Selected AbstractsFungal tyrosinases: new prospects in molecular characteristics, bioengineering and biotechnological applicationsJOURNAL OF APPLIED MICROBIOLOGY, Issue 2 2006S. Halaouli Abstract Tyrosinases are type-3 copper proteins involved in the initial step of melanin synthesis. These enzymes catalyse both the o -hydroxylation of monophenols and the subsequent oxidation of the resulting o -diphenols into reactive o -quinones, which evolve spontaneously to produce intermediates, which associate in dark brown pigments. In fungi, tyrosinases are generally associated with the formation and stability of spores, in defence and virulence mechanisms, and in browning and pigmentation. First characterized from the edible mushroom Agaricus bisporus because of undesirable enzymatic browning problems during postharvest storage, tyrosinases were found, more recently, in several other fungi with relevant insights into molecular and genetic characteristics and into reaction mechanisms, highlighting their very promising properties for biotechnological applications. The limit of these applications remains in the fact that native fungal tyrosinases are generally intracellular and produced in low quantity. This review compiles the recent data on biochemical and molecular properties of fungal tyrosinases, underlining their importance in the biotechnological use of these enzymes. Next, their most promising applications in food, pharmaceutical and environmental fields are presented and the bioengineering approaches used for the development of tyrosinase-overproducing fungal strains are discussed. [source] HEALTH BENEFITS OF APPLE PHENOLICS FROM POSTHARVEST STAGES FOR POTENTIAL TYPE 2 DIABETES MANAGEMENT USING IN VITRO MODELSJOURNAL OF FOOD BIOCHEMISTRY, Issue 1 2010I. ADYANTHAYA ABSTRACT An increasing number of studies indicate that regular intake of fruits and vegetables have clear links to reduced risk of chronic diseases like diabetes and cardiovascular disease. The beneficial effects in many cases have been attributed to the phenolic and antioxidant content of the fruits and vegetables. Apples are a major source of fiber and contain good dietary phenolics with antioxidant function. Previous epidemiological studies have indicated that intake of apples reduces the risk of developing type 2 diabetes. Our studies indicate that this reduced risk is potentially because of the modulation of postprandial glucose increase by phenolics present in apples via inhibition of, -glucosidase. Phenolic content was evaluated during 3 months of postharvest storage of four varieties of apples and results indicated positive linkage to enhanced postharvest preservation and, -glucosidase inhibition. These in vitro results along with existing epidemiological studies provide strong biochemical rationale for further animal or human clinical studies. PRACTICAL APPLICATIONS The understanding of phenolic-linked antioxidant enzyme responses during postharvest storage of apples has implications for using the same phenolic functional ingredients toward health benefits such as ,-glucosidase inhibition linked to glycemic index control associated with type 2 diabetes. Therefore strategies to understand phenolic-linked postharvest preservation and natural treatments to extend this preservation in selected varieties, such as McIntosh and Cortland in this study, can be basis for food ingredient design for health benefits. These strategies can then be extended to prolong postharvest preservation and enhance phenolic linked human health benefits of a wide variety of fruits and vegetables. [source] DEFINITION OF INTERNAL MORPHOLOGY AND STRUCTURAL CHANGES DUE TO DEHYDRATION OF RADISH (RAPHANUS SATIVUS L. CV. SUPRELLA) USING MAGNETIC RESONANCE IMAGING SPECTROSCOPYJOURNAL OF FOOD QUALITY, Issue 5-6 2005ANNA SALERNO ABSTRACT Magnetic resonance imaging (MRI) spectroscopy is a promising nondestructive analytical technique in food science. It offers the unique opportunity of studying vegetables, fruits and other foods in general, in their wholeness without any preparative manipulation of the sample. The aim of this study was to investigate the internal structure of radish and to monitor the variations induced by postharvest storage at low relative humidity. The MRI allowed for a clear definition of the internal structure of radishes with distinct visibility of xylematic and phloematic vessels distributed in a radial way. A decrease in water content, which results in the breakdown of tissues and the formation of large cavities with the detachment of the external cortex, is the main consequence of a few days' storage in low relative humidity. Both of these are factors that drastically decrease the quality of the radish's fleshy root. The MRI images give a novel insight into the internal organization of the hypocotyl, and this offers opportunities for further studies with regard to the structural differences related to the cultivars as well as the cultivation system. [source] Hydrogen Peroxide and Calcium Chloride Added to Irrigation Water as a Strategy to Reduce Bacterial Populations and Improve Quality of Fresh MushroomsJOURNAL OF FOOD SCIENCE, Issue 6 2005Naveen Chikthimmah ABSTRACT The quality and value of fresh mushrooms are often diminished by the presence of high bacterial populations that cause a brown, blotchy appearance. The objective of the present research was to evaluate the addition of hydrogen peroxide and/or calcium chloride to irrigation water as a means to reduce total bacterial populations on fresh mushrooms. Crops were grown using commercial mushroom growing practices except for the addition of 0.75% hydrogen peroxide and/or 0.3% calcium chloride irrigation water added to the crop starting 11 d after the casing layer was applied on top of mushroom compost. Irrigation water without the added treatments acted as the control. Mushrooms were aseptically sampled from the production beds for enumerating bacterial counts. Total aerobic bacterial populations were determined by standard microbiological plating procedures. Mushroom whiteness (L -value) and color (delta E) after harvest and postharvest storage were measured using a Minolta chromameter. Harvested mushrooms were separated by treatment and weighed to record yield. Mushrooms irrigated with water (control) had 7.3 log colony-forming units (CFU) of aerobic bacterial populations per gram of fresh mushroom tissue. Compared with the control, irrigation with 0.75% hydrogen peroxide and 0.3% calcium chloride reduced the bacterial populations on fresh mushrooms by 87% (6.4 log CFU/g). Irrigation with hydrogen peroxide and calcium chloride significantly enhanced mushroom whiteness after harvest as well as after 6 d of postharvest storage at 12 °C. The irrigation treatments did not have a significant effect on crop yields; hence, the addition of hydrogen peroxide and calcium chloride to irrigation water was demonstrated to have good potential as a practical strategy to reduce bacterial populations and to improve the quality of fresh mushrooms. [source] Mechanical behaviour and quality traits of highbush blueberry during postharvest storageJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 6 2009Valentina Chiabrando Abstract BACKGROUND: Berry firmness is one of the most important characteristics for fresh market consumption. Blueberry firmness is also an important attribute because it is considered to be a measure of its freshness. Berries lose their firmness by loss of water and by changes in their structure. RESULTS: The postharvest life of two highbush blueberry cultivars (Bluecrop and Coville) was investigated. Several parameters related to blueberry quality were evaluated during the postharvest storage period. To assess berry texture characteristics (firmness, hardness, cohesiveness, gumminess, chewiness, springiness, resilience), a rapid non-destructive penetrometer test by Durofel® and texture profile analysis (TPA) using a texture analyser were carried out. Low temperature inhibited the decrease of total soluble solids, total titratable acidity and increase of flesh pH value, thereby maintaining good taste quality. There was an increase in fruit firmness (Durofel index) and hardness and a decrease in chewiness and springiness during storage. CONCLUSION: There was a significant correlation among the TPA parameters and Durofel index. The Durofel index could therefore be used as a suitable indicator of fruit quality and storability, and low-temperature storage is beneficial to maintain the taste quality of blueberry fruit after harvest. Copyright © 2009 Society of Chemical Industry [source] Use of alginate or zein as edible coatings to delay postharvest ripening process and to maintain tomato (Solanum lycopersicon Mill) qualityJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 7 2008Pedro Javier Zapata Abstract BACKGROUND: Edible coatings could be effective tools for delaying the ripening process of fruits. Alginate or zein as edible coatings were assayed in tomato in order to maintain parameters related to quality during postharvest storage. RESULTS: Coated tomatoes showed lower respiration rate and ethylene production than control ones, with a twofold lower concentration of ethylene precursor. In addition, the evolution of parameters related to tomato quality losses, such as softening, colour evolution and weight loss, was significantly delayed (4,6 days on average) in coated tomatoes as compared to controls. Thereafter, sugars, organic acids (and especially ascorbic acid) and scores from sensory analysis remained at much higher levels at the end of storage in treated than in control tomatoes. CONCLUSIONS: Coatings based on alginate or zein could be effective tools for delaying the tomato-ripening process during postharvest storage, and in turn maintaining tomato quality and its acceptability by consumers. [source] Influence of growth stage and postharvest storage on ascorbic acid and carotenoid content and visual quality of baby spinach (Spinacia oleracea L.)JOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 3 2006Sara ÅM Bergquist Abstract To investigate the variations in quality with growth stage and postharvest storage, spinach was sown on three occasions. For each occasion, the spinach was harvested at three growth stages at 6-day intervals. The second stage corresponded to a growth period used for baby spinach by commercial growers. After harvest, the leaves were stored in polypropylene bags at 2 °C or 10 °C. The highest ascorbic acid content in fresh material was found at stage I. During storage, the ascorbic acid content decreased considerably and the dehydroascorbic acid/vitamin C ratio increased. Storage at 2 °C gave a smaller reduction in ascorbic acid content than storage at 10 °C. Total carotenoid content increased or remained stable during storage. Lutein was the major carotenoid, making up about 39% of the total carotenoid content, followed by violaxanthin, ,-carotene and neoxanthin. Visual quality decreased during storage in most cases, and was correlated to initial ascorbic acid and dry matter contents. The initial AA content might therefore be used as a parameter for predicting the shelf-life of baby spinach. The results also indicate that by harvesting baby spinach a few days earlier than the current commercial stage of harvest the postharvest visual quality and nutritional quality may be improved. Copyright © 2005 Society of Chemical Industry [source] Cell wall phenolics of white and green asparagusJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 6 2005Rocío Rodríguez Abstract Cinnamic acids influence physical and textural attributes of plant foods as they play an important role in lignification and cross-linking of cell-wall polymers. Several trans -cinnamic acids have been detected in asparagus cell wall (CW), and, of these, amounts of ferulic acid (FA) and its dehydrodimers have been found to increase significantly during post-harvest storage. The distribution of these compounds along the different sections of white and green spears, as well as their modifications during postharvest storage, has been investigated. It has been observed that the cell walls from the apical, middle and lower sections of the green spears contain equivalent amounts of FA derivatives (about 400 µg g,1 CW), while these compounds are mainly located in the middle (681 µg g,1 CW) and lower section (975 µg g,1 CW) of the white ones. During asparagus post-harvest storage a general increase of FA monomers and dimers took place that affected every section of both green and white spears. Major changes occurred in the middle and lower sections of the white asparagus, where the amounts of ferulic derivatives increased to 1700 and 1678 µg g,1 CW after storage. A similar but less pronounced trend was observed for the green spears. Copyright © 2005 Society of Chemical Industry [source] Effect of preharvest sprays containing calcium, magnesium and titanium on the quality of peaches and nectarines at harvest and during postharvest storageJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 11 2004María Serrano Abstract Peach and nectarine trees were foliar sprayed with a formulation containing Ca2+, Mg2+ and Ti4+. Parameters related to fruit quality were evaluated at harvest, after 7,28 days of cold storage and after subsequent ripening for 4 days at 20 °C (shelf life). At harvest, treated fruits from both cultivars had higher weight and pulp firmness than control fruits, while no effect was observed for either colour, total soluble solid content (TSS) and titratable acidity (TA) or the time required to ripen on the tree. During cold storage, lower levels of weight loss, colour evolution, TSS/TA ratio and ethylene production and higher pulp firmness were found in treated peaches and nectarines compared with control fruits. Also, the storability of treated fruits was extended for up to 14 days more than that of control fruits. During ripening at 20 °C after different periods of cold storage, parameters related to ripening evolved faster in control than in treated fruits. Also, the occurrence of the climacteric peak of ethylene production was delayed and its intensity was lower in treated peaches and nectarines than in controls. This method is sensitive, rapid and inexpensive and has been used to highlight the increases in sucrose loss following root stresses such as freezing or aggressive harvesting. Sugar beet roots lose 12.5% of their sucrose following an episode of impact damage greater than 2,J, rising to 19.7% loss after 8,J, with concurrent increases in glucose and fructose.
