    Resistance/susceptibility to Echinococcus multilocularis infection and cytokine profile in humans.

    Differences have been shown between HLA characteristics of patients with different courses of alveolar echinococcosis (AE). Notably the HLA B8, DR3, DQ2 haplotype was associated with more severe forms of this granulomatous parasitic disease. We compared IL-10, IL-5, interferon-gamma (IFN- ,) and tumour necrosis factor (TNF) secretion by peripheral blood mononuclear cells (PBMC) isolated from eight HLA-DR3+, DQ2+, B8+ AE patients and from 10 HLA-DR3,, DQ2,, B8, patients after non-specific mitogenic and specific Echinococcus multilocularis antigenic in vitro stimulation. PBMC from seven HLA-DR3+, DQ2+, B8+ healthy subjects and nine HLA-DR3,, DQ2,, B8, subjects were also studied as controls. PBMC from AE patients with HLA DR3+, DQ2+ haplotype secreted higher levels of IL-10 without any stimulation and after specific antigenic stimulation than did patients without this haplotype. Higher levels of IL-5 and IFN- , were also produced by these patients' PBMC after stimulation with non-purified parasitic antigenic preparations; however, the specific alkaline phosphatase antigen extracted from E. multilocularis induced only Th2-type cytokine secretion. A spontaneous secretion of TNF by HLA DR3+, DQ2+ B8+ AE patients was also found. These results suggest that HLA characteristics of the host can influence immune-mediated mechanisms, and thus the course of AE in humans; specific antigenic components of E. multilocularis could contribute to the preferential Th2-type cytokine production favoured by the genetic background of the host.     THE EFFECTS OF MATING SYSTEM AND GENETIC VARIABILITY ON SUSCEPTIBILITY TO TREMATODE PARASITES IN A FRESHWATER SNAIL, LYMNAEA STAGNALIS

    Abstract The amount and distribution of genetic variability in host populations can have significant effects on the outcome of host-parasite interactions. We studied the effect of mating system and genetic variability on susceptibility of Lymnaea stagnalis snails to trematode parasites. Mating system of snails from eight populations differing in the amount of genetic variability was manipulated, and self- and cross-fertilized offspring were exposed to naturally occurring trematode parasites in a controlled lake experiment. Susceptibility of snails varied between populations, but mating-system treatment did not have a significant effect. Heterozygosity of snails was negatively correlated with the probability of trematode infection, however, suggesting that parasitic diseases may pose a serious threat to populations lacking genetic variability.     RAPID GROWTH RESULTS IN INCREASED SUSCEPTIBILITY TO PREDATION IN MENIDIA MENIDIA

    Abstract Several recent studies have demonstrated that rapid growth early in life leads to decreased physiological performance. Nearly all involved experiments over short time periods (<1 day) to control for potentially confounding effects of size. This approach, however, neglects the benefits an individual accrues by growing. The net effect of growth can only be evaluated over a longer interval in which rapidly growing individuals are allowed the time required to attain the expected benefits of large size. We used two populations of Menidia menidia with disparate intrinsic growth rates to address this issue. We compared growth and survivorship among populations subject to predation in mesocosms under ambient light and temperature conditions for a period of up to 30 days to address two questions: Do the growth rates of fish in these populations respond differently to the presence of predators? Is the previously demonstrated survival cost of growth counterbalanced by the benefits of increased size? We found that growth was insensitive to predation risk: neither population appeared to modify growth rates in response to predation levels. Moreover, the fast-growing population suffered significantly higher mortality throughout the trials despite being 40% larger than the slow-growing population at the experiment's end. These results confirm that the costs of rapid growth extend over prolonged intervals and are not ameliorated merely by the attainment of large size. [source]

    Abstract, The susceptibility of Sogatella furcifera female adults on N22 to malathion increased as feeding time prolonged, while the day change of susceptibility to isoprocarb showed a inverse tendency. The activities of esterase and carboxylesterase were induced significantly after on N22 for one day, and then declined. The non-susceptible level of adults to insecticides and enzyme activities seemed to be heightened after feeding on ASD7, a variety resistant to Nilaparvata lugens. The total phenol content in rice leaf sheath was the highest in N22, and could be regaded as one of the factors, which caused chages in susceptibility of S. furicfera to insecticide after feeding different rice varieties. It was recommended that suitable insecticides and planthopper resistant varieties should be coordinately used in practice 90 that they contribute more effects respectively. [source]


    An F-box gene, CPR30, functions as a negative regulator of the defense response in Arabidopsis

    Summary Arabidopsis gain-of-resistance mutants, which show HR-like lesion formation and SAR-like constitutive defense responses, were used well as tools to unravel the plant defense mechanisms. We have identified a novel mutant, designated constitutive expresser of PR genes 30 (cpr30), that exhibited dwarf morphology, constitutive resistance to the bacterial pathogen Pseudomonas syringae and the dramatic induction of defense-response gene expression. The cpr30 -conferred growth defect morphology and defense responses are dependent on ENHANCED DISEASE SUSCEPTIBILITY 1 (EDS1), PHYTOALEXIN DEFICIENT 4 (PAD4), and NONRACE-SPECIFIC DISEASE RESISTANCE 1 (NDR1). Further studies demonstrated that salicylic acid (SA) could partially account for the cpr30 -conferred constitutive PR1 gene expression, but not for the growth defect, and that the cpr30 -conferred defense responses were NPR1 independent. We observed a widespread expression of CPR30 throughout the plant, and a localization of CPR30-GFP fusion protein in the cytoplasm and nucleus. As an F-box protein, CPR30 could interact with multiple Arabidopsis-SKP1-like (ASK) proteins in vivo. Co-localization of CPR30 and ASK1 or ASK2 was observed in Arabidopsis protoplasts. Based on these results, we conclude that CPR30, a novel negative regulator, regulates both SA-dependent and SA-independent defense signaling, most likely through the ubiquitin-proteasome pathway in Arabidopsis.     A GENETIC VARIANT OF APOLIPOPROTEIN M INCREASES SUSCEPTIBILITY TO CORONARY ARTERY DISEASE IN A CHINESE POPULATION

    SUMMARY 1High-density lipoprotein (HDL) is widely accepted as a lipoprotein that protects against coronary artery and other atherosclerotic diseases. Recently, a new apolipoprotein encoded by the APOM gene, which plays an important role in affecting the intrinsic properties of HDL, has been reported. Genetic variations exist in the APOM gene, but their significance is presently unclear. The aim of the present study was to elucidate whether the APOM T-855C mutant allele is implicated in coronary artery disease (CAD). 2In the present study, 418 patients with CAD and 372 controls were studied, all of whom were Han Chinese from Jiangsu Province, China. Plasma levels of triglycerides (TG), total cholesterol (TC), HDL,cholesterol (HDL-C) and low-density lipoprotein,cholesterol (LDL-C) were evaluated. Genomic DNA from the whole blood from these subjects was subjected to polymerase chain reaction amplification and restriction enzyme digestion to determine genotype with respect to the APOM T-855C polymorphism. 3The allelic frequencies were in Hardy,Weinberg equilibrium. Plasma HDL levels were significantly lower in subjects with CAD than in control subjects (1.08 ± 0.31 vs 1.25 ± 0.32, respectively; P < 0.001) and the distribution of genotypes and allelic frequencies was significantly different in the two groups (P = 0.013 and 0.005, respectively). Multiple logistic regression analysis after adjustment for age, gender, smoking, body mass index, hypertension and serum glucose showed that, compared with the wild-type TT genotype, carriers of the C allele had an increased risk of CAD (odds ratio = 1.819, 95% confidence interval 1.142,2.898; P = 0.012). 4In conclusion, the results of the present study suggest that the APOM T-855C polymorphism carries an increased risk for CAD in this Chinese population.     Activity of telithromycin and seven other agents against 1034 pediatric Streptococcus pneumoniae isolates from ten central and eastern European centers

    Objective, To test the activity of telithromycin against 1034 Streptococcus pneumoniae isolates from pediatric patients in ten centers from ten central and eastern European countries during 2000,2001, and to compare it with the activities of erythromycin A, azithromycin, clarithromycin, clindamycin, and quinupristin,dalfopristin. Methods, The minimum inhibitory concentrations (MICs) of telithromycin, erythromycin A, azithromycin, clarithromycin, clindamycin, levofloxacin, quinupristin,dalfopristin and penicillin G were tested by the agar dilution method with incubation in air, and mechanisms of resistance to macrolides and quinolones were investigated. Results, Strains were isolated from sputum, tracheal aspirates, ear, eye, blood, and cerebrospinal fluid. Among S. pneumoniae strains tested, 36% had raised penicillin G MICs (, 0.12 mg/L). Susceptibilities were as follows: telithromycin, quinupristin,dalfopristin and levofloxacin, , 99%; clindamycin, 83%; and erythromycin A, azithromycin and clarithromycin, 78%. Of 230 (22.3%) erythromycin A-resistant S. pneumoniae strains, 176 (79.6%) had erm(B), 38 (16.1%) had mef(A), and 10 (4.3%) had mutations in 23S ribosomal RNA or in ribosomal protein L4. The rates of drug-resistant S. pneumoniae are high in all centers except Kaunas, Riga, and Prague. Conclusion, Telithromycin had low MICs against all strains, irrespective of macrolide, azalide or clindamycin resistance. Ribosomal methylation was the most prevalent resistance mechanism among all resistant strains, except in Sofia, where the prevalence of the efflux mechanism was higher. [source]


    Susceptibility of Common and Rare Plant Species to the Genetic Consequences of Habitat Fragmentation

To date, most researchers dealing with genetic erosion in fragmented plant populations have focused on threatened or rare species. We raise the question whether common plant species are as susceptible to habitat fragmentation as rare species. We conducted a formal meta-analysis of habitat fragmentation studies that reported both population size and population genetic diversity. We estimated the overall weighted mean and variance of the correlation coefficients among four different measures of genetic diversity and plant population size. We then tested whether rarity, mating system, and plant longevity are potential moderators of the relationship between population size and genetic diversity. Mean gene diversity, percent polymorphic loci, and allelic richness across studies were positively and highly significantly correlated with population size, whereas no significant relationship was found between population size and the inbreeding coefficient. Genetic diversity of self-compatible species was less affected by decreasing population size than that of obligate outcrossing and self-compatible but mainly outcrossing species. Longevity did not affect the population genetic response to fragmentation. Our most important finding, however, was that common species were as, or more, susceptible to the population genetic consequences of habitat fragmentation than rare species, even when historically or naturally rare species were excluded from the analysis. These results are dramatic in that many more plant species than previously assumed may be vulnerable to genetic erosion and loss of genetic diversity as a result of ongoing fragmentation processes. This implies that many fragmented habitats have become unable to support plant populations that are large enough to maintain a mutation-drift balance and that occupied habitat fragments have become too isolated to allow sufficient gene flow to enable replenishment of lost alleles. Resumen:,Las poblaciones pequeñas de plantas son más propensas a la extinción debido a la pérdida de variación genética por medio de la deriva génica aleatoria, el incremento de autogamia y la reproducción entre individuos emparentados. A la fecha, la mayoría de los investigadores que trabajan con erosión genética en poblaciones fragmentadas de plantas se han enfocado en las especies amenazadas o raras. Cuestionamos si las especies de plantas comunes son tan susceptibles a la fragmentación del hábitat como las especies raras. Realizamos un meta análisis formal de estudios de fragmentación que reportaron tanto tamaño poblacional como diversidad genética. Estimamos la media general ponderada y la varianza de los coeficientes de correlación entre cuatro medidas de diversidad genética y de tamaño poblacional de las plantas. Posteriormente probamos si la rareza, el sistema reproductivo y la longevidad de la planta son moderadores potenciales de la relación entre el tamaño poblacional y la diversidad genética.     Susceptibility of a Northern Hardwood Forest to Exotic Earthworm Invasion

Upon invasion, earthworms have been shown to alter the surface soil environment and plant populations and communities. We sought to identify land-use factors in the Ottawa National Forest (ONF), Michigan (U.S.A.), that contribute to earthworm invasion in forest dominated by sugar maple (Acer saccharum Marsh.) so that the susceptibility to additional colonization could be evaluated. We sampled earthworm communities in Sylvania Wilderness Area, a unique old-growth hardwood forest, and nonwilderness sites influenced by recreational fishing, recent timber harvesting, or roads. All the nonwilderness sites contained one to five species of exotic earthworms. In contrast, only 50% of wilderness sites contained exotic earthworms, all of a single species. Nonwilderness sites also had thinner litter and duff layers, higher soil C and N content, and higher nitrogen mineralization potentials than Sylvania sites. Two central differences between Sylvania and nonwilderness sites were that all nonwilderness sites were in close contact with roads and had a history of timber harvest, whereas these factors were not present in Sylvania Wilderness Area. Using average rates of colonization, we constructed two geographic information system models to estimate the percentage of sugar maple on the ONF falling within a theoretical 100-year invasion distance of roads and of second-growth sugar maple as relative indices of susceptibility to invasion. Both models indicated high susceptibility to invasion, with 91.7% and 98.9% of sugar maple habitat falling within a theoretical 100-year invasion distance of roads or historical harvests, respectively. Por encima de la invasión, se ha demostrado que las lombrices alteran el ambiente superficial del suelo, así como a las comunidades y poblaciones de plantas. Tratamos de identificar factores de uso del suelo en el Bosque Nacional Ottawa (BNO), Michigan, E. U. A., que contribuyen a la invasión de lombrices en bosques dominados por arces (Acer saccharum Marsh.), para poder evaluar la susceptibilidad a futuras invasiones. Muestreamos comunidades de lombrices en Sylvania Wilderness Area, un bosque maduro único, y en sitios no silvestres influenciados por pesca deportiva, cosecha reciente de madera o caminos. Todas las áreas no silvestres contenían 1 - 5 especies de lombrices exóticas. En contraste, solo 50% de los sitios silvestres contenían lombrices exóticas, todas de una sola especie. Los sitios no silvestres también tenían capas de hojarasca y de mantillo más delgadas, mayor contenido de C y N del suelo y mayor potencial de mineralización del nitrógeno que los sitios en Sylvania.     Susceptibility and reactivity in polysensitized individuals following controlled induction

    Background: It is uncertain whether polysensitized patients acquire multiple allergies only because of a high degree of exposure to environmental allergens, or because of being highly susceptible to developing contact allergy. Objectives: The aim of this study was to investigate and compare susceptibility and reactivity in polysensitized and monosensitized individuals, and in healthy controls. Patients/methods: We sensitized 66 adult individuals (21 polysensitized, 22 monosensitized, and 23 healthy controls) with diphenylcyclopropenone and assessed challenge responses with visual scoring and ultrasound. We compared sensitization rates using a chi-square test and logistic regression analyses, and calculated linear regression lines of the elicitation responses for each individual. The mean values of the slopes and the intercepts for each group were used to measure the strength of the elicitation response, and were compared using the Mann,Whitney test. Results: Sensitization ratio was equal in the three groups: 57% for the polysensitized, 59% for the monosensitized, and 65% for the healthy control group. There was a lowered elicitation threshold in the polysensitized group compared with that in the monosensitized and healthy control groups and, although not statistically significant, a stronger elicitation response was observed in the polysensitized group. Conclusion: Increased reactivity was found in the polysensitized group, demonstrated by a lowered elicitation threshold, compared with that in the monosensitized and healthy control groups.     Susceptibility of isolated myofibrils to in vitro glutathionylation: Potential relevance to muscle functions,

    Abstract In this study we investigated the molecular mechanism of glutathionylation on isolated human cardiac myofibrils using several pro-glutathionylating agents. Total glutathionylated proteins appeared significantly enhanced with all the pro-oxidants used. The increase was completely reversed by the addition of a reducing agent, demonstrating that glutathione binding occurs by a disulfide and that the process is reversible. A sensitive target of glutathionylation was ,-actin, showing a different reactivity to the several pro-glutathionylating agents by ELISA. Noteworthy, myosin although highly sensitive to the in vitro glutathionylation does not represent the primary glutathionylation target in isolated myofibrils. Light scattering measurements of the glutathionylated ,-actin showed a slower polymerisation compared to the non-glutathionylated protein and force development was depressed after glutathionylation, when the myofibrils were mounted in a force recording apparatus. Interestingly, confocal laser scanning microscopy of cardiac cryosections indicated, for the first time, the constitutive glutathionylation of ,-cardiac actin in human heart. Due to the critical location of ,-actin in the contractile machinery and to its susceptibility to the oxidative modifications, glutathionylation may represent a mechanism for modulating sarcomere assembly and muscle functionality under patho-physiological conditions in vivo. © 2009 Wiley-Liss, Inc.     The acyltransferase gene bus-1 exhibits conserved and specific expression in nematode rectal cells and reveals pathogen-induced cell swelling

    Abstract Susceptibility to the rectal pathogen Microbacterium nematophilum provides a means of examining hindgut differentiation in C. elegans. Mutants of bus - 1 are resistant to infection with this pathogen. We show here that bus - 1 encodes a predicted acyltransferase expressed in rectal epithelial cells (K, F, and U), suggesting its involvement in regional surface modification. bus - 1 reporter genes were used to show spatial regulation by hindgut developmental control genes: egl - 38, mab - 9, and mab - 23. A bus - 1::GFP reporter reveals the conspicuous rectal epithelial swelling induced by M. nematophilum. The C. briggsae ortholog of bus - 1 exhibits conserved function and rectal expression, but it is expressed in vulval as well as rectal cells, correlated with pathogen adhesion to both vulval and rectal cells in this species. Another acyltransferase affecting bacterial adhesion, bus - 18/acl - 10, was also identified, which also shows strong rectal expression, but it is expressed in additional epithelial tissues and is required for general surface integrity.     SUMO4 and its role in type 1 diabetes pathogenesis

    Abstract Susceptibility to type 1 diabetes (T1D) is determined by interactions of multiple genes with unknown environmental factors. Despite the characterization of over 20 susceptibility regions for T1D, identification of specific genes in these regions is still a formidable challenge. In 2004, we first reported the cloning of a novel, small ubiquitin-like modifier (SUMO) gene, SUMO4, in the IDDM5 interval on chromosome 6q25, and presented strong genetic and functional evidence suggesting that SUMO4 is a T1D susceptibility gene. Subsequent studies have consistently confirmed this association in multiple Asian populations despite controversial observations in Caucasians. In this review, we will update the genetic evidence supporting SUMO4 as a T1D susceptibility gene and discuss the possible explanations for the discrepant associations observed in Caucasians.     Abstract The susceptibility of immature stages of the legume flower thrips, Megalurothrips sjostedti, to the entomopathogenic fungus Metarhizium anisopliae, was investigated under laboratory conditions. The adult stage was found to be more susceptible to infection than the larval and pupal stages. Mortality at all stages was dose-dependent, with the highest concentration of 1×108 conidia ml,1 producing the highest mortality (26, 46 and 100% for larvae, pupae and adults, respectively) at 8 days post-inoculation. At the same concentration, daily pollen consumption was significantly reduced at 2 days after treatment in infected adults but more slowly in infected larvae. Fecundity, egg fertility and longevity in adults surviving infection as larvae were significantly reduced compared to the control. [source]


    Effects of an organophosphorous insecticide on survival, fecundity, and development of Hylyphantes graminicola (Sundevall) (Araneae: Linyphiidae)

    Abstract The effects of an organophosphorous insecticide, methamidophos, on fecundity and development of the spider Hylyphantes graminicola (Sundevall) (Araneae: Linyphiidae) were assessed under laboratory conditions. Susceptibility of adults of both sexes to the insecticide and its influence on fecundity of females and development of offspring were investigated. At 48 h after topical application in adults, the median lethal dose (LD50) and 10% lethal dose (LD10) were 0.35 and 0.12 ,g/spider, respectively, for males and 0.52 and 0.16 ,g/spider, respectively, for females. Methamidophos had detrimental effects on fecundity of females; number of eggs per clutch, total egg mass, and clutch size decreased significantly. The hatching rate of eggs from LD10-treated females was slightly higher than the rate in the controls, but the hatching rate of eggs from LD50-treated females was lower than the rate in the controls. However, no significant differences were observed in hatching time and development time across treatments. Development time of spiderlings from LD50-treated females was significantly longer than the time in the controls, and body sizes of the first spiderlings from insecticide-treated females were larger than those in the controls. However, matured offspring were smaller than those in the controls. It was concluded that methamidophos has long-term effects on H. graminicola, and that this may affect the development of spider populations in the field. [source]


    Susceptibility of the leaf-eating beetle, Galerucella calmariensis, a biological control agent for purple loosestrife (Lythrum salcaria), to three mosquito control larvicides

    Abstract We evaluated the susceptibility of Galerucella calmariensis, a species used to control purple loosestrife (Lythrum salicaria), to three mosquito control larvicides. Larvae and adults were fed loosestrife cuttings dipped in Abate® (,375 g · L,1), Altosid® (,250 g · L,1), and Bacillus thuringiensis var israeliensis (Bti) (<110 g · L,1). Eggs on cuttings were dipped in the same concentrations. Pupae were immersed in Abate and Altosid solutions (,474.4 ,g · L,1 and ,1,169.2 ,g · L,1, respectively). Hatching success of eggs dipped in Abate (,3.75 g · L,1) was reduced significantly and survival was significantly lower among larvae and adults eating cuttings dipped in Abate (,0.17 g · L,1 and ,2.27 g · L,1, respectively). Hatching success of eggs dipped in Altosid (,2.52 g · L,1) was reduced significantly. With exposure to Altosid, larval survival to pupation and adult emergence was reduced significantly at concentrations of ,2.92 g · L,1 and ,0.63 g · L,1, respectively. Altosid (,0.23 g · L,1) also delayed the onset of pupation and adult emergence among larvae that survived to pupate. Larvae that survived with exposure to Altosid (,1.72 g · L,1) grew to 70% larger than those exposed to lower concentrations. Pupal survival was unaffected with exposure to Abate and Altosid and adult survival was unaffected with exposure to Altosid. Bacillus thuringiensis var israeliensis did not adversely affect any life stage of G. calmariensis. The mean Abate concentration on cuttings exposed to operational spraying was in the range that reduced egg hatchability and adult survival but was higher than concentrations that caused complete mortality of larvae. The mean Altosid concentration on cuttings exposed to operational spraying was in the range that reduced hatching success in eggs and delayed pupation and adult emergence of larvae. [source]


    Susceptibility of black fly larvae (Diptera: Simuliidae) to lawn-care insecticides individually and as mixtures

    Abstract Urban and suburban watersheds have the potential to be highly impacted by chemicals, especially insecticides to control insect pests on lawns, ornamental plants, and home gardens. Three of the most common lawn-care insecticides detected in urban watersheds, carbaryl, chlorpyrifos, and malathion, have been evaluated using an acute orbital shaker toxicity test to determine their respective concentrations that produce 50% mortality (LC50) in Simulium vittatum Zetterstedt cytospecies IS-7 larvae. Results of the 48-h LC50 tests show chlorpyrifos to be the most toxic to black fly larvae (LC50 = 0.28 ,g/L) followed by carbaryl (LC50 = 23.72 ,g/L) and malathion (LC50 = 54.20 ,g/L). These insecticides were also tested as binary and ternary mixtures using the toxic unit (TU) approach. Toxicity was shown to be greater than additive for the ternary mixture of chlorpyrifos,carbaryl,malathion (LC50 = 0.56 TU) and the binary mixtures of chlorpyrifos,malathion (LC50 = 0.72 TU) and carbaryl-malathion (LC50 = 0.78 TU). The binary combination of chlorpyrifos and carbaryl was shown to be additive (LC50 = 0.98 TU). These results indicate that aquatic invertebrate populations in urban and suburban streams may experience a higher-than-expected increase in toxicity-related effects when all three chemicals are present in the waterway.     Mice Carrying the Szt1 Mutation Exhibit Increased Seizure Susceptibility and Altered Sensitivity to Compounds Acting at the M-Channel

    Summary:,Purpose: Mutations in the genes that encode subunits of the M-type K+ channel (KCNQ2/KCNQ3) and nicotinic acetylcholine receptor (CHRNA4) cause epilepsy in humans. The purpose of this study was to examine the effects of the Szt1 mutation, which not only deletes most of the C-terminus of mouse Kcnq2, but also renders the Chnra4 and Arfgap-1 genes hemizygous, on seizure susceptibility and sensitivity to drugs that target the M-type K+ channel. Methods: The proconvulsant effects of the M-channel blocker linopirdine (LPD) and anticonvulsant effects of the M-channel enhancer retigabine (RGB) were assessed by electroconvulsive threshold (ECT) testing in C57BL/6J- Szt1/+ (Szt1) and littermate control C57BL/6J+/+ (B6) mice. The effects of the Szt1 mutation on minimal clonic, minimal tonic hindlimb extension, and partial psychomotor seizures were evaluated by varying stimulation intensity and frequency. Results:Szt1 mouse seizure thresholds were significantly reduced relative to B6 littermates in the minimal clonic, minimal tonic hindlimb extension, and partial psychomotor seizure models. Mice were injected with LPD and RGB and subjected to ECT testing. In the minimal clonic seizure model, Szt1 mice were significantly more sensitive to LPD than were B6 mice [median effective dose (ED50) = 3.4 ± 1.1 mg/kg and 7.6 ± 1.0 mg/kg, respectively]; in the partial psychomotor seizure model, Szt1 mice were significantly less sensitive to RGB than were B6 mice (ED50= 11.6 ± 1.4 mg/kg and 3.4 ± 1.3 mg/kg, respectively). Conclusions: These results suggest that the Szt1 mutation alters baseline seizure susceptibility and pharmacosensitivity in a naturally occurring mouse model. [source]


    Long-term Changes in Postnatal Susceptibility to Pilocarpine-induced Seizures in Rats Exposed to Gamma Radiation at Different Stages of Prenatal Development

    Summary:,Purpose: To determine whether brains irradiated at different stages of prenatal development also have different postnatal susceptibility to seizures evoked by pilocarpine. Methods: Pregnant Wistar rats were exposed to a single 1.0-Gy dose of gamma rays on gestation days 13, 15, 17, or 19 (E13, E15, E17, and E19, respectively). On postnatal day 60, their offspring received i.p. pilocarpine injections to evoke status epilepticus. Behavior of the animals was observed continuously for 6 h after the injection, and motor manifestations of seizure activity were rated, and survival times recorded. After 7-day survival, the animals were killed, and their brains were weighed. Results: The average brain weight of animals exposed to irradiation at earlier prenatal stages (E13 or E15) was significantly lower than that after irradiation on E17 or E19. However, effects of the irradiation on the susceptibility to pilocarpine-induced seizures were quite opposite. The intensity of status epilepticus evoked in rats irradiated on E13 or E15 was significantly lower than that in nonirradiated controls or in those irradiated on E17 or E19. Moreover, after irradiation on E13 or E15, survival of the animals was significantly higher in relation not only to other irradiated groups but also to the controls. Conclusions: The results suggest than the extent of neuronal deficit, even if relatively greater, cannot always lead to higher susceptibility of the dysplastic brain to seizures. Functional consequences of the deficit, even if its magnitude is relatively smaller but involving specific brain areas, appear to be critical for the epileptogenesis. [source]


    A Study of 43 Patients with Panayiotopoulos Syndrome, a Common and Benign Childhood Seizure Susceptibility

    Summary: ,Purpose: To determine prevalence, clinical, EEG features, and prognosis of Panayiotopoulos syndrome and to examine the proposition that clinical manifestations are more important than EEG findings. Methods: We analyzed retrospectively the clinical and EEG records of 1,340 children with one or more focal seizures seen in the last 18 years, supplemented with a prospective study from 1998. Panayiotopoulos syndrome was defined by clinical criteria, mainly ictal emesis, irrespective of EEG findings. Results: We analyzed 43 of 90 patients with Panayiotopoulos syndrome who were seizure free >2 years. Girls predominated. Mean age at first seizure was 5 years. Seizures consisted mainly of autonomic manifestations; ictal emesis was often the first symptom, culminating in vomiting in 86%. Of nonautonomic manifestations, lateral eye deviation was the most common; visual symptoms were exceptional. Impairment of consciousness ensued in all seizures, half of which ended with hemi or generalized convulsions. Nearly 46.5% of cases had at least one seizure >30 min, constituting autonomic status epilepticus. Seizures during sleep (84%) were more common than those in wakefulness. EEG showed occipital spikes in 29 patients. Of the other 14 cases, five had extraoccipital abnormalities or brief generalized discharges, and nine had normal awake and sleep EEG. Prognosis was excellent. All 43 children have been free of seizures for ,2 years, 53% having a single seizure, and 47%, an average two to three seizures. Conclusions: Panayiotopoulos syndrome is common and needs wider recognition. EEG shows occipital or extraoccipital abnormalities, is normal in one third of patients, and does not determine clinical manifestations or prognosis, which is excellent despite the high prevalence of lengthy seizures.     Caloric Restriction Inhibits Seizure Susceptibility in Epileptic EL Mice by Reducing Blood Glucose

    Summary: ,Purpose: Caloric restriction (CR) involves underfeeding and has long been recognized as a dietary therapy that improves health and increases longevity. In contrast to severe fasting or starvation, CR reduces total food intake without causing nutritional deficiencies. Although fasting has been recognized as an effective antiseizure therapy since the time of the ancient Greeks, the mechanism by which fasting inhibits seizures remains obscure. The influence of CR on seizure susceptibility was investigated at both juvenile (30 days) and adult (70 days) ages in the EL mouse, a genetic model of multifactorial idiopathic epilepsy. Methods: The juvenile EL mice were separated into two groups and fed standard lab chow either ad libitum (control, n = 18) or with a 15% CR diet (treated, n = 17). The adult EL mice were separated into three groups; control (n = 15), 15% CR (n = 6), and 30% CR (n = 3). Body weights, seizure susceptibility, and the levels of blood glucose and ketones (,-hydroxybutyrate) were measured over a 10-week treatment period. Simple linear regression and multiple logistic regression were used to analyze the relations among seizures, glucose, and ketones. Results: CR delayed the onset and reduced the incidence of seizures at both juvenile and adult ages and was devoid of adverse side effects. Furthermore, mild CR (15%) had a greater antiepileptogenic effect than the well-established high-fat ketogenic diet in the juvenile mice. The CR-induced changes in blood glucose levels were predictive of both blood ketone levels and seizure susceptibility. Conclusions: We propose that CR may reduce seizure susceptibility in EL mice by reducing brain glycolytic energy. Our preclinical findings suggest that CR may be an effective antiseizure dietary therapy for human seizure disorders. [source]

    Abstract The amount and distribution of genetic variability in host populations can have significant effects on the outcome of host-parasite interactions. We studied the effect of mating system and genetic variability on susceptibility of Lymnaea stagnalis snails to trematode parasites. Mating system of snails from eight populations differing in the amount of genetic variability was manipulated, and self- and cross-fertilized offspring were exposed to naturally occurring trematode parasites in a controlled lake experiment. Susceptibility of snails varied between populations, but mating-system treatment did not have a significant effect. Heterozygosity of snails was negatively correlated with the probability of trematode infection, however, suggesting that parasitic diseases may pose a serious threat to populations lacking genetic variability.     Reduced FAS transcription in clones of U937 cells that have acquired resistance to Fas-induced apoptosis

    Susceptibility to cell death is a prerequisite for the elimination of tumour cells by cytotoxic immune cells, chemotherapy or irradiation. Activation of the death receptor Fas is critical for the regulation of immune cell homeostasis and efficient killing of tumour cells by apoptosis. To define the molecular changes that occur during selection for insensitivity to Fas-induced apoptosis, a resistant variant of the U937 cell line was established. Individual resistant clones were isolated and characterized. The most frequently observed defect in the resistant cells was reduced Fas expression, which correlated with decreased FAS transcription. Clones with such reduced Fas expression also displayed partial cross-resistance to tumour necrosis factor-, stimulation, but the mRNA expression of tumour necrosis factor receptors was not decreased. Reintroduction of Fas conferred susceptibility to Fas but not to tumour necrosis factor-, stimulation, suggesting that several alterations could be present in the clones. The reduced Fas expression could not be explained by mutations in the FAS coding sequence or promoter region, or by silencing through methylations. Protein kinase B and extracellular signal-regulated kinase, components of signalling pathways downstream of Ras, were shown to be activated in some of the resistant clones, but none of the three RAS genes was mutated, and experiments using chemical inhibitors could not establish that the activation of these proteins was the cause of Fas resistance as described in other systems. Taken together, the data illustrate that Fas resistance can be caused by reduced Fas expression, which is a result of an unidentified mode of regulation. [source]


    Polarizability, Susceptibility, and Dielectric Constant of Nanometer-Scale Molecular Films: A Microscopic View

    Abstract The size-dependence of the polarizability, susceptibility, and dielectric constant of nanometer-scale molecular layers is explored theoretically. First-principles calculations based on density functional theory are compared to phenomenological modeling based on polarizable dipolar arrays for a model system of organized monolayers composed of oligophenyl chains. Size trends for all three quantities are primarily governed by a competition between out-of-plane polarization enhancement and in-plane polarization suppression. Molecular packing density is the single most important factor controlling this competition and it strongly affects the bulk limit of the dielectric constant as well as the rate at which it is approached. Finally, the polarization does not reach its "bulk" limit, as determined from the Clausius,Mossotti model, but the susceptibility and dielectric constant do converge to the correct bulk limit. However, whereas the Clausius,Mossotti model describes the dielectric constant well at low lateral densities, finite size effects of the monomer units cause it to be increasingly inaccurate at high lateral densities.     Susceptibility to Heterobasidion parviporum in Picea abies clones grown in different environments

    Summary Thirty-five Norway spruce, Picea abies, clones from Sweden were tested for resistance to Heterobasidion parviporum. Rooted cuttings of the clones were planted in Italy and Greece and cultivated for two growing seasons before inoculation with H. parviporum. Extent of infection was determined 6 weeks later. The results were compared to those of earlier inoculations in Sweden. Plant growth traits were under strong genetic control in all locations with broad sense heritability estimates between 0.14 and 0.54. Lesion and fungal extension heritabilities were moderate, H2 ranged from 0.09 to 0.20, and exhibited rather large genetic variation. There was significant genotypic correlation between Italy and Greece with respect to both lesions and fungal extension. No such correlations were found between Sweden and the two other countries. The lack of repeatability in testing susceptibility is unsatisfactory. It could be explained by C-effects associated with propagation of the host plants. Height growth correlated significantly and positively among all countries. [source]


    Finnish Cronartium ribicola does not infect alternate hosts of Cronartium flaccidum

    Summary Susceptibility of the main alternate hosts of Cronartium flaccidum, Vincetoxicum spp. and Melampyrum spp., to C. ribicola was investigated by artificial inoculations on detached leaves in the laboratory and on whole plants in the greenhouse. Neither uredinia nor telia developed on the leaves of either Vincetoxicum hirundinaria, Vincetoxicum nigrum, Melampyrum sylvaticum, Melampyrum pratense, Melampyrum nemorosum, Melampyrum arvense, Melampyrum cristatum or Melampyrum polonicum following inoculation by the 26 aeciospore sources collected from Pinus strobus, Pinus monticola, Pinus flexilis and Pinus peuce in 2000,2003. The results suggest that Finnish C. ribicola does not infect the main alternate hosts of C. flaccidum. Cronartium ribicola and C. flaccidum can thus be identified reliably by inoculations on their main alternate hosts.     Susceptibility of cones and seeds to fungal infection in a pine (Pinus spp.) collection

    Summary Thirty-one fungi, members of Ascomycota and Deuteromycota, were isolated from cones/seeds of 28 hosts (Pinus spp.) originating from East Asia, Europe and North America, and growing at the Montreal Botanical Garden, Canada. A total of 21 taxa of these isolated fungi are considered pathogens. The damage severity was most prevalent on Diploxylon pines of European provenance (Pinus nigra, Pinus mugo and Pinus sylvestris), and one indigenous North American species (Pinus albicaulis). Twelve fungi occurred on different hosts, i.e. Sphaeropsis sapinea (12), Herpotrichia juniperi (8), Truncatella hartigii (7), Ceuthospora sp. (5), Fusarium, spp. (5), Pestalotiopsis funerea (3), Phomopsis sp. (3), Valsa spp. (anamorph. Cytospora) (3), Diaporthe sp. (3), Fusicoccum sp. (2) and Sirococcus strobilinus (2). Sphaeropsis sapinea, Herpotrichia juniperi, Phomopsis conorum, Truncatella hartigii, Tubercularia sp. and Valsa spp. were related to high cone and seed damage. For the most frequently observed Sphaeropsis sapinea damage, the subgenus Haploxylon was less susceptible than Diploxylon, and P. sylvestris less than P. nigra and P. mugo. Pinus resinosa showed more tolerance to S. sapinea. Truncatella hartigii was more frequently associated with necrotic lesions on cones/seeds in comparison with Pestalotiopsis funerea. The results are discussed in the context of arboretum, seed orchard, nursery and quarantine management. [source]


    Parasites can cause selection against migrants following dispersal between environments

    Summary 1.,The potential for selection against migrants to promote population divergence and speciation is well established in theory, yet there has been relatively little empirical work that has explicitly considered selection against migrants as a form of reproductive barrier, and its importance in the accumulation of reproductive isolation between populations has been overlooked until recently. 2.,Parasites often differ between environments and can be an important source of selection on hosts, yet their contribution to population divergence in general, and selection against migrants in particular, is poorly understood. 3.,Selection against migrants might be reduced if organisms escape parasitism when they disperse (natural enemy release). Alternatively, parasites could increase selection against migrants if, when they disperse, organisms encounter parasites to which they are poorly adapted. 4.,Here we test experimentally the contribution that parasites could make to selection against migrants in the adaptive radiation of three-spined sticklebacks. These fish have repeatedly colonized freshwater environments from marine ones, and this has repeatedly lead to rapid speciation. 5.,We use transplant experiments of lab-raised fish to simulate dispersal, and antihelminthic treatment to show that ancestral-type marine sticklebacks contract higher burdens of novel parasites when introduced to freshwater, than in saltwater, and suffer a growth cost as a direct result. 6.,Susceptibility to parasites and their detrimental effect is less in derived, freshwater fish from evolutionarily young populations, possibly as a result of selection for resistance. 7.,Our results support a role for parasites in selection against migrants and population diversification.     Antimicrobial Susceptibility of Helicobacter pylori Strains in a Random Adult Swedish Population

    Abstract Background and Aim:, Antimicrobial resistance in Helicobacter pylori is a growing problem and has become an important factor leading to eradication failure. Information on antimicrobial susceptibility is important for selection of an optimum treatment regimen. The resistance rate in a random population has not been studied previously. Methods:, A random Swedish population sample (n = 3000, age 20,81 years) was surveyed using a mailed validated questionnaire assessing gastrointestinal symptoms (response rate of 74%). One-third of the responders was invited, in random order, and accepted an esophagogastroduodenoscopy with biopsies for H. pylori culture and histology. Subjects were not treated for their H. pylori infection but a minimum inhibitory concentration of metronidazole, clarithromycin, amoxicillin, and tetracycline for the H. pylori isolates (n = 333) was determined by agar dilution. Prescribed antibiotic in the area was recorded. Results:, Irrespective of symptomatology, 16.2% of the isolated H. pylori strains were resistant to metronidazole, 1.5% to clarithromycin, 0% to amoxicillin, and 0.3% to tetracycline. The antibiotic consumption was low from an international perspective.
