New reproductive anomalies in fruitless -mutant Drosophila males: Extreme lengthening of mating durations and infertility correlated with defective serotonergic innervation of reproductive organs

Abstract Several features of male reproductive behavior are under the neural control of fruitless (fru) in Drosophila melanogaster. This gene is known to influence courtship steps prior to mating, due to the absence of attempted copulation in the behavioral repertoire of most types of fru -mutant males. However, certain combinations of fru mutations allow for fertility. By analyzing such matings and their consequences, we uncovered two striking defects: mating times up to four times the normal average duration of copulation; and frequent infertility, regardless of the time of mating by a given transheterozygous fru -mutant male. The lengthened copulation times may be connected with fru -induced defects in the formation of a male-specific abdominal muscle. Production of sperm and certain seminal fluid proteins are normal in these fru mutants. However, analysis of postmating qualities of females that copulated with transheterozygous mutants strongly implied defects in the ability of these males to transfer sperm and seminal fluids. Such abnormalities may be associated with certain serotonergic neurons in the abdominal ganglion in which production of 5HT is regulated by fru. These cells send processes to contractile muscles of the male's internal sex organs; such projection patterns are aberrant in the semifertile fru mutants. Therefore, the reproductive functions regulated by fruitless are expanded in their scope, encompassing not only the earliest stages of courtship behavior along with almost all subsequent steps in the behavioral sequence, but also more than one component of the culminating events. © 2001 John Wiley & Sons, Inc. Microtubules in basalt glass from Hawaii Scientific Driling Project #2 phase 1 core and Hilina slope, Hawaii: evidence of the occurrence and behavior of endolithic microorganisms

ABSTRACT Elongate, fine tubes, ~1 µm wide and up to 200 µm long, extend from fractured surfaces, vesicle walls, and internal fractures into fragments of basalt glass in samples from the Hawaii Scientific Drilling Project #2 phase 1 (HSDP #21) core and the Hilina slope, Hawaii. Several features indicate that these tubes are microbial endolithic microborings: the tubes resemble many described microborings from oceanic basalt glass, their formation is postdepositional but restricted to certain but different ranges of time in the two sets of samples, and they are not uniformly distributed throughout glass fragments. Microtubules record several characteristic behaviors including boring into glass, mining, seeking olivine, and avoiding plagioclase. They also are highly associated with a particular form of glass-replacing smectite. Evidence of behavior should join morphological and geochemical criteria in indicating microbial alteration of basalt glass. In some samples, steeply conical tubes, ~10,20 µm in diameter tapering to 1 µm and commonly filled with smectite, appear to be modifications or elaborations of the microtubules. These also curve toward olivine and are associated with replacement smectite. In HSDP #21 samples, microtubules initiated at margins of shards before palagonite replaced those margins and are preserved during palagonitization. In fact, microtubules appear to have provided routes that enhanced the efficiency of water's reaching of unaltered glass. In Hilina Slope samples, the microtubules appear to postdate palagonitization because they initiate at the boundary between palagonite and unaltered sideromelane. Preservation of microtubules during palagonitization in samples together with recognition of other associated characteristics representing behavior suggests that such features may be recognizable in more heavily altered ancient rocks. Embryo development of Corticium candelabrum (Demospongiae: Homosclerophorida)

Abstract. Corticium candelabrum is a homosclerophorid sponge widespread along the rocky Mediterranean sublittoral. Scanning and transmission electron microscopy were used to describe the gametes and larval development. The species is hermaphroditic. Oocytes and spermatocytes are clearly differentiated in April. Embryos develop from June to July when the larvae are released spontaneously. Spermatic cysts originate from choanocyte chambers and spermatogonia from choanocytes by choanocyte mitosis. Oocytes have a nucleolate nucleus and a cytoplasm filled with yolk granules and some lipids. Embryos are surrounded by firmly interlaced follicular cells from the parental tissue. A thin collagen layer lies below the follicular cells. The blastocoel is formed by migration of blastomeres to the morula periphery. Collagen is spread through the whole blastocoel in the embryo, but is organized in a dense layer (basal lamina) separating cells from the blastocoel in the larva. The larva is a typical cinctoblastula. The pseudostratified larval epithelium is formed by ciliated cells. The basal zone of the ciliated cells contains lipid inclusions and some yolk granules; the intermediate zone is occupied by the nucleus; and the apical zone contains abundant electron-lucent vesicles and gives rise to cilia with a single cross-striated rootlet. Numerous paracrystalline structures are contained in vacuoles within both apical and basal zones of the ciliated cells. Several slightly differentiated cell types are present in different parts of the larva. Most cells are ciliated, and show ultrastructural particularities depending on their location in the larvae (antero-lateral, intermediate, and posterior regions). A few smaller cells are non-ciliated. Several features of the C. candelabrum larva seem to support the previously proposed paraphyletic position of homoscleromorphs with respect to the other demosponges. [source]


Obstructive sleep apnoea is associated with risk factors comprising the metabolic syndrome

ABSTRACT Background and objective: Several features of OSA syndrome suggest that it is a manifestation of the metabolic syndrome (MS). In this study, we investigated the prevalence of the MS among male Japanese patients with OSA, as well as the relationship between OSA in non-obese patients and components of the MS other than obesity (hypertension, dyslipidaemia and glucose intolerance). Methods: The study included 416 Japanese men who were diagnosed as having OSA by polysomnography. Among these, 101 non-obese patients were selected and the severity of OSA, as well as the prevalence of hypertension, dyslipidaemia and glucose intolerance, was assessed. Results: The MS was associated with OSA in 218/416 patients (52.4%). A significant increase in the prevalence of the MS was associated with increased severity of OSA, as categorized according to AHI. In the non-obese patients with OSA (mean age 57.6 years, BMI 22.7 kg/m2, AHI 34.3 events/h), hypertension, dyslipidaemia and glucose intolerance were identified in 70 (69.3%), 43 (42.6%) and 20 patients (19.8%), respectively. At least two of these factors were identified in 40 patients (39.6%). Non-obese patients with severe OSA had a significantly higher prevalence of two or more of these factors (33/59 patients, 55.9%). Conclusions: Although Asians are generally less obese than Caucasians, the prevalence of the MS was high among Japanese patients with OSA, and even among non-obese patients, OSA was associated with risk factors for the MS. Intercalary Elements, Treefrogs, and the Early Differentiation of a Complex System in the Neobatrachia

Abstract Intercalary elements are additional skeletal structures of digits of many anuran amphibians. Twelve terminal clades in the neobatrachian lineage of frogs have intercalary elements revealing it is a homoplastic character with five to seven gains and two to four losses along a consensus phylogeny of the Neobatrachia. We analyzed anatomical variation of intercalary elements, related structures (distal phalanges, tendons, and muscles), and articulations of digits of 45 anuran species, representing eight suprageneric terminal taxa. The intercalary elements are integrated in a complex system that is probably related to different types of movements, which are produced by a similar set of muscles and tendons with limited variation among the studied taxa. Species in the clades Hyloides and Ranoides show distinctive patterns of morphostructural features in their intercalary elements that are usually wedge-shaped and composed of hyaline cartilage in Ranoides, and biconcave and composed of embryonic cartilage in Hyloides. Features derived from the typical hyloid condition may only be interpreted in some Hylidae (Pseudis and Lysapsus) and Centrolenidae. In Ranoides, the described features of the intercalary elements are found in all taxa examined with the exception of Leptopelis, which have an intercalary element similar to the other Ranoides but formed by connective tissue. Several features are shared by all taxa having intercalary elements: (1) the intercalary elements differ from the phalanges by lacking terminal epiphyses, (2) they are present in hands and feet, and (3) they appear in all digits. This finding suggests that the genetic basis for presence of intercalary elements may be homologous in all these taxa and may have evolved only once early in neobatrachian history. Imagination and false memory inductions: investigating the role of process, content and source of imaginations

Three experiments examined the influence of imaginal encoding on false memory rates induced by the Deese-Roediger-McDermott (DRM) paradigm. Several features of imaginal encoding were investigated including the processes giving rise to the images (automatic or deliberate), the source of imagery scripts (participant or another person), and imagery content (e.g. individual or integrated images). Deliberately generating images led to a reduction in false memory rates relative to an imagery control condition. However, the effects on false memory rates very much depended on features characterizing the imaginal encoding. Most notably, when participants (rather than someone else) specified the scripts for guiding their imagery experiences, false memory rates were considerably less. Extending the generality of the distinctiveness heuristic as an explanation for the reduction in false memory rates following pictorial encoding, the results also speak to debates about the wisdom of using guided imagery techniques in applied settings. Research returns redux: a meta-analysis of the returns to agricultural R&D

A total of 289 studies of returns to agricultural R&D were compiled and these provide 1821 estimates of rates of return. After removing statistical outliers and incomplete observations, across the remaining 1128 observations the estimated annual rates of return averaged 65 per cent overall , 80 per cent for research only, 80 per cent for extension only, and 47 per cent for research and extension combined. These averages reveal little meaningful information from a large body of literature, which provides rate-of-return estimates that are often not directly comparable. This study was aimed at trying to account for the differences. Several features of the methods used by research evaluators matter, in particular assumptions about lag lengths and the nature of the research-induced supply shift. [source]


Effects of glucose and insulin on HepG2-C3A cell metabolism

Abstract HepG2, hepatocellular carcinoma cells, are used in drug toxicity studies and have also been explored for bioartificial livers. For these applications, the cells are under variable levels of nutrients and hormones, the effects of which on metabolism are poorly understood. In this study, HepG2-C3A cells were cultured under varying levels of glucose (high, low, and glucose-free) and insulin (without and with physiological levels of insulin) for 5 days. Cell growth was found to be comparable between high and low glucose media and lowest for glucose-free medium. Several features of central metabolism were affected profoundly by the medium glucose levels. Glucose consumption was greater for low glucose medium compared to high glucose medium, consistent with known glucose feedback regulation mechanisms. Urea productivity was highest in glucose-free medium. Further, it was seen that lactate acted as an alternative carbon source in the absence of glucose, whereas it acted as a sink for the high and low glucose media. Using a metabolic network flexibility analysis (MNFA) framework with stoichiometric and thermodynamic constraints, intracellular fluxes under varying levels of glucose and insulin were evaluated. The analysis indicates that urea production in HepG2-C3A cells arises via the arginase II pathway rather than from ammonia detoxification. Further, involvement of the putrescine metabolism with glutamine metabolism caused higher urea production in glucose-free medium consistent with higher glutamine uptake. MNFA indicated that in high and low glucose media, glycolysis, glutaminolysis, and oxidative phosphorylation were the main sources of energy (NADH, NADPH, and ATP). In the glucose-free medium, due to very low glycolytic flux, higher malate to pyruvate glutaminolytic flux and TCA cycle contributed more significantly to energy metabolism. The presence of insulin lowered glycerol uptake and corresponding fluxes involved in lipid metabolism for all glucose levels but otherwise exerted negligible effect on metabolism. HepG2-C3A cells thus show distinct differences from primary hepatocytes in terms of energy metabolism and urea production. This knowledge can be used to design media supplements and metabolically engineer cells to restore necessary hepatic functions to HepG2-C3A cells for a range of applications. Biotechnol. FEATURE-BASED KOREAN GRAMMAR UTILIZING LEARNED CONSTRAINT RULES

In this paper, we propose a feature-based Korean grammar utilizing the learned constraint rules in order to improve parsing efficiency. The proposed grammar consists of feature structures, feature operations, and constraint rules; and it has the following characteristics. First, a feature structure includes several features to express useful linguistic information for Korean parsing. Second, a feature operation generating a new feature structure is restricted to the binary-branching form which can deal with Korean properties such as variable word order and constituent ellipsis. Third, constraint rules improve efficiency by preventing feature operations from generating spurious feature structures. Moreover, these rules are learned from a Korean treebank by a decision tree learning algorithm. The experimental results show that the feature-based Korean grammar can reduce the number of candidates by a third of candidates at most and it runs 1.5 , 2 times faster than a CFG on a statistical parser. Evidence for distinct effects of GH and IGF-I in the metabolic syndrome

Abstract Aims, The metabolic syndrome is a cluster of cardiovascular risk factors which include central obesity, dyslipidaemia, glucose intolerance and hypertension. These risk factors are common in patients with growth hormone (GH) deficiency, suggesting a role for the somatotropic axis in the development of metabolic syndrome. Methods, We used factor analysis to investigate the relationships linking serum levels of GH and insulin-like growth factor I (IGF-I) to metabolic syndrome variables (high-density lipoprotein cholesterol, triglycerides, fasting glucose, blood pressure and waist circumference). We studied 359 men and 388 women from the Data from an Epidemiological Study on the Insulin Resistance syndrome (DESIR). Their age range was 30,64 years. Results, Three independent latent factors explained 61% of the total variance in women and four factors explained 73% in men. In both men and women, IGF-I showed a strong positive correlation with the lipid factor and a negative correlation with the obesity/glucose factor. In women, GH showed a strong negative correlation with the obesity/glucose factor but not the lipid factor. In men, GH was unrelated to the lipid and obesity/glucose factors. The blood pressure factor was not related to GH or IGF-I. In contrast with IGF-I, GH was significantly lower in women with metabolic syndrome (1575 ± 449 pg/ml) than in the other women (2121 ± 520 pg/ml, P = 0.002). No significant difference was observed in men for GH or IGF-I. Conclusion, Our results support a link between the somatotropic axis and several features of the metabolic syndrome, and suggest distinct effects of GH and IGF-I on these parameters. [source]


Elements of Resilience After the World Trade Center Disaster: Reconstituting New York City's Emergency Operations Centre

In this paper we examine the reconstitution of the Emergency Operations Centre (EOC) after its destruction in the World Trade Center attack, using that event to highlight several features of resilience. The paper summarises basic EOC functions, and then presents conceptions of resilience as understood from several disciplinary perspectives, noting that work in these fields has sought to understand how a natural or social system that experiences disturbance sustains its functional processes. We observe that, although the physical EOC facility was destroyed, the organisation that had been established to manage crises in New York City continued, enabling a response that drew on the resources of New York City and neighbouring communities, states and the federal government. Availability of resources , which substituted for redundancy of personnel, equipment and space , pre-existing relationships that eased communication challenges as the emergency developed and the continuation of organisational patterns of response integration and role assignments were among the factors that contributed to resilience following the attack. Experimentally Constrained Virulence is Costly for Common Cuckoo Chicks

Chicks of some avian brood parasites show high virulence by eliminating all host progeny in the nest whereas others develop in the presence of host nestmates. Common cuckoo (Cuculus canorus) chicks are typically highly virulent parasites as they attempt to evict all host eggs and chicks soon after hatching. However, several features of nest design, including steep walls and/or cavity nests, may effectively prevent cuckoo hatchlings from evicting nestmates. A previous observational study showed low success of cuckoo chicks in evicting progeny of a cavity nester host, the redstart (Phoenicurus phoenicurus) but cuckoo chicks showed low survival both when reared alone or in mixed broods with host nestmates. Whether poor cuckoo performance was caused by eviction costs and/or by the effect of presence of host chicks per se remains unclear. We experimentally cancelled any potential eviction costs by removing host eggs immediately after the cuckoo hatched and creating mixed broods 5 days later when the eviction instinct of the cuckoo already ceased. Cuckoos that were forced to compete with host nestlings experienced lower provisioning rates, poorer growth, and lower fledging success than control lone cuckoos. Cuckoos in mixed broods that survived until fledging fledged later, and at lower masses, than those in the sole cuckoo group. Thus, the cuckoo gens specializing on redstarts is similar to other cuckoo gentes, whose chicks are more successful in evicting host nestmates, and it does not benefit from the presence of host brood. Cohabitation with host nestlings then should be viewed as a maladaptive by-product of host cavity nest design. [source]

Characterization of the single-channel properties of NMDA receptors in laminae I and II of the dorsal horn of neonatal rat spinal cord

Abstract The single-channel properties of native NMDA receptors in laminae I and II of the dorsal horn of the neonatal rat spinal cord were studied using outside-out patch-clamp techniques. These receptors were found to have several features that distinguish them from native NMDA receptors elsewhere in the CNS. Single-channel currents activated by NMDA (100 nm) and glycine (10 µm) exhibited five distinct amplitude components with slope-conductance values of 19.9 ± 0.8, 32.9 ± 0.6, 42.2 ± 1.1, 53.0 ± 1.0 and 68.7 ± 1.5 pS. Direct transitions were observed between all conductance levels but transitions between 69-pS openings and 20-, 33- and 42-pS openings were rare. There was no significant difference in the frequency of direct transitions from 42- to 20-pS compared to 20- to 42-pS transitions. The Kb (0 mV) for Mg2+ was 89 µm. The Mg2+ unblocking rate constant was similar to other reported values. However, the Mg2+ blocking rate constant was larger than other reported values, suggesting an unusually high sensitivity to Mg2+. The NR2B subunit-selective antagonist, ifenprodil, had no significant effect on overall channel activity but significantly decreased the mean open time of 53-pS openings. These results suggest neonatal laminae I and II NMDA receptors are not simply composed of NR1 and NR2B subunits or NR1 and NR2D subunits. It is possible that these properties are due to an as yet uninvestigated combination of two NR2 subunits with the NR1 subunit or a combination of NR3A, NR2 and NR1 subunits. [source]

Abstract 2-Acyl- N -propargylindoles 1 and 2-acyl-3-propargylindoles 5 undergo aminobenzannulation reactions with pyrrolidine in the presence of an appropriate Lewis acid to give 9-aminopyrido[1,2- a]indoles 6 and 1-aminocarbazoles 7, respectively. The selection of the appropriate Lewis acid, TiCl4 or GaCl3 for 1 and InCl3 for 5, allows the domino process involving the initial formation of an enamine intermediate, followed by a regioselective 6- exo - dig intramolecular nucleophilic attack of the nucleophilic terminus of the unsaturated system (the ,-carbon of the enamino moiety) to the carbon,carbon triple bond. The changing face of HIV-associated lymphoma: what can we learn about optimal therapy inl the post highly active antiretroviral therapy era?

Abstract Epidemiological data indicate that the risk of developing non-Hodgkin lymphoma (NHL) in HIV positive individuals is related to age and CD4 count (i.e. degree of immunosuppression). The prognosis of patients with HIV-NHL has been shown to be linked to several features including age, stage, modified IPI, prior AIDS diagnosis, CD4 count, immunoblastic pathology, LDH, and HAART use. These features are, as would be expected, a mixture of prognostic factors relating to both the HIV, and to the NHL. Population studies indicate that the incidence of associated (HIV-NHL) may be reducing with the advent of HAART, although not all studies concur. However, most population-based studies have not as yet shown a significant improvement in the survival of patients with HIV-NHL with HAART. The optimal chemotherapy for these patients is unknown, although it is generally accepted that CNS prophylaxis is mandatory. There is currently no good evidence of any survival benefit with increased dose intensity from large RCT. However, it must be borne in mind that the large randomised studies comparing differing dose intensities were undertaken before the advent of effective HAART. There is some evidence that there may be a subset of good prognosis patients who may benefit from more intensive therapy.6 Given that the prognosis of patients with HIV can now be considerably improved with HAART, we cannot necessarily assume that the same results would apply with regard to chemotherapy dose intensity. There is some evidence that there is a survival benefit from the addition of HAART to chemotherapy, although this is retrospective. It is likely, however, that the reason for this is that the HAART improves the prognosis of the patients from their HIV, and therefore reduces the number of patients dying from other HIV-related illnesses whilst in remission from their lymphoma, as was seen in large numbers of patients in the earlier chemotherapy trials. It must not be forgotten that the prognosis of the patient's NHL is intimately linked to their prognosis with respect to the HIV. Although the number of patients with HIV-NHL is currently few, there is a need for more trials of chemotherapy, particularly now in the HAART era, when the prognosis from the point of view of the HIV has improved so much. In particular, the issue of dose intensity needs revisiting for patients whose overall prognosis can be improved by commencing HAART. Some accounts from Colchester relating to the collection of royal and local taxes between 1490 and 1502 illustrate several features of the way in which the operation was organized. The parishes of the borough and its dependent hamlets were adopted as units for the purpose, each with its assessors and collectors answerable to the borough chamberlain. An analysis of the lists of taxpayers, and the amounts they owed, reveals the extent to which the principles of assessment had departed from those practised up to 1334; in some ways they approached those subsequently adopted for assessing subsidies under the Tudors. Learning intravenous cannulation: a comparison of the conventional method and the CathSim Intravenous Training System

,,Intravenous cannulation is a nursing procedure carried out in some clinical units that may induce trauma and discomfort. Nurses should be well prepared before practising the procedure with clients. ,,Conventionally, a plastic arm was used for practice but, with innovative developments in technology, a computer program called the CathSim Intravenous Training System (CathSim ITS) is available for this purpose. This study was conducted to compare the effectiveness of learning using a plastic arm with the CathSim ITS. ,,Twenty-eight nurses were divided into two groups and randomly assigned to the two different methods: plastic arm (control group) or CathSim ITS (experimental group). Both groups were provided with 1 hour of theory input and 2 hours of nursing laboratory work. ,,When the nurses felt ready to practise on clients, their performances were assessed by researchers with an intravenous cannulation qualification using a validated checklist. Prior to the assessment, trait and state anxiety levels were measured using the State-Trait Anxiety Inventory (Hong Kong Chinese adaptation) to check whether anxiety would affect the performance. Following the assessment, a semi-structured interview was conducted to reveal any knowledge acquired through using the selected method. ,,Both the control and CathSim ITS groups demonstrated a high success rate, scoring 100% and 92.86%, respectively, with their first client. ,,The semi-structured interviews revealed that the CathSim ITS group appreciated several features of their assigned method. We report a 48-year-old male who presented with ulcerated plaques and nodules of the lower extremities. Skin biopsies revealed a dense lymphocytic infiltrate involving the dermis and the subcutis in a lobular and septal pattern. No overt cytological atypia was present. Notably, several features resembling lupus erythematosus were present, including vacuolar interface change and abundant dermal mucin deposition. The patient developed pulmonary nodules, and a lung biopsy showed a perivascular and interstitial lymphoid infiltrate without overt atypia. The cutaneous and pulmonary lymphoid infiltrates showed similar immunohistochemical profiles: CD3+CD4,CD8+/,CD56+. Monoclonal rearrangements of T-cell receptor gamma gene with similar migration patterns were identified from both locations. The patient developed fatal hemophagocytic syndrome, involving liver, spleen, lymph nodes, and bone marrow. This case is amongst rare reports of subcutaneous panniculitis-like T-cell Lymphoma (SPTCL) with systemic involvement. [source]


Wilson's disease with superimposed autoimmune features: Report of two cases and review

Abstract We describe two females, 15 and 23 years old, respectively, who presented with classical features of Wilson's disease (WD) and several features of autoimmune hepatitis (AIH). The first patient was initially diagnosed as AIH and treated with prednisolone which caused clinical improvement, with an increase of serum albumin from 22 to 30 g/L, and a decrease of aspartate aminotransferase from 103 to 47 U/L. Subsequent diagnosis of WD and introduction of penicillamine gave excellent improvement and complete normalization of liver function tests. The second patient, at first also diagnosed as having AIH, was treated with steroids and azathioprine with initial improvement, but subsequent deterioration. The diagnosis of WD was made 2 years after initial diagnosis of AIH, as the patient reached end-stage liver disease and required a transplant. Therefore, d -penicillamine treatment was not attempted. We conclude that, in patients with AIH, a thorough screening for WD is necessary, particularly when the response to steroid therapy is poor. Conversely, in patients suffering from WD with superimposed features of AIH, a combination of steroids and penicillamine may be of benefit. [source]


Alcohol Biomarkers in Applied Settings: Recent Advances and Future Research Opportunities

During the past decade, advances have been made in the identification, development, and application of alcohol biomarkers. This is important because of the unique functions that alcohol biomarkers can serve in various applied settings. To carry out these functions, biomarkers must display several features including validity, reliability, adequacy of temporal window of assessment, reasonable cost, and transportability. During the past two decades, several traditional alcohol biomarkers have been studied in multiple human studies. Meanwhile, several new, promising biomarkers, including various alcohol metabolites and alcohol biosensors, are being explored in human studies. In addition, researchers have explored using biomarkers in combination and using biomarkers in combination with self-reports, resulting in increased sensitivity with little sacrifice in specificity. Despite these advances, more research is needed to validate biomarkers, especially the new ones, in humans. Moreover, recent advances in high-throughput technologies for genomics, proteomics, and metabolomics offer unique opportunities to discover novel biomarkers, while additional research is needed to perfect newly developed alcohol sensors. Development of more accurate biomarkers will help practicing clinicians to more effectively screen and monitor individuals who suffer from alcohol use disorders. [source]


NeoHepatocytes From Alcoholics and Controls Express Hepatocyte Markers and Display Reduced Fibrogenic TGF-,/Smad3 Signaling: Advantage for Cell Transplantation?

Background:, Liver transplantation is the only definitive treatment for end stage liver disease. Donor organ scarcity raises a growing interest in new therapeutic options. Recently, we have shown that injection of monocyte-derived NeoHepatocytes can increase survival in rats with extended liver resection. In order to apply this technology in humans with chronic liver diseases in an autologous setting, we generated NeoHepatocytes from patients with alcoholic liver disease and healthy controls and compared those to human hepatocytes. Methods:, We generated NeoHepatocytes from alcoholics with Child A and B cirrhosis and healthy controls. Hepatocytes marker expression and transforming growth factor (TGF)-, signaling was investigated by RT-PCR, Western blot, immunofluorescent staining, and adenoviral reporter assays. Glucose and urea was measured photometrically. Phase I and II enzyme activities were measured using fluorogenic substrates. Neutral lipids were visualized by Oil Red O staining. Results:, There was no significant difference in generation and yield of NeoHepatocytes from alcoholics and controls. Hepatocyte markers, e.g., cytokeratin18 and alcohol dehydrogenase 1, increased significantly throughout differentiation. Glucose and urea production did not differ between alcoholics and controls and was comparable to human hepatocytes. During differentiation, phase I and II enzyme activities increased, however remained significantly lower than in human hepatocytes. Fat accumulation was induced by treatment with insulin, TGF-, and ethanol only in differentiated cells and hepatocytes. TGF-, signaling, via Smad transcription factors, critically required for progression of chronic liver disease, was comparable among the investigated cell types, merely expression of Smad1 and -3 was reduced (,30 and ,60%) in monocytes, programmable cells of monocytic origin, and NeoHepatocytes. Subsequently, expression of TGF-, regulated pro-fibrogenic genes, e.g., connective tissue growth factor and fibronectin was reduced. Conclusions:, Generation of NeoHepatocytes from alcoholics, displaying several features of human hepatocytes, offers new perspectives for cell therapeutic approaches, as cells can be obtained repeatedly in a noninvasive manner. Furthermore, the autologous setting reduces the need for immunosuppressants, which may support recovery of patients which are declined for liver transplantation. [source]


Mycoplasma genitalium: the aetiological agent of urethritis and other sexually transmitted diseases

ABSTRACT Mycoplasma genitalium was first isolated in 1980 from two of 13 men with non-gonococcal urethritis (NGU). It shares several features with M. pneumoniae, a recognized respiratory tract pathogen. It is extremely difficult to isolate by culture. The development of sensitive and specific polymerase chain reaction (PCR) assays in the early 1990s made clinical studies possible and a significant number of publications have shown a strong association between M. genitalium and NGU, independent of Chlamydia trachomatis. The purpose of this review is to evaluate the currently available information on the associations between M. genitalium and urogenital tract infections in men and women and assess their fulfilment of the Henle,Koch postulates. It is concluded that there is very strong evidence that M. genitalium is a cause of NGU in men and cervicitis in women. Evidence for upper genital tract infections in women has begun to accrue, but further studies are needed. The optimal treatment of M. genitalium infections remains to be determined, but antibiotics of the macrolide group appear to be more active than tetracyclines. [source]


Severe blunt trauma in dogs: 235 cases (1997,2003)

Abstract Objective , To evaluate population characteristics, injuries, emergency diagnostic testing, and outcome of dogs with blunt trauma requiring intensive care in an urban hospital. Design , Retrospective study 1997,2003. Setting , All data obtained from the University of Pennsylvania , Matthew J. Ryan Veterinary Hospital. Animals , Dogs admitted to the intensive care unit for treatment following blunt trauma. Interventions , None. Measurements and Main results , Of the 235 dogs that met inclusion criteria, 206 (88%) survived and 29 (12%) did not survive. Blunt vehicular trauma accounted for 91.1% of cases. Mild hyperglycemia and hyperlactatemia was common in both survivors and nonsurvivors. The chest was the most common region traumatized and the prevalence of polytrauma was 72.3%. Initial weight, vital signs, PCV, total plasma protein, BUN, glucose, lactate, acid-base status, and electrolytes did not differ between survivors and nonsurvivors. Nonsurvivors were significantly more likely to have had head trauma (P=0.008), cranium fractures (P<0.001), recumbency at admission (P<0.001), development of hematochezia (P<0.001), clinical suspicion of acute respiratory distress syndrome (P<0.001), disseminated intravascular coagulation (P<0.001), multiorgan dysfunction syndrome (P<0.001), development of pneumonia (P<0.001), positive-pressure ventilation (P<0.001), vasopressor use (P<0.001), and cardiopulmonary arrest (P<0.001). Conclusions , Outcome of severe blunt trauma in dogs treated with intensive care is very good. Despite the high survival rate, several features associated with poor outcome were identified. Neither admission lactate nor glucose was able to predict outcome. [source]


A scratch circle origin for the medusoid fossil Kullingia

Kullingia is considered a key taxon in demonstrating the presence of terminal Proterozoic-early Cambrian chondrophorine hydrozoans. However, Kullingia concentrica from the Lower Cambrian of northern Sweden possesses several features that show that it is not a body fossil but that it was formed by current or wave-induced rotation of an anchored tubular organism, possibly a sabelliditid. A scratch circle interpretation applies also to several other reports of Lower Cambrian Kullingia, including Kullingia delicata from the Chapel Island Formation of Newfoundland. Given the considerable number of problematic fossils that have been interpreted as chondrophorines, it is difficult to put an age on the oldest fossil chondrophorines, but it may be as late as the Devonian. Overall, scratch circles appear to be rarely preserved. The occurrence of these scratch circles in Lower Paleozoic storm deposits is probably related to low levels of bioturbation that enhanced both the likelihood of formation and preservation of these structures. Individual differences in the drive to overeat

Summary, Obesity is considered a public health crisis. In order to tackle this, an enhanced appreciation of what drives some consumers to overeat is required. It is clear that several features of the modern environment encourage over-consumption in many people, at least some of the time. However, there is variation in the extent to which this translates into weight gain. Understanding whether susceptibility is dispositional or learned will shape prevention and treatment programmes. This review identifies situational cues that promote overeating and dispositional traits, such as impulsivity and sensitivity to reward, which make some individuals specifically vulnerable to overeating. These traits are likely to contribute to the development of the obese state, but also present barriers to weight management. Personalised nutrition and behavioural strategies may provide a novel and innovative approach, as these take account of individual differences in vulnerability to overeating. [source]


THE AFFINITIES OF THE ENIGMATIC DINOSAUR ESHANOSAURUS DEGUCHIIANUS FROM THE EARLY JURASSIC OF YUNNAN PROVINCE, PEOPLE'S REPUBLIC OF CHINA

Abstract:,Eshanosaurus deguchiianus is based on a single left dentary from the Lower Lufeng Formation (Lower Jurassic) of Yunnan Province, China. It was originally identified as the earliest known member of Therizinosauroidea (Theropoda: Coelurosauria), a conclusion that results in a significant downward range extension for this clade (>65 million years) and for many other major lineages within Coelurosauria. However, this interpretation has been questioned and several authors have proposed that the anatomical features used to refer Eshanosaurus to Therizinosauroidea are more consistent with attribution to a basal sauropodomorph dinosaur. Detailed consideration of the holotype specimen suggests that several features of the dentary and dentition exclude Eshanosaurus from Sauropodomorpha and support its inclusion within Therizinosauroidea. If accepted as an Early Jurassic coelurosaur, Eshanosaurus has important implications for understanding the timing and tempo of early theropod diversification. Moreover, its provenance also suggests that substantial portions of the coelurosaur fossil record may be missing or unsampled. However, the Early Jurassic age of Eshanosaurus requires confirmation if this taxon is to be fully incorporated into broader evolutionary studies. [source]

After discussing the arguments for and against a fiscal stabilization program, I discuss several features a reasonable program should have. Applying those criteria to the German fiscal package, I find that while it satisfies most of them, it has two major flaws. First, the fiscal stimulus is likely to come too late, and second, the political marketing of the policy was disastrous. As a consequence the policy is likely to stabilize the economy in the second half of 2009 and in 2010, but did little to soften the initial blow of the recession. [source]


Chemically-mediated host-plant location and selection by root-feeding insects

Abstract., Recent studies have shown that root-feeding insects can be of considerable importance in terms of agricultural damage, their indirect impacts on above-ground herbivores and their efficacy as biocontrol agents of weeds. To date, isolated studies have made it difficult to identify the mechanisms by which soil-dwelling insects locate and select host-plant roots. This review synthesizes 78 studies describing root location and selection. Soil insect herbivores do not rely on encountering roots at random, but orientate towards them using semiochemicals that enable specialist insects to distinguish host-plants from unsuitable plants. Secondary plant metabolites released into the rhizosphere (alcohols, esters and aldehydes representing 37% of reported examples) underpin host-plant location and recognition, with 80% having ,attractant' properties. Insects feeding on a limited range of plants tend to exploit host-specific secondary metabolites, whereas nonspecialist feeders appear to use more general semiochemicals. When insects reach the roots, contact chemosensory cues act as either ,phagostimulants' (48% of the compounds being sugars) or feeding ,deterrents' (notably phenolic compounds). Twenty studies conclude that CO2 is the major primary plant metabolite that allows insects to locate to roots. However, several features of CO2 emissions from roots mitigate against it as a precise location cue. In addition to its lack of specificity, gradients of root emitted CO2 do not persist for long periods and vertical gradients of CO2 in the soil tend to be stronger than horizontal gradients. A conceptual model is presented, emphasizing the importance of soil properties (e.g. porosity, moisture) on chemical diffusion and insect motility. [source]


The 11 August 2006 squall-line system as observed from MIT Doppler radar during the AMMA SOP

Abstract On the evening of 9 August 2006, a mesoscale convective system (MCS) having a north-south oriented squall-line organization formed over the border between Chad and Nigeria. It propagated westward, intensified over Nigeria on 10 August, and reached Niamey (Niger) at 0320 UTC on 11 August. Its passage over Niamey was accompanied by dust lifting and was well tracked by the Massachusetts Institute of Technology (MIT) Doppler radar. The three-dimensional structure of the airflow and precipitation pattern is investigated from regular radar volume scans performed every ten minutes between 0200 and 0321 UTC. The 3D wind components are deduced from the multiple-Doppler synthesis and continuity adjustment technique (MUSCAT) applied to a set of three volume scans obtained over a time period of one hour, which are equivalent to a three-radar observation of the squall line when considering a reference frame moving with the system and the hypothesis of a stationary field. Results of the wind synthesis reveal several features commonly observed in tropical squall lines, such as the deep convective cells in front of the system, fed by the monsoon air and extending up to 15 km altitude, and the well-marked stratiform rain region at the rear, associated with mesoscale vertical motions. Forward and trailing anvils are clearly identified as resulting from the outflow of air reaching the tropopause and transported to this level by the sloping convective updraughts occurring in a sheared environment. In the northern part, a deeper and stronger front-to-rear flow at mid-levels is found to contribute to the rearward deflection of the leading line and to promote a broader (over 300 km) stratiform cloud region. Eddy vertical transports of the cross-line momentum mainly accounts for the mid-level flow acceleration due to a momentum redistribution from low to higher levels. The height distribution of hydrometeors and their associated production terms derived from a one-dimensional microphysical retrieval model indicate the distinct roles of the convective and stratiform regions in the formation of graupel and rain, and the respective contributions of cold (riming) and warm (coalescence, melting) processes. Cooling from melting, and heating/cooling from condensation/evaporation processes yield a net decrease and increase of the potential temperature at low and mid-to-upper levels, respectively, with respect to an environmental thermodynamic profile taken three hours ahead of the analysis. Finally, the upper-level rearward flow could convey the non-negligible proportion of ice particles farther from the leading deep convection to the trailing stratiform region, thereby favouring the extent of this region.
