Learning-dependent changes in sleep spindles and Stage 2 sleep

Summary It has become increasingly clear that sleep is necessary for efficient memory consolidation. Recently, it has been found that Stage 2 sleep disruption impairs procedural memory performance, and that memory performance is correlated with the duration of Stage 2 sleep; but the mechanisms involved in synaptic plasticity for procedural memory during sleep have not been identified. The present study examined the learning-dependent changes in sleep, including Stage 2 sleep spindles. Following an intense period of simple motor procedural learning, the duration of Stage 2 sleep and spindle density increased. There were no changes observed in the duration of any other stage of sleep or in the density of rapid eye movements. These findings support the hypothesis that sleep spindles are involved in the off-line reprocessing of simple motor procedural memory during Stage 2 sleep. [source]


OVERCOMING BARRIERS TO PAIN RELIEF IN THE CARIBBEAN

ABSTRACT This paper examines pain and pain relief in the Caribbean, where pain is widely perceived as an unavoidable part of life, and where unnecessary suffering results from untreated and under treated pain. Barriers to pain relief in the Caribbean include patient and family attitudes, inadequate knowledge among health professionals and unduly restrictive regulations on the medical use of opioids. Similar barriers exist all over the world. This paper urges medical, nursing and public health professionals, and educators to examine attitudes towards pain and pain relief and to work towards making effective pain relief and palliation more accessible. It recommends that i) health professionals and officials be better educated about pain, palliation and opioids, ii) regulatory restrictions be updated in light of clinical and scientific evidence, iii) opioid procurement policies be adjusted to facilitate increased medical use, iv) medical charts and records be modified to routinely elicit and document patients levels of pain, and v) educational campaigns be developed to inform the public that moderate and severe pain can be safely relieved at the end of life and other stages of life. The professional, respectful, and beneficent response to patients in pain is to provide rapid and aggressive pain relief or to urgently consult a pain or palliative specialist. When a health system hinders such efforts the ethical response is to identify, facilitate and advocate for overcoming barriers to improvement. [source]


Developmental changes in cell proliferation in the auditory midbrain of the bullfrog, Rana catesbeiana

Abstract We examined patterns of cell proliferation in the auditory midbrain (torus semicircularis) of the bullfrog, Rana catesbeiana, over larval and early postmetamorphic development, by visualizing incorporation of 5-bromo-2,-deoxyuridine (BrdU) in cycling cells. At all developmental stages, BrdU-labeled cells were concentrated around the optic ventricle. BrdU-labeled cells also appeared within the torus semicircularis itself, in a stage-specific manner. The mitotic index, quantified as the percent of BrdU-positive cells outside the ventricular zone per total cells available for label, varied over larval development. Mitotic index was low in hatchling, early larval, and late larval stages, and increased significantly in deaf period, metamorphic climax, and froglet stages. Cell proliferation was higher in metamorphic climax than at other stages, suggesting increased cell proliferation in preparation for the transition from an aquatic to an amphibious existence. The change in mitotic index over development did not parallel the change in the total numbers of cells available for label. BrdU incorporation was additionally quantified by dot-blot assay, showing that BrdU is available for label up to 72 h postinjection. The pattern of change in cell proliferation in the torus semicircularis differs from that in the auditory medulla (dorsal medullary nucleus and superior olivary nucleus), suggesting that cell proliferation in these distinct auditory nuclei is mediated by different underlying mechanisms. © 2006 Wiley Periodicals, Inc. Size-Related Advantages for Reproduction in a Slightly Dimorphic Raptor: Opposite Trends between the Sexes

Despite many comparative analyses and more than 20 proposed hypotheses, there is still little consensus over the factors promoting the evolution of reversed sexual dimorphism (RSD) in raptorial species. Furthermore, intrapopulation studies, which may elucidate how RSD is maintained once evolved, have been surprisingly scarce and only focused on a handful of species with medium to high dimorphism. We examined the reproductive advantages associated with body size and condition, measured in the pre-laying period, in a diurnal raptor with low sexual dimorphism, the black kite (Milvus migrans). The study population was essentially monomorphic in size. For females, there was an evidence of reproductive benefits associated with larger size and/or with better body condition. Larger females had also access to higher quality partners and territories, consistent with the ,intrasexual selection' hypothesis, by which members of the larger sex enjoy size-related advantages in intrasexual competition over a scarce resource, the smaller sex. Opposite trends emerged for males: smaller, leaner males had higher breeding output, consistent with the ,small efficient male' hypothesis. Overall, the fact that we observed in an essentially monomorphic population the same selection pressures previously found in species with marked dimorphism suggests that such reproductive advantages may be counterbalanced in our study model by opposite selection pressures during other stages of the life cycle. This casts some doubts on the evolutionary significance of studies focusing exclusively on reproduction and calls for the need of more comprehensive analyses incorporating trait-mediated differentials in survival and recruitment. Host,bacterial interaction in the development of gastric precancerous lesions in a high risk population for gastric cancer in Venezuela,

Abstract Helicobacter pylori (HP) infection affects over 50% of the world's population. The prevalence is over 90% in populations at high risk for gastric cancer, but clinical outcomes of the infection are highly variable and thus host genetic factors have been suggested to play a role in its outcomes in addition to bacterial factors. In this study, we examined the effects of common functional genetic polymorphisms of several proinflammatory cytokines known to be overexpressed in HP-infected gastric mucosa on the risk of various stages of gastric premalignant lesions. The odds ratios (ORs) and 95% confidence intervals (CI) for atrophic gastritis, intestinal metaplasia and dysplasia were estimated by multinominal logistic regression analysis among 2,033 Venezuelan subjects. There was a significant effect of IL8 -251A allele on the prevalence of dysplasia (p = 0.021). The OR associated with the A-allele was 1.34 (95% CI: 0.82,2.18) for heterozygotes and 2.00 (95% CI: 1.13,3.56) for homozygotes, compared with the TT genotype. Furthermore, there was a statistically significant interaction between the number of A-alleles and HP cag A genotype (p = 0.009), suggesting that the A-allele increased the risk of dysplasia only when cag A was present. The OR for the AA compared with TT genotype was 3.22 (95% CI: 1.60,6.52) in this group. There were no associations with other proinflammatory cytokines studied, i.e., IL1,, IL6, monocyte chemoattractant protein 1 (MCP1) and TNF,, or with other stages of premalignant lesions. Susceptibility of immature stages of Chrysoperla rufilabris (Neurop., Chrysopidae) to pyriproxyfen, a juvenile hormone analog

Effects of pyriproxyfen (Knack), a juvenile analog, at three concentrations (10, 50 and100 mg [AI]/l), on survival and development of all immature stages of Chrysoperla rufilabris (Burmeister), were determined in the laboratory. Pyriproxyfen significantly reduced the survival rates when eggs, first and third instars were treated, but not when the second instars and pupae were treated. When eggs were treated, the two higher concentrations reduced the eggs viability by 33.3,50%, and only 0.0,6.7% developed to adults. The lowest concentration of pyriproxyfen (10 mg [AI]/l) caused high mortality on third instars, not on other stages, indicating the third instar was the most vulnerable stage. Pyriproxyfen had significant effects on development for all immature C. rufilabris that successfully developed to adults with variations among the developmental stages and concentrations. The overall developmental duration from eggs to adults when eggs, first, and third instars were treated were 2.6,4.2, 2.4,4.1 respectively, and 6.0,7.1 d longer than those in water control, respectively. However, the overall developmental durations from eggs to adults were 0.5,1.2 d shorter than in water control when the second instars were treated with pyriproxyfen. The compatibility of pyriproxyfen with natural enemies in integrated pest management programs is discussed. [source]


Examination of the Validity of a Stages of Exercise Change Algorithm

The purpose of this study was to examine the validity of a stages of exercise change questionnaire with self-reported exercise, age, and body mass index (BMI) in 1,034 male and female community residents. It was found that the maintenance stage participants reported more strenuous exercise than did individuals in the other stages; and participants in the action stage reported more strenuous exercise than did individuals in the preparation and contemplation stages. There were no significant group differences among participants in the preparation, contemplation, and precontemplation stages for strenuous and moderate exercise. The maintenance stage participants engaged in more moderate exercise than did the preparation, contemplation, and precontemplation individuals; and the action stage participants reported more moderate exercise than did individuals in the preparation and precontemplation stages. The participants in the maintenance stage were older than were those in the action stage. No significant stage differences were found for mild exercise or BMI. Knowing an individual's stage of exercise change may provide researchers and practitioners with a mechanism for developing, testing, and delivering stage-matched exercise interventions. [source]


Seedling survival and seed size: a synthesis of the literature

Summary 1Large-seeded species have long been known to have higher survivorship during establishment than small-seeded species. Here, we assessed the size of this advantage by compiling published data on survival through seedling emergence, seedling establishment and sapling establishment. 2We found no relationship between seed mass and survival through the transition from viable seed in or on the soil to newly emerged seedlings (P = 0.47, n = 33 species). 3Synthesis of data from experimental studies on the advantages of large-seeded species establishing under particular hazards (such as shade, drought or herbivory) confirmed that seedlings of large-seeded species perform better than those of small-seeded species in most situations. However, the magnitude of this advantage was not sufficient to counterbalance the greater number of seeds produced by small-seeded species m,2 of canopy outline year,1. 4Synthesis of data from field studies of populations under natural conditions also showed that large-seeded species have higher survival through early seedling establishment than small-seeded species (P = 0.006, n = 112 species). However, the magnitude of this advantage would only be sufficient to counterbalance the greater number of seeds produced by small-seeded species m,2 of canopy outline year,1 if mortality continued at the same rate for some time. 5The time required for a species with 10-fold larger seeds to recoup the advantage gained by a smaller-seeded species during seed production ranged from 8.8 weeks for the smallest seeded species in the data set, up to an implausible 4.2 years for the largest-seeded species. Thus, while large-seeded species do have a survival advantage over small-seeded species during seedling establishment, the available evidence suggests that advantages must also accrue during other stages in the life cycle. One possibility is that the greater seed production of small-seeded species (m,2 of canopy outline year,1) is partly offset by larger canopies and longer reproductive life spans in large-seeded species. Shared decision making observed in clinical practice: visual displays of communication sequence and patterns

Abstract The aim of the study was to examine the communication strategies of general practitioners attempting to involve patients in treatment or management decisions. This empirical data was then compared with theoretical ,competences' derived for ,shared decision making'. The subjects were four general practitioners, who taped conducted consultations with the specific intent of involving patients in the decision-making process. The consultations were transcribed, coded into skill categorizations and presented as visual display using a specifically devised sequential banding method. The empirical data from these purposively selected consultation from clinicians who are experienced in shared decision making did not match suggested theoretical frameworks. The views of patients about treatment possibilities and their preferred role in decision making were not explored. The interactions were initiated by a problem-defining phase, statements of ,equipoise' consistently appeared and the portrayal of option information was often intermingled with opportunities to allow patients to question and reflect. A decision-making stage occurred consistently after approximately 80% of the total consultation duration and arrangements were consistently made for follow-up and review. Eight of the 10 consultations took more than 11 min , these specific consultations were characterized by significant proportions of time provided for information exchange and patient interaction. The results demonstrate that some theoretical competences are not distinguishable in practice and other stages, not previously described, such as the ,portrayal of equipoise', are observed. The suggested ideal of a shared decision-making interaction will either require more time than currently allocated, or alternative strategies to enable information exchange outside the consultation. Constitutive Phosphorylation of the Vesicular Inhibitory Amino Acid Transporter in Rat Central Nervous System

Abstract:,-Aminobutyric acid (GABA) and glycine are stored into synaptic vesicles by a recently identified vesicular inhibitory amino acid transporter [VIAAT, also called vesicular GABA transporter (VGAT)]. Immunoblotting analysis revealed that rat brain VIAAT migrated as a doublet during sodium dodecyl sulfate,polyacrylamide gel electrophoresis, with a predominant slower band in all areas examined except olfactory bulb and retina. The slower band corresponded to a phosphorylated form of VIAAT as it was converted to the faster one by treating brain homogenates with alkaline phosphatase or with an endogenous phosphatase identified as type 2A protein,serine/threonine phosphatase using okadaic acid. In contrast, the recombinant protein expressed in COS-7 or PC12 cells co-migrated with the faster band of the brain doublet and was insensitive to alkaline phosphatase. To investigate the influence of VIAAT phosphorylation on vesicular neurotransmitter loading, purified synaptic vesicles were treated with alkaline phosphatase and assayed for amino acid uptake. However, neither GABA nor glycine uptake was affected by VIAAT phosphorylation. These results indicate that VIAAT is constitutively phosphorylated on cytosolic serine or threonine residues in most, but not all, regions of the rat brain. This phosphorylation does not regulate the vesicular loading of GABA or glycine, suggesting that it is involved at other stages of the synaptic vesicle life cycle. [source]


Mixed-Gender Groups, Dating, and Romantic Relationships in Early Adolescence

This study examined dating-stage and developmental-contextual models of romantic relationships during early adolescence. Same-gender friendships, affiliation with mixed-gender groups, dating, and romantic relationships were investigated in a sample of 1,284 young adolescents of diverse ethnocultural backgrounds. Data were collected cross-sectionally in Grades 5 through 8, as well as longitudinally in the fall and spring of an academic year. Consistent with a stage model, affiliation with mixed-gender groups and dating were qualitatively distinct activities that were sequentially organized and facilitated the progression from same-gender friendships to dyadic romantic relationships. The results also provide insights on how the developmental context may alter stage pathways: Dating activities were incorporated with mixed-gender affiliations, group-based romantic stages showed more stability than other stages, and the ethnocultural context influenced romantic timing. Finally, results indicated that participation in romantic activities heightened adolescents' future interest in having a romantic relationship. [source]


Molecular control of mitochondrial function in preimplantation mouse embryos

Abstract Mitochondria play a key role in a number of physiological events during all stages of life, including the very first stages following fertilization. It is, therefore, important to understand the mechanisms controlling mitochondrial activity during early embryogenesis to determine their role in development outcome. The objective of this study was to investigate the molecular control of mitochondrial transcription and mitochondrial DNA (mtDNA) replication in mouse preimplantation embryos. We estimated the mtDNA copy number and characterized the expression patterns of two mitochondrial genes and several nuclear genes that encode mitochondrial transcription and replication factors throughout preimplantation development. Mitochondrial gene transcripts were present in larger quantities in morula and blastocyst stage embryos relative to other stages. A significant increase in the amount of mRNA for nuclear genes encoding mtDNA transcription factors was observed in eight-cell stage embryos. Although a similar increase in the mRNA levels of nuclear genes encoding mtDNA replication factors was observed in morula and blastocyst stage embryos, the number of mtDNA molecules remained stable during preimplantation stages, suggesting that nuclear-encoded mitochondrial transcription factors are involved in the regulation of mtDNA transcription during early development. Although transcripts of replication factors are abundant at the morula and blastocyst stage, mtDNA replication did not occur until the blastocyst stage, suggesting that the inhibition of mtDNA replication is controlled at the post-transcriptional level during early embryogenesis. Spontaneous platelet aggregation in Kawasaki disease using the particle counting method

AbstractBackground:,Platelet aggregation is generally measured by the optical density method. This method is not very sensitive in detecting platelet activation because of the poor correlation between the formation of platelet aggregates and light transmission, and inability to detect small platelet aggregates. Recently, a new method was developed that detects small platelet aggregates formed in the early phase of platelet aggregation by means of a particle counting technique using light scattering. Methods:,The present study used the particle counting method to measure spontaneous platelet aggregation (SPA) produced by agitation with a stirrer bar without agonists in patients with Kawasaki disease (KD) and children's thrombotic disease, and compared the results with the conventional optical density method. Results:,Using the particle counting method, significantly higher SPA rates were found before treatment than in healthy adults and at the other stages of treatment. A similar tendency was noted with the optical density method; however, the positive rate of SPA was significantly higher with the particle counting method in each stage except the second stage, which was the period from the start of gamma-globulin therapy to 20 days. Conclusion:,These results indicate that enhanced platelet aggregation is noted at a high frequency before treatment of KD and that the particle counting method is superior to the optical density method in detecting enhanced platelet aggregation. Proteomic comparison of four Eimeria tenella life-cycle stages: Unsporulated oocyst, sporulated oocyst, sporozoite and second-generation merozoite

Abstract We report the proteomes of four life-cycle stages of the Apicomplexan parasite Eimeria tenella. A total of 1868 proteins were identified, with 630, 699, 845 and 1532 found in early oocysts (unsporulated), late oocysts (sporulated), sporozoites and second-generation merozoites, respectively. A multidimensional protein identification technology shotgun approach identified 812 sporozoites, 1528 merozoites and all of the oocyst proteins, whereas 2-D gel proteomics identified 230 sporozoites and 98 merozoite proteins. Comparing the invasive stages, we find moving junction components RON2 in both, whereas AMA-1 and RON4 are found only in merozoites and AMA-2 and RON5 are only found in sporozoites, suggesting stage-specific moving junction proteins. During early oocyst to sporozoite development, refractile body and most "glideosome" proteins are found throughout, whereas microneme and most rhoptry proteins are only found after sporulation. Quantitative analysis indicates glycolysis and gluconeogenesis are the most abundant metabolic groups detected in all stages. The mannitol cycle "off shoot" of glycolysis was not detected in merozoites but was well represented in the other stages. However, in merozoites we find more protein associated with oxidative phosphorylation, suggesting a metabolic shift mobilising greater energy production. We find a greater abundance of protein linked to transcription, protein synthesis and cell cycle in merozoites than in sporozoites, which may be residual protein from the preceding massive replication during schizogony. Immunohistochemical Studies on Oestrogen Receptor Alpha (ER,) and the Proliferative Marker Ki-67 in the Sow Uterus at Different Stages of the Oestrous Cycle

Contents In order to better understand physiological changes during the different stages of the oestrous cycle, immunohistochemistry was used in the present study to investigate the distribution of oestrogen receptor alpha (ER,) as well as the proliferative marker Ki-67, in the sow uterus during the oestrous cycle. Uterine samples were collected from multiparous sows with normal reproductive performance at selected stages of the oestrous cycle: at late dioestrus (d 17), prooestrus (d 19), oestrous (d 1), early dioestrus (d 4) and dioestrus (d 11,12), respectively. The tissue samples were fixed in 10% formaldehyde, embedded in paraffin and subjected to immunohistochemistry using monoclonal antibodies against ER, (C-311) and Ki-67 (MM-1). In general, the immunostaining of both ER, and Ki-67 was confined to nuclei of the target cells. Variations were seen, not only at the different stages of the oestrous cycle, but also in the different tissue compartments of the uterus. In the epithelia, the strongest ER, staining and highest amount of positive Ki-67 cells were found at early dioestrus. In the myometrium, the highest levels of staining of both ER, and Ki-67 positive cells were found at pro-oestrus and oestrus. For the proliferative marker, Ki-67, no positive cells were found at dioestrus and late dioestrus in the epithelium and myometrium. In the connective tissue stroma (subepithelial layer), the highest number of ER, positive cells were found at oestrus, which was significantly different compared with other stages (p,0.05), whereas the levels of Ki-67 positive cells were relatively low and did not differ between the stages examined. Significant correlations between the number of ER, positive cells in the stroma and Ki-67 positive cells in the epithelia were observed. This suggests indirect regulatory mechanisms on epithelial proliferation via ER, in the stroma. In conclusion, these findings in the sow uterus show that the presence of ER, as well as Ki-67 protein varies not only between different stages of the oestrous cycle but also between different tissue compartments of the uterus. These findings indicate various regulatory mechanisms and stress the importance of localising ER, and proliferating cells in different uterine tissues. [source]


ORIGINAL ARTICLE: Lipopolysaccharide Alters the Vaginal Electrical Resistance in Cycling and Pregnant Mice

Problem, Lipopolysaccharide (LPS) has been postulated to exert harmful biologic effects during pregnancy. The objective of present investigation is to measure the vaginal electrical resistance (VER) in LPS-treated normal cycling and pregnant female mice. Method of study, Minimum dose (MD) of LPS (250 ,g/kg body weight) was injected in pregnant female mice through i.p. route on day 0.5 of pregnancy. VER was measured during different phases of reproductive cycle in female mice, which were pre-exposed to LPS and in untreated cycling female mice. VER was also measured in control pregnant female mice (saline-treated mice) through whole pregnancy and LPS-treated female mice in early stages of pregnancy. Results, Vaginal electrical resistance was significantly higher during proestrous or early estrous stage as compared with any other stages of reproductive cycle in mouse. One peak of VER was observed during peri-implantation period of pregnancy in control female mice. The significant differences in the pattern of VER were found between LPS-treated and control female mice during peri-implantation period of pregnancy, and between cycling female mice, which were pre-exposed to LPS and untreated cycling female mice during proestrus. Conclusion, The presented results demonstrate, for the first time, that LPS exposure during pregnancy may be determined by measuring VER in mothers without any adverse effect on ongoing pregnancy and may help in refining the assisted reproduction techniques. Rapid sequence spinal anaesthesia for category-1 urgency caesarean section: a case series

Summary General anaesthesia is the fastest method for anaesthetising a category-1 caesarean section but is associated with increased maternal morbidity and mortality. We describe the ,rapid sequence spinal' to minimise anaesthetic time. This consists of a no-touch spinal technique, consideration of omission of the spinal opioid, limiting spinal attempts, allowing the start of surgery before full establishment of the spinal block, and being prepared for conversion to general anaesthesia if there are delays or problems. We present a case series of 25 rapid sequence spinal anaesthetics for category-1 caesarean section. The mean (SD [range]) decision-delivery interval was 23 (6 [14,41]) min. After excluding cases where there was an identified delay, the median (IQR [range]) time to prepare and perform the spinal was 2 (2,3 [1,7]) min, and time to develop a ,satisfactory' block was 4 (3,5 [2,7]) min. The total time to induce spinal anaesthesia was 8 (7,8 [6,8]) min. There were three pre-operative conversions to general anaesthesia and three women had pain during surgery that did not require treatment. Our data indicate that one might expect to establish anaesthesia in 6,8 min using a rapid sequence spinal. Careful case selection is crucial. While rapid anaesthesia is important, the reduction of the decision-delivery interval also requires attention to other stages in the pre-operative process. [source]

Abstract The relative toxicity (LC50 values based on µg oil/cm2) is evaluated of aqueous nC24 agricultural mineral oil (AMO) emulsions to the egg, six-legged nymph (larva), eight-legged protonymph and adult stages of two-spotted mite (Tetranychus urticae) and its predator, Phytoseiulus persimilis, on French bean leaf discs, using a Potter spray tower to apply of the oil. The egg of P. persimilis was the least susceptible stage (LC50 444.84) and its LC50 was significantly higher than all other stages tested of either P. persimilis or T. urticae. The LC50 for adult female T. urticae (LC50 63.89) was significantly lower than the larva (LC50 93.86); however, there was no significant difference in response between the protonymph (LC50 70.44) and the larva, which were both higher than T. urticae eggs (LC50 17.55). LC50s for P. persimilis larva (LC50 43.87), protonymph (LC50 41.55) and adult female (LC50 53.34) were similar. Scanning electron microscopy showed that the egg surface of T. urticae is usually well covered with fine silk that may trap more oil and increase AMO efficacy. Other possible differences in AMO efficacy between T. urticae and P. persimilis may be due to differences in egg size, egg incubation period, egg surface structure and the presence of vulnerable respiratory cones in T. urticae eggs. Dose of 0.2,0.3% (w/w) is considered to be the most appropriate for nC24 AMOs use against T. urticae in combination with P. persimilis in integrated pest management programs. [source]
