A STUDY ON SUITABILITY OF FOUR ENRICHMENT BROTHS FOR PCR-BASED DETECTION OF LISTERIA MONOCYTOGENES FROM RAW MEAT

ABSTRACT Four enrichment broths were evaluated for their compatibility with the polymerase chain reaction (PCR) for detection of Listeria monocytogenes from raw meat after single-step enrichment. Standardized PCR protocols for listeriolysin O (hlyA) gene were used for the species-specific identification of L. monocytogenes. Four broths, namely, modified University of Vermont broth (MUVM), Listeria enrichment broth (LEB), Fraser broth (FB) and polymyxin, acriflavin, lithium chloride, ceftazidime, aesculin, mannitol, egg yolk broth (PALCAM) , were inoculated with L. monocytogenes. The enriched cultures were subjected for PCR. Similarly, meat samples were artificially spiked with various concentrations of L. monocytogenes, these spiked samples were enriched in the above-mentioned four broths and subjected to PCR to determine the medium that was most compatible for PCR-based detection of L. monocytogenes. The aliquots taken during different incubation periods were subjected to three different procedures for the concentration of the target organism for use in PCR. Results revealed that MUVM was better than other broths for the detection of L. monocytogenes by both PCR and cultural method; moreover, it was able to support the growth of as low as 10 cfu/g of meat. Concentration of the target organisms by centrifugation and washing with PCR buffer was the most suitable method for improving PCR performance for detection of L. monocytogenes. Goat (n = 67) and buffalo (n = 45) meat samples from local markets were also screened by both PCR and cultural method to validate the results obtained from the spiking studies. Both results were in agreement in spiking studies as well as screening of market meat samples. [source]


Anti-microbial hand washes for domestic use , their effectiveness in vitro and in normal use

Abstract The killing or removal of microbes from the hands is a critical factor in food safety as many studies have shown the hands to be both an important source of microbes and powerful agents of cross-contamination in hospital and domestic situations. In response to this concern, a number of novel hand-washing products have appeared on the market. These products contain anti-microbial agents and claim to be more effective at removing bacteria than soap bars and conventional liquid soaps. This study attempts to test these claims by comparing the effectiveness of a conventional soap bar, a conventional liquid soap and an anti-microbial liquid soap containing triclosan. In vitro tests demonstrate that the anti-microbial liquid soap is more effective than conventional liquid soaps in reducing the viability of six bacterial species and that this effect is both time and dose dependent. However, when the three soaps were compared for their ability to reduce microbial counts on the hands no differences were observed between the three products. For all three soaps, counts after washing sometimes went up and sometimes down when compared with pre-wash counts. This was the case both when the soaps were used ,normally', that is, with great variation in the time taken, water and soap volumes used and method of washing and after a standardized, rigorous wash recommended in clinical situations. Furthermore, reduction in microbial counts from hands contaminated by handling raw meat was no greater for the anti-microbial than for the conventional liquid soap. [source]


Cross-contamination in the kitchen: effect of hygiene measures

Abstract Aims:, To determine the effect of hygiene measures on cross-contamination of Campylobacter jejuni at home and to select a safe tracer organism for C. jejuni. Methods and Results: Comparative tests were conducted with nonpathogenic Escherichia coli and Lactobacillus casei and L. casei was chosen as the safe tracer organism. Salads containing chicken breast fillet contaminated with a known number of C. jejuni and L. casei were prepared according to different cross-contamination scenarios and contamination levels of salads were determined. Cross-contamination could be strongly reduced when cleaning cutting board and cutlery with hot water (68°C), but generally was not prevented using consumer-style cleaning methods for hands and cutting board. Conclusions:, Dish-washing does not sufficiently prevent cross-contamination, thus different cutting boards for raw meat and other ingredients should be used and meat,hand contact should be avoided or hands should be thoroughly cleaned with soap. Lactobacillus casei can be used as a safe tracer organism for C. jejuni in consumer observational studies. Significance and Impact of the Study:, Cross-contamination plays an important role in the transmission of food-borne illness, especially for C. jejuni. This study delivers suitable data to quantitatively assess the risk of campylobacteriosis caused by cross-contamination and it shows the effect of different preventive hygiene measures. [source]


PREVALENCE AND ANTIMICROBIAL RESISTANCE OF LISTERIA SPECIES IN FOOD PRODUCTS IN BANGKOK, THAILAND

ABSTRACT A total of 380 meat and meat products, dairy and dairy products, fresh vegetables, fresh seafood, and ready-to-eat food samples from supermarkets in Bangkok, Thailand were collected and analyzed for the occurrence of Listeria spp. and of Listeria monocytogenes. The overall incidence of Listeria spp. was 16.8%, most of them were isolated from raw meat and vegetables. L. monocytogenes was isolated from 18 (4.7%) out of 380 studied samples. Other species isolated were L. innocua (6.6%), L. ivanovii (0.8%), L. seeligeri (0.5%), L. grayi (1.6%) and L. welshimeri (2.6%). The antimicrobial susceptibilities of the 64 isolate of Listeria spp. were also examined by the standard disk diffusion method. Listeria spp. were resistant to penicillin (6.3%), chloramphenicol (3.1%) and tetracycline (1.6%), but sensitive to amoxicillin, vancomycin, ampicillin, rifampicin and sulfamethoxazole. PRACTICAL APPLICATIONS Listeria monocytogenes prevalence in food products in Bangkok has been documented. More studies on the occurrence of L. monocytogenes are needed to establish microbiological criteria of foods in the country. The findings of our study, increases in antibiotic resistance among Listeria spp. will provide useful information for the development of public health policy in the use of antimicrobials in food animal production. BOTULINAL TOXIN PRODUCTION IN VACUUM AND CARBON DIOXIDE PACKAGED MEAT DURING CHILLED STORAGE AT 2 AND 4C

ABSTRACT This study was undertaken to determine if carbon dioxide packaging of meat afforded a food safety advantage over vacuum packaging with respect to botulinal toxin production during chilled storage. A cocktail of washed spores from five toxigenic clostridial strains , four reference Clostridium botulinumstrains [types A, B (2 strains) and E] and a C. butyricum type E strain , was inoculated onto lamb chumps. Of these strains, two were psychrotolerant. The inoculated chumps were individually carbon dioxide packaged and duplicate packs were placed into storage at 10, 8, 6, 4 and 2C. All storage regimens included a weekly defrost cycle when meat surface temperatures increased by up to 6 to 7C during a 2 to 2.5 h period. After 84 days storage, packs were assessed for the presence of botulinal toxin using the mouse bioassay procedure. All packs contained botulinal toxin. To compare toxin production in vacuum and carbon dioxide packs at chill temperatures, the challenge trials were repeated for 4 and 2C storage. Packs were examined at regular intervals for toxin presence. Both pack types contained toxin after 21 and 48 days storage at 4 and 2C, respectively. In the unlikely, but not impossible, event that raw meat would be contaminated with psychrotolerant toxincapable clostridial spores, product safety, with respect to botulinal toxin presence after prolonged chilled storage, requires storage temperatures to be maintained below 2C for both vacuum and carbon dioxide packaged product. Effects of Vitamin E and Organic Selenium on Oxidative Stability of ,-3 Enriched Dark Chicken Meat during Cooking

ABSTRACT:, The influence of vitamin E and selenomethionine (SeMet) on lipid oxidation in frozen,raw and cooked ,-3 enriched dark chicken meat was evaluated. Feed was supplemented with 2 levels of vitamin E (250 and 50 IU/kg of feed) and selenium (0.1 mg of sodium selenite/kg of feed and 0.3 mg of SeMet/kg of feed). An extruded linseed product was used as the ,-linolenic acid source. Fatty acid (FA) profile, oxysterols, and thiobarbituric reactive acid substances (TBARs) were analyzed in frozen,raw, boiled, pan-fried, and roasted meat. After 6 mo of storage, oxysterols in frozen,raw meat remained stable with either high or low levels of dietary antioxidants. During cooking, high levels of vitamin E reduced oxysterol formation, whereas high levels of SeMet were inconsistent and even increased oxysterols during roasting. TBARs in frozen,raw meat stored for 6 mo were inhibited by high levels of either antioxidant. Conversely, no protective effect during cooking was observed at this time of storage. After 12 mo at ,30 °C no antioxidant protection was observed. [source]


Neosporosis and hammondiosis in dogs

The dog is a definitive host of the protozoan parasite Neospora caninum, and in many parts of the world, infection is relatively common as determined by serology. Reported seroprevalences usually range from 0 to 20 per cent, however, reports of clinically affected dogs are infrequent. Affected dogs are generally less than six months old and predominantly have signs of an ascending hindleg paralysis, with the associated lesions of polyradiculoneuritis and granulomatous polymyositis. Although any organ may be affected, infections are more common in the central nervous system, muscles, lungs and skin. Ante-mortem diagnosis is difficult but serology and cytology can aid diagnosis. The diagnosis can be confirmed by histology, immunohistochemistry, the use of molecular techniques on biopsy material, or on post-mortem examination. Neospora caninum oocysts are rarely found in faeces and must be differentiated from oocysts of related coccidians such as Hammondia heydorni and Toxoplasma gondii. Hammondia heydorni can cause diarrrhoea in immunosuppressed dogs. Neosporosis should be suspected in young pups with an ascending paralysis of the hindlegs. Treatment with clindamycin and potentiated sulphonamides may be useful in cases where muscular atrophy and fibrosis are absent. Feeding of raw meat is a potential risk factor for infection of dogs and should be discouraged. [source]


Playing the Stockmarket in Tana Toraja

This paper describes the players and the play in a weekly stockmarket in Tana Toraja. South Sulawesi, where up to 600 buffalo bulls are bought and sold for exorbitant prices by any standards. These prices are partly determined by external, global economic forces. The buffaloes are all intended for sacrifice in elaborate funerals. Although contributed by individuals or families they are conceived as part of mana. the common wealth of tongkonan. the origin houses of the Toraja. Live bulls cannot be given away but are able to be lent in ceremonial exchanges at funerals. After sacrifice, their raw meat is distributed to participants in the funeral in a version of potlatch. Bulls mediate all exchange. apart from mundane commodity exchange, and are liquid assets. A mature black bull is an object of general equivalence able to be exchanged as payment for certain symbolic objects. In the past the production, distribution and circulation of buffaloes, both on the hoof and as meat, were controlled by the nobility. Buffaloes were said to be in finite supply. They derived from the Upperworld and accompanied the ancestors of the Toraja nobility to this world and were replenished through the ritual of the ways of the ancestors. Now the advent of the market has democratised buffaloes. There has been a dispersion of wealth and the power that the bulls embody. Any man,noble, commoner or former slave-who has sufficient cash is able to buy and sell buffaloes: to have a share in the stockmarket. The marketplace has become a new field of power play, one where innovative methods are being found for increasing the supply of buffaloes by importing hundreds of inferior quality buffaloes and by a program of artificial insemination which has been instigated by the local government. The role of intramuscular connective tissue in meat texture

ABSTRACT The structure, composition and amount of intramuscular connective tissue (IMCT) vary tremendously between muscles, species and breeds, and certainly contribute to meat texture. With animal growth, collagen crosslinks become more stable, and the structural integrity of IMCT increases. These changes increase the mechanical properties of IMCT, contributing to the toughening of meat. Intramuscular fat deposits, mainly in the perimysium between muscle fiber bundles, result in marbling. This causes the remodeling of IMCT structures and reduces the mechanical strength of IMCT, contributing to the tenderization of beef. The IMCT has been thought to be rather immutable compared to myofibrils during postmortem ageing of meat. However, recent studies have shown the disintegration of IMCT during postmortem ageing of meat and its relationship to tenderization of raw meat, although its contribution to cooked meat is still controversial. Given the large influence of IMCT on meat texture, further elucidations of molecular mechanisms which change the structural integrity of IMCT during chronological ageing of animals and postmortem ageing of meat are needed. Effects of dietary supplementation with conjugated linoleic acid on fatty acid composition and lipid oxidation in chicken breast meat

ABSTRACT We investigated the effects of dietary conjugated linoleic acid (CLA) on fatty acid composition and lipid oxidation in breast meat of broiler chickens. Broiler chickens (28-day-old females) were fed diets containing experimental oils at 20 g/kg diet for 28 days. The experimental oils consisted of either a 2:0, 1:1, or a 0:2 (wt : wt) ratio of safflower oil (high linoleic acid content) to a commercial CLA mixture. In this study, dietary CLA supplementation significantly increased the composition and content of CLA in chicken meat. The predominant CLA in meat from birds with supplemented diets was the cis -9, trans -11 isomer. The proportion of saturated fatty acid in meat significantly increased with increasing CLA supplementation, with a corresponding decrease in monounsaturated fatty acid. Dietary CLA also reduced thiobarbituric acid reactive substances (TBARS) values in raw meat during storage at 4°C for 5 days. These results provide evidence that CLA feeding is a practical strategy not only for adding nutritional benefits to chicken meat but also for improving meat quality including oxidative stability. Successful treatment of butcher's warts with imiquimod 5% cream

Summary A unique correlation has been shown to exist between frequent occupational handling of raw meat and development of warts on the hands, leading to the designation of ,butcher's warts'. Numerous treatments are available for warts including cryotherapy, laser therapy and surgical excision. We present a case of a 31-year-old-butcher with a 4-year history of warts on his fingers. Imiquimod 5% cream was applied once daily to the warts for 3 months. Clinical improvement was noted after 1 month, with regression of the warts experienced after 3 months. During 12 months of follow-up, no recurrent lesions were observed. [source]


New and old risk-factors for Toxoplasma gondii infection: prospective cross-sectional study among military personnel in the Czech Republic

Abstract The aims of this study were to evaluate seroprevalence and the importance of various risk-factors for Toxoplasma infection in the Czech Republic. A prospective cross-sectional survey was conducted among military personnel in Prague. Consenting subjects (n = 3250) completed a questionnaire concerning demographics and risk-factors, and blood samples were taken to determine anti- Toxoplasma antibody titres according to complement fixation and ELISA IgG and IgM tests. The seroprevalence of toxoplasmosis was 23%. In multivariate analysis, independent predictors of Toxoplasma seropositivity were age (OR 1.03,/,year), consumption of raw meat (OR 1.35), owning a cat (OR 1.25), owning rabbits (OR 1.47), childhood residence in a town with a population of <10 000 inhabitants (OR 1.63) vs. location of the childhood residence in a town with population of >100 000 inhabitants, and blood group type A (OR 1.28), B (OR 1.33) or AB (OR 1.43) vs. O. These results suggested that horizontal toxoplasmosis transmission in the Czech Republic may occur through consumption of raw meat, contact with cat faeces and farming. [source]


Genotyping of Campylobacter spp. from retail meats by pulsed-field gel electrophoresis and ribotyping

Abstract Aims:, To determine the genetic relatedness of Campylobacter spp. from retail meat products, and compare the discriminatory power of pulsed-field gel electrophoresis (PFGE) and automatic ribotyping. Methods and Results:, A total of 378 Campylobacter isolates recovered from 159 raw meats (130 chicken, 25 turkey, three pork and one beef) sampled from 50 retail grocery stores of four supermarket chains in the Maryland suburban area from August 1999 to July 2000 were analysed by PFGE with SmaI, 120 isolates of which were also characterized by ribotyping with PstI using RiboPrinterŪ system. A total of 148 unique PFGE patterns were identified, 91 of which were present in multiple Campylobacter isolates and 24 in multiple meat samples. Nineteen Campylobacter clones with identical PFGE patterns recurred frequently (up to nine times) throughout the sampling period. Comparing ribotyping with PFGE, we identified 44 PFGE patterns and 22 RiboGroups among the 120 isolates tested. Multiple PFGE patterns within one RiboGroup were commonly observed, as well as multiple RiboGroups within one PFGE pattern. Conclusions:, Although Campylobacter present in retail meats were genetically diverse, certain clones persisted in poultry meats. PFGE had a greater discriminatory power than ribotyping, and the two methods were complementary in genotyping Campylobacter. Significance and Impact of the Study:, Genomic DNA fingerprinting of Campylobacter confirmed diverse and recurrent Campylobacter clones in the retail meats, which provides additional data for a better understanding of the epidemiological aspect of Campylobacter infection.
