    Xenopus, the next generation: X. Tropicalis genetics and genomics

    Abstract A small, fast-breeding, diploid relative of the frog Xenopus laevis, Xenopus tropicalis, has recently been adopted for research in developmental genetics and functional genomics. X. tropicalis shares advantages of X. laevis as a classic embryologic system, but its simpler genome and shorter generation time make it more convenient for multigenerational genetic, genomic, and transgenic approaches. Its embryos closely resemble those of X. laevis, except for their smaller size, and assays and molecular probes developed in X. laevis can be readily adapted for use in X. tropicalis. Genomic manipulation techniques such as gynogenesis facilitate genetic screens, because they permit the identification of recessive phenotypes after only one generation. Stable transgenic lines can be used both as in vivo reporters to streamline a variety of embryologic and molecular assays, or to experimentally manipulate gene expression through the use of binary constructs such as the GAL4/UAS system. Several mutations have been identified in wild-caught animals and during the course of generating inbred lines. A variety of strategies are discussed for conducting and managing genetic screens, obtaining mutations in specific sequences, achieving homologous recombination, and in developing and taking advantage of the genomic resources for Xenopus tropicalis. © 2002 Wiley-Liss, Inc.     Colony productivity and foundress behaviour of a native wasp versus an invasive social wasp

    Abstract., 1.,Colony productivity, prey utilisation, and foundress behaviour of a North American native wasp (Polistes fuscatus) versus an European invasive wasp (Polistes dominulus) were investigated in a controlled field experiment with optimal versus natural foraging conditions. Colonies with the optimal prey foraging conditions were provided with prey ad libitum within an enclosed area. The other colonies foraged in the adjacent field,woodland but had the same nest conditions as the other treatment. 2.,When given prey ad libitum, both wasp species captured similar amounts of prey and the conversion to total offspring biomass was similar. But P. dominulus colonies produced 2.5 times the number of workers as P. fuscatus colonies, reflecting the smaller size of P. dominulus wasps. 3.,Foundresses of P. dominulus were observed more often building or repairing the nest, thereby contributing to the production of colonies with twice as many cells as colonies of P. fuscatus. Foundresses of P. dominulus showed more acts of aggression toward workers than did P. fuscatus foundresses, which was not a function of adult density on the nest. 4.,At the end of the experiment, P. dominulus colonies with optimal prey foraging conditions still had a high level of egg-laying and peaked in the number of pupae then, whereas egg-laying and the number of pupae per colony of the other treatments began to decline 2,3 weeks earlier. These results indicate that P. dominulus is more opportunistic than P. fuscatus, which may account in part for P. dominulus's success as an introduced species in North America. [source]


    Reproduction biology of pikeperch (Sander lucioperca (L.)) , a review

    Abstract,,, The present review focuses on the reproduction biology of pikeperch (Sander lucioperca (L.)). Aspects like maturity, fecundity, spawning migrations, spawning habitats, onset of spawning, and development time of eggs were reviewed. The onset of maturity is reached at younger age in southern than northern populations due to higher growth rate in the south. Males mature at smaller size and are on average younger than females. Absolute fecundity is closely related to the length and weight, but no clear relationship could be found between relative fecundity and length. Statistically significant relationships were found between the onset of spawning and latitude, and between the duration of the development time of eggs and stable water temperature. Near the southern limits of distribution, the onset of spawning is in February while near the northern limits it is in June. The interannual variability in fecundity and in the onset of maturity and further the factors affecting them have not been studied much. Furthermore, it is not known whether these variations could affect the population dynamics of pikeperch. Little is also known about the actual spawning behaviour of pikeperch in natural habitats. This is probably due to the typical spawning habitats located at 1,3 m depth in waters with high turbidity and low visibility. Even though the homing behaviour to the same spawning areas is well developed in adults, it is not known whether the adults were actually born in the same area. [source]


    WORKFORCE COMPOSITION AND FIRM PRODUCTIVITY: EVIDENCE FROM TAIWAN

    We study the relationship between workforce composition and firm productivity based on a new employee-employer-matched data set, using an array of workforce characteristics and three alternative measures of firm productivity. While firm age is not essential for the performance of firms, those of smaller size and those in the steel and transportation industries outperform others. Moreover, labor quality, particularly the middle-aged with higher education, contributes significantly to firms' productivity. Furthermore, economic incentives and market competition both play important roles in the performance of firms.     Effects of atrazine and iridovirus infection on survival and life-history traits of the long-toed salamander (Ambystoma macrodactylum)

    Abstract Environmental contaminants and emerging infectious diseases are implicated as factors contributing to global amphibian declines. However, few studies have tested the interaction of these factors. We exposed six-week-old, larval long-toed salamanders (Ambystoma macrodactylum) to Ambystoma tigrinum virus (ATV; 0 or 103.5 plaque-forming units/ml) and sublethal concentrations of atrazine (0, 1.84, 18.4, and 184 ,g/L) in a 4 × 2 factorial design for 30 d. We tested the effects of atrazine and virus on mass and snout-vent length (SVL) at metamorphosis and larval period as well as on rates of mortality and viral infectivity. We confirmed ATV transmission to A. macrodactylum via polymerase chain reaction, but infection rates were lower than expected, consistent with the theory predicting lower pathogen transmission to nonnative hosts. Larvae exposed to both atrazine and ATV had lower levels of mortality and ATV infectivity compared to larvae exposed to virus alone, suggesting atrazine may compromise virus efficacy. The highest atrazine level (184 ,g/L) accelerated metamorphosis and reduced mass and SVL at metamorphosis significantly relative to controls. Exposure to ATV also significantly reduced SVL at metamorphosis. The present study suggests moderate concentrations of atrazine may ameliorate effects of ATV on long-toed salamanders, whereas higher concentrations initiate metamorphosis at a smaller size, with potential negative consequences to fitness.     Modelling the impacts of removing seal predation from Atlantic salmon, Salmo salar, rivers in Scotland: a tool for targeting conflict resolution

    Abstract, Bioenergetics were used to model the potential impacts on adult Atlantic salmon, Salmo salar L., stocks and rod fisheries of removing harbour seals, Phoca vitulina L., from three rivers of different scales in the Moray Firth, Scotland, viz: the Spey (large), Conon (medium) and Moriston (small). Overall, seals had the greatest impact on the Moriston, where removal of a single animal could increase cumulative catch by 17% during the fishing season. On the Conon and Spey the impacts were negligible, and resulted in increased catches of <1% annually. On all rivers eliminating seal predation had the greatest impact during the spring due to the smaller size of spring salmon sub-stocks. A generalised model of seal removal illustrated that stocks and catches increased by >33% in rivers with monthly rod catches ,10 fish, but declined to <10% for rivers with catches >34 fish. The outputs of the models are qualitative, but provide a management tool for targeting action to resolve seal-salmon fishery conflict. Smaller salmon population units, and spring salmon sub-stocks and fisheries in particular, are most vulnerable to predation. The merit of this approach is discussed regarding the management of Special Areas of Conservation for salmon and seals. [source]


    The effects of morphology and substrate diameter on climbing and locomotor performance in male spiders

    Summary 1.,Spiders are the most sexually size dimorphic terrestrial animals and the evolution of this dimorphism is controversial. Patterns of sexual size dimorphism (SSD) in spiders have been related to individual performance and size. In 2002 Moya-Laraño, Halaj & Wise proposed the ,gravity hypothesis' to explain patterns of sexual size dimorphism in spiders whereby species building webs high in the vegetation are predicted to show greater SSD than those that build lower down. They advocated an advantage in climbing speed in smaller males searching for females in high places. The gravity hypothesis predicts a negative relationship between male size and climbing speed. In 2007 Brandt & Andrade questioned this interpretation and proposed that the pattern of SSD in spiders is better explained by an advantage for larger males of low-dwelling species to run faster along the ground. 2.,We induced male spiders to run a standard distance up vertical poles of different diameters to examine the predicted relationship between size and climbing speed. We tested two species of extremely size-dimorphic orb-web spiders, Argiope keyserlingi and Nephila plumipes, that differ in the height at which females tend to build webs, and one species of jumping spider, Jacksonoides queenslandica, with low levels of size dimorphism. We also examined morphological determinants of horizontal motility by inducing males to run along a raceway. 3.,Substrate diameter was consistently found to influence climbing performance. In N. plumipes, climbing speed was slowest on the widest diameter substrate. In A. keyserlingi, size-adjusted leg length and substrate diameter interacted to determine climbing speed, while in J. queenslandica, there was an interaction between body size and substrate diameter on climbing speed. In the effect of substrate diameter, we have identified a potential bias in previous tests of the gravity hypothesis. 4.,Our results do not support the prediction of the gravity hypothesis. There was no evidence of a negative relationship between body size and climbing speed in the two orb-web species with high levels of SSD. Our results are also not consistent with a recent modification of the gravity hypothesis that suggests a curvilinear relationship between climbing speed and size. 5.,Body size was positively associated with maximum running speed only in the cursorial hunter J. queenslandica. For this spider, results are more consistent with Brandt & Andrade's explanation for variation in SSD in spiders, that larger males are selected for superior running ability in low-dwelling species, rather than selection for smaller size for climbing to females in high-dwelling species.     Reproductive strategies of Gammarus lacustris (Crustacea: Amphipoda) along an elevation gradient

    Abstract 1.,The number of eggs, their size, mass and development time, and the starvation time of newly hatched young, was examined in four populations of Gammarus lacustris along an elevation gradient from prairie to alpine lakes (730 m to > 2300 m above sea level). Water temperature and ice-free season decreased with increasing altitude. 2.,Females in the alpine lake produced fewer but larger and heavier eggs than females in the prairie lake. Eggs produced by females in montane and subalpine lakes were intermediate in size, mass and number. Within populations, egg size was not related to the number of eggs or female size. 3.,The development time of eggs declined with an increase in incubation temperature. At all incubation temperatures, large eggs had a longer incubation time than small eggs. All eggs incubated at 4 °C failed to produce young. Young from large eggs were larger in size than young from small eggs. 4.,The starvation time of newly hatched young increased with decreasing temperature. However, slopes of regressions relating starvation time to temperature differed among populations. At 4 °C young from large eggs survived longer than young from small eggs. 5.,The high phenotypic plasticity in reproductive traits contributes to the success of G. lacustris in a wide range of aquatic habitats. It is predicted that in response to climate-induced warming, populations in currently cold montane and alpine lakes would shift their reproduction to produce more eggs of smaller size. However, the accurate prediction of the fate of populations between ecoregions will require knowledge of the extent to which these traits are under genetic control. [source]


    Complete physical map and gene content of the human NF1 tumor suppressor region in human and mouse

    Duplicon-mediated microdeletions around the NF1 gene are frequently associated with a severe form of neurofibromatosis type I in a subgroup of patients who show an earlier onset of cutaneous neurofibromas, dysmorphic facial features, and lower IQ values. To clarify the discrepancies between published maps of the NF1 tumor-suppressor gene region as well as the length of gaps in these assemblies and to validate the recently described tandem duplication of the human NF1 locus, we assembled a contiguous high-density map of BAC and PAC clones from different genomic libraries. Although two WI-12393,derived low-copy fragments are known to occur at the proximal and distal boundaries of the 1.5-Mb segment that is usually deleted in NF1 microdeletion patients, we identified an additional WI-12393,related segment between the MGC13061 and the NF1 gene, which appears to trigger interstitial deletions of smaller size as observed in two patients. Moreover, we completed the genomic organization and cDNA structure of all functional genes, CYTOR4, FLJ12735, FLJ22729, CENTA2, MGC13061, NF1, OMG, EVI2B, EVI2A, KIAA1821, MGC11316, HCA66, KIAA0160, and WI-12393, from this region. A comparison of the human map to the orthologous region on mouse chromosome 11 revealed significant differences in the number and arrangement of genes, indicating that many chromosomal breaks with partial duplications, inversions, and deletions occurred predominantly in the primate lineage. © 2003 Wiley-Liss, Inc.     Direct, partitioned and projected solution to finite element consolidation models

    Abstract Direct, partitioned, and projected (conjugate gradient-like) solution approaches are compared on unsymmetric indefinite systems arising from the finite element integration of coupled consolidation equations. The direct method is used in its most recent and computationally efficient implementations of the Harwell Software Library. The partitioned approach designed for coupled problems is especially attractive as it addresses two separate positive definite problems of a smaller size that can be solved by symmetric conjugate gradients. However, it may stagnate and when converging it does not prove competitive with a global projection method such as Bi-CGSTAB, which may take full advantage of its flexibility in working on scaled and reordered equations, and thus may greatly improve its computational performance in terms of both robustness and convergence rate. The Bi-CGSTAB superiority to the other approaches is discussed and demonstrated with a few representative examples in two-dimensional (2-D) and three-dimensional (3-D) coupled consolidation problems.     Cooling load density characteristics of an endoreversible variable-temperature heat reservoir air refrigerator

    Abstract The performance optimization of an endoreversible air refrigerator with variable-temperature heat reservoirs is carried out by taking the cooling load density, i.e. the ratio of cooling load density to the maximum specific volume in the cycle, as the optimization objective in this paper. The analytical relations of cooling load, cooling load density and coefficient of performance are derived with the heat resistance losses in the hot- and cold-side heat exchangers. The maximum cooling load density optimization is performed by searching the optimum pressure ratio of the compressor, the optimum distribution of heat conductance of the hot- and cold-side heat exchangers for the fixed total heat exchanger inventory, and the heat capacity rate matching between the working fluid and the heat reservoirs. The influences of some design parameters, including the heat capacitance rate of the working fluid, the inlet temperature ratio of heat reservoirs and the total heat exchanger inventory on the maximum cooling load density, the optimum heat conductance distribution, the optimum pressure ratio and the heat capacity rate matching between the working fluid and the heat reservoirs are provided by numerical examples.     Stage 2 Pressure Ulcer Healing in Nursing Homes

    OBJECTIVES: To identify resident and wound characteristics associated with Stage 2 pressure ulcer (PrU) healing time in nursing home residents. DESIGN: Retrospective cohort study with convenience sampling. SETTING: One hundred two nursing homes participating in the National Pressure Ulcer Long-Term Care Study (NPULS) in the United States. PARTICIPANTS: Seven hundred seventy-four residents aged 21 and older with length of stay of 14 days or longer who had at least one initial Stage 2 (hereafter Stage 2) PrU. MEASUREMENTS: Data collected for each resident over a 12-week period included resident characteristics and PrU characteristics, including area when first reached Stage 2. Data were obtained from medical records and logbooks. RESULTS: There were 1,241 initial Stage 2 PrUs on 774 residents; 563 (45.4%) healed. Median time to heal was 46 days. Initial area was significantly associated with days to heal. Using Kaplan-Meier survival analyses, median days to heal was 33 for small (,1 cm2), 53 days for medium (>1 to ,4 cm2), and 73 days for large (>4 cm2) ulcers. Using Cox proportional hazard regression models to examine effects of multiple variables simultaneously, small and medium ulcers and ulcers on residents with agitation and those who had oral eating problem healed more quickly, whereas ulcers on residents who required extensive assistance with seven to eight activities of daily living (ADLs), who temporarily left the facility for the emergency department (ED) or hospital, or whose PrU was on an extremity healed more slowly. CONCLUSION: PrUs on residents with agitation or with oral eating problems were associated with faster healing time. PrUs located on extremities, on residents who went temporarily to the ED or hospital, and on residents with high ADL disabilities were associated with slower healing time. Interaction between PrU size and place of onset was also associated with healing time. For PrU onset before or after admission to the facility, smaller size was associated with faster healing time.     Stability of genetic parameter estimates for production traits in pigs

    Changes in variance component estimates in growing sets of performance data in two pig breeds were investigated. Data was used from the field and station test of Czech Landrace (LA: 75 099 observations) and the Slovakian breed, White Meaty swine (WM: 32 203 observations). In LA the traits analysed were estimated lean meat content (LM) and average daily gain (ADGF) on field test and average daily gain (ADGS) and weight of valuable cuts (VCW) on station test. In WM the traits analysed were backfat thickness on field and station test (BFF, BFS, respectively), proportion of valuable cuts (VCP) on station test, ADGF and ADGS. Covariance components were estimated from four- and five-trait animal models using the VCE software. Omitting data from factor levels with a low number of records led to 4.2% of LA records and 21.7% of WM records being deleted. Changes in genetic and residual variance estimates were less than 5% for all traits in LA and less than 12% for all traits except ADGS in WM. The changes in estimated genetic variances caused by 18 months (LA) or 24 months (WM) of new data were 2,25% and the changes in estimated residual variances were less than 5% in LA and less than 20% in WM. In both breeds, changes in heritability estimates did not exceed 0.06 in absolute value. In LA, it is reasonable to use genetic parameter estimates for 3 years before re-estimation. In WM the time interval should be shorter because of changes in the estimates caused by their lower accuracy arising from the smaller size of the data-set and smaller frequency of station testing. Folgende Merkmale wurden ausgewertet: Magerfleischanteil (LM) und Lebenstagszunahme (ADGF) aus der Feldprüfung sowie Prüftagszunahme (ADGS) und Gewicht wertvoller Teilstücke (VCW) aus der Stationsprüfung bei der LA; Rückenspeckdicke aus der Feld- und Stationsprüfung (BFF bzw. BFS), Anteil wertvoller Teilstücke (VCP) aus der Stationsprüfung sowie ADGF und ADGS bei der Rasse WM. Die Kovarianzkomponenten wurden für Vier- bzw. Fünf-Merkmals-Tiermodelle mit dem Programm VCE berechnet. Das Auslassen von Daten von Klassen mit geringer Besetzung führte dazu, daß in der LA 4,2% und in WM 21,7% der Daten gelöscht wurden. Die Änderungen in den genetischen und den Rest-Varianzen waren in der LA bei allen Merkmalen kleiner als 5% und in WM bei allen Merkmalen mit Ausnahme von ADGS kleiner als 12%. Durch Hinzufügen von Daten aus einem Zeitraum von 18 (LA) bzw. 24 (WM) Monaten änderten sich die genetischen Varianzen um 2 bis 25%. Die Änderungen in den Restvarianzen lagen unter 5% bei der LA und unter 20% bei WM. Die maximale Änderung der Heritabilitätskoeffizienten überstieg in beiden Rassen nicht 0,06. Bei der LA sollte ein Zeitintervall von drei Jahren zu einer Neuschätzung der genetischen Parameter ausreichen, bei WM sollte wegen der beobachteten Änderungen der Schätzwerte, der kleineren Datenmenge und des geringeren Anteils stationsgeprüfter Tiere das Zeitintervall kürzer sein.     Size at maturity and egg capsules of the softnose skates Bathyraja brachyurops (Fowler, 1910) and Bathyraja macloviana (Norman, 1937) (Elasmobranchii: Rajidae) in the SW Atlantic (37°00,,39°30,S)

    Summary The softnose skates Bathyraja brachyurops and Bathyraja macloviana represent an important portion of the skate catches of the Uruguayan trawling fleet in the southwestern Atlantic. From March to October 2004, specimens of these species were collected at 75,200 m depth range in the area situated between latitudes 37°00,,39°30,S. For B. brachyurops, total length at which 50% of the specimens were retained by the gear was 68.0 cm for both sexes; TL50 was estimated at 65.4 cm for males and 67.0 cm for females. For B. macloviana, total length at which 50% of the specimens were retained was 56.0,57.0 cm for both sexes; TL50 was estimated at 53.5 cm for males and 52.0 cm for females. Egg capsule length varied from 79,91 mm in B. brachyurops and 69,75.5 mm in B. macloviana. In both species, capsules displayed striated surfaces and similar gross morphology, although egg capsules of B. macloviana had more robust anterior horns and a smaller size than those of B. brachyurops. Egg capsules of the latter also exhibited microscopical prickles. Capsule edges were laterally keeled with a groove along the keel, and a straight and transverse velum was present in the egg capsules of both species. [source]


    Preparation and properties of dynamically cured PP/MAH- g -EVA/epoxy blends

    Abstract A method concerning with the simultaneous reinforcing and toughening of polypropylene (PP) was reported. Dynamical cure of the epoxy resin with 2-ethylene-4-methane-imidazole (EMI-2,4) was successfully applied in the PP/maleic anhydride-grafted ethylene-vinyl acetate copolymer (MAH- g -EVA), and the obtained blends named as dynamically cured PP/MAH- g -EVA/epoxy blends. The stiffness and toughness of the blends are in a good balance, and the smaller size of epoxy particle in the PP/MAH- g -EVA/epoxy blends shows that MAH- g -EVA was also used as a compatibilizer. The structure of the dynamically cured PP/MAH- g -EVA/epoxy blends is the embedding of the epoxy particles by the MAH- g -EVA. The cured epoxy particles as organic filler increases the stiffness of the PP/MAH- g -EVA blends, and the improvement in the toughness is attributed to the embedded structure. The tensile strength and flexural modulus of the blends increase with increasing the epoxy resin content, and the impact strength reaches a maximum of 258 J/m at the epoxy resin content of 10 wt %. DSC analysis shows that the epoxy particles in the dynamically cured PP/MAH- g -EVA/epoxy blends could have contained embedded MAH- g -EVA, decreasing the nucleating effect of the epoxy resin. Thermogravimetric results show the addition of epoxy resin could improve the thermal stability of PP, the dynamically cured PP/MAH- g -EVA/epoxy stability compared with the pure PP. Wide-angle x-ray diffraction analysis shows that the dynamical cure and compatibilization do not disturb the crystalline structure of PP in the blends. © 2009 Wiley Periodicals, Inc.     Effective thermal conductivity behavior of filled vulcanized perfluoromethyl vinyl ether rubber

    Abstract The effective thermal conductivity behavior of vulcanized perfluoromethyl vinyl ether (PMVE) rubber filled with various inorganic fillers was investigated and analyzed with thermal conductivity models. Experimental results showed that there was no significant improvement in the thermal conductivity of PMVE rubber if the intrinsic thermal conductivity of the fillers was greater than 100 times that of the rubber matrix, and this agreed with the prediction of Maxwell's equation. The thermal conductivity of PMVE rubber filled with larger size silicon carbide (SiC) particles was greater than that of PMVE filled with smaller size SiC because of the lower interfacial thermal resistance, and there existed a transition filler loading at about 60 vol %. It was also found that flocculent graphite was the most effective thermally conductive filler among the fillers studied.     Crystallization of poly(butylene terephthalate)/poly(ethylene octene) blends: Nonisothermal crystallization

    Abstract Poly(ethylene octene) (POE), maleic anhydride grafted poly(ethylene octene) (mPOE), and a mixture of POE and mPOE were added to poly(butylene terephthalate) (PBT) to prepare PBT/POE (20 wt % POE), PBT/mPOE (20 wt % mPOE), and PBT/mPOE/POE (10 wt % mPOE and 10 wt % POE) blends with an extruder. The melting behavior of neat PBT and its blends nonisothermally crystallized from the melt was investigated with differential scanning calorimetry (DSC). Subsequent DSC scans exhibited two melting endotherms (TmI and TmII). TmI was attributed to the melting of the crystals grown by normal primary crystallization, and TmII was due to the melting of the more perfect crystals after reorganization during the DSC heating scan. The better dispersed second phases and higher cooling rate made the crystals that grew in normal primary crystallization less perfect and relatively prone to be organized during the DSC scan. The effects of POE and mPOE on the nonisothermal crystallization process were delineated by kinetic models. The dispersed phase hindered the crystallization; however, the well- dispersed phases of an even smaller size enhanced crystallization because of the higher nucleation density.     In vivo evaluation of hydroxyapatite foams

    Abstract Hydroxyapatite (HA) is widely applied as bone graft material due to its osteoconductive potential and well-established biocompatibility. In this work, macroporous hydroxyapatite structures made through foaming of aqueous suspensions and gelcasting were tested for in vivo osteointegration. These foams are composed of a three-dimensional array of spherical pores with diameters of approximately 100,500 ,m, interconnected by windows of smaller size in the range of 30,120 ,m. The HA foams were implanted in the tibia of albino New Zealand rabbits and removed after a period of 8 weeks. Histological analysis revealed that the pores in the foams were partially or completely filled progressively with mature new bone tissue and osteoid after the implanted period. No immune or inflammatory reactions were detected. The high osteoconductive potential of the HA foams provides a potential structure for use as bone substitute in orthopedic, oral, and cranio-maxillofacial reconstructive surgery, and as dento-alveolar implants. © 2002 Wiley Periodicals, Inc.     Effects of exploitation on reproductive capacity of blackspot snapper, Lutjanus fulviflamma (Pisces: Lutjanidae) in Mafia Island, Tanzania

    Abstract There is paucity of information on the effects of exploitation on reproductive characteristics of blackspot snapper, Lutjanus fulviflamma (Forsskål 1775) in Tanzanian coastal waters. We compared size at first sexual maturity (LM50), sex ratio, fecundity, and breeding season of L. fulviflamma in least fished Mafia Island Marine Park (MIMP) and intensively fished areas (IFA) between May 1999 and April 2001. Fish in MIMP matured at significantly smaller size (female: LM50 = 206.3 mm; male: LM50 = 195.5 mm) than in IFA (female: LM50 = 216.7 mm; male: LM50 =212.1 mm) total body length. Sex ratio was balanced at 1.03 : 1 (female : male) in MIMP, but it was skewed 0.9 : 1 (female : male) in IFA. Size-related differences in sex ratio were observed with males predominating in the smaller sizes and females in the larger sizes. Total fecundity of fish in MIMP was determined at 45,200,430,200 oocytes in females of between 207 and 293 mm total length. Lutjanus fulviflamma in MIMP has a prolonged spawning season lasting from September to March peaking in December. None of the fish from IFA were in breeding state, suggesting recruitment overfishing is an added matter of concern for the long-term sustainability of the fishery at the current exploitation level. Résumé Il existe très peu d'informations sur les effets de l'exploitation sur les caractéristiques de la reproduction de Lutjanus fulviflamma (Forsskål 1775) dans les eaux côtières de Tanzanie. Nous avons comparé la taille au début de la maturité sexuelle (LM50), le sex-ratio, la fécondité et la saison de reproduction de L. fulviflamma dans le Parc Marin de l'Ile de Mafia (MIMP) où la pêche est moins intense et dans des eaux intensément exploitées (IFA) entre mai 1999 et avril 2001. Les poissons du MIMP deviennent mâtures tout en ayant une longueur totale significativement plus petite (femelles: LM50 = 206,3 mm; mâles: LM50 = 195,5 mm) que dans les IFA (femelles: LM50 = 216,7 mm; mâles: LM50 = 212,1 mm). Le sex-ratio est équilibréà 1,03/1 (femelles/mâles) dans le MIMP, mais il est de 0,9/1 (femelles/mâles) dans les IFA. On a observé des différences du sex-ratio liées à la taille, avec les mâles qui prédominent dans les plus petites tailles et les femelles dans les plus grandes. On a déterminé la fécondité totale dans le MIMP, entre 45.200,430.000 ovocytes à l'intérieur des femelles comprises entre 207 et 293 mm de longueur totale. Dans le MIMP, Lutjanus fulviflamma a une saison de frai prolongée, allant de septembre à mars avec un pic en décembre. Aucun des poissons des IFA n'était en période de reproduction, ce qui signifie une sur-pêche du potentiel reproducteur, ce qui est un motif d'inquiétude pour la durabilitéà long terme de la pêche à son niveau actuel.     Effects of aging and gender on the spatial organization of nuclei in single human skeletal muscle cells

    Summary The skeletal muscle fibre is a syncitium where each myonucleus regulates the gene products in a finite volume of the cytoplasm, i.e., the myonuclear domain (MND). We analysed aging- and gender-related effects on myonuclei organization and the MND size in single muscle fibres from six young (21,31 years) and nine old men (72,96 years), and from six young (24,32 years) and nine old women (65,96 years), using a novel image analysis algorithm applied to confocal images. Muscle fibres were classified according to myosin heavy chain (MyHC) isoform expression. Our image analysis algorithm was effective in determining the spatial organization of myonuclei and the distribution of individual MNDs along the single fibre segments. Significant linear relations were observed between MND size and fibre size, irrespective age, gender and MyHC isoform expression. The spatial organization of individual myonuclei, calculated as the distribution of nearest neighbour distances in 3D, and MND size were affected in old age, but changes were dependent on MyHC isoform expression. In type I muscle fibres, average NN-values were lower and showed an increased variability in old age, reflecting an aggregation of myonuclei in old age. Average MND size did not change in old age, but there was an increased MND size variability. In type IIa fibres, average NN-values and MND sizes were lower in old age, reflecting the smaller size of these muscle fibres in old age. It is suggested that these changes have a significant impact on protein synthesis and degradation during the aging process. [source]


    Morphological differences between two ecologically similar sympatric fishes

    Morphological differentiation and microhabitat segregation of two ecologically similar populations of pelagic planktivorous fishes, Coregonus albula and the smaller Coregonus fontanae, were studied in Lake Stechlin (northern Germany). Both populations performed diel vertical migrations, although C. fontanae was always situated in deeper pelagic water than C. albula both during day and night. Landmark-based geometric morphometrics revealed that sympatric C. albula and C. fontanae differ in external morphology, with main differences found in head length and eye position, as well as in length and width of the caudal peduncle. Moreover, while C. albula has a similar morphology over all sizes, the shape of C. fontanae changes with size. Accordingly, the morphology of the two is most different at smaller size. Although the morphological differences may reflect adaptations to the slightly differing microhabitats of the two populations, there is no conclusive evidence that this correspondence between ecology and morphology is the main mechanism behind the coexistence of the closely related coregonids in Lake Stechlin.     Pigment variability in larval Sebastes jordani off central California

    Atypical pigment was observed in pre-flexion, flexion and post-flexion larval Sebastes collected off central California in 1991 and 1992 that otherwise resembled Sebastes jordani. Atypical pigment occurred on the axillary region at the base of the pectoral fin (most prominent on the inner edge of the pectoral fin base adjacent to the gut), on the median and distal regions of the pectoral fin and on the median and distal regions of the pelvic fin. In addition, lower and upper jaw pigment was observed at a much smaller size in these specimens than previously described in the literature. Identifications of these atypical specimens as S. jordani were confirmed using meristic and otolith characters as well as mitochondrial DNA sequence data. The ontogenetic variability of S. jordani is described. Specimens collected north of Point Conception were more pigmented than larval S. jordani described in the literature (collected predominately south of Point Conception), suggesting geographic variation in pigment development during the larval stage.     Spawning and recruitment patterns of major fish species in Bontanga Reservoir, Ghana, West Africa

    Abstract The spawning and recruitment patterns of the major Cichlid fish species, including Hemichromis fasciatus, Oreochromis niloticus, Sarotherodon galilaeus, Tilapia zillii and other species, Auchenoglanis occidentalis, Brycinus nurse, Clarias gariepinus and Marcusenius senegalensis were studied for 24 months in Bontanga Reservoir, Ghana, using length-based fish stock assessment approaches. The species spawned throughout the year, with two spawning pulses described as major and minor spawning seasons. The major spawning season occurred from March to September for the Cichlids, and from May to September for the other species. The minor spawning season, indicative of extended spawning, occurred from October to March for all the fish stocks. Fish spawning began with the onset of the rains in April/May, peaking by June/July, before the rainfall peak in August for all the fish stocks studied. Recruitment was found to occur throughout the year, with major and minor pulses coinciding with the major and minor spawning seasons. Accordingly, the most appropriate time for a possible closed fishing period appears to be from June to August, 1 month after the start of, and before the end of, the rains. The estimated mean standard length (Lm) for first time spawners of A. occidentalis, B. nurse, C. gariepinus and H. faciatus were 11.7, 12.7, 2.7 and 7.5 cm respectively. The estimated maturity,length ratio of 0.4 and 0.2 for O. niloticus, S. galilaeus and T. zillii were lower than the known 0.7 for normal growth, suggesting the tilapias matured faster, and at a smaller size, in the reservoir. Apparent sexual precocity associated with early maturity, year-round spawning and recruitment were some important adaptations found to have sustained the reservoir fisheries, even during high fishing pressures. For conservation and sustainable exploitation of the fisheries, instituting a closed fishing season, mesh size regulations, withdrawal rights and a community-based fishery management system are recommended.     MRT Letter: Spatial distribution of vancomycin-induced damage in Staphylococcus epidermidis biofilm: An electron microscopic study

    Abstract This study was planned to elucidate the efficacy of antibiotics on Staphylococcus epidermidis and Staphylococcus aureus biofilms by scanning electron microscopy (SEM). Biofilms of S. epidermidis ATCC 35984 and S. aureus ATCC 29213 were grown on black, polycarbonate membranes placed on tryptic soy agar plates for 48 h at 37°C, and then exposed to vancomycin or amikacin or ciprofloxacin at clinically achievable levels for 24 h at 37°C. The morphology of antibiotic-treated and untreated biofilms was elucidated by SEM. SEM analysis indicated a differential affection of S. epidermidis ATCC 35984 in the center and periphery of biofilm upon treatment with vancomycin. The center of biofilm revealed damaged cells with sparse distribution, smaller size, and irregular shape, whereas cells in the periphery were unaffected. This differential distribution of susceptibility within S. epidermidis ATCC 35984 biofilms was specific for vancomycin only and was not observed on exposure to amikacin or ciprofloxacin. No such response was found in S.aureus ATCC 29213 biofilms. Thus, our study suggests a spatial distribution of vancomycin-induced damage in S. epidermidis biofilms. To our knowledge, this is the first report that indicates a differential affection of S. epidermidis in the center and periphery of biofilm upon treatment with vancomycin. Studies on the factors controlling this differential distribution could provide valuable insights into the mechanisms of antimicrobial resistance in S. epidermidis biofilms. Microsc. Res.     Design of a compact microstrip low-pass filter using defected ground structure

    Abstract A novel compact microstrip low-pass filter (LPF) is proposed in this article. The proposed LPF is composed of semicircle-defected ground structures (S-DGS) and semicircle stepped-impedance shunt stubs (S-SISS). An equivalent lumped L-C circuit model is also presented. With the S-DGS and S-SISS, better band-stop performance and much smaller size can be obtained.     Populations do not become less genetically diverse or more differentiated towards the northern limit of the geographical range in clonal Vaccinium stamineum (Ericaceae)

    Summary ,,Geographically peripheral populations are expected to exhibit lower genetic diversity and higher differentiation than central populations because of their smaller size and greater spatial isolation. In plants, a shift from sexual to clonal asexual reproduction may further reduce diversity and increase differentiation. ,,Here, these predictions were tested by assaying 36 inter-simple sequence repeat (ISSR) polymorphisms in 21 populations of the woody, clonal plant Vaccinium stamineum in eastern North America, from the range center to its northern limit where it has ,threatened' status. Populations decline in frequency, but not size or sexual reproductive output, across the range. ,,Within-population diversity did not decline towards range margins. Modest genetic differentiation among populations increased slightly towards range margins and in small populations with high clonal propagation and low seed production, although none of these trends was significant. Low seed production and high clonal propagation were not associated with large-scale clonal spread. ,,By combining demographic and genetic data, this study determined that increased population isolation, rather than reduced population size, can account for the weak increase in genetic differentiation at range margins.     k-space analysis of point-resolved spectroscopy (PRESS) with regard to spurious echoes in in vivo1H MRS

    Abstract The spurious echo artefact, not uncommon in 1H MRS in the brain, comes from refocusing outer volume signal. Application of MRS in small volumes in susceptibility-affected regions often results in large shim gradients. The artefact problem is accentuated when the global effect of the shim gradient shifts the water resonance outside the water suppression band in the outer volume. This scenario brings the issue of spurious echoes once again to the fore. In this paper, spurious signals of the point-resolved spectroscopy (PRESS) sequence are analysed using the concept of k-space. This new approach facilitates a more geometrical view of the problem, well suited for studying the effect of gradient spoiling and refocusing of signal. Several spoiling options are shown, and the probability of the global effects of shimming being a primary cause of the artefact is discussed. Fourier transform analysis of realistic slice profiles, combined with the k-space description of spurious echoes, shows that unsuppressed water signal in outer regions greatly increases the demands on spoiling. Gradient spoiling adequate for artefact suppression at a given size of MRS volume may not be sufficient at a smaller size.     Antenatal and postpartum depressive symptoms are differentially associated with early childhood weight and adiposity

Paediatric and Perinatal Epidemiology 2010; 24: 179,189. Antenatal depression is associated with small-for-gestational age, but few studies have examined associations with weight during childhood. Similarly, few studies address whether antenatal and postpartum depression differentially affect child weight. Among 838 mother,child dyads in Project Viva, a prospective cohort study, we examined relationships of antenatal and postpartum depression with child weight and adiposity. We assessed maternal depression at mid-pregnancy and 6 months postpartum with the Edinburgh Postnatal Depression Scale (score ,13 indicating probable depression). We assessed child outcomes at age 3 years: body mass index (BMI) z-score, weight-for-height z-score, sum of subscapular (SS) and triceps (TR) skinfold thickness (SS + TR) for overall adiposity, and SS : TR ratio for central adiposity. Sixty-nine (8.2%) women experienced antenatal depression and 59 (7.0%) postpartum depression. Mean (SD) outcomes at age 3 were: BMI z-score, 0.45 (1.01); SS + TR, 16.72 (4.03) mm; SS : TR, 0.64 (0.15). In multivariable models, antenatal depression was associated with lower child BMI z-score (,0.24 [95% confidence interval: ,0.49, 0.00]), but higher SS : TR (0.05 [0.01, 0.09]). There was no evidence of a dose,response relationship between antenatal depression and these outcomes. Postpartum depression was associated with higher SS + TR (1.14 [0.11, 2.18]). In conclusion, whereas antenatal depression was associated with smaller size and central adiposity at age 3 years, postpartum depression was associated with higher overall adiposity. [source]


    SOFT-TISSUE PRESERVATION OF THE HIND GUT IN A NEW GENUS OF CLADID CRINOID FROM THE MISSISSIPPIAN (VISEAN, ASBIAN) AT ST ANDREWS, SCOTLAND

    Abstract:, Soft-tissue preservation of the hind gut, or anal sac, in the tegmen of Tubulusocrinus (gen. nov.) doliolus (Wright) from the Mississippian (Visean, Asbian) Pittenweem Formation at St Andrews, Scotland, is the first of its kind known in crinoids; it sheds important new light on the nature of the cladid tegmen. Many cladid crinoids had a calcite-plated anal sac that may have functioned like a chimney to prevent fouling of the ambulacra. The tubular, uncalcified anal sac of Tubulusocrinus may have functioned like a hose by pointing down current between the arms to avoid fouling of the ambulacra. Its smaller size may also have required less energy to produce than a heavy-plated sac. The hind gut is apparently preserved by haematite replacement, which was probably altered from original authigenic pyrite that formed soon after catastrophic burial. [source]
