Selected AbstractsThe "Modified" Winch StitchDERMATOLOGIC SURGERY, Issue 10 2001J. Michael Casparian MD Background. The methods described to achieve intraoperative tissue expansion have drawbacks. Objective. To report an intraoperative tissue expansion technique using standard suture material and hemostats to create a "modified" winch stitch. Methods. A continuous running suture is placed with both ends left free. A "fixed" hemostat is placed above the skin at one end. After pulling at the other end with a second hemostat, a third hemostat is placed proximal to this hemostat just above the skin, which is left in place to produce stretching. The second and third hemostats are then used in an alternating pulling and holding fashion. Results. This method produces significant tissue expansion. If the desired degree of approximation is achieved, the ends of the suture can be tied together, as in the previously described winch stitch. Conclusion. The modified winch stitch is a simple and inexpensive means to facilitate closure of wounds under a significant amount of tension. Radin Abstract Compounds that can produce potent biological effects in cells encompass a variety of structural motifs. Many of these compounds share a structural feature that has rarely been noted. It is an allylic cluster of atoms, a 3-carbon chain with a double bond between two of the atoms and an oxygen atom at the other end. The oxygen can be in a hydroxyl group, or in an ether or ketal or ester linkage, or simply a carbonyl form. In the latter case, the linkage is an allylic ketone (ene-one) structure. Nitrogen is often seen in equivalent forms. Inclusion of at least one allylic moiety appears to be able to turn a modestly active or inert compound into an effective drug or toxin. Some compounds lack the allylic moiety but develop one by enzymatic action, usually via cytochrome P-450 enzymes. These metabolites probably represent the active drug forms. The above concepts seem to be radically simplistic and improbable, but the evidence supporting them and the explanations for the biological activities are hidden "in plain view." Comparisons with the pleiotropic activities of the allylic sphingolipid, ceramide, indicate that many allylic drugs operate by controlling the state of protein phosphorylation, by activating proteases, by generating reactive oxygen species, by slowing mitochondrial electron transport, or by lowering cellular glutathione concentrations. [source] Does amplitude scaling of ground motion records result in biased nonlinear structural drift responses?EARTHQUAKE ENGINEERING AND STRUCTURAL DYNAMICS, Issue 13 2007Nicolas Luco Abstract Limitations of the existing earthquake ground motion database lead to scaling of records to obtain seismograms consistent with a ground motion target for structural design and evaluation. In the engineering seismology community, acceptable limits for ,legitimate' scaling vary from one (no scaling allowed) to 10 or more. The concerns expressed by detractors of scaling are mostly based on the knowledge of, for example, differences in ground motion characteristics for different earthquake magnitude,distance (Mw,Rclose) scenarios, and much less on their effects on structures. At the other end of the spectrum, proponents have demonstrated that scaling is not only legitimate but also useful for assessing structural response statistics for Mw,Rclose scenarios. Their studies, however, have not investigated more recent purposes of scaling and have not always drawn conclusions for a wide spectrum of structural vibration periods and strengths. This article investigates whether scaling of records randomly selected from an Mw,Rclose bin (or range) to a target fundamental-mode spectral acceleration (Sa) level introduces bias in the expected nonlinear structural drift response of both single-degree-of-freedom oscillators and one multi-degree-of-freedom building. The bias is quantified relative to unscaled records from the target Mw,Rclose bin that are ,naturally' at the target Sa level. We consider scaling of records from the target Mw,Rclose bin and from other Mw,Rclose bins. The results demonstrate that scaling can indeed introduce a bias that, for the most part, can be explained by differences between the elastic response spectra of the scaled versus unscaled records. Casey Abstract Excitement with the publication of the human genome has served as catalyst for scientists to uncover the functions of specific genes. The main avenues for understanding gene function have been in behavioral genetics on one end and on the other end, molecular mouse models. Attempts to bridge these approaches have used brain imaging to conveniently link anatomical abnormalities seen in knockout/transgenic mouse models and abnormal patterns of brain activity seen in humans. Although a convenient approach, this article provides examples of challenges for imaging genetics, its application to developmental questions, and promises for future directions. Attempts to link genes, brain, and behavior using behavioral genetics, imaging genetics, and mouse models of behavior are described. Each of these approaches alone, provide limited information on gene function in complex human behavior, but together, they are forming bridges between animal models and human psychiatric disorders. Han Kim In the last months of 1997, the value of the Korean currency lost over half its value against the dollar, and the ruling party was swept from power in presidential elections. One of the fundamental causes of this national economic crisis was the widespread failure of Korean companies to earn their cost of capital, which contributed to massive shareholder losses and calls for corporate governance reform. Among the worst performers, and hence the main targets of governance reform, were family-controlled Korean business groups known as chaebol. Besides pursuing growth and size at the expense of value, such groups were notorious for expropriating minority shareholders through "tunneling" activities and other means. The reform measures introduced by the new administration were a mix of market-based solutions and government intervention. The government-engineered, large-scale swaps of business units among the largest chaebol,the so-called "big deals" that were designed to force each of the groups to identify and specialize in a core business,turned out to be failures, with serious unwanted side effects. At the same time, however, new laws and regulations designed to increase corporate transparency, oversight, and accountability have had clearly positive effects on Korean governance. Thanks to reductions in barriers to foreign ownership of Korean companies, such ownership had risen to about 37% at the end of 2006, up from just 13% ten years earlier. And in addition to the growing pressure for better governance from foreign investors, several newly formed Korean NGOs have pushed for increased transparency and accountability, particularly among the largest chaebol. The best governance practices in Korea today can be seen mainly in three kinds of corporations: (1) newly privatized companies; (2) large corporations run by professional management; and (3) banks with substantial equity ownership in the hands of foreign investors. The improvements in governance achieved by such companies,notably, fuller disclosure, better alignment of managerial incentives with shareholder value, and more effective oversight by boards,have enabled many of them to meet the global standard. And the governance policies and procedures of POSCO, the first Korean company to list on the New York Stock Exchange,as well as the recent recipient of a large equity investment by Warren Buffett,are held up as a model of best practice. At the other end of the Korean governance spectrum, however, there continue to be many large chaebol-affiliated or family-run companies that have resisted such reforms. And aided by the popular resistance to globalization, the lobbying efforts of such firms have succeeded not only in reducing the momentum of the Korean governance reform movement, but in reversing some of the previous gains. Most disturbing is the current push to allow American style anti-takeover devices, which, if successful, would weaken the disciplinary effect of the market for corporate control. [source] Corporate Cash Policy and How to Manage it with Stock RepurchasesJOURNAL OF APPLIED CORPORATE FINANCE, Issue 3 2008Amy Dittmar At the end of 2004 total U.S. corporate cash holdings reached an all-time high of just under $2 trillion,an amount equal to roughly 15% of the total U.S. GDP. And during the past 25 years, average cash holdings have jumped from 10% to 23% of total corporate assets. But at the same time their levels of cash have risen, U.S. companies have paid out dramatically increasing amounts of cash to buy back shares. This article addresses the following questions: What accounts for the dramatic increase in the average level of corporate cash holdings since 1980? And why do some companies keep so much cash (with one fourth of U.S. firms holding cash amounting to at least 36% of total assets) while others have so little (with another quarter having less than 3%)? Why do companies pay out excess cash in the form of stock repurchases (rather than, say, dividends), and what explains the significant increase in repurchases (both in absolute terms and relative to dividends) over time? The author begins by arguing that cash reserves provide companies with a buffer against possible shortfalls in operating profits,one that, especially during periods of financial trouble, can be used to avoid financial distress or provide funding for promising projects that might otherwise have to be put off. Such buffers are particularly valuable in the case of smaller, riskier companies with lots of growth opportunities and limited access to capital markets. And the dramatic increase in corporate cash holdings between 1980 and the present can be attributed mainly to an increase in the risk of publicly traded companies,an increase in risk that reflects in part a general increase in competition, but also a notable change over time in the kinds of companies (smaller, newer, less profitable, non-dividend paying firms) that have chosen to go public. At the other end of the corporate spectrum are large, relatively mature companies with limited growth opportunities. Although such companies tend to produce considerable free cash flow, they also tend to retain relatively small amounts of cash (as a percentage of total assets), in part because of shareholder concern about the corporate "free cash flow problem",the well-documented tendency of such companies to destroy value through overpriced (often diversifying) acquisitions and other misguided attempts to pursue growth at the expense of profitability. For companies with highly predictable earnings and investment plans, dividends provide one means of addressing the free cash flow problem. But for companies with more variable earnings and less predictable reinvestment, open-market stock repurchases provide a more flexible means of distributing cash to shareholders. Unlike the corporate "commitment" implied by dividend payments, an open market stock repurchase program creates what amounts to an option but not an obligation to distribute funds. The value of such flexibility, which increases during periods of increased risk and uncertainty, explains much of the apparent substitution of repurchases for dividends in recent years. [source] Studies on codon usage in Thermoplasma acidophilum and its possible implications on the occurrences of lateral gene transferJOURNAL OF BASIC MICROBIOLOGY, Issue 5 2005S. K. Gupta Codon usage studies have been carried out on the coding sequences of Thermoplasma acidophilum, which is an archaeon and grows at very low pH and high temperature. Overall codon usage data analysis indicates that all the four bases are almost equifrequent at the third position of codons, which is expected (since genomic GC % of this genome is about 46%). However, multivariate statistical analysis indicates that there are two major trends in the codon usage variation among the genes in this organism. In the first major trend it is observed that genes having G and C ending codons are clustered at one end while, A and T ending ones are clustered at the other end. We have also found a significant positive correlation between the expressivities of genes and GC contents at the synonymous third codon positions. In the second major trend, it is seen that the genes are clustered into three distinct parts. A comparative analyses of codon usage data of T. acidophilum and Sulfolobus solfataricus reveals that one of the three clusters of genes of T. acidophilum is very similar to a considerable number of S. solfataricus genes, suggesting possible occurrences of lateral gene transfer between these two microorganisms as reported by earlier workers. The aim of this paper is to explore some of the drivers for change and their impact and recommend a way forward to optimising nurses' work in the hospital environment. Background., The healthcare workplace has been transformed over the past 20 years in response to economic and service pressures. However, some of these reforms have had undesirable consequences for nurses' work in hospitals and the use of their time and skills. Results., As the pace and complexity of hospital care increases, nursing work is expanding at both ends of the complexity continuum. Nurses often undertake tasks which less qualified staff could do while at the other end of the continuum, are unable to use their high level skills and expertise. This inefficiency in the use of nursing time may also impact negatively on patient outcomes. Conclusions., Nurses' work that does not directly contribute to patient care, engage higher order cognitive skills or provide opportunity for role expansion may decrease retention of well-qualified and highly skilled nurses in the health workforce. Relevance to clinical practice., In this climate of nursing shortages, we need to use nurses in a cost-effective but also, intellectually satisfying manner, to achieve a sustainable nursing workforce. [source] A capillary holder for scanning detection of capillary isoelectric focusing with laser-induced fluorescenceJOURNAL OF SEPARATION SCIENCE, JSS, Issue 3 2009Katsuyoshi Takahashi Abstract A holder for a 12 cm long capillary was designed for scanning LIF detection of CIEF. The polyimide coat of a fused-silica capillary has been removed, and 1.5 mm diameter flanges have been attached near both ends. The holder is fixed on the stage of a fluorescence microscope via a translational stage, and a capillary guide is directly fixed on the microscope stage. The guide has a groove and a pressure plate for the capillary to slide in. The holder has two pulling plates with slits of 1 mm to accept the capillary just inside the flanges. The slits and the groove of the guide have been aligned. The motion of the translational stage brings the pulling plate into contact with the flange at the pulled side, and slides the capillary through the guide. The other end of the capillary is free and produces no strain on the capillary. When the motion of the stage is reversed, an unstrained contact is achieved at the other end. The baseline noise from scanning was only 50% larger than that without scanning. The fluorescence-signal variation during scanning was about 4% of the total signal, which was about twice that without scanning. Klessig Abstract Lake management is typically approached from a biophysical perspective. Lake managers ask how lakes can be managed to sustain their ecological functions. The social value of lakes is usually given less attention. The present paper begins the analysis at the other end of the lake and society connection by posing the question: what social needs must be met to sustain society? The primary social needs of sustainable societies are outlined and then the contribution of lakes to each need is discussed. Lakes can only provide optimal social benefits if management decisions recognize the full set of potential contributions lakes can make to society and those management decisions are integrated to provided balanced attention to all values that lakes provide. The present paper expands the domain of values beyond the traditional environmental and recreational contributions of lake management to also include aesthetics, education, economic opportunity, emotional security, cultural opportunity, individual freedom and spirituality. Citizen involvement is essential in broadening the conceptualization of the lake values and in implementing integrated management plans. Shubov Abstract We consider a class of non-selfadjoint operators generated by the equation and the boundary conditions, which govern small vibrations of an ideal filament with non-conservative boundary conditions at one end and a heavy load at the other end. The filament has a non-constant density and is subject to a viscous damping with a non-constant damping coefficient. The boundary conditions contain two arbitrary complex parameters. In our previous paper (Mathematical Methods in the Applied Sciences 2001; 24(15) : 1139,1169), we have derived the asymptotic approximations for the eigenvalues and eigenfunctions of the aforementioned non-selfadjoint operators when the boundary parameters were arbitrary complex numbers except for one specific value of one of the parameters. We call this value the critical value of the boundary parameter. It has been shown (in Mathematical Methods in the Applied Sciences 2001; 24(15) : 1139,1169) that the entire set of the eigenvalues is located in a strip parallel to the real axis. The latter property is crucial for the proof of the fact that the set of the root vectors of the operator forms a Riesz basis in the state space of the system. In the present paper, we derive the asymptotics of the spectrum exactly in the case of the critical value of the boundary parameter. We show that in this case, the asymptotics of the eigenvalues is totally different, i.e. both the imaginary and real parts of eigenvalues tend to ,as the number of an eigenvalue increases. We will show in our next paper, that as an indirect consequence of such a behaviour of the eigenvalues, the set of the root vectors of the corresponding operator is not uniformly minimal (let alone the Riesz basis property). In the forthcoming papers, based on the asymptotical results of the present paper, we will prove the Riesz basis property of the eigenfunctions. The spectral results obtained in the aforementioned papers will allow us to solve boundary and/or distributed controllability problems for the filament using the spectral decomposition method. Dawes Author's Introduction The article was provoked by recent discussion of the so-called ,conflict thesis': the idea that the Christian faith and the findings of modern science are necessarily at odds. This thesis is generally attributed to John William Draper (1811,1882) and Andrew Dickson White (1832,1918). Recent opposition to their work dates from a 1979 publication by James Moore. Moore argues that the warfare metaphor employed by Draper and White misrepresents the historical reality, by suggesting that the religion and science debates were clashes between distinct groups of people who were sharply polarized and violently antagonistic. Since then, similar criticisms have been made by historians, such as David Livingstone, Ronald Numbers, and David Lindberg. A key question here is: what does the conflict thesis entail? If it holds that Christian thinkers have invariably opposed scientific progress, while the defenders of science have been non-believers, it would be demonstrably false. But there exist more interesting forms of conflict thesis, which are philosophical rather than historical. These suggest that there is some tension between what Christians have traditionally believed and the findings of modern science, particularly Darwin's theory of evolution by natural selection. Even if the two are not, strictly speaking, incompatible, the truth of one may constitute evidence against the truth of the other. Darwin's theory also undercuts traditional arguments from design, and highlights the epistemological divide between religious and scientific conceptions of authority. The ASA ,does not take a position when there is honest disagreement between Christians', so it embraces a variety of perspectives. Does that mean there is no conflict between Darwinism and Christianity? 2Taken at face value, Genesis 1,3 tells the story of the origins of the world and of human beings. What aspects of that story would you consider essential to the Christian faith? 3If we have an entirely natural explanation of the origins of complex living organisms, do we still have reasons to believe in a creator God? 4If God could have created complex living beings by a simple command, why would he choose a lengthy and wasteful process such as natural selection? 5Could a Christian regard the existence of God in the same way as a scientific hypothesis, that is to say, to be accepted only in so far as it is supported by the evidence? [source] Interplay between dipolar, stacking and hydrogen-bond interactions in the crystal structures of unsymmetrically substituted esters, amides and nitriles of (R,R)- O,O,-dibenzoyltartaric acidACTA CRYSTALLOGRAPHICA SECTION B, Issue 3 2001Urszula Rychlewska The compounds analysed are the O,O,-dibenzoyl derivatives of (R,R)-tartaric acid, asymmetrically substituted by ester, amide and nitrile groups. Benzoylation does not introduce drastic changes to the molecular conformation. All investigated molecules adopt the planar T conformation of the four-carbon chain with noticeably smaller departures from the ideal conformation than observed in the nonbenzoylated analogs. Primary and secondary amides always orient the C=O bond antiperiplanar (a) with respect to the nearest C*,O bond, while methylester groups adjust their conformation to that adopted by the amide substituent situated at the other end of the molecule. Tertiary amides and carboxyl groups place their carbonyls at the same side as the nearest C*,O bond (the s form), but often deviations from coplanarity of the two bonds are significant and higher than those observed in the nonbenzoylated series. The results presented demonstrate the importance of dipole/dipole interactions between CO and ,C*H groups in stabilizing the molecular conformation, and between carbonyl groups in stabilizing crystal packing of the molecules that lack classical hydrogen-bond donor groups. An illustration is provided as to how a small change in mutual orientation of molecules arranged in a close-packed fashion causes a change in the character of intermolecular interactions from van der Waals to sandwich stacking between the benzoyloxy phenyls, and to dipolar between the benzoyloxy carbonyls. Hydrogen-bonded molecules tend to orient in a head-to-tail mode; the head-to-head arrangement being limited to cases in which terminal carbonyl groups are situated at one side of the molecule. The orientation of the benzoyloxy substituents with respect to the carbon main chain is such that the (O=)C,O,C,H bond system often deviates significantly from planarity. The 16th Congress of the International Union of Anthropological and Ethnological Sciences was held in China in summer 2009 in the southwestern state of Yunnan, where this diversity is most pronounced. Participants were encouraged to visit model communities, enjoy music and dance performances by the minorities, and admire their colourful costumes, as in this picture, which shows how VIP guests were welcomed to the closing ceremony of the Congress. Yet just a few weeks earlier, at the other end of the country in Xinjiang, almost 200 people were killed in the worst ethnic rioting since Liberation 60 years ago. While most economic and educational indicators suggest that socialist ethnic policies have been an impressive success, evidently not all is harmonious. Especially among Tibetans and Uyghurs, policies of recognition and the emphasis on a territorial homeland have fostered aspirations which cannot be satisfied by stepping up support for folklore and building ever larger ethnic museums and theme parks. In these regions, the evidence which power holders present as proof of progress and integration, and even as the accomplishment of an ancient civilizing mission, is interpreted locally in terms of assimilation. These tensions constrain the possibilities for anthropological research, for Chinese and foreign scholars alike. In his contribution to this issue, Chris Hann juxtaposes his impressions of the Congress in Kunming with his experiences of rural fieldwork among the Uyghurs of Xinjiang. [source] Effect of line shooter and mainline tension on the sink rates of pelagic longlines and implications for seabird interactionsAQUATIC CONSERVATION: MARINE AND FRESHWATER ECOSYSTEMS, Issue 4 2010Graham Robertson Abstract 1. The likelihood that seabirds will be hooked and drowned in longline fisheries increases when baited hooks sink slowly. Fishermen target different fishing depths by setting the mainline through a line shooter, which controls the tension (or slackness) in the line. An experiment was conducted in Australia's pelagic longline fishery to test the hypothesis of no difference in sink rates of baited hooks attached to mainline set under varying degrees of tension. 2. Mainline was set in three configurations typically used in the fishery: (a) surface set tight with no slackness astern; (b) surface set loose with 2,s of slack astern; and (c) deep set loose with 7,s of slack astern. 3. Tension on the mainline had a powerful effect on sink rates. Baited hooks on branch lines attached to tight mainlines reached 2,m depth nearly twice as fast as those on the two loose mainline tensions, averaging 5.8,s (0.35,m,s,1) compared with 9.9,s (0.20,m,s,1) and 11.0,s (0.18,m,s,1) for surface set loose and deep set loose tensions, respectively. 4. The likely reason for the difference is propeller turbulence. Tight mainline entered the water aft of the area affected by turbulence whereas the two loose mainlines and the clip ends of branch lines were set directly into it about 1,m astern of the vessel. The turbulence presumably slowed the sink rates of baited hooks at the other end of the branch lines. 5. The results suggest that mainline deployed with a line shooter (as in deep setting) into propeller turbulence at the vessel stern slows the sink rates of baited hooks, potentially increasing their availability to seabirds. Unless mainline can be set to avoid propeller turbulence the use of line shooters for deep setting should not be promoted as an effective deterrent to seabirds. [source] Crystallization and preliminary crystallographic study of triple-helical DNAACTA CRYSTALLOGRAPHICA SECTION D, Issue 1 2000Zong-Jin Han Single crystals of d(CTCCTSCCGCGCG)·d(CGCGCGGAG) have been grown by the vapor-diffusion method using 2-methyl-2,4-pentanediol as a precipitant. The crystals are tetragonal, space group P42, with unit-cell parameters a = b = 53.8, c = 43.1,Å, and diffract to 1.8,Å resolution at a synchrotron X-ray beamline. In the crystal, the asymmetric unit contains one copy of the construct. The two halves of the structure are related by non-crystallographic twofold symmetry. These observations are consistent with the conclusion that the sequences of the 12-mer and 9-mer oligonucleotides form a duplex DNA at one end and a triplex DNA at the other end. The inclusion behavior of these rotaxanes was investigated by using one- and two-dimensional NMR spectroscopy. The methyl group at the 2-position of the pyridinium moiety at the end of each axle molecule was found to control the rates of threading of the ,-CD onto the axle molecules. ,-CD can approach axle molecules from a particular direction to form inclusion complexes. Axle molecules that contain a 2-methylpyridinium moiety at one end and a bulky stopper at the other end can regulate the direction of approach to give a [2]pseudorotaxane such as 2,b,,-CD. A [3]pseudorotaxane in which two ,-CD molecules are arranged facing in the same direction at two stations of the tetracationic axle molecule was also obtained. These face-selective behaviors are dominated by kinetic processes rather than thermodynamic processes. [source] Surface-Functionalized Ultrasmall Superparamagnetic Nanoparticles as Magnetic Delivery Vectors for CamptothecinCHEMMEDCHEM, Issue 6 2009Feride Cengelli Abstract Drug,nanoparticle conjugates: The anticancer drug camptothecin (CPT) was covalently linked at the surface of ultrasmall superparamagnetic iron oxide nanoparticles (USPIOs) via a linker, allowing drug release by cellular esterases. Nanoparticles were hierarchically built to achieve magnetically-enhanced drug delivery to human cancer cells and antiproliferative activity. The linking of therapeutic drugs to ultrasmall superparamagnetic iron oxide nanoparticles (USPIOs) allowing intracellular release of the active drug via cell-specific mechanisms would achieve tumor-selective magnetically-enhanced drug delivery. To validate this concept, we covalently attached the anticancer drug camptothecin (CPT) to biocompatible USPIOs (iron oxide core, 9,10,nm; hydrodynamic diameter, 52,nm) coated with polyvinylalcohol/polyvinylamine (PVA/aminoPVA). A bifunctional, end-differentiated dicarboxylic acid linker allowed the attachment of CPT to the aminoPVA as a biologically labile ester substrate for cellular esterases at one end, and as an amide at the other end. These CPT,USPIO conjugates exhibited antiproliferative activity in,vitro against human melanoma cells. The intracellular localization of CPT,USPIOs was confirmed by transmission electron microscopy (iron oxide core), suggesting localization in lipid vesicles, and by fluorescence microscopy (CPT). An external static magnetic field applied during exposure increased melanoma cell uptake of the CPT,USPIOs. The ectopic expression of genes has proven to be an extremely valuable tool for biologists. The most widely used systems involve electrically or chemically mediated transfer of genes to immortalized cell lines and, at the other end of the spectrum, transgenic animal models. As would be expected, there are compromises to be made when using either of these broad approaches. Immortalized cell lines have limited ,physiological relevance' and transgenic approaches are costly and out of the reach of many laboratories. There is also significant time required for the de novo generation of a transgenic animal. 2. As a viable alternative to these approaches, we describe the use of recombinant adenovirus and Sindbis virus to deliver genes to cells and tissues. 3. We exemplify this approach with studies from our laboratories: (i) an investigation of Ca2+ handling deficits in cardiac myocytes of hypertrophied hearts using infection with recombinant adenovirus encoding either green fluorescent protein (GFP) or the sarcoplasmic/endoplasmic reticulum calcium-ATPase (Serca2a); (ii) a study of the mechanism of macrophage/microglial migration by infection of embryonic phagocytes with a GFP-encoding virus and coculture with brain slices to then track the movement of labelled cells; and (iii) we are also exploiting the natural tropism of the Sindbis virus to label neurons in hippocampal brain slices in culture to resolve high-resolution structure and to map neuronal connectivity. 4. Further development of these approaches should open new avenues of investigation for the study of physiology in a range of cells and tissues. [source] Visual detection of IS6110 of Mycobacterium tuberculosis in sputum samples using a test based on colloidal gold and latex beadsCLINICAL MICROBIOLOGY AND INFECTION, Issue 11 2006P. Upadhyay Abstract The IS6110 sequence was detected visually in sputum samples of tuberculosis patients using a bi-probe system. One of the probes was an oligonucleotide conjugated to colloidal gold particles, complementary to one end of the target strand. The other probe was an oligonucleotide conjugated to latex beads complementary to the other end of the target strand. In a reaction mix, these two probes bind to the target strand, and the latex beads are then separated by filtration. Bound latex beads have gold colloid particles at the other end of the target strand. These gold colloid particles were made visible to the naked eye by silver autometallography on the ,invisible' colloidal gold particles. The lower detection limit was 50 ng of genomic DNA of Mycobacterium tuberculosis. This new test, together with conventional PCR, was performed on DNA extracted from sputum samples of suspected tuberculosis patients. The new test was simple to perform, the results were visible to the naked eye, and the test was highly specific, as even single point mutations in the target strand sequence could be differentiated. The test could be useful in field-level laboratories because it requires no sophisticated equipment. [source] |
