Lead-induced alterations of apoptosis and neurotrophic factor mRNA in the developing rat cortex, hippocampus, and cerebellum

Abstract Previous reports have recently shown the prototypic neurotoxicant, lead, to induce apoptosis in the brains of developing organisms. In the current study, timed-pregnant rats were exposed to lead acetate (0.2% in the drinking water) 24 h following birth at postnatal day 1 (PND 1). Dams and pups were continuously exposed to lead through the drinking water of the dam until PND 20. Postnatal exposure in the pups resulted in altered mRNA levels of the following apoptotic and neurotrophic factors: caspase 2 and 3, bax, bcl-x, brain-derived neurotrophic factor (BDNF). Ribonuclease protection assays were conducted to measure the factors simultaneously at the following postnatal time points: 9, 12, 15, 20, 25, days. Our results suggest a brain region- and time-specific response following lead acetate exposure. The region most vulnerable to alterations occurs in the hippocampus with alterations beginning at PND 12, in which caspase 3, bcl-x, BDNF increase with lead exposure. Significant treatment effects were not observed for both the cortex and cerebellum. © 2007 Wiley Periodicals, Inc. Comparison of clozapine and haloperidol on some autonomic and psychomotor functions, and on serum prolactin concentration, in healthy subjects

Aims To compare the autonomic, neuroendocrine and psychomotor effects of single doses of the ,atypical' antipsychotic clozapine and the ,classical' antipsychotic haloperidol, in healthy male volunteers. Methods Clozapine (50 mg), haloperidol (3 mg) and placebo were administered to 12 healthy male volunteers at weekly intervals, according to a balanced double-blind design. Resting pupil diameter, salivary output, heart rate, blood pressure, plasma prolactin concentration, critical flicker fusion frequency and subjective ,alertness', ,contentedness' and ,anxiety' were measured at baseline and 2, 3, 4 and 5 h after drug ingestion. Data were analysed by analysis of variance with individual comparisons (Dunnett's test) with a significance criterion of P < 0.05. Results Significant treatment effects (difference from placebo [mean, 95% CI] 5 h after drug ingestion) were as follows: clozapine reduced pupil diameter (mm; ,3.02 [,3.56, ,2.47]), salivary output (g; ,0.34 [,0.60, ,0.08]), mean arterial blood pressure (mm Hg; ,8.7 [,14.3, ,3.1]), critical flicker fusion frequency (Hz; ,3.26 [,3.94, ,2.58]), and subjectively-rated ,alertness' (mm; ,20.94 [,29.21, ,12.67]) and ,contentedness' (mm; ,12.98 [,17.90, ,8.06]), whereas haloperidol increased prolactin concentration (mU l,1; 301.3 [196.7, 405.8]) and caused small reductions in pupil diameter (mm; ,0.68 [,1.23, ,0.14]), mean arterial blood pressure (mm Hg; ,7.0 [,12.6, ,1.4]) and critical flicker fusion frequency (Hz; ,1.15 [,1.83, ,0.47]). Conclusions The effects of the antipsychotics are in agreement with their receptor binding profiles: ,1 -adrenoceptor blockade by clozapine may contribute to reductions in pupil diameter, salivation, mean arterial blood pressure and sedation, and muscarinic cholinoceptor blockade by the drug may underlie the reduction in salivation. Conversely, D2 dopamine receptor blockade by haloperidol is likely to be responsible for the increase in prolactin secretion evoked by the drug. [source]


Naltrexone versus acamprosate in the treatment of alcohol dependence: a multi-centre, randomized, double-blind, placebo-controlled trial

ABSTRACT Aim To compare the efficacy of acamprosate and naltrexone in the treatment of alcohol dependence., Design A double-blind, placebo-controlled trial., Setting Three treatment centres in Australia., Participants A total of 169 alcohol dependent subjects were given naltrexone (50 mg/day), acamprosate (1998 mg/day) or placebo for 12 weeks. Intervention All subjects were offered manualized compliance therapy, a brief intervention that targets problems that may affect treatment compliance such as ambivalence and misperceptions about medication. Measurements Time to the first drink, time to first relapse, drinks per drinking day and cumulative abstinence. Findings In intention-to-treat analyses, there were no differences between groups on outcome measures of drinking, craving or biochemical markers. Similarly, analyses of the 94 subjects that completed the study in full and demonstrated 80% compliance, revealed no significant treatment effects. Differential treatment effects were identified after stratification according to scores on the Alcohol Dependence Scale (ADS) and Depression Anxiety and Stress Scale (DASS). A significant beneficial treatment effect on time to first relapse was revealed for subjects with ,no depression' allocated to naltrexone (n = 56; P < 0.01). In addition, a significant beneficial treatment effect was revealed in subjects with ,low dependence' allocated to naltrexone (n = 34; P < 0.05). Conclusions The results of this study support the efficacy of naltrexone in the relapse prevention of alcoholism amongst those with low levels of clinical depression and alcohol dependence severity. No effect of acamprosate was found in our sample. [source]


Effects of increased temperature and nutrient enrichment on the stoichiometry of primary producers and consumers in temperate shallow lakes

Summary 1. We studied the effects of increased water temperatures (0,4.5 °C) and nutrient enrichment on the stoichiometric composition of different primary producers (macrophytes, epiphytes, seston and sediment biofilm) and invertebrate consumers in 24 mesocosm ecosystems created to mimic shallow pond environments. The nutrient ratios of primary producers were used as indicative of relative nitrogen (N) or phosphorus (P) limitation. We further used carbon stable isotopic composition (,13C) of the different primary producers to elucidate differences in the degree of CO2 limitation. 2. Epiphytes were the only primary producer with significantly higher ,13C in the enriched mesocosms. No temperature effects were observed in ,13C composition of any primary producer. Independently of the treatment effects, the four primary producers had different ,13C signatures indicative of differences in CO2 limitation. Seston had signatures indicating negligible or low CO2 limitation, followed by epiphytes and sediment biofilm, with moderate CO2 limitation, while macrophytes showed the strongest CO2 limitation. CO2 together with biomass of epiphytes were the key variables explaining between 50 and 70% of the variability in ,13C of the different primary producers, suggesting that epiphytes play an important role in carbon flow of temperate shallow lakes. 3. The ratio of carbon to chlorophyll a decreased with increasing temperature and enrichment in both epiphytes and seston. The effects of temperature were mainly attributed to changes in algal Chl a content, while the decrease with enrichment was probably a result of a higher proportion of algae in the seston and epiphytes. 4. Macrophytes, epiphytes and seston decreased their C : N with enrichment, probably as an adaptation to the different N availability levels. The C : N of epiphytes and Elodea canadensis decreased with increasing temperature in the control mesocosms. Sediment biofilm was the only primary producer with lower C : P and N : P with enrichment, probably as a result of higher P accumulation in the sediment. 5. Independently of nutrient level and increased temperature effects the four primary producers had significantly different stoichiometric compositions. Macrophytes had higher C : N and C : P and, together with epiphytes, also the highest N : P. Seston had no N or P limitation, while macrophytes and epiphytes may have been P limited in a few mesocosms. Sediment biofilm indicated strong N deficiency. 6. Consumers had strongly homeostatic stoichiometric compositions in comparison to primary producers, with weak or no significant treatment effects in any of the groups (insects, leeches, molluscs and crustaceans). Among consumers, predators had significantly higher N content and lower C : N than grazers. [source]


Effects of bird predation on arthropod abundance and tree growth across an elevational gradient

Considerable uncertainty surrounds the conditions under which birds can cause trophic cascades. In a three-year experiment, we studied the direct and indirect effects of insectivorous birds on arthropod abundance, herbivory, and growth of striped maple Acer pensylvanicum saplings in a northern hardwood forest of central New Hampshire, USA. We manipulated bird predation by erecting exclosures around saplings and directly manipulated herbivory by removing herbivores. We also examined how climate modifies these interactions by replicating the experiment at three locations along an elevational gradient. Effects of bird predation were variable. Overall, mean arthropod biomass was 20% greater on saplings within bird exclosures than on controls (p<0.05). The mean biomass of leaf-chewing herbivores, primarily Lepidoptera larvae, was 25% greater within exclosures but not statistically different from controls. To a lesser degree, mean herbivore damage to foliage within exclosures exceeded that of controls but differences were not significant. We also did not detect significant treatment effects on sapling shoot growth. The high understory vegetation density relative to bird abundance, and low rate of herbivory during the study (mean 5% leaf area removed, controls), may have limited the ability of birds to affect sapling growth. Climate effects operated at multiple scales, resulting in a complex interplay of interactions within the food web. Regional synchrony of climatic conditions resulted in annual fluctuations in herbivore abundance and tree growth that were shared across elevations. At the same time, local environmental variation resulted in site differences in the plant, herbivore, and bird communities. These patterns resulted in a mosaic of top,down strengths across time and space, suggesting an overall pattern of limited effects of birds on plant growth, possibly interspersed with hotspots of trophic cascades. Effects of moisture, nitrogen, grass competition and simulated browsing on the survival and growth of Acacia karroo seedlings

Abstract The effects of irrigation, nitrogen fertilization, grass competition and clipping were investigated for one growing season at the research farm of the University of Fort Hare in the Eastern Cape Province of South Africa. The aim of the experiment was to assess the short-term performance of Acacia karroo seedlings under different environmental conditions and the implications of such factors on the long-term recruitment of plant species in savanna rangelands. There were no significant treatment effects on the survival of A. karroo seedlings. Using stem length and basal diameter as growth parameters, it was observed that irrigation enhanced both variables, while nitrogen fertilization did not have any significant effects. Clipping, grass competition and their interaction greatly suppressed the growth of the seedlings. Clipping increased the mean stem length when they were irrigated and fertilized. Control and fertilized plants had the highest stem length in the absence of grass competition, while grass competition combined with clipping resulted in the lowest stem length in both irrigated and nonirrigated plants. It was concluded that in the presence of grass competition, controlled browsing could be a viable solution to the problem of bush encroachment in savanna rangelands. Résumé Les effets de l'irrigation, d'une fertilisation à l'azote, de la compétition avec l'herbe et de la taille ont étéétudiés pendant une saison de croissance dans la ferme expérimentale de l'Université de Fort Hare, dans la Province du Cap oriental, en Afrique du Sud. Le but de l'expérience était d'évaluer les performances à court terme de jeunes plants d'Acacia karroo dans différentes conditions expérimentales, et les implications de ces facteurs sur le recrutement à long terme d'espèces végétales dans des pâturages de savane. Il n'y a pas eu d'effet significatif des traitements sur la survie des plants d'A. karroo. En utilisant la longueur des troncs et le diamètre de la base comme indicateurs de croissance, on a observé que l'irrigation augmentait les deux variables, alors que la fertilisation à l'azote n'avait aucun effet significatif. La taille, la compétition avec l'herbe et leur interaction supprimait fortement la croissance des jeunes plants. La taille augmentait la longueur moyenne des troncs lorsqu'ils étaient irrigués et fertilisés. Les plants témoins et les plants fertilisés avaient les troncs les plus longs en absence de toute compétition avec des herbes, alors que cette dernière, combinée avec la taille, aboutissait à la longueur la plus petite des troncs, chez les plants irrigués ou non. On en a conclu qu'en cas de compétition avec des herbes, un pâturage contrôlé pourrait être une solution viable au problème de l'envahissement des broussailles dans les pâturages de savane. Effects of bisacodyl on ascending colon emptying and overall colonic transit in healthy volunteers

Summary Background, The mechanism of action of bisacodyl in the unprepared human colon remains unclear. Aim, To evaluate the effect of oral bisacodyl on the overall and regional colonic transit in humans. Methods, In a double-blind, randomized, placebo-controlled study of 25 healthy participants, effects of oral bisacodyl (5 mg p.o. per day) and placebo on colonic transit were compared. A validated scintigraphy method was used to measure colonic transit. The primary transit endpoints, ascending colon emptying t1/2 and geometric centre of colon isotope at 24 h (overall transit), were compared (Wilcoxon rank sum test). Results, There were significant treatment effects on ascending colon t1/2, with the bisacodyl group demonstrating accelerated emptying [median 6.5 h, interquartile range 5.0,8.0 h] relative to the placebo group [11.0 h (7.0,17.1); P = 0.03]. Numerical differences in colonic geometric centre 24 h [bisacodyl median 3.0 (2.2,3.8), placebo 4.0 (3.1,4.6)] were not significant (P = 0.19). There were no significant differences observed in geometric centre 4 h. Conclusion, Oral 5 mg bisacodyl accelerates ascending colon in the unprepared colon in healthy adults; this action may contribute to the drug's efficacy in constipation. [source]


Competitive Interactions among First-Year and Second-Year Plants of the Invasive, Biennial Garlic Mustard (Alliaria petiolata) and Native Ground Layer Vegetation

We studied the effects of hand weeding of second-year plants of the biennial garlic mustard (Alliaria petiolata) on first-year plants (seedlings) and native ground layer vegetation. Garlic mustard is a Eurasian species that has invaded deciduous forest ground layers in eastern North America. Treatments consisted of a control and an early or late weeding of second-year garlic mustard. The early treatment (early March) was applied before garlic mustard seeds had germinated and when most native species were dormant. The late treatment (mid-May) occurred after plants had bolted, flowering was occurring, and most native species and new garlic mustard seedlings were actively growing. Pre-treatment data were obtained in 2004 and treated and control plots were sampled in 2005, 2006, and 2007. No significant treatment effects were observed in 2004 or 2005. In 2006, mean cover of first-year plants was higher in the early weeding treatment than in the late weeding treatment and control. In 2007, mean cover of first-year garlic mustard was higher in the control than in either of the two weeding treatments. There were no significant treatment effects in any year on native vegetation cover, bare ground, or the five most abundant native species. Our data indicate that (1) late weeding of garlic mustard provided more effective control than early weeding because late weeding allows second-year plants to compete with garlic mustard seedlings for a longer period of time and (2) competition between first- and second-year plants is responsible for alternating dominance of first-year and second-year garlic mustard plants. Feed intake, growth and nutrient utilization in Atlantic halibut (Hippoglossus hippoglossus) fed diets containing a bacterial protein meal

Abstract Triplicate groups of Atlantic halibut were fed diets containing 0%, 9% or 18% of a bacterial protein meal (BPM) produced from natural gas in a 9-week trial. Growth rates, relative feed intake, feed efficiency ratio and retention of all indispensable amino acids were significantly lower in fish fed the 18% BPM diets than in those fed the 0% and 9% BPM diets. There were no significant treatment effects on urea levels in plasma, liver or muscle, or in uric acid levels in plasma. The hepatosomatic index was lowest in fish fed the 18% BPM diet. Although the concentration of copper, an element abundant in BPM, increased in the liver as dietary BPM level increased, the total copper content in liver decreased. Fish fed the 0% and 9% BPM diets had a higher degree of supranuclear vacuolization of pyloric caeca and mid-intestine epithelia compared with fish fed the 18% BPM diet. In conclusion, the halibut fed the 9% BPM diet performed equally well as the control group regarding growth, feed intake and feed efficiency ratio, whereas performance was reduced in the fish fed the 18% BPM diet. Species-specific Seedling Responses to Hurricane Disturbance in a Puerto Rican Rain Forest1

ABSTRACT Seedling dynamics were followed in a Puerto Rican forest for 20 months following a severe hurricane to study the interactive effects of hurricane debris, nutrients, and light on seedling diversity, density, growth, and mortality. Three treatments (debris removal, an unaltered control with hurricane debris, and chemical fertilization added to hurricane debris) altered levels of forest debris and soil nutrients. Canopy openness was measured twice using hemispherical photographs of the canopy. We examined the demographic responses of six common species to treatments over time. Seedling densities increased for all six species but the only significant treatment effects were increased densities of the pioneer tree Cecropia and the shrub Palicourea in the debris removal treatment. Seedling growth declined with declining light levels for four species but not for the pioneer tree Alchornea or the non-pioneer tree Dacryodes. Only Cecropia and the non-pioneer tree Chionanthus had treatment effects on growth. Mortality also differed among species and tended to be highest in the fertilized plots for all but Cecropia and Dacryodes. We found only some of the expected differences between pioneer and non-pioneer plants, as each species had a unique response to the patchy distributions of organic debris, nutrients, and light following the hurricane. High local species diversity was maintained through the individualistic responses of seedlings after a disturbance. RESUMEN Seguimos la dinámica de plántulas en un bosque en Puerto Rico durante 20 meses después del huracán Hugo para estudiar el efecto de la interacción de hojarasca de huracán, nutrientes, y luz sobre la diversidad de especies, la densidad, el crecimiento, y la mortalidad. Establecimos tres tratamientos (remoción de hojarasca, control con la hojarasca de huracán inalterada, y fertilizante químico añadido a la hojarasca del huracán) para alterar los niveles de hojarasca en el bosque y los nutrientes en el suelo. Medimos luz directa e indirecta dos veces usando fotografias hemisféricas del dosel. La diversidad y la uniformidad en la distribución de especies pero no la riqueza de especies fueron reducidas en presencia de fertilización. Durante el estudio examinamos respuestas demográficas de seis especies communes a los tratamientos. La densidad de plántulas aumentó para todas las seis especies pero el único efecto de tratamiento fue el aumento en la densidad del árbol pionero Cecropia y el arbusto Palicourea en el tratamiento de remoción de hojarasca. El crecimiento de plántulas disminuyó según los niveles de luz disminuyeron para cuatro de las especies pero no para el árbol pionero Alchornea o el árbol nopionero Dacryodes. Sólo el crecimiento de Cecropia y del árbol no-pionero Chionanthus fue affectado por los tratamientos. La mortalidad fue diferente entre las especies y tuvo una tendencia a ser mayor en las parcelas fertilizadas. Sólo encontramos algunas de las diferencias esperadas entre plantas pioneras y no-pioneras. Cada especie respondió de forma única a la distribución en parches de hojarasca, nutrientes, y luz luego del huracán. La aha diversidad local de especies se mantuvo a través de las respuestas individualizadas de las plántulas después de la perturbación. [source]


Multivitamin-mineral supplementation and age-related cataract.

The Italian-American controlled clinical trial
Purpose To evaluate if a multivitamin-mineral supplement at RDI dosage can prevent incidence and progression of age-related cataract. Methods Randomized, double blind, single centre clinical trial. 1020 participants of Italian descent living in Parma, 55 to 75 years old, were assigned to placebo or to a daily tablet of Centrum. Lens status was assessed at baseline and at yearly follow-up visits by lens photography.Primary outcome was a prespecified increase from baseline in N, C, or PSC opacity grades or cataract surgery. Secondary outcomes were increase in type-specific opacity grades, cataract surgery, and visual acuity loss > 15 letters. Results After an average follow-up of 9 +- 2.4 years there was a decrease in total lens events in participants assigned to Centrum (HR 0.82, 95% CI 0.68-0.98, P = 0.03). Nuclear events were significantly less common (HR 0.66, 95% CI 0.50-0.88, P = 0.004) and PSC events significantly more common (HR 2.00, 95% CI 1.35-2.98, P =< 0.001) in participants taking Centrum. No statistically significant treatment effects were seen for cortical opacities, moderate visual acuity loss, or cataract surgery. Conclusion Lens events were less common in participants who took Centrum, but treatment had opposite effects on the development or progression of N and PSC opacities. This and the absence of effect on a functional outcome does not allow us to recommend the use of RDI nutritional supplement to affect the risk of cataract development (Ophthalmology 115;599,2008).
