Effects of climate change on labile and structural carbon in Douglas-fir needles as estimated by ,13C and Carea measurements

Abstract Models of photosynthesis, respiration, and export predict that foliar labile carbon (C) should increase with elevated CO2 but decrease with elevated temperature. Sugars, starch, and protein can be compared between treatments, but these compounds make up only a fraction of the total labile pool. Moreover, it is difficult to assess the turnover of labile carbon between years for evergreen foliage. Here, we combined changes in foliar Carea (C concentration on an areal basis) as needles aged with changes in foliar isotopic composition (,13C) caused by inputs of 13C-depleted CO2 to estimate labile and structural C in needles of different ages in a four-year, closed-chamber mesocosm experiment in which Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) seedlings were exposed to elevated temperature (ambient + 3.5 °C) and CO2 (ambient + 179 ppm). Declines in ,13C of needle cohorts as they aged indicated incorporation of newly fixed labile or structural carbon. The ,13C calculations showed that new C was 41 ± 2% and 28 ± 3% of total needle carbon in second- and third-year needles, respectively, with higher proportions of new C in elevated than ambient CO2 chambers (e.g. 42 ± 2% vs. 37 ± 6%, respectively, for second-year needles). Relative to ambient CO2, elevated CO2 increased labile C in both first- and second-year needles. Relative to ambient temperature, elevated temperature diminished labile C in second-year needles but not in first-year needles, perhaps because of differences in sink strength between the two needle age classes. We hypothesize that plant-soil feedbacks on nitrogen supply contributed to higher photosynthetic rates under elevated temperatures that partly compensated for higher turnover rates of labile C. Strong positive correlations between labile C and sugar concentrations suggested that labile C was primarily determined by carbohydrates. Labile C was negatively correlated with concentrations of cellulose and protein. Elevated temperature increased foliar %C, possibly due to a shift of labile constituents from low %C carbohydrates to relatively high %C protein. Decreased sugar concentrations and increased nitrogen concentrations with elevated temperature were consistent with this explanation. Because foliar constituents that vary in isotopic signature also vary in concentrations with leaf age or environmental conditions, inferences of ci/ca values from ,13C of bulk leaf tissue should be done cautiously. Tracing of 13C through foliar carbon pools may provide new insight into foliar C constituents and turnover. [source]


United Kingdom and Ireland precipitation variability and the North Atlantic sea-level pressure field

Abstract The relationship between UK and Ireland (UK&I) precipitation variability and the North Atlantic sea-level pressure (SLP) field is examined. Strong positive correlations between the North Atlantic Oscillation (NAO) and precipitation in the northwest of the UK&I, particularly in winter, are confirmed but correlations are insignificant at the 0.05 level in the southeast during all months. This paper identifies new patterns of SLP associated with precipitation variability both for regions and months where precipitation variability is not strongly linked with the NAO and for patterns that appear to be more closely related to UK&I precipitation than the NAO. Two indices of monthly UK&I precipitation variability are calculated using empirical orthogonal functions (EOFs) of monthly UK&I precipitation anomalies. EOF1 represents precipitation variability for the UK&I as a whole and EOF2 the variability in the north,south precipitation gradient across the UK&I. Correlations between both these monthly EOF derived precipitation indices and SLP show a north,south (sub-tropical/mid-latitude) dipole, which is particularly strong in winter. These correlation patterns are then used to construct new SLP indices, which necessarily relate more closely to UK&I precipitation. The first index resembles the East Atlantic pattern from September to April. The second may be thought of as an alternative index of the NAO, such that it is optimized with respect to precipitation variability and is located northeast of those centres of action most commonly used to calculate the NAO index. Stepwise linear regression models, incorporating the two new indices and the original NAOI, suggest that over 25% of UK&I precipitation variability this century (1900,1994) in each month can be explained by a simple index representation of the North Atlantic SLP field. Sex pheromone for monitoring flight periods and population densities of the pine sawfly, Diprion jingyuanensis Xiao et Zhang (Hym., Diprionidae)

Abstract:, The pine sawfly, Diprion jingyuanensis Xiao et Zhang, is a serious pest of Chinese pine (Pinus tabulaeformis Carr.) in the northern parts of China. The general biology of this recently described sawfly species is little known and in the present study we used a synthetic pheromone to monitor its flight period and to evaluate the possibility to use pheromone traps as a tool for estimating and predicting population densities. The attractant pheromone of D. jingyuanensis is (1S,2R,6R)-1,2,6-trimethyldodecyl propionate and in this study we used a four-isomer threo-mixture containing the active attractant. Both doses tested, 1 and 2 mg/trap, revealed the same seasonal flight pattern, the higher dosage demonstrating more clear flight peaks. The first flight peak of D. jingyuanensis occurred in mid-June during all 3 years, 1997,1999, and in 1997 and 1998 a second flight peak was also recorded in mid-July. The flight period was similar in time for populations located at higher (1400 m) and lower altitude (1100 m), from early June to late July or early August. Temperature was the main weather factor-affecting trap catches. Diurnal flight activity began at 9.00, peaked at 13.00 and lasted until 20.00. A series of tests with traps in different positions within stands and trees were conducted and the results demonstrated the importance of standardizing the trapping protocol in population monitoring studies. By using traps with 2 mg baits it is possible to detect sawfly occurrence at very low population densities, not detectable by other means. Strong positive correlations between trap catch and relative population density were found in 2000 and 2001, but not in 1998, when only few sites were monitored and the population was in a decreasing phase. The results are promising for future large-scale use of pheromone-based monitoring of D. jingyuanensis. [source]


Lipid peroxidation levels and total oxidant status in serum, saliva and gingival crevicular fluid in patients with chronic periodontitis

Abstract Objectives: Increased levels of reactive oxygen species lead to oxidative stress. Recent data suggest increased lipid peroxidation (LPO) levels and oxidative stress in periodontitis. Malondialdehyde (MDA), a significant LPO product, increases in oxidative stress. In this study, MDA levels and total oxidant status (TOS) in serum, saliva and gingival crevicular fluid (GCF) were investigated in patients with chronic periodontitis (CP). Materials and Methods: Thirty-six CP patients and 28 periodontally healthy controls were included in the study. Following clinical measurements and samplings, MDA and TOS levels were measured by high-performance liquid chromatography and a novel automatic colorimetric method, respectively. Results: While the saliva and GCF MDA levels, and serum, saliva and GCF TOS values were significantly higher in the CP group than the control group (p<0.05), no significant difference in serum MDA levels was found (p>0.05). Strong positive correlations were observed between periodontal parameters and MDA and TOS levels (p<0.05). Conclusions: The results revealed that LPO significantly increased locally in the periodontal pocket/oral environment, while TOS displayed both systemic and local increases in periodontitis. The findings suggest that increased LPO and TOS may play an important role in the pathology of periodontitis, and are closely related to the clinical periodontal status. [source]


Myopodia (postsynaptic filopodia) participate in synaptic target recognition

Abstract Synaptic partner cells recognize one another by utilizing a variety of molecular cues. Prior to neuromuscular synapse formation, Drosophila embryonic muscles extend dynamic actin-based filopodia called "myopodia." In wild-type animals, myopodia are initially extended randomly from the muscle surface but become gradually restricted to the site of motoneuron innervation, a spatial redistribution we call "clustering." Previous experiments with prospero mutant embryos demonstrated that myopodia clustering does not occur in the absence of motoneuron outgrowth into the muscle field. However, whether myopodia clustering is due to a general signal from passing axons or is a result of the specific interactions between synaptic partners remained to be investigated. Here, we have examined the relationship of myopodia to the specific events of synaptic target recognition, the stable adhesion of synaptic partners. We manipulated the embryonic expression of ,PS2 integrin and Toll, molecules known to affect synaptic development, to specifically alter synaptic targeting on identified muscles. Then, we used a vital single-cell labeling approach to visualize the behavior of myopodia in these animals. We demonstrate a strong positive correlation between myopodia activity and synaptic target recognition. The frequency of myopodia clustering is lowered in cases where synaptic targeting is disrupted. Myopodia clustering seems to result from the adherence of a subset of myopodia to the innervating growth cone while the rest are eliminated. The development of gaze following and its relation to language

We examined the ontogeny of gaze following by testing infants at 9, 10 and 11 months of age. Infants (N = 96) watched as an adult turned her head toward a target with either open or closed eyes. The 10- and 11-month-olds followed adult turns significantly more often in the open-eyes than the closed-eyes condition, but the 9-month-olds did not respond differentially. Although 9-month-olds may view others as ,body orienters', older infants begin to register whether others are ,visually connected' to the external world and, hence, understand adult looking in a new way. Results also showed a strong positive correlation between gaze-following behavior at 10,11 months and subsequent language scores at 18 months. Implications for social cognition are discussed in light of the developmental shift in gaze following between 9 and 11 months of age. [source]


Evidence for distinct effects of GH and IGF-I in the metabolic syndrome

Abstract Aims, The metabolic syndrome is a cluster of cardiovascular risk factors which include central obesity, dyslipidaemia, glucose intolerance and hypertension. These risk factors are common in patients with growth hormone (GH) deficiency, suggesting a role for the somatotropic axis in the development of metabolic syndrome. Methods, We used factor analysis to investigate the relationships linking serum levels of GH and insulin-like growth factor I (IGF-I) to metabolic syndrome variables (high-density lipoprotein cholesterol, triglycerides, fasting glucose, blood pressure and waist circumference). We studied 359 men and 388 women from the Data from an Epidemiological Study on the Insulin Resistance syndrome (DESIR). Their age range was 30,64 years. Results, Three independent latent factors explained 61% of the total variance in women and four factors explained 73% in men. In both men and women, IGF-I showed a strong positive correlation with the lipid factor and a negative correlation with the obesity/glucose factor. In women, GH showed a strong negative correlation with the obesity/glucose factor but not the lipid factor. In men, GH was unrelated to the lipid and obesity/glucose factors. The blood pressure factor was not related to GH or IGF-I. In contrast with IGF-I, GH was significantly lower in women with metabolic syndrome (1575 ± 449 pg/ml) than in the other women (2121 ± 520 pg/ml, P = 0.002). No significant difference was observed in men for GH or IGF-I. Conclusion, Our results support a link between the somatotropic axis and several features of the metabolic syndrome, and suggest distinct effects of GH and IGF-I on these parameters. [source]


The evolution of floral scent and insect chemical communication

Ecology Letters (2010) 13: 643,656 Abstract Plants have evolved a range of strategies to manipulate the behaviour of their insect partners. One powerful strategy is to produce signals that already have a role in the animals' own communication systems. To investigate to what extent the evolution of floral scents is correlated with chemical communication in insects, I analyse the occurrence, commonness, and evolutionary patterns of the 71 most common ,floral' volatile organic compounds (VOCs) in 96 plant families and 87 insect families. I found an overlap of 87% in VOCs produced by plants and insects. ,Floral' monoterpenes showed strong positive correlation in commonness between plants (both gymnosperms and angiosperms) and herbivores, whereas the commonness of ,floral' aromatics was positively correlated between angiosperms and both pollinators and herbivores. According to a multivariate regression analysis the commonness of ,floral' aromatics was best explained by their commonness in pollinators, whereas monoterpenes were best explained by herbivores. Among pollinator orders, aromatics were significantly more common in Lepidoptera than in Hymenoptera, whereas monoterpenes showed no difference among the two orders. Collectively, these patterns suggest that plants and insects converge in overall patterns of volatile production, both for attraction and defence. Monoterpenes seem to have evolved primarily for defence under selection by herbivores, whereas aromatics evolved signalling functions in angiosperms, primarily for pollinator attraction. [source]


Environmental determinants correlated to Vibrio harveyi -mediated death of marine gastropods

Summary Vibrio harveyi is an emerging pathogen that causes mass mortality in a wide variety of marine animal species; however, it is still unclear which environmental determinants correlate V. harveyi dynamics and the bacterium-mediated death of marine animal life. We conducted a correlation analysis over a 5-year period (2003,2007) analysing the following data: V. harveyi abundance, marine animal mortality and environmental variables (seawater temperature, salinity, pH, chlorophyll a, rainfall and total viable bacterial counts). The samples were collected from a coastal area in northern Japan, where deaths of a marine gastropod species (Haliotis discus hannai) have been reported. Our analysis revealed significant positive correlations between average seawater temperature and average V. harveyi abundance (R = 0.955; P < 0.05), and between average seawater temperature and V. harveyi -mediated abalone death (R = 0.931; P < 0.05). Based on the regression model, n°C rise in seawater temperature gave rise to a 21n -fold increase in the risk of mortality caused by V. harveyi infection. This is the first report providing evidence of the strong positive correlation between seawater temperature and V. harveyi -mediated death of marine species. [source]


Fluctuations in European eel (Anguilla anguilla) recruitment resulting from environmental changes in the Sargasso Sea

Abstract European eel decline is now widely observed and involves a large number of factors such as overfishing, pollution, habitat loss, dam construction, river obstruction, parasitism and environmental changes. In the present study, we analyzed the influence of environmental conditions in the Sargasso Sea and Atlantic ocean circulation on European glass eel recruitment success. Over a recent 11-yr period, we showed a strong positive correlation between an original index of glass eel recruitment and primary production (PP) in eel spawning area. Moreover, PP was negatively correlated with temperature in the Sargasso Sea. Therefore, we used sea temperature as an inverse proxy of marine production. A close negative relationship has been found over the last four decades between long-term fluctuations in recruitment and in sea temperature. These findings were reinforced by the detection of a regime shift in sea temperature that preceded the start of the decline in glass eel recruitment in the early 1980s. By contrast, variations in integrative indices measuring ocean circulation, i.e. latitude and strength of the Gulf Stream, did not seem to explain variations in glass eel recruitment. Our results support the hypothesis of a strong bottom-up control of leptocephali survival and growth by PP in the Sargasso Sea on short and long time scales. We argue that sea warming in the eel spawning area since the early 1980s has modified marine production and eventually affected the survival rate of European eels at early life stages. [source]


Lake age and water level affect the turbidity of floodplain lakes along the lower Rhine

SUMMARY 1. We sampled a set of 93 lakes situated in the floodplains of the lower River Rhine in search for morphometric and other factors that explain their variation in clarity. 2. Lakes with a drop in summer water level were less turbid at the time of sampling, mainly because of a lower concentration of inorganic suspended solids (ISS). 3. We also found that older lakes were more turbid than younger lakes and that this was largely because of an increase in phytoplankton. 4. Water clarity was positively related to lake depth and the presence of vegetation. 5. Model calculations indicated that the underwater light climate was strongly affected by chlorophyll and ISS, the latter being the dominant factor affecting Secchi depth. Dissolved organic carbon (DOC) was less important. 6. The high concentration of ISS suggests that intensive resuspension occurs in most of the lakes. Using a simple wave model, and assuming that vegetation protects sediments against resuspension, we could eliminate wind resuspension as an important process in 90% of the lakes, leaving resuspension by benthivorous fish as probably the most important factor determining transparency. 7. Chlorophyll a concentration showed a strong positive correlation to ISS concentration, suggesting that resuspension may also have a positive effect on phytoplankton biomass in these lakes. 8. In conclusion, in-lake processes, rather than river dynamics, seem to be driving the turbidity of floodplain lakes along the lower River Rhine. [source]


Fluctuations of Vanessa cardui butterfly abundance with El Niño and Pacific Decadal Oscillation climatic variables

Abstract Annual 4th of July Butterfly Count data spanning more than 20 years are examined to explore Vanessa cardui (Painted Lady) population fluctuations with ENSO (El Niño) and Pacific Decadal Oscillation (PDO) indices. California, Colorado and Nebraska censuses exhibit a strong positive correlation with the strong El Niño events of 1982,1983 and 1997,1998 and the weaker event of 1991,1992. Regression analysis shows the population fluctuations are strongly coupled to climate variations on both short (El Niño) and longer (Pacific Decadal Oscillation) time scales. Recognizing the sensitivity to these time scales is important for predicting longer-term global climate change effects. [source]


Contrasting soil respiration in young and old-growth ponderosa pine forests

Abstract Three years of fully automated and manual measurements of soil CO2 efflux, soil moisture and temperature were used to explore the diel, seasonal and inter-annual patterns of soil efflux in an old-growth (250-year-old, O site) and recently regenerating (14-year-old, Y site) ponderosa pine forest in central Oregon. The data were used in conjunction with empirical models to determine which variables could be used to predict soil efflux in forests of contrasting ages and disturbance histories. Both stands experienced similar meteorological conditions with moderately cold wet winters and hot dry summers. Soil CO2 efflux at both sites showed large inter-annual variability that could be attributed to soil moisture availability in the deeper soil horizons (O site) and the quantity of summer rainfall (Y site). Seasonal patterns of soil CO2 efflux at the O site showed a strong positive correlation between diel mean soil CO2 efflux and soil temperature at 64 cm depth whereas diel mean soil efflux at the Y site declined before maximum soil temperature occurred during summer drought. The use of diel mean soil temperature and soil water potential inferred from predawn foliage water potential measurements could account for 80% of the variance of diel mean soil efflux across 3 years at both sites, however, the functional shape of the soil water potential constraint was site-specific. Based on the similarity of the decomposition rates of litter and fine roots between sites, but greater productivity and amount of fine litter detritus available for decomposition at the O site, we would expect higher rates of soil CO2 efflux at the O site. However, annual rates were only higher at the O site in one of the 3 years (597 ± 45 vs. 427 ± 80 g C m,2). Seasonal patterns of soil efflux at both sites showed influences of soil water limitations that were also reflected in patterns of canopy stomatal conductance, suggesting strong linkages between above and below ground processes. Elevation of interleukin-18 in chronic hepatitis C: implications for hepatitis C virus pathogenesis

Summary The outcome of hepatitis C virus (HCV) infection is determined by the interplay between the virus and the host immune response. Interleukin (IL)-18, an interferon-,-inducing factor, plays a critical role in the T helper type 1 (Th1) response required for host defence against viruses, and antibodies to IL-18 have been found to prevent liver damage in a murine model. The present study was conducted to investigate the possible role of IL-18 in the pathogenesis and persistence of HCV. IL-18 levels were measured in sera of 50 patients at various stages of HCV infection (resolved, chronic and cirrhosis) and compared with those of normal controls. IL-18 gene expression was studied in peripheral blood mononuclear cells (PBMC) from each group, and in liver biopsy tissue from patients with chronic hepatitis C. The mean levels of IL-18 in sera were markedly elevated in patients with chronic hepatitis and cirrhosis, and were reduced in patients with resolved HCV infection. The serum IL-18 concentrations were related to the Child,Pugh severity of liver disease in cirrhotic patients. There also existed a strong positive correlation of IL-18 levels with histological activity score and necrosis. IL-18 mRNA expression was significantly up-regulated in the PBMC of cirrhotic patients when compared with other groups, while in the liver, higher levels of IL-18 transcripts were expressed in patients with chronic hepatitis C. The results of our study indicate that IL-18 levels reflect the severity and activity of HCV infection, and may contribute to the pathogenesis and progression of liver disease associated with HCV. Variability and trends in the directional wave climate of the Southern Hemisphere

Abstract The effect of interannual climate variability and change on the historic, directional wave climate of the Southern Hemisphere is presented. Owing to a lack of in situ wave observations, wave climate in the Southern Hemisphere is determined from satellite altimetry and global ocean wave models. Altimeter data span the period 1985 to present, with the exception of a 2-year gap in 1989,1991. Interannual variability and trends in the significant wave height are determined from the satellite altimeter record (1991 to present), and the dominant modes of variability are identified using an empirical orthogonal function (EOF) analysis. Significant wave heights in the Southern Ocean are observed to show a strong positive correlation with the Southern Annular Mode (SAM), particularly during Austral autumn and winter months. Correlation between altimeter derived significant wave heights and the Southern Oscillation Index is observed in the Pacific basin, which is consistent with several previous studies. Variability and trends of the directional wave climate are determined using the ERA-40 Waves Re-analysis for the period 1980,2001. Significant wave height, mean wave period and mean wave direction data are used to describe the climate of the wave energy flux vector. An EOF analysis of the wave energy flux vector is carried out to determine the dominant modes of variability of the directional seasonal wave energy flux climate. The dominant mode of variability during autumn and winter months is strongly correlated to the SAM. There is an anti-clockwise rotation of wave direction with the southward intensification of the Southern Ocean storm belt associated with the SAM. Clockwise rotation of flux vectors is observed in the Western Pacific Ocean during El-Nino events. Directional variability of the wave energy flux in the Western Pacific Ocean has previously been shown to be of importance to sand transport along the south-eastern Australian margin, and the New Zealand region. The directional variability of the wave energy flux of the Southern Ocean associated with the SAM is expected to be of importance to the wave-driven currents responsible for the transport of sand along coastal margins in the Southern Hemisphere, in particular those on the Southern and Western coastal margins of the Australian continent. Snow cover in western Poland and macro-scale circulation conditions

Abstract The aim of the study was to find out the connection between the nature of winters in the western part of Poland (excluding the Sudety mountains) and the fluctuation in the atmospheric circulation in the North Atlantic region determined by the North Atlantic Oscillation (NAO) index. An attempt was made to establish the correlation between the NAO index and specific meteorological parameters in Pozna,. The strongest positive correlation was obtained for the mean winter temperature (December,March) and strong negative correlation was found for the number of days with snow cover. Winter precipitation in Pozna, was least associated with the NAO. The correlation coefficient was small and not significant. In the next stage of the study, the area of western Poland was examined; however, only one parameter, i.e. the number of days with snow cover, was taken into consideration. At each of 29 stations distributed in the study area the number of days with snow cover was proved to be strongly negatively correlated with the NAO index. Finally, the frequency of air flow directions was taken into consideration and their association with the NAO was examined. A strong negative correlation was obtained for the frequency of northeasterly and easterly air flow directions and a strong positive correlation was calculated for the frequency of westerly and northwesterly airflow directions. Such findings are consistent with the westerly flow of air masses during the positive phase and with the northerly and easterly flows during the negative phase. The results lead to the conclusion that the positive phase of the NAO causes mild and less snowy winters, whereas the negative phase increases the probability of severe and snowy winters in western Poland. The aim of this study was to analyse the relationship between SOC and treatment results measured as glucolysed haemoglobine (HbA1c) in patients with type-2 diabetes. The aim was further to test the relationship between treatment results and an index of patients' participation in active management and emotional state. Eighty-eight patients answered a questionnaire containing 13 statements about sense of coherence (SOC-13), questions about self-assessed health, diabetes activity such as self-management of diet, exercise and self-control of blood sugar and emotional acceptance. There was no direct relationship between SOC-13 and treatment results measured as HbA1c but there was a positive correlation between SOC-13, self-assessed health and HbA1c (P < 0·02). Self-assessed health was seen as a mediating factor. The better patients' estimation of their own health, the higher were SOC-13 scores and the lower HbA1c. There was also a strong positive correlation between low levels of HbA1c and high levels of an index of active management and emotional acceptance of diabetes (P < 0·001). [source]


Imputation of 10-year osteoporotic fracture rates from hip fractures: A clinical validation study

Abstract The World Health Organization (WHO) fracture risk assessment system (FRAX) allows for calibration from country-specific fracture data. The objective of this study was to evaluate the method for imputation of osteoporotic fracture rates from hip fractures alone. A total of 38,784 women aged 47.5 years or older at the time of baseline femoral neck bone mineral density (BMD) measurement were identified in a database containing all clinical dual energy X-ray absorptiometry (DXA) results for the Province of Manitoba, Canada. Health service records were assessed for the presence of nontrauma osteoporotic fracture codes after BMD testing (431 hip, 787 forearm, 336 clinical vertebral, and 431 humerus fractures). Ten-year hip and osteoporotic fracture rates were estimated by the Kaplan-Meier method. The population was stratified by age (50 to 90 years, 5-year width strata) and again by femoral neck T -scores (,4.0 to 0.0, 0.5 SD width strata). Within each stratum, the ratio of hip to osteoporotic fractures was calculated and compared with the predicted ratio from FRAX. Increasing age was associated with greater predicted hip-to-osteoporotic ratios (youngest 0.07 versua oldest 0.41) and observed ratios (youngest 0.10 versus oldest 0.48). Lower T -scores were associated with greater predicted (highest 0.04 versus lowest 0.71) and observed ratios (highest 0.06 versus lowest 0.44). There was a strong positive correlation between predicted and observed ratios (Spearman r,=,0.90,0.97, p,<,.001). For 14 of the 18 strata, the predicted ratio was within the observed 95% confidence interval (CI). Since collection of population-based hip fracture data is considerably easier than collection of non,hip fracture data, this study supports the current emphasis on using hip fractures as the preferred site for FRAX model calibration. © 2010 American Society for Bone and Mineral Research [source]


Effect of Heat Treatment on Antioxidant Capacity and Flavor Volatiles of Mead

ABSTRACT: The objective of this study was to evaluate the effects of heat processing on the antioxidant capacity of mead (honey wine). Soy and buckwheat honey musts were subjected to 2 heat treatments and fermented into wine. Total phenolic concentration was determined. High-performance liquid chromatography (HPLC) phenolic profiling was performed on the methanol fraction of Amberlite extraction. Antioxidant capacity was evaluated using the oxygen radical absorbance capacity (ORAC) assay. Changes in volatile components were evaluated by headspace-solid phase microextraction/gas chromatography-mass spectrometry (H-SPME/GC-MS). ORAC values of experimental meads (3.62 mMTrolox equivalent) were comparable to those of commercial white wine (3.66 mMTrolox equivalent). No significant difference in antioxidant capacity due to heat treatment or honey type was observed. There was no difference in total phenolics between heat treatments in buckwheat mead; however, soy mead made from high-heated must had significantly greater phenolic concentration than the gently heated mead (,= 0.05). Linear regression analysis indicated a strong positive correlation between total phenolic concentration and antioxidant capacity by ORAC (r= 0.9077; P < 0.0001). HPLC analysis of phenolic profiles in the methanol fractions of Amberlite extraction of the meads indicated significantly higher levels of certain phenolics as a result of the high-heat process in buckwheat mead, but not in soy mead. Differences in volatile components that potentially impact flavor were noted between high and low heat treatments. Results of this study suggest dramatic heat treatments that are often avoided because their flavor impact in mead production have the potential to alter the antioxidant capacity of mead by changing phenolic profiles. Clinical and genotypic correlates of mutation K65R in HIV-infected patients failing regimens not including tenofovir

Abstract The mutation RT-K65R confers resistance to tenofovir (TDF). Although its prevalence is increasing with the use of this drug, clinical and genotypic correlates of K65R occurrence have yet to be fully identified. Clinical, virological and immunological and genotypic data of patients naïve for TDF who failed HAART regimens and underwent genotypic resistance test (GRT) during 1999,2003 were collected in a database and analyzed retrospectively. Out of 1392 GRT performed for 771 patients, 12 TDF-naïve patients had the K65R mutation with an overall prevalence of 1.6%. Previous AIDS, the use of abacavir, and treatment with efavirenz at GRT were independently associated with a greater risk of expressing K65R, while patients with longer exposure to lamivudine were less likely to present the mutation. Among genotypic correlates, the presence of M184V and NAMs seems to be protective for the emergence of K65R, while a strong positive correlation was found with the Q151M complex mutation. Moreover, the L100I mutation was independently associated with a higher probability of presenting K65R. The selection of mutation K65R in patients failing without TDF is rare. However, exposure to abacavir and/or efavirenz, presence of Q151M and/or L100I, and prior AIDS may favor the selection of this mutation. A NEW LARVAL FISH BIOASSAY FOR TESTING THE PATHOGENICITY OF PFIESTERIA SPP. (DINOPHYCEAE),

Water quality, microbial contamination, prior fish health, and variable results have been major impediments to identifying the cause and mechanism of fish mortality in standard aquarium-format Pfiesteria bioassays. Therefore, we developed a sensitive 96-h larval fish bioassay for assessing Pfiesteria spp. pathogenicity using six-well tissue culture plates and 7-day-old larval cyprinodontid fish. We used the assay to test pathogenicity of several clonal lines of Pfiesteria piscicida Steidinger and Burkholder and P. shumwayae Glasgow and Burkholder that had been cultured with algal prey for 2 to 36 months. The P. shumwayae cultures exhibited 80%,100% cumulative mortality in less than 96 h at initial zoospore densities of approximately 1000 cells·mL,1. No fish mortalities occurred with P. piscicida at identical densities or in controls. In a dose-response assay, we demonstrated a strong positive correlation between dinospore density and fish mortality in a highly pathogenic culture of P. shumwayae, generating a 96-h LD50 of 108 zoospores·mL,1. Additionally, we applied the assay to evaluate a 38-L P. shumwayae bioassay that was actively killing fish and compared results with those from exposures of juvenile tilapia (Oreochromis niloticus) in a 500-mL assay system. Water from the fish-killing 38-L assay was filtered and centrifuged to produce fractions dominated by dinoflagellates, bacteria, or presumed ichthyotoxin (cell-free fraction). After 96 h, the larval fish assay exhibited 50%,100% cumulative mortality only in fractions containing dinoflagellates, with no mortalities occurring in the other fractions. The 500-mL bioassay with tilapia produced inconsistent results and demonstrated no clear correlation between mortality and treatment. The new larval fish bioassay was demonstrated as a highly effective method to verify and evaluate dinoflagellate pathogenicity. [source]


Use of , -Glucuronidase Activity to Quantify the Growth of Fusarium oxysporum f. sp. radicis-lycopersici during Infection of Tomato

Abstract The , -glucuronidase (gus) reporter gene was integrated into the phytopathogenic fungus Fusarium oxysporum f. sp. radicis-lycopersici (FORL) in a co-transformation experiment using the hygromycin B resistance (hph) gene as selective marker, which resulted in the generation of 10 mitotically stable transformants. One transformant, F30, was selected based on the results of prior detailed characterization of the 10 transformants for growth rate, conidia production and pathogenicity in comparison with the wild-type strain. A strong positive correlation was found between GUS activity and accumulated biomass of in vitro -grown fungus and therefore GUS activity was used to study fungal growth quantitatively in two tomato lines. Although a parallel increase in lesion development and GUS activity was noted for both tomato lines, a correlation between the GUS activity and disease progression was not always possible. Interestingly, the levels of GUS activity obtained for the more resistant line were higher than those obtained for the susceptible line, indicating that disease progression in tomato caused by FORL may not be related only to the amount of fungal biomass within the root tissue. Contextualizing instruction: Leveraging students' prior knowledge and experiences to foster understanding of middle school science

Abstract Contextualizing science instruction involves utilizing students' prior knowledge and everyday experiences as a catalyst for understanding challenging science concepts. This study of two middle school science classrooms examined how students utilized the contextualizing aspects of project-based instruction and its relationship to their science learning. Observations of focus students' participation during instruction were described in terms of a contextualizing score for their use of the project features to support their learning. Pre/posttests were administered and students' final artifacts were collected and evaluated. The results of these assessments were compared with students' contextualizing scores, demonstrating a strong positive correlation between them. These findings provide evidence to support claims of contextualizing instruction as a means to facilitate student learning, and point toward future consideration of this instructional method in broader research studies and the design of science learning environments. © 2007 Wiley Periodicals, Inc. Mercury Accumulation in Periphyton of Eight River Ecosystems,

Abstract:, In 2003, the U.S. Geological Survey (USGS) National Water-Quality Assessment (NAWQA) program and U.S. Environmental Protection Agency studied total mercury (THg) and methylmercury (MeHg) concentrations in periphyton at eight rivers in the United States in coordination with a larger USGS study on mercury cycling in rivers. Periphyton samples were collected using trace element clean techniques and NAWQA sampling protocols in spring and fall from targeted habitats (streambed surface-sediment, cobble, or woody snags) at each river site. A positive correlation was observed between concentrations of THg and MeHg in periphyton (r2 = 0.88, in log-log space). Mean MeHg and THg concentrations in surface-sediment periphyton were significantly higher (1,333 ng/m2 for MeHg and 53,980 ng/m2 for THg) than cobble (64 ng/m2 for MeHg and 1,192 ng/m2 for THg) or woody snag (71 ng/m2 for MeHg and 1,089 ng/m2 for THg) periphyton. Concentrations of THg in surface-sediment periphyton had a strong positive correlation with concentrations of THg in sediment (dry weight). The ratio of MeHg:THg in surface-sediment periphyton increased with the ratio of MeHg:THg in sediment. These data suggest periphyton may play a key role in mercury bioaccumulation in river ecosystems. [source]


Evaluation of the clinical and immunohistological efficacy of the 585-nm pulsed dye laser in the treatment of psoriasis

Abstract Background, The 585-nm pulsed dye laser (PDL) therapy is useful for the patients with psoriasis. PDL treatment is based on selective photothermolysis of the dermal vasculature. Objective, The objectives of this study were to evaluate the clinical and immunohistological effects of PDL on psoriasis and to examine the association between psoriatic dermal vasculature and the clinical effects. Methods, Eleven patients with recalcitrant psoriasis were treated with 585-nm PDL. Biopsy specimens obtained before and after treatment were stained with CD31. All microvessels to the depth of 400 µm from the rete ridge were counted and the internal diameters were measured. Results, The mean percent reduction of plaque severity score was 42. The mean microvessel count decreased significantly from 63 to 35.6 (P < 0.001). There was a strong positive correlation between the plaque severity score and microvessel number (P < 0.001) and a strong negative correlation between the microvessel count of an untreated area and degree of the change in the microvessel count after treatment (P = 0.005). Conclusions, The findings of this study suggest that PDL treatment improves psoriasis. Moreover, PDL treatment decreased the number of dermal papillary microvessels. Dermal papillary microvessels are important pathogenetic targets of psoriasis, and PDL therapy, which selectively targets superficial vessels, is therefore a valid therapeutic approach. Soil moisture stress-induced alterations in bioconstituents determining tea quality

Abstract The impact of water stress on the biochemical constituents that determine black tea quality was investigated. Phenylalanine ammonia lyase (PAL) activity was highest in the drought tolerant ,Assam' cultivar UPASI-2, followed by UPASI-8 and UPASI-9, under non-stress conditions. Under soil moisture stress a reduction in PAL activity was found in all three clones investigated. A strong positive correlation was observed between an increase in soil moisture deficit and a decrease in PAL activity. Lower PAL activity correlated well with lower synthesis of flavanols such as epigallocatechin gallate (EGCG) and epicatechin gallate (ECG), which are important precursors of theaflavin-3,3,-digallate that determines final tea quality. Altered synthesis of EGCG and ECG could be due to their molecular rearrangement at elevated leaf temperature during drought. Synthesis of quality constituents such as gallic acid and caffeine declined significantly owing to both drought and waterlogging stress. The reduction in gallic acid due to water stress could lead to lower synthesis of theaflavin fractions such as epitheaflavic acid, epitheaflavic acid-3,-gallate and theaflavic acid and, thereby, quality deterioration. Mating system, philopatry and patterns of kinship in the cooperatively breeding subdesert mesite Monias benschi

Abstract In the first molecular study of a member of the threatened avian family, Mesitornithidae, we used nine polymorphic microsatellite loci to elucidate parentage, patterns of within-group kinship and occurrence of extra-group paternity in the subdesert mesite Monias benschi, of southwest Madagascar. We found this cooperatively breeding species to have a very fluid mating system. There was evidence of genetic monogamy and polygynandry: of the nine groups with multiple offspring, six contained one breeding pair with unrelated helpers and three contained multiple male and female breeders with related helpers. Although patterns of within-group kinship varied, there was a strong positive relationship between group size and relatedness, suggesting that groups form by natal philopatry. There was also a strong positive correlation between within-sex and between-sex relatedness, indicating that unlike most cooperatively breeding birds, philopatry involved both sexes. In contrast to predictions of kin selection and reproductive skew models, all monogamous groups contained unrelated individuals, while two of the three polygynandrous groups were families. Moreover, although between-group variation in seasonal reproductive success was related to within-group female relatedness, relatedness among males and between the sexes had no bearing on a group's reproductive output. While kin selection may underlie helping behaviour in females, factors such as direct long-term fitness benefits of group living probably determine helping in males. Of the 14 offspring produced by fully sampled groups, at least two were sired by males from neighbouring groups: one by a breeding male and one by a nonbreeding male, suggesting that males may augment their reproductive success through extra-group paternity. Relative contributions of nine genes in the pathway of histidine biosynthesis to the control of free histidine concentrations in Arabidopsis thaliana

Summary Despite the functional importance of histidine (His) as an essential amino acid in proteins and as a metal-coordinating ligand, comparatively little is known about the regulation of its biosynthesis in plants and the potential for metabolic engineering of this pathway. To investigate the contribution of different steps in the pathway to overall control of His biosynthesis, nine His biosynthetic genes were individually over-expressed in Arabidopsis thaliana to determine their effects on free amino acid pools. Constitutive, CaMV 35S -driven over-expression of the cDNAs encoding either isoform of ATP-phosphoribosyltransferase (ATP-PRT), the first enzyme in the pathway, was sufficient to increase the pool of free His by up to 42-fold in shoot tissue of Arabidopsis, with negligible effect on any other amino acid. In contrast, over-expression of cDNAs for seven other enzymes in the biosynthetic pathway had no effect on His content, suggesting that control of the pool of free His resides largely with ATP-PRT activity. Over-expression of ATP-PRT and increased His content had a negative pleiotropic effect on plant biomass production in 35S:PRT1 lines, but this effect was not observed in 35S:PRT2 lines. In the presence of 100 µM Ni, which was inhibitory to wild-type plants, a strong positive correlation was observed between free His content and biomass production, indicating that the metabolic cost of His overproduction was outweighed by the benefit of increased tolerance to Ni. His-overproducing plants also displayed somewhat elevated tolerance to Co and Zn, but not to Cd or Cu, indicating chemical selectivity in intracellular metal binding by His. [source]


How does photorespiration modulate leaf amino acid contents?

A dual approach through modelling, metabolite analysis
Abstract The aim of this work was to establish the quantitative impact of photorespiration on leaf amino acid contents. Attached leaves of wheat and potato were incubated for 30,40 min under defined conditions in which net CO2 uptake (A) was manipulated by irradiance, ambient CO2 or ambient O2. The incubated portion of the leaf was sampled by a rapid-quench method and photorespiratory flux (vo) was modelled from the measured rate of net CO2 uptake. In both wheat and potato, the ratio between glycine and serine showed a strong positive correlation with vo. Aspartate and alanine correlated negatively with vo but glutamate and glutamine showed less clear relationships. In potato, glutamate and glutamine did not correlate clearly with either A or vo. In wheat, glutamine showed a general increase with A but no relationship with vo, whereas 2-oxoglutarate contents correlated positively with vo and negatively with A. As a result, glutamine : glutamate and glutamine : 2-oxoglutarate increased with net CO2 uptake in wheat, observations that are attributed primarily to imperfect and variable coupling between the supply of NH3 in primary nitrogen assimilation and the associated delivery of 2-oxoglutarate to the chloroplast. A simple theoretical analysis is used to illustrate the potentially marked impact of primary nitrogen assimilation on leaf glutamine, even against a background of high rates of photorespiratory ammonia recycling. Carbon isotope discrimination and wood anatomy variations in mixed stands of Quercus robur and Quercus petraea

Abstract The two most common oak species in western Europe, Quercus robur and Quercus petraea, display different ecological behaviours, particularly with respect to their responses to drought. The ecophysiological basis of this niche difference is not understood well. Here we test the hypothesis that these two species present distinct water use efficiencies (WUEs), using the carbon isotope discrimination approach. Leaves and 13 dated ring sequences were sampled in 10 pairs of adult trees growing side by side. Carbon isotope composition was measured on cellulose extracts. In addition, relationships between carbon isotope discrimination and wood anatomy were assessed at the tree level. Quercus robur displayed a 1·0, larger isotopic discrimination than Q. petraea, and therefore a lower intrinsic WUE (,13%). This interspecific difference of isotopic discrimination was quite stable with time and independent of tree radial growth and climate fluctuations. A strong positive correlation was observed between average tree values of earlywood vessel surface area and 13C isotopic discrimination. This correlation was even higher with 13C of the 1976 dry year (r = 0·86). These observations led to the hypothesis that hydraulic properties of xylem could exert a constraint on leaf gas exchange, resulting in a larger WUE for individuals with smaller vessel cross-section area.
