Selected AbstractsOn the Weight-bearing Function of the Medial Coronoid Process in DogsANATOMIA, HISTOLOGIA, EMBRYOLOGIA, Issue 1 2006S. Breit Summary The shape of and proportions between the surface areas of the medial coronoid process (MCP) and the fovea of the radial head were determined in 88 juvenile dogs and 146 adult dogs grouped as giant, large, mid-sized, chondrodystrophic, or small dogs. Thereby, the longitudinal (length) and transverse (width) extension of the MCP and fovea of the radial head have been measured. Original values were used to describe changes of the parameters attributed to growth. Normalized values (i.e. values expected in case of a width of the fovea of the radial head of 20 mm) were used to determine potential differences between constitutional types. All original values increased during growth (P < 0.05) except for the width and length of the MCP in chondrodystrophic and small breeds. Normalized values revealed a proportional decrease in width and length of the MCP during growth (P < 0.05) compared with the radial head. In adults, the normalized MCP was widest in giant dogs followed by large, mid-sized, small, and chondrodystrophic breeds. The MCP was also longest in giant dogs but shortest in large and chondrodystrophic dogs with those of large dogs being significantly (P < 0.05) shorter than those in giant, mid-sized and small dogs. Present results suggest that a deficiency in length-growth of the MCP , which has been present especially in large dogs , results in smaller humeral contact areas and decreased weight-bearing capacity of the MCP. Because loading forces acting on the MCP increase with body weight, the condition noted in large dogs might increase the risk of fragmentation of the MCP in these. [source] Glassy Carbon Electrodes Modified with Multiwall Carbon Nanotubes Dispersed in PolylysineELECTROANALYSIS, Issue 15 2008Yamile Jalit Abstract We report the analytical performance of glassy carbon electrodes (GCE) modified with a dispersion of multiwall carbon nanotubes (MWCNT) in polylysine (Plys) (GCE/MWCNT-Plys). The resulting electrodes show an excellent electrocatalytic activity towards different bioanalytes like ascorbic acid, uric acid and hydrogen peroxide, with important decrease in their oxidation overvoltages. The dispersion of 1.0,mg/mL MWCNT in 1.0,mg/mL polylysine is highly stable, since after 2 weeks the sensitivity for hydrogen peroxide at GCE modified with this dispersion remained in a 90% of the original value. The MWCNT-Plys layer immobilized on glassy carbon electrodes has been also used as a platform to build supramolecular architectures by self-assembling of polyelectrolytes based on the polycationic nature of the polylysine used to disperse the nanotubes. The self-assembling of glucose oxidase has allowed us to obtain a supramolecular multistructure for glucose biosensing. The influence of glucose oxidase concentration and adsorption time as well as the effect of using polylysine or MWCNT-Plys as polycationic layers for further adsorption of GOx is also evaluated. [source] Pharmacodynamic Analysis of the Interaction between Tiagabine and Midazolam with an Allosteric Model That Incorporates Signal TransductionEPILEPSIA, Issue 3 2003Daniėl M. Jonker Summary: ,Purpose: The objective of this study was to characterize quantitatively the pharmacodynamic interaction between midazolam (MDL), an allosteric modulator of the ,-aminobutyric acid subtype A (GABAA) receptor, and tiagabine (TGB), an inhibitor of synaptic GABA uptake. Methods: The in vivo concentration,response relation of TGB was determined through pharmacokinetic/pharmacodynamic (PK/PD) modeling. Rats received a single intravenous dose of 10 mg/kg TGB in the absence and the presence of a steady-state plasma concentration of MDL. The EEG response in the 11.5- to 30-Hz frequency band was used as the pharmacodynamic end point. Results: Infusion of MDL resulted in a mean steady-state plasma concentration of 66 ± 3 ng/ml. A significant pharmacokinetic interaction with TGB was observed. MDL inhibited TGB clearance by 20 ± 7 ml/min/kg from the original value of 89 ± 6 ml/min/kg. However, no changes in plasma protein binding of both drugs were observed. The concentration,EEG relation of TGB was described by the sigmoid- Emax model. The pharmacodynamic parameter estimates of TGB were: Emax = 327 ± 10 ,V, EC50 = 392 ± 20 ng/ml, and nH = 3.1 ± 0.3. These values were not significantly different in the presence of MDL. Factors that may explain the lack of synergism were identified by a mechanism-based interaction model that separates the receptor activation from the signal-transduction process. High efficiency of signal transduction and the presence of a baseline response were shown to diminish the degree of synergism. Conclusions: We conclude that the in vivo pharmacodynamic interaction between MDL and TGB is additive rather than synergistic. This strongly suggests that allosteric modulation of the antiseizure activity of a GAT-1 inhibitor by a benzodiazepine does not offer a therapeutic advantage. [source] Water-Soluble Polyfluorenes as an Interfacial Layer Leading to Cathode-Independent High Performance of Organic Solar CellsADVANCED FUNCTIONAL MATERIALS, Issue 12 2010Seung-Hwan Oh Abstract Novel poly[(9,9-bis((6,-(N,N,N -trimethylammonium)hexyl)-2,7-fluorene)- alt -(9,9-bis(2-(2-(2-methoxyethoxy)ethoxy)ethyl)-9-fluorene)) dibromide (WPF-6-oxy-F) and poly[(9,9-bis((6,-(N,N,N -trimethylammonium)hexyl)-2,7-fluorene)- alt -(9,9-bis(2-(2-methoxyethoxy)ethyl)-fluorene)] dibromide (WPF-oxy-F) compounds are developed and the use of these water-soluble polymers as an interfacial layer for low-cost poly(3-hexylthiophene):phenyl-C61 butyric acid methyl ester (P3HT:PCBM) organic solar cells (OSCs) is investigated. When WPF-oxy-F or WPF-6-oxy-F is simply inserted between the active layer and the cathode as an interfacial dipole layer by spin-coating water-soluble polyfluorenes, the open-circuit voltage (Voc), fill factor (FF), and power-conversion efficiency (PCE) of photovoltaic cells with high work-function metal cathodes, such as Al, Ag, Au, and Cu, dramatically increases. For example, when WPF-6-oxy-F is used with Al, Ag, Au, or Cu, regardless of the work-function of the metal cathode, the Voc is 0.64, 0.64, 0.58, and 0.63,V, respectively, approaching the original value of the P3HT:PCBM system because of the formation of large interfacial dipoles through a reduction of the metal work-function. In particular, introducing WPF-6-oxy-F into a low-cost Cu cathode dramatically enhanced the device efficiency from 0.8% to 3.36%. [source] Confocal imaging of chromatographic fouling under flow conditionsJOURNAL OF CHEMICAL TECHNOLOGY & BIOTECHNOLOGY, Issue 10 2007Sun Chau Siu Abstract BACKGROUND: The fouling impact of selected fouling species was assessed by utilising confocal scanning laser microscopy (CSLM) to image a packed chromatographic bed during operation. A custom-made flow cell was packed with Q Sepharose FF and loaded with partially clarified E. coli homogenate. Selective, multicoloured fluorescent dyes were used to label a bovine serum albumin (BSA) test protein (Cy5.5), dsDNA (PicoGreen) and host cell proteins (HCPs) (Cy3). The fouling caused by the various fluorescently labelled components was visualised as a result of the fluorescence emitted by the PicoGreen-labelled dsDNA and the Cy3-labelled protein in the foulant stream, and by testing the adsorptive capacity of a test protein (BSA) onto the resin prior to and post-fouling as well as following the application of a common CIP procedure. RESULTS: Values for the effective diffusivity of BSA (De) were derived from the confocal images and the fouling impact was assessed by comparing De values obtained from different fouling scenarios. Under the most extreme conditions examined, fouling caused a 20% reduction in capacity compared to a fresh bed. BSA diffusivity did not appear to be affected by the fouling conditions studied. Sequential CIP using 15 CVs of 1 mol L,1 NaCl then 15 CVs of 1 mol L,1 NaOH was shown to be effective in removing nucleic acids and HCPs. Subsequent BSA adsorption showed that the CIP regime successfully restored the column capacity to its original value. In contrast, 15 CVs of 1 mol L,1 NaCl were ineffective in removing dsDNA but substantially removed HCPs. CONCLUSION: CSLM was demonstrated to be a useful tool for visualising fouling mechanisms. Comparing the results obtained by this technique using different modes of chromatographic operation provided insights into the fouling characteristics of finite baths versus packed beds. Bandorf Abstract To enhance the lifetime and reliability of microcomponents, thin microtribological films are applied to microparts. With reduction of the component size, investigation methods for tribological testing must be adapted. This paper studies the microtribological behaviour of thin diamond-like carbon (DLC) films using different testing methods. To tie in with macroscopic results, to determine friction we used the well-known pin-on-disc test with spherical surfaces of 10,mm diameter under a typical load of 3,N. For investigations of the behaviour under single asperity contact, Atomic Force Microscope (AFM) methods with applied loads of a few hundred micronewtons were used. Investigations on thin DLC films showed that the friction coefficient under single asperity contact is strongly dependent on the applied load and the resulting contact area. Especially for thin films (up to a few hundred nanometres) the friction coefficient is influenced by the substrate material. With decreasing substrate Young's modulus the friction coefficient also decreases. On the other hand, an increase in the abrasive wear resistance was observed using soft substrate materials. In this paper we show that the friction coefficient was also reduced by a simple surface structure. For investigations we used photolithography to create concentric circles in different substrates. This resulted in a behaviour like riding on rails for the pin-on-disc test. Depending on the tribological pairing the friction coefficient was reduced to more than 50% of the original value. [source] Design of Metabolic Engineering Strategies for Maximizing l -(,)-Carnitine Production by Escherichia coli.BIOTECHNOLOGY PROGRESS, Issue 2 2005Bioreactor Levels, Integration of the Metabolic In this work metabolic engineering strategies for maximizing l -(,)-carnitine production by Escherichia coli based on the Biochemical System Theory (1,3) and the Indirect Optimization Method are presented (4). The model integrates the metabolic and the bioreactor levels using power-law formalism. Based on the S-system model, the Indirect Optimization Method was applied, leading to profiles of parameter values that are compatible with both the physiology of the cells and the bioreactor system operating conditions. This guarantees their viability and fitness and yields higher rates of l -(,)-carnitine production. Experimental results using a high cell density reactor were compared with optimized predictions from the Indirect Optimization Method. When two parameters (the dilution rate and the initial crotonobetaine concentration) were directly changed in the real experimental system to the prescribed optimum values, the system showed better performance in l -(,)-carnitine production (74% increase in production rate), in close agreement with the modelapos;s predictions. The model shows control points at macroscopic (reactor operation) and microscopic (molecular) levels where conversion and productivity can be increased. In accordance with the optimized solution, the next logical step to improve the l -(,)-carnitine production rate will involve metabolic engineering of the E. coli strain by overexpressing the carnitine transferase, CaiB, activity and the protein carrier, CaiT, responsible for substrate and product transport in and out of the cell. By this means it is predicted production may be enhanced by up to three times the original value. Lokey Background The usefulness of rapid intraoperative monitoring of intact (1,84) parathyroid hormone (PTH) is not clearly defined in the surgical management of secondary HPT in the patients on haemodialysis. The aim of this study was to define the normal pattern of decay during surgery for secondary HPT using the rapid intact (1,84) PTH assay during operation. Methods Fifty patients on haemodialysis underwent neck exploration for secondary HPT. The therapeutic goal in all patients was the subtotal resection of four or more glands and bilateral transcervical thymectomy. PTH levels were monitored using a rapid immunochemiluminometric assay. Peripheral blood samples were assayed at induction of anaesthesia, after dissection but before resection, and 20 and 40 min after resection in all patients. All patients were followed up for at least 6 months. PTH levels were expressed as absolute values, as multiples of the upper limit of normal and as the percentage decline from pre-excision values. Results Forty-eight patients (96 per cent) were considered cured after surgery. Twenty patients (40 per cent) had a PTH level less than twice normal and 20 patients (40 per cent) had a PTH level between two and four times normal at 20 min. At late follow-up, all these patients were cured. Ten patients (20 per cent) had a PTH level greater than four times normal at 20 min. Eight of these patients were cured. Seven of these eight had a PTH level at 20 min, while not less than four times normal, less than 40 per cent of the original value. In contrast, the two failures had neither a decline to less than four times normal nor a decay to less than 40 per cent of the original value. One has been reoperated with resection of a fifth gland and one awaits reoperation. Conclusion The intraoperative decay of PTH during surgery for secondary HPT in patients on haemodialysis is slower than that in patients with normal renal function. However, 20 min after resection, a decline to less than four times the upper limit of normal is predictive of cure. Variability of decay slopes in individual patients may reflect molecular heterogeneity or biphasic metabolism of the hormone. © 2000 British Journal of Surgery Society Ltd [source] Simple Bisthiocarbonohydrazones as Sensitive, Selective, Colorimetric, and Switch-On Fluorescent Chemosensors for Fluoride AnionsCHEMISTRY - A EUROPEAN JOURNAL, Issue 10 2007Feng Han Abstract Bisthiocarbonohydrazones are found to be a class of sensitive, selective, ratiometric, and colorimetric chemosensors for anions such as fluoride (F,) or acetate (Ac,). The sensitivities, or the binding constants of the sensors with anions, were found to be strongly dependent on the substituents appended on the ,-conjugation framework, the delocalization bridge CHN, the aromatic moiety, and the hetero atom in the CX group (X=O, S) of the sensors. Single-crystal structures and 1H,NMR titration analysis shows that the CHN moiety is a hydrogen-bond donor, and it is proposed that an additional CH,,,F hydrogen bond is formed for the sensors in the presence F,. A sensor bearing anthracenyl groups is demonstrated as a switch-on fluorescent chemosensor for F, and Ac,. The recognition of F, in acetonitrile (MeCN) by a sensor with nitrophenyl substituents is tolerant to MeOH (MeCN/MeOH=10:1, v/v) and water (MeCN/H2O=30:1, v/v); at these solvent ratios the absorption intensity of the sensor,F, complex solution at maximal absorption wavelength was attenuated to half of the original value in pure MeCN. Hill Summary 1.,Ellenberg's indicator values scale the flora of a region along gradients reflecting light, temperature, continentality, moisture, soil pH, fertility and salinity. They can be used to monitor environmental change. 2.,Ellenberg values can be extended from central Europe, for which they were defined, to nearby parts of Europe. Given a database of quadrat samples, they can be repredicted by a simple algorithm consisting of two-way weighted averaging, followed by local regression. 3.,A database of British samples was assembled from two large surveys. Ellenberg values were repredicted. 4.,Except for the indicator of continentality, the correlation of repredicted and original values was in the range 0·72 (light) to 0·91 (moisture). The continentality indicator could not be adequately repredicted by the algorithm, and is unusable in Britain. 5.,Discrepancies between original and repredicted values can be attributed to various causes, including wrong original values, differing ecological requirements in Britain and central Europe, biased sampling of the British range of habitats, and the occurrence of small plants in shaded or basic microhabitats within well illuminated or predominantly acid quadrats. 6.,The repredicted values were generally reliable, but a small proportion was clearly wrong. [source] Ligament creep recruits fibres at low stresses and can lead to modulus-reducing fibre damage at higher creep stresses: a study in rabbit medial collateral ligament modelJOURNAL OF ORTHOPAEDIC RESEARCH, Issue 5 2002G. M. Thornton Ligaments are subjected to a range of loads during different activities in vivo, suggesting that they must resist creep at various stresses. Cyclic and static creep tests of rabbit medial collateral ligament were used as a model to examine creep over a range of stresses in the toe- and linear-regions of the stress,strain curve: 4.1 MPa (n =7), 7.1 MPa (n = 6), 14 MPa (n = 9) and 28 MPa (n = 6). We quantified ligament creep behaviour to determine if, at low stresses, modulus would increase in a cyclic creep test and collagen fibres would be recruited in a static creep test. At higher creep stresses, a decrease in measured modulus was expected to be a potential marker of damage. The increase in modulus during cyclic creep and the increase in strain during static creep were similar between the three toe-region stresses (4.1, 7.1, 14 MPa). However, at the linear-region stress (28 MPa), both these parameters increased significantly compared to the increases at the three toe-region stresses. A concurrent crimp analysis revealed that collagen fibres were recruited during creep, evidenced by decreased area of crimped fibres at the end of the static creep test. Interestingly, a predominance of straightened fibres was observed at the end of the 28 MPa creep test, suggesting a limited potential for fibre recruitment at higher, linear-region stresses. An additional 28 MPa (n = 6) group had mechanically detectable discontinuities in their stress,strain curves during creep that were related to reductions in modulus and suggested fibre damage. These data support the concept that collagen fibre recruitment is a mechanism by which ligaments resist creep at low stresses. At a higher creep stress, which was still only about a third of the failure capacity, damage to some ligaments occurred and was marked by a sudden reduction in modulus. In the cyclic tests, with continued cycling, the modulus increased back to original values obtained before the discontinuity suggesting that other fibres were being recruited to bear load. These results have important implications for our understanding of how fibre recruitment and stress redistribution act in normal ligament to minimize creep and restore modulus after fibre damage. © 2002 Orthopaedic Research Society. [source] Helium Isotope Geochemistry of Ore-forming Fluids from Furong Tin Orefield in Hunan Province, ChinaRESOURCE GEOLOGY, Issue 1 2006Zhao-li Li Abstract. The Furong tin orefield, located in southern Hunan, China, is a newly-discovered super-large tin orefield. In contrast to most other tin deposits associated with S-type granites, the Furong tin deposit is closely associated with the Qitianling A-type granite. The 3He/4He ratios of fluid inclusions in pyrite and arsenopyrite from this orefield range from 0.13 to 2.95 Ra. The influence of various post-mineralization processes on the helium isotopic composition of ore-forming fluid inclusions are estimated negligible. Thus, the analytical values of helium isotopic composition basically represent the original values of ore-forming fluids at the time they were trapped. The 3He/4He ratios of ore-forming fluids from the Furong orefield indicate the existence of mantle-source components. It supports the idea that both the Furong tin orefield and Qitianling granite formed under the geodynamic background of mantle upwelling and crustal extension.
