Christon PhD Dietary long chain omega-3 polyunsaturated fatty acids from fish oil appear to be clearly efficient in regulating endothelial dysfunction (or activation), which is the first stage of atherogenesis. Studies on endothelial cells in vitro have shown that the main dietary PUFA and oleic acid may prevent endothelium activation either by inhibiting the expression of adhesion molecules or by improving the nitric oxide production. Saturated fatty acids and also linoleic acid do not inhibit endothelium activation. The mechanisms involved in this inhibition could be related to endothelial cell membrane characteristics or redox status. However, these findings need to be confirmed in vivo. [source] Increased vigabatrin entry into the brain by polysorbate 80 and sodium caprateJOURNAL OF PHARMACY AND PHARMACOLOGY: AN INTERNATI ONAL JOURNAL OF PHARMACEUTICAL SCIENCE, Issue 2 2001D. Dimitrijevic The effects of a non-ionic surfactant, polysorbate 80, and the sodium salt of the saturated fatty acid, sodium caprate (C10), as potential brain absorption enhancers for vigabatrin were studied. Vigabatrin is an enzyme-activated irreversible inhibitor of ,-aminobutyric acid (GABA) transaminase that increases brain and cerebrospinal GABA concentrations in animals and man. Before intravenous administration, a range of concentrations of the surfactants were tested using erythrocyte lysis or the red blood cell lysis test to establish the non-toxic concentration range. Vigabatrin was dissolved in 0.1% polysorbate 80 and 0.1% sodium caprate and administered intravenously in doses of 4 mL kg,1 to male Wistar rats (230,250 g; n = 3). Rats were killed 2 h after drug and surfactant administration and the brains were immediately removed and homogenized in 0.4m perchloric acid. Selected ion monitoring electrospray mass spectrometry was used to determine the concentration of vigabatrin and GABA directly from the perchloric acid extract of the rat brain. This method was developed to increase the speed and efficiency of the analysis by removing the need for complex extraction and derivatization procedures while retaining the specificity of the mass spectrometer as a detector. The stability of both vigabatrin and GABA in perchloric acid was established by monitoring their pseudo molecular ions in standard solutions at timed intervals over 24 h. Although the detection level for vigabatrin and GABA was at least 50 pg, only GABA was detected in rat brain. Vigabatrin caused a small increase in whole brain GABA. However, GABA levels were higher in the samples with vigabatrin + enhancer than in the samples where vigabatrin alone was administered. One-way analysis of variance indicated a significant effect of the surfactants on GABA levels (F (5,17) = 11.86, P < 0.01) and vigabatrin absorption was presumed. The rectal temperature of the rats is lowered by the presence of vigabatrin in the brain. Vigabatrin alone decreased rectal temperature by 6%. When given with either polysorbate 80 or sodium caprate, the extent of temperature lowering was significantly greater (P < 0.001). There was no significant difference after 2 h between polysorbate 80 + vigabatrin, and sodium caprate + vigabatrin. [source] Dietary effects on growth, plasma lipid and tissues of rats fed with non-conventional oil of Telfairia occidentalisJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 13 2004Ibironke A Ajayi Abstract The nutritional value of Telfairia occidentalis seed and seed oil available in local market was studied in rats. The average values of the proximate analysis on dry matter basis were 203 g kg,1 crude protein, 536.3 g kg,1 fat, 39 g kg,1 ash, 95 g kg,1 crude fibre and 43.7 g kg,1 carbohydrate, while the moisture content was 75 g kg,1. The mineral analyses showed that T occidentalis seeds contain potassium (3280 mg kg,1), sodium (1230 mg kg,1), magnesium (60.8 mg kg,1), calcium (60.8 mg kg,1) and iron (13.0 mg kg,1). The oil extracted from the seed sample showed physicochemical properties that suggest its suitability for edible and industrial uses. The oil contained high levels of unsaturated fatty acids, oleic and linoleic, which accounted for 63.22% of total fatty acids recovered, while palmitic acid (27.4% of the oil by weight) was the most prevalent saturated fatty acid in the oil. No trace of eleostearic acid was detected in the oil. The effect on physical appearance, weight gain, organ weight, tissue and plasma cholesterol and triacylglycerol levels was determined in rats fed a diet containing 5% T occidentalis oil. These were compared with those of rats fed a control diet (0% T occidentalis oil) over an 8 week study period. Weekly monitoring of the rats showed good physical appearance and steady weight increase. The test rats were not anaemic and no mortality was recorded. While there was a significantly higher (p < 0.05) level of cholesterol in the heart of test rats compared with the control, there was a significantly lower (p < 0.05) plasma cholesterol level, suggesting that T occidentalis seed oil can be used to lower plasma levels of cholesterol. Histological examination of the sections of the heart, liver, kidney and spleen of the test rats revealed varying degrees of pathology. These include artherosclerotic changes in the walls of vasa vasori and lymphatics in the heart and hyaline degeneration of myofibrils, fatty infiltration of hepatocytes, and glomerular and tubular degeneration in the kidneys. These lesions may have been due to the effect of fatty acids, especially erucic acid, in the seed oil, which at 1.24% (less that the 5% conventional allowable limit for edible oils) may be intolerable for rats and may be unsuitable for human consumption. The seed oil may be used for industrial purposes such as soap making, lubrication and cosmetics for which it is well suited. Bondaruk Abstract A cDNA encoding palmitoyl-acyl carrier protein (ACP)-desaturase from cat's claw (Doxantha unguis-cati L.) was expressed in Arabidopsis thaliana and Brassica napus L. with the goal of decreasing the saturated fatty acid (FA) content of the seed oil. In general, transformation of Arabidopsis resulted in a greater change in the FA composition of the seed oil than for B. napus. An increase in palmitoleic acid (16:1cis,9) was obtained in transgenic lines, suggesting that the 16:0-ACP-desaturase cDNA was expressed in the manner originally intended. Other effects on lipid metabolism, however, were observed in the seed of transgenic plants. In Arabidopsis, there was a large increase in the proportions of cis -vaccenic acid (18:1cis,11) and cis -13-eicosenoic acid (20:1cis,13), possibly generated through elongation of 16:1cis,9. Elongation of 18:1cis,11 to 20:1cis,13, however, was not observed in B. napus indicating that certain aspects of lipid metabolism in the model plant, Arabidopsis, may not apply to B. napus. As well, the appearance of 18:1cis,11 was accompanied by a decrease in the proportion of oleic acid (18:1cis,9). Although the introduced ACP-desaturase resulted in synthesis of some unsaturated FAs, the overall saturated FA content was maintained at similar levels to the control or was enhanced. Increased levels of saturation were mainly associated with an increase in stearic acid, which unlike 16:0, is considered non-atherogenic. The results suggest that a mechanism exists further downstream in oil biosynthesis to counteract the decrease in saturation brought about by the 16:0-ACP-desaturase action. [source] Effects of GH gene polymorphism and sex on carcass traits and fatty acid compositions in Japanese Black cattleANIMAL SCIENCE JOURNAL, Issue 1 2009Astrid ARDIYANTI ABSTRACT To investigate the effects of bovine growth hormone (bGH) gene polymorphism on carcass traits and fatty acid compositions in Japanese Black cattle caused by nucleotide substitution of CTG (allele A)/GTG (allele B) at codon 127 and of ACG (allele A and B)/ATG (allele C) at codon 172 of bGH, GH genotypes of 135 cattle were determined using allele specific-multiplex polymerase chain reaction (PCR). Allele A gave greater rib thickness and lower melting point of fat (MP) while allele B gave higher C18:1% (P < 0.05). Allele C gave higher C18:1, monounsaturated fatty acid (MUFA), unsaturated fatty acid (USFA) percentages (P < 0.05). It also gave lower saturated fatty acid (SFA) percentages, higher MUFA/SFA and USFA/SFA ratios, and lower MP (P < 0.05). Interactions of sex and GH alleles were analyzed. In heifers, allele A gave higher carcass weight, daily carcass gain, rib eye area, rib thickness, subcutaneous fat thickness, and BMS while allele B gave greater rib eye area and rib thickness (P < 0.05). Allele C gave higher C18:1 (P < 0.01), MUFA (P < 0.01), USFA percentages (P < 0.05) and MUFA/SFA and USFA/SFA ratios (P < 0.01), and lower C16:0 and SFA percentages (P < 0.05) and MP (P < 0.01). GH gene polymorphism affected carcass traits and fatty acid compositions although the effects were more pronounced in heifers. [source] LIPID, CHOLESTEROL AND FATTY ACID PROFILE OF SOME COMMERCIALLY IMPORTANT FISH SPECIES FROM SOUTH CASPIAN SEAJOURNAL OF FOOD BIOCHEMISTRY, Issue 4 2010S. PIRESTANI ABSTRACT The fatty acid, lipid, cholesterol and energy contents in five commercially important fish species from South Caspian Sea (common kilka, Caspian kutum, golden gray mullet, common carp and pike perch) were evaluated. The fatty acid compositions of these five fish species ranged from 28.99 to 41.05% saturated fatty acids, 40.99,56.25% monounsaturated fatty acids and 14.22,23.03% polyunsaturated fatty acids. Among these, those occurring in the highest proportions were palmitic acid (20.42,27.9%), palmitoleic acid (11.09,26.26%), oleic acid (16.1,36.94%), eicosapentaenoic acid (3.22,7.53%) and docosahexaenoic acid (3.86,11.36%). The lipid, cholesterol and energy contents ranged from 1.97% to 10.23%, 57,302 mg/100 g and 4365.4,5544.2 cal g/dm, respectively. The obtained Statistical results showed that in these fishes, many of the above mentioned indices had significant differences (P , 0.01) and the cluster analysis results of fatty acid compositions showed that common carp and pike perch had good similarity, followed by the Caspian kutum and golden gray mullet. However, common kilka did not show any similarity to others. PRACTICAL APPLICATIONS Fish consumption has been linked to health benefits such as reduced risk of coronary heart disease. This is largely attributed to the lipid, cholesterol, energy contents, fatty acid compositions and the polyunsaturated fatty acids present in fish oils. The ,-3 : ,-6, polyunsaturated fatty acid/saturated fatty acid and eicosapentaenoic acid + docosahexaenoic acid/C16 ratios are considered to be useful criteria for comparing relative nutritional and oxidation values of fish oils. [source] ROLE OF SATURATED FATTY ACIDS IN LIPASE PRODUCTION , USING PSEUDOMONAS AERUGINOSAJOURNAL OF FOOD BIOCHEMISTRY, Issue 6 2007A.N. SARAVANAN ABSTRACT Complex substrates always induce substantial amount of enzyme production during hydrolysis by microorganisms. In this study, ghee was taken for its saturated fatty acid content and analyzed as an inducer for the production of lipase. With ghee emulsion, the bacterium Pseudomonas aeruginosa at optimal condition produced 60 units/min/L at 72 h. With olive oil emulsion, this organism produced only 41 units/min/L as maximum at 96 h. The saturated fatty acids present in ghee make it a hard substance for hydrolysis, which is the reason for the increased enzyme production. This was evaluated by the iodine number experiment. Ghee can also reduce the production cost whereas the costlier olive oil constitutes 25,50% of the total production cost for a commercial scale. The experimental results showed that the saturated fatty acids play an important role in lipase enzyme induction by P. aeruginosa. The use of ghee is cost-effective; hence, it can be used as a potential inducer for lipase production. PRACTICAL APPLICATIONS Lipases are industrially very important enzymes. They are used in pharmaceutical, food, soap and other industries. In lipase production, olive oil is the main constituent. Comparatively, olive oil is costlier; hence, it increases the production cost of lipase. So, this study was done to replace olive oil with a much cheaper ghee using Pseudomonas aeruginosa. The ghee-containing medium gave a very good result because of the presence of complex saturated fatty acids. The ghee-containing medium produced 60 units/min/L at 72 h. The olive oil medium, which contains mainly unsaturated fatty acids, produced only 41 units/min/L as maximum at 96 h. Hence, in the commercial scale, ghee can reduce raw material cost as well as operation time cost significantly when it is used as substrate. [source] The efficacy of dietetic intervention in patients with chronic obstructive pulmonary diseaseJOURNAL OF HUMAN NUTRITION & DIETETICS, Issue 4 2008L. Bottle Background:, Clinical trials have shown that pulmonary rehabilitation can improve the functional status and quality of life of chronic obstructive pulmonary disease (COPD) patients (Lacasse, 2006) but there is no research examining the efficacy of group dietetic intervention during standard 8 week rehabilitation courses. Current input is usually limited to a 1 h nutrition education session. This pilot study aimed to investigate whether patients receiving additional dietetic intervention during pulmonary rehabilitation significantly increased their general nutritional knowledge, thereby facilitating improvements in dietary intake and nutritional status. Methods:, Patients were recruited from two courses of pulmonary rehabilitation and randomly allocated to a control group or an intervention group. Anthropometry (height, weight, body mass index, mid arm circumference and triceps skinfold), 3 day food diaries and nutritional knowledge questionnaires covered guidelines, food groups, choosing healthy options and diet and COPD were completed at baseline and at the end of 8 weeks. In week 2 both groups received the same nutrition education session which covered healthy eating during periods of stability as well as advice on coping with loss of appetite and reduced intake during illness and exacerbations. The intervention group was followed up during weeks 4, 6 and 7 when further anthropometric measurements were taken and additional dietary advice was provided, which addressed issues raised by individual patients. Information from food diaries was converted to nutrients using Windiets dietary analysis software. Statistical analyses were carried out using SPSS (v14) and included Mann,Whitney U non parametric tests, paired t -tests and Spearman correlations used for comparisons over time and between groups. For analysis purposes patients were classified as normal weight (NW) and overweight (OW). Approval was obtained from the appropriate Ethics Committee. Results:, Changes reported were not statistically significant (P > 0.05). Complete data sets were obtained for six control (NW = 2, OW = 4) and five intervention (NW = 1, OW = 4) patients. Nutritional knowledge increased in the control group by 5% compared to 3% in the intervention group. Control NW patients increased their energy intake resulting in a mean weight gain of 0.5 kg (SD 3.3). OW control group patients increased their energy intake by 12.4% (16.9) with a mean weight gain of 0.2 kg (2.5). All control patients increased their intake of in total fat, saturated fatty acids (SFA), sugars and sodium. Conversely there was a decrease in energy intake in the intervention group of 14.4% (17.8) and a mean weight loss of 1.5 kg (1.2) (three out of four overweight patients lost weight). Improvements in diet were shown with reduced intakes of total fat, SFA, sugars and sodium. The NW patient in the intervention group regained weight that had previously been lost. These changes did not correlate with changes in nutritional knowledge. Discussion:, An increase in nutritional knowledge was expected to facilitate appropriate changes in dietary intake and nutritional status. Despite the lack of correlation between dietary knowledge and intake, beneficial outcomes were none-the-less observed in the intervention group. The trend for weight gain in OW control group patients, and weight loss in OW intervention group patients contrasted with results seen by Slinde et al. (2002) where the control OW patients lost weight, and OW intervention patients gained weight. It is possible that in the current study, patients in the intervention group were motivated to lose weight with repeated exposure to the dietitian, rather than an increase in nutritional knowledge. Significant anthropometrical changes were unlikely to be observed in 8 weeks, and further follow up may be necessary to establish sufficient evidence for the most efficacious level of dietetic intervention. The small sample sizes, especially with regard to weight sub groups, limits the conclusions which can be drawn. Further research is recommended, using a larger sample size, in order to make recommendations for dietetic best practice. Conclusion:, The results of this study did not show statistical significance and the association between nutritional knowledge and improved nutritional outcomes remains unclear. However, the findings may have clinical significance since they appear to show that additional dietetic intervention may benefit the nutritional status of patients with COPD attending pulmonary rehabilitation. For this purpose a total of 160 Lohmann Brown laying hens 36 weeks of age were allocated to four dietary treatments with one control group and three treatment groups. Black cumin seed (Nigella sativa L.) was used at the level of 5, 10 and 15 g kg,1 in the diets of the first, second and third treatment groups, respectively. RESULTS: Dietary treatments did not significantly affect body weight, feed intake, egg production, egg quality characteristics and blood parameters. Diets containing 10 and 15 g kg,1 black cumin seed increased egg weight (P < 0.01), improved feed efficiency (P < 0.01) and decreased egg yolk cholesterol, saturated fatty acids (% of total fatty acid methyl esters) and the ratio of saturated to unsaturated fatty acids (P < 0.05) compared to the diet of control group. CONCLUSION: Dietary black cumin seed at the level of 10 and 15 g kg,1 had beneficial effects on egg weight, feed efficiency, egg cholesterol content and egg yolk fatty acid composition. Therefore it can be used at the level of 10 and 15 g kg,1 in the diets of laying hens. Seeds contained on average 71.5 g kg,1 dry matter, 61.9 g kg,1 crude oil, 9.4 g kg,1 protein, 507.4 g kg,1 crude fibre, 12.3 g kg,1 ash and 409.0 g kg,1 carbohydrate. The fatty acid composition of prickly pear seed oil consisted of 1.3,1.9 g kg,1 myristic (14:0), 132.1,156.0 g kg,1 palmitic (16:0), 14.4,18.5 g kg,1 palmitoleic (16:1), 33.1,47.9 g kg,1 stearic (18:0), 210.5,256.0 g kg,1 oleic (18:1), 522.5,577.6 g kg,1 linoleic (18:2), 2.9,9.7 g kg,1 linolenic (18:3), 4.2,6.6 g kg,1 arachidic (20:0) and 2.1,3.0 g kg,1 behenic (22:0) acids, which is comparable with that of corn oil. No statistical difference in seed weight ratio was determined during the maturation period, whereas changes in the saturated fatty acids of the seed oil were observed. From this study it can be concluded that the seeds of prickly pear are suitable as animal feed. However, there will be much greater challenges in the future. Based on the current population growth rate of 1.4% per year, the world's population is forecast to increase from the current level of approximately six billion to nine to twelve billion in 50 years. In addition to continuously increasing demand for agricultural production, there is an urgent need to improve the nutritional quality of human diets for this rapidly growing human population. Malnutrition is still a worldwide health issue. Macronutrient and micronutrient deficiencies are prevalent in developing countries, and over-consumption of certain nutrients in developed countries (e.g., saturated fatty acids) is associated with high incidence of certain chronic diseases. Furthermore, there will be declining natural resources such as arable land and water, and the challenges to humans must be met without further degrading the environment. Biotechnology offers a valuable tool to help achieve these goals. This review focuses on the most recent advances in biotechnology, which promise to improve human nutrition by enhancing the nutrient density of plant foods. Issues relating to the safety of food products from genetically engineered crops are also discussed. [source] Changing of fat content and fatty acid profile of reared pike (Esox lucius) fed two different dietsAQUACULTURE RESEARCH, Issue 1 2006Balázs Kucska Abstract Fat content and fatty acid profile of two pike yearling groups grown on two different diets were compared. The groups originated from culture on artificial feed. One group (cultured pike fed first pellet then prey-fish (PF)) was fed with natural food, live prey-fish, while the other one (cultured pike fed exclusively pellet (PP)) by trout feed through a 3-month experimental period. Growth of pike was lower with PP than with PF. The trout feed resulted in an increase of the fat content of fillet and the formation of abdominal fat depots. Feeding of natural food decreased the fat content. The proportion of the saturated fatty acids in fillet was higher in the (PF) group. The n-6 fatty acids (arachidonic C20:4n-6 and docosatetraenic C22:4n-6 acids) were lowest in PP-fed pike. Regarding total n-3 fatty acids ratio there was no significant difference between the groups, but the level of ,-linolenic (C:18:3n-3) acid showed significant difference among groups. GREGG This paper presents evidence for increased recovery of organic residues from archaeological pottery through use of a microwave-assisted liquid chromatography protocol. C16:0 and C18:0 saturated fatty acids were obtained from archaeological potsherds recovered from nine Neolithic settlements in the Middle East dating between 4700 and 7300 cal bc, including materials that had not produced evidence for the survival of any lipid species through use of ,conventional' solvent extraction techniques. Compound-specific isotopic analyses of C16:0 and C18:0 fatty acids in potsherd extracts subsequently revealed ,13C/,12C compositions consistent with modern subcutaneous fats of wild boar and goats pastured on lands adjacent to the Jordan Valley, and residues from a modern pottery vessel used in the manufacturing of butter, cheese and yogurt in central Turkey. These results are presented as an illustration of capabilities of the microwave-assisted recovery protocol. [source] Evidence for two-step regulation of pheromone biosynthesis by the pheromone biosynthesis-activating neuropeptide in the moth Heliothis virescens,ARCHIVES OF INSECT BIOCHEMISTRY AND PHYSIOLOGY (ELECTRONIC), Issue 3 2007H. Eltahlawy Abstract The control of pheromone biosynthesis by the neuropeptide PBAN was investigated in the moth Heliothis virescens. When decapitated females were injected with [2- 14C] acetate, females co-injected with PBAN produced significantly greater quantities of radiolabeled fatty acids in their pheromone gland than females co-injected with saline. This indicates that PBAN controls an enzyme involved in the synthesis of fatty acids, probably acetyl CoA carboxylase. Decapitated females injected with PBAN showed a rapid increase in native pheromone, and a slower increase in the pheromone precursor, (Z)-11-hexadecenoate. Total native palmitate and stearate (both pheromone intermediates) showed a significant decrease after PBAN injection, before their titers were later restored to initial levels. In contrast, the acyl-CoA thioesters of these two saturated fatty acids increased during the period when their total titers decreased. When a mixture of labeled palmitic and heptadecanoic (an acid that cannot be converted to pheromone) acids was applied to the gland, PBAN-injected females produced greater quantities of labeled pheromone and precursor than did saline-injected ones. The two acids showed similar time-course patterns, with no difference in total titers of each of the respective acids between saline- and PBAN-injected females. When labeled heptadecanoic acid was applied to the gland alone, there was no difference in titers of either total heptadecanoate or of heptadecanoyl-CoA between PBAN- and saline-injected females, suggesting that PBAN does not directly control the storage or liberation of fatty acids in the gland, at least for this fatty acid. Overall, these data indicate that PBAN also controls a later step involved in pheromone biosynthesis, perhaps the reduction of acyl-CoA moieties.
