Petrounkina Content It is a general property of the intact animal cell to swell rapidly in response to hypo-osmotic conditions. The modified hypo-osmotic swelling test (HOS-test) is an indicative test to evaluate the integrity of the plasma membrane by means of an electronic cell counter, based on the relative increase of the cell volume in response to hypo-osmotic conditions. In this study the relationships between the osmotically induced changes of the cell volume of boar spermatozoa as determined by cell counter and the integrity of the membrane as determined by propidium iodide staining (PI) were studied. Boar sperm cell volume distributions were measured under iso-osmotic (300 mosmolar) conditions and after a hypo-osmotic stress (150 mosmolar). The relative volume shift of mean and modal volume were calculated as a proportion coefficient of modal and mean values of the cell volume distributions by transition from iso-osmotic to hypo-osmotic conditions. The volumetric parameters related to the different cell subpopulations were derived from the different peaks of cell volume distributions. PI-staining techniques were used for comparison. The values of the volume shift and of derived percentages of the osmotically inactive cells were correlated negatively and positively, respectively (p < 0.05) with the percentage of the PI-stained cells. This correlation indicates that a relationship exists between membrane functions of the different cell compartments (sperm head and tail) due to the circumstance that the increase of the cell volume in the HOS-test is associated with the morphological changes in the tail and the PI-staining is associated with the membrane integrity and permeability of the head region. The advantage of computer-assisted volume measurement is that a large number of cells (5000,50 000 spermatozoa) can be measured and evaluated during one procedure and in a very short time. The relative volume shift is a quantitative continuous parameter characterizing the osmotic reactivity and membrane functional competence of a cell population and of subpopulations within one ejaculate. This parameter could be useful to evaluate membrane functional competence rapidly and sensitively. Der auf der relativen Vergrößierungdes Zellvolumens basierende modifizierte hypoosmotischeSchwelltest ist ein indikativer Test zur Beurteilung der Membranintegrität mittels eines elektronischen Partikelzählers. In dieser Studie wurden die Zusammenhänge zwischen der mittels der Propidiumjodid-Färbung bestimmten Zellmembranintegrität und den osmotisch induzierten Veränderungen des Zellvolumens von Eberspermien untersucht. Volumenverteilungen von Eberspermien wurden unter isoosmotischen (300 mosmolar) und hypoosmotischen (150 mosmolar) Bedingungen gemessen. Die relative Volumenverschiebung der modalen und mittleren Werte der Volumenverteilung wurde als Quotient aus Modalwerten der Zellvolumenverteilungen und des mittleren Zellvolumens beim Übergang von isotonen zu hypotonen Bedingungen berechnet. Die auf verschiedene Subpopulationen bezogenen volumetrischen Parameter werden aus den originalen Volumenverteilungen berechnet. Der Betrag der Zellvolumenzunahme und die aus den Volumenverteilungen bestimmten Anteile an Zellen mit beschädigter Geißielmembran korrelierten signifikant negativ bzw. positiv (p < 0,05) mit dem Anteil an den Zellen mit beschädigter Kopfmembran, der sich aus der Propidiumjodid-Färbung ergab. Es wird geschlossen, daßi im Verhalten zwischen den Membranen der verschiedenen Zellkompartimente (Spermienkopf und-Geißiel) ein Zusammenhang besteht. Die beschriebene Methode ermöglicht die Analyse großier Zellpopulationen (5.000,50.000 Zellen). Die relative Volumenverschiebung stellt einen quantitativen kontinuierlichen Parameter dar, der den Membranzustand der Eberspermien einer Spermatozoenpopulation und Subpopulationen innerhalb eines Ejakulates charakterisiert. [source] Concrete canal lining cracking in low to medium plastic soils,IRRIGATION AND DRAINAGE, Issue 2 2002H. Rahimi sol plastique; gonflement; revêtement bétonique; Iran Abstract Failure of concrete irrigation canal linings in the form of cracking, rupture, uplifting and opening of joints, causes loss of water and money in many countries. Following the appearance of extensive cracking of concrete linings in one of the Shoeybieh sugar cane industry farms in the Khoozestan province of Iran, extensive research work was conducted to identify the main causes. In this paper, the final results of the research are presented. The testing program consisted of laboratory as well as field tests, including identification, chemical and mechanical tests of soil samples taken from the borrow pits and canal embankments. Dispersivity tests were performed using pin-hole and chemical methods. Swelling tests were conducted using the ASTM standard and ISSMFE method. The field test includes measurement of deformations of the concrete lining and embankment of an actual lined canal resulting from the filling of the canal with water. The canal was 60 m in length. The deformations were recorded by surveying the elevations of steel bars driven to different depths under the canal lining and at different points in the canal and its embankment sections. The results of laboratory tests showed the soil to have low to medium plasticity with a classification of CL-ML, having less than 1% soluble content, and being nondispersive. Swelling tests conducted by the ASTM and ISSMFE methods showed completely different results. The ISSMFE method resulted in a high free swell potential, while the ASTM method indicated a low to medium potential. The results of full-scale field tests were all in favor of the ISSMFE method. The greatest difference between the results of the two methods was found to be due to the different compaction methods used during construction of the canal, as well as the moisture content of the soil sample being prepared for the swell tests. The flocculent structure and lower compaction water content of soil samples in the ISSMFE method resulted in much higher free swell. The similarity between compaction methods used in the field and the static effort used in the ISSMFE method, as well as very low soil water content of the canal embankments during lining operations, were found to be the main reasons for swelling of the soil and the eventual cracking of the concrete linings. RÉSUMÉ L'écroulement du revêtement bétonique des canaux d'irrigation en forme de fissure, de la rupture, de la sous-pression et de l'ouverture des joints, est la cause de la perte d' eau et par conséquence des fonds publics de plusieurs pays dans le monde entier. Après avoir observé ce phénomène dans des canaux d'irrigation d'un grand projet de canne à sucre à Shoeybieh dans la région de Khozestan, située au sud de l'Iran, une équipe de recherche a été engagée pour trouver les causes principales de cetécroulement. Cet article présente les résultats finaux de cette recherche. Le programme consiste de deux types d'essai, en laboratoire et sur le terrain. Les échantillons du sol pour l'essai mécanique et chimique ont été pris dans des emprunts de terre et la digue du canal. Les essais de dispersivité ont été faits par les méthodes de pin-hole et de chimique. La méthode del ISSMFE et la standard del ASTM ont été utilisés pour les essais de gonflement du sol. Le mesurage de la déformation d'un canal revêté en béton et aussi de digue du canal a été fait par rempliz le canal avee de l'eau. Le canal avait une longueur de 60 m. La déformation a été observée en examinant les élévations à plusieurs points sur la profondeur du canal en dessous de la partie revêtée et aussi la digue du canal par un instrument spécifique. Les résultats des essais de laboratoire montrent que les sols qui ont une plasticité entre petite et moyenne situés dans la classification de CL-ML ayant moins d'un pour cent de contenude soluble sont non-dispersifes. Les essais de gonflement par les méthodes de l'ASTM et l'ISSME montrent des résultats tout à fait différents. La méthode ISSMFE montre un potentiel de gonflement assez élevé, tandis que la méthode ASTM montre un potentiel entre petit et moyen. Les résultats des essais en champs d'étude confirme la méthode ISSMFE. La plus grande différence entre le résultat de deux méthodes est à cause d'usage de plusieurs méthodes de compaction dans la période de la construction du canal. La structure floculée du sol ayant une humidité moins compactée dans la méthode ISSMFE montre un gonflement plus élevée du sol. Les méthodes similaires de compaction utilisées dans le champs d'étude et énergie statique utilisée par la méthode ISSFE, et aussi une petite humidité de digue du canal en période de revêtement sont les causes principales de gonflement et éventuellement de fissure. Bhattacharya Abstract Aim:, To study the relationship of various sperm characteristics and hypo-osmotic swelling test (HOS test) with repeat unexplained early pregnancy loss. Methods:, Semen samples from husbands of 74 couples with a history of repeat early pregnancy loss (group A) were analyzed according to World Health Organization criteria, and a HOS test was performed in each case. Semen samples from 65 husbands with proven fertility (group B) were also studied for comparison. Results:, No statistically significant differences were noted in the age of the husbands, sperm concentration, sperm morphology and percent motile sperm between groups A and B. The mean HOS test scores of the two groups were significantly different (group A: 60.4%; group B: 76.9%; P = 0.01 [normal value: >60%]). In group A, 33.8% of cases (25/74) and in group B, 12.3% of cases (8/65) showed low HOS test scores. Conclusion:, The sperm HOS test may be helpful to screen for any paternal factor associated with repeat embryonic or early fetal loss and in a resource-poor setting, and may be utilized in any clinical laboratory. [source] Influence of clay modification on the reinforcement of vinyl-terminated polydimethylsiloxane networksPOLYMER INTERNATIONAL, Issue 4 2010Eung Soo Kim Abstract A novel method was attempted to reinforce a vinyl-terminated polydimethylsiloxane (PDMS) with two commercially available clays, sodium montmorillonite and Cloisite® 25A. The two clays were functionalized with bis(3-triethoxysilylpropyl)tetrasulfide (TESPT) to prepare Na+MMTS4 and C25AS4, respectively. Incorporation of the tetrasulfide group-containing clays, especially Na+MMTS4, was found to be effective for the enhancement of the interfacial interaction between PDMS and the clays by way of a plausible chemical reaction between the tetrasulfide groups (TSS) and the vinyl-terminated PDMS. Compounding of PDMS with the TESPT-modified clays improved the mechanical properties significantly. In particular, the elongation at break of PDMS/Na+MMTS4 composite was almost twice as high as that of neat PDMS, even if the silicate layers were not fully exfoliated in the PDMS matrix. The tear strength of PDMS was also improved greatly as a result of the incorporation of Na+MMTS4. According to toluene swelling test results, the crosslinking density of the composites was lower than that of neat PDMS, indicating that the improved mechanical properties of the composites arise from enhanced compatibility between the constituents and not from increased crosslinking density. Copyright © 2009 Society of Chemical Industry [source] Effect of N-acetyl-L-cysteine Supplementation in Semen Extenders on Semen Quality and Reactive Oxygen Species of Chilled Canine SpermatozoaREPRODUCTION IN DOMESTIC ANIMALS, Issue 2 2010AJ Michael Contents The objective of this study was to evaluate the quality of chilled dog semen processed with extenders containing various concentrations of N-acetyl-L-cysteine (NAC). Ejaculates from five dogs were collected, pooled and evaluated for concentration, motility, rapid steady forward movement (RSF-movement), viability, acrosomal integrity and by the hypo-osmotic swelling test (HOST). In addition, superoxide anion (O2 -,) production, hydroxyl radicals (OH,) and total reactive oxygen species (tROS) were determined. The pool was divided into five aliquots, which were diluted to a final concentration of 66.66 × 106 spermatozoa/ml with Tris-glucose-egg yolk extender containing one of the following concentrations of NAC (0, 0.5, 1, 2.5 or 5 mm). The semen aliquots were chilled and preserved at 4°C. Semen quality was evaluated after rewarming at 72 h. Sperm motility was significantly higher with the 0.5 mm concentration compared with the control group (p = 0.001). Rapid steady forward movement was higher with the 0.5 and 1 mm concentrations compared with the control and 5 mm group (p < 0.001). Viability and HOST percentages were not significantly altered. Compared with the control, the 5 mm concentration showed significantly reduced percentages of spermatozoa with normal acrosomes (p = 0.049). None of the ROS values at 72 h were significantly affected by the presence of NAC in semen extenders, although all NAC concentrations showed lower O2 -, and OH, values compared with the control. Only the concentrations of 1 and 5 mm inhibited the significant increase of tROS values after 72 h, compared with the fresh semen value. In conclusion, NAC supplementation of semen extenders is beneficial to semen motility of canine spermatozoa during chilling with the 0.5 mm concentration being the most effective, although no significant ROS inhibition was observed at 72 h. Nevertheless, it has limited capacity to preview male fertility. As calorimetry may be used to evaluate directly the metabolic activity of a biological system, the aim of this study was to use microcalorimetry as an additive method for sperm metabolism evaluation of cooled equine semen. Two ejaculates of four stallions were collected and motility, viability (eosin 3%) and membrane functional integrity (hyposmotic swelling test) of spermatozoa were evaluated. Sperm samples were processed following different protocols and the metabolism of these samples was accessed by calorimetry. Centrifugation is part of some of these processing protocols and although this procedure has been deleterious for sperm viability and plasma membrane integrity, no decrease in sperm motility was observed. Microcalorimetry was capable of detecting the positive effect of re-suspending the sperm pellet with Kenney extender. Thus, the use of microcalorimetry offered additional information for equine sperm metabolism evaluation and was efficient in detecting important information from sperm cell metabolism. [source] Intracytoplasmic sperm injection with motile and immotile frozen-thawed testicular spermatozoa (the Hungarian experience)ANDROLOGIA, Issue 1 2005Sz. Mátyás Summary The authors summarize their experience in 75 in vitro fertilization cycles, where frozen-thawed testicular spermatozoa were used for intracytoplasmic sperm injection. In 32 cases, motile spermatozoa could be observed in the frozen-thawed sample. In 34 cases, motility could be induced by pentoxifylline and in nine cases immotile spermatozoa, selected with hypoosmotic swelling test, were used for fertilization. The fertilization rates obtained with motile and immotile spermatozoa (66.1% versus 52.3%) were not significantly different. Our data demonstrate that freezing of testicular spermatozoa opened new possibilities for the treatment of azoospermic men. The clinical pregnancy rate per embryo transfer (ET) (21.87%) was comparable with previous results use of fresh testicular spermatozoa (27.7%). The quality and number of transferred embryos had the most significant impact on the pregnancy rate. The fertilization rate and frequency distribution of good-quality embryos were lower in the case of immotile spermatozoa, and pregnancies were only achieved when motile spermatozoa had been used. [source] Influence of sexual stimulation on sperm parameters in semen samples collected via masturbation from normozoospermic men or cryptozoospermic men participating in an assisted reproduction programmeANDROLOGIA, Issue 3 2000Y. Yamamoto Summary. To evaluate the influence of sexual stimulation via sexually stimulating videotaped visual images (VIM) on sperm function, two semen samples were collected from each of 19 normozoospermic men via masturbation with VIM. Two additional samples were collected from each man via masturbation without VIM. The volume of seminal plasma, total sperm count, sperm motility, percentage of morphologically normal spermatozoa, outcome of hypo-osmotic swelling test and zona-free hamster oocyte sperm penetration assay, and markers of the secretory function of prostate were significantly larger in semen samples collected via masturbation with VIM than masturbation without VIM. The improved sperm parameters in the samples collected via masturbation with VIM may reflect an enhanced prostatic secretory function and increased loading of the vas deferens at that time. In a similar protocol, two semen samples were collected via masturbation with VIM from each of 22 non-obstructed azoospermic men. Semen samples from these men had been occasionally positive in the past for a very small number of spermatozoa (cryptozoospermic men). Two additional samples were collected from each cryptozoospermic man via masturbation without VIM. The volume of seminal plasma, total sperm count, sperm motility, and a marker of the secretory function of prostate were significantly larger in semen samples collected via masturbation with VIM. Fourteen out of the 22 men were negative for spermatozoa in both samples collected via masturbation without VIM. These men demonstrated spermatozoa in both samples collected via masturbation with VIM. Six men with immotile spermatozoa in both samples collected via masturbation without VIM exposed motile spermatozoa in both samples collected via masturbation with VIM. High sexual stimulation during masturbation with VIM results in recovery of spermatozoa of greater fertilizing potential both in normozoospermic and cryptozoospermic men. The appearance of spermatozoa after masturbation with VIM in the vast majority of cryptozoospermic men is of clinical significance in programmes applying intracytoplasmic sperm injections for the management of severe male infertility and obviates the need for testicular biopsy. [source] Do physical forces contribute to cryodamage?BIOTECHNOLOGY & BIOENGINEERING, Issue 4 2009Joseph Saragusty Abstract To achieve the ultimate goal of both cryosurgery and cryopreservation, a thorough understanding of the processes responsible for cell and tissue damage is desired. The general belief is that cells are damaged primarily due to osmotic effects at slow cooling rates and intracellular ice formation at high cooling rates, together termed the "two factor theory." The present study deals with a third, largely ignored component,mechanical damage. Using pooled bull sperm cells as a model and directional freezing in large volumes, samples were frozen in the presence or absence of glass balls of three different diameters: 70,110, 250,500, and 1,000,1,250,µm, as a means of altering the surface area with which the cells come in contact. Post-thaw evaluation included motility at 0,h and after 3,h at 37°C, viability, acrosome integrity, and hypoosmotic swelling test. Interactions among glass balls, sperm cells, and ice crystals were observed by directional freezing cryomicroscopy. Intra-container pressure in relation to volume was also evaluated. The series of studies presented here indicate that the higher the surface area with which the cells come in contact, the greater the damage, possibly because the cells are squeezed between the ice crystals and the surface. We further demonstrate that with a decrease in volume, and thus increase in surface area-to-volume ratio, the intra-container pressure during freezing increases. It is suggested that large volume freezing, given that heat dissipation is solved, will inflict less cryodamage to the cells than the current practice of small volume freezing. Biotechnol. [source] Design and In vitro evaluation of a film-controlled dosage form self-converted from monolithic tablet in gastrointestinal environmentJOURNAL OF PHARMACEUTICAL SCIENCES, Issue 11 2010Tingting Zhang Abstract The purpose of this study is to design an easily manufactured sustained drug delivery system, which can be converted to a film coated system during the dissolution process and then control the drug release according to near zero-order kinetics. Two kinds of pH-sensitive and oppositely charged hydrophilic polymers, chitosan and alginate, were physically mixed as the matrix. Slightly water-soluble drugs such as theophylline, aspirin, and acetaminophen were utilized as model drugs. In vitro drug release and swelling tests were undertaken in simulated gastrointestinal environments. The formation and properties of the film formed during the dissolution process were identified using different techniques. It was demonstrated that formation of the film was based on the interaction of the polymers on tablet surface with the change of system pH. In 0,4,h drug release depended on the intrinsic properties of the polymers, however, characteristics of the film played a leading role in controlling drug release after 4,h. By studying the ratio of relaxation over Fickian diffusion and relationship between tablets swelling and drug release, it was revealed that the film probably modified drug release behavior by limiting polymer erosion.
