Formal Synthesis of Olivacine via Indolylborate

Abstract Palladium-catalyzed tandem cyclization,cross-coupling reaction of indolylborate 2 and vinyl bromide 5 was successfully applied in a short formal synthesis of olivacine. The reaction of 2 with 5 in the presence of Pd(OAc)2 readily afforded three kinds of products, triene derivative 6 and vinylindole derivative 7, along with a small amount of the piperidine derivative 8 (Scheme,2). On the other hand, the reactions of 2 with bromide 10 or 15 were also examined (Schemes,4 and 5), and their outcome markedly depended on the relative ease of ring closure as a function of ring size. Irradiation of 6 with a high-pressure mercury lamp (,9; Scheme,2), followed by removal of the N -[(benzyloxy)carbonyl] group and subsequent oxidation afforded, after deprotection, pyridocarbazole 23, and the conversion of 23 to olivacine is known (Scheme,6). A radically new UV stabilizer for flexible PVC roofing membranes

Ultraviolet absorbers, such as those from the hydroxyphenylbenzotriazole and hydroxybenzophenone classes of compounds, are typically added to flexible PVC roofing membranes in order to protect the membranes during long-term exposure to sunlight. In this article a radically new UV stabilizer, one that is not just another UV absorber, is introduced. This new UV stabilizer, which is chemically a low-basicity hindered piperidine derivative, was found to dramatically increase the weatherability of PVC roofing membranes. The performance of the new light stabilizer is documented and compared to that of traditional PVC membranes containing just a UV absorber. Although the article focuses on PVC roofing membranes, examples are provided of other durable applications that could benefit from the new light stabilizer technology. J. VINYL ADDIT. Dose-Dependent Immunohistochemical Changes in Rat Cornea and Retina after Oral Methylphenidate Administration

Summary Methylphenidate hydrochloride (MPH), more commonly known as Ritalin, is a piperidine derivative and is the drug most often used to treat attention deficit/hyperactivity disorder, one of the most common behavioural disorders of children and young adults. The aim of this study was to investigate dose-dependent immunohistochemical Dopamine 2 receptor (D2) expression and apoptosis in the rat cornea and cornea. In this study, 27 female pre-pubertal Wistar albino rats, divided into three different dose groups (5, 10 and 20 mg/kg) and their control groups, were used. They were treated orally with methylphenidate dissolved in saline solution for 5 days per week during 3 months. At the end of the third month, after perfusion fixation, eye tissue was removed. Paraffin sections were collected for immunohistochemical and terminal deoxynucleotidyl-transferase-mediated dUTP-biotin nick end labelling assay studies. In our study, we observed that the cornea D2 receptor reactivity showed a dose-related increase after MPH treatment, especially in basal cells of the epithelium and a dose-dependent decrease in the retinal ganglion cell which was statistically meaningful. Analysis of the cornea thickness results showed no meaningful difference between groups. Apoptotic cell number showed a meaningful increase in the high dose treated group compared to the other groups of the study. The data suggest that Ritalin has degenerative effect on the important functional part of the eye, such as cornea and retina and its activating dopaminergic mechanism via similar neuronal paths, functionally and structurally, to induce morphological changes. As a result, we believe that this morphological changes negatively effecting functional organization of the affected cornea and retina. [source]

Synthesis of functionalized cyclic enamines from lithium alkylphenyl sulfones and N -carbo- tert -butoxy lactams

Alkylphenyl sulfones 3 are appropriate synthons for the synthesis of 2-phenylsulfonyl alkylidene pyrro-lidine or piperidine derivatives 1 in good to moderate yields. The lithium alkyl sulfones 4 are first reacted with the desired protected lactams and then subjected to acidic methanolysis to afford the unusual enam-ines 1a-e. NMR studies (COSY 1H- 1H, COSY 1H- 13C, NOE) showed the Z enamines to exist in a dynamic equilibrium with the corresponding imines 2. The stereochemistry of the described compounds was confirmed by molecular calculations. [source]


Design, Synthesis and Evaluation of N -Basic Substituted 3-Hydroxypyridin-4-ones: Orally Active Iron Chelators with Lysosomotrophic Potential

To investigate the possibility of targeting chelators into the lysosomal iron pool, nine bidentate 3-hydroxypyridin-4-ones with basic chains have been synthesized. As the turnover of ferritin iron is centred in the lysosome, such strategy is predicted to increase chelator efficacy of bidentate ligands. The pKa values of the ligands together with their distribution coefficients between 1-octanol and 4-morpholinepropane sulphonic acid (MOPS) buffer pH 7.4 have been determined. The in-vivo iron mobilization efficacy of these basic 3-hydroxypyridin-4-ones has been investigated in a 59Fe-ferritin-loaded rat model. No obvious correlation was observed between efficacy and the pKa value of the side chain, although those with pKa > 7.0 tended to be more efficient than those with pKa < 7.0. The imidazole-containing molecules are much less effective than the tertiary amine derivatives. A dose-response study suggested that basic pyridinones are relatively more effective at lower doses when compared with N -alkyl hydroxypyridinones. Optimal effects were observed with the piperidine derivatives 4h and 4i. The derivative 4i at a dose of 150 ,mol kg,1 was more effective than 450 ,mol kg,1 deferiprone, the widely adopted clinical dose. [source]


Anticonvulsant and Neurotoxicity Evaluation of Some Novel Kojic Acids and Allomaltol Derivatives

Abstract A series of new 3-hydroxy-6-hydroxymethyl/methyl-2-substituted 4H -pyran-4-ones were synthesized and prepared by the reaction of kojic acid or allomaltol with piperidine derivatives and formaline as potential anticonvulsant compounds. The structure of the synthesized compounds was confirmed using the elemental analysis results and the spectroscopic techniques such as IR, 1H-NMR, and ESI-MS. Anticonvulsant activities were examined by maximal electroshock (MES) and subcutaneous Metrazol (scMet)-induced seizure tests. Neurotoxicity was determined by the rotorod toxicity test. All these tests were performed in accordance with the procedures of the Antiepileptic Drug Development (ADD) program. According to the activity studies and at all doses, 3-hydroxy-2-[(4-hydroxy-4-phenylpiperidin-1-yl)methyl]-6-methyl-4H -pyran-4-one (compound 1), 2-{[4-(4-chlorophenyl)-3,6-dihydropyridin-1(2H)-yl]methyl}-3-hydroxy-6-methyl-4H -pyran-4-one (compound 6), 2-[(4-acetyl-4-phenylpiperidin-1-yl)methyl]-3-hydroxy-6-(hydroxymethyl)-4H -pyran-4-one (compound 11), and 2-{[4-(4-chlorophenyl)-3,6-dihydropyridin-1(2H)-yl] methyl}-3-hydroxy-6-hydroxymethyl-4H -pyran-4-one (compound 12) were found to have anticonvulsant activity against MES-induced seizures at 4 h. Also, 2-{[4-(4-bromophenyl)-4-hydroxypiperidin-1-yl]methyl}-3-hydroxy-6-(hydroxymethyl)-4H -pyran-4-one (compound 8) was determined to be the most active compound against scMet-induced seizures at all doses at 0.5 and 4 h. In the rotorod neurotoxicity screening, all compounds showed no toxicity at all doses. [source]


A New, Efficient and Stereoselective Synthesis of Tricyclic and Tetracyclic Compounds by Samarium Diiodide Induced Cyclisations of Naphthyl-Substituted Arylketones,An Easy Access to Steroid-Like Skeletons

Abstract In this report, we present the application of samarium diiodide induced cyclisations of naphthyl-substituted ketones towards an easy and stereoselective access to tri- and tetracyclic-functionalised compounds. Typical naphthalene derivatives were studied to investigate the scope and limitations of this novel cyclisation process. The model substrates studied demonstrate that the samarium ketyl cyclisations are essentially restricted to the formation of six-membered rings. The diastereoselectivity of these reactions is strongly influenced by the connection of the alkyl side chain to the naphthalene core. ,-Naphth-1-yl-substituted ketones furnished cyclisation products, such as 17 or 22,26, as single diastereomers, whereas ,-naphth-2-yl-substituted precursors gave mixtures of diastereomers,as demonstrated by the conversion of model compound 10 into tricyclic products 18,a/18,b, or that of cyclohexanone derivative 33 into tetracyclic diastereomers 34,a/34,b. Cyclic ketones as ketyl precursors furnished steroid-like tetracyclic skeletons; however, due to the cis/cis fusion of rings B/C and C/D these products have an "unnatural" bowl-like shape. Several of the cyclisation products have been identified by X-ray analyses, which not only proved the constitutions, but also the relative configurations and the preferred conformations. Steroid analogue 23 was subjected to subsequent transformations, which demonstrate that the styrene-like double bond of such compounds can be used for further structural diversification. First attempts to synthesise related azasteroids by incorporating nitrogen atoms into the ketone moiety are also reported. Thus, pyrrolidine derivatives 44 and 47 as well as piperidine derivatives 50 and 52 were subjected to samarium diiodide induced cyclisations. The expected tetracyclic products 48, 49,a/49,b, 51 and 53,a/53,b were obtained in moderate to good yields. The stereoselectivities observed follow the rules already established for the all-carbon precursors. The resulting products, bearing a nitrogen atom in ring D, are interesting azasteroid analogues with "unnatural" configuration. [source]


Bifunctional-Thiourea-Catalyzed Diastereo- and Enantioselective Aza-Henry Reaction

Abstract Bifunctional thiourea 1,a catalyzes aza-Henry reaction of nitroalkanes with N -Boc-imines to give syn -,-nitroamines with good to high diastereo- and enantioselectivity. Apart from the catalyst, the reaction requires no additional reagents such as a Lewis acid or a Lewis base. The N-protecting groups of the imines have a determining effect on the chirality of the products, that is, the reaction of N -Boc-imines gives R adducts as major products, whereas the same reaction of N -phosphonoylimines furnishes the corresponding S adducts. Various types of nitroalkanes bearing aryl, alcohol, ether, and ester groups can be used as nucleophiles, providing access to a wide range of useful chiral building blocks in good yield and high enantiomeric excess. Synthetic versatility of the addition products is demonstrated by the transformation to chiral piperidine derivatives such as CP-99,994. [source]
