[source] Rhodium-Catalyzed Asymmetric Nitroallylation of Arylmetallics with Cyclic Nitroallyl Acetates and Applications in Organic SynthesisEUROPEAN JOURNAL OF ORGANIC CHEMISTRY, Issue 18 2006Lin Dong Abstract Highly enantioselective rhodium-catalyzed nitroallylations of arylboronic acids and arylzinc chlorides with cyclic nitroallyl acetates are described. Catalyst screening indicated that the rhodium complex of [Rh(OH)(COD)]2 and optically pure binap is the optimal catalyst for the nitroallylation of arylboronic acids with 2-nitrocyclohex-2-enyl esters, providing good yields and high enantioselectivities of up to 99,% ee. The rhodium complex prepared from Rh(acac)(C2H4)2 and (R)-binap efficiently catalyzed the nitroallylation of arylzinc chlorides with 2-nitrocyclohex-2-enyl acetate at 0 °C in high yields of up to 93,% and with high enantioselectivities of up to 96,% ee. A number of synthetically useful intermediates with high optical purity were prepared with this reaction as starting point: concise total syntheses of optically pure (+)-,-lycorane in 53,% overall yield and of (+)-,-lycorane in 52,% overall yield were achieved by commencing with the asymmetric nitroallylation of 3,4-methylenedioxyphenylzinc chloride with 2-nitrocyclohex-2-enyl acetate. To apply this reaction to large-scale preparations of the said bicyclic acrylates, a flexible Pd catalyst system with tunable reactivity has been developed. The dependence of the product and diastereomer distribution on both the reaction conditions, including the type of palladium catalyst used, and on the nature of the substrate has been investigated. A variety of methyl 2-(bicyclo[3.1.0]hex-1-yl)acrylates and parent carboxylic acids as well as some of their derivatives of potential interest towards a technical application were prepared on a multigram scale. A general large-scale synthesis of the cyclization precursors bearing one or two carbonyl groups in the tether is also disclosed. Smooth and rugged surfaces can be created by varying the polymerization procedure. We have shown that the polyacrolein resins with a high loading of aldehyde groups serve as effective scavengers for primary amines and may be used to remove compounds bearing amino groups in the combinatorial synthesis of compound libraries. Copolymerization with styrene can help to separate the adjacent aldehyde groups, thus making the functional groups more available in organic reactions. The polyacrolein resins in the aldehyde form or after appropriate chemical modifications may also be useful as support materials in solid-phase synthesis. The influence of the Ca2+ channel antagonists on pancreatic , cell functions is dependent on lipophilicity, interactions with the cell membrane lipid bilayer, with SNAREs protein complexes in cell and vesicle membranes, with intracellular receptors, bioavailability and time of elimination from several organs and the bloodstream. In the present work we studied the effect at several doses of new compounds synthesized in the Latvian Institute of Organic Synthesis on blood glucose levels in normal and STZ-induced diabetic rats. The compounds tested were: 1,4-DHP derivatives cerebrocrast (1), etaftoron (2), OSI-1190 (3), OSI-3802 (4), OSI-2954 (5) and known 1,4-DHP derivatives: niludipine (6), nimodipine (7) and nicardipine (8) which possess different lipophilicities. Analysis of the structure,function relationships of the effect of 1,4-DHP derivatives on glucose metabolism showed that cerebrocrast could evoke qualitative differences in activity. Insertion of an OCHF2 group in position 2 of the 4-phenylsubstituent and propoxyethylgroup R in ester moieties in positions 3 and 5 of the DHP structure, as well as an increase in the number of carbon atoms in the ester moiety, significantly modified the properties of the compound. Thereby cerebrocrast acquired high lipophilicity and membranotropic properties. Cerebrocrast, in a single administration at low doses (0.05 and 0.5,mg,kg,1, p.o.), significantly decreased the plasma level of glucose in normal rats and in STZ-induced diabetic rats returned plasma glucose to basal levels. This effect was characterized by a slow onset and a powerful long-lasting influence on glucose metabolism, especially in STZ-induced diabetic rats. Copyright © 2004 John Wiley & Sons, Ltd.
