Reproductive features of the non-native Siganus luridus (Teleostei, Siganidae) during early colonization at Linosa Island (Sicily Strait, Mediterranean Sea)

Summary In July 2003, the finding of a newly settled population of Siganus luridus at Linosa Island (Sicily Strait, Mediterranean Sea) gave us the unusual opportunity to examine the reproductive condition of a Lessepsian migrant during early phases of colonization. Aspects of gonad morphology, fecundity, atresia and oocyte dynamics were investigated by using 43 pioneer specimens collected in concomitance with their first record in the Pelagie Islands. Ovarian development was consistent with the group-synchronous type, and testicular organization was of the unrestricted spermatogonial testis type, with cystic spermatogenesis. Both males and females had reached final stages of gonad maturation. The rates of follicular atresia were moderate: out of 17 adult females, 10 individuals did not present atretic oocytes; six exhibited <15.1% of secondary growth phase (SGP) oocytes in , -atresia, while one female presented 45.7% of SGP in , -atresia. Fecundity estimates did not diverge from what was observed in a reference population along the Lebanese coast. Absolute fecundity ranged from 115 739 to 740 433 oocytes per female (16.5,24.5 cm LT). Relative fecundity ranged from 1239 to 3162 oocytes g,1, with a mean of 1885 ± 868 oocytes g,1. Our observations indicated that these early settled siganids are reproductively active at Linosa and suggested the forthcoming of self-maintaining populations across the central Mediterranean area. Ovarian dynamics and oosorption in two species of predatory lady beetles (Coleoptera: Coccinellidae)

Abstract Ovarian development and oviposition dynamics of two species of lady beetle, Coccinella septempunctata L. and C. transversoguttata richardsoni Brown, are examined in laboratory experiments in which pea aphids (Acyrthosiphon pisum Harris) are provided ad libitum and then removed. Both species respond to prey removal by stabilizing at lower body weights, and by laying progressively fewer and smaller eggs of reduced viability, until oviposition ceases altogether after several days of starvation. Dissections of females after prey removal reveal similar patterns of oosorption in both species. However, C. septempunctata reduces oviposition more rapidly after prey removal than does C. transversoguttata. When prey are again provided, C. septempunctata soon lay as many eggs as previously but C. transversoguttata lay fewer. Females, especially of C. septempunctata, that stop producing and resorb eggs in the absence of prey lay more eggs subsequently than do females that feed continually on prey provided ad libitum and lay eggs throughout the experiment. Thus, although both species are responsive to a rapid change in prey availability, C. septempunctata appears to be especially responsive. Rapid responses to changes in prey availability may contribute to the greater abundance and reproductive success of this introduced species relative to the native C. transversoguttata in western North American alfalfa fields that exhibit widely varying pea aphid densities. However, both species engage in oosorption as a means of reserving resources under poor prey conditions and enhancing future reproductive effort when prey conditions improve. [source]


Sexual dimorphism of g-protein subunit Gng13 expression in the cortical region of the developing mouse ovary

Abstract In our search for genes required for the development and function of mouse gonads, we identified Gng13 (guanine nucleotide binding protein 13, gamma), a gene with an embryonic expression pattern highly restricted to the ovary. Based on reverse transcriptase-polymerase chain reaction (RT-PCR) and whole-mount in situ hybridization, Gng13 is expressed in both XX and XY gonads at embryonic day (E) 11.5, but becomes up-regulated in the XX gonad by E12.5. Expression is retained after treatment with busulfan, a chemical known to eliminate germ cells, pointing to the soma as a site of Gng13 transcription. In situ hybridization of embryonic ovarian tissue sections further localized the expression to the cortex of the developing XX gonad. Gng13 expression in the adult is also highly restricted. Northern blot analyses and Genomic Institute of the Novartis Research Foundation expression profiling of adult tissues detected very high expression in the cerebrum and cerebellum, in addition to, a weaker signal in the ovary. Gng13 belongs to a well-known family of signal transduction molecules with functions in many aspects of development and organ physiology. Here, we report that, in the developing mouse embryo, expression of Gng13 mRNA is highly restricted to the cortex of the XX gonad during sexual differentiation, suggesting a role for this gene during ovarian development. Neonatal estrogen exposure inhibits steroidogenesis in the developing rat ovary

Abstract Treatment of newborn female rats with estrogens significantly inhibits the growth and differentiation of the ovary. To understand the molecular mechanism of estrogen action in the induction of abnormal ovary, we examined the expression profiles of steroidogenic factor 1 (SF-1) and several of its target genes in the developing ovaries after neonatal exposure to synthetic estrogen, estradiol benzoate (EB) by using reverse transcriptase polymerase chain reaction, in situ hybridization, and immunohistochemistry. Morphologic examination indicated inhibitory effects of estrogen on the stratification of follicles and development of theca and interstitial gland during postnatal ovarian differentiation. The expression of the steroidogenic acute regulatory protein (StAR) and cholesterol side-chain cleavage cytochrome P450 (P450SCC), which are both essential for steroid biosynthesis, markedly decreased in theca and interstitial cells throughout the postnatal development of the EB-treated ovary. However, expression of the transcriptional activator of the two genes, SF-1 was unaffected in theca and interstitial cells, although the number of these cells was lower in the EB-treated ovary than in the control ovary. The expression of the estrogen mediator, estrogen receptor-, (ER-,), diminished specifically in theca cells at P6 and recovered by P14 in the EB-treated ovary. These results indicate that the effect of estrogens is mediated by means of ER-, resulting in the down-regulation of StAR and P450SCC genes during early postnatal development of the ovary. These results suggest that the abnormal ovarian development by neonatal estrogen treatment is closely correlated with the reduced steroidogenic activity, and the data obtained by using this animal model may account in part the mechanism for aberrant development and function of the ovary in prenatally estrogen-exposed humans. © 2001 Wiley-Liss, Inc. Bimodal life cycle of the burying beetle Nicrophorus quadripunctatus in relation to its summer reproductive diapause

Abstract 1. Under natural conditions in Kyoto, Japan, the reproductive activities of Nicrophorus quadripunctatus Kraatz (Coleoptera: Silphidae) decreased in summer and the species showed a bimodal life cycle. 2. In the laboratory, most adult pairs raised at 20 °C under a LD 12:12 h regime reproduced when provided with a piece of chicken. In adults raised at 20 °C under a LD 16:8 h regime, however, both reproductive behaviour and ovarian development were reduced. It is concluded that these adults entered a reproductive summer diapause. 3. High temperature (25 °C) also suppressed the reproductive behaviour even under a favourable LD 12:12 h regime. In the field, therefore, adults reduce their reproductive activity in summer because of diapause induced by long-day photoperiods and direct inhibition of reproduction by high temperatures. 4. When the temperature was changed from 20 °C to 25 °C immediately after hatching of larvae, they reached the wandering stage in 95% of adult pairs. When the temperature was changed from 20 °C to 25 °C immediately after oviposition, however, no larvae hatched in 85% of pairs. Egg mortality was significantly higher at 25 °C than at 20 and 22.5 °C; no eggs hatched at 27.5 °C. The physiological mechanisms for reducing reproduction probably prevent the beetles from inefficient oviposition in summer. [source]


Quantitative aspects of the regulation of ovarian development in selected anautogenous Diptera: integration of endocrinology and nutrition

Abstract Aspects of the influence of nutrition on the degree of ovarian development in selected anautogenous Diptera are reviewed. The Diptera considered are several mosquito species, with emphasis on Aedes aegypti, the house fly, Musca domestica, the Australian bush fly, M. vetustissima and the blowflies, Lucilia cuprina and Phormia regina. All the selected species display discrete ovarian cycles in which all oocytes destined to reach maturity in a particular ovarian cycle develop synchronously. In these species, the proportion of females maturing oocytes and, where such data exist, the number of oocytes they mature are positively correlated with the amount of any particular nitrogen-containing material ingested or given by enema or by infusion. In addition, the degree of ovarian development may be affected by the chemical composition of nitrogen-containing food. Possible physiological bases for the observed relationships are discussed. Available evidence suggests that whether or not a female matures any oocytes is hormonally regulated and that the number of oocytes matured is probably regulated by the availability of nutrients. Some approaches that might further elucidate the physiological regulatory mechanisms involved in ovarian development are outlined. [source]


Difference in the sensitivity to chemical compounds between female and male neonates of Daphnia magna

Abstract Daphnia magna usually produce female offspring by parthenogenesis, and thus only female neonates are used to evaluate the environmental toxicity to chemicals. Additionally, it is known that male daphnids are induced by exposure to a juvenile hormone, methyl farnesoate, during late ovarian development. In this study, we investigated the concentration of methyl farnesoate in a 24-h exposure producing 100% males, and the difference in sensitivity to chemical compounds, potassium dichromate, pentachlorophenol, and paraquat, between females and males, referring to OECD Test Guideline 202. The results show that the minimum concentration for 100%-male induction of methyl farnesoate in adult females was 50 ,g/L. In addition, acute toxicity tests (immobility test) with the other chemicals showed that male neonates have higher tolerance to potassium dichromate and pentachlorophenol than females for at least 24 h after birth, while no sex difference was observed in the sensitivity to paraquat. The differences in the median effective concentrations in these compounds between female and male neonates suggest two different overall modes of action. Using female daphnids for environmentally toxicity testing seems reasonable, since the females are more sensitive to chemicals than males. Gonadal development and spawning cycle in the digynic hermaphrodite sharpsnout seabream Diplodus puntazzo (Sparidae) off the Canary Islands, northwest of Africa

Summary Diplodus puntazzo is a species characterized by a rudimentary hermaphroditism, with a low level of protandry. The study found that the overall sex ratio was one male to 2.03 females, diverging significantly from 1 : 1, and that there was a ratio of one male to 0.11 transitional fish. The sex ratio varied with season: during the reproductive period the ratio was 1 : 1.86 (male : female), significantly lower than in the non-reproductive period in which the sex ratio was approximately 1 : 2.26. Both sexes had similar size distributions; however, females were predominant in all size intervals, especially in the larger ones. No significant difference in mean total length (TL) between females (330 mm), males (319 mm) or transitionals (321 mm) was found. The smallest functional female and male analysed were 159 and 157 mm TL, and the largest were 520 and 510 mm TL, respectively. No significant difference in length at first level of maturity, attained at the second year of life, was found between males (281 mm) and females (292 mm), respectively. A protracted spawning season was identified for the species: from September to February, with a peak in November. GSI values followed the same pattern for males and females, with values of females being similar to those of males; the highest values occurred between September and December, with a maximum in November. Occurrence of transitional fish was significantly higher during the month with highest values of GSI than in the resting period. The study found that vitellogenesis did not begin synchronously for all females, and the simultaneous presence of oocytes in all stages of development indicated an asynchronous mode of ovarian development. The presence of postovulatory, together with tertiary yolk vesicle oocytes, indicated that the species is a multiple spawner. [source]


Sonographic assessment of uterine and ovarian development in normal girls aged 1 to 12 years

Abstract Purpose. To provide normal references of sonographic uterine and ovarian size in girls aged 1,12 years. Method. Ninety-nine girls were enrolled in the study (mean age ± SD, 6.9 ± 2.4 years [range, 1,12 years]). Pubertal status was classified according to Tanner staging, whereas for height and weight assessment a standard stadiometer and weight scale were employed. All subjects underwent pelvic sonographic examination for the measurement of uterine length, volume, ratio of anteroposterior diameter at the fundus divided by the anteroposterior diameter at the cervix (fundal,cervical [F/C] ratio), and ovarian volume and morphology. Results. A gradual increase with age was observed in all uterine and ovarian measurements. Cubic model analysis provided the best curve estimation for uterine length, uterine volume, and ovarian volume in relation to age. Uterine length, uterine volume, ovarian volume and F/C ratio were significantly correlated to both age and height. With respect to ovarian morphology, there was a gradual decrease in frequency of the homogeneous and the paucicystic appearances with increasing age. The macrocystic appearance was observed after the age of 6 years, and its frequency increased gradually with age. Conclusion. There is a continuous increase in size of internal female genitalia from early childhood until the onset of puberty. Reproductive cycle and sex inversion in razor fish, a protogynous labrid in the southern Mediterranean Sea

The reproductive biology of the Mediterranean razor fish Xyrichthys novacula was investigated by demographic data and histological analysis of the female, intersexual and male gonads. Specimens were collected by bottom trawl on a monthly basis between June 2000 and July 2001 in a sandy bay in southern Thyrrenian. Gonad histology confirmed that the Mediterranean razor fish is a monandric, protogynous hermaphrodite. Females reached first sexual maturity at 100 mm (LT) and the estimated mean LT at first maturity (L50) was 125 mm. Females exhibited asynchronous ovarian development and multiple ovulations occurred over the spawning period. Vitellogenesis started in early May and spawning occurred from late May until late September. Sexual transition involved a large-scale atresia of all oocyte stages and a massive degeneration of ovarian tissue followed by primordial germ cells proliferation. Sex change began at spawning time (June) but transitional individuals tended to cluster at the end of the reproductive period (September). They accounted for 17·1% of the population sampled and were found in a broad size range (105,150 mm LT). [source]


Micro- and macroscopic characteristics to stage gonadal maturation of female Baltic cod

A set of histological characteristics to judge ovarian development was established and used to elaborate morphological criteria of 10 maturity stages of Baltic cod Gadus morhua sampled throughout the annual cycle to represent different macroscopic maturity stages. The applied characteristics confirmed most stages of the macroscopic scale, but the separation of late immature and resting mature females remained imprecise. Atretic vitellogenic oocytes or encapsulated residual eggs identified the resting condition morphologically, but not all ovaries with visible signs of previous spawning showed such features. One ovarian stage that was previously classified as ,ripening' was changed to ,spawning', owing to the prevalence of hydrated eggs and empty follicles. Ovaries with malfunctions were defined by a separate stage. Macroscopic criteria were revised by comparing the gross anatomy of ovaries with their histology. Female length and gonado-somatic index supported stage definitions, but substantial variation in Fulton's condition factor and the hepato-somatic index rendered these of little use for this purpose. The time of sampling influenced staging accuracy. A female spawner probability function based on the proportion of ripening and ripe specimens in early spring seems to be the most appropriate method to estimate spawner biomass and reproductive potential. Winged presoldiers induced by a juvenile hormone analog in Zootermopsis nevadensis: Implications for plasticity and evolution of caste differentiation in termites

Abstract To elucidate the switching mechanism of caste differentiation in termites and to examine the possible induction of soldier-reproductive intercastes experimentally, we investigated the effects of juvenile hormone on the morphologies of soldier caste by applying a juvenile hormone analog (JHA) to nymphs of the damp-wood termite Zootermopsis nevadensis (Isoptera : Termopsidae). JHA treatment for about 2 weeks induced a variety of intermediate castes, showing both alate and soldier morphological features. The principal component analysis (PCA) of those morphological characters showed that those intercastes were a deviation from the developmental line into alates to soldier differentiation, which is known to be triggered by juvenile hormone. Detailed morphological examination of the compound eyes, wing joint, and mandibles showed that those intercastes expressed soldier features, although they had started to develop alate characteristics. The morphology of the resultant intercastes seemed to be determined by the nymphal stage, at which JHA treatment was applied. The induced intercastes with exaggerated soldier-specific characteristics (e.g., mandibles) repressed alate-specific characteristics (e.g., wings), namely, the alate and soldier morphological characteristics in induced intercastes show opposite responses against the application of JHA. On the other hand, ovarian development was not suppressed by the JHA application, even in the soldier-like individuals. Naturally differentiated presoldiers also possessed developed ovarioles, although ovaries of mature soldiers were degenerated. Anatomy and functions of brain neurosecretory cells in diptera

Abstract In the larval brain of dipteran insects, there are two medial and three lateral groups of neurons innervating the ring gland. One lateral group extends fibers to the corpus allatum. After metamorphosis, a large cluster of the medial group in the pars intercerebralis and two lateral groups in the pars lateralis innervate the retrocerebral complex and some neurons from the lateral group and a few from the medial group extend fibers to the corpus allatum in the adults. Neuropeptides such as insulin-like peptides, FMRFamide related peptides, Locusta -diuretic hormone, ,-pigment dispersing hormone, Manduca sexta -allatostatin, ovary ecdysteroidogenic hormone, and proctolin have been immunocytochemically revealed in medial groups in the pars intercerebralis, and FMRFamide related peptides, ,-pigment dispersing hormone, corazonin, and M. sexta -allatostatin in lateral groups in the pars lateralis of dipteran brains. In mosquitoes after the blood meal, ovary ecdysteroidogenic hormone from 2,3 pairs of medial neurosecretory cells is released at the corpus cardiacum to stimulate the ovaries to secrete ecdysteroid to cause ovarian development. In addition to ovarian development, removal and implantation experiments have shown that neurosecretory cells in the pars intercerebralis and pars lateralis are involved in control of reproductive diapause, cuticular tanning, sugar metabolism, and diures. Expression and downregulation of WNT signaling pathway genes in rhesus monkey oocytes and embryos

Abstract Mammalian WNT genes encode secreted glycoproteins that are conserved homologues of the Drosophila Wingless gene, which plays a crucial role in Drosophila development. Recently, WNT pathway signaling has been implicated in ovarian development, oogenesis, and early development. We sought to evaluate whether these genes may contribute to the formation of healthy human oocytes or embryos, and whether the expression of these genes could provide informative markers of human oocyte and embryo quality. To do this, we employed the primate embryo gene expression resource (PREGER; www.preger.org) to examine expression of mRNAs encoding 38 components of the WNT signaling pathway in rhesus monkey oocytes and embryos as a nonhuman primate model. We observed considerable conservation between rhesus monkey and mouse of expression of WNT, FZD, and effector gene mRNAs, and a generalized downregulation of genes encoding key components of the WNT signaling pathway during preimplantation development. Our results support a role for WNT signaling during oocyte growth or maturation, but not during preimplantation development. Additionally, we observed differences between in vitro cultured and in vivo developing blastocysts, indicating possible effects of culture on WNT signaling during the peri-implantation period. Mol. Mode of action of methoprene in affecting female reproduction in the African malaria mosquito, Anopheles gambiae

Abstract BACKGROUND: One of the most studied actions of juvenile hormone (JH) is its ability to modulate ecdysteroid signaling during insect development and metamorphosis. Previous studies in mosquitoes showed that 20-hydroxyecdysone (20E) regulates vitellogenin synthesis. However, the action of JH and its mimics, e.g. methoprene, on female reproduction of mosquitoes remains unknown. RESULTS: Here, a major malaria vector, Anopheles gambiae Giles, was used as a model insect to study the action of methoprene on female reproduction. Ecdysteroid titers and expression profiles of ecdysone-regulated genes were determined before and after a blood meal. An ecdysteroid peak was detected at 12 h post blood meal (PBM). The maximum expression of ecdysone-regulated genes, such as ecdysone receptor (EcR), hormone receptor 3 (HR3) and vitellogenin (Vg) gene, coincided with the ecdysteroid peak. Interestingly, topical application of methoprene at 6 h PBM delayed ovarian development and egg maturation by suppressing the expression of ecdysone-regulated genes in female mosquitoes. CONCLUSION: The data suggest that ecdysteroid titers are correlated with Vg synthesis, and methoprene affects vitellogenesis by modulating ecdysteroid action in A. gambiae. Potential effects of tyramine on the transition to reproductive workers in honeybees (Apis mellifera L.)

Abstract To explore the role of brain tyramine in reproductive worker honeybees, its effects after injection and oral treatment on brain dopamine levels and ovarian development in queenless worker honeybees are determined. Both tyramine injection and oral treatment in 10-day-old queenless bees leads to tyramine transportation into the brain and significantly elevates brain dopamine levels as a function of the tyramine concentration. Ovarian diameters are significantly larger in 10-day-old queenless bees treated with tyramine compared with queenless bees of the same age without tyramine treatment. Results on yolk formation in the ovary support the finding of increased ovarian diameter, suggesting that oral tyramine treatment accelerates ovarian development through dopamine effects and/or direct effects of tyramine on the ovary in queenless bees. Thus, tyramine has potential effects on the enhancement of brain dopamine levels and the acceleration of ovarian development for the transition of normal workers to reproductive worker honeybees. Photoperiod and temperature affect the life cycle of a subtropical cockroach, Opisoplatia orientalis: seasonal pattern shaped by winter mortality

Abstract.,Opisoplatia orientalis is an ovoviviparous cockroach living in the subtropical areas in Japan. Both adults and nymphs overwinter on Hachijo Island (33°N). The nymphs sampled before and after overwintering showed a similar pattern in frequency distribution of head widths with a definite peak of fifth instars. The present study was conducted to determine how this pattern was formed by investigating the effects of photoperiod and temperature on development and reproduction. Photoperiod influenced the number of nymphal instars, resulting in a longer duration of nymphal development at LD 12 : 12 h than at LD 16 : 8 h. However, the rate of development at each instar was only affected to a small extent by photoperiod and no sign of diapause was detected. It was suggested that the photoperiodic response controlling the number of nymphal instars might have evolved to adjust the timing of adult emergence and reproduction to the favourable season. The prereproductive period and time intervals between nymph depositions were prolonged as temperature declined, but there was no evidence for diapause in adults. Mortality occurred in eggs and embryos inside of the body of the females during winter. Thus, it was inferred that female adults would reset ovarian development in spring and deposit nymphs in summer simultaneously, and these nymphs would reach the fifth instar before winter comes. This winter mortality hypothesis was supported by experiments in which reproductive activity and mortality were monitored for field-collected adults under either constant or changing temperature conditions simulating those in the field. The biosynthesis of Juvenile Hormone, its degradation and titres in females of the true armyworm: a comparison of migratory and non-migratory populations

Summary In a previous study [McNeil et al. (1996) Archives of Insect Biochemistry and Physiology, 32, 575,584], patterns of sexual maturation and Juvenile Hormone (JH) biosynthesis were compared in virgin females from migratory (North American) and non-migratory (Azorean) populations of the true armyworm moth, Pseudaletia unipuncta Haworth (Lepidoptera: Noctuidae). Sexual maturation occurred at a significantly earlier age after emergence in the non-migrant population, and the rates of biosynthesis of JH in vitro suggested that lower titres of JH may be required to initiate the onset of calling behaviour (pheromone emission) and ovarian development in Azorean females. To examine the physiological differences in the reproductive biology of migratory and non-migratory populations in greater detail, the haemolymph titres of JH and JH esterase activity were compared in virgin females as a function of age. In addition, the effects of mating on JH biosynthesis in vitro, JH titres, JH esterase activity and egg production were measured in the two populations. As expected, JH titres rose more rapidly after emergence in Azorean females than in their North American counterparts but, contrary to our prediction, the maximum levels were also higher in the non-migrant population. Activity of JH esterase was much higher in Azorean females on the day of emergence. However, by the second day both populations had similar activity levels (about 17 nmol JH/min/ml) and exhibited a similar age-related decline in subsequent days. Mating did not affect the rate of JH biosynthesis in vitro but resulted in a significant increase in the titres of JH in the haemolymph of both populations. The maximum titre (a five-fold increase) occurred within 24 h of mating in Azorean females. In North American individuals the increase was greater (seven-fold) but did not occur until 48 h after mating. No difference in the activity of JH esterase was observed between mated and virgin North American females. By contrast, while there was an age-related decline in the activity of JH esterase in mated Azorean females, as seen in both North American groups, activity levels in virgin females remained constant with age. In all females, mating resulted in a significant increase in egg production within 24 h. The Azores is a volcanic archipelago, so these non-migratory populations were probably founded by immigrants originating from migratory continental populations. It is clear from our results that the change from a life history that includes migration to a non-migratory one involved more than just a temporal shift in the timing of the production of JH. Furthermore, the interpopulation differences in titres of JH and mating-induced changes reported here cannot be fully explained by the observed differences in the patterns of activity of JH esterase and JH biosynthesis in vitro. Effects of dietary phospholipid and highly unsaturated fatty acid on the gonadal development, tissue proximate composition, lipid class and fatty acid composition of precocious Chinese mitten crab, Eriocheir sinensis

Abstract Precocious puberty is a major constraint to the economical rearing Eriocheir sinensis farming. Although dietary phospholipid (PL) and highly unsaturated fatty acids (HUFA) reportedly enhanced ovarian development in normal adult E. sinensis, it had opposite effects of reducing precocity in juveniles. This study investigated the effects of dietary PL and HUFA on survival, gonadal development and biochemical composition of precocious E. sinensis. Two diets with PL and HUFA supplementation (diet A) and deficient (diet B) were formulated and fed to precocious E. sinensis. Although no significant differences were found on survival and gonadosomatic index of crabs fed both diets, crabs fed diet B had significantly higher hepatosomatic index and hepatopancrean lipid content than crabs fed diet A. The percentages, 14:0, 18:1n-9, 20:5n-3 and 22:6n-3, in hepatopancreas, muscle and gonads were positively correlated to the diets. Interestingly, the crabs fed diet B accumulated significantly higher 18:2n-6 in gonads and muscle than those fed diet A. Furthermore, substantially higher 20:4n-6 was detected in the gonad and muscle than in the diets. These results suggested that dietary PL and HUFA had limited effects on male gonad biochemical composition. Meanwhile, the precocious E. sinensis may be capable of negating the negative effects of HUFA deficiency by enhanced tissue 18:2n-6 accumulation and converting 18:2n-6 to 20:4n-6. [source]


Precocious sex change and spermatogenesis in the underyearling Malabar grouper Epinephelus malabaricus by androgen treatment

Abstract The Malabar grouper Epinephelus malabaricus is an important candidate species for commercial aquaculture in tropical and subtropical areas. In nature, this species requires more than 10 years to change sex from female to male and have active spermatogenic tissues in the testis. Thus, it is essential to find a means of producing sperm for seed production. This is the first report of artificial sex change in underyearling E. malabaricus. Female E. malabaricus with immature ovaries at 144 days post-hatch (DPH) were fed a diet with 17,-methyltestosterone (MT) at 50 ,g g,1 diet for 6 months. Sex change occurred in most of the treated fish, which had testis with all stages of spermatogenic germ cells including spermatozoa. In contrast, most of the control fish had immature ovaries. These results, which reveal that germ cells in the underyearling grouper have the ability to produce spermatozoa in response to exogenous androgen, demonstrate that sex change can be artificially induced during ovarian development. Reproductive performance and offspring quality of Chinese mitten crab Eriocheir sinensis (H. Milne-Edwards) females fed an optimized formulated diet and the razor clam Sinonovacula constricta

Abstract After feeding female Eriocheir sinensis on an optimized formulated diet or fresh razor clam Sinonovacula constricta for 7 months, their reproductive performance and offspring quality were compared. To evaluate diet nutrient contents, the proximate, fatty acid and amino acid compositions of the formulated diet and the razor clam were analysed. The nutritional value of the diets was determined by assessing survival, gonadosomatic index (GSI) and hepatosomatic index (HSI) of female crabs from both diet treatments, together with the percentage of females that spawned, total egg production per female and fecundity (number of eggs g,1 female wet weight). Furthermore, the quality of eggs and newly hatched larvae from the two dietary treatments were determined using the following parameters: egg diameter, wet weight and dry weight, hatchability, proximate and fatty acid profile of eggs, larval carapace length, resistant to starvation and osmotic shock, larval survival and development to the zoea II stage. Higher protein, phospholipids (PL) and amino acids (AA) contents were found in the razor clam while the formulated diet contains higher levels of ash, total lipid (TL) and 18:1n-9, 18:2n-6 and 22:6n-3 fatty acids. Although female crabs fed the two different diets showed similar reproductive performances, newly hatched zoea I larvae produced by the crabs fed the formulated diet had significantly longer mean carapace length and shorter development time to the zoea II stage under identical culture condition (P<0.05). Moreover, dietary fatty acid appeared to have more significant effects on the fatty acid composition of the hepatopancreas than it did on mature ovaries or eggs. This suggests that the fatty acid profile of mature ovaries is indicative of the specific fatty acid required for ovarian development in E. sinensis. In conclusion, our results show that the optimized formulated diet developed in this laboratory can totally replace the razor clam, a broodstock food widely used in E. sinensis hatcheries in China. This encouraging result should facilitate more reliable hatchery production of this important aquaculture species. [source]


Dynamic changes in gene expression during vitellogenic stages of the white shrimp: Fenneropenaeus merguiensis de Man

Abstract Ovarian maturation is a crucial step for shrimp brood stock. A suppressive subtractive hybridization was used to identify differentially expressed genes in the ovaries during vitellogenesis of Fenneropenaeus merguiensis. Three- to sevenfold up-regulated genes were selected. A blast search identified nine unique genes. The genes that may be involved in ovarian maturation, namely translationally controlled tumour protein (TCTP), heat shock protein 70 (HSP70), H-L(3)MBT-LIKE, shrimp ovarian peritrophin (SOP), vitellin (Vn), thrombospondin (TSP) and ribosomal protein L10a (RPL10a), were further studied. The transcripts of HSP70, TCTP, SOP and RPL10a in the ovary showed their highest expression in the early stage and declined in the later stages. In contrast, the transcripts of the H-L(3)MBT-LIKE, TSP and Vn genes increased from the early stage to be significantly up-regulated during the late stage. A comparison of gene expression among organs during the vitellogenesis showed that the transcripts of HSP70, SOP, H-L(3)MBT-LIKE and TSP were down-regulated in the brain, intestine, hepatopancreas and lymphoid (except for TSP) when compared with their expression in shrimp with non-developed ovaries. The mRNA of TCTP and RPL10a was significantly over-expressed in the lymphoid and heart, whereas TCTP transcripts were significantly down-regulated in the brain during the vitellogenesis. The molecular behaviour of the transcripts in this study may, in the future, lead to an ability to stimulate the ovarian development in shrimp. [source]


Role of allatostatin-like factors from the brain of Tenebrio molitor females,

Abstract The effect of brain extract from females of freshly emerged Tenebrio molitor on ovary, oocyte development, total protein content of hemolymph, and ovary was studied in 4-day-old adult mealworm females. Injections of extracts of 2-brain equivalents into intact (unligatured) Tenebrio females did not affect ovarian and oocyte development. Injections of ligated females, however, with 2-brain equivalents on day 1 and 2 after adult emergence strongly inhibited ovarian growth and oocyte development. At day 4, ligated and injected females did not develop their ovaries and pre-vitellogenic oocytes were not found. The changes in ovarian development correlated with an increase in the concentration of soluble proteins in the hemolymph as compared with the saline-injected controls. Additionally, a strong reduction of total protein content in ovarian tissue was observed. Reverse phase HPLC separation of a methanolic brain extract of T. molitor females showed that fraction 5 has a similar retention time to synthetic cockroach allatostatin. Fraction 5 was eluted at 12.88,min, which was closest to the internal standard Dippu-AST I, which eluted at 12.77,min. An ELISA of fraction 5 from the methanolic brain extract using antibodies against allatostatins Grybi-AST A1 and Grybi-AST B1 from cricket Gryllus bimaculatus showed that fraction 5 cross-reacted with Grybi-AST A1 antibodies. The cross-reactivity was similar to the synthetic allatostatin from D. punctata, which was used as a positive control. These observations demonstrate a possible role for allatostatin-like brain factor(s) in regulating the reproductive cycle of Tenebrio molitor. © 2009 Wiley Periodicals, Inc. [source]


Effects of heat shock on ovary development and hsp83 expression in Tribolium castaneum (Coleoptera: Tenebrionidae)

Abstract Heat shock affects reproductive performance in insects including Tribolium castaneum. In this study, the effects of heat shock on ovary development and hsp83 expression in T. castaneum were investigated. Two lines of T. castaneum, H line and C line, from the same base population were established and maintained for five successive generations. In each generation, the newly hatched beetles (within 3,h after eclosion) in the H line were treated with a heat shock at 40°C for 1,h, and those in the C line were raised at normal temperature (30°C) as control treatment. Four traits related to ovary development were measured for the beetles of the fifth generation: days from eclosion to laying the first eggs (To), days from eclosion to laying the first hatchable eggs (Th), ovariole size on the third day after eclosion, and pupal mass of their offspring. The results showed that the beetles of the H line had a significantly longer pre-oviposition period (0.6 more days) and smaller ovariole size than those of the C line. No significant difference in pupal mass was observed. Applying heat shock to the offspring of the fifth generation of both lines led to significantly higher hsp83 expression in offspring of the C line than in offspring of the H line. Within each line, the hsp83 expression level in offspring with heat shock was significantly higher than that of offspring without heat shock, but the difference in the C line was much larger than that in the H line. We infer from these results that a tradeoff between heat resistance, registered as hsp83 expression, and ovarian development operates under heat stress in T. castaneum. 2009 Wiley Periodicals, Inc. Gonadoinhibitory effects of Neb-colloostatin and Neb-TMOF on ovarian development in the mealworm, Tenebrio molitor L.

Abstract The gonadostatic action of the peptides Neb-colloostatin (SIVPLGLPVPIGPIVVGPR) and Neb-TMOF (NPTNLH) from Neobellieria bullata was studied in female mealworm Tenebrio molitor. Both peptides potently inhibit ovarian development and terminal oocyte maturation of mated females during their first reproductive cycle. Injection of 4 ,g of Neb-colloostatin or Neb-TMOFNeb-TMOF reduced, at day 4 of the cycle, the size of the terminal oocytes to about half or one third of the normal size in saline-injected controls. In addition, follicular patency was arrested. The injections of Neb-colloostatin and Neb-TMOF also caused a delay to the first ovulation and oviposition as well as a reduction of the number of eggs by about 50% in the first 3 days of the oviposition period. At 4 days after adult emergence, none of the peptides had caused significant changes in protein concentration or composition of the haemolymph. However, both peptides reduced total protein content in ovaries and induced qualitative changes in ovarian protein patterns. Electrophoretic analyses indicated that Neb-colloostatin and Neb-TMOF caused a loss of two proteins (150, 180 kDa) and a drastic reduction of 4 others (39, 43, 47, 130 kDa), which are the most abundant ones in ovaries of control females. On the other hand, they increased the concentration of 2 other polypeptides (65, 70 kDa), which normally occur in insignificant quantities in ovaries. Our results indicate that both peptides have a very similar mode of action despite the differences in their amino acid sequence. They seem to interfere with vitellogenin production by the fat body as well as with vitellogen uptake by the oocytes through modification of patency. Arch. Insect Biochem.
