Burkel Abstract Actin filaments (F-actin) are protein polymers that undergo rapid assembly and disassembly and control an enormous variety of cellular processes ranging from force production to regulation of signal transduction. Consequently, imaging of F-actin has become an increasingly important goal for biologists seeking to understand how cells and tissues function. However, most of the available means for imaging F-actin in living cells suffer from one or more biological or experimental shortcomings. Here we describe fluorescent F-actin probes based on the calponin homology domain of utrophin (Utr-CH), which binds F-actin without stabilizing it in vitro. We show that these probes faithfully report the distribution of F-actin in living and fixed cells, distinguish between stable and dynamic F-actin, and have no obvious effects on processes that depend critically on the balance of actin assembly and disassembly. [source] The commonly used marker ELAV is transiently expressed in neuroblasts and glial cells in the Drosophila embryonic CNSDEVELOPMENTAL DYNAMICS, Issue 12 2007Christian Berger Abstract Glial cells in the Drosophila embryonic nervous system can be monitored with the marker Reversed-polarity (Repo), whereas neurons lack Repo and express the RNA-binding protein ELAV (Embryonic Lethal, Abnormal Vision). Since the first description of the ELAV protein distribution in 1991 (Robinow and White), it is believed that ELAV is an exclusive neuronal and postmitotic marker. Looking at ELAV expression, we unexpectedly observed that, in addition to neurons, ELAV is transiently expressed in embryonic glial cells. Furthermore, it is transiently present in the proliferating longitudinal glioblast, and it is transcribed in embryonic neuroblasts. Likewise, elav -Gal4 lines, which are generally used as postmitotic neuronal driver lines, show expression in neural progenitor cells and nearly all embryonic glial cells. Thus, in the embryo, elav can no longer be considered an exclusive marker or driver for postmitotic neurons. elav loss-of-function mutants show no obvious effects on the number and pattern of embryonic glia. [source] Vascular regression is required for mesenchymal condensation and chondrogenesis in the developing limbDEVELOPMENTAL DYNAMICS, Issue 3 2001Melinda Yin Abstract Vascular regression occurs during limb mesenchymal cell condensation and chondrogenesis, but it is unclear whether it is required for these processes or is a secondary phenomenon without major regulatory roles. To address this issue, beads presoaked with the potent angiogenic factor vascular endothelial growth factor (VEGF) were implanted in the vicinity of the prospective digit 2 in early chick embryo wing buds and the effects on angiogenesis and digit development were determined over time. We found that VEGF treatment caused a marked local increase in blood vessel number and density. Strikingly, this was accompanied by inhibition of digit 2 development as revealed by lack of expression of chondrogenic transcription factor Sox9 and absence of Alcian blue staining. Vascular distribution and skeletal development in adjacent areas remained largely unaffected. Inhibition of digit formation and excess vascularization were both reversible upon further embryonic growth and dissipation of VEGF activity. When supernumerary digits were induced at the anterior limb margin by retinoic acid treatment, their development was also preceded by vascular regression; interestingly, cotreatment with VEGF inhibited supernumerary digit development as well. Direct exposure of limb mesenchymal cells in micromass cultures to VEGF caused no obvious effects on condensation and chondrogenesis, indicating that VEGF effects are not due to direct action on skeletal cells. Our results are the first to provide evidence that vascular regression is required for mesenchymal condensation and chondrogenesis. A model of how patterning mechanisms and vascular regression may intersect and orchestrate limb skeletogenesis is proposed. © 2001 Wiley-Liss, Inc. [source] Inter-sexual differences in the immune response of Eurasian kestrel nestlings under food shortageECOLOGY LETTERS, Issue 1 2002Juan A. Fargallo When resources are limited, parents should decide the optimal number, size, and sex of progeny, and offspring should decide the optimal allocation of resources to different costly functions, such as growth and immunity. We manipulated clutch sizes of Eurasian kestrels by one egg to estimate possible cumulative effects of incubation and chick rearing costs on parental body condition, feeding effort, and offspring viability. No obvious effects of clutch size manipulations on feeding effort were found while feeding effort was adjusted to the original clutch size. Enlarged and control nests suffered from higher nestling mortality than reduced nests, and chicks of the enlarged group were in poorer body condition than chicks of the reduced group. Controlling for body mass, male chicks exhibited lower cell-mediated immunity assessed by a cutaneous hypersensitivity response than females, but only in treatments suffering from food restrictions, as indicated by chick starvation. These novel results reveal inter-sexual differences in physiological strategies in early life, suggesting sex-related differences in susceptibility to disease and consequently in survival prospects of offspring. [source] Engineered 5S ribosomal RNAs displaying aptamers recognizing vascular endothelial growth factor and malachite greenJOURNAL OF MOLECULAR RECOGNITION, Issue 2 2009Xing Zhang Abstract In previous work, Vibrio proteolyticus 5S rRNA was shown to stabilize 13,50 nucleotide "guest" RNA sequences for expression in Escherichia coli. The expressed chimeric RNAs accumulated to high levels in E. coli without being incorporated into ribosomes and without obvious effects on the host cells. In this work, we inserted sequences encoding known aptamers recognizing a protein and an organic dye into the 5S rRNA carrier and showed that aptamer function is preserved in the chimeras. A surface plasmon resonance competitive binding assay demonstrated that a vascular endothelial growth factor (VEGF) aptamer/5S rRNA chimera produced in vitro by transcriptional runoff could compete with a DNA aptamer for VEGF, implying binding of the growth factor by the VEGF "ribosomal RNA aptamer." Separately, a 5S rRNA chimera displaying an aptamer known to increase the fluorescence of malachite green (MG) also enhanced MG fluorescence. Closely related control rRNA molecules showed neither activity. The MG aptamer/5S rRNA chimera, like the original MG aptamer, also increased the fluorescence of other triphenyl methane (TPM) dyes such as crystal violet, methyl violet, and brilliant green, although less effectively than with MG. These results indicate that the molecular recognition properties of aptamers are not lost when they are expressed in the context of a stable 5S rRNA carrier. Inclusion of the aptamer in a carrier may facilitate production of large quantities of RNA aptamers, and may open an approach to screening aptamer libraries in vivo. [source] The Effects of Dietary Yeast Culture or Short-chain Fructo-oligosaccharides on the Intestinal Autochthonous Bacterial Communities in Juvenile Hybrid Tilapia, Oreochromis niloticus,×Oreochromis aureus,JOURNAL OF THE WORLD AQUACULTURE SOCIETY, Issue 4 2009Zhi-Gang Zhou The effects of dietary yeast culture (YC) or short-chain fructo-oligosaccharides (FOS) on intestinal autochthonous bacterial communities in juvenile hybrid tilapia, Oreochromis niloticus,×Oreochromis aureus, were studied by 16S rDNA denaturing gradient gel electrophoresis (DGGE). Ninety Tilapias in tanks (10 fish per tank) were randomly and equally divided into three groups. At the end of an 8-wk feeding period of CK (the control treatment), YC (3 g/kg), or FOS (1 g/kg), autochthonous gut bacteria were analyzed in intestinal samples of all fish in each tank of a recirculating aquaculture system. The clear differences in the banding patterns indicated the obvious effects of dietary prebiotics on intestinal communities in hybrid tilapia. Higher variation was detected within the dietary YC group. This difference might be due to the effects of certain immune-stimulating agents in YC on the immunity response of hybrid tilapia. It was concluded that dietary prebiotics, YC, and FOS obviously affected the intestinal bacterial community in hybrid tilapia with different patterns for different kinds. [source] Preparation and Characterization of Trialkoxysilane-Containing Acrylic Resin/Alumina Hybrid MaterialsMACROMOLECULAR MATERIALS & ENGINEERING, Issue 8 2006Mingqi Li Abstract Summary: In the present study (3-methacryloxypropyl)trimethoxysilane (MPMS) containing acrylic resin/alumina hybrid materials with various alumina contents were prepared. The effects of ethylacetoacetate (EAA) content, catalyst type, and water content during sol,gel process for alumina sols on the microstructure and properties of the hybrid materials were investigated by SAXS, AFM, DSC, TGA, and nano-indentation tester, respectively. It was found that the hybrid materials exhibited a homogeneity and the alumina phase of the hybrids had mass fractal dimension and open structure. The thermal and mechanical properties of the hybrid materials were obviously improved when alumina was incorporated. The EAA contents, catalyst type, and water content during sol,gel process for alumina sols had obvious effects on the microstructure and properties of the hybrid materials. [source] Differential expression and activation of Stat3 during mouse embryo implantation and decidualizationMOLECULAR REPRODUCTION & DEVELOPMENT, Issue 1 2004Chun-Bo Teng Abstract Signal transducer and activator of transcription (STATs) can be activated by many cytokines and growth factors. Stat3, a member of STAT family, is essential for embryonic development. Stat3 is specifically activated during mouse embryo implantation. This study was to investigate the expression, activation, and regulation of Stat3 in mouse uterus during early pregnancy, pseudopregnancy, delayed implantation, artificial decidualization, and hormonal treatments using in situ hybridization and immunohistochemistry. There was a strong level of Stat3 phosphorylation in the luminal epithelium only at the midnight of day 4 pregnancy, which coincides with attachment reaction between the blastocyst and luminal epithelium. However, there was no detectable Stat3 phosphorylation at the corresponding period during pseudopregnancy. On day 5 of pregnancy, Stat3 phosphorylation was strongly observed in the luminal epithelium and the stroma surrounding the implanting blastocyst at implantation sites, but not at the inter-implantation sites. Stat3 phosphorylation was also not detected on day 5 of pseudopregnancy. Stat3 phosphorylation was at a high level in the decidual cells on days 6,8 of pregnancy. Under artificial decidualization, Stat3 was also phosphorylated in the decidual cells. In the ovariectomized mice, there was no Stat3 expression and activation in the uterus. Progesterone had no obvious effects. However, Stat3 mRNA expression and phosphorylation were significantly stimulated by estrogen treatment. Our data suggest that Stat3 phosphorylation may be important for mouse embryo implantation and decidualization, and may also be regulated by maternal estrogen. Mol. Reprod. [source] In vitro cytotoxic, antiprotozoal and antimicrobial activities of medicinal plants from VanuatuPHYTOTHERAPY RESEARCH, Issue 6 2010Gesine Bradacs Abstract Sixty-three extracts obtained from 18 plants traditionally used in the South Pacific archipelago Vanuatu for the treatment of infectious diseases were screened for antimicrobial and antiprotozoal activities. In addition, the extracts were subjected to a detailed analysis on cytotoxic effects toward a panel of human cancer cell lines, designed as a smaller version of the NCI60 screen. Intriguingly, 15 plant extracts exhibited strong cytotoxic effects specific for only one cancer cell line. Extracts of the leaves of Acalypha grandis Benth. significantly affected Plasmodium falciparum without showing obvious effects against the other protozoa tested. The leaves of Gyrocarpus americanus Jacq. displayed significant activity against Trypanosoma b. brucei and the leaves of Tabernaemontana pandacaqui Lam. I as well as the stems of Macropiper latifolium (L.f.) against Trypanosoma cruzi. In contrast none of the extracts showed relevant antibacterial or antifungal activity.
