    Characterization of a family with dominant hypophosphatasia

    A kindred with dominant hypophosphatasia resulting from an alanine to threonine substitution at position 99 of the alkaline phosphatase protein is described. The clinical findings of individual members of the kindred were assessed by oral and physical examinations, or from the descriptions of multiple family members. The proband displayed enamel hypoplasia and premature loss of fully rooted primary anterior teeth, which were shown by histological examination to lack cementum. Serum alkaline phosphatase (ALP) and a vitamin B6 panel, and urine phosphoethanolamine (PEA) were measured on 21 family members. Based upon the clinical and laboratory tests, affected and unaffected status was assigned. Parametric linkage analysis of the kindred using different dominant models and frequency distributions for the disease allele and the mutation gave lodscores >4.2 and confirmed the strong linkage between the disease and the mutation. Assuming the defined mutation causes the disease, the reliability of clinical and laboratory tests is assessed. [source]


    Localization of the membrane-anchored MMP-regulator RECK at the neuromuscular junctions

    Abstract Nerve apposition on nicotinic acetylcholine receptor clusters and invagination of the post-synaptic membrane (i.e. secondary fold formation) occur by embryonic day 18.5 at the neuromuscular junctions (NMJs) in mouse skeletal muscles. Finding the molecules expressed at the NMJ at this stage of development may help elucidating how the strong linkage between a nerve terminal and a muscle fiber is established. Immunohistochemical analyses indicated that the membrane-anchored matrix metalloproteinase regulator RECK was enriched at the NMJ in adult skeletal muscles. Confocal and electron microscopy revealed the localization of RECK immunoreactivity in secondary folds and subsynaptic intracellular compartments in muscles. Time course studies indicated that RECK immunoreactivity becomes associated with the NMJ in the diaphragm at around embryonic day 18.5 and thereafter. These findings, together with known properties of RECK, support the hypothesis that RECK participates in NMJ formation and/or maintenance, possibly by protecting extracellular components, such as synaptic basal laminae, from proteolytic degradation.     LABOUR MARKET ACTIVITY OF FOREIGN SPOUSES IN TAIWAN: EMPLOYMENT STATUS AND CHOICE OF EMPLOYMENT SECTOR

    The present study examines the employment status and choice of employment sector of female foreign spouses from Southeast Asia and Mainland China in Taiwan. The conceptual framework is based on the family labour supply model, human and social capital theory, and immigrant assimilation theory. Our findings indicate that in regard to employment status, family background variables, including the presence of small children and husbands' characteristics, play a more significant role in determining the employment probability for these foreign spouses than do human capital variables. In particular, for spouses from Southeast Asia, each additional child is correlated with a decrease in working probability of 11.3%, whereas college education has an insignificant effect on their employment probability. Employment assimilation for these marriage immigrants may be confirmed by the finding that the employment probability of foreign spouses rises rapidly with the number of years that have elapsed since migration. As for the choice of employment sector, a strong linkage between the employment sector of the foreign spouses and their husbands' employment sector is found in this study. [source]


    Oxytocin as a "High Alert Medication": A Multilayered Challenge to the Status Quo

    ABSTRACT: Oxytocin is the drug most commonly associated with preventable adverse perinatal outcomes. In 2007 it was added to the Institute for Safe Medication Practices short list of medications "bearing a heightened risk of harm," which may "require special safeguards to reduce the risk of error." In January 2009 the American Journal of Obstetrics and Gynecology published a Clinical Opinion paper about oxytocin's inclusion on the list and how the obstetrics profession in the United States should respond. The authors call for the development of specific evidence-based guidelines to reduce the likelihood of patient harm by limiting elective use of oxytocin, decreasing the need for indicated use, reducing dosages during necessary use, giving more responsibility and authority for the patient's safety to the professional who is "at the bedside administering and monitoring the oxytocin infusion" (i.e., the nurse), and accepting that "more time rather than more oxytocin is generally preferable" once adequate uterine activity has been achieved. It is unfortunate that this important paper discounted the risk of harm from cesarean sections and did not mention the strong linkage between epidural analgesia and use of oxytocin. Physicians, midwives, nurses, and others should examine and discuss these issues further in view of increased alertness to the risk of harm from unsafe use of oxytocin. [source]


    Analyzing Factors Affecting U.S. Food Price Inflation

    Since the summer of 2007, U.S. food price inflation has increased dramatically. Given public anxiety over fast-rising food prices, this study attempts to analyze the effects of market factors,prices of energy and agricultural commodities and exchange rate,on U.S. food prices using a cointegration analysis. Results show that the agricultural commodity price and exchange rate play the key roles in determining the short- and long-run movement of U.S. food prices. It is also found that in recent years, energy price has been a significant factor affecting U.S. food prices in the long run, but has little effect in the short run. This implies the strong linkage between energy and agricultural markets in the long run over the recent years. Depuis l'été 2007, l'inflation des prix des aliments aux États-Unis a augmenté considérablement. En raison de l'anxiété que la hausse rapide des prix des aliments suscite au sein de la population, nous avons tenté d'évaluer, à l'aide d'une analyse de co-intégration, les répercussions de certains facteurs de marché, le prix de l'énergie, le prix des produits agricoles primaires et le taux change , sur les prix des aliments aux États-Unis. Les résultats ont montré que les prix des produits agricoles primaires et le taux de change jouent un rôle important dans la détermination des tendances à court et à long terme des prix des aliments aux États-Unis. Nous avons également constaté que, au cours des dernières années, le prix de l'énergie a eu une forte influence sur les prix des aliments aux États-Unis à long terme, mais peu d'influence à court terme. Cette observation montre l'étroit lien à long terme des marchés de l'énergie et des produits agricoles au cours des dernières années. [source]


    In vivo manipulation of V,9V,2 T cells with zoledronate and low-dose interleukin-2 for immunotherapy of advanced breast cancer patients

    Summary The potent anti-tumour activities of ,, T cells have prompted the development of protocols in which ,,-agonists are administered to cancer patients. Encouraging results from small Phase I trials have fuelled efforts to characterize more clearly the application of this approach to unmet clinical needs such as metastatic carcinoma. To examine this approach in breast cancer, a Phase I trial was conducted in which zoledronate, a V,9V,2 T cell agonist, plus low-dose interleukin (IL)-2 were administered to 10 therapeutically terminal, advanced metastatic breast cancer patients. Treatment was well tolerated and promoted the effector maturation of V,9V,2 T cells in all patients. However, a statistically significant correlation of clinical outcome with peripheral V,9V,2 T cell numbers emerged, as seven patients who failed to sustain V,9V,2 T cells showed progressive clinical deterioration, while three patients who sustained robust peripheral V,9V,2 cell populations showed declining CA15-3 levels and displayed one instance of partial remission and two of stable disease, respectively. In the context of an earlier trial in prostate cancer, these data emphasize the strong linkage of V,9V,2 T cell status to reduced carcinoma progression, and suggest that zoledronate plus low-dose IL-2 offers a novel, safe and feasible approach to enhance this in a subset of treatment-refractory patients with advanced breast cancer.     Contrasting soil respiration in young and old-growth ponderosa pine forests

    Abstract Three years of fully automated and manual measurements of soil CO2 efflux, soil moisture and temperature were used to explore the diel, seasonal and inter-annual patterns of soil efflux in an old-growth (250-year-old, O site) and recently regenerating (14-year-old, Y site) ponderosa pine forest in central Oregon. The data were used in conjunction with empirical models to determine which variables could be used to predict soil efflux in forests of contrasting ages and disturbance histories. Both stands experienced similar meteorological conditions with moderately cold wet winters and hot dry summers. Soil CO2 efflux at both sites showed large inter-annual variability that could be attributed to soil moisture availability in the deeper soil horizons (O site) and the quantity of summer rainfall (Y site). Seasonal patterns of soil CO2 efflux at the O site showed a strong positive correlation between diel mean soil CO2 efflux and soil temperature at 64 cm depth whereas diel mean soil efflux at the Y site declined before maximum soil temperature occurred during summer drought. The use of diel mean soil temperature and soil water potential inferred from predawn foliage water potential measurements could account for 80% of the variance of diel mean soil efflux across 3 years at both sites, however, the functional shape of the soil water potential constraint was site-specific. Based on the similarity of the decomposition rates of litter and fine roots between sites, but greater productivity and amount of fine litter detritus available for decomposition at the O site, we would expect higher rates of soil CO2 efflux at the O site. However, annual rates were only higher at the O site in one of the 3 years (597 ± 45 vs. 427 ± 80 g C m,2). Seasonal patterns of soil efflux at both sites showed influences of soil water limitations that were also reflected in patterns of canopy stomatal conductance, suggesting strong linkages between above and below ground processes.     Priority research areas for ecosystem services in a changing world

    Summary 1.,Ecosystem services are the benefits humans obtain from ecosystems. The importance of research into ecosystem services has been widely recognized, and rapid progress is being made. However, the prevailing approach to quantifying ecosystem services is still based on static analyses and single services, ignoring system dynamics, uncertainty and feedbacks. This is not only partly due to a lack of mechanistic understanding of processes and a dearth of empirical data, but also due to a failure to engage fully with the interdisciplinarity of the problem. 2.,We argue that there is a tendency to ignore the feedbacks between and within both social and ecological systems, and a lack of explicit consideration of uncertainty. Metrics need to be developed that can predict thresholds, which requires strong linkages to underlying processes, while the development of policy for management of ecosystem services needs to be based on a broader understanding of value and drivers of human well-being. 3.,We highlight the complexities, gaps in current knowledge and research, and the potentially promising avenues for future investigation in four priority research areas: agendas, processes, metrics and uncertainty. 4.,Synthesis and applications. The research interest in the field of ecosystem services is rapidly expanding, and can contribute significantly to the sustainable management of natural resources. However, a narrow disciplinary approach, or an approach which does not consider feedbacks within and between ecological and social systems, has the potential to produce dangerously misleading policy recommendations. In contrast, if we explicitly acknowledge and address uncertainties and complexities in the provision of ecosystem services, progress may appear slower but our models will be substantially more robust and informative about the effects of environmental change.     Politics And Gastropolitics: Gender And The Power Of Food In Two African Pastoralist Societies

    Male-centred aspects of political behaviour have generally remained the explanatory and interpretive focuses in analyses of the social organization of African pastoralists. While recent work on African pastoralists has shed increasing light on the lives of women, I argue that key assumptions underlying anthropological models of male dominance in these societies have been insufficiently challenged. Drawing on recent approaches in gender and social organization that highlight the mutual constitution of domestic and political domains, I examine comparative material from two well-known pastoralist societies: the Samburu of northern Kenya and the Nuer of southern Sudan. In doing so, I suggest strong linkages between male-dominated ,political spheres' and areas of domestic life in which the role of women is more significant , particularly processes of domestic food distribution. In re-examining central facets of Samburu politics , which are best known through Paul Spencer's seminal analysis of the gerontocratic aspects of Samburu political life , I suggest that the status and identities of Samburu men are in fundamental ways defined through their relationship to women as providers of food within Samburu households. Comparative material from the Nuer suggests, additionally, the strategic use of food by women in influencing male ,political spheres'. In comparing these cases, I suggest a more general model through which domestic processes of food allocation as realms of female-centred social action may be seen to play a central role in the forms and processes of pastoral ,political' life.
