Relations of sugar composition and ,13C in phloem sap to growth and physiological performance of Eucalyptus globulus (Labill)

ABSTRACT We characterized differences in carbon isotopic content (,13C) and sugar concentrations in phloem exudates from Eucalyptus globulus (Labill) plantations across a rainfall gradient in south-western Australia. Phloem sap ,13C and sugar concentrations varied with season and annual rainfall. Annual bole growth was negatively related to phloem sap ,13C during summer, suggesting a water limitation, yet was positively related in winter. We conclude that when water is abundant, variations in carboxylation rates become significant to overall growth. Concentrations of sucrose in phloem sap varied across sites by up to 600 mm, and raffinose by 300 mm. These compounds play significant roles in maintaining osmotic balance and facilitating carbon movement into the phloem, and their relative abundances contribute strongly to overall ,13C of phloem sap. Taken together, the ,13C and concentrations of specific sugars in phloem sap provide significant insights to functions supporting growth at the tree, site and landscape scale. [source]


Physiological changes in white lupin associated with variation in root-zone CO2 concentration and cluster-root P mobilization

ABSTRACT White lupin (Lupinus albus L.) mobilizes insoluble soil phosphorus through exudation of organic acids from ,cluster' roots. Organic acid synthesis requires anaplerotic carbon derived from dark CO2 fixation involving PEP-carboxylase. We tested the hypothesis that variation in root-zone CO2 concentration would influence organic acid synthesis and thus P mobilization. Root-zone CO2 concentrations and soil FePO4 concentrations supplied to sand-grown white lupin (cv. Kiev Mutant) were varied. More biomass accumulated in plants supplied with 360 µL L,1 CO2 to the root-zone, compared with those aerated with either 100 or 6000 µL L,1 CO2. Increased FePO4 in the sand resulted in greater leaf P concentrations, but root-zone [CO2] did not influence leaf P concentration. Suppression of cluster-root development in plants supplied with 100 µL L,1 root-zone CO2 was correlated with increased leaf [P]. However, at both 360 and 6000 µL L,1 CO2, cluster-root development was suppressed only at the highest leaf P concentration. Phloem sap [P] was significantly increased by greater [FePO4] in the sand, but was reduced with increased root-zone [CO2], and this may have triggered increased cluster-root initiation. Succinate was the major organic acid (carboxylate) in the phloem sap (minor components included malate, citrate, fumarate) and was increased at greater [FePO4], suggesting that this shoot-derived carboxylate might provide an important source of organic acids for root metabolism. Since cluster root development was inhibited by increasing concentrations of FePO4 in the sand, it is possible that succinate was utilized for the functioning of the root-nodules. [source]


Separating host-tree and environmental determinants of honeydew production by Ultracoelostoma scale insects in a Nothofagus forest

Abstract 1.,Sugar-rich honeydew excreted (,produced') by insects feeding on phloem sap is a key energy flow in a range of temperate and tropical ecosystems. The present study measured honeydew produced by Ultracoelostoma sp. (Homoptera: Coelostomidiidae) scale insects feeding on Nothofagus solandri var. solandri (Hook f.) Oerst. trees in a temperate evergreen forest in New Zealand. Simultaneous measurements of environmental variables and canopy photosynthesis were conducted to allow separation of host-tree and environmental determinants of honeydew production. These relationships were further examined in experiments where canopy photosynthesis was manipulated by shading or plant nitrogen levels increased by foliar spray. 2.,Rates of honeydew production varied nine-fold from a maximum (± 1 SE) of 64.4 ± 15.2 mg dry mass m,2 bark h,1 in early summer (December) to a minimum of 7.4 ± 4.2 mg m,2 h,1 in winter (August). Rates of production measured 1.4 m from the base of the trees' stems varied significantly with stem diameter, and were higher on medium-sized (18 cm diameter) than small or large stems. 3.,Rates of production were significantly related to environmental conditions over the hours preceding measurement (air temperature and air saturation deficit averaged over the preceding 24 and 12 h respectively). There was no evidence that rates of production were directly related to short-term changes in the supply of carbohydrates from the canopy (either when compared with measurements of unmanipulated photosynthetic rate, or after sugar levels were manipulated by shading 80% of host-trees' leaf area), or to changes in phloem nitrogen content. 4.,The results show that there is no clear effect of host-tree carbon supply on honeydew production; if production is related to photosynthesis, the effect of this is much less important that the large and significant direct effect of environmental conditions on honeydew production. Flexibility in the composition and concentration of amino acids in honeydew of the drepanosiphid aphid Tuberculatus quercicola

Abstract 1. Mutualistic interactions between aphids and ants are mediated by honeydew that aphids produce. Previous work showed that when attended by the ant Formica yessensis Forel (Hymenoptera: Formicidae), nymphs of the aphid Tuberculatus quercicola (Matsumura) (Homoptera: Aphididae) developed into significantly smaller adults with lower fecundity than did nymphs that were not ant attended. 2. This study tested the hypothesis that this cost of ant attendance arises through changes in the quality and quantity of honeydew. Ant-attended and ant-excluded aphid colonies were prepared in the field. The composition and concentration of amino acids were compared between the honeydew produced by ant-attended colonies and that produced by ant-excluded colonies. 3. The aphids excreted smaller droplets of honeydew, but also excreted them more frequently, in ant-attended colonies than in ant-excluded colonies. The honeydew of ant-attended aphids contained more types of amino acid, and a significantly higher total concentration of amino acids, than did the honeydew of ant-excluded aphids. 4. These results suggest that the increase in the concentration of amino acids in honeydew leads to a shortage of nitrogen available for aphid growth and reproduction, resulting in lower performance under ant attendance. 5. With the advance of seasons, a significant reduction was found in both the total free amino acid concentration in phloem sap and the frequency of honeydew excretion; however the total concentration of amino acids in the honeydew did not vary significantly during the seasons, suggesting that aphids keep the quality of honeydew constant in order to maintain ant visitation. Analysis by DC,EPG of the resistance to Bemisia tabaci on an Mi -tomato line

Abstract The tomato Mi gene confers resistance to nematodes, Meloidogyne spp., and to the potato aphid, Macrosiphum euphorbiae (Thomas). Previous greenhouse choice assays with Bemisia tabaci (Gennadius) showed that tomato commercial varieties carrying this gene had significantly lower values of host suitability and whitefly reproduction than varieties lacking Mi. This indicated that Mi, or another gene in its region, could regulate partial resistance. In order to characterise this resistance, probing and feeding behaviour of Bemisia tabaci B-biotype was studied with DC Electrical Penetration Graph (EPG) technique on the near-isogenic tomato lines Moneymaker (without Mi) and Motelle (carrying Mi). Significant differences (P < 0.05) between tomato lines were found in EPG parameters related to epidermis and/or mesophyll tissues. On Motelle, a lower percentage of whiteflies achieved phloem phase and they made more probes before attaining first phloem phase, had a higher ratio (number of probes before first phloem phase)/(total number of probes), had a longer total duration of non-probing time, and a longer time before making the first intracellular puncture and before making the first phloem phase. In contrast, most of the parameters related to phloem phase were found not to differ significantly between these near-isogenic lines. The behavioural data strongly suggest that the partial resistance in the variety Motelle is due to factors in the epidermis and/or mesophyll that inhibit the whiteflies from reaching phloem sieve elements. However, once the stylets reach a sieve element, whitefly behaviour did not differ between the two varieties. Thus, phloem sap of the two varieties appears to be equally acceptable to the whiteflies. Further studies are necessary to provide a better understanding of these mechanisms of resistance to whiteflies in tomatoes. [source]


Influence of parasitism by encyrtid parasitoids on the feeding behaviour of the cassava mealybug Phenacoccus herreni

Abstract Three encyrtid parasitoids Apoanagyrus (Epidinocarsis) diversicornis, Aenasius vexans, and Acerophagus coccois (Hymenoptera: Encyrtidae) are used to control the cassava mealybug Phenacoccus herreni Cox & Williams (Sternorrhyncha: Pseudococcidae), an important pest of cassava in South America. The influence of parasitism on the feeding behaviour of mealybugs was studied by observing honeydew secretion and by the electrical penetration graph technique (EPG, DC-system). Honeydew secretions were observed after parasitism until mummy transformation. No strong EPG parameter differences were found between parasitised and control insects. All results indicated that parasitised mealybugs keep feeding on the phloem sap after parasitism until mummy transformation. The main influence of parasitism on EPG parameters is the appearance of a new pattern resembling the E2 pattern at the extracellular level and labelled H. This pattern was also produced with control insects located on an unfavourable feeding site and could be associated with a stress response. It might be related to the still unclear E(c) pattern of aphids. The relationship of H to stylet activities is discussed. [source]


THE ROLE OF BACTERIAL SYMBIONTS IN AMINO ACID COMPOSITION OF BLACK BEAN APHIDS

Abstract To evaluate the role of bacterial symbionts (Buchnera spp.) in the black bean aphids (Aphis craccivora Koch), the aphids were treated with the antibiotic, rifampicin, to eliminate their intracellular symbiotic bacteria. Analysis of protein and amino acid concentration in 7-day-old of aposymbiotic aphids showed that the total protein content per mg fresh weight was significantly reduced by 29%, but free amino acid titers were increased by 17%. The ratio of the essential amino acids was in general only around 20% essential amino acids in phloem sap of broad bean, whereas it was 44% and 37% in symbiotic and aposymbiotic aphids, respectively, suggesting that the composition of the free amino acids was unbalanced. For example, the essential amino acid, threonine represented 21.6% of essential amino acids in symbiotic aphids, but it was only 16.7% in aposymbiotic aphids. Likewise, two nonessential amino acids, tyrosine and serine, represented 8.9% and 5.6% of total amino acids in symbiontic aphids, respectively, but they enhanced to 21.1% and 13.6% in aposymbiotic aphids. It seems likely that the elevated free amino acid concentration in aposymbiotic aphids was caused by the limited protein anabolism as the result of the unbalanced amino acid composition. [source]


Pathogen-induced resistance and alarm signals in the phloem

SUMMARY Despite a long-standing notion of long-distance signals triggering systemic acquired resistance (SAR), the translocation pathway and the identity of the signals involved have not been determined with any degree of certainty. A critical assessment indicates that, in parallel to signalling via the phloem, alternative modes for SAR induction such as signalling via the xylem or air-borne signalling by volatile substances may occur. This review further evaluates several classes of compounds as being functional in systemic resistance signalling. Evidence in favour of SAR involvement of phloem-mobile substances such as salicylic acid, lipid-derived molecules, reactive oxygen species and components of the antioxidant machinery is contradictory, circumstantial or inconclusive, at best. Nitric oxide bound to proteins or thiols seems a good candidate for signalling, but has not been found in phloem sap thus far. No convincing support of the involvement in SAR of phloem-mobile substances such as calcium, oligosaccharides, peptides or RNA species, which function in other systemic signalling cascades, has yet been produced. Nevertheless, phloem-mobile macromolecules are considered as potential tools for SAR given their pivotal role in remote gene expression under stress conditions. In this framework, the existence of several cascades for signal generation along the phloem pathway is envisaged. Finally, recent methods for detection of molecular signals in phloem sap and their expression in companion cells are presented. [source]


Honeydew amino acids in relation to sugars and their role in the establishment of ant-attendance hierarchy in eight species of aphids feeding on tansy (Tanacetum vulgare)

Abstract., The ratio of the concentration of honeydew total amino acids to total sugars in the honeydew of eight species of aphids, all feeding on tansy, Tanacetum vulgare (L.), was determined and correlated with honeydew production and ant-attendance. The honeydew of the five ant-attended aphid species [Metopeurum fuscoviride (Stroyan), Trama troglodytes (v. Hayd), Aphis vandergooti (Börner), Brachycardus cardui (L.), Aphis fabae (Scopoli)] was rich in total amino acids, ranging from 12.9 to 20.8 nmol µL,1 compared with the unattended aphid Macrosiphoniella tanacetaria (Kalt.) with only 3 nmol µL,1. Asparagine, glutamine, glutamic acid and serine (all nonessential amino acids) were the predominant amino acids in the honeydew of all species. The total concentration of amino acids in the phloem sap of tansy was much higher (78.7 nmol µL,1) then in the honeydew samples, and the predominant amino acids were glutamate (34.3%) and threonine (17.7%). A somewhat unexpected result was the finding that those aphid species with the highest total amino acid concentration in the honeydew always had the highest concentration of sugars. The lowest amino acid,sugar combined value was 104,28.8 nmol µL,1 in the non ant-attended species M. tanacetaria, and the highest value was an average of 270,89.9 nmol µL,1 for the three most intensely attended aphid species M. fuscoviride, A. vandergooti and T. troglodytes. There is no evidence that any single amino acid or group of amino acids in the honeydew acted as an attractant for ant-attendance in these eight aphid species. The richness of the honeydew (rate of secretion × total concentration of sugars), along with the presence of the attractant sugar melezitose, comprised the critical factors determining the extent of ant-attendance of the aphids feeding on T. vulgare. The high total amino acid concentration in sugar-rich honeydews can be explained by the high flow-through of nutrients in aphids that are particularly well attended by ants. [source]


Amino acid budgets in three aphid species using the same host plant

Abstract. Nutrient provisioning in aphids depends both on the composition of ingested phloem sap and on the biosynthetic capabilities of the aphid and its intracellular symbionts. Amino acid budgets for three aphid species, Rhopalosiphum padi (L.), Schizaphis graminum (Rondani) and Diuraphis noxia (Mordvilko), were compared on a single host plant species, wheat Triticum aestivum L. Ingestion of amino acids from phloem, elimination of amino acids in honeydew, and the content of amino acids in aphids tissue were measured. From these values, ingestion rates were estimated and compared to honeydew and to estimated composition of aphid proteins. Ingestion rate was lowest in D. noxia due to low growth rate and low honeydew production; intermediate in S. graminum due to higher growth rate and intermediate honeydew production; and highest in R. padi, which had the highest rates for both variables. Both D. noxia and S. graminum induced increases in the amino acid content of ingested phloem. These changes in phloem content, combined with differences in ingestion rates, resulted in large differences among aphids in estimated rates of ingestion of individual amino acids. In honeydew, most essential amino acids were found in low amounts compared with the amounts ingested, especially for methionine and lysine. A few amino acids (arginine, cystine, histidine and tryptophan) were more abundant in honeydew of some aphids, suggesting oversupply. Aphid species differed in the composition of free amino acids in tissue but showed very similar composition in protein, implying similar requirements among the aphids. In R. padi and D. noxia, most essential amino acids were ingested in amounts insufficient for growth, implying dependence on symbiont provisioning. In S. graminum, most amino acids were ingested in amounts apparently sufficient for growth. [source]

Evaporative enrichment and time lags between ,18O of leaf water and organic pools in a pine stand

ABSTRACT Understanding ecosystem water fluxes has gained increasing attention, as climate scenarios predict a drier environment for many parts of the world. Evaporative enrichment of 18O (,18O) of leaf water and subsequent enrichment of plant organic matter can be used to characterize environmental and physiological factors that control evaporation, based on a recently established mechanistic model. In a Pinus sylvestris forest, we measured the dynamics of oxygen isotopic composition (,18O) every 6 h for 4 d in atmospheric water vapour, xylem sap, leaf water and water-soluble organic matter in current (N) and previous year (N-1) needles, phloem sap, together with leaf gas exchange for pooled N and N-1 needles, and relevant micrometeorological variables. Leaf water ,18O showed strong diel periodicity, while ,18O in atmospheric water vapour and in xylem sap showed little variation. The ,18O was consistently lower for N than for N-1 needles, possibly related to phenological stage. Modelled leaf water ,18O showed good agreement with measured values when applying a non-steady state evaporative enrichment model including a Péclet effect. We determined the time lags between ,18O signals from leaf water to water-soluble foliar organic matter and to phloem sap at different locations down the trunk, which clearly demonstrated the relevance of considering these time-lag effects for carbon transport, source-sink and carbon flux partitioning studies. Simultaneous measurement of water flow velocity and solute transport in xylem and phloem of adult plants of Ricinus communis over a daily time course by nuclear magnetic resonance spectrometry

ABSTRACT A new method for simultaneously quantifying rates of flow in xylem and phloem using the FLASH imaging capabilities of nuclear magnetic resonance (NMR) spectrometry was applied in this study. The method has a time resolution of up to 4 min (for the xylem) and was used to measure the velocity of flows in phloem and xylem for periods of several hours to days. For the first time, diurnal time course measurements of flow velocities and apparent volume flows in phloem and xylem in the hypocotyl of 40-d-old Ricinus communis L were obtained. Additional data on gas exchange and the chemical composition of leaves, xylem and phloem sap were used to assess the role of leaves as sinks for xylem sap and sources for phloem. The velocity in the phloem (0·250 ± 0·004 mm s,1) was constant over a full day and not notably affected by the light/dark cycle. Sucrose was loaded into the phloem and transported at night, owing to degradation of starch accumulated during the day. Concentrations of solutes in the phloem were generally less during the night than during the day but varied little within either the day or night. In contrast to the phloem, flow velocities in the xylem were about 1·6-fold higher in the light (0·401 ± 0·004 mm s,1) than in the dark (0·255 ± 0·003 mm s,1) and volume flow varied commensurately. Larger delays were observed in changes to xylem flow velocity with variation in light than in gas exchange. The relative rates of solute transport during day and night were estimated on the basis of relative flow and solute concentrations in xylem and phloem. In general, changes in relative flow rates were compensated for by changes in solute concentration during the daily light/dark cycle. However, the major solutes (K+, NO3,) varied appreciably in relative concentrations. Hence the regulation of loading into transport systems seems to be more important to the overall process of solute transport than do changes in mass flow. Due to transport behaviour, the chemical composition of leaves varied during the day only with regard to starch and soluble carbohydrates. [source]


Effects of cold-girdling on flows in the transport phloem in Ricinus communis: is mass flow inhibited?

ABSTRACT The effects of cold girdling of the transport phloem at the hypocotyl of Ricinus communis on solute and water transport were investigated. Effects on the chemical composition of saps of phloem and xylem as well as of stem tissue were studied by conventional techniques and the water flow in the phloem was investigated by NMR imaging. Cold girdling reduced the concentration of sucrose but not that of inorganic solutes or amino acids in phloem saps. The possibility that cold treatment inhibited the retrieval of sucrose into the phloem, following leaching from the sieve tubes along a chemical gradient is discussed. Leaching of other solutes did not occur, as a result of missing promoting gradients in stem tissue. Following 3 d of cold girdling, sugar concentration increased and starch was synthesized and accumulated in stem tissue above the cold girdling region and along the cold-treated phloem pathway due to leaching of sugars from the phloem. Only in the very first period of cold girdling (< 15,30 min) was mass flow inhibited, but recovered in the rest of cold treatment period to values similar to the control period before and the recovery period after the cold treatment. It is concluded that cold treatment affected phloem transport through two independent and reversible processes: (1) a permanent leaching of sucrose from the phloem stem without normal retrieval during cold treatment, and (2) a short-term inhibition of mass flow at the beginning of cold treatment, possibly involving P proteins. Possible further mechanisms for reversible inhibition of water flow are discussed. [source]
