Selected AbstractsA regional impact assessment of climate and land-use change on alpine vegetationJOURNAL OF BIOGEOGRAPHY, Issue 3 2003Thomas Dirnböck Abstract Aim, Assessing potential response of alpine plant species distribution to different future climatic and land-use scenarios. Location, Four mountain ranges totalling 150 km2 in the north-eastern Calcareous Alps of Austria. Methods, Ordinal regression models of eighty-five alpine plant species based on environmental constraints and land use determining their abundance. Site conditions are simulated spatially using a GIS, a Digital Terrain Model, meteorological station data and existing maps. Additionally, historical records were investigated to derive data on time spans since pastures were abandoned. This was then used to assess land-use impacts on vegetation patterns in combination with climatic changes. Results, A regionalized GCM scenario for 2050 (+ 0.65 °C, ,30 mm August precipitation) will only lead to local loss of potential habitat for alpine plant species. More profound changes (+ 2 °C, ,30 mm August precipitation; + 2 °C, ,60 mm August precipitation) however, will bring about a severe contraction of the alpine, non-forest zone, because of range expansion of the treeline conifer Pinus mugo Turra and many alpine species will loose major parts of their habitat. Precipitation change significantly influences predicted future habitat patterns, mostly by enhancing the general trend. Maintenance of summer pastures facilitates the persistence of alpine plant species by providing refuges, but existing pastures are too small in the area to effectively prevent the regional extinction risk of alpine plant species. Main conclusions, The results support earlier hypotheses that alpine plant species on mountain ranges with restricted habitat availability above the treeline will experience severe fragmentation and habitat loss, but only if the mean annual temperature increases by 2 °C or more. Even in temperate alpine regions it is important to consider precipitation in addition to temperature when climate impacts are to be assessed. The maintenance of large summer farms may contribute to preventing the expected loss of non-forest habitats for alpine plant species. Conceptual and technical shortcomings of static equilibrium modelling limit the mechanistic understanding of the processes involved. [source] Expression of retinoid receptors in sebaceous cell carcinomaJOURNAL OF CUTANEOUS PATHOLOGY, Issue 1 2006Nitin Chakravarti Purpose:, The aim of this study is to investigate whether there are any abnormalities in the in vivo expression of retinoid acid receptors (RAR-,, RAR-, and RAR-,) and retinoid X receptors (RXR-,, RXR-, and RXR-,) in sebaceous cell carcinoma. Methods:, Expression of retinoid receptors in paired specimens of cancerous tissues (n = 10) and adjacent normal tissues (n = 10) from 10 patients with sebaceous cell carcinoma was studied immunohistochemically by using anti-retinoid receptor antibodies. Results:, In eight of the 10 normal tissue samples, all six receptors were expressed. In the other two samples, all receptors were expressed except RAR-, (one sample) or RXR-, (two samples). Five tumours (50%) lacked RAR-,; RAR-, expression was lower in tumours than in normal tissues in eight of 10 cases. RAR-, was expressed in the cytoplasm of nine of 10 tumours; RAR-, expression was at least as high in tumours as in normal tissue in eight of 10 cases. Two tumours lacked RAR-,; three tumours had lower RAR-, expression than paired normal epithelium; four had the same RAR-, expression, and one had higher RAR-, expression. RXR-, expression was strong in all normal tissues and tumour samples. Ten tumours lacked RXR-, and all 10 tumours lacked RXR-, expression. Conclusions:, Diminished RXR-, and RXR-, expression might be related to the development of sebaceous cell carcinoma. Additional studies are required to establish whether the defects in RAR expression in sebaceous cell carcinoma might affect the potential response of this tumour to treatment with retinoids. [source] Early signaling through the Arabidopsis pattern recognition receptors FLS2 and EFR involves Ca2+ -associated opening of plasma membrane anion channelsTHE PLANT JOURNAL, Issue 3 2010Elena Jeworutzki Summary The perception of microbes by plants involves highly conserved molecular signatures that are absent from the host and that are collectively referred to as microbe-associated molecular patterns (MAMPs). The Arabidopsis pattern recognition receptors FLAGELLIN-SENSING 2 (FLS2) and EF-Tu receptor (EFR) represent genetically well studied paradigms that mediate defense against bacterial pathogens. Stimulation of these receptors through their cognate ligands, bacterial flagellin or bacterial elongation factor Tu, leads to a defense response and ultimately to increased resistance. However, little is known about the early signaling pathway of these receptors. Here, we characterize this early response in situ, using an electrophysiological approach. In line with a release of negatively charged molecules, voltage recordings of microelectrode-impaled mesophyll cells and root hairs of Col-0 Arabidopsis plants revealed rapid, dose-dependent membrane potential depolarizations in response to either flg22 or elf18. Using ion-selective microelectrodes, pronounced anion currents were recorded upon application of flg22 and elf18, indicating that the signaling cascades initiated by each of the two receptors converge on the same plasma membrane ion channels. Combined calcium imaging and electrophysiological measurements revealed that the depolarization was superimposed by an increase in cytosolic calcium that was indispensable for depolarization. NADPH oxidase mutants were still depolarized upon elicitor stimulation, suggesting a reactive oxygen species-independent membrane potential response. Furthermore, electrical signaling in response to either flg22 or elf 18 critically depends on the activity of the FLS2-associated receptor-like kinase BAK1, suggesting that activation of FLS2 and EFR lead to BAK1-dependent, calcium-associated plasma membrane anion channel opening as an initial step in the pathogen defense pathway. [source] Restoring landscapes: the authenticity problemEARTH SURFACE PROCESSES AND LANDFORMS, Issue 13 2006Isis Brook Abstract Philosophical concerns about restoring landscapes often revolve around two, connected, issues. First is the idea that a restored landscape, even if it is a perfect replica, has lost some of its value. The claim might appeal to a break in the continuity of the landscape and that continuity is part of what is valuable. Alternatively, often in the case of natural landscapes, the appeal is that any human manipulation is inauthentic; here the analogy is sometimes made with the art world and the restoration is deemed a fake. The second problem highlighted in philosophical debates is that the greater the success of restoration projects, the more threatened natural landscapes become: any claim that something must be preserved in its pristine or historically layered state is undermined by the claim that it could be put back again. Initially I discuss two opposing potential responses to these claims: (1) that humans are part of nature and thus cannot be an alien dominating force outside of nature; and (2) that nature is itself a social construct. Neither of these positions is entirely satisfactory, but what they jointly reveal is the reality of our fluid and multifaceted relationship with the world. I then show that Elliot's claim of the additional value of pristine nature is actually not based on an inherent value, but is dependent on the human valuation of it. I propose an alternative that places the source of value in the thing itself and thus arrive at a positive role for restoration as the setting in train and guiding of positive relationships above and beyond their social or public amenity value to us. [source] Interaction between genioglossus and diaphragm responses to transcranial magnetic stimulation in awake humansEXPERIMENTAL PHYSIOLOGY, Issue 4 2007Wei Wang The modulation of activity of the upper airway dilator and respiratory muscles plays a key role in the regulation of ventilation, but little is known about the link between their neuromuscular activation processes in vivo. This study investigated genioglossus and diaphragm responses to transcranial magnetic stimulation applied in different facilitatory conditions. The amplitude and latency of motor-evoked potential responses and the stimulation intensity threshold leading to a motor response (motor threshold) were recorded with stimulation applied at the vertex and anterolateral area in 13 awake normal subjects. Stimuli were applied during inspiration with and without resistance, during expiration with and without maximal tongue protrusion and during deep inspiration. In each stimulation location and condition, no diaphragmatic response was obtained without previous genioglossus activity (diaphragmatic and genioglossus responses latencies during expiration: 18.1 ± 2.9 and 6.3 ± 2.6 ms, respectively, mean ±s.d., P < 0.01). Genioglossus motor-evoked potential amplitude, latency and motor threshold were significantly modified with tongue protrusion with a maximal effect observed for stimulation in the anterolateral area. Deep inspiration was associated with a significant facilitatory effect on both genioglossus and diaphragm motor responses. The facilitatory effects of respiratory and non-respiratory manoeuvres were also observed during focal stimulation where isolated genioglossus responses were observed. Genioglossus and diaphragm differed in their motor threshold both at baseline and following facilitatory manoeuvres. Conclusions: (1) transcranial magnetic stimulation-induced genioglossus response systematically precedes that of diaphragm; (2) this sequence of activation is not modified by respiratory and non-respiratory manoeuvres; and (3) the genioglossus and diaphragm are differently influenced by these manoeuvres in terms of latency of the motor response and of motor threshold. [source] Macroecology, global change and the shadow of forgotten ancestorsGLOBAL ECOLOGY, Issue 1 2008José Alexandre Felizola Diniz-Filho ABSTRACT Many recent studies have evaluated how global changes will affect biodiversity, and have mainly focused on how to develop conservation strategies to avoid, or at least minimize, extinctions due to shifts in suitable habitats for the species. However, these complex potential responses might be in part structured in phylogeny, because of the macroecological traits underlying them. In this comment, we review recent analytical developments in phylogenetic comparative methods that can be used to understand patterns of trait changes under environmental change. We focus on a partial regression approach that allows for partitioning the variance of traits into a fraction attributed to a pure ecological component, a fraction attributed to phylogenetically structured environmental variation (niche conservatism) and a fraction that may be attributed to phylogenetic effects only. We then develop a novel interpretation for linking these components for multiple traits with potential responses of species to global environmental change (i.e. adaptation, range shifts or extinctions). We hope that this interpretation will stimulate further research linking evolutionary components of multiple traits with broad-scale environmental changes. [source] Evidence for an endothelium-derived hyperpolarizing factor in the superior mesenteric artery from rats with cirrhosisHEPATOLOGY, Issue 5 2000Eric Barriere In cirrhosis, in splanchnic arteries, endothelium-dependent relaxation may persist even if overactive nitric oxide synthase (NOS) and cyclooxygenase (COX) are inhibited. In normal arteries, a significant endothelium-dependent relaxation to acetylcholine persists after NOS/COX inhibition. This relaxation is caused by smooth muscle cell (SMC) membrane hyperpolarization, which is sensitive to a combination of the potassium channel blockers apamin and charybdotoxin, and is mediated by an endothelium-derived hyperpolarizing factor (EDHF). The aim of this study was to detect EDHF and evaluate its pathophysiologic role in isolated superior mesenteric arteries from cirrhotic rats. Arterial rings were obtained and exposed to Nw -nitro-L-arginine (L-NNA, a NOS inhibitor) and indomethacin (a COX inhibitor). Acetylcholine-induced membrane potential responses and concentration-response curves to the relaxant of acetylcholine were obtained with and without apamin plus charybdotoxin. Acetylcholine-induced responses were measured in certain rings from endothelium-denuded arteries. Contractions caused by the ,1 -adrenoceptor agonist phenylephrine were obtained in cirrhotic and normal rings with and without apamin and charybdotoxin. Significant acetylcholine-induced, endothelium-dependent, apamin- and charybdotoxin-sensitive, SMC membrane hyperpolarization and relaxation were found. An apamin- and charybdotoxin-sensitive hyporesponsiveness to the contractile action of phenylephrine was found in cirrhotic rings. In conclusion, in cirrhotic rats, in the superior mesenteric artery exposed to NOS/COX-inhibitors, an EDHF exists that may replace NOS/COX products to induce endothelium-dependent arterial relaxation. [source] Hierarchical model of the population dynamics of hippocampal dentate granule cellsHIPPOCAMPUS, Issue 5 2002G.A. Chauvet Abstract A hierarchical modeling approach is used as the basis for a mathematical representation of the population activity of hippocampal dentate granule cells. Using neural field equations, the variation in time and space of dentate granule cell activity is derived from the summed synaptic potential and summed action potential responses of a population of granule cells evoked by monosynaptic excitatory input from entorhinal cortical afferents. In this formulation of the problem, we have considered a two-level hierarchy: the synapses of entorhinal cortical axons define the first level of organization, and dentate granule cells, which include these synapses, define the second, higher level of organization. The model is specified by two state field variables, for membrane potential and for synaptic efficacy, respectively, with both evolving according to different time scales. The two state field variables introduce new parameters, physiological and anatomical, which characterize the dentate from the point of view of neuronal and synaptic populations: (1) a set of geometrical constraints corresponding to the morphological properties of granule cells and anatomical characteristics of entorhinal-dentate connections; and (2) a set of neuronal parameters corresponding to physiological mechanisms. Assuming no interaction between granule cells, i.e., neither ephaptic nor synaptic coupling, the model is shown to be mathematically tractable and allows solution of the field equations leading to the determination of activity. This treatment leads to the definition of two state variables, volume of stimulated synapses and firing time, which describe observed activity. Numerical simulations are used to investigate the populational characterization of the dentate by individual parameters: (1) the relationship between the conditions of stimulation of active perforant path fibers, e.g., stimulating intensity, and activity in the granule cell layer; and (2) the influence of geometry on the generation of activity, i.e., the influence of neuron density and synaptic density-connectivity. [source] Cholinergic modulation of synaptic physiology in deep layer entorhinal cortex of the ratJOURNAL OF NEUROSCIENCE RESEARCH, Issue 1 2001Mi Young Cheong Abstract We have recently shown that cholinergic effects on synaptic transmission and plasticity in the superficial (II/III) layers of the rat medial entorhinal cortex (EC) are similar, but not identical, to those in the hippocampus (Yun et al. [2000] Neuroscience 97:671,676). Because the superficial and deep layers of the EC preferentially convey afferent and efferent hippocampal projections, respectively, it is of interest to compare cholinergic effects between the two regions. We therefore investigated the physiological effects of cholinergic agents in the layer V of medial EC slices under experimental conditions identical to those in the previous study. Bath application of carbachol (0.5 ,M) induced transient depression of field potential responses in all cases tested (30 of 30; 18.5% ± 2.3%) and rarely induced long-lasting potentiation (only 3 of 30; 20.4% ± 3.2% in successful cases). At 5 ,M, carbachol induced transient depression only (20 of 20, 48.9% ± 2.8%), which was blocked by atropine (10 ,M). Paired-pulse facilitation was enhanced during carbachol-induced depression, suggesting presynaptic action of carbachol. Long-term potentiation (LTP) could be induced in the presence of 10 ,M atropine by theta burst stimulation, but its magnitude was significantly lower (9.1% ± 4.7%, n = 15) compared to LTP in control slices (22.4% ± 3.9%, n = 20). These results, combined with our previous findings, demonstrate remarkably similar cholinergic modulation of synaptic transmission and plasticity across the superficial and deep layers of EC. J. [source] RESTRAINT ON REASONS AND REASONS FOR RESTRAINT: A PROBLEM FOR RAWLS' IDEAL OF PUBLIC REASONPACIFIC PHILOSOPHICAL QUARTERLY, Issue 1 2006MICAH LOTT I will argue, however, that in certain cases Rawls' ideal of public reason is unable to provide a person with good reason for exercising such restraint, even if the person is already committed to Rawls' ideal of public reason. Because it is plausible to believe that such cases are widespread, the issue I am raising represents a serious problem for Rawls' account of public reason. After posing this problem, I consider potential responses on behalf of Rawls' view, and I reply to those responses. The moral of this story, as I see it, is that the kind of duty an ideal of public reason aims to place on citizens must be more modest than Rawls supposes. [source] When Changing from Merit Pay to Variable/Bonus Pay: What Do Employees Want?PERFORMANCE IMPROVEMENT QUARTERLY, Issue 4 2004Jeremy B. Fox ABSTRACT This study examines potential responses to a change in an employee reward system from permanent merit pay increases to one-time bonus payments. Removing long term risks associated with escalating pay is an increasingly common compensation strategy. Often overlooked, however, are employee perceptions of reward fairness under such conditions of change. Receiving lump sum payments in lieu of permanent merit pay increase may de-motivate employees. There has been little or no research conducted on this topic. In this study, using samples of practicing HR managers and university students, an equity questionnaire gathered data on the perceived equivalence between a permanent merit pay increase and what might be demanded by employees as a single payment in its replacement. An analysis of the data collected indicate an approximate 1:2 ratio is needed, such that a proposed lump-sum payment of $2400 would be perceived as a fair replacement for a permanent merit pay allocation of $1200 per year. Our research indicates that this 1:2 ratio holds for both high and low job satisfaction levels. [source] Thermal characterisation of active layer across a soil moisture gradient in the McMurdo Dry Valleys, AntarcticaPERMAFROST AND PERIGLACIAL PROCESSES, Issue 1 2009Scott J. Ikard Abstract Heat transport into active layer soils is important to understanding potential responses to changes in surface energy balance, particularly in the context of changing climate. Here we present results of a study to characterise soil thermal properties along a soil moisture gradient adjacent to Lake Fryxell in Taylor Valley, Antarctica. Our goals were to characterise the thermal characteristics of these relatively wet soils (compared to the rest of the McMurdo Dry Valleys landscape), and to assess the response of the active layer to possible increases in soil moisture. We measured subsurface temperatures at depths from 3 to 50,cm at four locations along a natural gradient of wet to dry soils adjacent to Lake Fryxell from January 2006 to January 2007. We used a numerical model to estimate apparent thermal diffusivity (ATD) and simulate observed temperature time series. Calculations of ATD at discrete locations yielded values ranging from 1.0,×,10,9 , 2.4,×,10,5,m2,s,1, and the corresponding range of bulk (i.e. depth averaged at a single surface location) ATD was 2.9,×,10,9,1.2,×,10,7,m2,s,1. Thawed soils had a range of bulk ATD during warming of 2.9,×,10,9,3.8,×,10,8,m2,s,1, and during cooling of 2.9,×,10,9,4.8,×,10,8,m2,s,1. When soils were frozen, however, the range of bulk ATD was 7.6,×,10,9,1.2,×,10,7,m2,s,1 during warming, and 7.8,×,10,9,1.1,×,10,7,m2,s,1 during cooling. Estimated bulk ATD values were consistently greater in locations of enhanced soil moisture, so lakeside soils were more likely to conduct energy into the subsurface. Apparao Abstract Pre- and postnatal developmental studies of the lung have provided compelling evidence demonstrating multiple factors that orchestrate alveolar epithelial cell differentiation. The extent to which reactivation of certain developmental pathways in the adult might influence the course of differentiation of alveolar type 2 cells (AT2) into AT1 cells is not known. In this study, we examined selected members of the forkhead (Fox) family of transcription factors and the Wnt (wingless) family of signaling proteins for expression during human alveolar cell differentiation in vitro and determined their potential responses to sulfated components of extracellular matrix (ECM), like those shed from cell surfaces or found in basement membrane and modeled by heparin. Isolated adult human AT2 cells cultured over a 9-day period were used to define the temporal profile of expression of targeted factors during spontaneous differentiation to AT1-like cells. FoxA1 protein was upregulated at early to intermediate time points, where it was strongly elevated by heparin. Gene expression of wnt7A increased dramatically beginning on day 3 and was enhanced even further on days 7 and 9 by heparin, whereas protein expression appeared at days 7 and 9. These temporal changes of expression suggest that sulfated ECMs may act to enhance the increase in FoxA1 at the critical juncture when AT2 cells commence the differentiation process to AT1 cells, in addition to enhancing the increase in wnt7A when the AT1 cell phenotype stabilizes. Johnson Background:, An important theory of attention suggests that there are three separate networks that execute discrete cognitive functions. The ,alerting' network acquires and maintains an alert state, the ,orienting' network selects information from sensory input and the ,conflict' network resolves conflict that arises between potential responses. This theory holds promise for dissociating discrete patterns of cognitive impairment in disorders where attentional deficits may often be subtle, such as in attention deficit hyperactivity disorder (ADHD). Methods:, The Attentional Network Test (ANT), a behavioural assay of the functional integrity of attention networks, was used to examine the performance of 73 children with ADHD and 73 controls. Results:, Performance on the ANT clearly differentiated the children with and without ADHD in terms of mean and standard deviation (SD) of reaction time (RT), the number of incorrect responses made and the number of omission errors made. The ADHD group demonstrated deficits in the conflict network in terms of slower RT and a higher number of incorrect responses. The ADHD group showed deficits in the alerting network in terms of the number of omission errors made. There was no demonstration of a deficit in the orienting network in ADHD on this task. Conclusions:, The children with ADHD demonstrated deficits in the alerting and conflict attention networks but normal functioning of the orienting network. [source] |
