    THE EFFECTS OF PERSONALITY SIMILARITY ON PEER RATINGS OF CONTEXTUAL WORK BEHAVIORS

    The present field study investigates whether rater-ratee similarity in the Big Five personality factors influences peer ratings of contextual work behaviors. It overcomes problems that previous studies have had by using a polynomial regression analysis and by correcting the potential biases from nonindependence. Using more than 500 peer dyads, we found that rater-ratee similarity in Conscientiousness, but not in other dimensions, was positively associated with peer ratings even after controlling for interpersonal affect. These results suggest that the observed effect of personality similarity may reflect actual behavioral differences rather than biases due to interpersonal affect. Implications of the findings are discussed along with recommendations for future research. [source]


    AN EXACT TEST FOR HAZARD SIMILARITY

    We analyze life history narratives and structured data derived from a study of serious female and male offenders interviewed when incarcerated as adolescents and followed up thirteen years later. We highlight shifts in the influence of friends and in the nature of friendship choices, and suggest how these changes can facilitate desistance processes. While key events (e.g., marriage) are important to an understanding of such changes, shifts in the actor's perspective and identity are also integral to the process of making successful network realignments. Similarities and differences by gender in the effects of adult social influence processes are also examined. [source]


    Beta diversity of geometrid moths (Lepidoptera: Geometridae) in an Andean montane rainforest

    Abstract. Turnover in species composition of the extremely species-rich family Geometridae (Lepidoptera) was investigated along an elevational gradient ranging from 1040 m to 2677 m above sea level. Moths were sampled using weak light traps (30 W) in three field periods in 1999 and 2000 in an Andean montane rainforest in the province of Zamora-Chinchipe in southern Ecuador. A total of 13 938 specimens representing 1010 species were analysed. Similarities of ensembles of all geometrid moths and of the subfamilies Ennominae and Larentiinae were calculated using the NESS index (with mmax). Ordinations performed using nonmetric multidimensional scaling (NMDS) and correspondence analysis depicted a gradual change of the ensembles along the altitudinal gradient. Extracted ordination scores significantly correlate with altitude (,0.97 , r , ,0.95, P < 0.001) and with ambient air temperature (0.93 , r , 0.97, P < 0.001). Temperature is therefore assumed to be the most important abiotic determinant responsible for the species turnover among the moths. Matrix correlation tests were performed in order to compare faunal matrices with matrices derived from available environmental factors. Both tree diversity and vegetation structure significantly correlate with faunal data, but tree diversity explains considerably more of the data variability (range: Mantel r = 0.81,0.83, P < 0.001) than vegetation structure (range: Mantel r = 0.35, P < 0.005 to r = 0.43, P < 0.001). Tree diversity also changes gradually and scores of the first NMDS dimension are highly significantly correlated with altitude (r = 0.98, P < 0.001). A common underlying factor such as ambient temperature might also be responsible for such vegetation changes. Additionally, simulated model data was developed that assumed a constant turnover of moth species and equal elevational ranges of all species involved. Despite the simplicity of the models, they fit empirical data very well (Mantel r > 0.80 and P < 0.001 in all models). [source]


    Genital system anatomy and development of Ovatella myosotis by three-dimensional computer visualization

    Abstract Adult anatomy as well as organogenesis of the genital system of the ellobiid pulmonate Ovatella myosotis is investigated in detail by means of serial sectioning and three-dimensional computer reconstruction and visualization. From the middle portion of the adult, which has four nidamental glands, a spermoviduct leads to a common genital aperture. From here two separate structures, the vas deferens and a groove on the body surface, lead anteriorly. The latter is termed the egg groove because it carries the egg ribbon anteriorly, a function that is recognized here for the first time in the Ellobiidae. The evolution of this structure is discussed. In development, the organ system arises from four separate anlagen: (1) the ovotestis anlage, (2) the pallial anlage giving rise to the hermaphrodite duct, fertilization pouch,spermatheca complex, nidamental glandular complex and spermoviduct, (3) the bursa copulatrix anlage and (4) the anlage of the copulatory organ, vas deferens and egg groove. This development mode strongly resembles that of the siphonariid Williamia radiata, supporting its interpretation as a plesiomorphy in Pulmonata. Similarities in development of primitive pulmonates and evolution in gastropods lead to the assumption that ontogenesis of this organ system reflects evolution to some degree. [source]


    Bridging the Divides: The Need for a Pragmatic Semiotics of Teacher Knowledge Research

    In this essay, we consider four approaches to research on teacher knowledge: the scholarship of teaching, action research and teacher research, narrative inquiry, and critical-cultural teacher research. Similarities and differences among these four approaches are highlighted. The most salient difference lies in the way each approach identifies different discourses as sources of distortion in teacher knowledge research. Although some divergence within a field of study can be a valuable source of debate and dialogue, we believe the differences identified here risk dividing the field of teacher knowledge research in unproductive ways. What is needed, we propose, is a semiotic theory that acknowledges the way teacher knowledge is irreducibly mediated by multiple discourses while preserving a commitment to the idea that individual teachers' experiences can be a source of novel and useful knowledge. We examine two semiotic theories , French poststructuralism and Charles Sanders Peirce's pragmatic semiotics , and critically assess how they might facilitate more constructive dialogue among differing conceptions of teachers' knowledge research.     Comparative sediment quality guideline performance for predicting sediment toxicity in Southern California, USA

    Abstract Several types of sediment quality guidelines (SQGs) are used by multiple agencies in southern California (USA) to interpret sediment chemistry data, yet little information is available to identify the best approaches to use. The objective of this study was to evaluate the predictive ability of five SQGs to predict the presence and absence of sediment toxicity in coastal southern California: the effects range-median quotient (ERMq), consensus moderate effect concentration (consensus MEC), mean sediment quality guideline quotient (SQGQ1), apparent effects threshold (AET), and equilibrium partitioning (EqP) for organics. Large differences in predictive ability among the SQGs were obtained when each approach was applied to the same southern California data set. Sediment quality guidelines that performed well in identifying nontoxic samples were not necessarily the best predictors of toxicity. In general, the mean ERMq, SQGQ1q, and consensus MECq approaches had a better overall predictive ability than the AET and EqP for organics approaches. In addition to evaluating the predictive ability of SQGs addressing chemical mixtures, the effect of an individual SQG value (DDT) was also evaluated for the mean ERMq with and without DDT. The mean ERMq without DDT had a better ability to predict toxic samples than the mean ERMq with DDT. Similarities in discriminatory ability between different approaches, variations in accuracy among SQG values for some chemicals, and the presence of complex mixtures of contaminants in most samples underscore the need to apply SQGs in combination, such as the mean quotient. Management objectives and SQG predictive ability using regional data should be determined beforehand so that the most appropriate SQG approach and critical values can be identified for specific applications.     The structure of the human allo-ligand HLA-B*3501 in complex with a cytochrome p450 peptide: Steric hindrance influences TCR allo-recognition

    Abstract Virus-specific T cell populations have been implicated in allo-recognition. The subdominant T cell receptor JL12 recognizes both HLA-B*0801 presenting the Epstein,Barr virus-derived peptide FLRGRAYGL and also HLA-B*3501 presenting the cytochrome p450 self peptide KPIVVLHGY. This cross-reactivity could promote the rejection of HLA-B*3501-positive cells in Epstein,Barr virus-exposed HLA-B*0801 recipients. LC13, the dominant TCR against the HLA-B*0801:FLRGRAYGL complex, fails to recognize HLA-B*3501:KPIVVLHGY. We report the 1.75-Angstrom resolution crystal structure of the human allo-ligand HLA-B*3501:KPIVVLHGY. Similarities between this structure and that of HLA-B*0801:FLRGRAYGL may facilitate cross-recognition by JL12. Moreover, the elevated peptide position in HLA-B*3501:KPIVVLHGY would provide steric hindrance to LC13, preventing it from interacting in the manner in which it interacts with HLA-B*0801:FLRGRAYGL. These findings are relevant to understanding the basis of T cell cross-reactivity in allo-recognition, optimal transplant donor-recipient matching and developing specific molecular inhibitors of allo-recognition.     Early-onset Alzheimer's disease with presenilin-1 M139V mutation: clinical, neuropsychological and neuropathological study

    The clinical, neuropsychological and neuropathological features of a patient with early-onset Alzheimer's disease as a result of the M139V presenilin-1 (PSEN-1) mutation are presented, and compared with previous reports of patients with the same mutation. Similarities, such as the age at onset and the relative preservation of naming skills, and differences, such as the significant basal ganglia, thalamic and cerebellar pathology, are noted. This clinical and pathological heterogeneity in patients with the same PSEN-1 mutation suggests phenotype modulation by genetic and/or epigenetic factors. [source]


    Similarities of stress concentrations in contact at round punches and fatigue at notches: implications to fretting fatigue crack initiation

    A linear elastic model of the stress concentration due to contact between a rounded flat punch and a homogeneous substrate is presented, with the aim of investigating fretting fatigue crack initiation in contacting parts of vibrating structures including turbine engines. The asymptotic forms for the stress fields in the vicinity of a rounded punch-on-flat substrate are derived for both normal and tangential loading, using both analytical and finite element methods. Under the action of the normal load, P, the ensuing contact is of width 2b which includes an initial flat part of width 2a. The asymptotic stress fields for the sharply rounded flat punch contact have certain similarities with the asymptotic stress fields around the tip of a blunt crack. The analysis showed that the maximum tensile stress, which occurs at the contact boundary due to tangential load Q, is proportional to a mode II stress intensity factor of a sharp punch divided by the square root of the additional contact length due to the roundness of the punch, Q/(,(b,,,a),,b). The fretting fatigue crack initiation can then be investigated by relating the maximum tensile stress with the fatigue endurance stress. The result is analogous to that of Barsom and McNicol where the notched fatigue endurance stress was correlated with the stress intensity factor and the square root of the notch-tip radius. The proposed methodology establishes a ,notch analogue' by making a connection between fretting fatigue at a rounded punch/flat contact and crack initiation at a notch tip and uses fracture mechanics concepts. Conditions of validity of the present model are established both to avoid yielding and to account for the finite thickness of the substrate. The predictions of the model are compared with fretting fatigue experiments on Ti,6Al,4V and shown to be in good agreement. [source]


    Exploratory Analysis of Similarities in Solar Cycle Magnetic Phases with Southern Oscillation Index Fluctuations in Eastern Australia

    Abstract There is growing interest in the role that the Sun's magnetic field has on weather and climatic parameters, particularly the ~11 year sunspot (Schwab) cycle, the ~22 yr magnetic field (Hale) cycle and the ~88 yr (Gleissberg) cycle. These cycles and the derivative harmonics are part of the peculiar periodic behaviour of the solar magnetic field. Using data from 1876 to the present, the exploratory analysis suggests that when the Sun's South Pole is positive in the Hale Cycle, the likelihood of strongly positive and negative Southern Oscillation Index (SOI) values increase after certain phases in the cyclic ~22 yr solar magnetic field. The SOI is also shown to track the pairing of sunspot cycles in ~88 yr periods. This coupling of odd cycles, 23,15, 21,13 and 19,11, produces an apparently close charting in positive and negative SOI fluctuations for each grouping. This Gleissberg effect is also apparent for the southern hemisphere rainfall anomaly. Over the last decade, the SOI and rainfall fluctuations have been tracking similar values to that recorded in Cycle 15 (1914,1924). This discovery has important implications for future drought predictions in Australia and in countries in the northern and southern hemispheres which have been shown to be influenced by the sunspot cycle. Further, it provides a benchmark for long-term SOI behaviour. [source]


    Pseudomigraine With Lymphocytic Pleocytosis: A Calcium Channelopathy?

    Clinical Description of 10 Cases, Genetic Analysis of the Familial Hemiplegic Migraine Gene CACNA1A
    Objective.,To report the clinical findings of 10 patients diagnosed with pseudomigraine with lymphocytic pleocytosis and the results of mutational analysis of the CACNA1A gene in 8 of these patients. Background.,Pseudomigraine with lymphocytic pleocytosis, also referred to as headache with neurologic deficits and cerebrospinal fluid lymphocytosis (HaNDL), is characterized by episodic transient neurologic dysfunction associated with moderate to severe headache and cerebrospinal fluid lymphocytic pleocytosis. Episodes are recurrent and the condition is self-limiting. The etiology of this sporadic condition remains unknown, but the episodic nature and its ability to be triggered by angiography is somewhat reminiscent of the phenotypic features of familial hemiplegic migraine, a condition caused by mutations in the CACNA1A gene. Design/Methods.,Utilizing retrospective chart review, we describe the clinical features of pseudomigraine with lymphocytic pleocytosis in 10 patients. Whole blood was taken from 8 patients (2 were lost to follow-up) and used for DNA testing. The CACNA1A gene was screened for mutations using heteroduplex analysis and direct DNA sequencing. Results.,Clinical features of pseudomigraine with lymphocytic pleocytosis included transient episodes of weakness, sensory and visual symptoms, aphasia, and confusion lasting minutes up to 4 hours. Sensory symptoms, typically affecting the face and arm, were the most common presentation. Localization of symptoms did not conform to vascular territories. Headache was typically throbbing and most often bilateral. Genetic analysis did not identify any mutations in the CACNA1A gene. Conclusions.,Similarities between familial hemiplegic migraine and pseudomigraine with lymphocytic pleocytosis include recurrent headache with reversible neurologic deficit, cerebrospinal fluid lymphocytic pleocytosis, and triggers such as angiography. Even so, heteroduplex analysis and DNA sequencing failed to identify any sporadic mutations or shared polymorphisms in the exons or the intron/exon boundaries of the CACNA1A gene. These results do not support a role of the CACNA1A gene in the etiology of pseudomigraine with lymphocytic pleocytosis. [source]


    ,We didn't know it would get that bad': South Asian experiences of dementia and the service response

    Abstract The aim of the present paper was to examine some views and experiences of dementia among older South Asian people, as well as their families and carers, and to explore central issues of service support. Data were collected in Scotland through interviews with 11 professionals working with South Asian people with dementia, and four case studies of South Asian people with a diagnosis of dementia, as well as their families and carers. The case studies demonstrated overwhelmingly negative experiences of dementia, with poor quality of life, desperate needs for support, lack of access to appropriate services, little knowledge of dementia, and isolation from community and family life. The interviews with professionals described a strong demand for services, a need to develop awareness and knowledge about dementia in South Asian communities, and a need to promote more culturally sensitive, individually responsive services. Similarities between South Asian people and the non-South Asian population include stress on carers, increasing isolation, problematic diagnostic practices, lack of knowledge and demand for service support. Differences include limited use of non-National Health Service (NHS) support, dealing with later stages of dementia at home, particularly negative views about residential care, culturally based attitudinal differences and use of the term ,dementia' in English as neutral rather than stigmatising. The present authors suggest that there is little knowledge and experience of dementia in South Asian communities, as well as restricted access to appropriate services, despite the efforts of voluntary sector and NHS special projects. There is demand for services, especially at home. Services need to develop individual responsiveness for effective working in a diverse society. [source]


    The patient movement as an emancipation movement

    Abstract Objective, To suggest that the patient movement is an emancipation movement. Background, The patient movement is young and fragmented; and it can seem confusing because it lacks an explicit ideology with intellectual and theoretical underpinnings. Methods, Drawing mainly on the experiences and the published writings of patient activists, the author identified eight aspects of the patient movement that could be compared with aspects of recognized emancipation movements: the radicalization of activists; the creation of new knowledge; the identification of guiding principles; the sense of direction; the unmasking of new issues; schisms within the movement and allies outside it; and the gradual social acceptance of some of the ideas (here standards of health care) that activists work to promote. Results, Similarities between certain aspects of the patient movement and of the recognized emancipation movements were close. Conclusion, The patient movement can be regarded as an emancipation movement, albeit an immature one. [source]


    Bilateral medial temporal lobe damage does not affect lexical or grammatical processing: Evidence from amnesic patient H.M.

    Abstract In the most extensive investigation to date of language in global amnesia, we acquired data from experimental measures and examined longitudinal data from standardized tests, to determine whether language function was preserved in the amnesic patient H.M. The experimental measures indicated that H.M. performed normally on tests of lexical memory and grammatical function, relative to age- and education-matched control participants. Longitudinal data from four Wechsler subtests (Information, Comprehension, Similarities, and Vocabulary), that H.M. had taken 20 times between 1953 (preoperatively) and 2000, indicated consistent performance across time, and provided no evidence of a lexical memory decrement. We conclude that medial temporal lobe structures are not critical for retention and use of already acquired lexical information or for grammatical processing. They are, however, required for acquisition of lexical information, as evidenced in previous studies revealing H.M.'s profound impairment at learning new words.     Endodontic teaching in Philippine dental schools

    Abstract Aim The aim of this study was to evaluate the pattern of undergraduate endodontic teaching in Philippine dental schools. Methodology Data were gathered by sending questionnaires to the deans of the 23 dental schools in the country to determine details of the teaching of root canal treatment in permanent teeth. The covering letter requested that endodontic staff complete the questionnaire. Results Twenty of 23 dental schools returned completed questionnaires. Similarities were observed in the timing of undergraduate endodontic teaching, working length determination, and root canal preparation technique. Irrigating fluids recommended included one or a combination of the following: sodium hypochlorite, hydrogen peroxide, distilled water and EDTA. The root canal medicaments popularly employed were CMCP and eugenol. Most schools used slow-setting zinc oxide eugenol cement as sealer. Differences between schools were noted in the laboratory component of the course. The time allotted for the laboratory exercises, as well as the number of teeth used, differed greatly between each school. An inadequate tutor to student ratio was noted in the majority of schools. Teaching aids were limited and, in most instances, produced by faculty members. Most faculty members teaching endodontics had no specialist training. Conclusion The results of this study have demonstrated that there is a need to review endodontic teaching in the majority of the Philippine dental schools to ensure that the course content and curriculum employed by all schools meet specified standards and that appropriate measures should be considered to enhance the learning experience of students.     Capital Asset Pricing Model and the Risk Appetite Index: Theoretical Differences, Empirical Similarities and Implementation Problems,

    We perform a thorough analysis of the Risk Appetite Index (RAI), a measure of changes in risk aversion proposed by Kumar and Persaud (2002). Building on Misina's study (2003), we first argue that the theoretical assumptions granting that the RAI correctly distinguishes between changes in risk and changes in risk aversion are very restrictive. Then, by comparing the RAI with a measure of risk aversion obtained from the Capital Asset Pricing Model (CAPM), we find that the estimates are surprisingly similar. We prove that if the variance of returns is sufficiently smaller than the variance of asset riskiness, then RAI and CAPM provide essentially the same information about risk aversion. We also show, however, that RAI and CAPM suffer from exactly the same implementation problems , the main one being the difficulty in measuring ex-ante returns. At high and medium frequencies, the standard method of measuring ex-ante with ex-post returns may generate negative risk aversion and other inconsistencies. Hence, future research is needed to address this problem. [source]


    Development and testing of a comprehensive chemical mechanism for the oxidation of methane

    A comprehensive chemical mechanism to describe the oxidation of methane has been developed, consisting of 351 irreversible reactions of 37 species. The mechanism also accounts for the oxidation kinetics of hydrogen, carbon monoxide, ethane, and ethene in flames and homogeneous ignition systems in a wide concentration range. It has been tested against a variety of experimental measurements of laminar flame velocities, laminar flame species profiles, and ignition delay times. The highest sensitivity reactions of the mechanism are discussed in detail and compared with the same reactions in the GRI, Chevalier, and Konnov mechanisms. Similarities and differences of the four mechanisms are discussed.     Expectations of the child health nurse in Sweden: two perspectives

    Abstract Background: the child health service exists to support and stimulate parents in order to reduce stress and to encourage an advantageous development of the preschool child. Aim: To explore and describe similarities and differences in expectations of the child health nurse, from the perspective of the recently delivered first-time mother, as compared to an expression of what the child health nurse believed mothers of infants expected of them. The data consisted of 15 interviews with child health nurses and 20 interviews with first-time mothers. Thematic content analysis resulted in seven categories of expectations. The child health nurse was expected to be someone to approach, who could assess the child's development and give immunizations and to be a supporter, counsellor, safety provider and a parent group organizer with knowledge. Similarities between the mothers' and the nurses' statements occurred more frequently than differences, which is suggested to depend on the Swedish tradition among new mothers of visiting the child health clinic. The mothers expected participation in parent groups to a higher degree than the nurses thought they did. But there are also some fundamental differences, not only technically but also institutionally. Since industrialization (around 1800), inland navigation became a major user on both river systems and later flood control started developing more fundamentally. Large intervention works were needed, mainly developed and based on trial and error. In both cases it has led to irreversible effects, which demand continuous attention. For the alluvial stretches in both river basins, a review is given on the most important developments in river engineering over the last two centuries. For both rivers, nautical management and flood control were held in one institutional hand at national/federal level, because both uses/sectors need the creation and regular maintenance of one similar issue: a stable and deep main channel. But the way in which in particular flood management gradually developed institutionally, as an essential part of integrated water management in the alluvial flat lower reaches of both river systems, has diverged. Discussions on financing, priorities, public disclosure, multifunctional aspects, etc. have led in both basins to lengthy procedures and complicated policy making. This paper elaborates on the historic development of fundamentals in river engineering and river management in the alluvial plains of the Rhine Basin and the Lower Mississippi Basin, more in particular focusing on the development of flood protection dikes, and on the stabilization of major channels.     Similarities and Differences Between African Americans' and European Americans' Attitudes, Knowledge, and Willingness to Communicate About Organ Donation,

    While little is known about African Americans' attitudes and knowledge about organ donation, even less is known about how African Americans' attitudes, values, and beliefs affect their behavior and behavioral intentions regarding organ donation; or how African Americans' views are similar to or different from those of European Americans. Adults working 2 sites of a national corporation were randomly selected to complete a survey about organ donation willingness, intention to sign an organ donor card, knowledge and attitudes toward organ donation, and level of altruism. Results indicate that African Americans differ significantly from Whites on several individual attitude and knowledge items. However, the basic relationship between knowledge, attitudes, values, and behaviors regarding organ donation between the 2 groups appears the same. Furthermore, these results indicate that future organ donation promotion campaigns must focus on increasing basic knowledge and countering myths about organ donation for both populations. [source]


    Biogeography of Plagiochila (Hepaticae): natural species groups span several floristic kingdoms

    Abstract Aim This paper presents a synthesis of our recent results regarding the biogeography of Plagiochila using a molecular approach, and documents intercontinental ranges within this largest genus of the hepatics. Methods A maximum likelihood analysis of sixty-one nrITS sequences of Plagiochila was performed and the molecular topology obtained was compared with morphological, phytochemical and geographical data. Results Our molecular data set allowed the identification of eleven Plagiochila sections, the majority of which cover at least two floristic kingdoms. Seven sections have species in Europe (sect. Arrectae, Carringtoniae, Fuscoluteae, Glaucescentes, Plagiochila, Rutilantes, Vagae). Plagiochila species from Atlantic Europe are usually close to or conspecific with neotropical taxa, whereas species widespread in Europe are closely related to Asian ones and not to those in the Neotropics. Plagiochila sect. Arrectae represents a neotropical , Atlantic European clade. The section is not closely related , as has often been suggested , to the morphologically similar sect. Zonatae from Asia and western North America. Sequence data show that the African P. integerrima and the neotropical P. subplana are members of the Asian sect. Cucullatae (sect. Ciliatae, syn. nov.), which becomes pantropical in distribution. An ITS sequence of P. boryana from Uganda confirms the Afro-American range of the primarily neotropical sect. Hylacoetes. Similarities in sporophyte morphology between the sect. Cucullatae and sect. Hylacoetes are the result of parallel evolution. Main conclusions Our results indicate that intercontinental ranges at section and species level are common in Plagiochila. Carl's (1931) subdivision of Plagiochila into sections restricted to one floristic kingdom is outdated. Biogeographical patterns in Plagiochila are not dissimilar to those of other groups of bryophytes but elucidation of the geographical ranges of the taxa requires a molecular approach. Contrary to earlier belief, most Plagiochila species from Atlantic Europe do not have close relatives in Asia but are conspecific with or closely related to species from tropical America. [source]


    Comparative Pharmacology of Guinea Pig Cardiac Myocyte and Cloned hERG (IKr) Channel

    Introduction: This study used whole-cell, patch clamp techniques on isolated guinea pig ventricular myocytes and HEK293 cells expressing cloned human ether-a-go-go-related gene (hERG) to examine the action of drugs causing QT interval prolongation and torsades de pointes (TdP) in man. Similarities and important differences in drug actions on cardiac myocytes and cloned hERG IKr channels were established. Qualitative actions of the drugs on cardiac myocytes corresponded with results obtained from Purkinje fibers and measurement of QT interval prolongation in animal and human telemetry studies. Methods and Results: Adult guinea pig ventricular myocytes were isolated by enzymatic digestion. Cells were continuously perfused with Tyrode's solution at 33,35°C. Recordings were made using the whole-cell, patch clamp technique. Action potentials (APs) were elicited under current clamp. Voltage clamp was used to study the effect of drugs on IKr (rapidly activating delayed rectifier potassium current), INa (sodium current), and ICa (L-type calcium current). Dofetilide increased the myocyte action potential duration (APD) in a concentration-dependent manner, with a pIC50 of 7.3. Dofetilide 1 ,M elicited early afterdepolarizations (EADs) but had little affect on ICa or INa. E-4031 increased APD in a concentration-dependent manner, with a pIC50 of 7.2. In contrast, 10 ,M loratadine, desloratadine, and cetirizine had little effect on APD or IKr. Interestingly, cisapride displayed a biphasic effect on myocyte APD and inhibited ICa at 1 ,M. Even at this high concentration, cisapride did not elicit EADs. A number of AstraZeneca compounds were tested on cardiac myocytes, revealing a mixture of drug actions that were not observed in hERG currents in HEK293 cells. One compound, particularly AR-C0X, was a potent blocker of myocyte AP (pIC50 of 8.4). AR-C0X also elicited EADs in cardiac myocytes. The potencies of the same set of drugs on the cloned hERG channel also were assessed. The pIC50 values for dofetilide, E-4031, terfenadine, loratadine, desloratadine, and cetirizine were 6.8, 7.1, 7.3, 5.1, 5.2, and <4, respectively. Elevation of temperature from 22 to 35°C significantly enhanced the current kinetics and amplitudes of hERG currents and resulted in approximately fivefold increase in E-4031 potency. Conclusion: Our study demonstrates the advantages of cardiac myocytes over heterologously expressed hERG channels in predicting QT interval prolongation and TdP in man. The potencies of some drugs in cardiac myocytes were similar to hERG, but only myocytes were able to detect important changes in APD characteristics and display EADs predictive of arrhythmia development. We observed similar qualitative drug profiles in cardiac myocytes, dog Purkinje fibers, and animal and human telemetry studies. Therefore, isolated native cardiac myocytes are a better predictor of drug-induced QT prolongation and TdP than heterologously expressed hERG channels. Isolated cardiac myocytes, when used with high-throughput patch clamp instruments, may have an important role in screening potential cardiotoxic compounds in the early phase of drug discovery. This would significantly reduce the attrition rate of drugs entering preclinical and/or clinical development. The current kinetics and amplitudes of the cloned hERG channel were profoundly affected by temperature, significantly altering the potency of one drug (E-4031). This finding cautions against routine drug testing at room temperature compared to physiologic temperature when using the cloned hERG channel. [source]


    Cultural Similarities and Differences in the Design of University Web sites

    This study examines cultural differences and similarities in design of university Web sites using Hofstede's model of cultural dimensions. Graphical elements on a sample of university home pages from Malaysia, Austria, the United States, Ecuador, Japan, Sweden, Greece and Denmark are compared using content analysis methods. The home pages were analyzed on the basis of two criteria: organization and graphical design. Element frequency scores were correlated with Hofstede's indexes and interpreted on the basis of the existing literature. The results suggest that similarities and differences in Web site design can be brought out through Hofstede's cultural model. Computed correlations between Hofstede's scores and frequency counts of interface elements were weaker than anticipated, but in most cases occurred in the hypothesized direction. [source]

    Rather than addressing ethnicity through a pre,set cultural lens, I discuss how my experiences as a family therapy trainer in Singapore, Hong Kong, Taiwan and Shanghai taught me to understand culture through the lens of the family. Similarities and differences among these cultural domains were reviewed. I also demonstrate how each encounter shaped my emotional responses and ways of intervention in the course of family interviews. Even though I belong to the same ethnic background, I had to interact differently in different arenas, despite my cultural values and theoretical orientation. [source]


    Interspecific variations of otolith chemistry in estuarine fish nurseries

    Otolith chemical composition differed between juveniles of five fish species (Solea solea, Solea senegalensis, Platichthys flesus, Diplodus vulgaris and Dicentrarchus labrax) in nursery areas of six estuaries along the Portuguese coast (Douro, Ria de Aveiro, Mondego, Tejo, Sado and Mira). Spatially consistent patterns in the concentration of some elements were responsible for differences between species. Discrimination of estuaries using data from all species simultaneously averaged 44·7% of correctly classified cases, whilst discrimination of species averaged 76·2%. Moreover, species-related patterns in otolith fingerprints were highlighted when comparing species for each estuarine nursery area, with intra estuarine species discrimination averages ranging from 86·2 to 100·0%. Similarities in the otolith elemental fingerprints were larger between species with close phylogeny and ecology, particularly between flatfish and perciform species. In addition to the differences in physiological regulation of species, specific microhabitat use in a common environment was suggested as a relevant factor for the differentiation of otolith chemistry among species occurring in the same locations. Despite positive results in specific estuaries, variation in otolith composition limited the use of species as proxies to classify others to their system of origin. [source]


    Temperature and prey quality effects on growth of juvenile walleye pollock Theragra chalcogramma (Pallas): a spatially explicit bioenergetics approach

    A bioenergetics model for juvenile age-0 year walleye pollock Theragra chalcogramma was applied to a spatially distinct grid of samples in the western Gulf of Alaska to investigate the influence of temperature and prey quality on size-specific growth. Daily growth estimates for 50, 70 and 90 mm standard length (LS) walleye pollock during September 2000 were generated using the bioenergetics model with a fixed ration size. Similarities in independent estimates of prey consumption generated from the bioenergetics model and a gastric evacuation model corroborated the performance of the bioenergetics model, concordance correlation (rc) = 0·945, lower 95% CL (transformed) (L1) = 0·834, upper 95% CL (transformed) (L2) = 0·982, P < 0·001. A mean squared error analysis (MSE) was also used to partition the sources of error between both model estimates of consumption into a mean component (MC), slope component (SC), and random component (RC). Differences between estimates of daily consumption were largely due to differences in the means of estimates (MC= 0·45) and random sources (RC= 0·49) of error, and not differences in slopes (SC= 0·06). Similarly, daily growth estimates of 0·031,0·167 g day,1 generated from the bioenergetics model was within the range of growth estimates of 0·026,0·190 g day,1 obtained from otolith analysis of juvenile walleye pollock. Temperature and prey quality alone accounted for 66% of the observed variation between bioenergetics and otolith growth estimates across all sizes of juvenile walleye pollock. These results suggest that the bioenergetics model for juvenile walleye pollock is a useful tool for evaluating the influence of spatially variable habitat conditions on the growth potential of juvenile walleye pollock.     Planned and Unplanned Complex Suicides: A 5-year Retrospective Study

    ABSTRACT: The notion of planned and unplanned complex suicides first appeared in 1974 by Marcinkowski and, since then, no systematic study of complex suicides has been published in the English forensic literature. Here, the authors present a 5-year retrospective study of complex suicides. Nineteen complex suicides were reviewed: five unplanned and 14 planned, including the first case of an unplanned complex suicide in a woman. All cases were analyzed in terms of gender, age, methods of suicide, the presence of a suicide note, and past suicide attempts, and statistically compared with a 50-case sample of simple suicides. A further comparison was established with compiled data from the literature. Similarities were revealed regarding incidence of complex suicides, male gender predominance, and types of methods used. In contrast, results showed a higher average age for planned complex suicide victims. Finally, the authors discuss the application of the complex suicide definition. [source]

The flexibility approach based on the active set strategy is used to study the same examples from the flexibility point of view.     Disentangling Value Similarities and Transmissions in Established Marriages: A Cross-Lagged Longitudinal Study

    This study examined spousal value similarity and transmission across a 5-year period on four value orientations: traditional family values, self-determination, social criticism, and hedonism. Participants were 685 Dutch couples in established marriages. Structural equation modeling results indicated that spouses were moderately similar on all value orientations. Over time, spousal similarity remained for traditional family values, self-determination, and social criticism and decreased for hedonism. Direct spousal transmission occurred on social criticism and hedonism with wives influencing their husbands. Multiple group analyses revealed that wives' value transmission to husbands occurred only within couples with similar social positions (in education and religion) and with higher degrees of marital satisfaction. Findings confirm that experiences in one's family of destination contribute to midlife value development. [source]
