Hippocampal long-term depression as an index of spatial working memory

Abstract Long-term potentiation (LTP), a form of synaptic plasticity in the hippocampus, is a cellular model for the neural basis of learning and memory, but few studies have investigated the contribution of long-term depression (LTD), a counterpart of LTP. To address the possible relationship between hippocampal LTD and spatial performance, the spatial cognitive ability of a rat was assessed in a spontaneous alternation test and, thereafter, LTD in response to low-frequency burst stimulation (LFBS) was monitored in the dentate gyrus of the same rat under anaesthesia. To enhance a divergence in the ability for spatial performance, some of the animals received fimbria,fornix (FF) transection 14 days before the experiments. LTD was reliably induced by application of LFBS to the medial perforant path of intact rats, while no apparent LTD was elicited in rats with FF lesions. The behavioural parameters of spatial memory showed a significant correlation with the magnitude of LTD. We found no evidence that the cognitive ability correlated with other electrophysiological parameters, e.g. basal synaptic responses, stimulus intensity to produce half-maximal responses, paired-pulse facilitation or paired-pulse depression. These results suggest that the magnitude of LTD in the dentate gyrus serves as a reliable index of spatial cognitive ability, providing insights into the functional significance of hippocampal LTD. Short Sales Constraints and Momentum in Stock Returns

Abstract: We show that stock characteristics identified by D'Avolio (2002) provide a reliable index of the mostly unobservable short sales constraints. Specifically, we find that this index is positively related to the level of short interest and to short selling costs implied by the disparity in prices in the options and stock markets, and is negatively related to future returns. Using this index, we show that the magnitude of momentum returns for the period 1984 to 2001 is positively related to short sales constraints, and loser stocks rather than winner stocks drive this result. We conclude that short sales constraints are important in preventing arbitrage of momentum in stock returns. [source]


Shared governance as vertical alignment of nursing group power and nurse practice council effectiveness

Aim(s), This study validates an instrument for measuring the effectiveness of nursing practice councils and offers a framework for measuring and understanding shared governance. Background, Empowerment results from the vertical alignment of nursing group power with nursing unit power practices. The field lacks an instrument for measuring nurses' practice of power. Method(s), Two studies (n1 = 119; n2 = 248) are used to validate the Nursing Practice Council effectiveness scale (NPCes). Results, NPCes is a valid and reliable index of nursing practice council effectiveness. This study suggests specific diagnostic tools to understand two levels for actualized power, one at the group or departmental level and one at the unit level. Conclusion(s), NPCes and the Sieloff-King Assessment of Group Power within Organizations (SKAGPO) can be used together to improve examination of shared governance. Examining group power as well as unit-level practices may give a more complete view of barriers to nurse empowerment. Implications for nursing management, Changing nursing power and practices in an organization may be made more effective by engaging and monitoring vertical alignment of strategies fostering power competencies among nurse leaders and simultaneously supporting nursing practice councils as a means of exercising nurse authority at the unit level. THE APPLICATION OF ELECTRON SPIN RESONANCE AS A GUIDE TO THE MATURATION AND TYPING OF ORGANIC MATTER IN THE NORTH SEA

In early electron spin resonance (ESR) analysis of North Sea wells, maturation of organic matter (OM) was expressed in terms of maximum palaeotemperature (MPT) based on North American calibrations that did not consider the influences of kerogen composition or overpressure. In the North Sea, the MPTs were anomalous in overpressured sequences and relative to other indices of OM maturation such as vitrinite reflectance, so the ESR method was abandoned there in geochemical studies. However, early empirical study of North Sea ESR data indicated that, in relation to functions that linked temperature and pore pressure, some ESR parameters were predictable without reference to MPTs. In order to re-evaluate ESR parameters as indices of OM maturation, the physical factors (temperature and pressure) which affect OM maturation are related in the present paper to the ESR parameters "g" (spectral position) and Ng (spin density) at six well locations in the northern North Sea. A third ESR parameter, W (line width), is not an effective guide to maturation levels due to its complex relationship to the physical factors and kerogen types. However, cross-plots of W versus "g" and Ng appear to be as effective as pyrolysis for kerogen typing. Levels of maturation investigated in the North Sea wells range through the equivalent vitrinite reflectance values of about 0.50,1.50%. The values of "g" and Ng have been differentiated for kerogen type, but undifferentiated values of "g" have also been studied. Regression analysis has shown that there are linear relationships between the ESR parameters "g" and Ng, and the physical factors present-day temperature (To), "effective" temperature (Te), and differential pressure (Pd). Correlation coefficients for both "g" (undifferentiated and differentiated) and Ng (differentiated) relative to the physical factors are high; the highest values are for "g" and Ng relative to Te and Pd (r =,0.950 for "g" differentiated or undifferentiated, r = 0.944,0.976 for Ng differentiated, respectively). However, correlation coefficients were lower for "g" and Ng relative to To. More frequent high correlation coefficients and larger sample populations suggest that "g" (undifferentiated) is a more reliable index of OM maturation than Ng(differentiated). However, the estimation of levels of OM maturation is improved if both indices are used together. The ESR method appears to be effective both for estimating levels of OM maturation and for kerogen typing. It has a number of potential advantages over other geochemical methods: firstly, it is more sensitive for estimating OM maturation than most other methods; secondly, it can be used to analyze organic matter which is as old as Proterozoic; thirdly, it does not destroy the samples analyzed. EPR oximetry in the beating heart: Myocardial oxygen consumption rate as an index of postischemic recovery

Abstract Oxygen plays a critical role in the pathophysiology of myocardial injury during both ischemia and subsequent reperfusion (I/R). Thus, oxygen concentration is an important variable to measure during I/R. In the present work, electron paramagnetic resonance (EPR)-based oximetry was used to measure the oxygen concentration during a series of I/R episodes and oxygenation levels were correlated with the contractile and hemodynamic functions of the heart. A custom-developed electronically tunable surface coil resonator working at 1.1 GHz was used to determine tissue pO2 in the beating heart. Microcrystalline particulate of lithium phthalocyanine was used as an EPR oximetry probe. Isolated and perfused rat hearts were subjected to 1 or 3 hr durations of preischemic perfusion, followed by 15-min I/R cycles. In hearts perfused for 3 hr prior to 15-min I/R cycles, the myocardial pO2 decreased gradually on subsequent reperfusions of three successive I/R cycles. However, in hearts perfused for 1 hr there was almost 100% recovery of myocardial pO2 in all three I/R cycles. The extent of oxygenation recovered in each reperfusion cycle correlated with the recovery of hemodynamic and contractile function. The results also showed that the oxygen consumption rate of the heart at the end of each I/R episode decreased in direct proportion to the functional recovery. In summary, it was observed that the amount of myocardial oxygen consumption during I/R could provide a reliable index of functional impairment in the heart. Relationship between serum leptin and thyroid hormones in children

Abstract Background: Because leptin decreases food intake and increases energy expenditure, the possible influence of thyroid status on the leptin system has been investigated mainly in adults and animals. However, the data available at present are very confusing. The aim of the present study was to assess the possible interaction of thyroid hormones with the leptin system. Methods: Serum free thyroxine (FT4), a biologically active thyroid hormone, and thyroid stimulating hormone (TSH), a sensitive and reliable index of thyroid status, were examined in 51 children (19 males, 32 females) with mass screening-detected congenital hypothyroidism on continuous L -thyroxine (L-T4) substitution therapy. The subjects were divided into younger (n=35, aged 1 month , 5 years) and older (n=16, 6 years , 11 years) children groups. Serum levels of leptin and thyroid hormones were measured in the subjects. Body mass index (BMI) was estimated by the formula bodyweight (kg)/height`height (m 2), which is known as the Kaup index in younger children and BMI in older children and adults. Results: In the younger children group, serum leptin levels showed no correlation with serum TSH, FT4 or T4. In the older children group, serum leptin concentrations significantly correlated with T4 (r=0.510, P<0.05) and BMI (n=16, r=0.647, P<0.01), but not with TSH or FT4. Conclusion: The role of thyroid hormones in modulating leptin synthesis and secretion seems to have little, if any, clinical or biological relevance. [source]


Strategic shotgun proteomics approach for efficient construction of an expression map of targeted protein families in hepatoma cell lines

Abstract An expression map of the most abundant proteins in human hepatoma HepG2 cells was established by a combination of complementary shotgun proteomics approaches. Two-dimensional liquid chromatography (LC)-nano electrospray ionization (ESI) tandem mass spectrometry (MS/MS) as well as one-dimensional LC-matrix-assisted laser desorption/ionization MS/MS were evaluated and shown that additional separation introduced at the peptide level was not as efficient as simple prefractionation of protein extracts in extending the range and total number of proteins identified. Direct LC-nanoESI MS/MS analyses of peptides from total solubilized fraction and the excised gel bands from one-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis fractionated insolubilized fraction afforded the best combination in efficient construction of a nonredundant cell map. Compiling data from multiple variations of rapid shotgun proteomics analyses is nonetheless useful to increase sequence coverage and confidence of hits especially for those proteins identified primarily by a single or two peptide matches. While the returned hit score in general reflects the abundance of the respective proteins, it is not a reliable index for differential expression. Using another closely related hepatoma Hep3B as a comparative basis, 16 proteins with more than two-fold difference in expression level as defined by spot intensity in two-dimensional gel electrophoresis analysis were identified which notably include members of the heat shock protein (Hsp) and heterogeneous nuclear ribonucleoprotein (hnRPN) families. The observed higher expression level of hnRNP A2/B1 and Hsp90 in Hep3B led to a search for reported functional roles mediated in concert by both these multifunctional cellular chaperones. In agreement with the proposed model for telomerase and telomere bound proteins in promoting their interactions, data was obtained which demonstrated that the expression proteomics data could be correlated with longer telomeric length in tumorigenic Hep3B. This biological significance constitutes the basis for further delineation of the dynamic interactions and modifications of the two protein families and demonstrated how proteomic and biological investigation could be mutually substantiated in a productive cycle of hypothesis and pattern driven research. Saccade generation and suppression in schizophrenia: Effects of response switching and perseveration

Abstract Poor antisaccade performance is a reliable index of action control deficits in schizophrenia. To further elucidate the underlying cognitive impairments, the current study aimed to confirm effects of switching the response direction on saccadic performance and to investigate whether response switch effects relate to perseveration. Fourteen schizophrenia patients and 14 healthy controls performed sequences of 1 to 3 simple volitional saccades to one direction and a subsequent volitional saccade with distractor to the same or the opposite direction. Response switches increased error rates in schizophrenia if they followed 3 saccades to the opposite side, suggesting that response switching affects performance on conditions of strong persisting response programs. The increase of response switch error rates with multiple repetitions of the prior response points to a relationship between perseveration and response selection. [source]


Estimation of Maximum Ventricular Elastance Under Assistance With a Rotary Blood Pump

Abstract The maximum ventricular elastance is a reliable index for assessing the cardiac function from changes in its pressure-volume relationship. The advantage of this index is that it can represent the contractility of either unassisted hearts or native hearts assisted with rotary blood pumps. However, there are situations in which changes in the ventricular load required for the conventional estimation method might be risky. For example, in a bridge-to-recovery the cardiac function should also be continuously observed after the implantation of a rotary blood pump. In this article, we present the results of the estimation of the maximum elastance with in vivo data using the parameter optimization method, which is a single-beat estimation method. The estimated values for the normal cardiac function (6.8 ± 0.6, 4.5 ± 0.9, 4.2 ± 1.8 mm Hg/mL) were significantly different from those for the low cardiac function (3.2 ± 1.5, 1.9 ± 1.0, 1.9 ± 1.2 mm Hg/mL) from the data of the three animals that were analyzed. Besides, the maximum elastance values were independent of the pump rotational speed. These results indicate that this index might be useful for the detection of the myocardial recovery. [source]


Using a GnRH agonist to obtain an index of testosterone secretory capacity in the cockatiel (Nymphicus hollandicus) and sulphur-crested cockatoo (Cacatua galerita)

Objective Validation of a stimulation test for determining the steroidogenic capacity of the parrot testis. The major aim was to characterise testosterone secretion after injection of a gonadotropin-releasing hormone agonist (GnRHa), then use the test to investigate seasonal reproduction in the male cockatiel. Procedure A synthetic GnRHa (buserelin; 8.0 µg of peptide/kg bodyweight) was injected IM into male cockatiels (n = 7) and sulphur-crested cockatoos (n = 3) and serial blood samples collected at 0, 30, 60, 90 and 120 min after administration. Once validated, the technique was subsequently used to examine seasonal changes (23 months) in the testosterone profile of a captive cockatiel population. Results Injection of buserelin resulted in a significant increase in the testosterone concentration of cockatiel plasma, with maximal concentrations occurring at approximately 60 (1.33 ± 0.08 ng/mL) to 90 min (1.22 ± 0.08 ng/mL) after injection. Although no clear pattern of seasonal variation in testosterone secretion was detected in cockatiel plasma, samples taken 60 and 90 min after administration showed a significant increase in all seasons. Injection of buserelin in the sulphur-crested cockatoo also resulted in increased testosterone secretion, with maximal concentrations obtained after 90 min. Conclusion Buserelin can be used to obtain a reliable index of the prevailing testosterone capacity of the cockatiel and cockatoo testis. With further studies, this test may be incorporated into clinical assessment of reproductive status. [source]


Reliability of knee extension and flexion measurements using the Con-Trex isokinetic dynamometer

Summary The aim of this study was to evaluate the reliability of isokinetic and isometric assessments of the knee extensor and the flexor muscle function using the Con-Trex isokinetic dynamometer. Thirty healthy subjects (15 males, 15 females) were tested and retested 7 days later for maximal strength (isokinetic peak torque, work, power and angle of peak torque as well as isometric maximal voluntary contraction torque and rate of torque development) and fatigue (per cent loss and linear slope of torque and work across a series of 20 contractions). For both the knee extensor and the flexor muscle groups, all strength data , except angle of peak torque , demonstrated moderate-to-high reliability, with intraclass correlation coefficients (ICC) higher than 0·86. The highest reliability was observed for concentric peak torque of the knee extensor muscles (ICC = 0·99). Test,retest reliability of fatigue variables was moderate for the knee extensor (ICC range 0·84,0·89) and insufficient-to-moderate for the knee flexor muscles (ICC range 0·78,0·81). The more reliable index of muscle fatigue was the linear slope of the decline in work output. These findings establish the reliability of isokinetic and isometric measurements using the Con-Trex machine. [source]

A conceptual framework for the quality of equine healthcare, based on court cases against equine practitioners in The Netherlands
Summary Quality control and client satisfaction are topics of considerable current interest in both human and veterinary healthcare. Crucial to this discussion is a proper definition of ,quality of care'. This study applied, to (equine) veterinary care, a conceptual framework designed for assessing the quality of human healthcare and featuring structural deficiency, procedural inadequacy and disappointing outcome as key elements. This was done using court cases against equine practitioners as a yardstick for client dissatisfaction. Applying a system for evaluating the quality of healthcare may be a good way of monitoring and improving (equine) veterinary services, particularly once reliable indices for client satisfaction have been validated. Detecting Technological Catch-Up in Economic Convergence

We address the problem of measuring, in the absence of reliable indices of technology levels, how much of the convergence we observe is due to convergence in technology or in capital,labour ratios. We first develop a growth model where technology accumulation in lagging economies depends on their propensity to innovate and on technological spillovers, with convergence due both to capital-deepening and to technological diffusion. Then we study the transitional dynamics of the model to show how to discriminate empirically between the following three hypotheses: (i) convergence is due to capital-deepening with technology levels uniform across economies, as in Mankiw, Romer and Weil; (ii) convergence is due to capital-deepening with stationary differences in individual technologies, as in Islam; (iii) convergence is due to both technological catch-up and capital-deepening. Our main findings are as follows. First, we show that it may be difficult to distinguish between hypotheses (ii) and (iii) in cross-section or panel data. This problem has often been overlooked in the empirical literature on convergence. Second, we suggest how the problem can be overcome by noting that hypothesis (iii) does (and hypothesis (ii) does not) imply that the initial differences in technology levels may tend to decrease over time. A careful analysis of the fixed-effects estimates obtained by means of panel data methodology proposed in Islam should allow researchers to discriminate between the two competing hypotheses. Electrocortical and electrodermal responses covary as a function of emotional arousal: A single-trial analysis

Abstract Electrophysiological studies of human visual perception typically involve averaging across trials distributed over time during an experimental session. Using an oscillatory presentation, in which affective or neutral pictures were presented for 6 s, flickering on and off at a rate of 10 Hz, the present study examined single trials of steady-state visual evoked potentials. Moving window averaging and subsequent Fourier analysis at the stimulation frequency yielded spectral amplitude measures of electrocortical activity. Cronbach's alpha reached values >.79, across electrodes. Single-trial electrocortical activation was significantly related to the size of the skin conductance response recorded during affective picture viewing. These results suggest that individual trials of steady-state potentials may yield reliable indices of electrocortical activity in visual cortex and that amplitude modulation of these indices varies with emotional engagement.
