Structural Correlates of Functional Language Dominance: A Voxel-Based Morphometry Study

ABSTRACT BACKGROUND AND PURPOSE The goal of this study was to explore the structural correlates of functional language dominance by directly comparing the brain morphology of healthy subjects with left- and right-hemisphere language dominance. METHODS Twenty participants were selected based on their language dominance from a cohort of subjects with known language lateralization. Structural differences between both groups were assessed by voxel-based morphometry, a technique that automatically identifies differences in the local gray matter volume between groups using high-resolution T1-weighted magnetic resonance images. RESULTS The main findings can be summarized as follows: (1) Subjects with right-hemisphere language dominance had significantly larger gray matter volume in the right hippocampus than subjects with left-hemisphere language dominance. (2) Leftward structural asymmetries in the posterior superior temporal cortex, including the planum temporale (PT), were observed in both groups. CONCLUSIONS Our study does not support the still prevalent view that asymmetries of the PT are related in a direct way to functional language lateralization. The structural differences found in the hippocampus underline the importance of the medial temporal lobe in the neural language network. They are discussed in the context of recent findings attributing a critical role of the hippocampus in the development of language lateralization. [source]


Precursor complex structure of pseudouridine synthase TruB suggests coupling of active site perturbations to an RNA-sequestering peripheral protein domain

Abstract The pseudouridine synthase TruB is responsible for the universally conserved post-transcriptional modification of residue 55 of elongator tRNAs. In addition to the active site, the "thumb," a peripheral domain unique to the TruB family of enzymes, makes extensive interactions with the substrate. To coordinate RNA binding and release with catalysis, the thumb may be able to sense progress of the reaction in the active site. To establish whether there is a structural correlate of communication between the active site and the RNA-sequestering thumb, we have solved the structure of a catalytically inactive point mutant of TruB in complex with a substrate RNA, and compared it to the previously determined structure of an active TruB bound to a reaction product. Superposition of the two structures shows that they are extremely similar, except in the active site and, intriguingly, in the relative position of the thumb. Because the two structures were solved using isomorphous crystals, and because the thumb is very well ordered in both structures, the displacement of the thumb we observe likely reflects preferential propagation of active site perturbations to this RNA-binding domain. One of the interactions between the active site and the thumb involves an active site residue whose hydrogen-bonding status changes during the reaction. This may allow the peripheral RNA-binding domain to monitor progress of the pseudouridylation reaction. [source]


EXTENDING SOCIAL DISORGANIZATION THEORY: A MULTILEVEL APPROACH TO THE STUDY OF VIOLENCE AMONG PERSONS WITH MENTAL ILLNESSES,

Prior studies of violence among individuals with mental illnesses have focused almost exclusively on individual-level characteristics. In this study, I examine whether the structural correlates of neighborhood social disorganization also explain variation in violence. I use data on 270 psychiatric patients who were treated and discharged from an acute inpatient facility combined with tract-level data from the 1990 U.S. Census. I find that living in a socially disorganized neighborhood increased the probability of violence among the sample, an effect that was not mediated by self-reported social supports. Implications for future research in the areas of violence and mental illness are discussed. [source]


SOCIAL DISORGANIZATION OUTSIDE THE METROPOLIS: AN ANALYSIS OF RURAL YOUTH VIOLENCE,

In order to extend the study of community social disorganization and crime beyond its exclusive focus on large urban centers, we present an analysis of structural correlates of arrest rates for juvenile violence in 264 nonmetropolitan counties of four states. Findings support the generality of social disorganization theory: Juvenile violence was associated with rates of residential instability, family disruption, and ethnic heterogeneity. Though rates of poverty were not related to juvenile violence, this is also in accord with social disorganization theory because, unlike urban settings, poverty was negatively related to residential instability. Rates of juvenile violence varied markedly with population size through a curvilinear relationship in which counties with the smallest juvenile populations had exceptionally low arrest rates. Analyses used negative binomial regression (a variation of Poisson regression) because the small number of arrests in many counties meant that arrest rates would be ill suited to least-squares regression. Preliminary crystallographic study of the Streptococcus agalactiae sortases, sortase A and sortase C1

Sortases are cysteine transpeptidases that are essential for the assembly and anchoring of cell-surface adhesins in Gram-positive bacteria. In Streptococcus agalactiae (GBS), the pilin-specific sortase SrtC1 catalyzes the polymerization of pilins encoded by pilus island 1 (PI-1) and the housekeeping sortase SrtA is necessary for cell-wall anchoring of the resulting pilus polymers. These sortases are known to utilize different substrates for pilus polymerization and cell-wall anchoring; however, the structural correlates that dictate their substrate specificity have not yet been clearly defined. This report presents the expression, purification and crystallization of SrtC1 (SAG0647) and SrtA (SAG0961) from S. agalactiae strain 2603V/R. The GBS SrtC1 has been crystallized in three crystal forms and the GBS SrtA has been crystallized in one crystal form. [source]
