CLÁUDIO ABSTRACT A numerical prediction of the life of a gas turbine model disc by means of the finite-element technique is presented and the solution is compared with an experimental rim-spinning test. The finite-element method was used to obtain the K solution for a disc with two types of cracks, both at the notch root of the blade insert and located in the corner and in the centre. A crack aspect ratio of (a/c) = 1 was assumed. The fracture mechanics parameters J -integral and K were used in the assessment, which were computed with linear elastic and elastic,plastic material behaviour. Using a crack propagation program with appropriate fatigue-creep crack growth-rate data, previously obtained in specimens for the nickel-based superalloy IN718 at 600 °C, fatigue life predictions were made. The predicted life results were checked against experimental data obtained in real model discs. The numerical method, based on experimental fatigue data obtained in small laboratory specimens, shows great potential for development, and may be able to reduce the enormous costs involved in the testing of model and full-size components. [source] Coupled ghost fluid/two-phase level set method for curvilinear body-fitted gridsINTERNATIONAL JOURNAL FOR NUMERICAL METHODS IN FLUIDS, Issue 9 2007Juntao Huang Abstract A coupled ghost fluid/two-phase level set method to simulate air/water turbulent flow for complex geometries using curvilinear body-fitted grids is presented. The proposed method is intended to treat ship hydrodynamics problems. The original level set method for moving interface flows was based on Heaviside functions to smooth all fluid properties across the interface. We call this the Heaviside function method (HFM). The HFM requires fine grids across the interface. The ghost fluid method (GFM) has been designed to explicitly enforce the interfacial jump conditions, but the implementation of the jump conditions in curvilinear grids is intricate. To overcome these difficulties a coupled GFM/HFM method was developed in which approximate jump conditions are derived for piezometric pressure and velocity and pressure gradients based on exact continuous velocity and stress and jump in momentum conditions with the jump in density maintained but continuity of the molecular and turbulent viscosities imposed. The implementation of the ghost points is such that no duplication of memory storage is necessary. The level set method is adopted to locate the air/water interface, and a fast marching method was implemented in curvilinear grids to reinitialize the level set function. Validations are performed for three tests: super- and sub-critical flow without wave breaking and an impulsive plunging wave breaking over 2D submerged bumps, and the flow around surface combatant model DTMB 5512. Comparisons are made against experimental data, HFM and single-phase level set computations. The proposed method performed very well and shows great potential to treat complicated turbulent flows related to ship flows. Copyright © 2007 John Wiley & Sons, Ltd. [source] Development of a benzaldehyde sensor utilizing chemiluminescence on nanosized Y2O3LUMINESCENCE: THE JOURNAL OF BIOLOGICAL AND CHEMICAL LUMINESCENCE, Issue 6 2008Yayan Wu Abstract A chemiluminescence sensor has been proposed for sensitive determination of benzaldehyde, with nanosized Y2O3 as the sensing material. Under optimized conditions, the linear range of the CL intensity vs. the concentration of benzaldehyde vapour is 1.8 ng/mL,10.8 µg/mL (r2 = 0.9996), with a detection limit of 0.90 ng/mL (signal:noise ratio = 3:1). The sensor also exhibits high selectivity to benzaldehyde because no or weak CL signals have been detected when foreign substances are introduced into the sensor. In addition, the sensor also shows good stability and longer lifetime within 100 h. The results indicate that the proposed sensor, which has high sensitivity and selectivity, shows great potential for the detection of benzaldehyde. Copyright © 2008 John Wiley & Sons, Ltd. [source] Gene therapy for psoriasis in the K14-VEGF transgenic mouse model by topical transdermal delivery of interleukin-4 using ultradeformable cationic liposomeTHE JOURNAL OF GENE MEDICINE, Issue 6 2010Jiong Li Abstract Background Topical transdermal gene delivery to the skin shows great potential for painless, non-invasive administration of vaccines and therapeutic agents. Interleukin (IL)-4 strategies have shown a good antipsoriatic effect in clinic trials. To date, no information has been acquired on the effectiveness of gene therapy for psoriasis in the K14-VEGF transgenic mouse model by topical transdermal penetration of murine IL-4 (mIL-4) using ultradeformable cationic liposome (UCL). Methods In the present study, we synthesized an UCL and determined a suitable formula for transdermally delivering plasmid DNA to mouse skin. We then tested the antipsoriatic efficacy in the K14-VEGF transgenic mouse model by transdermal delivery of mIL-4 using UCL. Results We found that plasmid DNA was transdermally delivered to vicinal sites of epidermis and hair follicles using this optimized formula. Plasmid DNA expression was detected in ear skin. Twenty-four hours after topical application, plasmid DNA was not detected in blood serum and liver, which may decrease the risk of insertion of promoter from plasmid to genomic DNA. Mice treated with UCL/mIL-4 displayed a mild psoriasis phenotype. Histological analysis of pathological score using the Baker scoring system revealed an antipsoriatic effect. Immunohistochemical analysis revealed that hyperplastic and inflamed vessels were suppressed. Conclusions These observations provide evidence of antipsoriatic efficacy by topical transdermal delivery of mIL-4. Therefore, topical transdermal gene transfer is attractive and offers future potential for application in human patients with other dermatogic diseases. Copyright © 2010 John Wiley & Sons, Ltd. [source] Effect of single nucleotide polymorphisms of CAPN1 and CAST genes on meat traits in Nellore beef cattle (Bos indicus) and in their crosses with Bos taurusANIMAL GENETICS, Issue 4 2009R. A. Curi Summary The objectives of this work were to study the segregation of single nucleotide polymorphisms of the calpain 1, large subunit (CAPN1) and calpastatin (CAST) genes in Nellore (Bos indicus) and Nellore ×Bos taurus beef cattle, as well as to evaluate their effects on meat traits. For this, 300 animals, including 114 Nellore, 67 Angus × Nellore, 44 Rubia Gallega × Nellore, 41 Canchim, 19 Brangus three-way crosses and 15 Braunvieh three-way crosses, were genotyped for the CAPN4751 [AF_248054.2:g.6545C>T (GenBank accession AF248054.2)] and CAST/DdeI [AF_159246.1:g.2959A>G (GenBank accession AF159246.1)] polymorphisms and phenotyped for Ribeye Area, Backfat Thickness, Intramuscular Fat, Shear Force (SF) and Myofibrillar Fragmentation Index (MFI). In relation to the CAPN4751 polymorphism, a frequency of 10.5% was observed for the C allele in the Nellore breed. In the total sample of studied animals, a significant association was found between genotypes and meat tenderness, assessed by SF (P = 0.005) and MFI (P = 0.008), with genotype CT being more favourable than TT. For the CAST/DdeI polymorphism, a frequency of 55.7% was found for the A allele in the Nellore breed. In the total sample, a significant association was observed between genotypes and meat tenderness , SF (P = 0.004) and MFI (P = 0.006), with genotype AA being more favourable than AG. The relationship between genotypes and aged meat tenderness in confluence with the distribution of favourable alleles shows great potential for application of the CAPN4751 and CAST/DdeI polymorphisms in the genetic improvement of the Nellore breed, whilst contributing to the validation, in this breed and in its crosses with B. taurus, of the association results previously described in the literature.
