Reduced fertility of mouse epididymal sperm lacking Prss21/Tesp5 is rescued by sperm exposure to uterine microenvironment

Although the acrosome reaction and subsequent penetration of sperm through the egg zona pellucida (ZP) are essential for mammalian fertilization, the molecular mechanism is still controversial. We have previously identified serine protease Tesp5 identical to Prss21 on the mouse sperm surface as a candidate enzyme involved in sperm penetration through the ZP. Here we show that despite normal fertility of male mice lacking Prss21/Tesp5, the epididymal sperm penetrates the ZP only at a very low rate in vitro, presumably owing to the reduced ability to bind the ZP and undergo the ZP-induced acrosome reaction. The ability of Prss21-null sperm to fuse with the egg in vitro was also impaired severely. Intriguingly, the reduced fertility of Prss21-null epididymal sperm was rescued by exposure of the sperm to the uterine microenvironment and by in vitro treatment of the sperm with uterine fluids. These data suggest the physiological importance of sperm transport through the uterus. [source]

Summary Treatment with cytotoxic chemotherapy and radiotherapy is associated with significant gonadal damage in men. Alkylating agents, such as cyclophosphamide and procarbazine, are the most common agents implicated. The vast majority of men receiving procarbazine-containing regimens for the treatment of lymphomas are rendered permanently infertile. Treatment with adriamycin, bleomycin, vinblastine and dacarbazine (ABVD) appears to have a significant advantage in terms of testicular function, with a return to normal fertility in the vast majority of patients. Cisplatin-based chemotherapy for testicular cancer results in temporary azoospermia in most men with a recovery of spermatogenesis in about 50% after 2 years and 80% after 5 years. There is also evidence of chemotherapy-induced Leydig cell impairment in a proportion of these men, although this appears to be of no clinical significance in the majority of patients. The germinal epithelium is very sensitive to radiation-induced damage with changes to spermatogonia following as little as 0.1 Gy, and permanent infertility after fractionated doses of 2 Gy and above, whereas clinically significant Leydig cell impairment occurs rarely with doses of less than 20 Gy. Sexual and Reproductive Health Knowledge in Cystic Fibrosis Female Patients and Their Parents

ABSTRACT Introduction., The changing outcomes for young cystic fibrosis (CF) patients means that reproductive health issues have become an integral part of CF management. Aim., The aim of this study was to investigate the knowledge and experiences of reproductive and sexual health issues in women with CF and to investigate the knowledge and reproductive health attitudes of their parents. Main Outcome Measures., Assessment of reproductive and sexual health knowledge in female CF patients and their parents. Methods., A questionnaire study directed to 120 Polish women with CF aged 16 years and older and their parents. Results., Sixty-four patients and their parents responded to the questionnaire. Sixty-eight percent of the patients started sexual intercourse at a mean age of 19.2 years. Eighty-four percent of all sexually active women reported that they did not use any form of contraception. Only 32.8% of women understood the problems connected with their own and male fertility in CF. Popular scientific publications and other CF patients were identified as the most important source of information. Only 23% of parents understood the problems connected with female fertility in CF; 44% of parents thought that man with CF had normal fertility. Seventy-five percent of the women and 40% of the parents felt that sexual health discussions should begin between age 12 and 14 years with a CF doctor and the mother. Conclusions., Our study showed that significant knowledge gaps exist regarding fertility issues in both CF patients and CF parents. Women with CF have some general knowledge about sexual issues but insufficient knowledge to have a safe sexual life. The results helped us to develop the educational program for CF patients. Analysis of some factors affecting fertility levels in a high-producing dairy herd in south-western Japan

ABSTRACT The present study aimed to know whether all cows have been showing declining fertility or only a proportion of cows are attributed to the declining fertility, and to describe factors affecting the level of fertility. A total of 131 cows calved from February 2005 to December 2007 in a dairy herd were examined. Fourteen cows were excluded from the study because of early culling. Of the remaining 117 cows, 47 (40%) conceived within 115 days postpartum after 1,3 artificial insemination (AI) (normal fertility cows), 42 (36%) conceived after 115 days postpartum following 1,3 AI or were culled after 1,2 (sub-fertility cows/culled), and 28 (24%) were inseminated more than three times without detectable genital tract abnormalities (repeat breeders). Calving to conception interval in the normal fertility group was 72 + 3 days, while in the sub-fertility/culled and repeat breeding groups the intervals were 170 + 8 and 259 + 16 days, respectively. Endometritis was the risk factor for sub-fertility/culled (odds ratio (OR) = 3.76). Prolonged luteal phase (OR = 4.08), delayed first ovulation (OR = 6.02), and delayed corpus luteum formation after AI (OR = 8.55) were the risk factors for repeat breeding. In conclusion, 60% cows showed reduced fertility in a herd, while the other 40% had normal fertility. Uterine infection and some ovarian disorders contributed to reduced fertility. [source]


Surgical correction of rectovaginal fistula in mares and subsequent fertility

Objective To evaluate the fertility of mares bred at various intervals relative to surgical management of rectovaginal fistula (RVF). Materials and Methods Surgical repair of RVF was performed in 28 mares at variable times relative to foaling (30 days to 24 months) and also relative to rebreeding (same cycle or delayed). Postoperative fertility was then evaluated. Results Two mares were already pregnant at the time of surgery and 20 of 23 mares (87%) that were bred immediately prior to or following surgery conceived from their first service. When mares were bred in the same cycle as surgery, the next cycle following surgery or in the following breeding season after surgery the pregnancy rate was 5/5, 5/6 and 10/12, respectively, and the foaling rates were 4/5, 4/6 and 7/12. The two mares already pregnant at the time of surgery foaled successfully. Conclusions Excellent fertility can be achieved following surgical repair of RVF and our results suggest that delaying breeding until the following breeding season is not necessary. In addition, breeding in the same cycle as the surgical repair is a previously unreported technique that should be considered to maintain normal fertility and a yearly foaling interval. [source]


Influence of antibiotic cefotaxime on somatic embryogenesis and plant regeneration in indica rice

Abstract An antibiotic, cefotaxime (OmnataxÔ) has been found to promote somatic embryogenesis and subsequent plant regeneration in vitro in indica-type basmati rice cultures. Response was highly genotype specific. The number, mass and morphology of the calli formed on the scutellar tissues were dependent on the growth medium (with or without cefotaxime). The embryogenic nature of nodular calli was confirmed through histological analysis and their plant regeneration ability. The calli of variety Pusa basmati 1 grown on medium supplemented with cefotaxime (100 mg/L) exhibited up to 70.5% plant regeneration as compared to control (51.51%). Plants regenerated from emryogenic calli were phenotypically normal and identical to seed-derived plants and exhibited normal fertility. A limited humidity and an optimal aeration of the culture tubes further enhanced the frequency of somatic embryogenesis and plant regeneration. [source]
