Alcohol-Induced Neurodegeneration: When, Where and Why?

Abstract: This manuscript reviews the proceedings of a symposium organized by Drs. Antonio Noronha and Fulton Crews presented at the 2003 Research Society on Alcoholism meeting. The purpose of the symposium was to examine recent findings on when alcohol induced brain damage occurs, e.g., during intoxication and/or during alcohol withdrawal. Further studies investigate specific brain regions (where) and the mechanisms (why) of alcoholic neurodegeneration. The presentations were (1) Characterization of Synaptic Loss in Cerebella of Mature and Senescent Rats after Lengthy Chronic Ethanol Consumption, (2) Ethanol Withdrawal Both Causes Neurotoxicity and Inhibits Neuronal Recovery Processes in Rat Organotypic Hippocampal Cultures, (3) Binge Drinking-Induced Brain Damage: Genetic and Age Related Effects, (4) Binge Ethanol-Induced Brain Damage: Involvement of Edema, Arachidonic Acid and Tissue Necrosis Factor , (TNF,), and (5) Cyclic AMP Cascade, Stem Cells and Ethanol. Taken together these studies suggest that alcoholic neurodegeneration occurs through multiple mechanisms and in multiple brain regions both during intoxication and withdrawal. [source]


MHC Gene Related Effects on Microglia and Macrophages in Experimental Autoimmune Encephalomyelitis Determine the Extent of Axonal Injury

Myelin-oligodendrocyte-glycoprotein (MOG)-induced experimental autoimmune encephalomyelitis (EAE) in rats is a chronic inflammatory demyelinating disease of the central nervous system (CNS) strongly mimicking multiple sclerosis (MS). We determined the involvement of macrophages and microglia in the lesions of MOG-EAE in relation to different major histocompatibility complex (MHC, RT1 in rat) haplotypes. We used intra-RT1 recombinant rat strains with recombinations between the RT1a and RT1u haplotypes on the disease permissive LEW non-MHC genome. Activated microglia and macrophages were identified morphologically and by expression of ED1 and allograft inhibitory factor-1 (AIF-1), and differentiated by their morphological phenotype. White matter lesions contained more macrophages and less microglia compared to grey matter lesions. Similarly active lesions were mainly infiltrated by macrophages, while microglia were abundant in inactive demyelinated plaques. In addition, we found a highly significant genetic association between a macrophage or microglia dominated lesional phenotype, which was independent from location and activity of the lesions. This was not only the case in demyelinating plaques of chronic EAE, but also in purely inflammatory lesions of acute passive transfer EAE. Rat strains with an u-haplotype in both the Class II and the telomeric non-classical Class I region revealed inflammatory and demyelinating lesions, which were dominated by activated microglia. The a-haplotype in any of these regions was associated with macrophage dominated lesions. A comparison of lesions, exactly matched for stages of demyelinating activity in these different rat strains, showed that in spite of a similar extent of demyelination, axonal injury was significantly less in microglia compared to macrophage dominated lesions. Thus, our studies document a genetic influence of the MHC-region on the relative contribution of macrophages versus microglia in the pathogenesis of EAE. [source]


Is improved high speed performance following frusemide administration due to diuresis-induced weight loss or reduced severity of exercise-induced pulmonary haemorrhage?

Summary Reasons for performing study: Prerace administration of frusemide to horses has been linked with a significant improvement in racing performance, but the basis for this improvement is unclear. Objective: To test whether improved performance with prerace administration of frusemide is due to the drug's diuresis-induced weight loss rather than its apparent alleviation of exercise-induced pulmonary haemorrhage (EIPH). Methods: Eight thoroughbred horses underwent 3 trials in a random order, 2 or 3 weeks apart: control (C), frusemide/unburdened (FU), and frusemide/burdened (FB). None of the horses were known to have exhibited post-exercise epistaxis or endoscopic evidence of EIPH. Endoscope-guided bronchoalveolar lavages (BALs) were performed before and after each horse completed a standardised exercise test (SET) on an inclined treadmill to assess semi-quantitatively the volume of EIPH. For C, horses received an i.v. saline placebo injection (5 ml) and were unburdened while performing the SET. With FU, horses received frusemide (0.5 mg/kg) and were also unburdened. For FB, horses received frusemide and were burdened with weight equal to that lost during the 4 h post frusemide injection period. Erythrocyte number in BAL fluid, mass specific VO2max, time and distance for the entire SET as well as at maximum speed were recorded. A one-way repeated measures analysis of variance was conducted on all results. Results: Mass specific VO2max was significantly higher for the FU than for FB or C. Mass specific VO2max for FB and C were not different. More RBCs were found in BAL samples after C runs than after both FU and FB trial runs. Horses with the frusemide treatment (either burdened or unburdened) produced less EIPH than in the C trial, but their mass specific VO2max values were higher on the FU trial alone. For FU, horses ran longer at 115% VO2max than under C or FB conditions. Conclusion and potential relevance: Improvement of performance in the furosemide trials was due more to the weight-loss related effects of the drug than its apparent alleviation of EIPH. Further research is warranted with the same or similar project design, but with a larger sample size and with horses known to have more severe EIPH. [source]

Statistics and orientation correlations of embedded/bulk material grains were deduced from two-dimensional X-ray diffraction patterns and give deep insight into the formation of the microstructure. The effect of respiration variations on independent component analysis results of resting state functional connectivity

Abstract The analysis of functional connectivity in fMRI can be severely affected by cardiac and respiratory fluctuations. While some of these artifactual signal changes can be reduced by physiological noise correction routines, signal fluctuations induced by slower breath-to-breath changes in the depth and rate of breathing are typically not removed. These slower respiration-induced signal changes occur at low frequencies and spatial locations similar to the fluctuations used to infer functional connectivity, and have been shown to significantly affect seed-ROI or seed-voxel based functional connectivity analysis, particularly in the default mode network. In this study, we investigate the effect of respiration variations on functional connectivity maps derived from independent component analysis (ICA) of resting-state data. Regions of the default mode network were identified by deactivations during a lexical decision task. Variations in respiration were measured independently and correlated with the MRI time series data. ICA appears to separate the default mode network and the respiration-related changes in most cases. In some cases, however, the component automatically identified as the default mode network was the same as the component identified as respiration-related. Furthermore, in most cases the time series associated with the default mode network component was still significantly correlated with changes in respiration volume per time, suggesting that current methods of ICA may not completely separate respiration from the default mode network. An independent measure of the respiration provides valuable information to help distinguish the default mode network from respiration-related signal changes, and to assess the degree of residual respiration related effects. Pharmaceutical antibiotic compounds in soils , a review

Antibiotics are highly effective, bioactive substances. As a result of their consumption, excretion, and persistence, they are disseminated mostly via excrements and enter the soils and other environmental compartments. Resulting residual concentrations in soils range from a few ,g upto g kg,1 and correspond to those found for pesticides. Numerous antibiotic molecules comprise of a non-polar core combined with polar functional moieties. Many antibiotics are amphiphilic or amphoteric and ionize. However, physicochemical properties vary widely among compounds from the various structural classes. Existing analytical methods for environmental samples often combine an extraction with acidic buffered solvents and the use of LC-MS for determination. In soils, adsorption of antibiotics to the organic and mineral exchange sites is mostly due to charge transfer and ion interactions and not to hydrophobic partitioning. Sorption is strongly influenced by the pH of the medium and governs the mobility and transport of the antibiotics. In particular for the strongly adsorbed antibiotics, fast leaching through soils by macropore or preferential transport facilitated by dissolved soil colloids seems to be the major transport process. Antibiotics of numerous classes are photodegraded. However, on soil surfaces this process if of minor influence. Compared to this, biotransformation yields a more effective degradation and inactivation of antibiotics. However, some metabolites still comprise of an antibiotic potency. Degradation of antibiotics is hampered by fixation to the soil matrix; persisting antibiotics were already determined in soils. Effects on soil organisms are very diverse, although all antibiotics are highly bioactive. The absence of effects might in parts be due to a lack of suitable test methods. However, dose and persistence time related effects especially on soil microorganisms are often observed that might cause shifts of the microbial community. Significant effects on soil fauna were only determined for anthelmintics. Due to the antibiotic effect, resistance in soil microorganisms can be provoked by antibiotics. Additionally, the administration of antibiotics mostly causes the formation of resistant microorganisms within the treated body. Hence, resistant microorganisms reach directly the soils with contaminated excrements. When pathogens are resistant or acquire resistance from commensal microorganisms via gene transfer, humans and animals are endangered to suffer from infections that cannot be treated with pharmacotherapy. The uptake into plants even of mobile antibiotics is small. However, effects on plant growth were determined for some species and antibiotics. Die Molekülstruktur von Antibiotika besteht häufig aus einem unpolaren Kern und polaren Randgruppen. Das Verteilungsverhalten hängt dabei entscheidend vom pH-Wert des Mediums ab und beeinflusst die Mobilität und Verlagerung der Antibiotika. Bei vielen, insbesondere stark adsorbierten Antibiotika sind v.,a. schnelle Fließvorgänge wie durch präferenziellen und Makroporenfluss sowie der Cotransport mit gelösten Bodenkolloiden von besonderer Bedeutung. Antibiotika vieler Strukturklassen können durch Licht abgebaut werden. Dieser Abbaupfad spielt auf Bodenoberflächen jedoch nur eine untergeordnete Rolle. Hingegen kommt es insbesondere durch biologische Transformationsprozesse zu einer intensiven Degradation und Inaktivierung der Antibiotika. Verschiedene Metaboliten weisen jedoch ebenfalls ein antibiotisches Potential auf. Der Abbau der Antibiotika wird durch die Festlegung in Böden gehemmt; dementsprechend wurde eine Persistenz verschiedener Antibiotika nachgewiesen. Trotz der starken bioaktiven Wirkung aller Antibiotika sind die festgestellten Effekte auf Bodenorganismen sehr unterschiedlich. Dies liegt nicht zuletzt an einem Mangel an geeigneten Testmethoden. In der Regel sind jedoch von Dosis und Wirkungsdauer abhängige Effekte insbesondere auf Mikroorganismen festzustellen, die zu Veränderungen der Mikroorganismenpopulation führen können. Lediglich durch Anthelmintika wurden deutliche Wirkungen auf Vertreter der Bodenfauna hervorgerufen. Infolge der antibiotischen Wirkung der Pharmazeutika kann eine Resistenzbildung bei Bodenorganismen ausgelöst werden. Zudem hat die Medikation von Antibiotika die Bildung resistenter Mikroorganismen bereits im behandelten Organismus zur Folge. Durch deren anschließende Ausscheidung gelangen resistente Keime auch direkt in die Böden. Handelt es sich um resistente Pathogene oder kommt es zur Übertragung der Resistenzgene zwischen kommensalen und pathogenen Mikroorganismen, so besteht das erhebliche Risiko einer nicht therapierbaren Infektion von Mensch und Tier. Die Aufnahme selbst mobiler Antibiotika in die Pflanzen ist sehr gering. Dennoch wurden bei einigen Pflanzenarten Wirkungen von Antibiotika auf das Wachstum nachgewiesen. [source]


Development of Mathematics Interest in Adolescence: Influences of Gender, Family, and School Context

This study investigated adolescents' developmental trajectories of mathematics interest and explored related effects of gender, family, and school context. Latent growth curve modeling was used to analyze longitudinal data of N=3,193 students (51% female) from grades 5 to 9 from all 3 ability tracks of the German state school system. Annual assessments involved student questionnaires on interest in mathematics, perceptions of classroom characteristics (classroom values for mathematics, mathematics teacher enthusiasm), as well as parent questionnaires regarding family values for mathematics. Results indicated a downward trend of students' mathematics interest that plateaued in later years, with high variability in mean levels, but little variability in the shape of the growth trajectories. Boys reported higher mathematics interest than girls, but similar downward growth trajectories. Students from the lowest ability track showed more favorable interest trajectories than students from the middle and highest tracks. Family values and classroom characteristics were positively related to within-person levels of interest over time and to average individual levels of interest, but not to growth parameters. Theoretical and practical implications are discussed. [source]


Temperature related effects on embryonic development of the Mediterranean locust, Dociostaurus maroccanus

Summary Laboratory studies were conducted to assess the effect of temperature on the development of the eggs of Dociostaurus maroccanus (Thunberg) (Orthoptera, Acrididae) during anatrepsis (stages I,XIV) and during catatrepsis (stages XV,XX). The developmental rates of anatrepsis were studied at five constant temperatures ranging from 10 to 30°C. Egg development occurred over the entire range but at 10°C the embryos were unable to complete anatrepsis. The relationship between temperature and developmental times for completing anatrepsis was analysed by the non-linear Logan type III model. The optimal temperature estimated for the development of eggs during anatrepsis was 24.7°C; the lower and upper thermal thresholds were 9°C and 31°C, respectively. Once the embryos completed anatrepsis, only those incubated at 15°C continued morphogenesis beyond stage XIV (diapause stage) without a low-temperature exposure period. The developmental rate of catatrepsis was studied at four constant temperatures ranging from 15°C to 30°C after exposure to low-temperature, 10°C, for 30, 60 or 90 days. For catatrepsis, temperature and developmental time were linearly and inversely related. Linear regression was used to estimate the lower developmental threshold and the degree days requirements for catatrepsis. Both decreased with longer exposure to the low temperature; the former from 13.8°C to 10.5°C and the latter from 212.8 to 171.5 degree days, following 30 and 90 days at 10°C, respectively. Our results improve the ability of decision support systems for Mediterranean locust pest management by providing better forecasts to land managers and pest advisors. [source]


Juvenile idiopathic arthritis and HLA Class I and Class II interactions and age-at-onset effects

Objective The aim of this study was to quantitate risk and to examine heterogeneity for HLA at high resolution in patients with the most common subtypes of juvenile idiopathic arthritis (JIA), IgM rheumatoid factor,negative polyarticular JIA and oligoarticular JIA. Use of 4-digit comprehensive HLA typing enabled great precision, and a large cohort allowed for consideration of both age at disease onset and disease subtype. Methods Polymerase chain reaction,based high-resolution HLA typing for class I and class II loci was accomplished for 820 patients with JIA and 273 control subjects. Specific HLA epitopes, potential interactions of alleles at specific loci and between loci (accounting for linkage disequilibrium and haplotypic associations), and an assessment of the current International League of Associations for Rheumatology classification criteria were considered. Results An HLA,DRB1/DQB1 effect was shown to be exclusively attributable to DRB1 and was similar between patients with oligoarticular JIA and a younger subgroup of patients with polyarticular JIA. Furthermore, patients with polyarticular JIA showed age-specific related effects, with disease susceptibility in the group older than age 6 years limited to an effect of the HLA,DRB1*08 haplotype, which is markedly different from the additional susceptibility haplotypes, HLA,DRB1*1103/1104, found in the group with oligoarticular JIA and the group of younger patients with polyarticular JIA. Also in contrast to findings for oligoarticular JIA, patients with polyarticular arthritis had no evidence of an HLA class I effect. Markers associated with a reduced risk of disease included DRB1*1501, DRB1*0401, and DRB1*0701. DRB1*1501 was shown to reduce risk across the whole cohort, whereas DRB1*0401 and DRB1*0701 were protective for selected JIA subtypes. Surprisingly, the disease predisposition mediated by DPB1*0201 in individuals without any disease-predisposing DRB1 alleles was great enough to overcome even the very strong protective effect observed for DRB1*1501. Conclusion Inherited HLA factors in JIA show similarities overall as well as differences between JIA subtypes. [source]


The effects of hormonal contraceptives on bone turnover markers and bone health

Summary Sex hormones are important regulators of bone metabolism. As hormonal contraceptives contain either oestrogens or progestins, or a combination thereof, it is conceivable that these widely used agents have an effect on bone metabolism and bone health. The main users of hormonal contraceptives, adolescent girls and young women, are still building bone and accruing bone mass and may therefore be particularly susceptible to the effects of hormonal contraceptives on bone. Despite these concerns, the effects of hormonal contraceptives on bone health are still poorly understood. As biochemical markers of bone turnover have been proven useful tools in the assessment and monitoring of bone metabolism, we reviewed the effects of combined and gestagen-only hormonal contraceptives on bone turnover markers and related effects on bone mineral density and fracture risk in premenopausal women, as documented in the literature until January 2009.
