    Demographic and life-history correlates for Amazonian trees

    Abstract Questions: Which demographic and life-history differences are found among 95 sympatric tree species? Are there correlations among demographic parameters within this assemblage? Location: Central Amazonian rain forest. Methods: Using long-term data from 24 1,ha permanent plots, eight characteristics were estimated for each species: wood density, annual mortality rate, annual recruitment rate, mean stem diameter, maximum stem diameter, mean stem-growth rate, maximum stem-growth rate, population density. Results: An ordination analysis revealed that tree characteristics varied along two major axes of variation, the major gradient expressing light requirements and successional status, and the second gradient related to tree size. Along these gradients, four relatively discrete tree guilds could be distinguished: fast-growing pioneer species, shade-tolerant sub-canopy species, canopy trees, and emergent species. Pioneers were uncommon and most trees were canopy or emergent species, which frequently had low mortality and recruitment. Wood density was negatively associated with tree mortality, recruitment, and growth rates when all species were considered. Growth rates varied markedly among and within species, with pioneers exhibiting far faster and less variable growth rates than did the other species. Slow growth in subcanopy species relative to canopy and emergent trees was not a simple consequence of mean tree size, but apparently resulted from physiological constraints imposed by low-light and other conditions in the forest understorey. Conclusions: Trees of Amazonian rain forests could be classified with some success into four relatively distinctive guilds. However, several demographic and life-history traits, such as those that distinguish early and late successional species, probably vary along a continuum, rather than being naturally grouped into relatively discrete categories.     Overcoming merohedral twinning in crystals of bacteriorhodopsin grown in lipidic mesophase

    Twinning is one of the most common crystal-growth defects in protein crystallography. There are neither efficient rational approaches for the growth of nontwinned protein crystals nor are there examples of systematic studies of the dependence of the twinning-ratio distribution on crystallization conditions. The description of the twinning phenomenon has been covered even less for membrane-protein crystals and is non-existent for crystals grown using lipidic phases (in meso). In the present work, possibilities for overcoming merohedral twinning are investigated for crystals of the membrane protein bacteriorhodopsin (bR) grown in meso. It is shown that traditional crystallization additives are not effective in the case of the in meso crystallization of bR. The twinning ratio was determined for 310 crystals grown under different crystallization conditions. A correlation of the twinning ratio with the growth rate of the crystals was observed. Slow growth indicated that crystals had a noticeable chance of avoiding twinning. Model calculations were performed in order to rationalize this observation. The calculations confirmed the experimental observation that most crystals consist of two twin domains and showed that under this condition small changes in the probability of twin-domain formation lead to dramatic changes in the number of nontwinned crystals, which explains why slow crystal growth results in a considerable number of nontwinned crystals.     Recombinant hGH replacement therapy and the hypothalamus,pituitary,thyroid axis in children with GH deficiency: when should we be concerned about the occurrence of central hypothyroidism?

    Summary objective, Recombinant hGH treatment may alter thyroid hormone metabolism and we have recently reported that 50% of patients with GH deficiency (GHD) due to organic lesions, previously not treated with thyroxine, developed hypothyroidism during treatment with recombinant human GH (rhGH). These results prompted us to evaluate the impact of rhGH treatment on thyroid function in children with GHD. design, Open study of GH treatment up to 12 months. Investigations were performed at baseline, and after 6 and 12 months of GH therapy. measurement and study subjects, Serum TSH, FT4, FT3, AbTg and AbTPO, IGF-I, height and weight, were evaluated in 20 euthyroid children (group A) with idiopathic isolated GHD and in six children (group B) with multiple pituitary hormone deficiencies (MPHD) due to organic lesions. Among the latter, four already had central hypothyroidism and were on adequate LT4 replacement therapy, while two were euthyroid at the beginning of the study. results, Serum IGF-I levels normalized in all patients. In both groups, a significant reduction in FT4 levels (P < 0·01) occurred during rhGH therapy. No patient in group A had FT4 values into the hypothyroid range, while in four of six patients in group B, fell FT4 levels into the hypothyroid range during rhGH. In particular, the two euthyroid children developed central hypothyroidism during rhGH treatment, and their height velocities did not normalize until the achievement of euthyroidism through appropriate LT4 substitution. No variation in serum FT3 and TSH levels was recorded in either groups. conclusion, Contrary to that observed in patients with MPHD, rhGH replacement therapy does not induce central hypothyroidism in children with idiopathic isolated GHD, further supporting the view that in children with MPHD, as in adults, GHD masks the presence of central hypothyroidism. Slow growth (in spite of adequate rhGH substitution and normal IGF-I levels) is an important clinical marker of central hypothyroidism, therefore a strict monitoring of thyroid function is mandatory in treated children with MPHD.     WHAT KIND OF BENEFIT DO WE EXPECT FOR PERORAL PANCREATOSCOPY IN THE DIAGNOSIS OF INTRADUCTAL PAPILLARY MUCINOUS TUMOR OF THE PANCREAS?

    Intraductal papillary mucinous tumor (IPMT) of the pancreas is characterized by slow growth and a relatively favorable prognosis, however, invasive cancer originating in an IPMT is associated with a poor prognosis. Although various parameters in imaging modalities have been advocated to differentiate between benign IPMN and malignant ones, it is not easy to obtain definite diagnosis based on these parameters. Peroral pancreatoscopy (POPS) allows a clear and direct visualization of the pancreatic duct, providing useful information regarding tumor nature in IPMT. The authors have studied the usefulness of POPS in the diagnosis of IPMT. Nevertheless, its usefulness is not necessarily widely accepted and the significance of POPS is still controversial. In this review, the authors intended to address the diagnostic value of POPS and to clarify its role in the diagnosis of IPMT. The authors think treatment of IPMT can be improved by introducing POPS because the determination of surgical procedure as well as the area of resection based on the preoperative diagnosis of the involvement of the main pancreatic duct and branch duct is inevitable.     KIN-BASED RECOGNITION AND SOCIAL AGGREGATION IN A CILIATE

    Aggregative groups entail costs that must be overcome for the evolution of complex social interactions. Understanding the mechanisms that allow aggregations to form and restrict costs of cheating can provide a resolution to the instability of social evolution. Aggregation in Tetrahymena thermophila is associated with costs of reduced growth and benefits of improved survival through "growth factor" exchange. We investigated what mechanisms contribute to stable cooperative aggregation in the face of potential exploitation by less-cooperative lines using experimental microcosms. We found that kin recognition modulates aggregative behavior to exclude cheaters from social interactions. Long-distance kin recognition across patches modulates social structure by allowing recruitment of kin in aggregative lines and repulsion in asocial lines. Although previous studies have shown a clear benefit to social aggregation at low population densities, we found that social aggregation has very different effects at higher densities. Lower growth rates are a cost of aggregation, but also present potential benefits when restricted to kin aggregations: slow growth and crowd tolerance allow aggregations to form and permit longer persistence on ephemeral resources. Thus in highly dynamic metapopulations, kin recognition plays an important role in the formation and stability of social groups that increase persistence through cooperative consumptive restraint.     Decreased expression of thymidine phosphorylase/platelet-derived endothelial cell growth factor in basal cell carcinomas

    Abstract:, Thymidine phosphorylase (TP)/platelet-derived endothelial cell growth factor is associated with tumor angiogenesis. We evaluated the TP mRNA and protein expression in basal cell carcinomas (BCC) and in various skin tumors including numerous BCC histological simulants. Immunohistochemistry was performed on 99 paraffin sections of formalin-fixed skin tumors using monoclonal antibodies (mAb) against TP. TP mRNA levels were measured by real time RT-PCR in whole BCCs (wBCC) and laser capture microdissected (LCM) BCC tumor cells. TP immunostaining was negative in all BCC variants and in most of the benign trichogeneic tumors studied. By contrast, TP was constantly immunodetected in actinic keratosis (AK), squamous cell carcinomas (SCC), syringomatous carcinomas (SC), basosquamous carcinomas (BSC) and melanomas. TP mRNA levels were low and statistically not different in wBCC and normal skin but were strongly downregulated in LCM-BCC as compared with LCM-normal epidermis. We concluded that (i) anti-TP mAb is an useful marker to differentiate BCC from AK, SCC, BSC and SC but not from trichoblastic tumors, (ii) the lack of TP protein expression in BCC tumoral cells is linked to transcriptional regulatory mechanisms, (iii) the low TP mRNA levels in whole BCC may be related to the low intra-tumoral microvessel density, the slow growth and the very low metastatic potential of these tumors.     Pronounced drought tolerance characterizes the early life stages of the epiphytic bromeliad Tillandsia flexuosa

    Summary 1Germination and seedling performance may set the limits for plant distributions, particularly in stressful habitats. Stressful conditions at these early stages may be avoided by opportunistic germination and growth, or may be tolerated. Many epiphytic plants are frequently exposed to severe drought. Adult epiphytes endure such dry periods in various ways, but little is known about strategies employed during germination and early life. 2Epiphyte seedlings could show either opportunistic fast growth to quickly attain the benefits of being larger or inherently slow growth and early drought tolerance. Here we address the question: which of these strategies characterizes the early life stages of the epiphytic bromeliad Tillandsia flexuosa, a species typical for dry tropical habitats? 3We studied growth and drought tolerance of germinating seeds, of the emerging seedlings and of 2-month-old seedlings under controlled conditions. Additionally, we studied drought hardening in 6-month-old seedlings. 4Germination of T. flexuosa was reduced by intermittent dry periods. However, compared to the congeneric T. fasciculata, which typically occurs in wetter habitats, the depression of germination by drought was small. Seedling growth was fastest at intermediate moisture levels: both prolonged drought and continuous moisture depressed growth. Prolonged drought had a less negative effect on drought-hardened seedlings than on previously well-watered seedlings. After a 3-week drought treatment the previously well-watered seedlings had lost their growth advantage entirely. Had drought continued, they would have probably been starved, indicated by the low level of their non-structural carbohydrate pool. 5Tillandsia flexuosa employs a stress-tolerance strategy both during germination and during the seedling stage. In its epiphytic habitat this strategy is clearly adaptive, considering the predictable briefness of moisture availability throughout the year and the low competition pressure that allows the very slow growth typically seen in adults and seedlings. These conditions characterize not only the dry-forest habitat of T. flexuosa, but all exposed epiphytic growing sites, so we expect this early stress-tolerance to be common among epiphytes in general. Still, a lower stress tolerance in species from wetter habitats may at least partly explain why T. flexuosa shares its dry-forest habitat with so few other vascular epiphytes. [source]


    FLR-2, the glycoprotein hormone alpha subunit, is involved in the neural control of intestinal functions in Caenorhabditis elegans

    The intestine plays an essential role in organism-wide regulatory networks in both vertebrates and invertebrates. In Caenorhabditis elegans, class 1 flr genes (flr-1, flr-3 and flr-4) act in the intestine and control growth rates and defecation cycle periods, while class 2 flr genes (flr-2, flr-5, flr-6 and flr-7) are characterized by mutations that suppress the slow growth of class 1 flr mutants. This study revealed that flr-2 gene controls antibacterial defense and intestinal color, confirming that flr-2 regulates intestinal functions. flr-2 encoded the only glycoprotein hormone alpha subunit in C. elegans and was expressed in certain neurons. Furthermore, FLR-2 bound to another secretory protein GHI-1, which belongs to a family of lipid- and lipopolysaccharide-binding proteins. A ghi-1 deletion mutation partially suppressed the short defecation cycle periods of class 1 flr mutants, and this effect was enhanced by flr-2 mutations. Thus, FLR-2 acts as a signaling molecule for the neural control of intestinal functions, which is achieved in a functional network involving class 1 and class 2 flr genes as well as ghi-1. These results are informative to studies of glycoprotein hormone signaling in higher animals. [source]


    Functional overlap between RecA and MgsA (RarA) in the rescue of stalled replication forks in Escherichia coli

    Escherichia coli RecA protein plays a role in DNA homologous recombination, recombination repair, and the rescue of stalled or collapsed replication forks. The mgsA (rarA) gene encodes a highly conserved DNA-dependent ATPase, whose yeast orthologue, MGS1, plays a role in maintaining genomic stability. In this study, we show a functional relationship between mgsA and recA during DNA replication. The mgsA recA double mutant grows more slowly and has lower viability than a recA single mutant, but they are equally sensitive to UV-induced DNA damage. Mutations in mgsA and recA cause lethality in DNA polymerase I deficient cells, and suppress the temperature-dependent growth defect of dnaE486 (Pol III ,-catalytic subunit). Moreover, recAS25P, a novel recA allele identified in this work, does not complement the slow growth of ,mgsA ,recA cells or the lethality of polA12 ,recA, but is proficient in DNA repair, homologous recombination, SOS mutagenesis and SOS induction. These results suggest that RecA and MgsA are functionally redundant in rescuing stalled replication forks, and that the DNA repair and homologous recombination functions of RecA are separated from its function to maintain progression of replication fork.     A GyrB-GyrA fusion protein expressed in yeast cells is able to remove DNA supercoils but cannot substitute eukaryotic topoisomerase II

    Background: Type II topoisomerases are a highly conserved class of enzymes which transport one double-stranded DNA segment through a transient break in another. Whereas the eukaryotic enzymes are homodimers of a single polypeptide, their bacterial homologues are homodimers of two independently coded protein subunits. Unlike eukaryotic topoisomerase II and bacterial topoisomerase IV, DNA gyrase is a bacterial type II topoisomerase which specializes in intramolecular DNA transport. Results: We have fused the Escherichia coli coding sequences for the proteins GyrB and GyrA, which comprise DNA gyrase. This fusion was expressed in yeast cells and yielded the expected full-length protein product. When it was expressed in ,top1- top2-4 yeast cells, the fusion protein compensated their slow growth and reverted their elevated chromosomal excision of ribosomal genes. Furthermore, it removed DNA positive supercoils. The fusion protein, however, was unable to complement the temperature-dependent lethality of top2-4 cells. Conclusion: Fusion of the E. coli GyrB and GyrA proteins leads to a catalytically active topoisomerase which compensates several phenotypic traits attributed to unconstrained DNA supercoiling in topoisomerase-deficient cells. However, since the fusion protein cannot substitute for topoisomerase II, it may be efficient in intramolecular but not intermolecular DNA passage, resembling the catalytic properties of DNA gyrase.     GROWTH OF CRUSTOSE LICHENS: A REVIEW

    ABSTRACT. Crustose species are the slowest growing of all lichens. Their slow growth and longevity, especially of the yellow-green Rhizocarpon group, has made them important for surface-exposure dating (lichenometry). This review considers various aspects of the growth of crustose lichens revealed by direct measurement including: 1) early growth and development; 2) radial growth rates (RGR, mm yr,1); 3) the growth rate,size curve; and 4) the influence of environmental factors. Many crustose species comprise discrete areolae that contain the algal partner growing on the surface of a non-lichenized fungal hypothallus. Recent data suggest that ,primary' areolae may develop from free-living algal cells on the substratum while ,secondary' areolae develop from zoospores produced within the thallus. In more extreme environments, the RGR of crustose species may be exceptionally slow but considerably faster rates of growth have been recorded under more favourable conditions. The growth curves of crustose lichens with a marginal hypothallus may differ from the ,asymptotic' type of curve recorded in foliose and placodioid species; the latter are characterized by a phase of increasing RGR to a maximum and may be followed by a phase of decreasing growth. The decline in RGR in larger thalli may be attributable to a reduction in the efficiency of translocation of carbohydrate to the thallus margin or to an increased allocation of carbon to support mature ,reproductive' areolae. Crustose species have a low RGR accompanied by a low demand for nutrients and an increased allocation of carbon for stress resistance; therefore enabling colonization of more extreme environments.     Development of a swine model of secondary liver tumor from a genetically induced swine fibroblast cell line

    Abstract Aim. Metastatic disease is the most common liver tumor. Although alternative therapies have been developed for non-surgical candidates, those therapies lacked ideal testing prior to clinical application because of a paucity of large animal models. The purpose of the present study was to develop a model for secondary liver tumor in a large animal. Material and methods. Fibroblasts were isolated from swine ear lobules and then transfected with amphotrophic retroviruses encoding human or murine genetic material (hTERT, p53DD, cyclinD-1, CDK4R24C, Myc T58A, RasG12V). Transformed cell lines were finally inoculated subcutaneously (s.c.) into: 1) immunodeficient mice (nude), 2) immunocompetent mice (wild type), 3) immunosuppressed swine (under tacrolimus or corticosteroids), 4) immunocompetent swine, and 5) into the liver and portal circulation of swine under steroid-based immunosuppression. Results. In the murine model, tumor growth was evident in 100% of the nude mice (n=5), with a peak size of 20 mm (15.22±4.5 mm; mean±SD) at the time of sacrifice (3 weeks). Tumor growth was evident in 71% of the wild mice (n=21), with a peak size of 7.8 mm (4.19±1.1 mm) by the third week of growth. In the swine model, tumor growth was evident in 75% (3/4 ears; n=2) of swine under tacrolimus-based immunosuppression versus 50% of swine under steroids-based immunosuppression (n=2). Tumor growth was slow in two animals, while in one animal the tumor was larger with a peak growth of 42 mm at 3 weeks. The tumor pattern in the ear lobules was characterized by slow growth, with a peak size of 6,8 mm in the immunocompetent swine at 3 weeks. All tumors were shown to be malignant by histology. In contrast, inoculums of the transformed fibroblast cell line in swine livers showed no evidence of tumor growth at 3 weeks. Conclusions. Development of a transformed swine fibroblast cell line was successful, resulting in an in vivo malignant tumor. Cell line inoculums had tumorigenic properties in nude mice, wild-type mice, and immunosuppressed swine, as judged by uncontrolled cell growth, invasion of surrounding tissue, neoangiogenesis, and invasion of normal vasculature, resulting in the formation of tumor nodules. Such properties were not observed in swine upon inoculation into the liver/portal circulation. [source]


    Life Cycle, Feeding and Production of Isoptena serricornis(Pictet, 1841) (Plecoptera, Chloroperlidae)

    Abstract Some aspects of the biology and ecology (life cycle, feeding and production) of a population of Isoptena serricornis in the Rudava River (Slovakia) are studied, reported and discussed. The life cycle is annual, with slow growth in autumn-winter and fast growth in late summer and spring. The growth decreased two weeks before the Fall Equinox and increased two weeks after the Spring Equinox. The flight period spans from the end of May to the beginning of July. The presence of large sand particles in the gut of all studied nymphs is of note, and indicates that I. serricornis acts as a deposit-collector species. Nymphal food is principally composed of detritus, unicellular organisms and, in nymphs of intermediate or large size, Chironomidae larvae. Adult food is composed fundamentally of different types of pollen grains. Males usually have lower food content than females. Annual production of this species (,694,750 mg · m,2) is very high in relation to other previously studied Chloroperlidae. This is probably largely responsible for I. serricornis being one of the most abundant components of the macroinvertebrate community in its habitat in the Rudava River. A negative correlation between production and temperature was observed. [source]


    An econometric model of nonlinear dynamics in the joint distribution of stock and bond returns

    This paper considers a variety of econometric models for the joint distribution of US stock and bond returns in the presence of regime switching dynamics. While simple two- or three-state models capture the univariate dynamics in bond and stock returns, a more complicated four-state model with regimes characterized as crash, slow growth, bull and recovery states is required to capture their joint distribution. The transition probability matrix of this model has a very particular form. Exits from the crash state are almost always to the recovery state and occur with close to 50% chance, suggesting a bounce-back effect from the crash to the recovery state.     Two modes of mitochondrial dysfunction lead independently to lifespan extension in Caenorhabditis elegans

    Summary In Caenorhabditis elegans, longevity is increased by a partial loss-of-function mutation in the mitochondrial complex III subunit gene isp-1. Longevity is also increased by RNAi against the expression of a variety of mitochondrial respiratory chain genes, including isp-1, but it is unknown whether the isp-1(qm150) mutation and the RNAi treatments trigger the same underlying mechanisms of longevity. We have identified nuo-6(qm200), a mutation in a conserved subunit of mitochondrial complex I (NUDFB4). The mutation reduces the function of complex I and, like isp-1(qm150), results in low oxygen consumption, slow growth, slow behavior, and increased lifespan. We have compared the phenotypes of nuo-6(qm200) to those of nuo-6(RNAi) and found them to be distinct in crucial ways, including patterns of growth and fertility, behavioral rates, oxygen consumption, ATP levels, autophagy, and resistance to paraquat, as well as expression of superoxide dismutases, mitochondrial heat-shock proteins, and other gene expression markers. RNAi treatments appear to generate a stress and autophagy response, while the genomic mutation alters electron transport and reactive oxygen species metabolism. For many phenotypes, we also compared isp-1(qm150) to isp-1(RNAi) and found the same pattern of differences. Most importantly, we found that, while the lifespan of nuo-6, isp-1 double mutants is not greater than that of the single mutants, the lifespan increase induced by nuo-6(RNAi) is fully additive to that induced by isp-1(qm150), and the increase induced by isp-1(RNAi) is fully additive to that induced by nuo-6(qm200). Our results demonstrate that distinct and separable aspects of mitochondrial biology affect lifespan independently. [source]


    The relationship between growth and mortality for seven co-occurring tree species in the southern Appalachian Mountains

    Summary 1Slow growth is associated with high mortality risk for trees, but few data exist to assess interspecific differences in the relationship between growth and mortality. Here we compare low growth tolerance for seven co-occurring species in the southern Appalachian Mountains: Acer rubrum, Betula lenta, Cornus florida, Liriodendron tulipifera, Quercus prinus, Quercus rubra and Robinia pseudo-acacia. 2For all species, mortality was greater for understorey individuals than for canopy trees. Species varied widely in the length of growth decline prior to death, ranging from 6 years for L. tulipifera to more than 12 years for Q. rubra. 3Growth-mortality functions differ among species, but we found little evidence of a trade-off between tolerance of slow growth and an ability to show rapid growth in high light conditions. 4A. rubrum stands out in its ability both to grow rapidly and to tolerate slow growth, suggesting that its density may increase at our study site as in other parts of the eastern United States. In contrast, C. florida shows high mortality (15% per annum) as a result of infection with dogwood anthracnose. 5We modified a forest simulation model, LINKAGES (which assumes that all species have the same ability to tolerate slow growth), to include our functions relating growth and mortality. The modified model gives radically altered predictions, reinforcing the need to rethink and re-parameterize existing computer models with field data. [source]


    Population structure, age and growth of macrourid fish from the upper slope of the Eastern-Central Mediterranean

    Hymenocephalus italicus, Nezumia sclerorhynchus and Coelorhynchus coelorhynchus were found in 80, 75 and 69% of trawl hauls carried out between 250 and 750 m on the upper slope of the Ionian Sea. The abundance of H. italicus and N. sclerorhynchus increased with depth while in C. coelorhynchus the highest densities were observed in the uppermost 500 m. In all three grenadiers the average size increased with depth. The populations had a multimodal sizefrequency distribution. In H. italicus adults were generally more represented in the population and the abundance of juveniles varied with seasons. In N. sclerorhynchus and mostly in C. coelorhynchus the bulk of the population was generally made up of small individuals the year round. In N. sclerorhynchus a seasonal pattern was shown in the depth distribution of juveniles. The sex ratio was in favour of females in larger specimens and in each bathymetric stratum. Seasonal growth was detected in the otoliths of the three species. Maximum ages were around 9 years in H. italicus and N. sclerorhynchus; 8 years in C. coelorhynchus. Although some differences have been detected in the population ecology of the three species, they are characterized by a prolonged recruitment during the year, slow growth, longevity and delayed maturity.     Advances in Research on Genetically Engineered Plants for Metal Resistance

    Abstract The engineering application of natural hyperaccumulators in removing or inactivating metal pollutants from soil and surface water in field trials mostly presents the insurmountable shortcoming of low efficiency owing to their little biomass and slow growth. Based on further understanding of the molecular mechanism of metal uptake, translocation, and also the separation, identification, and cloning of some related functional genes, this article highlights and summarizes in detail the advances in research on transgenic techniques, such as Agrobacterium tumefaciens -mediated transformation and particle bombardment, in breeding of plants for metal resistance and accumulation, and points out that deepening the development of transgenic plants is one of the efficient approaches to improving phytoremediation efficiency of metal-contaminated environments.     High-Temperature Tensile Strength of Er2O3 -Doped ZrO2 Single Crystals

    The deformation and fracture mechanisms in tension were studied in single-crystal Er2O3 -doped ZrO2 monofilaments processed by the laser-heated floating zone method. Tensile tests were carried out between 25° and 1400°C at different loading rates and the dominant deformation and fracture mechanisms were determined from the shape of the stress,strain curves, the morphology of the fracture surfaces, and the evidence provided by monofilaments deformed at high temperature and broken at ambient temperature. The tensile strength presented a minimum at 600°,800°C and it was controlled by the slow growth of a crack from the surface. This mechanism was also dominant in some monofilaments tested at 1000°C and above, while others showed extensive plastic deformation before fracture at these temperatures. The strength of plastically deformed monofilaments was significantly higher than those which failed by slow crack growth due to the marked strain hardening capacity of this material. [source]


    Abnormal Grain Growth in Alumina with Anorthite Liquid and the Effect of MgO Addition

    Abnormal grain growth (AGG) in alumina with anorthite liquid has been observed with varying anorthite and MgO contents, at 1620°C. When only anorthite is added to form a liquid matrix, the grain,liquid interfaces have either flat or hill-and-valley shapes indicating atomically flat (singular) structures. The large grains grow at accelerated rates to produce AGG structures with large grains elongated along their basal planes. This is consistent with the slow growth at low driving forces and accelerated growth above a critical driving force predicted by the two-dimensional nucleation theory of surface steps. With increasing temperature, the AGG rate increases. The number density of the abnormally large grains increases with increasing anorthite content. The addition of MgO causes some grain,liquid interfaces to become curved and hence atomically rough. The grains also become nearly equiaxed. With increasing MgO content the number density of the abnormally large grains increases until the grain growth resembles normal growth. This result is qualitatively consistent with the decreasing surface step free energy associated with partial interface roughening transition. [source]


    Land under pressure: soil conservation concerns and opportunities for Ethiopia

    Abstract This paper evaluates the future impact of soil degradation on national food security and land occupation in Ethiopia. It applies a spatial optimization model that maximizes national agricultural revenues under alternative scenarios of soil conservation, land accessibility and technology. The constraints in the model determine whether people remain on their original site, migrate within their ethnically defined areas or are allowed a transregional migration. Key to this model is the combination of a water erosion model with a spatial yield function that gives an estimate of the agricultural yield in its geographical dependence of natural resources and population distribution. A comparison of simulated land productivity values with historical patterns shows that results are interpretable and yield more accurate outcomes than postulating straightforward reductions in yield or land area for each geographic entity. The results of the optimization model show that in absence of soil erosion control, the future agricultural production stagnates and results in distressing food shortages, while rural incomes drop dramatically below the poverty line. Soil conservation and migration support a slow growth, but do not suffice to meet the expected food demand. In a transregional migration scenario, the highly degraded areas are exchanged for less affected sites, whereas cultivation on already substantially degraded soils largely continues when resettlement is confined to the original ethnic,administrative entity. A shift to modern technology offers better prospects and moderates the migration, but soil conservation remains indispensable, especially in the long term. Finally, an accelerated growth of non-agricultural sectors further alleviates poverty in the countryside, contributing to higher income levels of the total population and, simultaneously, relieving the pressure on the land through rural,urban migration.     Flow cytometric method for quantifying viable Mycoplasma agassizii, an agent of upper respiratory tract disease in the desert tortoise (Gopherus agassizii)

    Abstract Aims:,Mycoplasma agassizii can cause upper respiratory tract disease in the threatened desert tortoise of the Southwestern United States. Two technical challenges have impeded critical microbiological studies of this microorganism: (i) its small size limits the use of light microscopy for cell counting and (ii) its extremely slow growth in broth and agar cultures impedes colony counting. Our aim was to develop a rapid and sensitive flow cytometric method using a vital fluorescent dye to enumerate viable M. agassizii cells. Methods and Results:, Here, we demonstrate that the nonfluorescent molecule 5-carboxyfluorescein (5-CF) diacetate acetoxymethyl ester penetrates M. agassizii cell membranes and it is converted in the cytoplasm to the fluorescent molecule 5-CF by the action of intracellular esterases. Labelled mycoplasma cells can be easily detected by flow cytometry, and cultures with as few as 100 viable mycoplasma cells ml,1 can be labelled and counted in less than 1 h. Experiments using temperature-induced cell death demonstrated that only viable M. agassizii cells are labelled with this procedure. Conclusions:, A rapid and sensitive flow cytometric technique has been developed for enumerating viable M. agassizii cells. Significance and Impact of the Study:, This technique should facilitate basic immunological, biochemical and pharmacological studies of this important pathogen which may lead to new diagnostic and therapeutic methods.     Unusual evolution of an Epstein,Barr virus-associated leiomyosarcoma occurring after liver transplantation

    Abstract: We report the case of a child who developed, 2 yr after orthotopic liver transplantation (OLTx) for biliary atresia, a multi-focal hepatic tumor with lymphonodular metastases, identified as an Epstein,Barr virus (EBV)-associated leiomyosarcoma. Chemotherapy was given without tumor response. Subsequently, slow growth of the tumor was observed. Immunosuppression was tapered and stopped 9 yr after transplantation. At the present time, 12 yr after the discovery of the first hepatic lesions, the patient is alive and completely symptom-free, the abdominal masses are stable, and liver function tests are completely normal. Smooth muscle tumors are increasingly recognized in children with various immunodeficiencies occurring after organ transplantation. This unusual evolution of a clinically aggressive tumor into a stable disease after restoration of immunity confirms that the immune status of the patient is a crucial factor. [source]


    Chorionic villus sampling (CVS) and fluorescence in situ hybridization (FISH) for a rapid first-trimester prenatal diagnosis

    Abstract Objectives Early diagnosis of unbalanced chromosomal abnormalities can be crucial in minimizing the trauma caused by an elective abortion. Chorionic villus sampling (CVS) can be performed from 9 weeks of gestation. However, two major problems are encountered in fetal karyotyping using cultured cells from chorionic villi: the relatively slow growth of these cells in culture, which delays the diagnosis, and the occurrence of maternal cell contamination (MCC). With FISH, a result can be obtained within 24 h, and, as no cell culturing is involved, the problem of MCC is minimized. Methods Thirty-two women undergoing CVS between 9 and 12 weeks of gestation were offered FISH analysis in addition to the standard chromosome analysis. Results FISH was informative in all of the cases tested. Eleven aneuploidies were detected in cases of hygroma or abnormal nuchal translucency and two out of four fetuses from parental translocation were unbalanced. The decision to perform early termination of these chromosomally abnormal pregnancies was based on FISH results and ultrasound abnormalities, without waiting for karyotype results. Conclusion The present study confirms that the association of FISH and CVS allows a rapid and early prenatal diagnosis, and emphasizes that this association is of great benefit in cases of known parental balanced translocation or when hygroma is detected by ultrasonography.     Phytoremediation of arsenic in mining-contaminated areas: The future of transgenic technology

    A considerable number of contaminated mining sites in Europe and other parts of the world pose environmental hazards. Given the multifaceted benefits of phytoremediation, screening of plant communities grown in contaminated areas is being conducted to identify hyperaccumulating plant species. A few arsenic (As) hyperaccumulating plants are found in tropical countries; however, generally, they are not grown in contaminated mining sites of cold and temperate countries (Europe and other parts of the world). The transgenic plants identified to date are not believed to be suitable for commercial use of phytoremediation. A few tolerant plant species in mining sites that are found to have elevated As levels primarily concentrate As in their roots. The remediation potential of many of these tolerant plants is limited because of their slow growth and low biomass. It is the first paper to calculate world distribution for individuals based entirely on household surveys from 91 countries, and adjusted for differences in purchasing power parity between countries. Measured by the Gini index, inequality increased from 63 in 1988 to 66 in 1993. The increase was driven more by differences in mean incomes between countries than by inequalities within countries. The most important contributors were rising urban-rural differences in China, and slow growth of rural incomes in South Asia compared to several large developed economies. [source]


    Modelling life history strategies with capture,recapture data: Evolutionary demography of the water skink Eulamprus tympanum

    Abstract Matrix population models, elasticity analysis and loop analysis can potentially provide powerful techniques for the analysis of life histories. Data from a capture,recapture study on a population of southern highland water skinks (Eulamprus tympanum) were used to construct a matrix population model. Errors in elasticities were calculated by using the parametric bootstrap technique. Elasticity and loop analyses were then conducted to identify the life history stages most important to fitness. The same techniques were used to investigate the relative importance of fast versus slow growth, and rapid versus delayed reproduction. Mature water skinks were long-lived, but there was high immature mortality. The most sensitive life history stage was the subadult stage. It is suggested that life history evolution in E. tympanum may be strongly affected by predation, particularly by birds. Because our population declined over the study, slow growth and delayed reproduction were the optimal life history strategies over this period. Although the techniques of evolutionary demography provide a powerful approach for the analysis of life histories, there are formidable logistical obstacles in gathering enough high-quality data for robust estimates of the critical parameters.     Induction of mitotic cell death in cancer cells by small interference RNA suppressing the expression of RecQL1 helicase

    RecQL1 DNA helicase of the human RecQ helicase family participates in DNA repair and recombination pathways during cell-cycle replication. When we examined the effect of RecQL1 suppression on cell growth, we found that RecQL1 silencing by small interference RNA efficiently prevented proliferation of a wide range of cancer cells by inducing mitotic catastrophe and mitotic cell death. In contrast, such mitotic cell death was not seen in the growing normal fibroblasts used as controls, even if RecQL1 expression was fully downregulated. Our results support the hypothesis that endogenous DNA damage that occurs during DNA replication and remains unrepaired in cancer cells due to RecQL1 silencing induces cancer cell-specific mitotic catastrophe through a less-strict checkpoint in cancer cells than in normal cells. We speculate that normal cells are exempt from such mitotic cell death, despite slow growth, because cell-cycle progression is controlled strictly by a strong checkpoint system that detects DNA damage and arrests progression of the cell cycle until DNA damage is repaired completely.     3465: Medical cancer therapy of lacrimal gland tumours

    Purpose The most common malignant epithelial cancer of the lacrimal gland is the adenoid cystic carcinoma (ACC). Despite a slow growth, ACCs are ultimately associated with a poor outcome. Methods Given the rarity of this disease, there are actually no conclusive recommendations for optimal therapy of this tumor. Results Surgery and postoperative radiation therapy is commonly used in the initial local treatment of ACC of the lacrimal gland. In high-risk recurrence patients, concomitant platinum-based chemoradiation should be discussed in an attempt to enhance radiosensitivity. While encouraging responses were reported with intraarterial neoadjuvant chemotherapy, this strategy was associated with substantial toxicity and should not be recommended outside of clinical trials. In the metastatic setting, systemic therapy is the only available option if no surgery and/or radiation is feasible. Although some tumour shrinkage has been reported with intravenous chemotherapy, only dismal objective response rates were achieved. Most active drugs remain anthracyclines and platinum compounds. Drug combinations do not seem to add much efficacy. More recently, non-cytotoxic molecularly targeted agents have emerged and demonstrated significant efficacy in several tumour types. These agents modulate specific targets thought to be essential for tumour proliferation and/or angiogenesis. c-KIT, PDGFR,, EGFR, and VEGFR are transmembrane receptors with oncogenic tyrosine kinase activity that are commonly overexpressed in ACC. The use of drugs triggering these targets has been disappointing so far. Conclusion The recent identification of a hallmark gene fusion transcript thought to activate critical targets involved in apoptosis, cell cycle control, cell growth and angiogenesis, heralds new treatment promise.
