Neil Thomas Abstract Background Ageing promotes increases in the prevalence of components of the metabolic syndrome, which obesity often underlies. Methods We report the relationship between ageing, obesity and other cardiovascular risk factors in 694 community-based Chinese subjects in gender-specific groups of three age ranges: 20.0,39.9 (young), 40.0,59.9 (middle-aged) and 60.0,79.9 (old-aged) years. Results Body mass index (BMI) values were similar in males in each age group, but waist and percentage body fat increased (6.6, and 39.5%, both p < 0.001, respectively), from young to old-age groups, as did blood pressure and glycated haemoglobin levels (all p < 0.001). In the females, increases (all p < 0.001) in percentage body fat (29.3%) were accompanied by greater increases in BMI (10.3%) and waist (19.2%) than the males. Blood pressure, glycated haemoglobin, total and LDL-cholesterol and triglyceride levels increased linearly with age (all p < 0.001). Conclusion Age-related increases in central adiposity and percentage body fat were associated with increasingly adverse cardiovascular risk factor profiles. Copyright © 2005 John Wiley & Sons, Ltd. [source] Leptin receptor 170 kDa (OB-R170) protein expression is reduced in obese human skeletal muscle: a potential mechanism of leptin resistanceEXPERIMENTAL PHYSIOLOGY, Issue 1 2010T. Fuentes To examine whether obesity-associated leptin resistance could be due to down-regulation of leptin receptors (OB-Rs) and/or up-regulation of suppressor of cytokine signalling 3 (SOCS3) and protein tyrosine phosphatase 1B (PTP1B) in skeletal muscle, which blunt janus kinase 2-dependent leptin signalling and signal transducer and activator of transcription 3 (STAT3) phosphorylation and reduce AMP-activated protein kinase (AMPK) and acetyl-coenzyme A carboxylase (ACC) phosphorylation. Deltoid and vastus lateralis muscle biopsies were obtained from 20 men: 10 non-obese control subjects (mean ±s.d. age, 31 ± 5 years; height, 184 ± 9 cm; weight, 91 ± 13 kg; and percentage body fat, 24.8 ± 5.8%) and 10 obese (age, 30 ± 7 years; height, 184 ± 8 cm; weight, 115 ± 8 kg; and percentage body fat, 34.9 ± 5.1%). Skeletal muscle OB-R170 (OB-R long isoform) protein expression was 28 and 25% lower (both P < 0.05) in arm and leg muscles, respectively, of obese men compared with control subjects. In normal-weight subjects, SOCS3 protein expression, and STAT3, AMPK, and ACC, phosphorylation, were similar in the deltoid and vastus lateralis muscles. In obese subjects, the deltoid muscle had a greater amount of leptin receptors than the vastus lateralis, whilst SOCS3 protein expression was increased and basal STAT3, AMPK, and ACC, phosphorylation levels were reduced in the vastus lateralis compared with the deltoid muscle (all P < 0.05). In summary, skeletal muscle leptin receptors and leptin signalling are reduced in obesity, particularly in the leg muscles. [source] The Effect of Honey Compared to Sucrose, Mixed Sugars, and a Sugar-Free Diet on Weight Gain in Young RatsJOURNAL OF FOOD SCIENCE, Issue 3 2007L.M. Chepulis ABSTRACT:, To determine whether honey, sucrose, and mixed sugars as in honey have different effects on weight gain, 40 6-wk-old Sprague-Dawley rats were fed a powdered diet that was either sugar free or contained 8% sucrose, 8% mixed sugars as in honey, or 10% honey freely for 6 wk. Weight gain and food intake were assessed weekly, and at completion of the study blood samples were removed for measurement of blood sugar (HbA1c) and a fasting lipid profile. The animals were then minced and total percentage body fat and protein measured. Overall percentage weight gain was significantly lower in honey-fed rats than those fed sucrose or mixed sugars, despite a similar food intake. Weight gains were comparable for rats fed honey and a sugar free diet although food intake was significantly higher in honey-fed rats. HbA1c and triglyceride levels were significantly higher in all sugar treatments compared with rats fed a sugar free diet, but no other differences in lipid profiles were reported. No differences in percentage body fat or protein levels were reported. Zaveri Abstract Background:, With the increasing prevalence of being overweight and obesity, dietary strategies to curb hunger levels and increase satiety at lower energy intakes are sought. The frequency of eating and type of snack may influence total energy intake. The present study aimed to assess the impact of providing either a conventional snack (cereal bar) or a nonconventional snack (almonds) on eating frequency, hunger rating, dietary intake, body weight and blood lipids. Methods:, Forty-five healthy men (aged 25,50 years, body mass index = 25,35 kg m,2) were recruited and allocated to a control, cereal bar or almond snack group. Two packets of cereal bars and almonds were introduced for 12 weeks to the cereal bar group and the almond snack group, respectively. Dietary intakes and eating frequency were assessed by 4-day unweighed diet diaries; visual analogue scales were used to assess hunger ratings; and fasting blood parameters (i.e. glucose, insulin, total cholesterol, triglycerides, low-density lipoprotein cholesterol and high-density lipoprotein cholesterol) were measured at baseline, 6 and 12 weeks. In addition, anthropometric measures (height, weight, skinfold thickness, waist and hip circumference) were measured at baseline, 6 and 12 weeks. Results:, The present study found no significant change in the eating frequency within groups at 12 weeks. However, the almond snack group had a significantly higher eating frequency than the control group (P , 0.05) and cereal bar group (P , 0.01). This did not result in higher energy intake, body weight or percentage body fat in the almond snack group. Conclusion:, The present study demonstrated that snacking on almonds, in comparison to cereal bars, promoted a higher eating frequency, but not a higher energy intake. Advice to snack on either almonds or cereal bars did not result in weight gain, suggesting that energy compensation took place. LIBERATO Abstract Aim:, To quantitatively analyse energy intake reported by young men and the accuracy of the Goldberg cut-off method for identifying misreporters. Methods:, This was a cross-sectional study in which: food intake was assessed by a four-day food record; resting metabolic rate was assessed by indirect calorimetry; percentage body fat was measured by dual-energy X-ray absorptiometry; and energy expenditure was assessed by physical activity record completed simultaneously with food intake measurements. Energy intake was analysed by direct comparison of energy intake and energy expenditure and by the Goldberg cut-off. Subjects: 34 healthy men aged 18,25 years. Setting: Queensland University of Technology, Queensland, Australia. Main outcome measures: percentage of misreporters in a group of young men using different methods. Statistical analyses: data are presented as means and standard deviations. The analyses were conducted using Statistic for Windows 5.5 software. Results:, Seven underreporters were identified by direct comparison of energy intake and energy expenditure. The Goldberg cut-off found six out of the seven underreporters identified by direct comparison of energy intake and energy expenditure, but wrongly identified two acceptable reporters as underreporters. The sensitivity and specificity of the Goldberg cut-off method were 0.86 and 0.93, respectively. Conclusions:, Seven out of 34 participants underreported their energy intake. In the absence of physical activity measurements, the Goldberg cut-off method identified underreporters in this group of young men with assessed resting metabolic rate. [source] Threshold values of visceral fat and waist girth in Japanese obese childrenPEDIATRICS INTERNATIONAL, Issue 5 2005Kohtaro Asayama AbstractBackground:,In order to define the diagnostic criteria for visceral adipose tissue (VAT) accumulation and abdominal obesity in Japanese youths, a cross-sectional, multicenter study was conducted. Methods:,Subjects were 194 boys and 96 girls ranging in age from 6 to 15 years. Obese youths were classified according to the occurrence of abnormal values in serum triglyceride, alanine aminotransferase or insulin level. A threshold value of each criterion was calculated, using the analysis of receiver operating characteristic (ROC) curve. The areas of total abdominal adipose tissue (AT), VAT and subcutaneous adipose tissue (SAT) were estimated by single slice computed tomography at the level of umbilicus. Results:,VAT area was greater in boys than it was in girls. The critical values for VAT area and waist circumference in all subjects were 54.8 cm2 and 83.5 cm, respectively. The values for the area under the ROC curves were VAT area > total AT area > waist circumference > SAT area > percentage overweight > percentage body fat. The sensitivity and specificity for VAT area were 90.5 and 79.5%, respectively. Those for waist circumference were high enough (> 70%) for clinical use. In the linear regression analysis assigning VAT area as an independent variable and waist circumference as a dependent variable, the expected value for the waist circumference was 82 cm. Conclusion:,In Japanese obese youths ranging in age from 6 to 15 years, the diagnostic criteria for the waist circumference was 82 cm, and that for VAT area was 55 cm2. Arngrímsson The aim of this study was to determine the relation between change in body mass index (BMI) and changes in fat mass (FM), lean soft tissue (LST), and percentage body fat (%Fat) in elderly (67.6 ± 6.0 years) women varying in race (53 black, 144 white) who underwent measurements of BMI, FM, LST, and %Fat at baseline and after 2 years. The group did not markedly change body composition over 2 years (BMI = ,0.1 ± 1.5 kg/m2, P = 0.53; FM = 0.0 ± 2.8 kg, P = 0.95; LST = ,0.4 ± 1.7 kg, P < 0.001; %Fat = 0.3 ± 2.0%, P = 0.06). Change in BMI predicted change in FM (r = 0.90, SEE = 1.19 kg FM, P < 0.001) but was less predictive of change in %Fat (r = 0.64, SEE = 1.54%Fat, P < 0.001). Change in BMI was curvilinearly related to change in LST adjusted for change in height (R = 0.76, SEE = 1.10 kg LST, P < 0.001). [source] High fitness is associated with a healthier programming of body composition at adolescenceAMERICAN JOURNAL OF HUMAN BIOLOGY, Issue 6 2008Francisco B. Ortega The programming effect of birth weight on later body composition has become of increasing interest in recent years. This programming effect is affected by factors such as gender. Physical fitness could be another factor of influence. This study aimed to examine whether handgrip strength (HG) or cardiovascular fitness (CVF) modify the associations between birth weight and body composition in adolescents. A sample of 1,740 (942 females) adolescents aged 13 to 18.5 years, born at more than 35 weeks of gestation, from the AVENA study was studied. Waist circumference was measured, percentage body fat was calculated from skinfold thicknesses and fat free mass was derived by subtracting fat mass from total body weight. HG and CVF were assessed using the HG test and the 20 m shuttle run test, respectively. Birth weight was positively associated with fat free mass in females with high (above the median) CVF (P < 0.001), but not in those with low (below the median) CVF. In contrast, birth weight was positively associated with total and central adiposity in males with low HG (both P = 0.002), but not in those with high HG. These results suggest that the programming effect of birth weight on later body composition is dependent on gender and fitness level. A high birth weight may have a healthy programming effect on body composition in those adolescents with a high fitness level, being associated with increased fat free mass levels in females and not showing the increased adiposity levels observed in unfit males. [source] Change in leptin, body composition and other hormones around menarche , a visual representationACTA PAEDIATRICA, Issue 10 2008LG Bandini Abstract Aim: To present a visual representation of changes in body composition, leptin, insulin, estradiol and follicular stimulating hormone (FSH) levels in relation to menarche in girls. Methods: Participants were a subset of healthy girls (n = 108) enrolled in a longitudinal study of growth and development conducted at the General Clinical Research Center at the Massachusetts Institute of Technology (MIT). Participants were seen annually from before menarche until 4 years postmenarche for measures of body composition and serum levels of leptin, insulin, estradiol and FSH. Body composition was determined by bioelectrical impedance. Standardized body composition and hormone levels were smoothed and plotted relative to menarche to visualize patterns of change. Results: At menarche, the mean percentage body fat (%BF) of girls was 24.6% (SD = 4.1%) after menarche %BF was ,27%. Leptin levels averaged 8.4 ng/mL (SD = 4.6) at menarche and were ,12 ng/mL after menarche. Changes in leptin levels closely paralleled changes in %BF. Insulin, estradiol and FSH levels followed expected patterns relative to menarche. Leptin began rising closer to menarche than did insulin or the other sex hormones. Conclusion: We provide a visual presentation of hormonal and body composition changes occurring throughout the pubertal period in girls which may be useful in generating new hypotheses related to the timing of menarche. [source] Fasting concentrations of nesfatin-1 are negatively correlated with body mass index in non-obese malesCLINICAL ENDOCRINOLOGY, Issue 4 2010Takafumi Tsuchiya Summary Background, We recently identified a novel anorexigenic protein, nesfatin-1, which is processed from nesfatin/nucleobindin-2 (NUCB2). However, the clinical importance of this protein has not been determined. Objective, To investigate its clinical significance in humans, we have established a new specific enzyme-linked immunosorbent assay (ELISA) for human nesfatin-1 in peripheral blood and measured its circulating concentration in healthy subjects. Design, The new sandwich-type ELISA method was validated and then used to measure nesfatin-1 levels in plasma samples, under overnight fasting conditions, followed by oral glucose tolerance and meal tests. Patients and measurements, A total of 43 nonobese males (age: 24·5 ± 0·6 , body mass index (BMI); 21·1 ± 0·3 kg/m2) were recruited to the study for evaluating fasting concentrations of nesfatin-1. In those, fifteen subjects underwent a 75- g oral glucose tolerance test (OGTT) and another 15 underwent a meal test. In addition, fasting concentrations of nesfatin-1 were measured in nine males with high BMI (age: 32·4 ± 3·7 , BMI; 37·3 ± 3·8 kg/m2). Results, Peripheral concentrations of nesfatin-1 showed a significant negative correlation with BMI, percentage body fat, body fat weight and blood glucose (P < 0·05). Nesfatin-1 concentrations were not significantly changed during OGTT and meal tests. Fasting nesfatin-1 levels were significantly lower in subjects with high BMI compared to nonobese subjects (P < 0·05). Conclusions, A new specific and sensitive ELISA for nesfatin-1 was established. Further accumulation of clinical observations is necessary to clarify the role of circulating nesfatin-1 in various metabolic disorders. Travison Summary Introduction, Similarities in the symptomatic expressions of excess adiposity and hypercortisolaemic conditions suggest that elevated glucocorticoid exposure may influence the pathogenesis of obesity. Circulating cortisol levels are not typically elevated in obese subjects, but data from large prospective samples are rare. We undertook an analysis to determine both cross-sectional and longitudinal associations between body composition and serum cortisol concentrations in a randomly chosen group of 999 community-dwelling men, aged 40,79 years. Methods, Data were obtained from the two follow-up waves of the Massachusetts Male Ageing Study (T2: 1995,97; T3: 2002,04). Partial correlation and multivariate regression analyses were used to estimate cross-sectional (T2) and longitudinal associations between serum cortisol concentrations and a range of measures of subjects' body composition, including weight, body mass index (BMI), waist circumference (WC), waist-to-hip girth ratio (WHR), and percentage body fat (measured by bioelectrical impedance at T3); similar analyses were conducted to assess the association between change (T2 to T3) in serum cortisol and simultaneous change in body composition parameters. Results, We observed weak negative associations between cortisol concentrations and all body composition parameters, with the exception of percentage body fat. Longitudinal results demonstrated similar relationships but associations were of lesser magnitude. T2 cortisol concentrations were not associated with change in body composition over time, whereas T2 body size was positively associated with longitudinal changes in cortisol concentrations, providing limited evidence that weight change drives changes in cortisol concentrations, rather than vice versa. Results were unchanged when age and other covariate effects were controlled. Conclusions, Circulating cortisol concentrations are somewhat lower in obese than in nonobese community-dwelling men. There is some evidence that excess adiposity presages increases in cortisol concentrations, rather than the reverse. However, this observation should be greeted with caution, as age-related weight loss , and not gain , was associated with simultaneous increases in serum cortisol concentrations. [source] Effects of resistance training on cardiovascular responses to lower body negative pressure in the elderlyCLINICAL PHYSIOLOGY AND FUNCTIONAL IMAGING, Issue 5 2001Lynn B. Panton The purpose of the present study was to determine whether resistance training alters the cardiovascular responses to submaximal lower body negative pressure (LBNP) in the elderly. Twenty-one subjects were randomized into a control (C: n=10; 70 ± 3 years, mean ± SD) or a resistance training (TR: n=11; 67 ± 7 years) group. Subjects in the TR underwent 12 weeks of training consisting of three sets of 8,12 contractions at ,60,80% of their initial maximal one repetition, three times per week, on 10 different machines. Before (Pre) and after (Post) training, all subjects underwent exposures of LBNP of ,10, ,20 and ,40 Torr and muscle biopsy sampling at the vastus lateralis. TR increased (P,0·05) knee extension (Pre=379 ± 140 N, Post=534 ± 182 N) and chest press (Pre=349 ± 137 N, Post=480 ± 192 N) strength. Neither body weight nor percentage body fat were altered (P,>0·05) by training. Resistance training increased (P,0·05) cross-sectional area in both Type I (4203 ± 1196 to 5248 ± 1728 ,m2) and Type II (3375 ± 1027 to 4286 ± 1892 ,m2) muscle fibres. Forearm blood flow, forearm vascular conductance, mean arterial pressure, and heart-rate responses to LBNP were not altered by the training. These data suggest that the cardiovascular responses of elderly to LBNP are unaffected by 12 weeks of whole-body resistance training despite increases in muscle strength and size. [source] |
