[source] Differentiating Atrioventricular Nodal Reentrant Tachycardia from Junctional Tachycardia: Novel Application of the Delta H-A IntervalJOURNAL OF CARDIOVASCULAR ELECTROPHYSIOLOGY, Issue 1 2008KOMANDOOR SRIVATHSAN M.D. Introduction: Junctional tachycardia (JT) and atrioventricular nodal reentrant tachycardia (AVNRT) can be difficult to differentiate. Yet, the two arrhythmias require distinct diagnostic and therapeutic approaches. We explored the utility of the delta H-A interval as a novel technique to differentiate these two tachycardias. Methods: We included 35 patients undergoing electrophysiology study who had typical AVNRT, 31 of whom also had JT during slow pathway ablation, and four of whom had spontaneous JT during isoproterenol administration. We measured the H-A interval during tachycardia (H-AT) and during ventricular pacing (H-AP) from the basal right ventricle. Interobserver and intraobserver reliability of measurements was assessed. Ventricular pacing was performed at approximately the same rate as tachycardia. The delta H-A interval was calculated as the H-AP minus the H-AT. Results: There was excellent interobserver and intraobserver agreement for measurement of the H-A interval. The average delta H-A interval was ,10 ms during AVNRT and 9 ms during JT (P < 0.00001). For the diagnosis of JT, a delta H-A interval , 0 ms had the sensitivity of 89%, specificity of 83%, positive predictive value of 84%, and negative predictive value of 88%. The delta H-A interval was longer in men than in women with JT, but no gender-based differences were seen with AVNRT. There was no difference in the H-A interval based on age , 60 years. Conclusion: The delta H-A interval is a novel and reproducibly measurable interval that aids the differentiation of JT and AVNRT during electrophysiology studies. [source] Immunoisolated Chromaffin Cells Implanted Into the Subarachnoid Space of Rats Reduce Cold Allodynia in a Model of Neuropathic Pain: A Novel Application of Microencapsulation TechnologyARTIFICIAL ORGANS, Issue 12 2004Yu Mi Kim Abstract:, Intrathecal transplants of adrenal medullary chromaffin cells relieve chronic pain by secreting catecholamines, opioids, and other neuroactive substances. Recently, macrocapsules with semipermeable membranes were used to isolate immunologically xenogenic chromaffin cells, but the poor viability in vivo of the encapsulated chromaffin cells limited the usefulness of this method. In this study, we used a novel method of encapsulation to increase the viability of chromaffin cells. We found that microencapsulated chromaffin cells that were implanted into the subarachnoid space of rats relieved cold allodynia in a model of neuropathic pain. Furthermore, microencapsulated chromaffin cells were morphologically normal and retained their functionality. These findings suggest that the intrathecal placement of microencapsulated chromaffin cells might be a useful method for treating chronic pain. [source] Novel application of flow cytometry: Determination of muscle fiber types and protein levels in whole murine skeletal muscles and heartCYTOSKELETON, Issue 12 2007Connie Jackaman Abstract Conventional methods for measuring proteins within muscle samples such as immunohistochemistry and western blot analysis can be time consuming, labor intensive and subject to sampling errors. We have developed flow cytometry techniques to detect proteins in whole murine heart and skeletal muscle. Flow cytometry and immunohistochemistry were performed on quadriceps and soleus muscles from male C57BL/6J, BALB/c, CBA and mdx mice. Proteins including actins, myosins, tropomyosin and ,-actinin were detected via single staining flow cytometric analysis. This correlated with immunohistochemistry using the same antibodies. Muscle fiber types could be determined by dual labeled flow cytometry for skeletal muscle actin and different myosins. This showed similar results to immunohistochemistry for I, IIA and IIB myosins. Flow cytometry of heart samples from C57BL/6J and BALB/c mice dual labeled with cardiac and skeletal muscle actin antibodies demonstrated the known increase in skeletal actin protein in BALB/c hearts. The membrane-associated proteins ,-sarcoglycan and dystrophin could be detected in C57BL/6J mice, but were decreased or absent in mdx mice. With the ability to label whole muscle samples simultaneously with multiple antibodies, flow cytometry may have advantages over conventional methods for certain applications, including assessing the efficacy of potential therapies for muscle diseases. Stok Abstract A new technique for characterization of the three-dimensional morphology of murine articular cartilage is proposed. The technique consists of a novel application of confocal laser scanning microscopy (CLSM), where the objective was to develop and validate it for cartilage measurements in murine joints. Murine models are used in arthritis research, because they are well-described for manipulating the disease pathophysiology, facilitating our understanding of the disease, and identifying new targets for therapy. A calibration and reproducibility study was carried out to provide a consistent testing methodology for quantification of murine joints. The proximal tibial condyles from male C57BL/6 mice were scanned using a CLS microscope with an isotropic voxel size of 5.8 ,m. Measurements and analyses were repeated three times on different days, and in a second step the analysis was repeated three times for a single measurement. Calculation of precision errors (coefficient of variation) for cartilage thickness and volume was made. The bias of the system was estimated through comparison with histology. This technique showed good precision, with errors in the repeated analysis ranging from 0.63% (lateral thickness) to 3.48% (medial volume). The repeated analysis alone was robust, with intraclass correlations for the different compartments between 0.918 and 0.991. Measurement bias was corrected by scaling the confocal images to 32% of their width to match histology. CLSM provided a fast and reproducible technique for gathering 3D image data of murine cartilage and will be a valuable tool in understanding the efficacy of arthritis treatments in murine models. [source] Novel application of oxygen-transferring membranes to improve anaerobic wastewater treatmentBIOTECHNOLOGY & BIOENGINEERING, Issue 4 2005Anthony S. Kappell Abstract Anaerobic biological wastewater treatment has numerous advantages over conventional aerobic processes; anaerobic biotechnologies, however, still have a reputation for low-quality effluents and operational instabilities. In this study, anaerobic bioreactors were augmented with an oxygen-transferring membrane to improve treatment performance. Two anaerobic bioreactors were fed a synthetic high-strength wastewater (chemical oxygen demand, or COD, of 11,000 mg l,1) and concurrently operated until biomass concentrations and effluent quality stabilized. Membrane aeration was then initiated in one of these bioreactors, leading to substantially improved COD removal efficiency (>95%) compared to the unaerated control bioreactor (,65%). The membrane-augmented anaerobic bioreactor required substantially less base addition to maintain circumneutral pH and exhibited 75% lower volatile fatty acid concentrations compared to the unaerated control bioreactor. The membrane-aerated bioreactor, however, failed to improve nitrogenous removal efficiency and produced 80% less biogas than the control bioreactor. A third membrane-augmented anaerobic bioreactor was operated to investigate the impact of start-up procedure on nitrogenous pollutant removal. In this bioreactor, excellent COD (>90%) and nitrogenous (>95%) pollutant removal efficiencies were observed at an intermediate COD concentration (5,500 mg l,1). Once the organic content of the influent wastewater was increased to full strength (COD = 11,000 mg l,1), however, nitrogenous pollutant removal stopped. This research demonstrates that partial aeration of anaerobic bioreactors using oxygen-transferring membranes is a novel approach to improve treatment performance. Additional research, however, is needed to optimize membrane surface area versus the organic loading rate to achieve the desired effluent quality. © 2005 Wiley Periodicals, Inc. [source] Investigating Burkholderia cepacia complex populations recovered from Italian maize rhizosphere by multilocus sequence typingENVIRONMENTAL MICROBIOLOGY, Issue 7 2007Claudia Dalmastri Summary The Burkholderia cepacia complex (BCC) comprises at least nine closely related species of abundant environmental microorganisms. Some of these species are highly spread in the rhizosphere of several crop plants, particularly of maize; additionally, as opportunistic pathogens, strains of the BCC are capable of colonizing humans. We have developed and validated a multilocus sequence typing (MLST) scheme for the BCC. Although widely applied to understand the epidemiology of bacterial pathogens, MLST has seen limited application to the population analysis of species residing in the natural environment; we describe its novel application to BCC populations within maize rhizospheres. 115 BCC isolates were recovered from the roots of different maize cultivars from three different Italian regions over a 9-year period (1994,2002). A total of 44 sequence types (STs) were found of which 41 were novel when compared with existing MLST data which encompassed a global database of 1000 clinical and environmental strains representing nearly 400 STs. In this study of rhizosphere isolates approximately 2.5 isolates per ST was found, comparable to that found for the whole BCC population. Multilocus sequence typing also resolved inaccuracies associated with previous identification of the maize isolates based on recA gene restriction fragment length polymorphims and species-specific polymerase chain reaction. The 115 maize isolates comprised the following BCC species groups, B. ambifaria (39%), BCC6 (29%), BCC5 (10%), B. pyrrocinia (8%), B. cenocepacia IIIB (7%) and B. cepacia (6%), with BCC5 and BCC6 potentially constituting novel species groups within the complex. Closely related clonal complexes of strains were identified within B. cepacia, B. cenocepacia IIIB, BCC5 and BCC6, with one of the BCC5 clonal complexes being distributed across all three sampling sites. Overall, our analysis demonstrates that the maize rhizosphere harbours a massive diversity of novel BCC STs, so that their addition to our global MLST database increased the ST diversity by 10%. In sequel, the paper presents an application of the conceptually new computer program, based on the symbolic analysis of load flows in power system networks. The network parameters and variables are defined as symbols. The symbolic analyzer which models analytically the power system DC load flows enables the sensitivity analysis of the power system to parameter and variable variations (costs, transfers, injections), a valuable tool for the expansion planning analysis. That virtue could not be found within the conventional approach, relying on compensation methods, precalculated distribution factors, and so on. This novel application sheds some light on the traditional power system network expansion planning method, as well as on its possible application within the system network expansion planning in the new environment assuming the competitive electric power market. Bleakley How and why cooperation evolves, particularly among nonrelatives, remains a major paradox for evolutionary biologists and behavioral ecologists. Although much attention has focused on fitness consequences associated with cooperating, relatively little is known about the second component of evolutionary change, the inheritance of cooperation or reciprocity. The genetics of behaviors that can only be expressed in the context of interactions are particularly difficult to describe because the relevant genes reside in multiple social partners. Indirect genetic effects (IGEs) describe the influence of genes carried in social partners on the phenotype of a focal individual and thus provide a novel approach to quantifying the genetics underlying interactions such as reciprocal cooperation. We used inbred lines of guppies and a novel application of IGE theory to describe the dual genetic control of predator inspection and social behavior, both classic models of reciprocity. We identified effects of focal strain, social group strain, and interactions between focal and group strains on variation in focal behavior. We measured ,, the coefficient of the interaction, which describes the degree to which an individual's phenotype is influenced by the phenotype of its social partners. The genetic identity of social partners substantially influences inspection behavior, measures of threat assessment, and schooling and does so in positively reinforcing manner. We therefore demonstrate strong IGEs for antipredator behavior that represent the genetic variation necessary for the evolution of reciprocity. With this methodology, different to the conventional finite element method, discretization of the domain with a mesh adapted to the geometry of the discontinuity is not required. On the other hand, in traditional fracture mechanics all variables have been considered to be deterministic (uniquely defined by a given numerical value). However, the uncertainty associated with these variables (external loads, geometry and material properties, among others) it is well known. This paper presents a novel application of the perturbation method along with the extended finite element method to treat these uncertainties. The methodology has been implemented in a commercial software and results are compared with those obtained by means of a Monte Carlo simulation. Oliver Abstract Hybrid organic-inorganic glasses exhibit unique electro-optical properties along with excellent thermal stability. Their inherently mechanically fragile nature, however, which derives from the oxide component of the hybrid glass network together with the presence of terminal groups that reduce network connectivity, remains a fundamental challenge for their integration in nanoscience and energy technologies. We report on a combined synthesis and computational strategy to elucidate the effect of molecular structure on mechanical properties of hybrid glass films. We first demonstrate the importance of rigidity percolation to elastic behavior. Secondly, using a novel application of graph theory, we reveal the complex 3-D fracture path at the molecular scale and show that fracture energy in brittle hybrid glasses is fundamentally governed by the bond percolation properties of the network. The computational tools and scaling laws presented provide a robust predictive capability for guiding precursor selection and molecular network design of advanced hybrid organic-inorganic materials. The thermally-treated N,N, -ditridecyl perylene diimide (PTCDI-C13)-based n-type FETs using a COC/SiO2 gate dielectric show remarkably enhanced atmospheric performance and stability. The COC gate dielectric layer displays a hydrophobic surface (water contact angle = 95° ± 1°) and high thermal stability (glass transition temperature = 181 °C) without producing crosslinking. After thermal annealing, the crystallinity improves and the grain size of PTCDI-C13 domains grown on the COC/SiO2 gate dielectric increases significantly. The resulting n-type FETs exhibit high atmospheric field-effect mobilities, up to 0.90 cm2 V,1 s,1 in the 20 V saturation regime and long-term stability with respect to H2O/O2 degradation, hysteresis, or sweep-stress over 110 days. By integrating the n-type FETs with p-type pentacene-based FETs in a single device, high performance organic complementary inverters that exhibit high gain (exceeding 45 in ambient air) are realized. [source] Non-uniqueness with refraction inversion , the Mt Bulga shear zoneGEOPHYSICAL PROSPECTING, Issue 4 2010Derecke Palmer ABSTRACT The tau-p inversion algorithm is widely employed to generate starting models with many computer programs that implement refraction tomography. However, this algorithm can frequently fail to detect even major lateral variations in seismic velocities, such as a 50 m wide shear zone, which is the subject of this study. By contrast, the shear zone is successfully defined with the inversion algorithms of the generalized reciprocal method. The shear zone is confirmed with a 2D analysis of the head wave amplitudes, a spectral analysis of the refraction convolution section and with numerous closely spaced orthogonal seismic profiles recorded for a later 3D refraction investigation. Further improvements in resolution, which facilitate the recognition of additional zones with moderate reductions in seismic velocity, are achieved with a novel application of the Hilbert transform to the refractor velocity analysis algorithm. However, the improved resolution also requires the use of a lower average vertical seismic velocity, which accommodates a velocity reversal in the weathering. The lower seismic velocity is derived with the generalized reciprocal method, whereas most refraction tomography programs assume vertical velocity gradients as the default. Although all of the tomograms are consistent with the traveltime data, the resolution of each tomogram is comparable only with that of the starting model. Therefore, it is essential to employ inversion algorithms that can generate detailed starting models, where detailed lateral resolution is the objective. Non-uniqueness can often be readily resolved with head wave amplitudes, attribute processing of the refraction convolution section and additional seismic traverses, prior to the acquisition of any borehole data. It is concluded that, unless specific measures are taken to address non-uniqueness, the production of a single refraction tomogram that fits the traveltime data to sufficient accuracy does not necessarily demonstrate that the result is either correct, or even the most probable. [source] Inter- alu PCR detects high frequency of genetic alterations in glioma cells exposed to sub-lethal cisplatinINTERNATIONAL JOURNAL OF CANCER, Issue 4 2005Tapasya Srivastava Abstract Increased genomic instability contributes to higher frequency of secondary drug resistance and neoplastic progression in tumors as well as in cells exposed to sub-lethal concentrations of chemotherapeutic agents. We have used PCR based DNA fingerprinting techniques of randomly amplified polymorphic DNA (RAPD) and inter- alu PCR to study this phenomenon in the tumor genome. The choice of the primer, either random (for RAPD) or specific (inter- alu PCR) can determine the nature of alterations being assessed. We have compared the inter- alu PCR and RAPD profiles of U87MG glioblastoma cells exposed to sequentially increasing low doses of cisplatin for 24 passages to that of untreated controls. Inter- alu PCR, with 2 primers, demonstrated a number of alterations in the treated cells, in the form of loss / gain and changes in the intensity of bands. No changes were observed by RAPD analysis with 5 primers, however, indicating a preferential increase in the alu mediated recombination frequency in the treated cells (p = 1.866 × 10,4). The number of changes observed with respect to the corresponding leucocyte DNA in the inter- alu PCR profile of 26 primary tumors (Grade II = 13; Grade IV = 13), resected before chemotherapy, for the 2 inter- alu primers was very small. We present a novel application of the inter- alu PCR in detecting alterations in long term cultured cells at low dose exposure to a chemotherapeutic agent. Our results suggest that alu mediated recombination may be important in cells exposed to sub-lethal doses of cisplatin but not in the genesis of primary glioma. © 2005 Wiley-Liss, Inc. [source] Flight control of a rotary wing UAV using backsteppingINTERNATIONAL JOURNAL OF ROBUST AND NONLINEAR CONTROL, Issue 6 2010Bilal Ahmed Abstract This paper presents a novel application of backstepping controller for autonomous landing of a rotary wing UAV (RUAV). This application, which holds good for the full flight envelope control, is an extension of a backstepping algorithm for general rigid body velocity control. The nonlinear RUAV model used in this paper includes the flapping and servo dynamics. The backstepping-based controller takes advantage of the ,decoupling' of the translation and rotation dynamics of the rigid body, resulting in a two-step procedure to obtain the RUAV control inputs. The first step is to compute desired thrusts and flapping angles to achieve the commanded position and the second step is to compute control inputs, which achieve the desired thrusts and flapping angles. This paper presents a detailed analysis of the inclusion of a flapping angle correction term in control. The performance of the proposed algorithm is tested using a high-fidelity RUAV simulation model. The RUAV simulation model is based on miniature rotorcraft parameters. The closed-loop response of the rotorcraft indicates that the desired position is achieved after a short transient. The Eagle RUAV control inputs, obtained using high-fidelity simulation results, clearly demonstrate that this algorithm can be implemented on practical RUAVs. Copyright © 2009 John Wiley & Sons, Ltd. Reed Summary 1In order to reproduce successfully in a temporally varying environment, iteroparous animals must exhibit considerable behavioural flexibility across their lifetimes. By adjusting timing of breeding each year, parents can ensure optimal overlap between the energy intensive period of offspring production and the seasonal peak in favourable environmental conditions, thereby increasing their chances of successfully rearing young. 2Few studies investigate variation among individuals in how they respond to fluctuating conditions, or how selection acts on these individual differences, but this information is essential for understanding how populations will cope with rapid environmental change. 3We explored inter-annual trends in breeding time and individual responses to environmental variability in common guillemots Uria aalge, an important marine top predator in the highly variable California Current System. Complex, nonlinear relationships between phenology and oceanic and climate variables were found at the population level. Using a novel application of a statistical technique called random regression, we showed that individual females responded in a nonlinear fashion to environmental variability, and that reaction norm shape differed among females. 4The pattern and strength of selection varied substantially over a 34-year period, but in general, earlier laying was favoured. Females deviating significantly from the population mean laying date each year also suffered reduced breeding success, with the strength of nonlinear selection varying in relation to environmental conditions. 5We discuss our results in the wider context of an emerging literature on the evolutionary ecology of individual-level plasticity in the wild. Better understanding of how species-specific factors and local habitat features affect the timing and success of breeding will improve our ability to predict how populations will respond to climate change. [source] Dispersal pattern of domestic cats (Felis catus) in a promiscuous urban population: do females disperse or die?JOURNAL OF ANIMAL ECOLOGY, Issue 2 2003Sébastien Devillard Summary 1The domestic feral cat (Felis catus L.) is a good model for studying intraspecific variability of dispersal patterns in mammals because cats live under a large diversity of socio-ecological conditions. We analysed both the natal and breeding dispersal patterns of domestic cats in a promiscuous urban population and tested whether or not it differed from the male-biased natal dispersal pattern observed for polygynous rural populations. 2During an 8-year study we recorded the exact date of in situ death for 148 marked cats and the exact date of disappearance from the population for 99 other cats. Because undiscovered deaths might over-estimate dispersal probabilities when considering only disappearance probabilities, we made an novel application of multistrata capture,recapture methods in order to disentangle dispersal from true mortality. 3We showed that mature females dispersed, both before and after their first reproduction, at 1 and 2 years old. Contrary to females, no dispersal seemed to occur in males. Before sexual maturity, females that disappeared at 1 and 2 years old were in worse body condition than females that stayed in the population area after 2 years old. However, they did not reproduce less successfully before their disappearance than females that died later in the population area. 4The female-biased and low natal dispersal pattern in this population was atypical compared to other promiscuous/polygynous mammals and differed from that observed in rural polygynous populations of domestic cat. Neither local mate competition nor inbreeding avoidance appeared to be sufficient pressures to counterbalance ecological constraints on dispersal in an urban environment. However, local resource competition for den sites between potential matriarchies could lead to the breeding dispersal of less competitive females. Ellis Abstract Aim, To quantify the interaction between climate and woodland continuity in determining the bioclimatic response of lichen epiphytes. Location, Northern Britain (Scotland). Methods, Indicator-species analysis was used to pre-select lichen epiphytes along parallel gradients in climate and the extent of old-growth woodland. Nonparametric multiplicative regression was used to describe in a predictive model the individualistic response of selected species, which were projected based on climate-change scenarios and contrasting patterns of simulated woodland loss or gain. Species with a similar response were grouped using a novel application of cluster analysis to summarize the potentially huge number of projected outcomes. Projected patterns of occurrence under climate-change scenarios were examined for different levels of old-growth woodland extent. Results, Forty-two lichen species were statistically significant indicator species in oceanic woodlands, and old-growth indicators under suboptimal climatic conditions. Responses to climate-change scenarios were contrasting, with one group comprising species projected to increase in extent in response to climate warming, and other response groups projected to decrease in occurrence, possibly in response to shifting rainfall patterns. The occurrence of all response groups had a positive relationship with old-growth woodland extent. Main conclusions, An ,oceanic' biogeographical group of epiphytes identified using the baseline climatic and present-day woodland setting comprised species with a cyanobacterial photobiont or tropical phytogeographical affinities. However, within this group the individual species responses to climate-change scenarios were contrasting. Additionally, group responses may be poorly matched with simple ecological traits. However, the studied interaction between climate and habitat continuity suggests that the impact of climate change might be offset for certain lichen epiphytes by appropriate management of woodland resources, for example, expansion of native woodland around remnant old-growth stands. Encapsulation of metal nanoparticles for catalysis, drug delivery and light harvesting are only some applications of dendrimers that are breaking new ground. A novel application of dendrimer technology is described in the present paper that relates to industrial water treatment. Industrial water systems often suffer from undesirable inorganic deposits. These can form either in the bulk or on metallic surfaces, such as heat exchangers or pipelines. Silica (SiO2) scale formation and deposition is a major problem in high-silica-containing cooling waters. Scale prevention rather than removal is highly desired. In this paper, benchtop screening tests on various silica inhibition chemistries are reported, with emphasis on materials with a dendrimeric structure. Specifically, the inhibition properties of commercially available STARBURST® polyaminoamide (PAMAM) dendrimers generations 0.5, 1, 1.5, 2, and 2.5 are investigated in detail together with other commonly-used scale inhibitors. Experimental results show that inhibition efficiency largely depends on structural features of PAMAM dendrimers such as generation number and nature of the end groups. PAMAM dendrimers are effective inhibitors of silica scale growth at 40 ppm dosage levels. PAMAM dendrimers also act as silica nucleators, forming SiO2,PAMAM composites. This occurs because the SiO2 formed by incomplete inhibition interacts with cationic PAMAM-1 and -2. The general scope of silica formation and inhibition in industrial waters is also discussed. Benton Derrick This paper presents a novel application of a model reference adaptive control (MRAC) system to control the lateral position of a farm tractor tracking a straight path. Farm tractors can be configured with various implements, and the tractor yaw rate dynamics vary with each implement. It is desired that the lateral position response of the farm tractor remain consistent with respect to different implement configurations. Therefore, a MRAC system is implemented on the farm tractor to compensate for yaw rate plant variations by adapting the feed-forward yaw rate controller. Simulation results of the algorithm are shown that display poor performance due to neglected steering actuator dynamics and saturation. Modifications are made to the algorithm to account for the steering actuator properties, and more simulated results are presented that display ideal performance. The authors assess the level of food safety performance in one accession country, Hungary, in one food sector (the fresh produce importing chain), and evaluate the capacity of the system to demonstrate quality assurance to the satisfaction of private customers and public regulators. The analysis of food safety performance has been undertaken through gap analysis using a novel application of a benchmarking methodology taking the United Kingdom fresh produce importing chain as the benchmark. In this study, fluorescent differential cell viability staining and flow cytometry were used to determine chondrocyte viability following laser heating. Study Design/Materials and Methods Porcine septal cartilages were irradiated with an Nd:YAG laser (,,= 1.32 ,m, 25 W/cm2) while surface temperature, stress relaxation, and diffuse reflectance were recorded. Each slab received one, two, or three laser exposures (respective exposure times of 6.7, 7.2, 10 seconds). Irradiated samples were then divided into two groups analyzed immediately and at 5 days following laser exposure. Chondrocytes were isolated following serial enzymatic digestion, and stained using SYTO®/DEAD RedÔ (Molecular Probes, Eugene, OR). A flow cytometer was then used to detect differential cell fluorescence; size; granularity; and the number of live cells, dead cells, and post-irradiation debris in each treatment population. Results Nearly 60% of chondrocytes from reshaped cartilage samples isolated shortly after one irradiation, were viable while non-irradiated controls were 100% viable. Specimens irradiated two or three times demonstrated increasing amounts of cellular debris along with a reduction in chondrocyte viability: 31 and 16% after two and three exposures, respectively. In those samples maintained in culture medium and assayed 5 days after irradiation, viability was reduced by 28,88%, with the least amount of deterioration in untreated and singly irradiated samples. Conclusions Functional fluorescent dyes combined with flow cytometric analysis successfully determines the effect of laser irradiation on the viability of reshaped cartilage. Lasers Surg. Dusane Abstract Aim:, The ability of enzymatically synthesized lauroyl glucose to disrupt fungal (Candida albicans, Candida lipolytica) and bacterial (Pseudomonas aeruginosa PAO1, Pseudomonas aureofaciens) biofilms was investigated. Methods and Results:, Preformed biofilms of C. albicans and C. lipolytica in polystyrene microtitre plates were disrupted upto 45% and 65%, respectively, while P. aeruginosa and P. aureofaciens biofilms were disrupted by 51% and 57%. Precoating of the microtitre wells with lauroyl glucose affected cell attachment and biofilm growth of all the cultures to a lesser extent. With C. albicans and C. lipolytica, there was 11% and 32% decrease in the development of biofilms, respectively. With P. aeruginosa and P. aureofaciens, the reduction was 21% and 12% after 48 h. Lauroyl glucose effectively inhibited the formation of biofilms on glass slide surfaces when added along with the inoculum. Analysis by confocal laser scanning microscopy showed that the growth of the biofilms was lesser as compared with the control experiments. Lauroyl glucose displayed minimum inhibitory concentration values >500 ,g ml,1 for the test cultures and was comparable to that obtained with acetyl salicylate. Conclusion:, Lauroyl glucose reduces biofilm growth of all the four test cultures on polystyrene and glass surfaces. Stok Abstract A new technique for characterization of the three-dimensional morphology of murine articular cartilage is proposed. The technique consists of a novel application of confocal laser scanning microscopy (CLSM), where the objective was to develop and validate it for cartilage measurements in murine joints. Murine models are used in arthritis research, because they are well-described for manipulating the disease pathophysiology, facilitating our understanding of the disease, and identifying new targets for therapy. A calibration and reproducibility study was carried out to provide a consistent testing methodology for quantification of murine joints. The proximal tibial condyles from male C57BL/6 mice were scanned using a CLS microscope with an isotropic voxel size of 5.8 ,m. Measurements and analyses were repeated three times on different days, and in a second step the analysis was repeated three times for a single measurement. Calculation of precision errors (coefficient of variation) for cartilage thickness and volume was made. The bias of the system was estimated through comparison with histology. This technique showed good precision, with errors in the repeated analysis ranging from 0.63% (lateral thickness) to 3.48% (medial volume). The repeated analysis alone was robust, with intraclass correlations for the different compartments between 0.918 and 0.991. Measurement bias was corrected by scaling the confocal images to 32% of their width to match histology. CLSM provided a fast and reproducible technique for gathering 3D image data of murine cartilage and will be a valuable tool in understanding the efficacy of arthritis treatments in murine models. [source] A mathematical programming approach for improving the robustness of least sum of absolute deviations regressionNAVAL RESEARCH LOGISTICS: AN INTERNATIONAL JOURNAL, Issue 4 2006Avi Giloni Abstract This paper discusses a novel application of mathematical programming techniques to a regression problem. While least squares regression techniques have been used for a long time, it is known that their robustness properties are not desirable. Specifically, the estimators are known to be too sensitive to data contamination. In this paper we examine regressions based on Least-sum of Absolute Deviations (LAD) and show that the robustness of the estimator can be improved significantly through a judicious choice of weights. The problem of finding optimum weights is formulated as a nonlinear mixed integer program, which is too difficult to solve exactly in general. We demonstrate that our problem is equivalent to a mathematical program with a single functional constraint resembling the knapsack problem and then solve it for a special case. We then generalize this solution to general regression designs. Furthermore, we provide an efficient algorithm to solve the general nonlinear, mixed integer programming problem when the number of predictors is small.
