Developmental, metabolic and immunological costs of flea infestation in the common vole

Summary 1Parasites use resources from their hosts, which can indirectly affect a number of host functions because of trade-offs in resource allocation. In order to get a comprehensive view of the costs imposed by blood sucking parasites to their hosts, it is important to monitor multiple components of the development and physiology of parasitized hosts over long time periods. 2The effect of infestation by fleas on body mass, body length growth, haematocrit, resistance to oxidative stress, resting metabolic rate and humoral immune response were experimentally evaluated. During a 3-month period, male common voles, Microtus arvalis, were either parasitized by rat fleas (Nosopsyllus fasciatus), which are naturally occurring generalist ectoparasites of voles, or reared without fleas. Then voles were challenged twice by injecting Keyhole Limpet Haemocyanin (KLH) to assess whether the presence of fleas affects the ability of voles to produce antibodies against a novel antigen. During the immune challenge we measured the evolution of body mass, haematocrit, resistance to oxidative stress and antibody production. 3Flea infestation negatively influenced the growth of voles. Moreover, parasitized voles had reduced haematocrit, higher resting metabolic rate and lower production of antibodies against the KLH. Resistance to oxidative stress was not influenced by the presence of fleas. 4During the immune challenge with KLH, body mass decreased in both groups, while the resistance to oxidative stress remained stable. In contrast, the haematocrit decreased only in parasitized voles. 5Our experiment shows that infestation by a haematophageous parasite negatively affects multiple traits like growth, energy consumption and immune response. Fleas may severely reduce the survival probability and reproductive success of their host in natural conditions. [source]


Gregarious development of the solitary endo-parasitoid, Microplitis rufiventris in its habitual host, Spodoptera littoralis

Abstract:, Earlier research has shown that the koinobiont parasitoid, Microplitis rufiventris, attacks and can develop on early instars of Spodoptera littoralis larvae with preference to third instars. However, the present study was carried out using the newly moulted sixth instar larvae at two different temperatures (20 ± 1 and 27 ± 1°C) to study the developmental interaction between the parasitoid and the last instar host larvae. Parasitoid eggs laid in singly parasitized host larvae invariably died. As the number of parasitoid eggs/host larvae increased, the proportion of eggs that hatched and number of viable parasitoid larvae successfully reached to their final instar increased. The effect of superparasitization seems to be dose (no. of eggs + parasitoid factors)-temperature-dependent. The results demonstrate a kind of ,Allee effect' suggesting that superparasitized last instar S. littoralis larvae provide a better host environment than singly parasitized hosts for the parasitoid, M. rufiventris. This may be due to host's hormone and/or low dose of factors injected with parasitoid eggs. The supernumerary individuals of wasp larvae developed normally to the point of emergence but most did not successfully emerge from the host. The improvement of both hatchability and post-embryonic development of parasitoid wasp was significantly (P < 0.01) greater at 20°C than at 27°C. The results of the present study are useful in understanding the evolution of life-history strategies and host range in parasitic hymenoptera. [source]


Evidences of non-additive effects of multiple parasitoids on Diatraea saccharalis Fabr. (Lep., Crambidae) populations in sugarcane fields in Brazil

Abstract: Biological control is a relatively benign method of pest control. However, considerable debate exists over whether multiple natural enemies often interact to produce additive or non-additive effects on their prey or host populations. Based on the large data set stored in the São João and Barra sugarcane mills (state of São Paulo, Brazil) regarding the programme of biological control of Diatraea saccharalis using the parasitoids Cotesia flavipes and tachinid flies, in the present study the author investigated whether the parasitoids released into sugarcane fields interfered significantly with the rate of parasitized D. saccharalis hosts. The author also observed whether there was an additive effect of releasing C. flavipes and tachinids on the rate of parasitized hosts, and looked for evidence of possible negative effects of the use of multiple parasitoid species in this biological control programme. Results showed that C. flavipes and the tachinids were concomitantly released in the Barra Mill, but not in the São Jão Mill. Furthermore, in the Barra Mill there was evidence that the parasitoids interacted because the percentage of parasitism did not increase after the release of either C. flavipes or tachinids. In the São João Mill, when both parasitoid species were released out of synchrony, both the percentage of parasitism by C. flavipes as well as that of the tachinids increased. When large numbers of tachinids were released in the Barra Mill, they caused a significant lower percentage of parasitism imposed by C. flavipes. The implications of the results as evidence of non-additive effects of C. flavipes plus tachinids on D. saccharalis populations are discussed. [source]


Parasitic castration of Plutella xylostella larvae induced by polydnaviruses and venom of Cotesia vestalis and Diadegma semiclausum

Abstract In the present study, we used ,-ray to irradiate the female parasitoids to make wasp eggs infertile, resulting in pseudoparasitization, which allowed the analysis of maternal secretions such as polydnaviruses (PDVs) and venom in the absence of larval secretions or teratocytes by the growing parasitoids. We then investigated the spermatogenesis and components of testicular proteins of male Plutella xylostella larvae pseudoparasitized by two endoparasitoids (Cotesia vestalis and Diadegma semiclausum). The results showed that pseudoparasitism by the two endoparasitoids at the early third instar host larvae both induced smaller testes in size than those of nonparasitized host larvae. Both of them caused parasitic castration, and the degree of castration is almost as severe as in naturally parasitized hosts. This suggested that PDVs and venom played a major role in the degeneration of host testes. There are significant differences in the degree of castration induced by the two endoparasitoids, with respect to testicular growth, testicular protein concentrations, and histological changes of germ cells. Cotesia vestalis bracovirus always has a significantly stronger effect on host testicular growth and development than D. semiclausum ichnovirus. SDS-PAGE analysis indicated that synthesis of P 65 and P 67 proteins were clearly inhibited in testes of hosts that were pseudoparasitized by C. vestalis while reduction in synthesis of other proteins was not evident.
