facilitated by dissolved soil colloids seems to be the major transport process. Antibiotics of numerous classes are photodegraded. However, on soil surfaces this process if of minor influence. Compared to this, biotransformation yields a more effective degradation and inactivation of antibiotics. However, some metabolites still comprise of an antibiotic potency. Degradation of antibiotics is hampered by fixation to the soil matrix; persisting antibiotics were already determined in soils. Effects on soil organisms are very diverse, although all antibiotics are highly bioactive. The absence of effects might in parts be due to a lack of suitable test methods. However, dose and persistence time related effects especially on soil microorganisms are often observed that might cause shifts of the microbial community. Significant effects on soil fauna were only determined for anthelmintics. Due to the antibiotic effect, resistance in soil microorganisms can be provoked by antibiotics. Additionally, the administration of antibiotics mostly causes the formation of resistant microorganisms within the treated body. Hence, resistant microorganisms reach directly the soils with contaminated excrements. When pathogens are resistant or acquire resistance from commensal microorganisms via gene transfer, humans and animals are endangered to suffer from infections that cannot be treated with pharmacotherapy. The uptake into plants even of mobile antibiotics is small. However, effects on plant growth were determined for some species and antibiotics. Antibiotika vieler Strukturklassen können durch Licht abgebaut werden. Dieser Abbaupfad spielt auf Bodenoberflächen jedoch nur eine untergeordnete Rolle. Hingegen kommt es insbesondere durch biologische Transformationsprozesse zu einer intensiven Degradation und Inaktivierung der Antibiotika. Verschiedene Metaboliten weisen jedoch ebenfalls ein antibiotisches Potential auf. Der Abbau der Antibiotika wird durch die Festlegung in Böden gehemmt; dementsprechend wurde eine Persistenz verschiedener Antibiotika nachgewiesen. Trotz der starken bioaktiven Wirkung aller Antibiotika sind die festgestellten Effekte auf Bodenorganismen sehr unterschiedlich. Dies liegt nicht zuletzt an einem Mangel an geeigneten Testmethoden. In der Regel sind jedoch von Dosis und Wirkungsdauer abhängige Effekte insbesondere auf Mikroorganismen festzustellen, die zu Veränderungen der Mikroorganismenpopulation führen können. Lediglich durch Anthelmintika wurden deutliche Wirkungen auf Vertreter der Bodenfauna hervorgerufen. Infolge der antibiotischen Wirkung der Pharmazeutika kann eine Resistenzbildung bei Bodenorganismen ausgelöst werden. Zudem hat die Medikation von Antibiotika die Bildung resistenter Mikroorganismen bereits im behandelten Organismus zur Folge. Durch deren anschließende Ausscheidung gelangen resistente Keime auch direkt in die Böden. Handelt es sich um resistente Pathogene oder kommt es zur Übertragung der Resistenzgene zwischen kommensalen und pathogenen Mikroorganismen, so besteht das erhebliche Risiko einer nicht therapierbaren Infektion von Mensch und Tier. Die Aufnahme selbst mobiler Antibiotika in die Pflanzen ist sehr gering. Dennoch wurden bei einigen Pflanzenarten Wirkungen von Antibiotika auf das Wachstum nachgewiesen. [source]


The European standard for sun-protective clothing: EN 13758

Abstract Clothing is considered one of the most important tools for sun protection. Contrary to popular opinion, however, some summer fabrics provide insufficient ultraviolet (UV) protection. The European Committee for Standardization (CEN), has developed a new standard on requirements for test methods and labelling of sun-protective garments. This document has now been completed and is published. Within CEN, a working group, CEN/TC 248 WG14 ,UV protective clothing', was set up with the mission to produce standards on the UV-protective properties of textile materials. This working group started its activities in 1998 and included 30 experts (dermatologists, physicists, textile technologists, fabric manufacturers and retailers of apparel textiles) from 11 European member states. Within this working group, all medical, ethical, technical and economical aspects of standardization of UV-protective clothing were discussed on the basis of the expertise of each member and in consideration of the relevant literature in this field. Decisions were made in consensus. The first part of the standard (EN 13758-1) deals with all details of test methods (e.g. spectrophotometric measurements) for textile materials and part 2 (EN 13758-2) covers classification and marking of apparel textiles. UV-protective cloths for which compliance with this standard is claimed must fulfill all stringent instructions of testing, classification and marking, including a UV protection factor (UPF) larger than 40 (UPF 40+), average UVA transmission lower than 5%, and design requirements as specified in part 2 of the standard. A pictogram, which is marked with the number of the standard EN 13758-2 and the UPF of 40+, shall be attached to the garment if it is in compliance with the standard. The dermatology community should take cognizance of this new standard document. Garment manufacturers and retailers may now follow these official guidelines for testing and labelling of UV-protective summer clothes, and the sun-aware consumer can easily recognize garments that definitely provide sufficient UV protection. Approaches in the safety evaluations of veterinary antimicrobial agents in food to determine the effects on the human intestinal microflora

The administration of antimicrobial agents to livestock creates potential for antibiotic residues to enter the food supply and be consumed by humans. Therefore, as a process of food animal drug registration, national regulatory agencies and international committees evaluate data regarding the chemical, microbiologic, pharmacokinetic, pharmacodynamic, pharmacologic, toxicologic, and antimicrobial properties of veterinary drugs to assess the safety of ingested antimicrobial residues to consumers. Currently, European, Australian and United States guidelines for veterinary drug registration require a safety assessment of microbiologic hazards from consumption of antimicrobial residues taking into account the potentially adverse effects on human intestinal microflora. The main concerns addressed are selection of resistant bacteria in the gastrointestinal tract and disruption of the colonization barrier of the resident intestinal microflora. Current requirements differ among national agencies. Efforts are ongoing internationally to review and harmonize approaches and test methods and protocols for application to these microbiologic safety evaluations of antimicrobial drug residues in food. This review describes the background to current regulatory approaches used in applying in vitro and in vivo methods to set a microbiologic acceptable daily intake for residues in food derived from animals treated with an antimicrobial agent. Evaluation of the usefulness of a new direct immunofluorescence assay (ScanVIT-LegionellaÔ) for monitoring hospital water systems contaminated with Legionella spp.

Abstract Aims:, To compare the efficiency of the ScanVIT-LegionellaÔ test (Vermicon, Munich, Germany) vs a conventional culture method for the quantification of Legionella spp. in hospital water samples in daily hospital practice. Methods and Results:, The detection of Legionella spp. takes place on a cultivated filter brought into contact with dye-marked gene probes. The results are analysed under fluorescence microscopy. Bacteria that light up green belong to the genus Legionella; those that light up both green and red belong to the species Legionella pneumophila. Our results showed that the ScanVIT test has a sensitivity of 90%; agreement between the two methods was 82%. In the 48 samples that tested positive with both methods, the Legionella concentration detected by the culture method was consistently higher. A statistically significant difference between the results obtained with the two test methods emerged at the Wilcoxon test (P < 0·001). Conclusion:, The ScanVIT test may be recommended for investigating the presence of Legionella by qualitative testing. Significance and Impact of the Study:, Given the simplicity of colony identification by fluorescence, the ScanVIT test can be used in laboratories where staffs are not experienced in identifying typical colonies of Legionella. The repeatability of friction and wear results obtained from unlubricated reciprocating sliding tests

Abstract Laboratory tests can help in the analysis of tribological failures of elements, and improve tribo-systems by choosing appropriate materials. In order to characterise the friction and wear behaviour of candidate materials, various different test methods have been developed in the past and are still in use. One such method is the reciprocating sliding of a ball against a disc. In the work reported here, the repeatability of friction and wear results was evaluated with ten tests under identical conditions with a steel (100Cr6) or alumina (Al2O3) ball against a steel (100Cr6) disc under unlubricated conditions at room temperature. The influence of ambient humidity on friction and wear behaviour was determined in three additional tests in dry and in moist air, respectively. The repeatability of friction coefficient in normal air was better than 5% for alumina/100Cr6 and 12% for 100Cr6/100Cr6, while the repeatability of volumetric wear was slightly better than 10% for alumina/steel, and slightly worse than 10% for steel/steel. For both couples the coefficient of friction is lowest in moist air and about 50% higher in dry air. The coefficient of wear is also least in moist air and higher by a factor of 3(5) in dry air for tests with a 100Cr6 (alumina) ball. [source]

Abstract Methods for service life prediction of reinforced concrete structures exposed to chloride-bearing environments require, amongst other parameters, the knowledge of the chloride threshold for pitting corrosion initiation (Clth). Nowadays, although the main factors influencing the chloride threshold are well known, it is often difficult to quantify a value of the chloride threshold, partly because of its intrinsic high variability, and partly because of the different test methods that have been used to measure it. All the experimental tests rely on the detection of steel depassivation and simultaneous measurement of chloride content or steel potential. This paper deals with the methods that can be used to detect steel depassivation in relation with the determination of the chloride threshold. Tests in concrete-pore-simulating solutions as well as tests in concrete will be considered, and advantages and limitations will be discussed. [source]


Atopy patch test in patients with atopic eczema/dermatitis syndrome: comparison of petrolatum and aqueous solution as a vehicle

Background:, The atopy patch test (APT) is an in vivo model to study the induction of eczema by inhalant allergens. This study was designed to compare two commonly used APT methods. Methods:, In the first method, the allergen is dissolved in aqueous solution, which is applied on tape-stripped skin. In the second method, the allergen is dissolved in petrolatum and applied without tape stripping. Thirteen patients with atopic dermatitis sensitized to inhalant allergens were patch tested using both methods. Reactions were evaluated macroscopically and microscopically after 48 h. Results:, Nine out of 13 patients displayed a positive reaction for both methods. One patient had a positive APT for the aqueous method alone and three for the petrolatum method alone. Reactions were significantly stronger when using the petrolatum method. Histological evaluation of the nine patients positive for both methods showed no significant differences in number of eosinophils, T-cells and neutrophils. Conclusion:, The APT using the petrolatum vehicle induces a higher number of positive reactions and is significantly stronger relative to the APT using allergen in aqueous vehicle. The cellular influx in both test methods is comparable. Both methods can be used to study the mechanisms in the induction of eczema by inhalant allergens. [source]
