Identification of an operon and inducing peptide involved in the production of lactacin B by Lactobacillus acidophilus

Abstract Aim: To determine if a 9·5-kb region on the Lactobacillus acidophilus NCFM genome, encoded the genetic determinants for regulation and production of lactacin B, a class II bacteriocin. Methods: Transcriptional analysis was used to identify a 9·5-kb polycistronic region suspected of encoding the lab operon. The 12 putative open reading frames (LBA1803,LBA1791) were organized into three clusters: a production and regulation cluster encoding a putative two-component signal transduction system; an export cluster encoding a putative ABC transporter and a final cluster composed of three unknown proteins. Seven genes were typical of bacteriocins, encoding small, cationic peptides, each with an N-terminal double-glycine leader motif. Inactivation of a predicted ABC transporter completely abolished bacteriocin activity. When cloned and expressed together, LBA1803,LBA1800 resulted in markedly higher levels of lactacin B activity. The four peptides were chemically synthesized but exhibited no bacteriocin activity, alone or in combination. Only LBA1800 induced lactacin B production in broth cultures. Conclusions: Lactacin B production is encoded within the 9·5-kb lab operon of 12 genes that are transcribed in a single transcript. LBA1800 is an inducing peptide of bacteriocin production. Significance and Impact of the Study: A three-component regulatory system common to class II bacteriocins regulates the production of this bacteriocin by Lact. acidophilus. [source]


Disciplinary Observance and Sanctions on German and Danish Auditors

This paper presents the results of a comparative study on disciplinary observance systems of the auditing profession within two member states of the European Union: Germany and Denmark. Disciplinary observance is an important factor in reducing the hidden action problem, but could also be used by the profession to signal quality. In Germany, the Wirtschaftsprüferkammer is the supervisory body which oversees compliance with standards and professional duties. It is entitled to sanction the minor violations of duties by auditors. Only more severe types of misbehaviour are dealt with by courts. In Denmark, a special court (Disciplinæernævn) is concerned with disciplinary actions against auditors. The results of this study will demonstrate that audit regulations and in particular disciplinary laws remain basically national, despite efforts to harmonise auditing. This study identifies characteristics of disciplinary systems common to both countries and provides information on the functioning of both systems that may be useful in a number of ways. The results presented might initiate a more systematic comparison of disciplinary systems within member states of the European Union, which would enhance institutional knowledge of the European market for auditing services. This in turn could promote the process of achieving a single European market for auditing services and thus reduce market inefficiencies. [source]


Floodplain agricultural systems: functionality, heritage and conservation

Abstract Floodplain infrastructural features reduce flood risk and have the potential to enhance habitat, biodiversity, water quality and provide societal benefits. Man-made water management systems common in southern England are both flood tolerant and form part of the functional floodplain. Historic floodplain features should be incorporated into agri-environmental policy, as climate change and increasing climate variability makes flood detention areas ever more desirable. Of major importance are floodplain meadows, grazing marshes, water meadows and riparian vegetation, and there is a trend to restore river channels to more natural conditions. This paper describes the operation of historic floodplain water management systems and considers the features associated with canals and mills. The major themes in achieving conservation and restoration goals are presented, and it is demonstrated that a refinement of policies on the ground is required. [source]


Normal and Abnormal Personality Traits: Evidence for Genetic and Environmental Relationships in the Minnesota Study of Twins Reared Apart

ABSTRACT Recent studies have demonstrated substantial correlations between normal and abnormal personality traits. Yet little is known about how these correlations are mediated genetically and environmentally: Do normal and abnormal personality traits stem from the same underlying genes and environments? We addressed this question using data from 128 monozygotic and dizygotic twin pairs in the Minnesota Study of Twins Reared Apart (MISTRA). Additive genetic and nonshared environmental correlations between scales of the Minnesota Multiphasic Personality Inventory (MMPI),an index of abnormal personality,and the Multidimensional Personality Questionnaire (MPQ),an index of normal personality,were estimated. Results indicated that phenotypic correlations between normal and abnormal personality were mediated by genetic as well as environmental factors, although the magnitude of genetic mediation tended to be larger overall. Moreover, the patterns of phenotypic, genetic, and environmental relationships among the scales were similar, suggesting that influences on normal and abnormal personality act through systems common to both. It is suggested that future research focus on the neurogenetic substrates of these shared systems and how dysfunction in these systems influences development of disordered personality. [source]
