Erectile Function Recovery Rate after Radical Prostatectomy: A Meta-Analysis

ABSTRACT Introduction., Erectile function recovery (EFR) rates after radical prostatectomy (RP) vary greatly based on a number of factors, such as erectile dysfunction (ED) definition, data acquisition means, time-point postsurgery, and population studied. Aim., To conduct a meta-analysis of carefully selected reports from the available literature to define the EFR rate post-RP. Main Outcome Measures., EFR rate after RP. Methods., An EMBASE and MEDLINE search was conducted for the time range 1985,2007. Articles were assessed blindly by strict inclusion criteria: report of EFR data post-RP, study population ,50 patients, ,1 year follow-up, nerve-sparing status declared, no presurgery ED, and no other prostate cancer therapy. Meta-analysis was conducted to determine the EFR rate and relative risks (RR) for dichotomous subgroups. Results., A total of 212 relevant studies were identified; only 22 (10%) met the inclusion criteria and were analyzed (9,965 RPs, EFR data: 4,983 subjects). Mean study population size: 226.5, standard deviation = 384.1 (range: 17,1,834). Overall EFR rate was 58%. Single center series publications (k = 19) reported a higher EFR rate compared with multicenter series publications (k = 3): 60% vs. 33%, RR = 1.82, P = 0.001. Studies reporting ,18-month follow-up (k = 10) reported higher EFR rate vs. studies with <18-month follow-up (k = 12), 60% vs. 56%, RR = 1.07, P = 0.02. Open RP (k = 16) and laparoscopic RP (k = 4) had similar EFR (57% vs. 58%), while robot-assisted RP resulted in a higher EFR rate (k = 2), 73% compared with these other approaches, P = 0.001. Patients <60 years old had a higher EFR rate vs. patients ,60 years, 77% vs. 61%, RR = 1.26, P = 0.001. Conclusions., These data indicate that most of the published literature does not meet strict criteria for reporting post-RP EFR. Single and multiple surgeon series have comparable EFR rates, but single center studies have a higher EFR. Secured Creditor Recovery Rates from Management Buy-outs in Distress

G33; G32 Buy-out literature suggests that secured creditors will recoup substantial proportions of the funds they extend to finance the initial buy-out. This paper uses a unique dataset of 42 failed MBOs to examine the extent of credit recovery by secured lenders under UK insolvency procedures and the factors that influence the extent of this recovery. On average, secured creditors recover 62 per cent of the amount owed. The percentage of secured credit recovered is increased where the distressed buy-out is sold as a going concern and where the principal reason for failure concerns managerial factors. The presence of a going concern qualification in the audit report and the size of the buy-out reduce the recovery rate by secured creditors. [source]


Ovum Pick-up in Sheep: a Comparison between Different Aspiration Devices for Optimal Oocyte Retrieval

Contents In vivo ovum pick-up (OPU) in sheep may be improved with a proper choice of aspiration elements (needle and tubing) and aspiration vacuum pressure. In the present study, two experiments were carried out. In Expt 1, visible follicles in ovaries of slaughtered ewes (treated separately according to their diameters: small <3 mm, medium 3,5 mm and large >5 mm) were aspirated using different combinations of the three studied factors such as aspiration flow rate (10, 20, 30, 40 and 50 ml water/min), needle gauge (18 and 20 G) and tubing inner diameter (1, 2 or 3 mm internal diameter). In Expt 2, a study with two 18 G needles of different lengths (18 G: 82 mm; 18 GL: 600 mm) was carried out, using ovaries obtained post-mortem, and performing in vivo laparoscopic follicular aspiration on ewes. We considered good quality oocytes as those with both complete compact cumulus and a homogeneous cytoplasm. Recovery rate, proportion of good quality oocytes (good quality oocytes/100 oocytes recovered) and overall efficiency (good quality oocytes/100 follicles aspirated) were noted. In Expt 1, aspiration flow rate affect remarkable proportion of good quality oocytes (69.5%, 50.5%, 44.8%, 36.5% and 28.3% for flows from 10 to 50 ml/min respectively, p < 0.05). Needle gauge did not affect aspiration device efficiency. Thin and intermediate tubings were more effective (overall efficiency rates: 34.9%, 32.3% and 28.1% for 1, 2 and 3 mm respectively, p < 0.05). Follicle size did not affect recovery rate, but proportion of good quality oocytes was higher for large (77.9%) and medium (64.4%) follicles (p < 0.05). Finally, some combinations of the aspiration device showed greater effectiveness. In Expt 2, needle length did not influence recovery rate, but good quality oocytes rate was significantly modified both post-mortem and in vivo (good quality rate for 18 G vs 18 GL needles: 69.5% vs 47.7% and 58.1% vs 25.4%, post-mortem and in vivo respectively, p < 0.05). We conclude that low-aspiration flow rates (10 and 20 ml/min) with thin or intermediate tubings (1 and 2 mm), and any short needle (18 G or 20 G) are the most adequate aspiration factors for OPU in sheep. A Novel Process for the Recovery of Polyphenols from Grape (Vitis vinifera L.) Pomace

ABSTRACT: A novel process for enzyme-assisted extraction of polyphenols from winery by-products was established on a pilot-plant scale. Optimization of enzymatic hydrolysis of grape skins, that is, selection of pectinolytic and cellulolytic enzymes, enzyme-substrate ratio, and time-temperature regime of enzymatic treatment, was conducted on a laboratory scale. Enzyme activities were monitored by viscosity measurement of resuspended grape pomace and by quantification of oligomeric pectin and cellulose degradation products released from cell wall material. Optimal conditions were obtained with 5000 ppm (based on dry matter) of a pectinolytic and 2500 ppm of a cellulolytic enzyme preparation, respectively, at 50°C, which were also applied in pilot-plant scale experiments. Concomitant determination of individual polyphenolics demonstrated a significantly improved yield for most compounds when compared with experiments without enzyme addition. Recovery rates were comparable to those obtained when grape pomace was extracted using sulfite. Pre-extraction of the pomace with hot water followed by treatment with cell wall degrading enzymes even increased yields of phenolic compounds. Only some quercetin glycosides and malvidin coumaroylglucoside were partly hydrolyzed due to enzyme side activities. This new process may provide a valuable alternative to the application of sulfite, which is considered crucial in food processing. [source]


Determination of polycyclic aromatic hydrocarbons in olive oil by a completely automated headspace technique coupled to gas chromatography-mass spectrometry

Abstract A new and completely automated method for the determination of ten relevant polycyclic aromatic hydrocarbons (PAHs) in olive oil is proposed using an extraction by the headspace (HS) technique. Quantification and confirmation steps are carried out by gas chromatography-mass spectrometry (GC-MS) combining simultaneously selected-ion monitoring (SIM) and tandem mass spectrometry (MS/MS). This combination offers on one hand an increased sensitivity and on the other hand, selective and reliable qualitative information. Sample pretreatment or clean-up are not necessary because the olive oil sample is put directly into an HS vial, automatically processed by HS and introduced into the GC-MS instrument for analysis. Because of its high selectivity and sensitivity, a triple-quadrupole (QqQ) detector coupled with the gas chromatograph allows us to limit handling. Each sample is completely processed in approximately 63 min (45 min for HS isolation and 18 min for GC-MS determination), a reduced time compared with previously published methods. The chemical and instrumental variables were preliminarily optimized using uncontaminated olive oil samples spiked with 25 µg kg,1 of each target compound. The final method was validated to ensure the quality of the results. The precision was satisfactory, with relative standard deviation (RSD) values in the range 3,9%. Recovery rates ranged from 96 to 99%. Limits of detection (LOD) were calculated as 0.02,0.06 µg kg,1 and the limits of quantification (LOQ) were obtained as 0.07,0.26 µg kg,1. It must be mentioned that the LOD and LOQ are much lower than the maximum levels established by the European Union (EU) in oils and fats intended for direct human consumption or for use as an ingredient in foods, which are set at 2 µg kg,1. All the figures of merit are completely in accordance with the latest EU legislation. Immuno-ultrafiltration as a new strategy in sample clean-up of aflatoxins

Abstract The present paper describes the development of a new clean-up strategy for the analysis of aflatoxins (AFs) in food. The sample preparation method is based on immuno-ultrafiltration (IUF) which, in contrast to immunoaffinity chromatography, makes use of antibodies in free form. After selecting an appropriate ultrafiltration (UF) device and optimizing different operation conditions the IUF method was applied to the clean-up of maize and rice. Quantification of AFs was carried out by HPLC and fluorescence detection, after postcolumn derivatization in a Kobracell. The IUF method was shown to be as selective as sample clean-up using commercial immunoaffinity columns. Recovery rates and RSD for the AFs G2, G1, B2 and B1 in spiked rice were found to be 76 ± 3, 76 ± 2, 83 ± 5 and 99 ± 14%, respectively. The analysis of a FAPAS (food analysis performance assessment scheme) maize material resulted in AFs concentrations which were in the range assigned by the producer of the reference material. In vitro degradation by mixed rumen bacteria of 17 mono- and sesquiterpenes typical of winter and spring diets of goats on Basilitica rangelands (southern Italy)

Abstract BACKGROUND: Nine monoterpenes (,-3-carene, p -cymene, limonene, ,-myrcene, (E)- and (Z)-,-ocimene, ,-phellandrene, ,-terpinene, ,-terpinene), seven oxygenated monoterpenes (1,8-cineole, linalool, (E)- and (Z)-linalool oxide, 4-terpinenol, ,-terpineol, ,-terpinolene) and one sesquiterpene (,-cedrene) were investigated for their degradability in the rumen microbial ecosystem. These molecules were identified as dominant terpenes in the winter and spring diets of milking goats in Basilicata (southern Italy). RESULTS: All terpenes were tested at 3.33 µL L,1 for 24 h using in vitro incubation with mixed rumen bacteria from dairy goats. Oxygen-containing compounds were those recovered at the highest levels (89% of (E)-linalool oxide, 93% of (Z)-linalool oxide, 91% of 1,8-cineole, 82% of terpineol and 72% of 4-terpinenol), except linalool. The linear alkenes ,-myrcene and ,-ocimene almost completely disappeared. Results were more variable among cyclic alkenes, with recovery rates ranging from 50% in the case of limonene to less than 1% for ,-phellandrene. 17% of the only sesquiterpene of the group, ,-cedrene, was recovered. CONCLUSION: Recovery rates differed markedly among terpenes, partly in relation to the presence of oxygen and rings in the molecules. Effects of the commercial probiotic Lactobacillus casei on the growth, protein content of skin mucus and stress resistance of juveniles of the Porthole livebearer Poecilopsis gracilis (Poecilidae)

Abstract A 11-week feeding trial was carried out to determine the effects of the probiotic bacteria Lactobacillus casei from the commercial product Yakult® on the growth performance, proximal composition, protein content of skin mucus and stress resistance of juvenile Porthole livebearer Poeciliopsis gracilis. Triplicate groups of 15 juveniles per tank with an initial weight of 47 ± 9 mg (mean ± standard deviation) were fed with Artemia nauplii enriched with the probiotic, by using the bacteria cells plus the fermented milk (group ProN) and the other (group ProC) by using only the bacterial cells, eliminating the fermented milk by centrifugation. A control of fish was set up, by feeding non-enriched Artemia nauplii. Growth performance and survival rates did not show significantly differences among the treatments and control group, but a slightly tendency of higher values for body weight, weight gain and specific growth rate was observed in the juveniles of ProC treatment. Whole body proximate composition did not show significant differences among the groups, but higher values of protein and lipid contents were observed in the groups fed with the probiotic. Content of protein in the skin mucus were significantly higher in the ProC treatment than control group. Recovery rates after an air-dive test were significantly higher on the fish fed with the probiotic cells than the control group. These results show that L. casei might be used as a probiotic for fish and would help during the culture of juvenile of the Porthole livebearer P. gracilis. [source]


Protein kinase A modulates A-type potassium currents of larval zebrafish (Danio rerio) white muscle fibres

Abstract Aims:, Potassium (K+) channels are involved in regulating cell excitability and action potential shape. To our knowledge, very little is known about the modulation of A-type K+ currents in skeletal muscle fibres. Therefore, we sought to determine whether K+ currents of zebrafish white skeletal muscle were modulated by protein kinase A (PKA). Methods:, Pharmacology and whole-cell patch clamp were used to examine A-type K+ currents and action potentials associated with zebrafish white skeletal muscle fibres. Results:, Activation of PKA by a combination of forskolin + 3-isobutyl-1-methylxanthine (Fsk + IBMX) decreased the peak current density by ,60% and altered the inactivation kinetics of A-type K+ currents. The specific PKA inhibitor H-89 partially blocked the Fsk + IBMX-induced reduction in peak current density, but had no effect on the change in decay kinetics. Fsk + IBMX treatment did not shift the activation curve, but it significantly reduced the slope factor of activation. Activation of PKA by Fsk + IBMX resulted in a negative shift in the V50 of inactivation. H-89 prevented all Fsk + IBMX-induced changes in the steady-state properties of K+ currents. Application of Fsk + IBMX increased action potential amplitude, but had no significant effect on action potential threshold, half width or recovery rate, when fibres were depolarized with single pulses, paired pulses or with high-frequency stimuli. Conclusion:, PKA modulates the A-type K+ current in zebrafish skeletal muscle and affects action potential properties. Our results provide new insights into the role of A-type K+ channels in muscle physiology. [source]


Advancing Loss Given Default Prediction Models: How the Quiet Have Quickened

We describe LossCalcÔ version 2.0: the Moody's KMV model to predict loss given default (LGD), the equivalent of (1 , recovery rate). LossCalc is a statistical model that applies multiple predictive factors at different information levels: collateral, instrument, firm, industry, country and the macroeconomy to predict LGD. We find that distance-to-default measures (from the Moody's KMV structural model of default likelihood) compiled at both the industry and firm levels are predictive of LGD. We find that recovery rates worldwide are predictable within a common statistical framework, which suggests that the estimation of economic firm value (which is then available to allocate to claimants according to each country's bankruptcy laws) is a dominant step in LGD determination. LossCalc is built on a global dataset of 3,026 recovery observations for loans, bonds and preferred stock from 1981 to 2004. This dataset includes 1,424 defaults of both public and private firms , both rated and unrated instruments , in all industries. We demonstrate out-of-sample and out-of-time LGD model validation. The model significantly improves on the use of historical recovery averages to predict LGD. [source]


Influence of isolation on the recovery of pond mesocosms from the application of an insecticide.

Abstract The immediate response and recovery of the macrobenthic communities of nonisolated and isolated freshwater outdoor 9 m3 mesocosms following an acute stress caused by the addition of deltamethrin were studied over a 14-month period. To discriminate between internal and external recovery mechanisms, half of the treated ponds were covered by 1-mm mesh lids that restricted aerial recolonization. Both structural (abundance of the different taxonomic groups) and functional (litter breakdown) parameters were monitored. Insects were broadly reduced in numbers by deltamethrin addition. In general, noninsect groups were not affected or increased in abundance in deltamethrin-treated ponds, probably because of relative insensitivity to deltamethrin, reduced predation, and lower competition for food. No major change in litter breakdown rates were seen, probably because of functional redundancy among the macrobenthic community. Chironominae larvae recovered in open, treated mesocosms 62 d after deltamethrin addition and most insect groups recovered 84 d after the treatment date. However, the presence of lids significantly reduced insect recovery rate, suggesting that it largely depends on the immigration of winged forms (i.e., external recovery) from surrounding non- or less affected systems. These results indicate that the recovery time of macrobenthic communities in an affected natural pond would depend on spatial characteristics of the landscape and also the season that exposure occurs. Isolated ecosystems would display posttreatment insect recovery dynamics very different from highly connected ones, evolving toward alternate pseudoequilibrium states, possibly with lower biodiversity but with preserved functionality. Consequences for higher tier risk assessment of pesticides are discussed. [source]

Episodic ataxia type 1 mutations in the KCNA1 gene impair the fast inactivation properties of the human potassium channels Kv1.4-1.1/Kv,1.1 and Kv1.4-1.1/Kv,1.2

Abstract Episodic ataxia type 1 (EA1) is an autosomal dominant neurological disorder characterized by constant muscle rippling movements (myokymia) and episodic attacks of ataxia. Several heterozygous point mutations have been found in the coding sequence of the voltage-gated potassium channel gene KCNA1 (hKv1.1), which alter the delayed-rectifier function of the channel. Shaker -like channels of different cell types may be formed by unique hetero-oligomeric complexes comprising Kv1.1, Kv1.4 and Kv,1.x subunits. Here we show that the human Kv,1.1 and Kv,1.2 subunits modulated the functional properties of tandemly linked Kv1.4-1.1 wild-type channels expressed in Xenopus laevis oocytes by (i) increasing the rate and amount of N-type inactivation, (ii) slowing the recovery rate from inactivation, (iii) accelerating the cumulative inactivation of the channel and (iv) negatively shifting the voltage dependence of inactivation. To date, the role of the human Kv1.4-1.1, Kv1.4-1.1/Kv,1.1 and Kv1.4-1.1/Kv,1.2 channels in the aetiopathogenesis of EA1 has not been investigated. Here we also show that the EA1 mutations E325D, V404I and V408A, which line the ion-conducting pore, and I177N, which resides within the S1 segment, alter the fast inactivation and repriming properties of the channels by decreasing both the rate and degree of N-type inactivation and by accelerating the recovery from fast inactivation. Furthermore, the E325D, V404I and I177N mutations shifted the voltage dependence of the steady-state inactivation to more positive potentials. The results demonstrate that the human Kv,1.1 and Kv,1.2 subunits regulate the proportion of wild-type Kv1.4-1.1 channels that are available to open. Furthermore, EA1 mutations alter heteromeric channel availability which probably modifies the integration properties and firing patterns of neurones controlling cognitive processes and body movements. Abstract:, We previously demonstrated that the electrical state of the skin surface influences epidermal permeability barrier homeostasis. At the interface between different materials, electrons are localized heterogeneously and induce electrical potential. In the present study, we evaluated the effects of metals on the barrier recovery. When we put pure gold plate on skin immediately after tape stripping, the barrier recovery rate was faster than the control. The acceleration of barrier recovery was blocked when the plate was earthed (grounded). When a plastic membrane was sandwiched between the plate and the skin, the recovery was delayed in comparison with the control. We then used a germanium diode to regulate the current flow between the plate and the earth. When the current was blocked, the barrier recovery was accelerated, but when the current was not blocked, the recovery was not accelerated. These results suggest that localization of electron might affect for the barrier recovery rate. The level of interfacial electric potential would be different due to the electrochemical property of metal. Thus, we next evaluated the effects of other metals. With samarium, zirconium, iridium and silver, the barrier recovery rate was faster than in the case of gold, while a platinum plate induced slower recovery than in the case of gold. There was a significant correlation between work function of each metal and barrier recovery rate. These results suggest that electron donation from outside accelerated the skin barrier recovery. Effect of glucose crosslinking on thermomechanical properties and shape memory effect of PET-PEG copolymers

Abstract Poly(ethylene terephthalate) (PET) and poly (ethylene glycol) (PEG) copolymers crosslinked with glucose as a crosslinker are prepared to improve their mechanical and shape memory properties compared to the one without crosslinking. Composition of PEG and glucose is varied to search for the one with the best mechanical and shape memory properties. The highest shape recovery rate is found in the copolymer composed of 25 mol % PEG-200 and 2.0 mol % glucose. The result that crosslinking by glucose improves the shape recovery rate and supports the high shape recovery rate under the repetitive cyclic test conditions, compared to the one without crosslinking, will be discussed in the points of the structure and shape memory mechanism. © 2008 Wiley Periodicals, Inc. Rarity, specialization and extinction in primates

Aim To determine and explain biological traits that distinguish rare from common primate taxa. Location Africa, Americas, Asia, Madagascar. Methods We compare the biology of rare primate taxa with the biology of common taxa. Rarity is defined by (1) small size of geographic range; (2) small geographic range plus low local population density; and (3) small geographic range plus low local density plus narrow habitat specificity. After a linear comparison of size of geographic range with various biological traits, globally and by realm, extremes of rarity and commonness per realm are identified, and then combined for a global analysis. Tests are done both with genera treated as independent data points (n=62), and also with phylogenetic control by use of an independent contrasts test. Extinction risk in vertebrates, including primates, often correlates with high resource requirements, slow population recovery rate, and specialization. The three indices of rarity are therefore compared with these three general traits. Measures of resource use are body mass, local density, annual range size, and group size; of recovery rate, interbirth interval, and maximum intrinsic rate of natural population increase; and of degree of specialization, variety of diet, of habitats, maximum latitude, and morphological variety. All data come from the literature. Because several measures are compared, probabilities are Bonferroni corrected. Results If rarity in primates correlates with any biological attribute, it consistently correlates with only measures of specialization, and not with measures of high resource use, or slow population recovery rate. Without phylogenetic correction, the first two indices of rarity associate significantly with all four measures of specialization, and the third with maximum latitude. With phylogenetic correction, the first index still associates with all four, the second with two (maximum latitude, number of species per genus), and the third shows no significant associations. While the four measures of specialization are strongly interrelated, stepwise regressions on geographic range indicate that maximum latitude has the strongest effect, followed by dietary variety and number of species per genus and, finally, habitat variety. Main conclusions The most commonly demonstrated traits of susceptibility to extinction are those of high resource use, slow recovery rate, and specialization. Yet, while rarity (almost however, it is defined) is an inevitable precursor to extinction, specialization is the only trait found to correlate with rarity in this study. We cannot explain this apparent contradiction. [source]


Rapid screening assay of congenital adrenal hyperplasia by measuring 17,-hydroxyprogesterone with high-performance liquid chromatography/electrospray ionization tandem mass spectrometry from dried blood spots

Abstract A rapid, simple, and specific method was developed for the diagnosis of congenital adrenal hyperplasia (CAH) from dried blood spots on newborn screening cards based on high-performance liquid chromatography/electrospray ionization tandem mass spectrometry (HPLC/ESI-MS/MS). The usefulness of 17,-hydroxyprogesterone (17OH-P) determination on dried filter-paper blood samples from patients with CAH caused by 21-hydroxylase deficiency was evaluated. The LC/MS/MS detection of 17OH-P was rapid, <4 min. The intra- and interday accuracy and precision of the method were <7%. Our procedure maintained good linearities (R2 > 0.992) and recovery rate (>83%). We used this new method to directly determine the 17OH-P levels in dried blood specimens from abnormal children of various ages, with a detection limit of 20 ng/ml (,240 pg), to avoid the time-consuming derivatization steps required by the gas-chromatography/mass spectrometry (GC/MS) method. Four dried filter-paper blood samples of CAH patients (three girls and one boy, 1,14 years old) were all quantified in an LC/MS/MS study and revealed high 17OH-P levels (>90 ng/ml). After treatment, all of the elevated 17OH-P levels either decreased or disappeared. Compared with CAH patients, 17OH-P was nearly undetectable (<20 ng/ml) in the normal infants by LC/MS/MS. This LC/MS/MS assay is not only useful for both diagnosis and monitoring of treatment of CAH in all other age groups, it also can be used as a screening test for CAH infants. Novel interpenetrating networks with shape-memory properties

Abstract Novel polyesterurethane/poly(ethylene glycol) dimethacrylate (PEGDMA) interpenetrating networks (IPNs) with good shape-memory properties were synthesized using solvent casting method. The star-shaped oligo[(rac -lactide)- co -glycolide] was coupled with isophorone diisocyanate to form a polyesterurethane network (PULG), and PEGDMA was photopolymerized to form another polyetheracrylate network. IPNs were transparent and gel content exceeded 92%. The values of strain fixity rate and strain recovery rate were above 93%. PULG and PEGDMA networks in IPNs were amorphous and did not show any characteristic diffraction peaks in X-ray diffraction spectra. Only one glass transition temperature (Tg) of the IPNs between Tg of PEGDMA and PULG was observed, which was proportional to PEGDMA content. PULG and PEGDMA networks were miscible when PEGDMA content was below 50 wt %. The hydrophilicity, transition temperatures, and mechanical properties of IPNs could be conveniently adjusted through variation of network compositions to match the promising potential clinical or medical applications. © 2007 Wiley Periodicals, Inc. Shape-memory polymer networks from oligo(,-caprolactone)dimethacrylates

Abstract Polymer networks showing a thermally induced shape-memory effect were prepared through the crosslinking of oligo(,-caprolactone)dimethacrylates under photocuring with or without an initiator. The influence of the molecular weight of the oligo(,-caprolactone)dimethacrylates and the initiator concentration on the macroscopic properties of the polymer networks was investigated. The isothermal and nonisothermal crystallization behavior of the polymer networks was evaluated as a basic principle of the functionalization process. Determination of seventeen main flavonoids and saponins in the medicinal plant Huang-qi (Radix Astragali) by HPLC-DAD-ELSD

Abstract A simple, rapid, and reliable method, namely high-performance liquid chromatography coupled with diode array and evaporative light scattering detectors (HPLC-DAD-ELSD), was developed to simultaneously determine twelve major flavonoids and five main saponins in different parts of the medicinal plant Huang-qi (Radix Astragali). The DAD wavelength was set at 280 nm for the UV detection of flavonoids, while the drift tube temperature was set at 105°C and the nebulizing gas flow rate at 2.7 L/min for ELSD detection of saponins. The method was fully validated with respect to linearity (r2 >0.998), sensitivity, precision, and accuracy (recovery rate between 93.3 and 104.2%). The analytical results of different parts of the medicinal plant Huang-qi revealed that the levels of total flavonoids or saponins in individual parts can vary considerably and the concentration of each compound in different parts is also significantly different. The aerial parts (stems and leaves) contain even higher total contents of flavonoids (although of different kinds) than the commonly used roots of the plants. In addition, the concentration of total flavonoids and saponins in the extract of the fibrous roots was surprisingly highest among all parts of Astragalus species. All of these findings provide clear evidence and scientific support for utilization of different parts of the medicinal plant Huang-qi and also for reduction in waste of plant resources. [source]


Profiling human gut bacterial metabolism and its kinetics using [U- 13C]glucose and NMR

Abstract This study introduces a stable-isotope metabolic approach employing [U- 13C]glucose that, as a novelty, allows selective profiling of the human intestinal microbial metabolic products of carbohydrate food components, as well as the measurement of the kinetics of their formation pathways, in a single experiment. A well-established, validated in vitro model of human intestinal fermentation was inoculated with standardized gastrointestinal microbiota from volunteers. After culture stabilization, [U- 13C]glucose was added as an isotopically labeled metabolic precursor. System lumen and dialysate samples were taken at regular intervals. Metabolite concentrations and isotopic labeling were determined by NMR, GC, and enzymatic methods. The main microbial metabolites were lactate, acetate, butyrate, formate, ethanol, and glycerol. They together accounted for a 13C recovery rate as high as 91.2%. Using an NMR chemical shift prediction approach, several minor products that showed 13C incorporation were identified as organic acids, amino acids, and various alcohols. Using computer modeling of the 12C contents and 13C labeling kinetics, the metabolic fluxes in the gut microbial pathways for synthesis of lactate, formate, acetate, and butyrate were determined separately for glucose and unlabeled background substrates. This novel approach enables the study of the modulation of human intestinal function by single nutrients, providing a new rational basis for achieving control of the short-chain fatty acids profile by manipulating substrate and microbiota composition in a purposeful manner. Gated dynamic 31P MRS shows reduced contractile phosphocreatine breakdown in mice deficient in cytosolic creatine kinase and adenylate kinase

Abstract We developed a new dedicated measurement protocol for dynamic 31P MRS analysis in contracting calf muscles of the mouse, using minimally invasive assessment of the contractile force combined with the acquisition of spectroscopic data gated to muscle contraction and determination of phosphocreatine (PCr) recovery rate and ATP contractile cost. This protocol was applied in a comparative study of six wild type (WT) mice and six mice deficient in cytosolic creatine kinase and adenylate kinase isoform 1 (MAK,/, mice) using 70 repeated tetanic contractions at two contractions per minute. Force levels during single contractions, and metabolite levels and tissue pH during resting conditions were similar in muscles of MAK,/, and WT mice. Strikingly, muscle relaxation after contraction was significantly delayed in MAK,/, mice, but during repeated contractions, the decrease in the force was similar in both mouse types. Gated data acquisition showed a negligible PCr breakdown in MAK,/, immediately after contraction, without a concomitant decrease in ATP or tissue pH. This protocol enabled the determination of rapid PCr changes that would otherwise go unnoticed due to intrinsic low signal-to-noise ratio (SNR) in mouse skeletal muscles combined with an assessment of the PCr recovery rate. Our results suggest that MAK,/, mice use alternative energy sources to maintain force during repeated contractions when PCr breakdown is reduced. Furthermore, the absence of large increases in adenosine diphosphate (ADP) or differences in force compared to WT mice in our low-intensity protocol indicate that creatine kinase (CK) and adenylate kinase (AK) are especially important in facilitating energy metabolism during very high energy demands. Patch dynamics in a landscape modified by ecosystem engineers

Ecosystem engineers, organisms that modify the environment, have the potential to dramatically alter ecosystem structure and function at large spatial scales. The degree to which ecosystem engineering produces large-scale effects is, in part, dependent on the dynamics of the patches that engineers create. Here we develop a set of models that links the population dynamics of ecosystem engineers to the dynamics of the patches that they create. We show that the relative abundance of different patch types in an engineered landscape is dependent upon the production of successful colonists from engineered patches and the rate at which critical resources are depleted by engineers and then renewed. We also consider the effects of immigration from either outside the system or from engineers that are present in non-engineered patches, and the effects of engineers that can recolonize patches before they are fully recovered on the steady state distribution of different patch types. We use data collected on the population dynamics of a model engineer, the beaver, to estimate the per-patch production rate of new colonists, the decay rate of engineered patches, and the recovery rate of abandoned patches. We use these estimated parameters as a baseline to determine the effects of varying parameters on the distribution of different patch types. We suggest a number of hypotheses that derive from model predictions and that could serve as tests of the model. [source]


Stability of ecosystem properties in response to above-ground functional group richness and composition

While there has been a rapidly increasing research effort focused on understanding whether and how composition and richness of species and functional groups may determine ecosystem properties, much remains unknown about how these community attributes affect the dynamic properties of ecosystems. We conducted an experiment in 540 mini-ecosystems in glasshouse conditions, using an experimental design previously shown to be appropriate for testing for functional group richness and composition effects in ecosystems. Artificial communities representing 12 different above-ground community structures were assembled. These included treatments consisting of monoculture and two- and four-species mixtures from a pool of four plant species; each plant species represented a different functional group. Additional treatments included two herbivore species, either singly or in mixture, and with or without top predators. These experimental units were then either subjected to an experimentally imposed disturbance (drought) for 40 d or left undisturbed. Community composition and drought both had important effects on plant productivity and biomass, and on several below-ground chemical and biological properties, including those linked to the functioning of the decomposer subsystem. Many of these compositional effects were due to effects both of plant and of herbivore species. Plant functional group richness also exerted positive effects on plant biomass and productivity, but not on any of the below-ground properties. Above-ground composition also had important effects on the response of below-ground properties to drought and thus influenced ecosystem stability (resistance); effects of composition on drought resistance of above-ground plant response variables and soil chemical properties were weaker and less consistent. Despite the positive effects of plant functional group richness on some ecosystem properties, there was no effect of richness on the resistance of any of the ecosystem properties we considered. Although herbivores had detectable effects on the resistance of some ecosystem properties, there were no effects of the mixed herbivore species treatment on resistance relative to the single species herbivore treatments. Increasing above-ground food chain length from zero to three trophic levels did not have any consistent effect on the stability of ecosystem properties. There was no evidence of either above-ground composition or functional group richness affecting the recovery rate of ecosystem properties from drought and hence ecosystem resilience. Our data collectively point to the role of composition (identity of functional group), but not functional group richness, in determining the stability (resistance to disturbance) of ecosystem properties, and indicates that the nature of the above-ground community can be an important determinant of the consistency of delivery of ecosystem services. The effects of MRI signal intensity changes and clinical manifestations on prognosis after surgical intervention for cervical spondylotic myelopathy

Objective:, To investigate whether the magnetic resonance (MR) T2 image signal intensity ratio and clinical manifestations can predict the prognosis in patients with cervical spondylotic myelopathy (CSM). Methods:, A total of 73 patients treated with anterior, posterior, or posterior-anterior combined surgery for compressive cervical myelopathy were enrolled retrospectively in this study. 1.5 T magnetic resonance imaging (MRI) was performed on all patients before surgery. T2-weighted images (T2WI) of sagittal signal intensity were obtained of the cervical spinal cord, and the regions of interest (ROI) were taken by 0.05 cm2. MR T2WI of sagittal normal cord signal at the C7-T1 disc level were also obtained, and the ROI were taken by 0.3 cm2. Signal value was measured by computer and the signal ratio between regions 0.05 cm2 and 0.3 cm2 calculated. Where no intramedullary high signal intensity was noted on MR T2WI, the ROI were taken by 0.05 cm2 of the region of most severe spinal cord compression. The 73 patients were divided into three groups by hierarchical clustering analysis with signal intensity ratio (group 1: low signal intensity ratio; group 2: middle signal intensity ratio; group 3: high signal intensity ratio). Statistical analyses were performed with SPSS 11.0. Results:, There were significant differences between the three groups according to the recovery rate (P < 0.001), age (P= 0.003), duration of disease (P= 0.001), Babinski sign (P < 0.001), pre- and postoperative Japanese Orthopaedic Association (JOA) score (P= 0.006). With increases in both signal intensity ratio grade and age, the recovery rate and pre- and postoperative JOA scores gradually decreased, and the incidence of Babinski sign increased. There was no significant difference in sex among the three groups (P= 0.387). Multiple comparison tests further supported the above-mentioned results. Conclusion:, Patients with light or no intramedullary signal changes on T2WI had a good surgical outcome. However, increase of signal intensity ratio and occurrence of the pyramidal sign were associated with a poor prognosis after surgery. [source]


Thermomechanical studies of aluminum nitride filled shape memory polymer composites

High thermal conductivity polyurethane shape memory polymer (SMP) composites filled with aluminum nitride (AlN) were fabricated, and their thermal and thermomechanical properties were studied. The purpose of this microstructure is to improve the thermal properties of the SMPs at low filler content. Morphology of AlN filler in polyurethane SMP matrix and the resulting thermal conductivity was also investigated. Thermal studies have shown that AlN is an effective filler for reinforcement of the polyurethane SMP and that it does not deteriorate the stable physical crosslink structure of the polyurethane, which is necessary to store the elastic energy in the service process of the shape memory material. The thermal conductivities of these SMP composites in relation to filler concentration and temperature were investigated, and it was found that the thermal conductivity can increase up to 50 times in comparison with that of the pure SMP. Furthermore, differential scanning calorimetry tests have shown a significant decrease in the glass transition temperature of the switching segment. Dynamic mechanical studies have shown that the storage modulus of the composites increase with higher AlN content in both glassy and rubbery state. Damping peak decreases and also the curve of damping becomes broader with increasing filler content. Strain fixity rate which expresses the ability of the specimens to fix their strain has been improved slightly in the presence of AlN filler but the final recovery rate of the shape memory measurement has decreased evidently. Polydimethylsiloxane,cristobalite composite adhesive system for aerospace applications

Abstract The effect of phase-pure cristobalite (a high temperature crystalline polymorph of silica) on the adhesive characteristics of hydroxyl terminated polydimethylsiloxane (PDMS) was studied. The potential advantages of PDMS/cristobalite composite system as an adhesive for aerospace applications are also discussed. A PDMS/cristobalite composite adhesive system containing different filler contents (0,46 volume percentage, vol%) was prepared. The filler material, phase-pure cristobalite, was synthesized by the pyrolysis of fused silica at 1400°C. The mechanical, rheological, and thermal characteristics of the composites were studied. A high yield stress (0.151,Pa), shear-thinning index (1.051), and fast recovery rate were observed for ,34,vol% cristobalite loading, which indicate that PDMS retains its excellent adhesive and flow characteristics even at high filler loading with enhanced mechanical characteristics. Thermal analysis shows the onset of degradation of PDMS shifts to higher temperatures, 372,438°C and 317,417°C in nitrogen and air atmosphere respectively, which shows excellent thermal stability. The residual component yields after thermal degradation of PDMS/cristobalite composite system in nitrogen and air atmosphere show different degradation mechanisms. Diffusion technique for 15N and inorganic N analysis of low-N aqueous solutions and Kjeldahl digests,

Diffusion of ammonia is a common sample preparation method for the stable isotope analysis of inorganic nitrogen in aqueous solution. Classical diffusion methods usually require 6,12 days of diffusion and often focus on 15N/14N analysis only. More recent studies have discussed whether complete N recovery was necessary for the precise analysis of stable N isotope ratios. In this paper we present a newly revised diffusion technique that allows correct and simultaneous determination of total N and 15N at% from aqueous solutions and Kjeldahl digests, with N concentrations down to sub-0.5-mg,N,L,1 levels, and it is tested under different conditions of 15N isotope labelling. With the modification described, the diffusion time was reduced to 72,h, while the ratios of measured and expected 15N at% were greater than 99% and the simultaneous recovery of total N was >95%. Analysis of soil microbial biomass N and its 15N/14N ratio is one of the most important applications of this diffusion technique. An experiment with soil extracts spiked with 15N-labelled yeast showed that predigestion was necessary to prevent serious N loss during Kjeldahl digestion of aqueous samples (i.e. soil extracts). The whole method of soil microbial biomass N preparation for 15N/14N analysis included chloroform fumigation, predigestion, Kjeldahl digestion and diffusion. An experiment with soil spiked with 15N-labelled yeast was carried out to evaluate the method. Results showed a highly significant correlation of recovered and added N, with the same recovery rate (0.21) of both total N and 15N. A kN value of 0.25 was obtained based on the data. In conclusion, the diffusion method works for soil extracts and microbial biomass N determination and hence could be useful in many types of soil/water studies. Procedure for Maximizing Oocyte Harvest for In Vitro Embryo Production in Small Ruminants

Contents Possible effects of repeated hormonal treatments and laparoscopic ovum pick-up (LOPU) on the efficiency of oocyte recovery rate and quality were determined in sheep and goats. In six adult Merino sheep and five Criolla goats, ovarian status was synchronized by a prostaglandin F2, analogue and the insertion of an intravaginal sponge 48 h later. Follicle development was stimulated by a single dose of FSH (60 mg NIH-FSH-P1) plus a single dose of equine chorionic gonadotrophin (eCG; 300 UI). The first FSH/eCG doses were administered 48 h after the sponge insertion, being repeated every 4 days to complete a total of four treatments in sheep and three in goats. Follicles in both ovaries were categorized according to their diameter and follicular fluid was aspirated under laparoscopic observation without a vacuum pump. In sheep, during a 12-day-period, a total of 347 follicles were aspirated with a recovery rate of 46.9%. In goats, during an 8-day-period, 219 follicles were aspirated with a recovery rate of 45.6%. In both species, there were no significant differences in the number of aspirated follicles, oocyte recovery rate and good quality oocyte recovery rate. However, in sheep the oocyte recovery rate was higher for large follicles, whereas in goats no such effect was detected. In summary, current results indicate that retrieval of oocytes can be maximized, without affecting oocyte quality, by repeating ,oneshot' FSH/eCG regimes and LOPUs at intervals as short as 4 days. Effect of Days Post-Partum, Breed and Ovum Pick-Up Scheme on Bovine Oocyte Recovery and Embryo Development

Contents The objective of this study was to investigate (i) the effect of two different ovum pick-up (OPU) schemes (once vs twice weekly aspirations) on oocyte recovery rate, quality and subsequent in vitro embryo development, (ii) the influence of days post-partum on oocyte recovery and (iii) possible differences in OPU results from two different herds. In group A, OPU was performed twice weekly in two Holstein Friesian (HF) and three Danish Red and White (DRW) cows from a private herd. In the research herd, two groups of eight HF cows were investigated: group B (OPU once weekly) and group C (OPU twice weekly). The collected oocytes were subsequently submitted to in vitro embryo production. More oocytes were recovered from the private herd when compared with the research herd. In the research herd, the twice weekly scheme aspirated more oocytes than the once weekly scheme. The quality of the retrieved oocytes was significantly different between groups B and C but not between groups A and C, and HF cows yielded higher quality oocytes than DRW cows (p = 0.029). Oocytes from group C showed higher level of embryonic development than group B oocytes. No differences in blastocyst rates were observed between groups A and C. Session affected the number of retrieved oocytes and subsequent developmental rates, with these being lower in the first compared with the last sessions. Finally, there was no significant effect of days post-partum in the number and quality of the retrieved oocytes, likely because of the small group size and high variation between sessions. [source]
