Selected AbstractsGlucagon-like peptide 1(GLP-1) in biology and pathologyDIABETES/METABOLISM: RESEARCH AND REVIEWS, Issue 2 2005Juris J. Meier Abstract Post-translational proteolytic processing of the preproglucagon gene in the gut results in the formation of glucagon-like peptide 1 (GLP-1). Owing to its glucose-dependent insulinotropic effect, this hormone was postulated to primarily act as an incretin, i.e. to augment insulin secretion after oral glucose or meal ingestion. In addition, GLP-1 decelerates gastric emptying and suppresses glucagon secretion. Under physiological conditions, GLP-1 acts as a part of the ,ileal brake', meaning that is slows the transition of nutrients into the distal gut. Animal studies suggest a role for GLP-1 in the development and growth of the endocrine pancreas. In light of its multiple actions throughout the body, different therapeutic applications of GLP-1 are possible. Promising results have been obtained with GLP-1 in the treatment of type 2 diabetes, but its potential to reduce appetite and food intake may also allow its use for the treatment of obesity. While rapid in vivo degradation of GLP-1 has yet prevented its broad clinical use, different pharmacological approaches aiming to extend the in vivo half-life of GLP-1 or to inhibit its inactivation are currently being evaluated. Therefore, antidiabetic treatment based on GLP-1 may become available within the next years. This review will summarize the biological effects of GLP-1, characterize its role in human biology and pathology, and discuss potential clinical applications as well as current clinical studies. [source] Determination of 1-methylhistidine and 3-methylhistidine by capillary and chip electrophoresis with contactless conductivity detectionELECTROPHORESIS, Issue 13 2007Petr T, ma Dr. Abstract CE with capacitively coupled contactless detection (C4D) was used to determine 3-methylhistidine (3-MH) and 1-methylhistidine (1-MH). The C4D response to 3-MH was studied in a BGE consisting of 500,mM acetic acid and ammonia at varying concentration and the results were compared with the theory. Complete separation of a model mixture of 3-MH, 1-MH, and histidine (His) was attained in two optimized BGEs, one containing 500,mM HAc, 20,mM NH4OH, and 0.1 % m/v hydroxyethylcellulose (HEC), pH,3.4 (I) and the other consisting of 100,mM morpholinoethanesulfonic acid (MES), 25,mM LiOH, and 0.1 % m/v HEC, pH,5.5 (II). These optimized BGEs were tested in CE/C4D analyses of urine. Promising results were obtained for separation and determination of 3-MH, 1-MH, and His on a silicon microchip, using aluminum strips as the C4D electrodes; the three analytes were baseline-separated within less than 30,s with a separation channel effective length of 38,mm. The LOD were satisfactory and amounted to 26.4,,M for 3-MH and 18.3,,M for 1-MH. [source] Columnar Mesomorphism in Hexacatenar Tetrahedral (2,2,-Bipyridine)zinc Complexes and Homologous Palladium DerivativesEUROPEAN JOURNAL OF INORGANIC CHEMISTRY, Issue 1 2005Giovanna Barberio Abstract The synthesis and characterisation of novel liquid crystals which display columnar mesomorphism induced upon complexation of a series of nonmesomorphic hexacatenar 4,4,-disubstituted 2,2,-bipyridines (Ln) are reported. The introduction of different metal centres (Zn, Pd) causes the appearance of mesomorphism in all complexes regardless of the geometry around the metal ion. We therefore report the first examples of mesomorphism in tetrahedral zinc derivatives. The nature of the columnar phases is related to the self-assembly of the half-disc shaped [LnMCl2] (M = Zn, Pd) complexes into full disc-shaped supramolecules. The molecular organisation in the mesophase is mainly driven by intermolecular attractive interactions, as shown by the crystal structure of the model compound [LPdCl2]. Preliminary measurements of photoconductivity have been performed on samples of [LnMCl2] complexes doped with C60 to increase absorption. Promising results were obtained. Lassouaoui Recently, the clinical role of image processing has been developed considerably. The resources of this new technology were exploited for the needs of doctors in their practice. In this study, we propose a computer vision for tracking the uterine collar cancer. Here, we present three stages: preprocessing, segmentation, and classification. The segmentation stage uses a multifractal algorithm based on the computation of the singularity exponents; its role is separating each cell on its core and its cytoplasm, which permits the analysis of each one in the recognition stage for deducing a response about the malignity of the cell. However, the classification is performed by an algorithm of area growth. Knowing that there are four layers in the epithelium, the classification allows for learning the type of each cell in an image for organizing the research in the recognition stage. Thus, we contribute to the creation of a database for the recognition stage. This base contains the core and cytoplasm images with information about the type of each cell. Promising results were obtained with a short execution time that permits the start of the recognition stage. © 2003 Wiley Periodicals, Inc. [source] ABSINTH: A new continuum solvation model for simulations of polypeptides in aqueous solutionsJOURNAL OF COMPUTATIONAL CHEMISTRY, Issue 5 2009Andreas Vitalis Abstract A new implicit solvation model for use in Monte Carlo simulations of polypeptides is introduced. The model is termed ABSINTH for self- Assembly of Biomolecules Studied by an Implicit, Novel, and Tunable Hamiltonian. It is designed primarily for simulating conformational equilibria and oligomerization reactions of intrinsically disordered proteins in aqueous solutions. The paradigm for ABSINTH is conceptually similar to the EEF1 model of Lazaridis and Karplus (Proteins 1999, 35, 133). In ABSINTH, the transfer of a polypeptide solute from the gas phase into a continuum solvent is the sum of a direct mean field interaction (DMFI), and a term to model the screening of polar interactions. Polypeptide solutes are decomposed into a set of distinct solvation groups. The DMFI is a sum of contributions from each of the solvation groups, which are analogs of model compounds. Continuum-mediated screening of electrostatic interactions is achieved using a framework similar to the one used for the DMFI. Promising results are shown for a set of test cases. These include the calculation of NMR coupling constants for short peptides, the assessment of the thermal stability of two small proteins, reversible folding of both an ,-helix and a ,-hairpin forming peptide, and the polymeric properties of intrinsically disordered polyglutamine peptides of varying lengths. The tests reveal that the computational expense for simulations with the ABSINTH implicit solvation model increase by a factor that is in the range of 2.5,5.0 with respect to gas-phase calculations. © 2008 Wiley Periodicals, Inc. Ingram Summary When considering all possible aging interventions evaluated to date, it is clear that calorie restriction (CR) remains the most robust. Studies in numerous species have demonstrated that reduction of calories 30,50% below ad libitum levels of a nutritious diet can increase lifespan, reduce the incidence and delay the onset of age-related diseases, improve stress resistance, and decelerate functional decline. A current major focus of this research area is whether this nutritional intervention is relevant to human aging. Evidence emerging from studies in rhesus monkeys suggests that their response to CR parallels that observed in rodents. To assess CR effects in humans, clinical trials have been initiated. However, even if results from these studies could eventually substantiate CR as an effective pro-longevity strategy for humans, the utility of this intervention would be hampered because of the degree and length of restriction required. As an alternative strategy, new research has focused on the development of ,CR mimetics'. The objective of this strategy is to identify compounds that mimic CR effects by targeting metabolic and stress response pathways affected by CR, but without actually restricting caloric intake. For example, drugs that inhibit glycolysis (2-deoxyglucose), enhance insulin action (metformin), or affect stress signaling pathways (resveratrol), are being assessed as CR mimetics (CRM). Promising results have emerged from initial studies regarding physiological responses which resemble those observed in CR (e.g. reduced body temperature and plasma insulin) as well as protection against neurotoxicity (e.g. enhanced dopamine action and up-regulated neurotrophic factors). Ultimately, lifespan analyses in addition to expanded toxicity studies must be accomplished to fully assess the potential of any CRM. Nonetheless, this strategy clearly offers a very promising and expanding research endeavor. [source] ADVANCES IN CLINICAL PRACTICE: New endoscopic and surgical treatment options for early esophageal adenocarcinomaJOURNAL OF GASTROENTEROLOGY AND HEPATOLOGY, Issue 9 2010Susan Gan Abstract Although the outcome for advanced stage esophageal cancer is poor, the early detection and treatment of early stage disease is usually associated with a much better outcome. Until recently, esophagectomy has been the treatment of choice in fit patients. However, morbidity is significant, and this has encouraged the development of newer endoscopic treatments that preserve the esophagus. These techniques include ablation and mucosal resection. Promising results are described, and endoscopic methods might provide a reasonable alternative for the treatment of early esophageal cancer. However, follow-up remains short and endoscopic treatment does not deal with potential lymphatic spread. Hence, careful selection is required. Minimally invasive techniques for esophageal resection have also been shown to be feasible, although there is only limited evidence that they reduce postoperative morbidity. Better data are still required to demonstrate improved outcomes from endoscopic treatment and minimally invasive esophagectomy. Wasiak Abstract Background Burn injury increases the body's metabolic demands, and therefore nutritional requirements. Provision of an adequate supply of nutrients is believed to lower the incidence of metabolic abnormalities, thus reducing septic morbidity, and improving survival rates. Enteral nutrition support is the best feeding method in a patient who is unable to achieve an adequate oral intake, but optimal timing of its introduction after burn injury (i.e. early versus late) needs to be established. The purpose of this review is to examine evidence for the effectiveness and safety of early versus late enteral nutrition support in adults with burn injury. Methods An examination of randomized and controlled clinical trials using various medical databases such as The Cochrane Library (Issue 3, 2006), MEDLINE (from 1950), CINHAL (from 1982) and EMBASE (from 1980). Results The trial evidence about the benefit of early enteral nutritional support on standardized clinical outcomes such as length of hospital stay and mortality remained inconclusive. Similarly, the question of whether early enteral feeding influenced or decreased metabolic rate, reduced septic and other complications remained uncertain. Conclusions Promising results suggest early enteral nutrition support may blunt the hypermetabolic response to thermal injury, but it is insufficient to provide clear guidelines for practice. Further research incorporating larger sample sizes and rigorous methodology that utilizes valid and reliable outcome measures is essential. [source] Image-based crystal detection: a machine-learning approachACTA CRYSTALLOGRAPHICA SECTION D, Issue 12 2008Roy Liu The ability of computers to learn from and annotate large databases of crystallization-trial images provides not only the ability to reduce the workload of crystallization studies, but also an opportunity to annotate crystallization trials as part of a framework for improving screening methods. Here, a system is presented that scores sets of images based on the likelihood of containing crystalline material as perceived by a machine-learning algorithm. The system can be incorporated into existing crystallization-analysis pipelines, whereby specialists examine images as they normally would with the exception that the images appear in rank order according to a simple real-valued score. Promising results are shown for 319,112 images associated with 150 structures solved by the Joint Center for Structural Genomics pipeline during the 2006,2007 year. Overall, the algorithm achieves a mean receiver operating characteristic score of 0.919 and a 78% reduction in human effort per set when considering an absolute score cutoff for screening images, while incurring a loss of five out of 150,structures. [source] Determination of quinocide as impurity in primaquine tablets by capillary zone electrophoresisBIOMEDICAL CHROMATOGRAPHY, Issue 5 2009Abdalla A. Elbashir Abstract A capillary zone electrophoretic method has been developed and validated for the determination of the impurity quinocide (QC) in the antimalarial drug primaquine (PQ). Different buffer additives such as native cyclodextrins and crown ethers were evaluated. Promising results were obtained when either , -cyclodextrin (, -CD) or 18-crown-6 ether (18C6) were used. Their separation conditions such as type of buffer and its pH, buffer additive concentration, applied voltage capillary temperature and injection time were optimized. The use of 18C6 offers slight advantages over , -CD such as faster elution times and improved resolution. Nevertheless, migration times of less than 5 min and resolution factors (Rs) in the range of 2,4 were obtained when both additives were used. The method was validated with respect to selectivity, linearity, limits of detection and quantitation, analytical precision (intra- and inter-day variability) and repeatability. Concentrations of 2.12 and 2.71% (w/w) of QC were found in pharmaceutical preparations of PQ from two different manufacturers. [source] Automatic guidance of a four-wheel-steering mobile robot for accurate field operationsJOURNAL OF FIELD ROBOTICS (FORMERLY JOURNAL OF ROBOTIC SYSTEMS), Issue 6-7 2009Christophe Cariou As world population growth requires an increasing level of farm production at the same time that environmental preservation is a priority, the development of new agricultural tools and methods is required. In this framework, the development of robotic devices can provide an attractive solution, particularly in the field of autonomous vehicles. Accurate automatic guidance of mobile robots in farming constitutes a challenging problem for researchers, mainly due to the low grip conditions usually found in such a context. From assisted-steering systems to agricultural robotics, numerous control algorithms have been studied to achieve high-precision path tracking and have reached an accuracy within ±10 cm, whatever the ground configuration and the path to be followed. However, most existing approaches consider classical two-wheel-steering vehicles. Unfortunately, by using such a steering system, only the lateral deviation with respect to the path to be followed can be satisfactorily controlled. Indeed, the heading of the vehicle remains dependent on the grip conditions, and crabwise motions, for example, are systematically observed on a slippery slope, leading to inaccurate field operations. To tackle this drawback, a four-wheel-steering (4WS) mobile robot is considered, enabling servo of both lateral and angular deviations with respect to a desired trajectory. The path tracking control is designed using an extended kinematic representation, allowing account to be taken online of wheel skidding, while a backstepping approach permits management of the 4WS structure. The result is an approach taking advantage of both rear and front steering actuations to fully compensate for sliding effects during path tracking. Moreover, a predictive algorithm is developed in order to address delays induced by steering actuators, compensating for transient overshoots in curves. Experimental results demonstrate that despite sliding phenomena, the mobile robot is able to automatically and accurately achieve a desired path, with lateral and angular errors, respectively, within ±10 cm and ±2 deg, whatever its shape and whatever the terrain conditions. [source] Photodynamic Treatment of the Dermatophyte Trichophyton rubrum and its Microconidia with Porphyrin Photosensitizers,PHOTOCHEMISTRY & PHOTOBIOLOGY, Issue 2 2004Threes G. M. Smijs ABSTRACT The application of photosensitizers for the treatment of fungal infections is a new and promising development within the field of photodynamic treatment (PDT). Dermatophytes, fungi that can cause infections of the skin, hair and nails, are able to feed on keratin. Superficial mycoses are probably the most prevalent of infectious diseases in all parts of the world. One of the most important restrictions of the current therapeutic options is the return of the infection and the duration of the treatment. This is especially true in the case of infections of the nail (tinea unguium) caused by Trichophyton rubrum, an anthropophilic dermatophyte with a worldwide distribution. Recently, we demonstrated that 5,10,15-tris(4-methylpyridinium)-20-phenyl-[21H,23H]-porphine trichloride (Sylsens B) and deuteroporphyrin monomethylester were excellent photosensitizers toward T. rubrum when using broadband white light. This study demonstrates the photodynamic activity of these photosensitizers with red light toward both a suspension culture of T. rubrum and its isolated microconidia. The higher penetration depth of red light is important for the PDT of nail infections. In addition, we tested the photodynamic activity of a newly synthesized porphyrin, quinolino-[4,5,6,7-efg]-7-demethyl-8-deethylmesoporphyrin dimethylester, displaying a distinct peak in the red part of the spectrum. However, its photodynamic activity with red light toward a suspension culture of T. rubrum appeared to be only fungistatic. Sylsens B was the best photosensitizer toward both T. rubrum and its microconidia. A complete inactivation of the fungal spores and destruction of the fungal hyphae was found. In studies into the photostability, Sylsens B appeared to be photostable under the conditions used for fungal PDT. A promising result of this study is the demonstration of the complete degradation of the fungal hyphae in the time after the PDT and the inactivation of fungal spores, both with red light. These results offer the ingredients for a future treatment of fungal infections, including those of the nail. [source] Long-term after-effects of fertilisation on the restoration of calcareous grasslandsAPPLIED VEGETATION SCIENCE, Issue 2 2008N.A.C. Smits Question: What are the long-term implications of former fertilisation for the ecological restoration of calcareous grasslands? Location: Gerendal, Limburg, The Netherlands. Methods: In 1970, ten permanent plots were established in just abandoned agricultural calcareous grassland under a regime of annual mowing in August. From 1971 to 1979, two different fertiliser treatments were applied twice a year to a subset of the plots (artificial fertiliser with different proportions of nitrogen and phosphorus). The vegetation of the plots was recorded yearly and vegetation biomass samples were taken for peak standing crop and total amounts of nitrogen, phosphorus and potassium. Species composition and floristic diversity were analysed over the research period (1970,2006) and between the treatments, including the use of multivariate techniques (Detrended Correspondence Analysis). Results: In terms of species number, there is a clear optimum 10 to 20 years after fertilisation has been terminated. Afterwards, there is a slow decrease; no new species appear and species of more nutrient-rich conditions gradually disappear. For the fertilised plots that received a relatively high proportion of N, effects are found only in the first years, whereas, for the plots that received a relatively high proportion of P, long-term after-effects are found in species composition, peak standing crop, total amounts of phosphorus in biomass, and in soil phosphorus data. Conclusions: The effect of artificial fertiliser with a large amount of nitrogen disappears in less than ten years when mown in August, including removal of the hay. This is a promising result for restoration of N-enriched calcareous grasslands, as the applied dose of nitrogen in this experiment largely exceeds the extra input of nitrogen via atmospheric deposition. Application of fertiliser with a large amount of phosphorus, however, has effects even more than 25 years after the last addition. There are no prospects that this effect will become reduced in the near future under the current mowing management. [source] Experimental Study on the Molecular Dimension and Configuration of Polymer and Its Flow Characteristics from Electrolyte EffectCHINESE JOURNAL OF CHEMISTRY, Issue 4 2009Xiangguo LU Abstract Molecular clew dimension and configuration of polymer, and flow characteristics of polymer solution were studied from electrolyte effect, by making use of dynamic light scattering (DLS), scanning electron microscopy (SEM), apparent viscosity method and core flow experiment. It can be observed that with the electrolyte concentration increasing, there exists a variation trend of "decreasing, increasing and decreasing again" to the molecular clew dimension of the polymer. The compression action of Ca2+ or Mg2+ to the double electrode layer of polymer molecules is more powerful by comparison against Na+, which results in that Ca2+ and Mg2+ have a more extensive effect on the viscosity of polymer solution, and clew dimensions and their distribution. With the electrolyte concentration increasing, the polymer molecular configuration of multi-layer stereoscopic random reticulation transformed into a dendritic one. During the succeeding water flooding, the variation degree of injection pressure of core was mainly determined by the swelling extent of molecular clew of retained polymer and the produced amount of polymer. And the bigger the molecular weight of polymer is, the stronger the compression or swelling action of electrolyte to the molecule clews is, and the greater the increasing degree of injection pressure during succeeding water flooding is. The greater difference of electrolyte concentrations in used water between polymer flooding and succeeding water flooding can result in greater increasing degree of injection pressure during the succeeding water flooding. So, an advisable increasing in difference of electrolyte concentrations in used water between the polymer flooding and succeeding water flooding was proposed when designing the polymer flooding performance in oilfields, which has promising result for improving effect of polymer flooding. [source] Antiepileptogenesis and Seizure Prevention Trials with Antiepileptic Drugs: Meta-Analysis of Controlled TrialsEPILEPSIA, Issue 4 2001Nancy R. Temkin Summary: ,Purpose: To synthesize evidence concerning the effect of antiepileptic drugs (AEDs) for seizure prevention and to contrast their effectiveness for provoked versus unprovoked seizures. Methods: Medline, Embase, and The Cochrane Clinical Trials Register were the primary sources of trials, but all trials found were included. Minimal requirements: seizure-prevention outcome given as fraction of cases; AED or control assigned by random or quasi-random mechanism. Single abstracter. Aggregate relative risk and heterogeneity evaluated using Mantel,Haenszel analyses; random effects model used if heterogeneity was significant. Results: Forty-seven trials evaluated seven drugs or combinations for preventing seizures associated with fever, alcohol, malaria, perinatal asphyxia, contrast media, tumors, craniotomy, and traumatic brain injury. Effective: Phenobarbital for recurrence of febrile seizures [relative risk (RR), 0.51; 95% confidence interval (CI), 0.32,0.82) and cerebral malaria (RR, 0.36; CI, 0.23,0.56). Diazepam for contrast media,associated seizures (RR, 0.10; CI, 0.01,0.79). Phenytoin for provoked seizures after craniotomy or traumatic brain injury (craniotomy: RR, 0.42; CI, 0.25,0.71; TBI: RR, 0.33; CI, 0.19,0.59). Carbamazepine for provoked seizures after traumatic brain injury (RR, 0.39; CI, 0.17,0.92). Lorazepam for alcohol-related seizures (RR, 0.12; CI, 0.04,0.40). More than 25% reduction ruled out valproate for unprovoked seizures after traumatic brain injury (RR, 1.28; CI, 0.76,2.16), and carbamazepine for unprovoked seizures after craniotomy (RR, 1.30; CI, 0.75,2.25). Conclusions: Effective or promising results predominate for provoked (acute, symptomatic) seizures. For unprovoked (epileptic) seizures, no drug has been shown to be effective, and some have had a clinically important effect ruled out. [source] Permanganate Oxidation Revisited: Synthesis of 3-Deoxy-2-uloses via Indium-Mediated Chain Elongation of CarbohydratesEUROPEAN JOURNAL OF ORGANIC CHEMISTRY, Issue 25 2010Christoph Schmölzer Abstract Application of the Barbier-type indium-mediated allylation method to suitable substrates offers access to carbohydrates bearing a terminal olefin moiety. The C,C bond forming reaction generates a defined stereochemistry of the new chiral center and tolerates a wide variety of starting aldehydes thus allowing modifications in the carbohydrate backbone. Further transformations of the alkene moiety via an environmentally benign and subtle controlled protocol using potassium permanganate gives rise to the structural motif of 3-deoxy-2-uloses in good yields. The final part of the reaction sequence focuses on the deprotection of the acetyl groups essential for the success of the oxidation step. The acidic and labile 3-deoxy position of the target molecule is prone to elimination applying standard deacetylation conditions and therefore demands derivatisation of the molecule. The introduction of a thioketal moiety using microwave conditions shows promising results and subsequent standard transformations are applicable leading to the desired products. [source] Dilute Doping, Defects, and Ferromagnetism in Metal Oxide SystemsADVANCED MATERIALS, Issue 29 2010Satishchandra B. Ogale Over the past decade intensive research efforts have been carried out by researchers around the globe on exploring the effects of dilute doping of magnetic impurities on the physical properties of functional non-magnetic metal oxides such as TiO2 and ZnO. This effort is aimed at inducing spin functionality (magnetism, spin polarization) and thereby novel magneto-transport and magneto-optic effects in such oxides. After an early excitement and in spite of some very promising results reported in the literature, this field of diluted magnetic semiconducting oxides (DMSO) has continued to be dogged by concerns regarding uniformity of dopant incorporation, the possibilities of secondary ferromagnetic phases, and contamination issues. The rather sensitive dependence of magnetism of the DMSO systems on growth methods and conditions has led to interesting questions regarding the specific role played by defects in the attendant phenomena. Indeed, it has also led to the rapid re-emergence of the field of defect ferromagnetism. Many theoretical studies have contributed to the analysis of diverse experimental observations in this field and in some cases to the predictions of new systems and scenarios. In this review an attempt is made to capture the scope and spirit of this effort highlighting the successes, concerns, and questions. Ucan Abstract In this paper, in order to improve error performance, we introduce a new type of turbo codes, called ,multilevel-turbo codes (ML-TC)' and we evaluate their performance over wide-sense stationary uncorrelated scattering (WSSUS) multipath channels. The basic idea of ML-TC scheme is to partition a signal set into several levels and to encode each level separately by a proper component of the turbo encoder. In the considered structure, the parallel input data sequences are encoded by our multilevel scheme and mapped to any modulation type such as MPSK, MQAM, etc. Since WSSUS channels are very severe fading environments, it is needed to pass the received noisy signals through non-blind or blind equalizers before turbo decoders. In ML-TC schemes, noisy WSSUS corrupted signal sequence is first processed in equalizer block, then fed into the first level of turbo decoder and the first sequence is estimated from this first Turbo decoder. Subsequently, the other following input sequences of the frame are computed by using the estimated input bit streams of previous levels. Here, as a ML-TC example, 4PSK 2 level-turbo codes (2L-TC) is chosen and its error performance is evaluated in WSSUS channel modelled by COST 207 (Cooperation in the field of Science & Technology, Project #207). It is shown that 2L-TC signals with equalizer blocks exhibit considerable performance gains even at lower SNR values compared to 8PSK-turbo trellis coded modulation (TTCM). The simulation results of the proposed scheme have up to 5.5 dB coding gain compared to 8PSK-TTCM for all cases. It is interesting that after a constant SNR value, 2L-TC with blind equalizer has better error performance than non-blind filtered schemes. Malayeri Abstract Energy lost due to fouling of heat exchangers accounts for at least 2% of the total world energy production per year. The overwhelming proportion of these losses is compensated by additional consumption of fossil energy carriers. Not surprisingly, this comes with an enormous energy price-tag as well as considerable green-house gas emissions, acidification of water resources and release of chemical fouling inhibitors. Any solution towards the mitigation of fouling will, therefore, provide significant economic and environmental benefits. In the present paper, the performance of innovative nano-modified surfaces is described when subjected to calcium sulphate scale deposition during convective heat transfer. Two types of non-structured and structured nano-modified surfaces are examined. The experimental results demonstrate that such coatings will significantly increase the induction time before fouling starts and also reduce the subsequent fouling rate, in comparison with untreated stainless steel surfaces. Considering these promising results and the potential application of nanotechnology to combat fouling, the paper continues by discussing the demands on thermal and mechanical stabilities that such coatings will have to satisfy. The Lennard,Jones and torsional parameters were optimized to be consistent with the ABEEM/MM fluctuating charge electrostatic potential. The hydrogen bond was specially treated with an electrostatic fitting function. Molecular dipole moments, dimerization energies, and hydrogen bond lengths of complexes are reasonably achieved by our model, compared to ab initio results. The ABEEM/MM fluctuating charge model reproduces both the peptide conformational energies and structures with satisfactory accuracy with low computer cost. The transferability is tested by applying the parameters of our model to the tetrapeptide of alanine and another four dipeptides. The overall RMS deviations in conformational energies and key dihedral angles for four di- or tetrapeptide, is 0.39 kcal/mol and 7.7°. The current results agree well with those by the accurate ab initio method, and are comparable to those from the best existing force fields. The results make us believe that our fluctuating charge model can obtain more promising results in protein and macromolecular modeling with good accuracy but less computer cost. © 2005 Wiley Periodicals, Inc. BLOCK M.D. Mitral regurgitation (MR) associated with, ischemic, and degenerative (prolapse) disease, contributes to left ventricular (LV) dysfunction due to remodeling, and LV dilation, resulting in worsening of MR. Mitral valve (MV) surgical repair has provided improvement in survival, LV function and symptoms, especially when performed early. Surgical repair is complex, due to diverse etiologies and has significant complications. The Society for Thoracic Surgery database shows that operative mortality for a 1st repair is 2% and for re-do repair is 4 times that. Cardiopulmonary bypass and cardiac arrest are required. The attendant morbidity prolongs hospitalization and recovery. Alfieri simplified mitral repair using an edge-to-edge technique which subsequently has been shown to be effective for multiple etiologies of MR. The MV leaflers are typically brought together by a central suture producing a double orifice MV without stenosis. Umana reported that MR decreased from grade 3.6 +/,0.5 to0.8 +/,0.4 (P < 0.0001)and LV ejection fraction increased from 33 +/,13% to 45 +/,11%(P = 0.0156). In 121 patients, Maisano reported freedom from re-operation of 95 +/,4.8% with up to 6 year follow-up. Oz developed a MV "grasper" that is directly placed via a left ventriculotomy and coapts both leaflets which are then fastened by a graduated spiral screw. An in-vitro model using explanted human valves showed significant reduction in MR and in canine studies, animals followed by serial echo had persistent MV coaptation. At 12 weeks the device was endothelialized. These promising results have paved the way for a percutaneous or minimally invasive off pump mitral repair. Evalve has developed catheter-based technology, which, by apposing the edges of a regurgitant MV, results in edge-to-edge repair. Release of the device is done after echo and fluoroscopic evaluation under normal loading conditions. If the desired effect is not produced the device can be repositioned or retrieved. Animal studies show excellent healing, with incorporation of the device into the leaflets at 6,10 weeks with persistent coaptation. Another percutaneous approach has been to utilize the proximity of the coronary sinus (CS) to the mitral annulus (MA). Placement of a self-compressing device in the CS along the region of the posterior MA has, in canine models, reduced MR and addresses the issues of MA dilation and its contribution to MR. Cappendijk MD Abstract Purpose To investigate the potential difference in the size of the lipid-rich necrotic core (LRNC) in carotid plaques of symptomatic patients versus asymptomatic patients. Pathological studies established that a large LRNC is an important feature of vulnerable atherosclerotic plaque. Previously, we have demonstrated a high correlation between semiquantitative analysis of the LRNC size in T1-weighted (w) turbo field echo (TFE) MR images and histology. Materials and Methods Thirty-seven patients with carotid stenosis >70% with (n = 26) or without (n = 11) symptoms were included. Three independent MR readers quantified the amount of LRNC with a T1w TFE pulse sequence. The relative amount of LRNC (LRNC score) was defined as sum of cross-sectional area percentages LRNC per carotid plaque. Results Interreader agreement for the three MR readers was good, with an intraclass correlation coefficient (ICC, 95% confidence interval [CI]) of 0.72 (0.57,0.83). All three MR readers on average found a larger LRNC in the symptomatic group of patients, although this was not statistically significant. The mean LRNC score was 116 ± 129 and 59 ± 62 for symptomatic and asymptomatic patients, respectively (P = 0.13). Symptomatic patients showed wide ranges in LRNC scores (0,424), while the range was much lower in the asymptomatic group (0,170). Conclusion Single-sequence T1w TFE may be a promising technique to study atherosclerotic plaque at risk of stroke. Larger studies are warranted to confirm these promising results. J. Magn. Reson. Rezvani Background:, Since 1994, when naltrexone (Revia®) was approved by the FDA for the treatment of alcoholism, only 2 other drugs (Campral® and Topamax®) have been approved for alcoholism treatment. However, various experimental drugs, including antiepileptic medications, have been tested in both animal models and in humans with some promising results. The purpose of this project was to study the effect of the novel neuromodulator carisbamate, which is in development for epilepsy treatment, on alcohol intake in selectively bred alcohol-preferring rats. Methods:, Male alcohol-preferring inbred P rats were allowed to freely drink water or alcohol (10%, v/v) using a 2-bottle choice procedure. After stable baselines for alcohol and water intakes were established, the acute effects of oral carisbamate (0, 10, 30, 45, 60, and 90 mg/kg) were assessed. Then, the chronic effect of the compound (60 mg/kg/day for 14 consecutive days) on alcohol intake was assessed in a separate group of male P rats. In another set of experiments, the effects of carisbamate and naltrexone on alcohol withdrawal-induced elevated drinking of alcohol, an index of craving, were compared. Rats were withdrawn from alcohol for 24 hours and were given vehicle, 20 mg/kg naltrexone or 60 mg/kg carisbamate 30 minutes before re-exposure to alcohol. Alcohol and water intake was measured 6 hours after alcohol re-exposure. To determine the effects of carisbamate on saccharin preference, rats were put on a 2-bottle choice of water versus a solution of 2% saccharin. Then, the effect of the highest dose of carisbamate (90 mg/kg) and naltrexone (20 mg/kg) and the vehicle on saccharin preference was determined. Results:, Our results showed that there was a selective dose-dependent reduction in alcohol intake and preference in the alcohol-preferring P rat after an acute oral administration of carisbamate. There were no significant effects on food or water intake. Chronic administration of carisbamate significantly reduced alcohol intake and preference initially, but partial tolerance developed after the 10th treatment. The degree of tolerance development was less than that observed for naltrexone. Acute administration of carisbamate was more effective than naltrexone in reducing enhanced alcohol intake after a period of alcohol deprivation. Compared with control vehicle neither carisbamate nor naltrexone had a significant effect on saccharin intake and preference. Conclusion:, The novel neuromodulator compound carisbamate has a favorable profile of effects on alcohol intake and related measures and should be considered for testing on human alcoholics. Methods:, An expert panel, convened by the Centers for Disease Control and Prevention (CDC) in 2005, reviewed and provided expertise on an earlier version of this article. The panel comprised experts in public health, education, school counseling, school medical care, and a parent organization. This article describes the purposes of BMI measurement programs, examines current practices, reviews existing research, summarizes the recommendations of experts, identifies concerns, and provides guidance including a list of safeguards and ideas for future research. Results:, The implementation of school-based BMI measurement for surveillance purposes, that is, to identify the percentage of students in a population who are at risk for weight-related problems, is widely accepted; however, considerable controversy exists over BMI measurement for screening purposes, that is, to assess the weight status of individual students and provide this information to parents with guidance for action. Although some promising results have been reported, more evaluation is needed to determine whether BMI screening programs are a promising practice for addressing obesity. Conclusions:, Based on the available information, BMI screening meets some but not all of the criteria established by the American Academy of Pediatrics for determining whether screening for specific health conditions should be implemented in schools. Schools that initiate BMI measurement programs should evaluate the effects of the program on BMI results and on weight-related knowledge, attitudes, and behaviors of youth and their families; they also should adhere to safeguards to reduce the risk of harming students, have in place a safe and supportive environment for students of all body sizes, and implement science-based strategies to promote physical activity and healthy eating. All three give results that correlate significantly with the research productivity of the target institution. The directory range model, which is based upon summing the number of distinct directories targeted by each other university, produces the most promising results of any link metric yet. Based upon an analysis of changes between models, it is suggested that range models measure essentially the same quantity as their predecessors but are less susceptible to spurious causes of multiple links and are therefore more robust. Although victims were more satisfied than offenders for some outcomes, all participants reported exceptionally high satisfaction. [source] Radioimmunodiagnosis of lymph node metastases in head and neck cancerORAL DISEASES, Issue 5 2003R de Bree Introduction:, Reliable staging of the neck remains a diagnostic challenge in head and neck squamous cell carcinoma (HNSCC) patients. Monoclonal antibodies (MAbs) directed against tumour-associated antigens can be used for selective tumour targeting. When labelled with a , -emitting radionuclide like 99mTechnetium, such MAbs can be used for tumour detection by radioimmunoscintigraphy (RIS). Objective:, The aim of this study was to assess the potential of RIS for the detection of lymph node metastases in HNSCC patients. Patients and methods:, In 49 patients with HNSCC, who were scheduled to undergo surgery including neck dissection, RIS using 99mTc-labelled squamous cell specific MAb E48 or U36 administered intravenously was compared with clinical palpation, computed tomography (CT), magnetic resonance imaging (MRI) and histopathological outcome. Results:, RIS detected lymph node metastases in 35 of 51 positive sides (sensitivity 69%). Interpretation of RIS was correct in 47 of 65 sides (accuracy 72%). Accuracy of palpation, CT and MRI were comparable. Immunohistochemical staining of lymph node metastases missed by RIS showed that the injected MAb had targeted these small tumour deposits but these were not visualized. Conclusions:, RIS at its current stage of development is not superior to CT or MRI for the detection of lymph node metastases. As small tumour deposits were probably not visualized because of the limited sensitivity and/or spatial resolution of the gamma camera, positron emission tomography (PET) using MAbs labelled with positron emitters may improve the detection. As MAb-PET studies in an animal model showed promising results we will soon start a clinical MAb-PET study. Cardiac stem cell transplantation has become a promising therapy to treat heart failure in adult patients. Based on these promising results, the cardiac stem cell therapy might also represent a new therapeutic option particularly in young children. The present case documents for the first time intracoronary administration of autologous bone marrow-derived progenitor cells in a critically ill two-yr-old child with severe heart failure caused by DCM. Because of progressive worsening of the clinical condition despite maximal anticongestive treatment, the decision to perform autologous stem cell therapy was made. Cardiac stem cell therapy proved to be technically feasible, was associated with improvement in cardiac function, and might represent an option before heart transplantation in children with severe heart failure. Buso Abstract The degree of heterosis for total tuber yield (TTY) and total solids (TS) in 4x-2x crosses was estimated by comparing the performance of 12 families with their respective parents in two locations in Wisconsin (USA). The parental 2x clones were Phureja-haploid Tuberosum hybrids with 2n -pollen production by first-division restitution. The general combining ability (GCA) and specific combining ability (SCA) were estimated for TTY, TS, vine maturity (VM), length of tuber sprout dormancy (LD), and tuber eye depth (ED). Family performance for TTY ranged from 74 to 146% at Hancock (E#1) and from 77 to 287 at Rhinelander (E#2) when compared with that of the 4x parent group. For VM, the families were late maturing, but a few precocious ones were identified. For TS, the families had heterosis of 5.1% over the 4x parent group. The families had slightly higher ED values than the 4x parents, but families with values within the commercial range were identified. The family average for LD (54 days) was closer to the 2x group (51 days) than to the 4x group (88 days). The direction and magnitude of the parent-family relationships were variable. The 4x parent TTY was correlated with progeny in E#1 but not E#2. The 2x parent VM had correlation with the offspring at E#2 but not at E#1. The type of gene action had a trait-specific expression. Significant SCA and GCA variances were observed, suggesting that additive as well as non-additive genetic effects were operating. The 4x-2x crosses were able to generate heterotic families for TTY and TS in combination with other useful traits. However, no promising results were found for LD because of the apparent dominance of the short-dormancy phenotype. This result indicates the need of additional selection and breeding efforts for some specific traits when using S. phureja -derived germplasm. Nanostructured coatings have shown most promising results in this connection. High oxidation resistance, hot hardness, and loW friction are just a few benefits that these coatings offer. The deposition of nanostructured coatings is only possible within a small deposition process window. Most cutting tool surfaces are complex shaped and include, for instance, small corner radii at the cutting edge or chip breakers. The local process window and the deposition parameters must be adapted to the actual shape of the cutting tools in order to obtain a hard nanocomposite coating with adequate adhesion properties.
