Probabilistic risk assessment of reproductive effects of polychlorinated biphenyls on bottlenose dolphins (Tursiops truncatus) from the Southeast United States coast

Abstract High levels of polychlorinated biphenyls (PCBs) have been reported in the tissues of some species of marine mammals. The high concentrations are of concern because a growing body of experimental evidence links PCBs to deleterious effects on reproduction, endocrine homeostasis, and immune system function. Much of the recent research has focused on determining the exposure of marine mammal populations to PCBs, but very little effort has been devoted to the actual risk assessments that are needed to determine the expected impacts of the documented exposures. We describe a novel risk assessment approach that integrates measured tissue concentrations of PCBs with a surrogate dose-response relationship and leads to predictions of health risks for marine mammals as well as to the uncertainties associated with these predictions. Specifically, we use PCB tissue residue data from three populations of bottlenose dolphins (Tursiops truncatus), study the feasibility of published dose-response data from a surrogate species, and combine this information to estimate the risk of detrimental reproductive effects in female dolphins. Our risk analyses for dolphin populations near Beaufort (NC, USA), Sarasota (FL, USA), and Matagorda Bay (TX, USA) indicate a high likelihood that reproductive success, primarily in primiparous females, is being severely impaired by chronic exposure to PCBs. Excess risk of reproductive failure, measured in terms of stillbirth or neonatal mortality, for primiparous females was estimated as 60% (Beaufort), 79% (Sarasota), and 78% (Matagorda Bay). Females of higher parity, which have previously off-loaded a majority of their PCB burden, exhibit a much lower risk. [source]


A Method for Predicting Chloride Concentrations in Leachate at Natural Attenuation Landfills in the Precambrian Shield Regions of Ontario, Canada

Natural attenuation landfill sites continue to be the preferred method of domestic waste disposal in the Precambrian Shield regions of Ontario due to economic factors. The main challenge in siting these landfills is ensuring that there will be no adverse impact on off-site water resources. Impact risk assessments are generally based on estimated volumes and strengths of chloride in the leachate. While volumes can be estimated using simple water balances, peak chloride concentration predictions are based on judgment and are quite variable. Since design chloride strengths dictate the size of the required attenuation zone, overestimating concentrations will typically make it impossible to find a suitable site, while underestimating concentrations increases the potential for adverse off-site impacts occurring. Hydrogeological data from active and closed landfills in the Precambrian Shield region were collected to help develop a reliable method of predicting peak chloride concentrations in leachate. This study focused on 21 sites located on relatively permeable sandy soils since landfills underlain by low permeability clayey soils retain leachate similar to lined facilities. Linear regression analyses were conducted to determine if source chloride concentrations at the "sand" sites are significantly influenced by waste thickness, fill area, waste volume, waste deposition rate, hydraulic conductivity, upgradient flow length, depth to the water table, and moisture surplus. A strong relationship (R = 0.957) was found to exist between source chloride concentrations and waste volume. This empirical volume versus chloride regression equation can be used as the basis for establishing design chloride concentrations at new natural attenuation landfills developed over sandy soils in the Precambrian Shield regions of Ontario. An alternative risk assessment approach is required for sites developed over clay soils. [source]

Abstract Although it generally is agreed on in the scientific community that an evaluation of sediment quality solely based on total contaminant levels should be regarded only as a 1st tier or 1 LOE in a WOE framework, not all regulatory frameworks follow this approach. As will be discussed later, dredged material disposal in Venice Lagoon currently is regulated based upon the use of regionally developed SQGs as strict pass/fail criteria. The implications of this policy upon the management of the lagoon are under investigation, but studies on contaminant mobility and bioavailability and on sediment-related effects on biota and exposure (bioassays, biomarkers, bioaccumulation, biomagnification) having the characteristics (duration, number of sites and samples, frequency, number of chemicals and endpoints, etc.) to properly support (e.g., preliminary and/or detailed quantitative risk assessment) decisions on a basin scale have been carried out only recently in the Venice Lagoon (Thetis 2003; Losso et al. 2004; Carrer et al. 2005; Thetis 2005c). Cytotoxic Bacillus spp. belonging to the B. cereus and B. subtilis groups in Norwegian surface waters

Abstract Aims:, To investigate the presence and numbers of Bacillus spp. spores in surface waters and examine isolates belonging to the B. cereus and B. subtilis groups for cytotoxicity, and to discuss the presence of cytotoxic Bacillus spp. in surface water as hazard identification in a risk assessment approach in the food industry. Methods and Results:, Samples from eight different rivers with variable degree of faecal pollution, and two drinking water sources, were heat shocked and examined for the presence of Bacillus spp. spores using membrane filtration followed by cultivation on bovine blood agar plates. Bacillus spp. was present in all samples. The numbers varied from 15 to 1400 CFU 100 ml,1. Pure cultures of 86 Bacillus spp. isolates representing all sampling sites were characterized using colony morphology, atmospheric requirements, spore and sporangium morphology, and API 50 CHB and API 20E. Bacillus spp. representing the B. cereus and B. subtilis groups were isolated from all samples. Twenty-one isolates belonging to the B. cereus and B. subtilis groups, representing eight samples, were screened for cytotoxicity. Nine strains of B. cereus and five strains belonging to the B. subtilis group were cytotoxic. Conclusions:, The presence of cytotoxic Bacillus spp. in surface water represents a possible source for food contamination. Filtration and chlorination of surface water, the most common drinking water treatment in Norway, do not remove Bacillus spores efficiently. This was confirmed by isolation of spores from tap water samples. Significance and Impact of the Study:, Contamination of food with water containing low numbers of Bacillus spores implies a risk for bacterial growth in foods. Consequently, high numbers of Bacillus spp. may occur after growth in some products. High numbers of cytotoxic Bacillus spp. in foods may represent a risk for food poisoning. [source]


Assessing violence risk in Tarasoff situations: a fact-based model of inquiry,

Although significant advances in risk assessment research and practice have been made in recent years, there has not been any analysis in the professional literature regarding how and whether the emerging practice recommendations apply in Tarasoff -type situations. We suggest that, when faced with a Tarasoff -type situation, the appraisal of risk should be guided by a method that is primarily fact-based and deductive, rather than by the more inductive risk assessment approach for general violence recidivism, which is guided primarily by base rates and historical risk factors. We review the principles underlying a fact-based, or threat assessment, approach and outline six areas of inquiry that can guide the appraisal of risk: A,attitudes that support or facilitate violence, C,capacity, T,thresholds crossed, I,intent, O,other's reactions, and N,noncompliance with risk reduction interventions.
