A taxometric study of alcohol abuse and dependence in a general population sample: evidence of dimensional latent structure and implications for DSM-V

ABSTRACT Aims To explore, with the aid of taxometric analysis, whether alcohol abuse and alcohol dependence are each conceptualized most effectively as single latent dimensions or distinct latent categories. Design Data were taken from a nationally representative cross-sectional epidemiological survey of psychiatric and substance use disorders. Setting General population of Australia. Participants A subsample of all respondents who had consumed at least 12 drinks in the year prior to the survey and who had consumed at least three drinks on at least one single day (n = 4920 of a possible 10 641). Measurements DSM-IV criteria for alcohol abuse and dependence were assessed with the Composite International Diagnostic Interview, version 2.1. Two independent taxometric procedures, MAXimum EIGenvalue (MAXEIG) and mean above minus below a cut (MAMBAC), together with analysis of simulated dimensional and categorical data sets, were carried out. Findings Consistent evidence was found for a single latent dimension underlying the symptoms of alcohol dependence. Less consistent evidence of dimensionality was found for the symptoms of alcohol abuse. Conclusions These findings support the growing consensus regarding the need for continuous measures of alcohol use disorders to complement the traditional categorical representations in upcoming versions of the major psychiatric classification systems. Influence of predator stress on the consolidation versus retrieval of long-term spatial memory and hippocampal spinogenesis

Abstract We have studied the influence of predator stress (30 min of cat exposure) on long-term (24 h) spatial memory and the density of spines in basilar dendrites of CA1 neurons. Predator stress occurred either immediately before water maze training (Stress Pre-Training) or before the 24 h memory test (Stress Pre-Retrieval). The Control (nonstress) group exhibited excellent long-term spatial memory and a robust increase in the density of stubby, but not mushroom, shaped spines. The Stress Pre-Training group had impaired long-term memory and did not exhibit any changes in spine density. The Stress Pre-Retrieval group was also impaired in long-term memory performance, but this group exhibited an increase in the density of stubby, but not mushroom, shaped spines, which was indistinguishable from the control group. These findings indicate that: (1) A single day of water maze training under control conditions produced intact long-term memory and an increase in the density of stubby spines in CA1; (2) Stress before training interfered with the consolidation of information into long-term memory and suppressed the training-induced increase in spine density; and (3) Stress immediately before the 24 h memory test trial impaired the retrieval of the stored memory, but did not reverse the training-induced increase in CA1 spine density. Overall, this work provides evidence of structural plasticity in dendrites of CA1 neurons which may be involved in the consolidation process, and how spinogenesis and memory are modulated by stress. © 2006 Wiley-Liss, Inc. Eggload dynamics and oviposition rate in a wild population of a parasitic wasp

Summary 1.,This paper develops and tests in the field a model that predicts the oviposition rate and the rate of egg maturation of a synovigenic parasitic wasp during a foraging period. The parasitoid is Aphytis melinus, the highly successful biological control agent of California redscale (Aondiniella aurantii), a pest of citrus worldwide. 2.,Females were sampled in the interior canopy of grapefruit trees either just before and at the end of the foraging period over 2 days and were dissected to determine the starting and ending eggload distributions. A group of females was caught before the onset of activity and kept in vials in the field in trees with honey but without access to hosts during the foraging period. Their eggload at the end of the day was used to estimate the egg maturation rate during the foraging period. 3.,Two stochastic models are used to predict the eggload distribution at the end of the day. Both use the observed starting eggload distribution, the observed length of the foraging period and the estimated rate of egg maturation. The model providing the better fit uses an oviposition rate which is an increasing function of the eggload. 4.,The eggload does not attain a steady state distribution during a foraging period. One-third to one-half of the population is predicted to experience egg-limitation at some time during the foraging period. Five percent of the population will experience egg-limitation a second time within a single day. 5.,The common occurrence of egg-limitation over a single day and the relatively high rate of change of states between egg- and time-limitation imply that the rate of nutrient acquisition and use are likely to be subject to strong evolutionary pressures. Disparities in Alcohol-Related Problems Among White, Black, and Hispanic Americans

Background:, This study assesses racial/ethnic disparities in negative social consequences of drinking and alcohol dependence symptoms among white, black, and Hispanic Americans. We examine whether and how disparities relate to heavy alcohol consumption and pattern, and the extent to which social disadvantage (poverty, unfair treatment, and racial/ethnic stigma) accounts for observed disparities. Methods:, We analyzed data from the 2005 U.S. National Alcohol Survey, a nationally representative telephone-based survey of adults ages 18 and older (N = 6,919). Given large racial/ethnic differences in abstinence rates, core analyses were restricted to current drinkers (N = 4,080). Logistic regression was used to assess disparities in alcohol-related problems at 3 levels of heavy drinking, measured using a composite variable incorporating frequency of heavy episodic drinking, frequency of drunkenness, and maximum amount consumed in a single day. A mediational approach was used to assess the role of social disadvantage. Results:, African American and Hispanic drinkers were significantly more likely than white drinkers to report social consequences of drinking and alcohol dependence symptoms. Even after adjusting for differences in heavy drinking and demographic characteristics, disparities in problems remained. The racial/ethnic gap in alcohol problems was greatest among those reporting little or no heavy drinking, and gradually diminished to nonsignificance at the highest level of heavy drinking. Social disadvantage, particularly in the form of racial/ethnic stigma, appeared to contribute to racial/ethnic differences in problems. Conclusions:, These findings suggest that to eliminate racial/ethnic disparities in alcohol-related problems, public health efforts must do more than reduce heavy drinking. Future research should address the possibility of drink size underestimation, identify the particular types of problems that disproportionately affect racial/ethnic minorities, and investigate social and cultural determinants of such problems. DNA extraction method for PCR in mycorrhizal fungi

Aims: To develop a simple and rapid DNA extraction protocol for PCR in mycorrhizal fungi. Methods and Results: The protocol combines the application of rapid freezing and boiling cycles and passage of the extracts through DNA purification columns. PCR amplifiable DNA was obtained from a number of endo- and ecto-mycorrhizal fungi using minute quantities of spores and mycelium, respectively. Conclusions: DNA extracted following the method, was used to successfully amplify regions of interest from high as well as low copy number genes. The amplicons were suitable for further downstream applications such as sequencing and PCR-RFLPs. Significance and Impact of the Study: The protocol described is simple, short and facilitates rapid isolation of PCR amplifiable genomic DNA from a large number of fungal isolates in a single day. The method requires only minute quantities of starting material and is suitable for mycorrhizal fungi as well as a range of other fungi. [source]


Lifespan and patterns of accumulation and mobilization of nutrients in the sugar-fed phorid fly, Pseudacteon tricuspis

Abstract., The effect of sugar feeding on the survival of adult phorid fly Pseudacteon tricuspis is investigated. Flies fed 25% sucrose in aqueous solution continuously throughout their lifespan have greater longevity (mean ± SE longevity: female = 7.9 ± 0.8 days, male = 8.9 ± 0.9 days) than completely starved (provided no water and no sugar solution) flies, sugar-starved (provided water only) flies, or flies fed sugar solution only on their first day of adult life. Completely starved flies rarely lived beyond one day. Provision of water increases longevity by 2 days, and one full day of sugar feeding further increases longevity by an additional 1,2 days. Flies fed 50% sucrose have similar survivorship as those fed 25% sucrose. The temporal patterns of nutrient accumulation and utilization are also compared in P. tricuspis fed different diets: sugar-starved, sucrose-fed on the first day of adult life only, and sucrose-fed continuously. Adult P. tricuspis emerge with no gut sugars, and only minimal amounts of body sugars and glycogen. Although the levels of body sugars and glycogen decline gradually in sugar-starved flies, a single day of sugar feeding results in the accumulation of maximum amounts of gut sugars, body sugars and glycogen. High levels of these nutrients are maintained in female and male phorid flies fed on sucrose continuously over the observation period, whereas nutrient levels decline in flies fed only on the first day of life, beginning 1 day postfeeding. Female and male P. tricuspis emerge with an estimated 12.3 ± 2.3 and 7.2 ± 1 g of lipid reserves per fly, respectively. These teneral amounts represent the highest lipid levels detected in adult flies, irrespective of their diet, and are maintained over the life times of sucrose-fed female and male flies, but declined steadily in sugar-starved females. These data suggest that adult P. tricuspis are capable of converting dietary sucrose to body sugars and glycogen, but not lipids. [source]


Computer-aided NMR assay for detecting natively folded structural domains,

Abstract Structural genomics projects require strategies for rapidly recognizing protein sequences appropriate for routine structure determination. For large proteins, this strategy includes the dissection of proteins into structural domains that form stable native structures. However, protein dissection essentially remains an empirical and often a tedious process. Here, we describe a simple strategy for rapidly identifying structural domains and assessing their structures. This approach combines the computational prediction of sequence regions corresponding to putative domains with an experimental assessment of their structures and stabilities by NMR and biochemical methods. We tested this approach with nine putative domains predicted from a set of 108 Thermus thermophilus HB8 sequences using PASS, a domain prediction program we previously reported. To facilitate the experimental assessment of the domain structures, we developed a generic 6-hour His-tag-based purification protocol, which enables the sample quality evaluation of a putative structural domain in a single day. As a result, we observed that half of the predicted structural domains were indeed natively folded, as judged by their HSQC spectra. Furthermore, two of the natively folded domains were novel, without related sequences classified in the Pfam and SMART databases, which is a significant result with regard to the ability of structural genomics projects to uniformly cover the protein fold space. Flow cytometry-assisted purification and proteomic analysis of the corticotropes dense-core secretory granules

Abstract The field of organellar proteomics has emerged as an attempt to minimize the complexity of the proteomics data obtained from whole cell and tissue extracts while maximizing the resolution on the protein composition of a single subcellular compartment. Standard methods involve lengthy density-based gradient and/or immunoaffinity purification steps followed by extraction, 1-DE or 2-DE, gel staining, in-gel tryptic digestion, and protein identification by MS. In this paper, we present an alternate approach to purify subcellular organelles containing a fluorescent reporter molecule. The gel-free procedure involves fluorescence-assisted sorting of the secretory granules followed by gentle extraction in a buffer compatible with tryptic digestion and MS. Once the subcellular organelle labeled, this procedure can be done in a single day, requires no major modification to any instrumentation and can be readily adapted to the study of other organelles. When applied to corticotrope secretory granules, it led to a much enriched granular fraction from which numerous proteins could be identified through MS. [source]


Off label and unlicensed prescribing in a specialist oncology center in Australia

Abstract Aim: Oncology is an area with a high prevalence of off-label and unlicensed prescribing. A previous audit conducted in 2001 at the Peter MacCallum Cancer Centre, a specialist oncology hospital, showed that 22% of all prescriptions were either off-label or unlicensed. This study aimed to determine if the rates of off-label and unlicensed prescribing in oncology had changed between 2001 and 2008. Methods: All prescriptions at the Peter MacCallum Cancer Centre were reviewed on a single day in March 2008. Each prescription was classified as licensed, off-label or unlicensed by comparing to the Approved Product Information (API). Results: The medications of 132 patients were assessed. Among the 1094 prescriptions, 382 (35%) were off-label and 44 (4%) were unlicensed. 112 (85%) patients received at least one off-label or unlicensed drug. Conclusion: The results of the audit suggest that the level of off-label prescribing increased by 17% since 2001. This study confirmed the extensive off-label and unlicensed drug usage as suggested by the literature and demonstrated a higher rate of off-label prescribing than the previous audit in 2001. The results demonstrate that 85% of all cancer patients in the study were prescribed at least one drug that had not been fully tested by the regulatory approval process. [source]


Gestational stage sensitivity to ultrasound effect on postnatal growth and development of mice

Abstract BACKGROUND: An experiment was conducted to find out whether ultrasound exposure leads to changes in postnatal growth and development in the mouse. METHODS: A total of 15 pregnant Swiss albino mice were exposed to diagnostic levels of ultrasound (3.5 MHz, 65 mW/cm2, ISPTP = 1 mW/cm2 Intensity(Spatial Peak-Temporal Peak), ISATA = 240 mW/cm2 Intensity(Spatial Average-Temporal Average)) for 30 min for a single day between days 10 and 18 of gestation (GD 10,18). Virgin female mice were placed with same age group males for mating in the ratio 2 females : 1 male and examined the next morning for the presence of vaginal plug, a sign of successful copulation. The females with vaginal plugs were separated and labeled as 0-day pregnant. Maternal vaginal temperature was also measured. A sham exposed control group of 15 pregnant mice was maintained for comparison. All exposed as well as control animals were left to complete gestation and parturition. Their offspring were used in our further studies. They were monitored during early postnatal life for standard developmental markers, postnatal mortality, body weight, body length, head length, and head width, and growth restriction was recorded up to 6 weeks of age. RESULTS: An exposure to ultrasound induced nonsignificant deviations in the maternal vaginal temperature or developmental markers. Significant low birth weight was observed in the present study, after exposure at GD 11, 12, 14, and 16. However, 14 and 16 days postcoitus during the fetal period appears to be the most sensitive to the ultrasound effect, in view of the number of different effects as well as severity of most of the observed effects when exposed on these gestation days. CONCLUSIONS: The results indicate that diagnostic ultrasound can induce harmful effects on mouse growth and development when given at certain critical periods of gestation. Rainfall distribution is the main driver of runoff under future CO2 -concentration in a temperate deciduous forest

Abstract Reduced stomatal conductance under elevated CO2 results in increased soil moisture, provided all other factors remain constant. Whether this results in increased runoff critically depends on the interaction of rainfall patterns, soil water storage capacity and plant responses. To test the sensitivity of runoff to these parameters under elevated CO2, we combine transpiration and soil moisture data from the Swiss Canopy Crane FACE experiment (SCC, 14 30,35 m tall deciduous broad-leaved trees under elevated CO2) with 104 years of daily precipitation data from an adjacent weather station to drive a three-layer bucket model (mean yearly precipitation 794 mm). The model adequately predicts the water budget of a temperate deciduous forest and runoff from a nearby gauging station. A simulation run over all 104 years based on measured sap flow responses resulted in only 5.5 mm (2.9%) increased ecosystem runoff under elevated CO2. Out of the 37 986 days (1 January 1901,31 December 2004), only 576 days produce higher runoff in the elevated CO2 scenario. Only 1 out of 17 years produces a CO2 -signal >20 mm a,1, which mostly depends on a few single days when runoff under elevated CO2 exceeds runoff under ambient conditions. The maximum signal for a double preindustrial CO2 -concentration under the past century daily rainfall regime is an additional runoff of 46 mm. More than half of all years produce a signal of <5 mm a,1, because trees consume the ,extra' moisture during prolonged dry weather. Increased runoff under elevated CO2 is nine times more sensitive to variations in rain pattern than to the applied reduction in transpiration under elevated CO2. Thus the key driver of increased runoff under future CO2 -concentration is the day by day rainfall pattern. We argue that increased runoff due to a first-order plant physiological CO2 -effect will be very small (<3%) in a landscape dominated by temperate deciduous forests, and will hardly increase flooding risk in forest catchments. Monthly rainfall sums are unsuitable to realistically model such CO2 effects. These findings may apply to other ecosystems with comparable soil water storage capacity. [source]
