Effects of Diet and Water Source on the Nursery Production of Pacific White Shrimp Litopenaeus vannamei

Penaeid shrimp reared in eutrophic pond water grow significantly faster than shrimp in clear well water, and this growth enhancement is especially pronounced in postlarval shrimp. The objective of this study was to determine if the nutritional benefits of pond water could supplement a lower protein feed for postlarval Pacific white shrimp Litopenaeus vannamei. Sixteen 230-L tanks were stocked with 10-d postlarvae at a density of 350 shrimp/tank. Four treatments (four replicates/treatment) were tested for 6 wk and consisted of: 1) shrimp grown in well water and fed a commercially available 45%-protein feed (W/45); 2) shrimp grown in pond water and fed the same 45%-protein feed (P/45); 3) shrimp grown in well water and fed a commercially available 52%-protein feed (W/ 52); and 4) shrimp grown in pond water and fed the same 52%-protein feed (P/52). At the end of the experiment. mean weight gain (± SE) for shrimp in pond water (1.85 ± 0.03 g) was significantly greater (P > 0.0001) than shrimp in well water (0.98 ± 0.10 g). Mean weight gain for shrimp fed the 52%-protein feed (1.56 ± 0.13 g) was significantly greater (P > 0.0001) than shrimp fed the 45%-protein feed (1.26 ± 0.20 g). In addition, there was a significant interaction effect between water source and feed (P > 0.0001). Mean weight gain for shrimp in the W/52 treatment (1.23 ± 0.04 g) was 68% greater than shrimp in the W/45 treatment (0.73 ± 0.03 g). However, mean weight gain for shrimp in the P/52 treatment (1.90 ± 0.03 g) was only 5% greater than shrimp in the P/45 treatment (1.80 ± 0.04 g). These results suggest that organically rich pond water provides postlarval shrimp with sufficient nutrients to compensate for nutritional deficiencies associated with a lower protein feed. [source]


Different expression of Litopenaeus vannamei (Boone) haemocytes to Vibrio and abiotic particle inoculation

Abstract Penaeid shrimps are among the most studied crustaceans, mainly regarding their immune system, and several proteins involved in the defense against pathogens have been described. Haemocytes are very dynamic cells responsible of recognition, phagocytosis, degranulation and nodule formation, but still little is known about their gene expression. Using differential display, we found modification in haemocyte gene expression after inoculation with an abiotic particle (DEAE-Sephadex) or potential pathogenic bacteria (Vibrio alginolyticus). We also noticed that some of the newly expressed genes are exclusive from a specific treatment. Here we report that haemocytes from Litopenaeus vannamei are capable of recognizing and distinguishing different foreign materials, and respond specifically to each treatment, indicating some specificity in shrimp immune response. Evaluation of In Vitro Apparent Protein Digestibility by Shrimp Using Gut Enzyme Extracts

Knowledge of apparent protein digestibility (APD) is required for optimization of feed formulae for the production of marine penaeid shrimp. The purpose of this study was to evaluate an in vitro method for determining APD in marine penaeid shrimp using gut enzyme extracts. A high correlation (r2 = 0.95) was shown between single-ingredient APD values for fish meal diets using in vivo methodology and those derived from in vitro testing of ingredients. A second study showed positive correlation (r2 = 0.71) between in vitro APD of selected purified and semipurified ingredients and their reported in vivo APDs. This correlation was much higher for purified ingredients (r2 = 0.93) versus less-refined ingredients (r2 = 0.24). A third trial compared in vitro APD at three different enzyme extract pH values and showed that for most protein sources, APD was significantly highest (P < 0.05) at pH = 7.0 and lower at pH = 6.1 or 7.9, indicating a neutral pH optimum for this methodology. Inbreeding Effects on Hatchery and Growout Performance of Pacific White Shrimp, Penaeus (Litopenaeus) vannamei

In animal breeding programs, selection coupled with a narrow genetic base can cause high levels of inbreeding to occur rapidly (in one or two generations). Although the effects of inbreeding have been studied extensively in terrestrial animals and to a lesser extent in aquaculture species, little is known about the effects of inbreeding on penaeid shrimp. The objective of this study was to investigate the effects of inbreeding on hatchery and growout performance of the Pacific white shrimp, Penaeus vannamei. The experiment was conducted over 2 yr, and data from two successive generations (G2 and G3) of inbred (sibling,sibling mating) and outbred families were analyzed. There were 11 inbred and 12 outbred families in G2 and 9 inbred and 10 outbred families in G3. Inbreeding coefficients (F) for outbred and inbred families were 0.00 and 0.25, respectively, for G2 and 0.00 and 0.375, respectively, for G3. Growth rates for outbreds and inbreds were similar in both G2 and G3. Hatch rate for inbred families was 33.1% lower than for outbred families in G2 and 47.1% lower in G3. Inbreeding depression (IBD) (relative change in phenotype per 0.1 increase in F) ± 95% CI for hatch rate was ,12.3 ± 10.1%. Hatchery survival for inbred families was 31.4% lower than for outbred families in G2 and 38.8% lower in G3. IBD for hatchery survival was ,11.0 ± 5.7%. Growout survival was 1.9% lower for inbred families than for outbred families in G2 and 19.6% lower in G3. IBD for growout survival was ,3.8 ± 2.9%. There was also a significant linear relationship between IBD estimates for survival traits and mean outbred survival. At high outbred survival, IBD was low (e.g., growout survival in G2), but IBD appeared to become more severe when outbred survival was lower. This suggests that stress (related to environment and/or life stage) may worsen IBD for survival traits. Results also indicate that moderate to high levels of inbreeding (>10%) should be avoided in commercial shrimp hatcheries because the cumulative effect of IBD on hatch rate and hatchery survival will significantly reduce postlarvae production. Thus, IBD can be significant enough to justify the use of inbreeding as a germplasm protection strategy (under certain scenarios) for genetic improvement programs. [source]


Vibrio harveyi: a significant pathogen of marine vertebrates and invertebrates

Abstract Vibrio harveyi, which now includes Vibrio carchariae as a junior synonym, is a serious pathogen of marine fish and invertebrates, particularly penaeid shrimp. In fish, the diseases include vasculitis, gastro-enteritis and eye lesions. With shrimp, the pathogen is associated with luminous vibriosis and Bolitas negricans. Yet, the pathogenicity mechanisms are imprecisely understood, with likely mechanisms involving the ability to attach and form biofilms, quorum sensing, various extracellular products including proteases and haemolysins, lipopolysaccharide, and interaction with bacteriophage and bacteriocin-like substances. Identification of shrimp species in raw and processed food products by means of a polymerase chain reaction-restriction fragment length polymorphism method targeted to cytochrome b mitochondrial sequences

Abstract A novel PCR-RFLP method has been developed for the identification of six commercially relevant penaeid shrimp species in raw and processed food products. The method can be completed within 8,h. To implement the method, PCR amplification with the crustF/crustR primers, targeted to the amplification of a ca. 181,bp region of the cytochrome b (cytb) mitochondrial gene in penaeid shrimps, was coupled to restriction analysis with CviJI, DdeI and NlaIV. The method was also applied successfully to the identification of shrimp species in complex processed foods, including this type of shellfish as an added-value food ingredient. The small size of this molecular target facilitates amplification from fresh, frozen, or precooked samples, where DNA fragmentation may be relevant and fragment size critical. We also report the first cytb mitochondrial sequences described to date for the species Farfantepenaeus notialis, Parapenaeus longirostris and Pleoticus muelleri, and these nearly triplicate current knowledge of reference nucleotide sequences in this mitochondrial region for this group of species. The cytb mitochondrial gene may also be considered as a molecular marker for identification and phylogenetic purposes in penaeid shrimp species. [source]


Spermatozoal ultrastructure in three Atlantic solenocerid shrimps (Decapoda, Dendrobranchiata)

Abstract The spermatozoal ultrastructure in three solenocerid shrimps (Solenocera membranacea, S. africana, and Pleoticus muelleri) from different Atlantic locations was examined with the aim of increasing understanding of the phylogenetic relationships in the Dendrobranchiata. A considerable structural similarity between the sperm of these species and those of penaeid shrimps supports a close affinity between the Penaeidae and Solenoceridae. However, significant differences in the sperm morphology of the previously investigated sicyoniids (namely, a greater complexity of the acrosomal complex) suggest evolutionary separation of the Sicyoniidae from the assemblage Penaeidae-Solenoceridae. Two ultrastructural features distinguish the spermatozoa of the three studied solenocerids from penaeid sperm: 1) separation of the plasma and acrosome membranes at the base of the spike and anterior region of the cap, and 2) asymmetry of the acrosomal cap, which appears to be a synapomorphy of the group. No striking ultrastructural differences were found between the spermatozoa of the closely related species S. membranacea and S. africana, whereas a great number of morphological differences separate the spermatozoa of Pleoticus from those of Solenocera (e.g., shape of the acrosomal cap, structural arrangement of the contents of the whole acrosome vesicle, thickness and distribution of the cytoplasm, and external shape of the spike). Comparative study of the intracellular superoxide anion production in different penaeid species through the NBT-reduction assay

Abstract The capacity of reactive oxygen intermediates production upon haemocyte stimulation is one of the most important immunoparameter utilized to assess the health status in cultivated shrimps. In the present study, we compared oxidative stress potential, by measuring the superoxide anion production in three penaeid shrimps: two wild Atlantic species, the pink shrimp Farfantepenaeus paulensis and the white shrimp Litopenaeus schmitti and one cultivated Pacific species, the white shrimp, Litopenaeus vannamei, through the nitro-blue-tetrazolium-reduction assay. We also proposed an optimized experimental protocol for this assay, that produces rapid and consistent results with low levels of basal superoxide anion (O2,) production by unstimulated haemocytes and high levels of this oxygen radical after cell stimulation. Among the different cell elicitors used (zymosan, laminarin, lipopolysaccharide and phorbol myristate acetate), laminarin (,-1,3-glucans , 2 mg mL,1) was the most potent cell activator for the haemocytes of all three penaeids and we recommend this immunostimulant to routinely evaluate shrimp respiratory burst. In general terms, the most elevated levels of O2, production, after cell stimulation with microbial components, were detected in L. schmitti. Interestingly, the stimulation profile of the haemocytes of L. vannamei was more similar to F. paulensis, than to L. schmitti, which is more phylogenetically related. [source]
