Selected AbstractsNovel haplotypes in 17q21 are associated with progressive supranuclear palsyANNALS OF NEUROLOGY, Issue 2 2004Pau Pastor MD Progressive supranuclear palsy (PSP) and corticobasal degeneration (CBD) are sporadic neurodegenerative diseases presenting as atypical parkinsonian disorders, characterized by the presence of tau-positive neurofibrillary tangles. Recently, an extended haplotype (H1E) of 787.6kb that comprises several genes including MAPT showed increased association with PSP. The objective of this study was to determine the size of the H1E haplotype associated with PSP and CBD in different populations and to identify specific subhaplotypes in the background of H1E haplotype. Nineteen single nucleotide polymorphisms (SNPs) in the 17q21 region were genotyped in two case,control samples. The SNPs that were associated with higher risk for the disease in the homozygous state delimit a region of more that 1Mb. Haplotype analyses in the Spanish sample showed that the most frequent haplotype found among the patients (H1E,), which extends 1.04Mb and contains several genes such as MAPT, CRHR1, IMP5, Saitohin, WTN3, and NSF. A specific subhaplotype (H1E,A) was present in 16% of PSP patients but was not observed in the controls. Furthermore, the H2E,A haplotype, was rarely present in the disease group suggesting that it plays a protective role. The identification of these specific subhaplotypes that modify risk for PSP/CBD supports the hypothesis that a pathogenic allele exists in a subgroup of PSP patients. Berrios-Hernandez Abstract The purpose of this study was to examine the psychometric properties of a Spanish version of the Bulimia Test-Revised (BULIT-R). The goal was to test the factor-structure equivalence of the BULIT-R across two samples of college students from two different cultures, Spain and the US. Researchers using exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) have reported different model solutions for the factor-structure of the BULIT-R: a one-factor model, a four-factor model, a five-factor model and a six-factor model. For the two samples, CFA did not support any of the models previously reported in the literature. [source] Replicable types and subtypes of personality: Spanish NEO-PI samplesEUROPEAN JOURNAL OF PERSONALITY, Issue S1 2002Bettina Boehm Three major personality prototypes were derived on the basis of a Big Five instrument (NEO-PI) by means of replicated cluster analysis in two Spanish samples (a sample from the general population and a student sample). The replicability of the three prototypes within and their consistency between the two samples were evaluated. In addition, subtypes were analysed in a similar way. Finally, the relation between prototype assignment and level of education was examined in the sample from the general population. Within-study replicability was satisfactory only for the student sample. Comparison with the results for a similar instrument (NEO-PI-R) applied to a German sample showed satisfactory consistency only for the student sample. [source] Evaluating cluster analysis solutions: an application to the Italian NEO personality inventoryEUROPEAN JOURNAL OF PERSONALITY, Issue S1 2002Claudio Barbaranelli This paper is concerned with the evaluation of cluster analysis solutions. Internal criteria and replication issues are compared and applied to empirical data collected from an Italian sample of 421 young adults, using the NEO Personality Inventory. The following internal criteria were considered: C, gamma, and G(,+,) indices, and point-biserial correlation. Replication was examined (i) ,internally' using double cross-validation and bootstrap approaches and (ii) ,externally' by comparing the solution obtained on the Italian sample with the results obtained in German and Spanish samples. While replication analyses supported three- and four-cluster solutions, internal criteria (with the exception of point-biserial correlation) tended to privilege solutions with a much larger number of groups. [source] Analysis of mitochondrial DNA diversity in Burkina Faso populations confirms the maternal genetic homogeneity of the West African goatANIMAL GENETICS, Issue 3 2009L. J. Royo Summary To date, no comprehensive study has been performed on mitochondrial genetic diversity of the West African goat. Here, we analysed a 481-bp fragment of the HVI region of 111 goats representing four native West African populations, namely the three main Burkina Faso breeds, zoo-farm kept Dwarf goats and endangered Spanish goat breeds used as the outgroup. Analyses gave 83 different haplotypes with 102 variable sites. Most haplotypes (65) were unique. Only three haplotypes were shared between populations. Haplotypes were assigned to cluster A except for H45 (belonging to the Spanish Bermeya goat) which was assigned to cluster C. amova analysis showed that divergence between groups (,CT) was not statistically significant regardless of whether the partition in two hierarchical levels that was fitted included Spanish samples or not. The West African goat scenario shown here is consistent with that previously reported for the species: haplogroup A is predominant and has a very high haplotype diversity regardless of the geographic area or sampled breed. The large phenotypic differences observable between the West African Dwarf and Sahelian long-legged goat populations are not detectable with mitochondrial markers. Moreover, a previously suggested introgression of Sahelian goat southwards because of desertification could not be assessed using mtDNA information. Esteves Abstract Pneumonia caused by the opportunistic organism Pneumocystis jirovecii is a clinically important infection affecting AIDS and other immunocompromised patients. The present study aimed to compare and characterise the frequency pattern of DNA sequences from the P. jirovecii mitochondrial large-subunit rRNA (mtLSU rRNA) gene, the dihydropteroate synthase (DHPS) gene and the internal transcribed spacer (ITS) regions of the nuclear rRNA operon in specimens from Lisbon (Portugal) and Seville (Spain). Total DNA was extracted and used for specific molecular sequence analysis of the three loci. In both populations, mtLSU rRNA gene analysis revealed an overall prevalence of genotype 1. In the Portuguese population, genotype 2 was the second most common, followed by genotype 3. Inversely, in the Spanish population, genotype 3 was the second most common, followed by genotype 2. The DHPS wild-type sequence was the genotype observed most frequently in both populations, and the DHPS genotype frequency pattern was identical to distribution patterns revealed in other European studies. ITS types showed a significant diversity in both populations because of the high sequence variability in these genomic regions. The most prevalent ITS type in the Portuguese population was Eg, followed by Cg. In contrast to other European studies, Bi was the most common ITS type in the Spanish samples, followed by Eg. A statistically significant association between mtLSU rRNA genotype 1 and ITS type Eg was revealed. [source] |
