Polanska Abstract Fibroblast growth factors (FGFs) and their receptors (FGFRs) regulate a multitude of biological functions in embryonic development and in adult. A major question is how does one family of growth factors and their receptors control such a variety of functions? Classically, specificity was thought to be imparted by alternative splicing of the FGFRs, resulting in isoforms that bind specifically to a subset of the FGFs, and by different saccharide sequences in the heparan sulfate proteoglycan (HSPG) co-receptor. A growing number of noncanonical co-receptors such as integrins and neural cell adhesion molecule (NCAM) are now recognized as imparting additional complexity to classic FGFR signaling. This review will discuss the noncanonical FGFR ligands and speculate on the possibility that they provide additional and alternative means to determining the functional specificity of FGFR signaling. [source] Heparan sulfate proteoglycans in experimental models of diabetes: a role for perlecan in diabetes complicationsDIABETES/METABOLISM: RESEARCH AND REVIEWS, Issue 6 2001Karin Conde-Knape Abstract Proteoglycans are ubiquitous extracellular proteins that serve a variety of functions throughout the organism. Unlike other glycoproteins, proteoglycans are classified based on the structure of the glycosaminoglycan carbohydrate chains, not the core proteins. Perlecan, a member of the heparan sulfate proteoglycan (HSPG) family, has been implicated in many complications of diabetes. Decreased levels of perlecan have been observed in the kidney and in other organs, both in patients with diabetes and in animal models. Perlecan has an important role in the maintenance of the glomerular filtration barrier. Decreased perlecan in the glomerular basement membrane has a central role in the development of diabetic albuminuria. The involvement of this proteoglycan in diabetic complications and the possible mechanisms underlying such a role have been addressed using a variety of models. Due to the importance of nephropathy among diabetic patients most of the studies conducted so far relate to diabetes effects on perlecan in different types of kidney cells. The various diabetic models used have provided information on some of the mechanisms underlying perlecan's role in diabetes as well as on possible factors affecting its regulation. However, many other aspects of perlecan metabolism still await full elucidation. The present review provides a description of the models that have been used to study HSPG and in particular perlecan metabolism in diabetes and some of the factors that have been found to be important in the regulation of perlecan. E. Koenig Summary The integron/gene cassette systems identified in bacteria comprise a class of genetic elements that allow adaptation by acquisition of gene cassettes. Integron gene cassettes have been shown to facilitate the spread of drug resistance in human pathogens but their role outside a clinical setting has not been explored extensively. We sequenced 2145 integron gene cassettes from four marine sediment samples taken from the vicinity of Halifax Nova Scotia, Canada, increasing the number of gene cassettes obtained from environmental microbial communities by 10-fold. Sequence analyses reveals that the majority of these cassettes encode novel proteins and that this study is consistent with previous claims of high cassette diversity as we estimate a Chao1 diversity index of ,3000 cassettes from these samples. The functional distribution of environmental cassettes recovered in this study, when compared with that of cassettes from the only other source with significant sampling (Vibrio genomes) suggests that alternate selection regimes might be acting on these two gene pools. The majority of cassettes recovered in this study encode novel, unknown proteins. In instances where we obtained multiple alleles of a novel protein we demonstrate that non-synonymous versus synonymous substitution rates ratios suggest relaxed selection. Cassette-encoded proteins with known homologues represent a variety of functions and prevalent among these are isochorismatases; proteins involved in iron scavenging. Phylogenetic analysis of these isochorismatases as well as of cassette-encoded acetyltransferases reveals a patchy distribution, suggesting multiple sources for the origin of these cassettes. Finally, the two most environmentally similar sample sites considered in this study display the greatest overlap of cassette types, consistent with the hypothesis that cassette genes encode adaptive proteins. [source] Territorial behaviour and immunity are mediated by juvenile hormone: the physiological basis of honest signalling?FUNCTIONAL ECOLOGY, Issue 1 2009Jorge Contreras-Garduño Summary 1The role of the juvenile hormone (JH) as a potential mediator in the trade-off between male,male competition and immune response has not been tested, but its study could reveal a potential mechanism that mediates resource allocation between these two traits. 2Controlling for body size, we tested whether males of the territorial damselfly Calopteryx virgo administrated with methoprene acid, an analog of the JH (JHa), compared to control males, increased their aggression and occupation time on territories but decreased their phenoloxidase (PO) activity (a key enzyme used during immune response after a bacterial challenge). We found an increase in aggression in JHa treated males compared to control males, but the opposite was found for PO activity. 3As fat load and muscle mass are also important traits during a contest, we tested whether JHa males compared to control males showed more fat and muscle content 2 h after JHa administration. Our results did not show a significant difference between both male groups, suggesting that JHa only increased aggression. 4These results and a review of other published articles, which have documented an effect of JH on a variety of functions in insects, suggest that JH may be a target of sexual selection: this hormone not only promotes the expression of secondary sexual characters but also seems condition-dependent and so its titers may indicate male condition. [source] Isolation of the salmonid rhamnose-binding lectin STL2 from spores of the microsporidian fish parasite Loma salmonaeJOURNAL OF FISH DISEASES, Issue 8 2005A Booy Abstract The microsporidian parasite, Loma salmonae, is the causative agent of gill disease in both wild and netpen-reared salmonids worldwide. In this paper we report the finding of a rhamnose-binding lectin from steelhead trout, Oncorhynchus mykiss, which was found bound in high concentration to the surface coat of L. salmonae spores. SDS-PAGE, immunoblot, N-terminal sequencing and mass spectrometric analyses were used to determine that the dominant 24 kDa protein lectin observed on SDS-PAGE analysis of intact spore extracts is the O. tshawytscha variant of the previously identified rhamnose-binding lectin STL2 from rainbow trout, O. mykiss. Although the physiological role of these lectins has not been clearly delineated, they have been implicated in a variety of functions, including inhibition of pathogenic bacteria by opsonization and macrophage-mediated tumour lysis. Harrison Abstract Legacy software maintenance is a significant cost item for many engineering organizations. This study is a preliminary report on work to investigate maintenance data, usage, and source code for legacy software used by an engineering design company to support a variety of functions, including electromagnetic, thermal, mechanical loading, vibration, and aerodynamic analysis. The results verify the applicability to legacy engineering software of previous research that concluded that size and structural metrics alone are not good indicators of high maintenance costs. Unlike previous research, the study also evaluates the effect of program usage on maintenance cost. Over the three-year period of this study of 71 legacy engineering programs, 11 of the programs (15%) accounted for 80% of all maintenance and 67% of all program runs. The highest maintenance programs were not always the largest programs or the worst structured programs. 49% of the programs accounted for only 1% of total maintenance but 42% of the total lines of code (LOC) thus invalidating LOC as an indicator for maintenance cost. While additional work is needed to validate these findings across other organizations and other code sets, these preliminary results provide strong evidence that expected program usage can be a useful indicator of long-term maintenance cost. [source] Redox regulation of cyclophilin A by glutathionylationPROTEINS: STRUCTURE, FUNCTION AND BIOINFORMATICS, Issue 3 2006Pietro Ghezzi Abstract Using redox proteomics techniques to characterize the thiol status of proteins in human T lymphocytes, we identified cyclophilin,A (CypA) as a specifically oxidized protein early after mitogen activation. CypA is an abundantly expressed cytosolic protein, target of the immunosuppressive drug cyclosporin,A (CsA), for which a variety of functions has been described. In this study, we could identify CypA as a protein undergoing glutathionylation in vivo. Using MALDI-MS we identified Cys52 and Cys62 as targets of glutathionylation in T,lymphocytes, and, using bioinformatic tools, we defined the reasons for the susceptibility of these residues to the modification. In addition, we found by circular dichroism spectroscopy that glutathionylation has an important impact on the secondary structure of CypA. Finally, we suggest that glutathionylation of CypA may have biological implications and that CypA may play a key role in redox regulation of immunity. Waterman Summary Salmonella enterica uses two functionally distinct type III secretion systems encoded on the pathogenicity islands SPI-1 and SPI-2 to transfer effector proteins into host cells. A major function of the SPI-1 secretion system is to enable bacterial invasion of epithelial cells and the principal role of SPI-2 is to facilitate the replication of intracellular bacteria within membrane-bound Salmonella -containing vacuoles (SCVs). Studies of mutant bacteria defective for SPI-2-dependent secretion have revealed a variety of functions that can be attributed to this secretion system. These include an inhibition of various aspects of endocytic trafficking, an avoidance of NADPH oxidase-dependent killing, the induction of a delayed apoptosis-like host cell death, the control of SCV membrane dynamics, the assembly of a meshwork of F-actin around the SCV, an accumulation of cholesterol around the SCV and interference with the localization of inducible nitric oxide synthase to the SCV. Several effector proteins that are translocated across the vacuolar membrane in a SPI-2-dependent manner have now been identified. These are encoded both within and outside SPI-2. The characteristics of these effectors, and their relationship to the physiological functions listed above, are the subject of this review. The emerging picture is of a multifunctional system, whose activities are explained in part by effectors that control interactions between the SCV and intracellular membrane compartments. [source] |
