    A double-blind, randomized, parallel group study to compare the efficacy, safety and tolerability of slow-release oral morphine versus methadone in opioid-dependent in-patients willing to undergo detoxification

    ABSTRACT Aims Evaluation of the efficacy and safety of slow-release oral morphine (SROM) compared with methadone for detoxification from methadone and SROM maintenance treatment. Design Randomized, double-blind, double-dummy, comparative multi-centre study with parallel groups. Setting Three psychiatric hospitals in Austria specializing in in-patient detoxification. Participants Male and female opioid dependents (age > 18 years) willing to undergo detoxification from maintenance therapy in order to reach abstinence. Interventions Abstinence was reached from maintenance treatment by tapered dose reduction of either SROM or methadone over a period of 16 days. Measurements Efficacy analyses were based on the number of patients per treatment group completing the study, as well as on the control of signs and symptoms of withdrawal [measured using Short Opioid Withdrawal Scale (SOWS)] and suppression of opiate craving. In addition, self-reported somatic and psychic symptoms (measured using Symptom Checklist SCL-90-R) were monitored. Findings Of the 208 patients enrolled into the study, 202 were eligible for analysis (SROM: n = 102, methadone: n = 100). Completion rates were 51% in the SROM group and 49% in the methadone group [difference between groups: 2%; 95% confidence interval (CI): ,12% to 16%]. The rate of discontinuation in the study was high mainly because of patients voluntarily withdrawing from treatment. No statistically significant differences between treatment groups were found in terms of signs and symptoms of opiate withdrawal, craving for opiates or self-reported symptoms. SROM and methadone were both well tolerated. Conclusions Detoxification from maintenance treatment with tapered dose reduction of SROM is non-inferior to methadone. [source]


    Safety, efficacy, and long-term results of a modified version of rapid opiate detoxification under general anaesthesia: A prospective study in methadone, heroin, codeine and morphine addicts

    Background: In the present study a method of rapid opiate detoxification under general anaesthesia has been evaluated regarding the safety, the efficacy in preventing withdrawal symptoms, and the long-term results. In addition, it was investigated whether the profile and severity of withdrawal symptoms depend on the type of opiate abused (methadone, heroin, codeine, morphine). Methods: Seventy-two opiate addicts were detoxified in an intensive care unit (ICU). Anaesthesia was induced and maintained using propofol infusion. Patients were endotracheally intubated. The opiate receptor antagonist naltrexon was administered into the stomach via a nasogastric tube. Withdrawal symptoms before and after the detoxification treatment were assessed using an objective and a subjective opiate withdrawal scale (OOWS, SOWS). After detoxification patients entered a long-term naltrexone maintenance programme as well as a supportive psychotherapy programme. Vital organ function was monitored using haemodynamic and respiratory parameters as well as body temperature. Results: Organ function parameters were stable during the whole treatment in all patients and no anaesthetic complications were registered. Minor side effects such as bradycardia or hypotension were observed in 20 patients. Compared to patients with pre-existing heroin, codeine, or morphine abuse respectively, patients from the methadone maintenance programme had significantly higher (P<0.01) OOWS as well as SOWS values after the treatment. Twelve months after the detoxification 49 patients (68%) were abstinent from opiates whereas 17 patients had relapsed during the period of follow-up. Six patients were lost during follow-up. Conclusions: Rapid opiate detoxification under general anaesthesia is a safe and efficient method to suppress withdrawal symptoms. This treatment may be of benefit in patients who particularly suffer from severe withdrawal symptoms during detoxification and who have failed repeatedly to complete conventional withdrawal. Methadone patients have more withdrawal symptoms than other opiate addicts. [source]


    Effects of Exogenous ACTH during Oestrus on Early Embryo Development and Oviductal Transport in the Sow

    Contents This study was conducted to assess the effects of ACTH injections on the early development of embryos and their transportation to the uterus. Fifteen sows were monitored for ovulation using transrectal ultrasonography during the first two oestrous periods after weaning. The sows were randomly divided into a control group (C group, n = 8) and an ACTH-treated group (ACTH group, n = 7), and were all surgically fitted with intra-jugular catheters. From the onset of the second standing oestrus after weaning, the sows were injected (NaCl/synthetic ACTH) every 4 h. Blood samples were collected immediately before and 45 min after each injection. All sows were inseminated once 10,33 h before ovulation in their second oestrus after weaning. At 48 (n = 4) or 60 (n = 11) h after ovulation during their second oestrus, the sows were killed and the embryos retrieved from the oviduct and uterus. The embryos were counted and compared with the number of corpora lutea, cleavage rate was noted and, finally, the embryos were prepared for confocal laser scanning microscopy and transmission electron microscopy. There was no difference between the groups regarding cleavage rate, the cytoskeleton, or the number of active nucleoli. However, the ACTH group had significantly (p < 0.05) fewer ova/embryos retrieved (51%) than the C group (81%), and there was a tendency towards faster transportation to the uterus in the ACTH group, possibly because of high progesterone concentrations during treatment. To conclude, administration of ACTH every 4 h from onset of oestrus to 48 h caused significant loss of oocytes or embryos, and possibly faster transportation through the oviduct. [source]


    Welfare Index and Reproductive Performance in the Sow

    Contents To study the relationship between on-farm welfare and reproductive performance in the sow, the TGI 35L Animal Needs Index was modified for use in Finnish pig production. The modified index had a maximal total score of 100. It was comprised of six categories: ,locomotion' (maximal score 21 for dry sows and 11 for lactating sows), ,social interaction' (12/8), ,floor quality' (16/9), ,stable climate' (16/21), ,feeding' (16/23) and ,health and stockmanship' (19/29). Index scorings were performed separately in farrowing, breeding and gestation units on 28 representative Finnish sow farms. One-year production parameters were collected. Multiple linear regression was used for statistical analysis. Total ANI-points varied between 36.5,68.0 for lactating and 39.5,86.0 for dry sows. Litter size increased with increasing scores for ,feeding' in the dry sow unit. Controlling for breed, high scores for ,health and stockmanship' and ,floor quality' shortened the reproductive cycle, probably because of good leg health. The number of weaned piglets per sow per year (PSY) increased with increasing scores for dry sow ,health and stockmanship', ,floor quality' and an interaction of ,feeding' in the farrowing and mating units. PSY increased with decreasing scores for farrowing pen ,climate'. High-quality floors and stockmanship appear to correlate positively with reproductive performance in the sow. Effects of a welfare-promoting feeding strategy on reproduction are contradictory. [source]


    Nursing Synchronization and Milk Ejection Failure as Maternal Strategies to Reduce Allosuckling in Pair-Housed Sows (Sus scrofa domestica)

    Non-offspring nursing (allosuckling) is costly for lactating females. It is reasonable to assume, therefore, that mechanisms have evolved to control the frequency of allosuckling. It is suggested that the synchronization of nursing between females could be such a mechanism. The aim of this study was to investigate whether nursing synchronization is related to the timing of nursing behaviour of paired sows and the behaviour of alien piglets in relation to allosuckling. Specifically, we predicted that: (i) nursing synchronization will increase if there is a real risk of allosuckling compared with a situation where there is none, (ii) nursing synchronization indeed reduces allosuckling and, therefore, those nursings with allosuckling attempts will be less closely synchronized than those without allosuckling attempts and (iii) in non-synchronized nursings, the sows will more often employ another strategy to reduce allosuckling, namely, they will make the nursings non-nutritive by not releasing milk. Ten pairs of sows and their litters were used in this study. For each pair, the two sows were housed individually until their litters were weaned; however, their litters had access to both sows from day 11 post-partum. The behaviour of the 10 pairs of sows with their litters was videotaped for 6 h on days 10, 11, 17 and 24 post-partum. The frequency of allosuckling attempts, non-nutritive nursings and allosuckling during milk ejection was analysed for every pair for every day of observation. The observed frequency of nursing synchronization was high. The median interval between the start of a nursing within one pair of sows was 32 s. The first prediction was supported as synchronization became significantly tighter when piglets were given the possibility to allosuckle on day 11 [Cochran,Mantel,Haenszel (CMH) statistics, p < 0.0001]. The second prediction was also supported; the nursings were less closely synchronized when there were allosuckling attempts (CMH statistics, p < 0.05). Of all nursings, 36% had an allosuckling attempt; however, only 17% resulted in real allosuckling. Evidence in favour of the third prediction was also found as non-nutritive nursings were more frequent in non-synchronized nursings (CMH statistics, p < 0.05). Finally, real allosuckling during milk ejection was unrelated to the degree of nursing synchronization, as in less synchronized nursings, the sows responded to allosuckling attempts by not releasing milk. Our results suggest that sows are not able to completely suppress allosuckling; however, they can reduce its frequency through nursing synchronization and non-nutritive nursings. [source]


    Supplementation of diets for lactating sows with zinc amino acid complex and gastric nutriment-intubation of suckling pigs with zinc methionine on mineral status, intestinal morphology and bacterial translocation in lipopolysaccharide-challenged weaned pigs

    Summary Sixty-four pigs from 16 sows were used to evaluate addition of zinc amino acid complex (ZnAA) to lactating sows and gastric nutriment-intubation of zinc methionine (ZnMet) to suckling pigs on mineral status, intestinal morphology and bacterial translocation after weaning. Sows were fed a barley-based diet supplying 120 ppm zinc (Zn; control) or the control diet supplemented with 240 ppm Zn from ZnAA. At birth, day-10 and day-21 (weaning) of age, pigs from each litter were nutriment-intubated with 5 ml of an electrolyte solution without or with 40 mg Zn from ZnMet. At weaning, 24 h prior to the collection of small and large intestinal lymph nodes and sections of the duodenum, jejunum and ileum, the pigs received an intramuscular injection of saline without or with 150 ,g/kg body weight of Escherichia coli O26:B6 lipopolysaccharide (LPS). With the exception of a tendency (p = 0.09) for lower serum concentration of copper in pigs at weaning from ZnAA-supplemented sows, there were no differences (p > 0.1) than for pigs from control-fed sows for mineral status or intestinal morphology. Nutriment-intubation of ZnMet increased serum (p = 0.001) and liver (p = 0.003) Zn concentrations, number of goblet cells per 250 ,m length of jejunal villous epithelium (p = 0.001) and tended (p = 0.06) to enhance jejunum mucosa thickness. Interactive effects (p < 0.05) for higher jejunal villi height and villi:crypt ratio and increased ileal goblet cell counts were apparent for pigs from ZnAA-supplemented sows that also received nutriment-intubation of ZnMet. Challenge with LPS increased (p = 0.05) ileal villous width. Nutriment-intubation of ZnMet decreased (p = 0.05) anaerobic bacteria colony forming unit counts in the large intestinal mesenteric lymph nodes. In conclusion, nutriment-intubation of ZnMet increased serum and liver tissue concentrations of Zn and resulted in limited improvement to intestinal morphology of weaned pigs. [source]


    Expression of Progesterone Receptor in the Utero-tubal Junction After Intra-uterine and Deep Intra-uterine Insemination in Sows

    Contents The aim of this study was to investigate the expression of progesterone receptor (PR) in the utero-tubal junction (UTJ) of sows at 24 h after intra-uterine insemination (IUI) and deep intra-uterine insemination (DIUI) compared with conventional artificial insemination (AI) in pigs. Fifteen multiparous sows were used: AI (n = 5), IUI (n = 5) and DIUI (n = 5). The sows were inseminated with a single dose of diluted semen during the second oestrus after weaning at 6,8 h prior to ovulation (AI: 3000 × 106 spermatozoa, IUI: 1000 × 106 spermatozoa and DIUI: 150 × 106 spermatozoa). The UTJ was collected and subject to immunohistochemical staining using avidin-biotin immunoperoxidase technique with mouse monoclonal antibody to PR. In the oviductal part of the UTJ, the intensity of PR in the tunica muscularis and the proportion of PR-positive cells in the surface epithelium after DIUI were lower than AI (p < 0.05). The intensity and the proportion of PR-positive cells between AI and IUI in all compartments of the UTJ did not differ significantly (p > 0.05). When comparing between tissue compartments, prominent staining was observed in the muscular layer of the UTJ. It could be concluded that the expression of PR in the UTJ prior to fertilization after DIUI with a reduced number of spermatozoa was lower than that after AI. This might influence sperm transportation and the fertilization process. [source]


    Number of Spermatozoa in the Crypts of the Sperm Reservoir at About 24 h After a Low-Dose Intrauterine and Deep Intrauterine Insemination in Sows

    Contents The aim of this study was to investigate the number of spermatozoa in the crypts of the utero-tubal junction (UTJ) and the oviduct of sows approximately 24 h after intrauterine insemination (IUI) and deep intrauterine insemination (DIUI) and compared with that of conventional artificial insemination (AI). Fifteen crossbred Landrace × Yorkshire (LY) multiparous sows were used in the experiment. Transrectal ultrasonography was performed every 4 h to examine the time of ovulation in relation to oestrous behaviour. The sows were inseminated with a single dose of diluted fresh semen by the AI (n = 5), IUI (n = 5) and DIUI (n = 5) at approximately 6,8 h prior to the expected time of ovulation, during the second oestrus after weaning. The sperm dose contained 3000 × 106 spermatozoa in 100 ml for AI, 1,000 × 106 spermatozoa in 50 ml for IUI and 150 × 106 spermatozoa in 5 ml for DIUI. The sows were anaesthetized and ovario-hysterectomized approximately 24 h after insemination. The oviducts and the proximal part of the uterine horns (1 cm) on each side of the reproductive tracts were collected. The section was divided into four parts, i.e. UTJ, caudal isthmus, cranial isthmus and ampulla. The spermatozoa in the lumen in each part were flushed several times with phosphate buffer solution. After flushing, the UTJ and all parts of the oviducts were immersed in a 10% neutral buffered formalin solution. The UTJ and each part of the oviducts were cut into four equal parts and embedded in a paraffin block. The tissue sections were transversely sectioned to a thickness of 5 ,m. Every fifth serial section was mounted and stained with haematoxylin and eosin. The total number of spermatozoa from 32 sections in each parts of the tissue (16 sections from the left side and 16 sections from the right side) was determined under light microscope. The results reveal that most of the spermatozoa in the histological section were located in groups in the epithelial crypts. The means of the total number of spermatozoa in the sperm reservoir (UTJ and caudal isthmus) were 2296, 729 and 22 cells in AI, IUI and DIUI groups, respectively (p < 0.01). The spermatozoa were found on both sides of the sperm reservoir in all sows in the AI and the IUI groups. For the DIUI group, spermatozoa were not found on any side of the sperm reservoir in three out of five sows, found in unilateral side of the sperm reservoir in one sow and found in both sides of the sperm reservoir in one sow. No spermatozoa were found in the cranial isthmus, while only one spermatozoon was found in the ampulla part of a sow in the IUI group. In conclusion, DIUI resulted in a significantly lower number of spermatozoa in the sperm reservoir approximately 24 h after insemination compared with AI and IUI. Spermatozoa could be obtained from both sides of the sperm reservoir after AI and IUI but in one out of five sows inseminated by DIUI. [source]


    Effect of Weaning to Oestrus Interval and Equine Chorionic Gonadotropin on Vaginal Electrical Impedance During Peri-oestrus in Sows

    Contents The influence of weaning to oestrus interval, its interaction with parity and equine chorionic gonadotropin (eCG) on changes of vaginal impedance in sows after weaning was examined. The impedance measurements were carried out by a four-terminal method. Sows were monitored for oestrus via exposure to a sexually mature boar. The interval from weaning to oestrus was longer in primiparous than multiparous sows (p < 0.01). A significant negative correlation was found between the interval from weaning to oestrus and parity. Repeated measures analysis showed that the interval from weaning to oestrus and parity and their interactions had a significant effect on the vaginal impedance in peri-oestrus. The vaginal impedance during pro-oestrus gradually decreased in all groups of sows with the weaning to oestrus interval from 4 to 8 days (p < 0.05). In the subsequent period, the vaginal impedance increased and was significantly lower from 1 to 3 days after oestrus onset in sows with the weaning to oestrus interval 7,8 days than 4,6 days. Similarly, the vaginal impedance during pro-oestrus gradually decreased in all groups of sows with parity 1,5 (p < 0.01). In the next period, the vaginal impedance increased and was significantly lower from 2,3 days after oestrus onset in sows of parity 1 than parity 2,5. Repeated measures analysis showed that eCG treatment had a significant effect on the vaginal impedance in peri-oestrus. Sows treated with eCG displayed the decrease and increase of vaginal impedance due to oestrus onset earlier than untreated sows. The results indicate that the weaning to oestrus interval, its interaction with parity and eCG markedly affect the vaginal impedance in sows during peri-oestrus. [source]


    Factors Affecting Fertility in Loosely Housed Sows and Gilts: Vulvar Discharge Syndrome, Environment and Acute-phase Proteins

    Contents The effect of vulvar discharge syndrome (VDS) on sow and gilt fertility was studied on 26 farms. Of 824 animals inspected in 21 randomly selected and five VDS problem farms, 19 (2.3%) were afflicted with VDS. Altogether 542/799 of the examined animals (67.8%) farrowed thereafter. Nine of the 19 VDS animals (47.4%) and 533/780 non-VDS animals (68.3%) farrowed at the first chance after the examination (p = 0.05). None of the unmated gilts in this study had VDS. Environmental and individual factors likely to be associated with fertility and VDS were tested. In multivariate analysis, factors associated with farrowing were VDS, reproductive status, availability of roughage and confinement to individual stalls. None of the variables tested was associated with VDS. However, all of the VDS problem farms were overcrowded and had concrete, partly slatted floors with little or no bedding. The median value of haptoglobin (Hp) was 2.5 (range 1.3,3.1) g/l in VDS animals and 2.3 (0.5,4.3) g/l in controls (p = 0.6). The median C-reactive protein (CRP) in VDS animals was 30.3 (3.3,171.3) mg/l and in controls 25.9 (3.3,361.1) mg/l (p = 0.7). In conclusion, VDS decreased fertility of gilts and sows in the absence of a systemic acute-phase response, as indicated by stable concentrations of Hp and CRP. [source]


    Postovulatory Effect of Intravenous Administration of Lipopolysaccharide (E. coli, O55:B5) on the Contractile Activity of the Oviduct, Ova Transport, Binding of Accessory Spermatozoa to the Zona Pellucida and Embryo Development in Sows

    Contents The effect of lipopolysaccharide (LPS) (E. coli, O55:B5), administered 18 h after ovulation in the second oestrus after weaning, on the contractile activity of the oviduct, ova transport, sperm binding to zona pellucida (ZP) and embryo development, was studied in 14 Swedish crossbred (Landrace Yorkshire) multiparous sows. The endotoxin group (E-group) sows were administered with 300 ng/kg of LPS while the control group (C-group) sows were administered with 5 ml of saline i.v. via an indwelling jugular cannula. Immediately after evidence of standing oestrus, a Millar® pressure transducer was placed intraluminally about 3 cm into the mid-isthmus, via laparotomy. Pressure recordings of the oviduct were collected from all conscious sows until slaughter. After slaughter, the genital tract opposite to the side with the transducer was retrieved, and three equal isthmic segments and the first third of the uterine horn part adjacent to the utero-tubal-junction (UTJ) were flushed separately to recover the ova. The intervals (mean±SD) from ovulation to slaughter (OS) and insemination to ovulation (IO) were not different between the E-group (44.5±5.7 h; 13.3±6.5 h) and the C-group (42.7±5.9 h; 14.8±4.1 h), respectively. Ova recovery rate (RR) in the E-group (80.2±22.9%) did not differ from that in the C-group (85.2±4.5%). The frequency distribution of ova recovered in the different segments did not significantly (p>0.05) differ between the groups. The E-group showed higher cleavage rate than controls. A higher proportion of spermatozoa bound to the ZP was also found in the E-group compared with controls. The isthmic intraluminal pressure slightly increased (p=0.07) 18 h after ovulation and immediately following LPS in the E-group, compared with the C-group. The frequencies of phasic pressure fluctuations were significantly (p<0.05) lower at 30 and 38 h after ovulation in the E- than in the C-group. It can be concluded from the present study that a single i.v. administration of LPS (300 ng/kg body weight) to sows, 18 h after ovulation might be associated with changes in isthmic pressure and the frequency of phasic pressure fluctuations, increased numbers of spermatozoa attached to the ZP and an enhanced embryo development but not with ova transport rates.     Influence of different phosphorus levels and phytase supplementation in gestation diets on sow performance

    Summary A total of 104 sows of different parities were studied. They were fed four diets with different phosphorus (P) levels during gestation for two reproductive cycles, while the same diet was fed during lactation. The aim was to decrease the total P level in the diet during gestation and to evaluate the effect on sow performance. The gestation treatments were low P (LP,; 3.7 g P/kg feed), low P with phytase (LP+, Ronozyme® P; 765 FTU/kg feed), medium P (MP; 4.5 g P/kg feed) and high P (HP; 6.0 g P/kg feed). Daily feed allowances were 2.6 kg during gestation and 9.2 kg during lactation. Number of born piglets and piglet mortality were higher (p < 0.05) in the LP treatments than in the MP and HP treatments. No difference (p > 0.05) in the numbers of live-born piglets, piglet birthweights, sow weights or piglet weight gains was found between the treatments. Phosphorus level in sow milk was the highest (p < 0.05) in the MP treatment, while no effects (p > 0.05) of treatment were found on milk Ca levels, P and Ca levels in serum of sows and piglets, nor on the analysed mineral, fat and protein contents of piglets. The estimated average requirement of P for the entire gestation period was 4.4,4.5 g/day. In conclusion, a reduction of dietary total P content during gestation did not result in negative effects on sow or piglet performance. This suggests that it should be possible to lower the dietary P content for gestating sows, compared with earlier recommendations, and thereby reduce the environmental P pollution. [source]


    Placental transfer and pharmacokinetics of allopurinol in late pregnant sows and their fetuses

    Xanthine oxidoreductase (XOR) is a key enzyme in the evolvement of reperfusion injury resulting from birth asphyxia, a common cause of decreased viability and perinatal mortality in newborn piglets under farm conditions. At present no standard pharmacological intervention strategy is available to reduce these adverse effects of birth asphyxia. In the present study we aimed to evaluate placental transfer of allopurinol, an inhibitor of XOR. For this purpose, fetal catheterization of the jugular vein was conducted in five late pregnant sows (one fetus per sow). At 24,48 h after surgery, sows received allopurinol (15 mg/kg body weight; i.v.) and pharmacokinetics of allopurinol and its active metabolite oxypurinol were measured in both late pregnant sows and fetuses. Maternal and fetal blood samples were collected during and after allopurinol administration. Maternal Cmax values averaged 41.90 ,g/mL (allopurinol) and 3.68 ,g/mL (oxypurinol). Allopurinol crossed the placental barrier as shown by the average fetal Cmax values of 5.05 ,g/mL at 1.47 h after allopurinol administration to the sow. In only one fetus low plasma oxypurinol concentrations were found. Incubations of subcellular hepatic fractions of sows and 24-h-old piglets confirmed that allopurinol could be metabolized into oxypurinol. In conclusion, we demonstrated that allopurinol can cross the placental barrier, a prerequisite for further studies evaluating the use of allopurinol as a neuroprotective agent to reduce the adverse effects following birth asphyxia in neonatal piglets.     Contents The aim of this study was to investigate the number of spermatozoa in the crypts of the utero-tubal junction (UTJ) and the oviduct of sows approximately 24 h after intrauterine insemination (IUI) and deep intrauterine insemination (DIUI) and compared with that of conventional artificial insemination (AI). Fifteen crossbred Landrace × Yorkshire (LY) multiparous sows were used in the experiment. Transrectal ultrasonography was performed every 4 h to examine the time of ovulation in relation to oestrous behaviour. The sows were inseminated with a single dose of diluted fresh semen by the AI (n = 5), IUI (n = 5) and DIUI (n = 5) at approximately 6,8 h prior to the expected time of ovulation, during the second oestrus after weaning. The sperm dose contained 3000 × 106 spermatozoa in 100 ml for AI, 1,000 × 106 spermatozoa in 50 ml for IUI and 150 × 106 spermatozoa in 5 ml for DIUI. The sows were anaesthetized and ovario-hysterectomized approximately 24 h after insemination. The oviducts and the proximal part of the uterine horns (1 cm) on each side of the reproductive tracts were collected. The section was divided into four parts, i.e. UTJ, caudal isthmus, cranial isthmus and ampulla. The spermatozoa in the lumen in each part were flushed several times with phosphate buffer solution. After flushing, the UTJ and all parts of the oviducts were immersed in a 10% neutral buffered formalin solution. The UTJ and each part of the oviducts were cut into four equal parts and embedded in a paraffin block. The tissue sections were transversely sectioned to a thickness of 5 ,m. Every fifth serial section was mounted and stained with haematoxylin and eosin. The total number of spermatozoa from 32 sections in each parts of the tissue (16 sections from the left side and 16 sections from the right side) was determined under light microscope. The results reveal that most of the spermatozoa in the histological section were located in groups in the epithelial crypts. The means of the total number of spermatozoa in the sperm reservoir (UTJ and caudal isthmus) were 2296, 729 and 22 cells in AI, IUI and DIUI groups, respectively (p < 0.01). The spermatozoa were found on both sides of the sperm reservoir in all sows in the AI and the IUI groups. For the DIUI group, spermatozoa were not found on any side of the sperm reservoir in three out of five sows, found in unilateral side of the sperm reservoir in one sow and found in both sides of the sperm reservoir in one sow. No spermatozoa were found in the cranial isthmus, while only one spermatozoon was found in the ampulla part of a sow in the IUI group. In conclusion, DIUI resulted in a significantly lower number of spermatozoa in the sperm reservoir approximately 24 h after insemination compared with AI and IUI. Spermatozoa could be obtained from both sides of the sperm reservoir after AI and IUI but in one out of five sows inseminated by DIUI. [source]


    Welfare Index and Reproductive Performance in the Sow

    Contents To study the relationship between on-farm welfare and reproductive performance in the sow, the TGI 35L Animal Needs Index was modified for use in Finnish pig production. The modified index had a maximal total score of 100. It was comprised of six categories: ,locomotion' (maximal score 21 for dry sows and 11 for lactating sows), ,social interaction' (12/8), ,floor quality' (16/9), ,stable climate' (16/21), ,feeding' (16/23) and ,health and stockmanship' (19/29). Index scorings were performed separately in farrowing, breeding and gestation units on 28 representative Finnish sow farms. One-year production parameters were collected. Multiple linear regression was used for statistical analysis. Total ANI-points varied between 36.5,68.0 for lactating and 39.5,86.0 for dry sows. Litter size increased with increasing scores for ,feeding' in the dry sow unit. Controlling for breed, high scores for ,health and stockmanship' and ,floor quality' shortened the reproductive cycle, probably because of good leg health. The number of weaned piglets per sow per year (PSY) increased with increasing scores for dry sow ,health and stockmanship', ,floor quality' and an interaction of ,feeding' in the farrowing and mating units. PSY increased with decreasing scores for farrowing pen ,climate'. High-quality floors and stockmanship appear to correlate positively with reproductive performance in the sow. Effects of a welfare-promoting feeding strategy on reproduction are contradictory. [source]


    Factors Affecting Fertility in Loosely Housed Sows and Gilts: Vulvar Discharge Syndrome, Environment and Acute-phase Proteins

    Contents The effect of vulvar discharge syndrome (VDS) on sow and gilt fertility was studied on 26 farms. Of 824 animals inspected in 21 randomly selected and five VDS problem farms, 19 (2.3%) were afflicted with VDS. Altogether 542/799 of the examined animals (67.8%) farrowed thereafter. Nine of the 19 VDS animals (47.4%) and 533/780 non-VDS animals (68.3%) farrowed at the first chance after the examination (p = 0.05). None of the unmated gilts in this study had VDS. Environmental and individual factors likely to be associated with fertility and VDS were tested. In multivariate analysis, factors associated with farrowing were VDS, reproductive status, availability of roughage and confinement to individual stalls. None of the variables tested was associated with VDS. However, all of the VDS problem farms were overcrowded and had concrete, partly slatted floors with little or no bedding. The median value of haptoglobin (Hp) was 2.5 (range 1.3,3.1) g/l in VDS animals and 2.3 (0.5,4.3) g/l in controls (p = 0.6). The median C-reactive protein (CRP) in VDS animals was 30.3 (3.3,171.3) mg/l and in controls 25.9 (3.3,361.1) mg/l (p = 0.7). In conclusion, VDS decreased fertility of gilts and sows in the absence of a systemic acute-phase response, as indicated by stable concentrations of Hp and CRP. [source]


    The Peritoneal Mesothelium Covering the Genital Tract and its Ligaments in the Female Pig Shows Signs of Active Function

    Abstract The aim of this study was to describe the surface features of the peritoneal mesothelium covering the genital tract and adjacent ligaments of the sow to find signs of biosynthetic activation of cells. Surface features of the serosa covering the genital tract and adjacent ligaments from 14 cyclic sows, 7 in the follicular phase and 7 in the luteal phase of the estrous cycle, were examined by histology and scanning electron microscopy. Five additional sows, three in the follicular phase and two in the luteal phase of the estrous cycle, were examined by transmission electron microscopy (TEM). In this study, the presence of cells of the oviductal epithelium in the serosa of the infundibulum and the ampulla, as well as indications of a high functional activity of the mesothelial cells in the areas studied were two aspects that differed from the findings of previous works. Presence of endosalpingeal cells was observed in the serosal surface, showing cyclical variations with a predominance of either ciliated cells during the follicular phase or secretory cells during the luteal phase. Signs of high functional activity of the mesothelial cells included the predominance of cuboidal over flattened cells, a cytoplasm richly supplied with organelles, a dense microvillous coat, numerous primary cilia, and many secretory structures on the surface of cells. These results indicate that the serosa covering the genital area and the adjacent ligaments in the sow has an active epithelium whose coordinating role between reproductive tissues may be far more significant than previously thought.     Indirect effect of IGF2 intron3 g.3072G>A mutation on prolificacy in sows

    Summary A QTL located in the paternally expressed insulin-like growth factor 2 (IGF2) gene is known to increase muscle growth and reduce fat deposition in pigs. This makes the QTL in IGF2 a good marker for use in pig breeding programmes. However, care has to be taken as it is postulated that increased leanness and lowered fat deposition may have a negative effect on the prolificacy and longevity of sows. Selection of sire and dam lines for different alleles of the mutation in the paternally imprinted IGF2 gene could actually provide a solution to this problem. Therefore, in this study, the effect of the IGF2 QTL on prolificacy-related traits in sows was investigated. It was found that the paternal IGF2 wild-type allele was associated with higher reproduction performance in the sow. Moreover, it was also examined whether the difference in prolificacy in sows could be a consequence of differential IGF2 expression in the ovarian follicles of the sow or whether it is mainly a secondary effect caused by differences in fatness traits. Therefore, IGF2 expression was measured in follicles of different sizes from sows with different genotypes for the paternal IGF2 allele. It was observed that, however, while the size of the follicles was associated with follicular IGF2 expression level, the IGF2 genotype was not. It could be concluded that the difference in prolificacy of sows with a different paternal IGF2 genotype could be a secondary effect, resulting from differences in fat deposition. [source]


    Nursing Synchronization and Milk Ejection Failure as Maternal Strategies to Reduce Allosuckling in Pair-Housed Sows (Sus scrofa domestica)

    Non-offspring nursing (allosuckling) is costly for lactating females. It is reasonable to assume, therefore, that mechanisms have evolved to control the frequency of allosuckling. It is suggested that the synchronization of nursing between females could be such a mechanism. The aim of this study was to investigate whether nursing synchronization is related to the timing of nursing behaviour of paired sows and the behaviour of alien piglets in relation to allosuckling. Specifically, we predicted that: (i) nursing synchronization will increase if there is a real risk of allosuckling compared with a situation where there is none, (ii) nursing synchronization indeed reduces allosuckling and, therefore, those nursings with allosuckling attempts will be less closely synchronized than those without allosuckling attempts and (iii) in non-synchronized nursings, the sows will more often employ another strategy to reduce allosuckling, namely, they will make the nursings non-nutritive by not releasing milk. Ten pairs of sows and their litters were used in this study. For each pair, the two sows were housed individually until their litters were weaned; however, their litters had access to both sows from day 11 post-partum. The behaviour of the 10 pairs of sows with their litters was videotaped for 6 h on days 10, 11, 17 and 24 post-partum. The frequency of allosuckling attempts, non-nutritive nursings and allosuckling during milk ejection was analysed for every pair for every day of observation. The observed frequency of nursing synchronization was high. The median interval between the start of a nursing within one pair of sows was 32 s. The first prediction was supported as synchronization became significantly tighter when piglets were given the possibility to allosuckle on day 11 [Cochran,Mantel,Haenszel (CMH) statistics, p < 0.0001]. The second prediction was also supported; the nursings were less closely synchronized when there were allosuckling attempts (CMH statistics, p < 0.05). Of all nursings, 36% had an allosuckling attempt; however, only 17% resulted in real allosuckling. Evidence in favour of the third prediction was also found as non-nutritive nursings were more frequent in non-synchronized nursings (CMH statistics, p < 0.05). Finally, real allosuckling during milk ejection was unrelated to the degree of nursing synchronization, as in less synchronized nursings, the sows responded to allosuckling attempts by not releasing milk. Our results suggest that sows are not able to completely suppress allosuckling; however, they can reduce its frequency through nursing synchronization and non-nutritive nursings. [source]


    Characterization of the porcine Kisspeptins receptor gene and evaluation as candidate for timing of puberty in sows

    Summary Kisspeptins receptor (KISS1R), also called GPR54, is a key regulator of puberty in many species. KISS1R and its genetics in pigs remain unexplored. The objective of this study was to characterize the porcine KISS1R gene and evaluate the association of KISS1R mutations with age at puberty in sows. KISS1R was assigned to pig chromosome 2q21-24 by radiation hybrid mapping. It has a 1438 bp full-length cDNA and spans 3349 bp genomic sequence consisting of five exons and four introns. Semi-quantitative RT-PCR showed that KISS1R transcripts was particularly abundant in the adrenal, prostate, testis, thymus, pituary and hypothalamus. KISS1R mRNA content in the hypothalamus was determined by real-time quantitative RT-PCR, and it fluctuated during the oestrous cycle with the highest level in the luteal phase. Anoestrus sows had markedly lower hypothalamic KISS1R mRNA content than cyclic animals. Seven KISS1R SNPs were identified in the founder animals of a White Duroc × Erhualian intercross. One missense mutation (T/C245) showed quite different allele distribution in Chinese and Western breeds. All F0, F1 animals and 367 detailed phenotyped cyclic F2 sows in the White Duroc × Erhualian intercross were genotyped for three KISS1R polymorphisms. No significant association of KISS1R haplotypes and haplotype pairs with age at puberty was observed in the resource population, indicating that mutations in KISS1R are not responsible for divergent age at puberty in White Duroc and Erhualian pigs.     Fixed or random contemporary groups in genetic evaluation for litter size in pigs using a single trait repeatability animal model

    Summary The importance of using fixed or random contemporary groups in the genetic evaluation of litter size in pigs was analysed by using farm and simulated data. Farm data were from four Spanish pig breeding populations, two Landrace (13 084 and 13 619 records) and two Large White (2762 and 8455 records). A simulated population (200 sows and 10 boars) selected for litter size, in which litter size was simulated using a repeatability animal model with random herd,year,season (HYS), was used to obtain simulated data. With farm data, the goodness-of-fit and the predictive ability of a repeatability animal model were calculated under several definitions of the HYS effect. A residual maximum likelihood estimator of the HYS variance in each population was obtained as well. In this sense, HYS was considered as either fixed or random with different number of individuals per level. Results from farm data showed that HYS variance was small in relation to the total variance (ranging from 0.01 to 0.04). The treatment of HYS effect as fixed, reduced the residual variance but the size of HYS levels does not explain by itself the goodness-of-fit of the model. The results obtained by simulation showed that the predictive ability of the model is better for random than for fixed HYS models. However, the improvement of predictive ability does not lead to a significant increase of the genetic response. Finally, results showed that random HYS models biased the estimates of genetic response when there is an environmental trend. Zusammenfassung Fixe oder zufällige Vergleichsgruppen bei der Zuchtwertschätzung für Wurfgröße beim Schwein mit einem Wiederholbarkeits-Tiermodell Der Einfluss von fixen oder zufälligen Vergleichgruppen bei der Zuchtwertschätzung für Wurfgröße beim Schwein wurde an realen Betriebsdaten und an simulierten Daten untersucht. Die Betriebsdaten stammen von vier spanischen Zuchtpopulationen, zwei Landrasse Populationen (13084 und 13619 Datensätze) und zwei Large White Populationen (2762 und 8455 Datensätze). Für die Simulation wurde eine Population (200 Sauen und 10 Eber), die auf Wurfgröße selektiert wurde, unter Berücksichtigung eines Wiederholbarkeitsmodelles und mit zufälligen Herden-Jahr-Saisonklassen simuliert. Anhand der Betriebsdaten wurde die Güte des Modells und die Vorhersagegenauigkeit des Wiederholbarkeitsmodelles mit verschiedenen Definitionen der Herden-Jahr-Saisonklassen geprüft. Mittels der REML-Methode wurden auch Varianzkomponenten für die Herden-Jahr-Saisonklassen geschätzt. Die Herden-Jahr-Saisonklassen wurden als fixer bzw. zufälliger Effekt mit unterschiedlicher Anzahl an Tieren pro Klasse im Modell berücksichtigt. Die Ergebnisse der Betriebsdaten ergaben, dass die Varianz für die Herden-Jahr-Saisonklassen nur einen kleinen Teil der Totalvarianz (von 0,01 bis 0,04) ausmachte. Mit den Herden-Jahr-Saisonklassen als fixer Effekt reduzierte sich die Restvarianz, aber die Größe der Herden-Jahr-Saisonklassen bestimmte nicht allein die Güte des Modells. Die Erhöhung der Vorhersagegenauigkeit ergab keinen signifikanten Anstieg des genetischen Fortschrittes. Abschließend bleibt festzustellen, dass Modelle mit zufälligen Herde-Jahr-Saisonklassen zu einem Bias des geschätzten genetischen Erfolges führten, wenn ein Umwelttrend vorhanden war. [source]


    A new HpaII PCR-RFLP within the porcine prolactin receptor (PRLR) gene and study of its effect on litter size and number of teats

    DNA polymorphism of the porcine prolactin receptor gene (PRLR) was investigated and used to study its effect on litter size and number of teats in pigs. By means of PRLR gene sequence homology in pig, human and other species, primers were designed for PCR amplification within 5, unknown (to date) part of the prolactin receptor gene in pigs. In this part of the gene, a new polymorphism with HpaII restriction endonuclease was detected. AluI polymorphism described before and our new HpaII polymorphism were used to study the associations with reproduction traits. The PCR restriction fragment length polymorphism (PCR-RFLP) method was used to genotype AluI and HpaII loci of the PRLR gene in line A with 83 sows of Landrace breed and in two lines (B and C) with 75 and 86 Large White sows, respectively. Statistical analysis of 1020 litters showed that AluI locus was associated with litter size mainly in Landrace and affected the first parities, while HpaII locus of the gene was associated with the same traits in Landrace and Large White pigs and mainly affected numbers of weaned of pigs. The magnitude of the effect varied by population with the effects exceeding two pigs per litter in Landrace line and 1 pig per litter in Large White populations. Ein neuer HpaII PCR-RFLP innerhalb des porcinen Prolaktionrezeptorgens (PRLR), und Zusammenhänge zur Wurfgröße und Zitzenzahl DNA-Polymorphismen im porcinen Prolaktionrezeptorgen (PRLR) wurden untersucht und für die Analyse von Einflüssen auf Wurfgröße und Zitzenzahl bei Schweinen verwendet. Auf der Basis der PRLR -Gensequenzhomologie zwischen Schwein, Mensch und anderen Spezies wurden Primer für die PCR-Amplifikation aus dem 5, Bereich des Prolaktionrezeptorgens abgeleitet, der bisher beim Schwein noch unbekannt ist. In diesem Teil des Gens wurde mittels HpaII-Restriktionsendonuklease ein neuer Polymorphismus dargestellt. AluI Polymorphismus und der neue HpaII Polymorphismus wurden für Assoziationsstudien in Bezug auf Reproduktionsmerkmale verwendet. Mittels PCR-RFLP wurden in Linie A 83 Sauen der Landrasse und die Linien B und C mit 75 bzw. 86 Large White Sauen unter Verwendung von AluI und HpaII am PRLR -Gen genotypisiert. Die statistische Analyse von 1.020 Würfen zeigte, dass der AluI-Polymorphismus insbesondere in der Landrasse mit der Wurfgröße assoziiert ist, sowie die ersten Trächtigkeiten beeinflusst, während der HpaII Polymorphismus die gleichen Merkmale in der Landrasse und Large White Schweinen und insbesondere die Zahl an abgesetzten Ferkeln beeinflusste. Die Auswirkungen des Effekts variierten innerhalb Population, wobei der Effekt 2 Ferkel je Wurf in der Landrasse-Linie und 1 Ferkel je Wurf in der Large White Populationen überstieg. [source]


    Comparative study on the consequences of culling badgers (Meles meles) on biometrics, population dynamics and movement

    1.,Capture,mark,recapture data were used to describe the process of recovery from a typical badger removal operation (BRO) at North Nibley, Gloucestershire, UK, which was carried out as part of the government's strategy to control bovine tuberculosis. Data on biometrics, demographics and movement from this low-density disturbed population were compared with those of two nearby high-density undisturbed populations (Wytham Woods and Woodchester Park, UK) in order to study fundamental principles of population dynamics and density-dependence. 2.,Badgers moved more between social groups at North Nibley than in the other study areas, particularly in the immediate aftermath of the removal operation. 3.,Recolonization of the vacated habitat occurred in the first instance by young females. 4.,Although in the first year after the BRO no cubs had been reared in any of the culled groups, and although the shortage of sexually mature boars may have limited the reproductive output of sows in the following year, the population took only 3 years to recover to its (already lowered) preremoval density. 5.,Losses from the adult (and cub) population due to mortality or emigration were smaller at North Nibley than at the other sites. 6.,There was much evidence that during 1995 and 1996 density-dependent effects constrained the reproductive output of the high-density populations, and some support for the hypothesis that badgers exhibit the non-linear ,large mammal' type of functional response to density. 7.,Badgers at North Nibley were younger, heavier and in better condition than badgers at Wytham Woods and Woodchester Park. 8.,We argue that the disease dynamics are likely to be different in disturbed compared with undisturbed badger populations, and that this could affect the effectiveness of BROs.     Supplementation of diets for lactating sows with zinc amino acid complex and gastric nutriment-intubation of suckling pigs with zinc methionine on mineral status, intestinal morphology and bacterial translocation in lipopolysaccharide-challenged weaned pigs

    Summary Sixty-four pigs from 16 sows were used to evaluate addition of zinc amino acid complex (ZnAA) to lactating sows and gastric nutriment-intubation of zinc methionine (ZnMet) to suckling pigs on mineral status, intestinal morphology and bacterial translocation after weaning. Sows were fed a barley-based diet supplying 120 ppm zinc (Zn; control) or the control diet supplemented with 240 ppm Zn from ZnAA. At birth, day-10 and day-21 (weaning) of age, pigs from each litter were nutriment-intubated with 5 ml of an electrolyte solution without or with 40 mg Zn from ZnMet. At weaning, 24 h prior to the collection of small and large intestinal lymph nodes and sections of the duodenum, jejunum and ileum, the pigs received an intramuscular injection of saline without or with 150 ,g/kg body weight of Escherichia coli O26:B6 lipopolysaccharide (LPS). With the exception of a tendency (p = 0.09) for lower serum concentration of copper in pigs at weaning from ZnAA-supplemented sows, there were no differences (p > 0.1) than for pigs from control-fed sows for mineral status or intestinal morphology. Nutriment-intubation of ZnMet increased serum (p = 0.001) and liver (p = 0.003) Zn concentrations, number of goblet cells per 250 ,m length of jejunal villous epithelium (p = 0.001) and tended (p = 0.06) to enhance jejunum mucosa thickness. Interactive effects (p < 0.05) for higher jejunal villi height and villi:crypt ratio and increased ileal goblet cell counts were apparent for pigs from ZnAA-supplemented sows that also received nutriment-intubation of ZnMet. Challenge with LPS increased (p = 0.05) ileal villous width. Nutriment-intubation of ZnMet decreased (p = 0.05) anaerobic bacteria colony forming unit counts in the large intestinal mesenteric lymph nodes. In conclusion, nutriment-intubation of ZnMet increased serum and liver tissue concentrations of Zn and resulted in limited improvement to intestinal morphology of weaned pigs. [source]


    Influence of different phosphorus levels and phytase supplementation in gestation diets on sow performance

    Summary A total of 104 sows of different parities were studied. They were fed four diets with different phosphorus (P) levels during gestation for two reproductive cycles, while the same diet was fed during lactation. The aim was to decrease the total P level in the diet during gestation and to evaluate the effect on sow performance. The gestation treatments were low P (LP,; 3.7 g P/kg feed), low P with phytase (LP+, Ronozyme® P; 765 FTU/kg feed), medium P (MP; 4.5 g P/kg feed) and high P (HP; 6.0 g P/kg feed). Daily feed allowances were 2.6 kg during gestation and 9.2 kg during lactation. Number of born piglets and piglet mortality were higher (p < 0.05) in the LP treatments than in the MP and HP treatments. No difference (p > 0.05) in the numbers of live-born piglets, piglet birthweights, sow weights or piglet weight gains was found between the treatments. Phosphorus level in sow milk was the highest (p < 0.05) in the MP treatment, while no effects (p > 0.05) of treatment were found on milk Ca levels, P and Ca levels in serum of sows and piglets, nor on the analysed mineral, fat and protein contents of piglets. The estimated average requirement of P for the entire gestation period was 4.4,4.5 g/day. In conclusion, a reduction of dietary total P content during gestation did not result in negative effects on sow or piglet performance. This suggests that it should be possible to lower the dietary P content for gestating sows, compared with earlier recommendations, and thereby reduce the environmental P pollution. [source]


    Field evaluation of the efficacy of a probiotic containing Bacillus licheniformis and Bacillus subtilis spores, on the health status and performance of sows and their litters

    Summary The aim of this study was to assess the efficacy of BioPlus 2B, a probiotic containing Bacillus licheniformis and Bacillus subtilis spores, on the health status and productivity of sows and their litters. A total of 109 gilts and sows were allocated into two experimental groups, as follows: untreated controls (UC) and BioPlus 2B (same feeding as the UC group plus BioPlus 2B) at a dose of 400 g/ton of feed (equal to 1.28 × 106 viable spores/g of feed). Treatment started from the day of allocation (14 days prior to the expected farrowing) up to the weaning day. Homogeneity of the groups was satisfied with regard to the parity. From the results it was evident that BioPlus 2B supplementation of the feed improved gilt/sow performance as shown by: (i) the increase of sow feed consumption during the first 14 days postpartum and (ii) the decrease of sow weight loss during the suckling period. Certain blood and milk parameters were significantly improved, as shown by higher serum cholesterol and total lipids concentrations and higher milk fat and protein content at mid-suckling period. As a consequence, a positive effect was also noticed as regard litter health and performance characteristics in terms of: (i) decrease in piglet diarrhoea score, (ii) decrease in pre-weaning mortality thus leading to increase in the number of weaned piglets per litter and (iii) increase in piglet body weight at weaning. Moreover, BioPlus 2B tended to improve the health status and fertility of sows demonstrating: (i) tendency to a lower proportion of sows with Mastitis-Metritus-Agalactia (MMA) problems and (ii) lower proportion of sows returning to oestrus.     Effect of L -carnitine supplementation on performance parameters in gilts and sows

    The effect of L-carnitine supplementation during pregnancy and lactation on performance parameters of sows was studied. The trial comprised a total of 127 sows (40 gilts, 87 mature sows) which were divided into a control and a treatment group. All animals were fed individually and received basic feed mixtures for pregnancy and lactation with low carnitine concentrations (gestation diet: 4.7 mg/kg feed, lactation diet: 12.5 mg/kg feed). The rations of the sows in the treated group were supplemented with 125 mg L -carnitine per head and day during pregnancy and 250 mg L -carnitine per head and day during lactation. The animals of the control group received identical feed mixtures in identical amounts, but without the L -carnitine supplement. L -carnitine supplementation resulted in higher sow liveweight gains between day 1 and day 85 of pregnancy. The number of piglets per litter and the number born alive did not differ between the control sows and those treated with L -carnitine. However, the L -carnitine-supplemented sows produced only half as many non-viable piglets as the control animals. Moreover, litter weight and mean birth weight of piglets from L -carnitine-treated sows were higher than in the control sows. This effect was more marked in gilts (+8% higher litter weight, +9% higher piglet weight) than in sows (+7% and +6%, respectively). Piglets from sows whose ration was supplemented with L -carnitine showed higher liveweight gains during the suckling period (+12% for gilts, +4% for sows), which is why litter weights post weaning were also higher among the sows treated with L -carnitine than in the control sows (+14% for gilts, +10% for sows). Overall, the study shows that dietary supplementation with L -carnitine during pregnancy and lactation improves the reproductive performance of sows. [source]


    Effect of quinolone treatment on selection and persistence of quinolone-resistant Escherichia coli in swine faecal flora

    Abstract Aims:, To study the effect of oral administration of a quinolone on emergence of resistance in an indicator bacterial species from faecal flora. Methods and Results:, Quinolone resistance was studied in Escherichia coli obtained from the faecal contents of pigs housed in nine commercial farrow-to-finish herds in France after administration of flumequine to sows. The percentage of quinolone-resistant E. coli increased in the faeces of sows after administration of flumequine (mean 21·78% at day 7 vs 6·42% before treatment for nalidixic acid) and then decreased (mean 12·6 and 10·4 at days 30 and 60, respectively for nalidixic acid), being not significantly different from initial values 1 month post-treatment. In young pigs, the proportion of resistant strains was lower and decreased over rearing period. Moreover, changes over time of both total E. coli and the proportion of resistant bacteria exhibited great inter-individual variability. Conclusions:, Restoration of susceptible faecal flora occurred within 2 months after flumequine treatment.     The effect of adding cadmium and lead alone or in combination to the diet of pigs on their growth, carcase composition and reproduction

    Abstract Limits for cadmium and lead concentrations in food animal products have been established independently, whereas these two toxic metals often co-exist in polluted regions. Weaned pigs (60) were allocated to ten treatments: control; low (0.5 mg kg,1), medium (1 mg kg,1) or high cadmium (2.5 mg kg,1) in feed; low (5 mg kg,1) medium (10 mg kg,1) or high (25 mg kg,1) lead in feed; and low, medium and high cadmium plus lead in feed. Growth rates and concentrations of cadmium and lead in body tissues (kidney, liver, spleen, lungs, heart, testicle, ribs, hair and teeth) were measured after 137 days. There was a similar reduction in weight gain for pigs in the cadmium and lead treatments, compared with the control, and a greater reduction for the pigs in the cadmium plus lead treatments. The reduction increased with the level of metal included. There was an increase in cadmium concentration of all tissues and blood with increasing feed cadmium concentration, which was usually less when lead was also included in the feed. There was also an increase in tissue lead concentration with increasing dietary lead, and this was in most cases increased when cadmium was also included in the feed. The most sensitive tissues for cadmium and lead exposure were the kidney, liver, hair and teeth, and regression equations were developed for the accumulation rates in these tissues. Tissue and blood cadmium concentrations increased gradually with increasing dietary lead, whereas tissue lead concentration was not sensitive to dietary cadmium, except in the ribs and heart. In a second experiment, 10 sows were allocated to a control diet or the same diet but with a supplement of cadmium and lead. The birth weight of piglets was decreased by the supplement and their mortality increased. Lead accumulated most in the ovary and oviduct of the sows, and there were increases in the lead and, to a lesser extent, cadmium concentrations of tissues of the piglets from these sows.     Placental transfer and pharmacokinetics of allopurinol in late pregnant sows and their fetuses

    Xanthine oxidoreductase (XOR) is a key enzyme in the evolvement of reperfusion injury resulting from birth asphyxia, a common cause of decreased viability and perinatal mortality in newborn piglets under farm conditions. At present no standard pharmacological intervention strategy is available to reduce these adverse effects of birth asphyxia. In the present study we aimed to evaluate placental transfer of allopurinol, an inhibitor of XOR. For this purpose, fetal catheterization of the jugular vein was conducted in five late pregnant sows (one fetus per sow). At 24,48 h after surgery, sows received allopurinol (15 mg/kg body weight; i.v.) and pharmacokinetics of allopurinol and its active metabolite oxypurinol were measured in both late pregnant sows and fetuses. Maternal and fetal blood samples were collected during and after allopurinol administration. Maternal Cmax values averaged 41.90 ,g/mL (allopurinol) and 3.68 ,g/mL (oxypurinol). Allopurinol crossed the placental barrier as shown by the average fetal Cmax values of 5.05 ,g/mL at 1.47 h after allopurinol administration to the sow. In only one fetus low plasma oxypurinol concentrations were found. Incubations of subcellular hepatic fractions of sows and 24-h-old piglets confirmed that allopurinol could be metabolized into oxypurinol. In conclusion, we demonstrated that allopurinol can cross the placental barrier, a prerequisite for further studies evaluating the use of allopurinol as a neuroprotective agent to reduce the adverse effects following birth asphyxia in neonatal piglets.
