K. Moss Nursery production may be enhanced by the addition of artificial substrate to increase the surface area upon which shrimp graze and to serve as refuge. The objective of this study was to assess the effects of the artificial substrate, AquaMatsTM, on the performance of postlarval Pacific white shrimp Litopenaeus vannamei stocked at three densities. Eighteen 230-L tanks were stocked with 10-d postlarvae (mean weight < 0.01 g). Six treatments were evaluated and consisted of shrimp stocked at three densities (778 shrimp/m2, 1,167 shrimp/ m2, and 1,556 shrimp/m2) with and without access to artificial substrate. Shrimp in all treatments received a commercial diet ad libitum. After 6 wk, shrimp were harvested from each nursery tank, counted, and batch weighed. Mean final weight, survival, production, feed conversion ratio, and water quality parameters were analyzed by 2-way ANOVA. There were highly significant (P < 0.001) density and substrate effects on final weight, but there was no significant interaction effect. Final weight was 26.0, 17.4, and 34.5% greater in treatments with substrate than without substrate when stocked at 778, 1,167, and 1,556 shrimp/m2, respectively. There was no significant density, substrate, or interaction effect on survival or water quality. Mean survival was ± 89.1% for all treatments. Increased shrimp growth in the presence of added substrate was likely due to the availability of attached particulate organic matter on the AquaMatsTM that served as an additional food source. Results from this study indicate that artificial substrate can be used to mitigate the potential negative effects of high stocking density on growth of L. vannamei in nursery systems. Periphytic diatoms are used as indicators of water quality because their ecological tolerances or preferences to environmental variables are thought to be predictable. However, much of the present autecological information for periphytic diatoms has been derived from studies conducted in the northern hemisphere. In this present study we used periphytic diatoms to determine the impacts of inorganic nutrient pollution in a tidal river system in the temperate latitudes of south-east Australia. In so doing, we assess the suitability of the use of the ,northern hemisphere' ecological tolerance/preference data for periphytic diatoms. 2. Artificial substrates were used to collect periphytic diatoms at 35 sites, which were positioned along the riverbanks and the middle of the river at various distances upstream and downstream of the sewage outfall. The sampling design took into account tidal excursions and the observed sewage plume dynamics. Periphytic diatoms were collected during the austral winter month of August and the austral spring months of September and October. We deployed the artificial substrates for 4 weeks to allow the periphytic diatoms to recruit and colonise, before identifying and enumerating the assemblages. 3. Data analysis included two approaches: multivariate visualisations of combinations of environmental and biological data to investigate shifts in species structure of the periphytic diatom assemblage and multimetric indices based on ecological tolerance/preference data. 4. We found that the spatial patterns inferred from multivariate and multimetric analyses were consistent. Temporal variation in the composition of the periphytic diatom assemblage was greater than the spatial variation along horizontal sections of the river (in any one deployment) due mainly to shifts between winter and spring species. 5. Outfall effects were most apparent in winter, possibly because subsequent deployments were swamped by growth of spring periphytic diatoms. The outfall effects included a shift towards pollutant tolerant species and a reduction in the variability of the periphytic diatom assemblage across the river. 6. We conclude that the use of periphytic diatoms and associated ecological tolerance/preference data as a means of assessing impacts of point source inorganic nutrient pollution is effective. An understanding of river and sewage flow patterns is essential to the design of appropriate monitoring programmes and to the interpretation of results, especially as periphytic diatoms are sensitive to many environmental variables. [source] Sperm surface arylsulfatase A can disperse the cumulus matrix of cumulus oocyte complexes,JOURNAL OF CELLULAR PHYSIOLOGY, Issue 1 2007Alexander Wu Cumulus cell layers of expanded cumulus oocyte complexes (COCs) are interlinked with networks of hyaluronic acid, chondroitin sulfate B proteoglycans and link proteins, and they can be dispersed by sperm surface hyaluronidases. In this report, we showed that arylsulfatase A (AS-A), existing on the sperm head surface, also had this dispersion action. Purified AS-A free of protease, hyaluronidase and chondroitinase activities could disperse the cumulus matrix of expanded COCs. However, this COC dispersion action was not associated with AS-A desulfation activity, assayed by using p -nitrocatecholsulfate (artificial substrate). COCs incubated for 1 h with sperm pretreated with anti-AS-A IgG in the presence of apigenin (a hyaluronidase inhibitor) did not exhibit matrix dispersion, whereas several cumulus layers were already dispersed in COCs incubated with sperm pretreated with preimmune IgG. Furthermore, sperm from AS-A null mice showed a significant delay in COC dispersion, compared with wild-type sperm. Within 1 h of sperm-COC co-incubation, the size of COCs incubated with AS-A null sperm was 65% of the original dimension, whereas that of COCs inseminated with wild-type sperm was only 17%. A further delay in COC dispersion by AS-A(,/,) mouse sperm was observed when apigenin was present in the co-incubation. We also showed for the first time that AS-A had a specific affinity for chondroitin sulfate B, a component of cumulus matrix proteoglycan networks; this might provide a mechanism of cumulus matrix destabilization induced by sperm surface AS-A. [source] Two Ser/Thr protein kinases essential for efficient aggregation and spore morphogenesis in Myxococcus xanthusMOLECULAR MICROBIOLOGY, Issue 6 2006Emily A. Stein Summary Myxococcus xanthus has a complex life cycle that involves vegetative growth and development. Previously, we described the espAB locus that is involved in timing events during the initial stages of fruiting body formation. Deletion of espA caused early aggregation and sporulation, whereas deletion of espB caused delayed aggregation and sporulation resulting in reduced spore yields. In this study, we describe two genes, pktA5 and pktB8, that flank the espAB locus and encode Ser/Thr protein kinase (STPK) homologues. Cells deficient in pktA5 or pktB8 formed translucent mounds and produced low spore yields, similar in many respects to espB mutants. Double mutant analysis revealed that espA was epistatic to pktA5 and pktB8 with respect to aggregation and fruiting body morphology, but that pktA5 and pktB8 were epistatic to espA with respect to sporulation efficiency. Expression profiles of pktA5,lacZ and pktB8,lacZ fusions and Western blot analysis showed that the STPKs are expressed under vegetative and developmental conditions. In vitro kinase assays demonstrated that the RD kinase, PktA5, autophosphorylated on threonine residue(s) and phosphorylated the artificial substrate, myelin basic protein. In contrast, autophosphorylation of the non-RD kinase, PktB8, was not observed in vitro; however, the phenotype of a pktB8 kinase-dead point mutant resembled the pktB8 deletion mutant, indicating that this residue was important for function and that it likely functions as a kinase in vivo. Immunoprecipitation of Tap-tagged PktA5 and PktB8 revealed an interaction with EspA during development in M. xanthus. These results, taken together, suggest that PktA5 and PktB8 are STPKs that function during development by interacting with EspA and EspB to regulate M. xanthus development. [source] Interactions between neighboring algae and snail grazing in structuring microdistribution patterns of periphytonOIKOS, Issue 3 2001Masakado Kawata The micro-distribution of periphyton (filamentous algae) on homogeneous substrates was examined in experimental tanks with and without the pressure of grazing snails. The growth of periphyton attached to artificial substrate was estimated at a small spatial scale (9.3 mm×9.3 mm cells) by varying the number of grazers (0, 5, or 10 snails per tank), using image processing analysis without removing the periphyton. The results suggest that periphyton growth within a cell was negatively affected by the biomass of periphyton in the cell but was positively affected by the biomass of periphyton in neighboring cells. A semivariogram analysis indicated that spatial heterogeneity increased with increasing grazing pressure. The size of patches was not clearly related to the number of snails, but there was a tendency for relative patch size to increase with snail density. Computer simulations were also conducted to examine factors affecting the degree of spatial heterogeneity. The simulation studies indicated that snails should graze a site that was previously grazed in order to produce the observed spatial heterogeneity of periphyton. The results also indicated that the positive effects of neighboring periphyton on the growth of algae might create patches. The interactions among neighboring algae and snail grazing might be an important factor creating the spatial heterogeneity of periphyton even on homogeneous substrates. [source] The chitinolytic system of Lactococcus lactis ssp. lactis comprises a nonprocessive chitinase and a chitin-binding protein that promotes the degradation of ,- and ,-chitinFEBS JOURNAL, Issue 8 2009Gustav Vaaje-Kolstad It has recently been shown that the Gram-negative bacterium Serratia marcescens produces an accessory nonhydrolytic chitin-binding protein that acts in synergy with chitinases. This provided the first example of the production of dedicated helper proteins for the turnover of recalcitrant polysaccharides. Chitin-binding proteins belong to family 33 of the carbohydrate-binding modules, and genes putatively encoding these proteins occur in many microorganisms. To obtain an impression of the functional conservation of these proteins, we studied the chitinolytic system of the Gram-positive Lactococcus lactis ssp. lactis IL1403. The genome of this lactic acid bacterium harbours a simple chitinolytic machinery, consisting of one family 18 chitinase (named LlChi18A), one family 33 chitin-binding protein (named LlCBP33A) and one family 20 N -acetylhexosaminidase. We cloned, overexpressed and characterized LlChi18A and LlCBP33A. Sequence alignments and structural modelling indicated that LlChi18A has a shallow substrate-binding groove characteristic of nonprocessive endochitinases. Enzymology showed that LlChi18A was able to hydrolyse both chitin oligomers and artificial substrates, with no sign of processivity. Although the chitin-binding protein from S. marcescens only bound to ,-chitin, LlCBP33A was found to bind to both ,- and ,-chitin. LlCBP33A increased the hydrolytic efficiency of LlChi18A to both ,- and ,-chitin. These results show the general importance of chitin-binding proteins in chitin turnover, and provide the first example of a family 33 chitin-binding protein that increases chitinase efficiency towards ,-chitin. [source] Unravelling the microbial role in ooid formation , results of an in situ experiment in modern freshwater Lake Geneva in SwitzerlandGEOBIOLOGY, Issue 4 2008K. PLEE ABSTRACT The microbial role in the formation of the cortex of low-Mg calcite freshwater ooids in western part of Lake Geneva in Switzerland has been suggested previously, but not demonstrated conclusively. Early work mostly concentrated in hypersaline milieus, and hence little is known about their genesis in freshwater environments. We designed an in situ experiment to mimic the natural process of low-Mg calcite precipitation. A special device was placed in the ooid-rich bank of the lake. It contained frosted glass (SiO2) slides, while quartz (SiO2) is the most abundant mineral composition of ooid nuclei that acted as artificial substrates to favour microbial colonization. Microscopic inspection of the slides revealed a clear seasonal pattern of carbonate precipitates, which were always closely associated with biofilms that developed on the surface of the frosted slides containing extracellular polymeric substance, coccoid and filamentous cyanobacteria, diatoms and heterotrophic bacteria. Carbonate precipitation peaks during early spring and late summer, and low-Mg calcite crystals mostly occur in close association with filamentous and coccoid cyanobacteria (e.g. Tolypothrix, Oscillatoria and Synechococcus, Anacystis, respectively). Further scanning electron microscope inspection of the samples revealed low-Mg calcite with crystal forms varying from anhedral to euhedral rhombohedra, depending on the seasons. Liquid cultures corroborate the in situ observations and demonstrate that under the same physicochemical conditions the absence of biofilms prevents the precipitation of low-Mg calcite crystals. These results illustrate that biofilms play a substantial role in low-Mg calcite ooid cortex formation. It further demonstrates the involvement of microbes in the early stages of ooid development. Combined with ongoing microbial cultures under laboratory-controlled conditions, the outcome of our investigation favoured the hypothesis of external microbial precipitation of low-Mg calcite as the main mechanism involved in the early stage of ooid formation in freshwater Lake Geneva. [source] In vitro evaluation of sun protection factors of sunscreen agents using a novel UV spectrophotometric techniqueINTERNATIONAL JOURNAL OF COSMETIC SCIENCE, Issue 4 2008M. D. Bleasel Synopsis A method for the in vitro determination of low- and high-value sun protection factors (SPF) of sunscreens using artificial substrates and a novel pseudo double beam (PDB) mode of operation of a standard double beam UV spectrophotometer is described. The method allows transmittance to be calculated from detector responses of reference and sample beams measured at different gain levels and facilitates the accurate quantification of low levels of electromagnetic radiation transmitted through highly absorbing samples. The spectrophotometer was modified to hold quartz diffusing plates on which a substrate [TransporeÔ adhesive tape or human stratum corneum obtained from a skin surface biopsy (SSB)] and the sunscreens to be tested were applied. The PDB mode of operation increased the effective linear range of the detector response of the spectrophotometer by a factor of approximately 20000-fold, enabling the in vitro SPF determination technique to be applied to both high and low SPF value sunscreens. Eight commercial sunscreens with known SPF values ranging from 4 to 77, previously determined by in vivo methods, were tested in vitro using both test substrates and correlations between the in vivo and in vitro values were determined. SPF values determined using the in vitro method correlated well with the known in vivo results (TransporeÔ tape, R2 = 0.611; SSB, R2 = 0.7928). The in vitro SPF obtained for one of the tested products differed substantially from the cited in vivo SPF value. Independent in vitro and in vivo re-evaluation of the SPF of this product matched the value predicted by the present method much more closely than the originally cited in vivo value. All determined SPFs were ordered correctly in comparison to in vivo ranking and the technique appeared to correctly identify a sunscreen that had a labelled SPF value that was significantly higher than its true SPF. Résumé Une méthode destinée à déterminer in vitro les facteurs de protection solaire (SPF) d'écrans solaires de faible et haut indice est décrite. Elle met en ,uvre des substrats artificiels et un nouveau mode opératoire reposant sur l'utilisation du pseudo double faisceau (PDB) d'un spectrophotomètre UV double faisceau standard. La méthode permet le calcul de la transmittence à partir des réponses du détecteur de référence et la mesure en simple faisceau à différents niveaux de gain facilitant ainsi la quantification précise des faibles niveaux de radiation électromagnétique (EMR) transmis à travers des échantillons hautement absorbants. Le spectrophotomètre a été modifié de façon à fixer des plaques diffusantes en quartz sur lesquelles un substrat (ruban adhésif Transport TM ou du stratum corneum humain obtenu à partir de biopsie de surface de peau (SSB) et les écrans solaires testés ont été appliqués. Le mode opératoire PTB augmente la gamme linéaire effective de la réponse du détecteur du spectrophotomètre d'un facteur approximatif 20.000 permettant, à cette technique de détermination des SPF in vitro, d'être appliquée à la fois sur les écrans solaires de haut et bas SPF. Huit écrans solaires commerciaux de SPF connus allant de 4 à 77, préalablement déterminés par des méthodes in vivo, ont été testés in vitro en utilisant les deux substrats, et les corrélations entre les valeurs in vivo et in vitro ont été déterminées. Les valeurs SPF déterminées en utilisant la méthode in vitro est bien corrélée avec les résultats in vivo connus (ruban transport, R2 = 0.611; SSB, R2 = .7928). McCabe Summary 1We used natural and manipulative field experiments to examine the effects of caddisfly (Trichoptera) diapause aggregations on benthic macroinvertebrates communities in a Vermont river. 2Natural substrates with aggregations of Neophylax and Brachycentrus (Trichoptera: Uenoidae and Brachycentridae) had higher species richness than did substrates lacking aggregations. Aggregations of caddisfly cases added to artificial substrates (bricks) also accumulated greater abundance, species density (number of species per unit area), and species richness (number of species per standard number of individuals) than did control bricks. 3Low-density, uniformly spaced, Brachycentrus cases accumulated higher species density and species richness than did an equivalent density of clumped cases. Similarly, empty Neophylax cases accumulated higher diversity than did cases still occupied by Neophylax pupae. 4Although natural substrates had higher species richness than artificial substrates, substrate type did not change qualitatively the effect of caddisfly aggregations on species richness. 5We subsampled individuals randomly from aggregations and control surfaces to provide an estimate of species richness unbiased by abundance. Expected species richness was higher in aggregations than on control surfaces. These results suggest that caddisfly aggregations increase species density by altering the shape of the species,abundance distribution as well as by accumulating individuals and species passively. 6We conclude that caddisfly diapause aggregations increase habitat complexity and facilitate colonization of other benthic species.
