    Ultrastructure of the Male Reproductive System of Cotesia vestalis (Hymenoptera: Braconidae) with Preliminary Characterization of the Secretions

    Abstract The morphology and ultrastructure of testes and accessory glands along with the characterization of their secretions were investigated for a braconid species Cotesia vestalis (Hymenoptera: Braconidae) using light and electron microscopes. The male internal reproductive system of this species is distinguished by a pair of testes, one vas deferens, and a pair of male accessory glands. The testes are separate, and the accessory glands are oval and not fused. It was observed that the secretory cells of testes have characteristic smooth and rough endoplasmic reticulum, and that the cytoplasm is filled with an array of granule droplets usually of two to three types. The secretory cells in the case of accessory glands are typified by the presence of microvilli on their apical cell surfaces and numerous mitochondria in their cytoplasm. SDS-PAGE profile when performed depicted a similarity in most bands of the secretions from both testes and accessory glands, except for four proteins of which two were present only in testes, while the other two only appeared in accessory glands. Their molecular weights were 117 and 55 kDa for testes and 196 and 30 kDa for accessory glands, respectively. This study gives new insights into the intriguing features of male internal reproductive system and it especially constitutes the first report of its kind about the secretion properties of these organs in C. vestalis.     Macroscopic Features of the Arterial Supply to the Reproductive System of the Male Ostrich (Struthio camelus)

    Summary The macroscopic features of the arterial supply to the reproductive system of the male ostrich was studied in 16 pre-pubertal and eight sexually mature and active birds. The left and right cranial renal arteries arise from the aorta, between the cranial divisions of the kidneys. These vessels supply the cranial divisions of the kidneys, the testes, the epididymides and the cranial segments of the ducti deferentia. Accessory testicular arteries which arise directly from the aorta are present in 45.8% of the specimens. They supply the testes and cranial parts of the ducti deferentia. They are variable in number and origin, and four variants are identified. A cranial ureterodeferential branch originates from the cranial renal artery, supplies the cranial portion of the ductus deferens and ureter, and runs caudally to anastomose with the middle renal artery. The sciatic artery arises laterally from the aorta, just caudal to the acetabulum, and gives rise, ventrally, to a common trunk, the common renal artery, which divides into the middle and caudal renal arteries. The middle renal artery gives rise to the middle ureterodeferential branch which supplies the middle part of the ductus deferens and ureter. A few centimetres caudal to the kidney, the aorta terminates in three branches, namely, the left and right internal iliac arteries and the median caudal artery. The internal iliac artery divides into the lateral caudal artery and the pudendal artery; the latter gives off caudal ureterodeferential branches that supply the caudal segments of the ductus deferens and ureter. In addition, the pudendal artery gives off vessels that supply the cloaca, some of which continue to the base of the phallus, where they form an arterial network. In conclusion, the pattern of the blood supply to the reproductive organs of the male ostrich is, in general, similar to that of the domestic fowl and pigeon, although there are a few highlighted distinctive features. Am J Reprod Immunol 2010 The dynamic changes that characterize the female reproductive system are regulated by hormones. However, local cell-to-cell interactions may mediate responsiveness of tissues to hormonal signals. The corpus luteum (CL) is an excellent model for understanding how immune cells are recruited into tissues and the role played by those cells in regulating tissue homeostasis or demise. Leukocytes are recruited into the CL throughout its lifespan, and leukocyte-derived cytokines have been found in corpora lutea of all species examined. The proinflammatory cytokines inhibit gonadotropin-stimulated steroidogenesis, profoundly stimulate prostaglandin synthesis by luteal cells, and promote apoptosis. However, there is mounting evidence that leukocytes and luteal cells communicate in different ways to maintain homeostasis within the functional CL. Domestic animals have provided important information regarding the presence and role of immune cells in the CL. [source]


    Antiandrogenic effects of dibutyl phthalate and its metabolite, monobutyl phthalate, in rats

    ABSTRACT, Developmental toxicity following administration of dibutyl phthalate (DBF) and its major metabolite, monobutyl phthalate (MBuP), by gavage was determined in Wistar rats. DBF on days 0,8 of pregnancy induced an increase in the incidence of preimplantation loss at 1250 mg/kg and higher and postimplantation loss at 750 mg/kg and higher. MBuP on days 0,8 of pregnancy produced an increase in the incidence of pre-and postimplantation loss at 1000 mg/kg. DBF on days 7,15 of pregnancy caused an increase in the incidence of fetuses with malformations at 750 mg/kg. MBuP on days 7,15 of pregnancy produced an increased incidence of fetuses with malformations at 500 mg/kg and higher. DBF on days 15,17 of pregnancy resulted in a decrease in the anogenital distance (AGD) of male fetuses and increase in the incidence of fetuses with undescended testes at 500 mg/kg and higher. MBuP on days 15,17 of pregnancy caused a decreased male AGD and increased incidence of fetuses with undescended testes at 250 mg/kg and higher. No effect of DBF and MBuP on the AGD was found in female offspring. The spectrum of fetal malformations, dependence of gestational days of treatment on the manifestation of teratogenicity, and alterations in development of the male reproductive system observed after administration of DBF were in good agreement with those observed after administration of MBuP. These findings suggest that MBuP may be responsible for the induction of developmental toxic effects of DBP. The doses that produced a decrease in the AGD and undescended testes in male offspring were lower than those producing maternal toxicity, fetal malformations after administration during major organogenesis, and embryonic loss. The male reproductive system may be more susceptible than other organ systems to DBP and MBuP toxicity after maternal exposure. [source]


    Gross anatomy of the musculature and a new description of the reproductive system of Tanaisia bragai and Tanaisia inopina (Trematoda: Eucotylidae) analysed by confocal laser scanning microscopy

The goal of this work was to describe the morphology of the reproductive system and the gross anatomy of the musculature of adults of the species Tanaisia bragai and Tanaisia inopina by means of confocal scanning laser microscopy. The helminths were found parasitizing the kidney collection ducts of Columba livia, in the municipality of Juiz de Fora, Brazil. These helminths were stained with Mayer's carmalum, mounted on permanent slides and observed through a confocal scanning laser microscope. The tomographic images showed the morphology of the organs and glands of the reproductive system, along with the general morphology of the musculature of the body wall, oral sucker, acetabulum, pharynx, intestinal caeca, oesophagus and reproductive system ducts. The present work is the first re-characterization of the reproductive tract of T. bragai and T. inopina. We also present the first description of the general morphology of the gametes and cells of the glands associated with the reproductive apparatus of these species. [source]


    Perspectives of multi-modal contribution of honeybee resources to our life

    Abstract The European honeybee, Apis mellifera, has been introduced to all continents and their products like honey, propolis, royal jelly and beeswax are well known. However, its contribution is not restricted to such direct products but extends into a much wider area. For example, the economic value of seed production by pollination exceeds the above-mentioned bee products. The application of F1 hybrid is increased to as much as 70% of commercial crops and flowers in Japan and honeybees are important pollinators in the F1 seed production. Incorporation into the large-scale biodiesel fuel production system by culturing rape and sunflower seeds etc. is relied on because it is good to construct possible zero-emission systems that reduce carbon dioxide and increase the rich by-products like honey and royal jelly. Bees' higher brain function and sophisticated social system of the colony opens new perspectives as a model system. Their individual ability to recognize even abstract concepts is comparable to that of higher primates. Rats or mice have no such ability. High performance learning ability of bees associated with proboscis extension reflex can be used to detect drugs at the airport. Function of the colony, on the other hand, is an excellent model for social physiology or a self-organization system. After the whole genome of A. mellifera was read in 2006 by the world consortium, consisting of more than 90 institutions from all over the world, many molecular biologists are coming into bee world. Nobody has yet succeeded in the challenge to make transgenic honeybee, so far, because of the difficulty in controlling the reproductive system headed by the queen. However, if someone succeeded in a breakthrough we will have stingless honeybees and a disease-resistant strain in the future. [source]


    Female reproductive biology of Platygaster diplosisae (Hymenoptera: Platygastridae) and Aprostocetus procerae (Hymenoptera: Eulophidae), two parasitoids associated with the African Rice Gall Midge, Orseolia oryzivora (Diptera: Cecidomyiidae)

    Abstract We investigated the female reproductive system of Platygaster diplosisae (Hymenoptera: Platygastridae) and Aprostocetus procerae (= Tetrastichus pachydiplosisae) (Hymenoptera: Eulophidae), two parasitoids associated with the African rice gall midge, Orseolia oryzivora (Diptera: Cecidomyiidae). Both optical and electron microscopy were used. The female reproductive system of P. diplosisae includes two large ovaries of the meristic polytrophic-type, each composed of several tens of ovarioles. The system includes also a venomous gland that extends to a common oviduct. This gland had a filiform secretory portion, in which the epithelium was thin and surrounded a common evacuation canal. The secretory cells secrete into a large reservoir. Parasitism due to P. diplosisae is gregarious. The female reproductive system of A. procerae includes two ovaries of the meristic polytrophic-type, and each ovary has a few ovarioles. Each ovariole includes one or two oocytes, which can be seen in the vitellarium. Two accessory glands, which extend to the oviduct, are also visible. The secretory epithelium of the accessory gland is made up of a dense network of secretory cells surrounded by muscle fibers. Females of A. procerae pierce the tissues of the gall and probably deposit one egg on or close to the pupa of the midge. Aprostocetus procerae is a solitary parasitoid of the midge. The two parasitoids exploit the same host at different developmental stages. These findings improve our knowledge of the reproductive biology of these two parasitoids associated with the African rice gall midge, an important pest in Africa. [source]


    Fireflies with or without prespermatophores: Evolutionary origins and life-history consequences

    Abstract During mating, some male North American fireflies produce spermatophores from prespermatophores in their paired reproductive accessory glands. Other species of fireflies have neither prespermatophores nor spermatophores. To establish a pattern of spermatophore occurrence across firefly species, we examined the male internal reproductive system in 20 Japanese species belonging to 10 genera for the presence or absence of prespermatophores. Twelve species from seven genera produced prespermatophores, while eight species from three genera did not. Superimposed on a molecular phylogeny of Japanese fireflies based on mitochondrial 16S ribosomal DNA sequences, the basal group was prespermatophore producers. Prespermatophores appear to have been lost in two different lineages. Species without prespermatophores are characterized by degeneration of both the forewings and hindwings, and by body gigantism in females. [source]


    Microcystin extracts induce ultrastructural damage and biochemical disturbance in male rabbit testis

    Abstract In the present research, the changes of ultrastructures and biochemical index in rabbit testis were examined after i.p. injection with 12.5 ,g/kg microcystin (MC) extracts. Ultrastructural observation showed widened intercellular junction, distention of mitochondria, endoplasmic reticulum, and Golgi apparatus. All these changes appeared at 1, 3, and 12 h, but recovered finally. In biochemical analyses, the levels of lipid peroxidation (MDA) and H2O2 increased significantly at 1 h, indicating MC-caused oxidative stress. Finally, H2O2 decreased to the normal levels, while MDA remained at high levels. The antioxidative enzymes (CAT, SOD, GPx, GST) and antioxidants (GSH) also increased rapidly at 1 h, demonstrating a quick response of the defense systems to the oxidative stress. Finally, the activity of CAT, SOD, and GPX recovered to the normal level, while the activity of GST and the concentration of GSH remained at a high level. This suggests that the importance of MCs detoxification by GST via GSH, and the testis of rabbit contained abundant GSH. The final recovery of ultrastructure and some biochemical indexes indicates that the defense systems finally succeeded in protecting the testis against oxidative damage. In conclusion, these results indicate that the MCs are toxic to the male rabbit reproductive system and the mechanism underlying this toxicity might to be the oxidative stress caused by MCs. Although the negative effects of MCs can be overcome by the antioxidant system of testis in this study, the potential reproductive risks of MCs should not be neglected because of their wide occurrence. © 2009 Wiley Periodicals, Inc.     Evolution and human tissue expression of the Cres/Testatin subgroup genes, a reproductive tissue specific subgroup of the type 2 cystatins

    SUMMARY The cystatin family comprises a group of generally broadly expressed protease inhibitors. The Cres/Testatin subgroup (CTES) genes within the type 2 cystatins differs from the classical type 2 cystatins in having a strikingly reproductive tissue-specific expression, and putative functions in reproduction have therefore been discussed. We have performed evolutionary studies of the CTES genes based on gene searches in genomes from 11 species. Ancestors of the cystatin family can be traced back to plants. We have localized the evolutionary origin of the CTES genes to the split of marsupial and placental mammals. A model for the evolution of these genes illustrates that they constitute a dynamic group of genes, which has undergone several gene expansions and we find indications of a high degree of positive selection, in striking contrast to what is seen for the classical cystatin C. We show with phylogenetic relations that the CTES genes are clustered into three original groups, a testatin, a Cres, and a CstL1 group. We have further characterized the expression patterns of all human members of the subfamily. Of a total of nine identified human genes, four express putative functional transcripts with a predominant expression in the male reproductive system. Our results are compatible with a function of this gene family in reproduction. [source]


    An evolutionary concept for altered steroid hormone metabolism in patients with rheumatoid arthritis

    The pathogenesis of chronic disabling inflammatory diseases (CDIDs) is partly understood. The presently used concepts focus mainly on abnormalities of the immune system but this view is incomplete. The presented concept is a new framework for the pathogenesis of CDIDs. It integrates evolutionary theories with the classical immunological standpoint, which is further linked with a neuroendocrine immune view of erroneous homeostatic adaptation of the other supersystems (nervous system, endocrine system, reproductive system): 1. In CDIDs, the loss of tolerance against self and harmless foreign antigens leads to continuous immune aggression which is dependent on a multifactorial genetically polymorph background (the initiation). 2. However, advantageous or disadvantageous adaptation to CDIDs were not evolutionary conserved because CDIDs severely impaired reproduction or appeared after the reproductive phase and, thus, imply a strong negative selection pressure. 3. Reactions of all supersystems are evolutionary conserved for transient inflammatory reactions such as the elimination of infectious agents, wound healing, foreign body reaction and many others. 4. The sum of the false reactions of all supersystems , conserved for transient inflammation , provide the pathogenetic background for the chronification of CDIDs because a continuous aggressive situation is created (the chronification). The human disease of rheumatoid arthritis is used as a prototypic CDID to illustrate the integrated view point. The synovial tissue innervation is in the focus of this concept. [source]


    The expression of glutathione reductase in the male reproductive system of rats supports the enzymatic basis of glutathione function in spermatogenesis

    Glutathione reductase (GR) recycles oxidized glutathione (GSSG) by converting it to the reduced form (GSH) using an NADPH as the electron source. The function of GR in the male genital tract of the rat was examined by measuring its enzymatic activity and examining the gene expression and localization of the protein. Levels of GR activity, the protein, and the corresponding mRNA were the highest in epididymis among testes, vas deferens, seminal vesicle, and prostate gland. The localization of GR, as evidenced by immunohistochemical techniques, reveals that it exists at high levels in the epithelia of the genital tract. In testis, GR is mainly localized in Sertoli cells. The enzymatic activity and protein expression of GR in primary cultured testicular cells confirmed its predominant expression in Sertoli cells. Intracellular GSH levels, expressed as mol per mg protein, was higher in spermatogenic cells than in Sertoli cells. As a result of these findings, the effects of buthionine sulfoximine (BSO), an inhibitor for GSH synthesis, and 1,3-bis(2-chlorethyl)-1-nitrosourea (BCNU), an inhibitor for GR, on cultured testicular cells were examined. Sertoli cells were prone to die as the result of BCNU, but not BSO treatment, although intracellular levels of GSH declined more severely with BSO treatment. Spermatogenic cells were less sensitive to these agents than Sertoli cells, which indicates that the contribution of these enzymes is less significant in spermatogenic cells. The results herein suggest that the GR system in Sertoli cells is involved in the supplementation of GSH to spermatogenic cells in which high levels of cysteine are required for protamine synthesis. In turn, the genital tract, the epithelia of which are rich in GR, functions in an antioxidative manner to protect sulfhydryl groups and unsaturated fatty acids in spermatozoa from oxidation during the maturation process and storage.     Sex ratio, reproductive mode and genetic diversity in Triops cancriformis

    Summary 1. Aquatic invertebrates display a wide array of alternative reproductive modes from apomixis to hermaphroditism and cyclical parthenogenesis. These have important effects on genetic diversity and population structure. Populations of the ,living fossil'Triops cancriformis display a range of sex ratios, and various reproductive modes are thought to underlie this variation. Using sex ratio information and histological analyses European populations have been inferred to be gonochoric (with separate males and females), selfing hermaphroditic and androdioecious, a rare reproductive mode in which selfing hermaphrodites coexist with variable proportions of males. In addition, some populations have been described as meiotic parthenogens. 2. Here we use population genetic analysis using microsatellite loci in populations with a range of sex ratios including a gonochoric population, and marker segregation patterns in heterozygote individuals reared in isolation, to clarify the reproductive mode in this species. 3. Our data show that populations in general have very low levels of genetic diversity. Non-gonochoric populations show lower genetic diversity, more heterozygote deficiencies, higher inbreeding coefficients and stronger linkage disequilibria than the gonochoric population. The maintenance of some heterozygosity in populations is consistent with some male influence in T. cancriformis populations, as would be expected from an androdioecious reproductive system. Results of marker segregation in eggs produced in isolation from non-gonochoric populations indicate that meiosis occurs and are consistent with two reproductive modes: selfing hermaphroditism and a type of ameiotic parthenogenesis. 4. Overall, our data indicate that androdioecy and selfing hermaphroditism are the most likely reproductive modes of non-gonochoric European Triops populations. Triops populations are strongly structured, suggesting high genetic drift and low levels of gene flow. [source]

    The distribution, metabolism and function of creatine in the male mammalian reproductive tract: a review

    Creatine is widely distributed throughout the male reproductive system in several mammalian species, and proteins involved in its metabolism and transport have been reported in a number of cell and tissue types. Creatine is synthesized within some organs, incorporating nitrogen from amino acid metabolism. Although creatine metabolism is obligatory for the motility of sea urchin spermatozoa, this does not appear to be the case for mammals. The possible functions of creatine within the reproductive tract are discussed. [source]


    Effect of systemic hormonal cyclicity on skin

    Fluctuations in estrogen and progesterone during the menstrual cycle can cause changes in body systems other than the reproductive system. We conducted several studies to determine a possible correlation between phases of the menstrual cycle and specific skin properties. Healthy Caucasian women (ages 21,48), who had a typical 26,29 day menstrual cycle, participated in the studies. Measurements of skin barrier strength, dryness, response to lactic acid stinging, skin surface lipids, and microflora were obtained every week for 2 to 3 months. Ultraviolet B (UV-B) susceptibility in terms of minimal erythemal dose was also studied. The skin barrier was the weakest between days 22 and 26 of the cycle. Elevated neuronal response (lactic acid sting) was not observed to vary much with the cycle. Skin was driest between day 1 and day 6, while skin surface lipid secretion appeared to be highest on days 16,20 of the hormonal cycle. The highest microbial count was around days 16,22, and there was a high UV-B susceptibility between days 20 and 28 of the menstrual cycle. [source]


    Anatomy and ultrastructure of the reproductive organs in Dactylopodola typhle (Gastrotricha: Macrodasyida) and their possible functions in sperm transfer

    Abstract. The reproductive anatomy of gastrotrichs is well known for several species, especially for the marine taxon Macrodasyida. However, there is little information on the reproductive organs and the modes of mating and sperm transfer in putative basal taxa, which is necessary for accurate reconstruction of the ground pattern of the Gastrotricha. We present the first detailed morphological investigation of the reproductive system of a putative basal gastrotrich, Dactylopodola typhle, using transmission and scanning electron microscopy, histology, and microscopic observations of living specimens. Dactylopodola typhle is a hermaphrodite that possesses paired female and male gonads, an unpaired uterus with an outlet channel that we call the cervix, and an additional accessory reproductive organ, the so-called caudal organ. We hypothesize that the hollow, secretory caudal organ serves for picking up autospermatozoa (self-sperm), for spermatophore formation, and finally for transferring the autospermatophore to a mating partner. The allospermatophore (foreign spermatophore) is stored within the uterus where fertilization occurs. We think that the mature and fertilized egg is released through the cervix and the dorsolateral female gonopore, and not by rupture of the body wall. Based on the morphology, we provide a plausible hypothesis for spermatophore formation and transfer in D. typhle. Preliminary phylogenetic considerations indicate that the stem species of Macrodasyida, perhaps that of all Gastrotricha, had paired ovaries and paired testes, an unpaired uterus, and only one accessory reproductive organ.     A histochemical study of the reproductive structures in the flatworm Dugesia leporii (Platyhelminthes, Tricladida)

    Abstract. The functional morphology and the topographic distribution of tissues in the reproductive system of specimens of Dugesia leporii, an endemic Sardinian free-living planarian, are investigated. Data are provided on the nature of epithelial and glandular secretions, spermatophores, and cocoons by histochemistry, light microscopy, and scanning electron microscopy. All secreting epithelial cells produce strongly acidic sulfated glycoproteins. Glandular cells secrete strongly acidic sulfated glycoproteins or keratohyalin-like material in the penis bulb, and prekeratin-like material in atrial glands. Secretions of the bursa copulatrix may be involved in the activation of sperm while material produced by the bursa canal and oviducts probably serves to propel spermatophores or sperm and eggs. Mucous secretion of the seminal vesicle may serve to dilute and activate sperm before copulation. The viscous secrete of the ejaculatory duct and vasa deferentia may play a protective role to maintain sperm viability. Materials produced by the penis papilla and atrium probably lubricate the epithelial surface. The bilayered wall of spermatophore made of keratohyalin-like material and strongly acidic sulfated glycoproteins is produced by two gland types of the penis bulb. The bilayered shell of cocoon made of prekeratin-like and keratohyalin-like materials is secreted by both atrial glands and vitelline cells. The cocoon stalk is made of keratohyalin-like material produced by cement glands. Shell glands, producing GAG, are not involved in cocoon formation, but they may be implicated in the dilution and activation of seminal material to favor sperm movement toward the oviducts.     Effect of lindane on CYP-mediated steroid hormone metabolism in male mice following in utero exposure

    Abstract A wide number of pesticides, including highly persistent organochlorinated compounds, such as lindane (LIN), may induce reproductive and developmental alterations by directly binding to the estrogen/androgen receptors or altering steroid hormone metabolism. In the present work, we have investigated whether LIN in utero exposure of CD1 mice affects the reproductive system in male offspring by causing an impairment of the CYP-dependent steroid hormone metabolism. Dam exposure to 25 mg kg,1 b.w. LIN occurred during critical developmental periods, from gestational days 9 to 16. Effects on hepatic CYP-mediated testosterone (TST) hydroxylase, aromatase activities and testicular parameters were tested at postnatal days (PND 50, 65,69, 100) that are critical for sexual maturation in CD1 mice. In the adult F1 mice significant changes of male reproductive endpoints (testis weight, spermatid number) as well as dramatic effects on CYP-mediated TST metabolism were observed on PND 65,69, in the absence of any of systemic toxicity. The levels of TST 6, - and 2, -hydroxylation and dehydrogenation showed the highest level of reduction, suggesting CYP 3A and 2C families as the major target of LIN induced effects. All changes were almost recovered on PND 100. No effects on aromatase activity were evidenced. Overall, these findings provide useful information for a better characterization of the LIN mode of action. They suggest that LIN-induced toxicity in males is linked to an impairment of steroid hormone homeostasis, due to CYP-mediated TST catabolism modulation and differs from LIN receptor-mediated mechanism previously reported in females.     Differential developmental toxicities of di- n -hexyl phthalate and dicyclohexyl phthalate administered orally to rats

    Abstract The objective of this study was to evaluate the developmental toxic potential of di- n -hexyl phthalate (DnHP) and dicyclohexyl phthalate (DCHP) in rats. Pregnant Sprague,Dawley rats were exposed to DnHP or DCHP at doses of 0 (olive oil), 250, 500 and 750 mg kg,1 per day, by gavage, on gestational days (GD) 6,20. Maternal food consumption and body weight gain were significantly reduced at 750 mg kg,1 per day of DnHP and at the two high doses of DCHP. Slight changes in liver weight associated with peroxisomal enzyme induction were seen in dams treated with DnHP or DCHP. DnHP caused dose-related developmental toxic effects, including marked embryo mortality at 750 mg kg,1 per day, and presence of malformations (mainly cleft palate, eye defects and axial skeleton abnormalities) and significant decreases in fetal weight at 500 and 750 mg kg,1 per day. Significant delay of ossification and increase in the incidence of skeletal variants (e.g. supernumerary lumbar ribs) also appeared at 250 mg kg,1 per day. DCHP produced fetal growth retardation at 750 mg kg,1 per day, as evidenced by significant reduction of fetal weight. DnHP and DCHP induced a significant and dose-related decrease in the anogenital distance of male fetuses at all doses, and there was a significant increase in the incidence of male fetuses with undescended testis at 500 and 750 mg kg,1 per day of DnHP. In conclusion, DnHP showed clear embryolethality and teratogenicity, but not DCHP. There was evidence that both phthalates could alter the development of the male reproductive system after in utero exposure, DnHP being much more potent than DCHP.     Argemone oil induced cellular damage in the reproductive tissues of transgenic Drosophila melanogaster: Protective role of 70 kDa heat shock protein

    Abstract We explored the reproductive toxicity of argemone oil and its principal alkaloid fraction in transgenic Drosophila melanogaster (hsp70-lacZ) Bg9. The toxicity of argemone oil has been attributed to two of its physiologically active benzophenanthridine alkaloids, sanguinarine and dihydrosanguinarine. Freshly eclosed first instar larvae of transgenic Drosophila melanogaster were transferred to different concentrations of argemone oil and its alkaloid fraction contaminated food. Virgin flies that eclosed from the contaminated food were pair-mated to look into the effect on reproduction. The study was further extended by investigating hsp70 expression and tissue damage in larval gonads, genital discs, and reproductive organs of adult fly. Our results showed that argemone oil was more cytotoxic than its principal alkaloid fraction. Moreover, it was the male fly that was more affected compared to its opposite number. The accessory glands of male reproductive system of the fly, which did not express hsp70, exhibited severe damage as evidenced by Trypan blue staining. This prompted us to explore the ultrastructural morphology of the gland, which showed acute signs of necrosis in both the cell types as evident by necrotic nuclei, higher vacuolization, and disorganized endoplasmic reticulum, decrease in the number of Golgi vesicles and disorganized, loosely packed filamentous structures in the lumen of the accessory gland, at the higher concentrations of the adulterant. The study showed the reproductive toxicity of argemone oil and its alkaloid fraction in transgenic Drosophila melanogaster and further confirmed the cytoprotective role of hsp70. © 2003 Wiley Periodicals, Inc.     Novel identification of peripheral dopaminergic D2 receptor in male germ cells,

    Abstract Dopamine is a recognized modulator in the central nervous system (CNS) and peripheral organ functions. The presence of peripheral dopamine receptors outside the CNS has suggested an intriguing interaction between the nervous system and other functional systems, such as the reproductive system. In the present study we analyzed the expression of D2R receptors in rat testis, rat spermatogenic cells and spermatozoa, in different mammals. The RT-PCR analysis of rat testis mRNA showed specific bands corresponding to the two dopamine receptor D2R (L and S) isoforms previously described in the brain. Using Western blot analysis, we confirmed that the protein is present in rat testis, isolated spermatogenic cells and also in spermatozoa of a range of different mammals, such as rat, mouse, bull, and human. The immunohistochemistry analysis of rat adult testis showed that the receptor was expressed in all germ cells (pre- and post-meiotic phase) of the tubule with staining predominant in spermatogonia. Confocal analysis by indirect immunofluorescence revealed that in non-capacitated spermatozoa of rat, mouse, bull, and human, D2R is mainly localized in the flagellum, and is also observed in the acrosomal region of the sperm head (except in human spermatozoa). Our findings demonstrate that the two D2 receptor isoforms are expressed in rat testis and that the receptor protein is present in different mammalian spermatozoa. The presence of D2R receptors in male germ cells implies new and unsuspected roles for dopamine signaling in testicular and sperm physiology. J. Cell.     Reproduction and Resistance to Stress: When and How

    Abstract Environmental and social stresses have deleterious effects on reproductive function in vertebrates. Global climate change, human disturbance and endocrine disruption from pollutants are increasingly likely to pose additional stresses that could have a major impact on human society. Nonetheless, some populations of vertebrates (from fish to mammals) are able to temporarily resist environmental and social stresses, and breed successfully. A classical trade-off of reproductive success for potential survival is involved. We define five examples. (i) Aged individuals with minimal future reproductive success that should attempt to breed despite potential acute stressors. (ii) Seasonal breeders when time for actual breeding is so short that acute stress should be resisted in favour of reproductive success. (iii) If both members of a breeding pair provide parental care, then loss of a mate should be compensated for by the remaining individual. (iv) Semelparous species in which there is only one breeding period followed by programmed death. (v) Species where, because of the transience of dominance status in a social group, individuals may only have a short window of opportunity for mating. We suggest four mechanisms underlying resistance of the gonadal axis to stress. (i) Blockade at the central nervous system level, i.e. an individual no longer perceives the perturbation as stressful. (ii) Blockade at the level of the hypothalamic-pituitary-adrenal axis (i.e. failure to increase secretion of glucocorticosteroids). (iii) Blockade at the level of the hypothalamic-pituitary-gonad axis (i.e. resistance of the reproductive system to the actions of glucocorticosteroids). (iv) Compensatory stimulation of the gonadal axis to counteract inhibitory glucocorticosteroid actions. Although these mechanisms are likely genetically determined, their expression may depend upon a complex interaction with environmental factors. Future research will provide valuable information on the biology of stress and how organisms cope. Such mechanisms would be particularly insightful as the spectre of global change continues to unfold. [source]


    Increased Galanin Synapses onto Activated Gonadotropin-Releasing Hormone Neuronal Cell Bodies in Normal Female Mice and in Functional Preoptic Area Grafts in Hypogonadal Mice

    Abstract Galanin synaptic input onto gonadotropin-releasing hormone (GnRH) neuronal cell bodies was analysed in female mice using the presynaptic vesicle-specific protein, synaptophysin (Syn) as a marker. In the first experiment, forebrain sections from normal ovariectomized ovarian steroid-primed mice exhibiting a surge of luteinizing hormone were processed for immunohistochemical labelling for GnRH, synaptophysin, galanin and Fos. Two representative sections from each brain, one passing through the anterior septum (anterior section) and the other through the organum vasculosum lamina terminalis-preoptic area (posterior section), were analysed under the confocal microscope. None of the GnRH cells analysed in the anterior sections were Fos immunoreactive (IR) or received input from galanin-IR fibres. In contrast, the majority of GnRH cells in the posterior sections analysed were Fos-positive. The number of galanin synapses onto the Fos-positive GnRH cells was significantly higher than that in the Fos-negative cells in this area of the brain, even though the number of Syn-IR appositions was comparable to each other. Transplantation of preoptic area (POA) into the third cerebral ventricle of hypogonadal (HPG) mice corrects deficits in the reproductive system. In the second experiment, synaptic input to GnRH cells was compared between HPG/POA mice with (functional graft) or without (nonfunctional graft) gonadal development. The mean numbers of Syn-IR appositions and galanin synapses per GnRH cell and the proportion of GnRH cells with galanin input were significantly higher in the functional than in the nonfunctional grafts. The results suggest that galanin can act directly on the GnRH cell bodies and may have an important regulatory role on the GnRH system. [source]


    Actions and Interactions of Alcohol and Insulin-Like Growth Factor-1 on Female Pubertal Development

    Alcohol (ALC) is a drug that is capable of disrupting reproductive function in adolescent humans, as well as immature rhesus monkeys and rats. Critical to determining the mechanism(s) of the effects of ALC on the pubertal process is to have a better understanding of the important events involved in the initiation of puberty. For years it has been hypothesized that there may be metabolic signals capable of linking somatic growth to the activation of the reproductive system at the time of puberty. In recent years it has been shown that insulin-like growth factor-1 (IGF-1) is one such signal that plays an early role in the pubertal process. In this review, we will describe the actions and interactions of ALC and IGF-1 on molecular and physiological processes associated with pubertal development. [source]


    The odour of pyrazine increases the egg mass of domestic chickens (Gallus gallus domesticus L.)

    Abstract Forty leghorn chickens at the commencement of egg laying, were divided into two groups, each with 10 females and 10 males. One group was exposed to the odour of synthetic pyrazine (2-methoxy-3-isobutylpyrazine) for 16 weeks while the other acted as a control. During the first 4 weeks the hens exposed to pyrazine odour laid significantly fewer eggs (unfertilized) than the controls, but thereafter both groups laid a similar number. Mean egg mass of the pyrazine exposed hens was significantly (P= 0.012) greater (5.5%) than that of the control group. No significant difference was found in body mass or eggshell thickness. Similarly, there were no consistent significant differences between groups in oestrogen and testosterone concentrations in the blood of females and males, respectively. No pyrazine could be detected in cloacal extracts. The experiment shows that an external odour can affect the internal reproductive system of the chicken. It is suggested that the pyrazine-engendered increase in egg mass involves neuroendocrine regulation within the hypothalamus rather than hormonal interactions ,downstream' of the brain. [source]


    Ultrastructure of the Male Reproductive System of Cotesia vestalis (Hymenoptera: Braconidae) with Preliminary Characterization of the Secretions

    Abstract The morphology and ultrastructure of testes and accessory glands along with the characterization of their secretions were investigated for a braconid species Cotesia vestalis (Hymenoptera: Braconidae) using light and electron microscopes. The male internal reproductive system of this species is distinguished by a pair of testes, one vas deferens, and a pair of male accessory glands. The testes are separate, and the accessory glands are oval and not fused. It was observed that the secretory cells of testes have characteristic smooth and rough endoplasmic reticulum, and that the cytoplasm is filled with an array of granule droplets usually of two to three types. The secretory cells in the case of accessory glands are typified by the presence of microvilli on their apical cell surfaces and numerous mitochondria in their cytoplasm. SDS-PAGE profile when performed depicted a similarity in most bands of the secretions from both testes and accessory glands, except for four proteins of which two were present only in testes, while the other two only appeared in accessory glands. Their molecular weights were 117 and 55 kDa for testes and 196 and 30 kDa for accessory glands, respectively. This study gives new insights into the intriguing features of male internal reproductive system and it especially constitutes the first report of its kind about the secretion properties of these organs in C. vestalis.     Population structure and intraspecific aggression in the invasive ant species Anoplolepis gracilipes in Malaysian Borneo

    Abstract Invasive species are one of the main sources of the ongoing global loss of biodiversity. Invasive ants are known as particularly damaging invaders and their introductions are often accompanied by population-level behavioural and genetic changes that may contribute to their success. Anoplolepis gracilipes is an invasive ant that has just recently received increased attention due to its negative impact on native ecosystems. We examined the behaviour and population structure of A. gracilipes in Sabah, Malaysia. A total of 475 individuals from 24 colonies were genotyped with eight microsatellite markers. Intracolonial relatedness was high, ranging from 0.37 to 1 (mean ± SD: 0.82 ± 0.04), while intercolonial relatedness was low (0.0 ± 0.02, range ,0.5,0.76). We compared five distinct sampling regions in Sabah and Brunei. A three-level hierarchical F-analysis revealed high genetic differentiation among colonies within the same region, but low genetic differentiation within colonies or across regions. Overall levels of heterozygosity were unusually high (mean HO = 0.95, mean HE = 0.71) with two loci being entirely heterozygous, indicating an unusual reproductive system in this species. Bioassays revealed a negative correlation between relatedness and aggression, suggesting kinship as one factor facilitating supercolony formation in this species. Furthermore, we genotyped one individual per nest from Sabah (22 nests), Sarawak (one nest), Brunei (three nests) and the Philippines (two nests) using two mitochondrial DNA markers. We found six haplotypes, two of which included 82.1% of all sequences. Our study shows that the sampled area in Sabah consists of a mosaic of differently interrelated nests in different stages of colony establishment. While some of the sampled colonies may belong to large supercolonies, others are more likely to represent recently introduced or dispersed propagules that are just beginning to expand. [source]


    Isolation and characterization of microsatellite markers from the Hypoptychus dybowskii (Pisces, Gasterosteiformes)

    Abstract Hypoptychus dybowskii (Gasterosteiformes) exhibits allopaternal care frequently caused by various types of male reproductive tactics (sneaking, egg desertion and taking over). In order to understand this interesting reproductive system, we isolated microsatellites loci from H. dybowskii. Five microsatellites showed 2,10 alleles and expected heterozygosities ranged from 0.15 to 0.84. These were not significant deviations from Hardy,Weinberg expectations. These results suggest that these novel polymorphic loci should be useful for parentage analysis of H. dybowskii. [source]


    Cigarette smoking and aneuploidy in human sperm

    Abstract Cigarette smoke contains chemicals which are capable of inducing aneuploidy in experimental systems. These chemicals have been shown to reach the male reproductive system, increasing oxidative DNA damage in human sperm and lowering semen quality. We have examined the association between smoking and aneuploid sperm by studying 31 Chinese men with similar demographic characteristics and lifestyle factors except for cigarette smoking. None of the men drank alcohol. These men were divided into three groups: nonsmokers (10 men), light smokers (<,20 cigarettes/day, 11 men), and heavy smokers (,,20 cigarettes/day, 10 men). There were no significant differences in semen parameters or in age across groups. Two multi-color fluorescence in situ hybridizations (FISH) were performed: two-color FISH for chromosomes 13 and 21, and three-color FISH for the sex chromosomes using chromosome 1 as an internal autosomal control for diploidy and lack of hybridization. The mean hybridization efficiency was 99.78%. The frequency of disomy 13 was significantly higher in light and heavy smokers than in non-smokers, while no significant differences in the frequency of disomy 21, X or Y were observed across groups. Significant inter-donor heterogeneity in every category of disomic sperm examined was found in both light and heavy smokers, while in nonsmokers only XY disomy showed significant inter-donor differences. Thus, we conclude that cigarette smoking may increase the risk of aneuploidy only for certain chromosomes and that men may have different susceptibilities to aneuploidy in germ cells induced by cigarette smoking.
