    Is Helicobacter pylori a True Microaerophile?

    Abstract Background:, There is no general consensus about the specific oxygen and carbon dioxide requirements of the human pathogen Helicobacter pylori. This bacterium is considered a microaerophile and consequently, it is grown under atmospheres at oxygen tensions 5,19% and carbon dioxide tensions 5,10%, both for clinical and basic and applied research purposes. The current study compared the growth of H. pylori in vitro, under various gas atmospheres, and determined some specific changes in the physiology of bacteria grown under different oxygen partial pressures. Methods:, Measurements of bacterial growth under various conditions were carried out employing classical solid and liquid culture techniques. Enzymatic activities were measured using spectrophotometric assays. Results:,H. pylori and all the other Helicobacter spp. tested had an absolute requirement for elevated carbon dioxide concentrations in the growth atmosphere. In contrast with other Helicobacter spp., H. pylori can tolerate elevated oxygen tensions when grown at high bacterial concentrations. Under 5% CO2, the bacterium showed similar growth in liquid cultures under oxygen tensions from microaerobic (< 5%) to fully aerobic (21%) at cell densities higher than 5 × 105 cfu/ml for media supplemented with horse serum and 5 × 107 cfu/ml for media supplemented with ,-cyclodextrin. Evidence that changes occurred in the physiology of H. pylori was obtained by comparing the activities of ferredoxin:NADH (nicotinamide adenine dinucleotide) oxidoreductases of bacteria grown under microaerobic and aerobic atmospheres. Conclusions:,H. pylori is a capnophile able to grow equally well in vitro under microaerobic or aerobic conditions at high bacterial concentrations, and behaved like oxygen-sensitive microaerophiles at low cell densities. Some characteristics of H. pylori cells grown in vitro under microaerobic conditions appeared to mimic better the physiology of organisms grown in their natural niche in the human stomach. [source]


    Demonstration of salmon farming as a net producer of fish protein and oil

    Abstract To date aquaculture's reliance on dietary marine sources has been calculated on a fish weight-to-weight basis without considering the absolute amounts of nutrients but this approach neglects the often considerable differences in the nutritional value of fish. We propose simple nutrient-to-nutrient-based dependency measures that take into account these nutritional differences. In the first study reported here, individually tagged Atlantic salmon (Salmo salar) were reared in seawater supplied tanks with feed collection facilities. In the second, commercial net pens were used to grow over 200 000 fish. For both studies, a low marine ingredient feed containing approximately 165 g kg,1 fishmeal was compared to a control feed (approx 300 g kg,1 fishmeal) whilst fish oil inclusion was less markedly reduced. The low marine feeds supported similar growth and feed efficiency compared to the control feeds. With the low marine ingredient feeds, the weight of salmon protein and lipid produced through growth exceeded the weight of marine protein and lipid consumed by the fish meaning that salmon farming can be a net producer of fish protein and oil. The amount of n-3 long-chain polyunsaturated fatty acids deposited was sufficient to meet current recommendations from human health organizations. [source]


    Influence of dietary palm oil on growth, tissue fatty acid compositions, and fatty acid metabolism in liver and intestine in rainbow trout (Oncorhynchus mykiss)

    Abstract This study aimed to investigate the effects of dietary crude palm oil (CPO) on fatty acid metabolism in liver and intestine of rainbow trout. Triplicate groups of rainbow trout for 10 weeks at 13 °C were fed on diets in which CPO replaced fish oil (FO) in a graded manner (0,100%). At the end of the trial, fatty acid compositions of flesh, liver and pyloric caeca were determined and highly unsaturated fatty acid (HUFA) synthesis and fatty acid oxidation were estimated in isolated hepatocytes and caecal enterocytes using [1- 14C]18:3n-3 as substrate. Growth performance and feed efficiency were unaffected by dietary CPO. Fatty acid compositions of selected tissues reflected the dietary fatty acid composition with increasing CPO resulting in increased proportions of 18:1n-9 and 18:2n-6 and decreased proportions of n-3HUFA, 20:5n-3 and 22:6n-3. Palmitic acid, 16:0, was also increased in flesh and pyloric caeca, but not in liver. The capacity of HUFA synthesis from 18:3n-3 increased by up to threefold in both hepatocytes and enterocytes in response to graded increases in dietary CPO. In contrast, oxidation of 18:3n-3 was unaffected by dietary CPO in hepatocytes and reduced by high levels of dietary CPO in enterocytes. The results of this study suggest that CPO can be used at least to partially replace FO in diets for rainbow trout in terms of permitting similar growth and feed conversion, and having no major detrimental effects on lipid and fatty acid metabolism, although flesh fatty acid compositions are significantly affected at an inclusion level above 50%, with n-3HUFA reduced by up to 40%.     Partial substitution of dietary fish meal with soybean meal for speckled shrimp, Metapenaeus monoceros (Fabricius, 1798) (Decapoda: Penaeidae) juvenile

    Abstract A 63-day feeding study was conducted to evaluate the effect of partial substitution (0%, 20%, 40% and 60%) of dietary fish meal (FM) with soybean meal (SBM) on the growth performance and feed utilization of speckled shrimp, Metapenaeus monoceros juveniles. A total of 180 M. monoceros juveniles with an average weight of 0.67 ± 0.18 g were equally distributed over 12 glass aquaria (30 L each, triplicate per treatment). All aquaria were filled with filtered sea water (30,35 ,, salinity). Four isonitrogenous (400 g kg,1 crude protein) and isocaloric (20.5 MJ GE kg,1) test diets were formulated. The control diet contained FM (D1) as the main protein source. In the other diets, FM was partially replaced with 20 (D2), 40% (D3) or 60% (D4) SBM. The results indicate that the diet in which 40% of the FM is replaced by SBM yields similar growth and production, and is as cost effective, as the control diet. [source]


    Carbohydrate utilization by juvenile silver perch, Bidyanus bidyanus (Mitchell).

    Abstract The ability of juvenile silver perch (Bidyanus bidyanus) to utilize dietary raw wheat meal, raw wheat starch, gelatinized wheat starch and dextrin as energy sources to spare protein for growth was quantified. Energy utilization and protein sparing were assessed by comparing the weight gain, energy retention efficiency, protein retention and body composition of silver perch that had been fed a series of diets in which the basal diet (low carbohydrate) was systematically replaced with graded levels of each carbohydrate ingredient or an inert diluent, diatomaceous earth. The protein content decreased as the carbohydrate content increased, giving four different protein to energy ratios for each of the four carbohydrate sources (except for the 60% inclusion level, at which only three carbohydrate sources were tested). Silver perch were efficient at utilizing carbohydrate for energy to spare protein. Silver perch fed diets containing up to 30% wheat meal, raw wheat starch, gelatinized wheat starch or dextrin exhibited similar growth, protein retention and energy retention efficiency to the fish fed the basal diet. Weight gain of silver perch fed diets containing wheat meal or carbohydrates at 45% inclusion content had significantly reduced weight gain when compared with fish fed the basal diet. However, protein retention and energy retention efficiency were similar or better. Whole-body protein levels of silver perch remained constant regardless of carbohydrate sources, and there was no evidence of increasing whole-body lipid concentrations for fish fed diets with up to 60% dietary carbohydrate. Silver perch were more efficient at utilizing processed starch (either gelatinized starch or dextrin) than wheat meal or raw wheat starch. [source]
