This experimental study investigates the effects of computer-delivered, explicit information on the acquisition of Spanish word order by comparing four groups comprised of [+/,Explanation] and [+/,Explicit Feedback]. Results showed that all groups improved significantly and similarly on interpretation and production tests. It is suggested that explicit information may not necessarily facilitate second Language acquisition and that exposing learners to task-essential practice is sufficient to promote acquisition. [source] Consequences of Positive and Negative Feedback: The Impact on Emotions and Extra-Role BehaviorsAPPLIED PSYCHOLOGY, Issue 2 2009Frank D. Belschak These studies examine employees' emotional reactions to performance feedback from their supervisors as well as subsequent effects on attitudes and (intentions to show) affect-driven work behaviors (counterproductive behavior, turnover, citizenship, and affective commitment). A pre-study (N= 72) illustrates that employees regularly receive performance feedback from supervisors and that this feedback elicits different positive and negative emotions. Next, a scenario experiment (Study 1) comparing the effects of positive/negative feedback given in public/private was conducted, with a student sample (N= 240) and a sample of working adults (N= 107). In both samples, feedback has an impact on emotions and subsequently on work attitudes and behavioral intentions. The results from the scenario experiment were validated in a survey study (Study 2) among employees of a for-profit research firm (N= 86) who reported on recalled emotions and work behaviors after receiving performance feedback during appraisals. Again, different types of feedback relate to different emotions. In turn, these emotions were related to subsequent work behaviors and attitudes. Together, these studies show that feedback affects recipients' emotions and that such emotional reactions mediate the relationship between feedback and counterproductive behavior, turnover intentions, citizenship, and affective commitment. Une préenquête (N= 72) a montré que les salariés reçoivent régulièrement des informations sur leurs résultats de la part de leur supérieur et que cette situation provoque des émotions à la fois positives et négatives. Ensuite, une expérience (Etude n° 1) comparant les conséquences d'un feedback positif ou négatif exprimé en public ou en privé a été menée à bien sur un échantillon d'étudiants (N= 240) et sur un échantillon de travailleurs (N= 107). Simons Abstract Negative feedback, either internal or external, is a fundamental guide to human learning and performance. The neural system that underlies the monitoring of performance and the adjustment of behavior has been subject to multiple neuroimaging investigations that uniformly implicate the anterior cingulate cortex and other prefrontal structures as crucial to these executive functions. The present article describes a series of experiments that employed event-related potentials to study a variety of processes associated with internal or external feedback. Three medial-frontal negativities (error-related negativity, correct-response negativity, feedback-related negativity) are highlighted, each of which plays an important role in the monitoring and dynamic adjustment of behavior. Extensions of basic research on these ERPs to questions relevant to clinical-science are also provided. [source] Escalation: The Role of Problem Recognition and Cognitive BiasDECISION SCIENCES, Issue 3 2007Mark Keil ABSTRACT Escalation of commitment to a failing course of action is an enduring problem that remains central to the study of managerial behavior. Prior research suggests that escalation behavior results when decision makers choose to ignore negative feedback concerning the viability of a previously chosen course of action. Previous work has also suggested that certain cognitive biases might promote escalation behavior, but there has been little attempt to explore how biases other than framing affect escalation. In this article, we explore the extent to which decision makers actually perceive negative feedback as indicative of a problem and how this influences their decision to escalate. Although problem recognition and cognitive biases have been intensively studied individually, little is known about their effect on escalation behavior. In this research, we construct and test an escalation decision model that incorporates both problem recognition and two cognitive biases: selective perception and illusion of control. Our results revealed a significant inverse relationship between problem recognition and escalation. Furthermore, selective perception and illusion of control were found to significantly affect both problem recognition and escalation. The implications of these findings for research and practice are discussed. To improve problem recognition and reduce the incidence of escalation, practicing managers should implement modern project management practices that can help to identify and highlight potential problems while guarding against these two key cognitive biases that promote the behavior. [source] Impaired estrogen-induced negative feedback on gonadotropin secretion in patients with gonadotropin-secreting and nonfunctioning pituitary adenomasEUROPEAN JOURNAL OF CLINICAL INVESTIGATION, Issue 5 2002A. Lania Abstract Background Several in vitro studies suggest that gonadotropin-secreting pituitary adenomas (Gn-omas) and non functioning pituitary adenomas (NFPA) originate from gonadotroph cells. Patients with Gn-oma and NFPA frequently show abnormal gonadotropin response to TRH. The aim of the study was to investigate whether the estrogen-induced negative feedback is operating in either patients with Gn-oma or NFPA. Materials and methods Serum gonadotropin levels were evaluated at 24 h after ethinylestradiol administration (1 mg per os; EE2 test) in seven patients with a diagnosis of Gn-oma, based on the presence of high follicle-stimulating hormone (FSH) and/or lutenising hormone (LH) levels with normal or high levels of sex steroids, in 22 patients with NFPA with normal or low levels of gonadotropin and sex steroids, and 30 sex- and age-matched healthy subjects. A normal response to EE2 test was arbitrarily defined as a serum LH and FSH decrease of at least 40 and 30% below basal levels. Results Among patients with Gn-oma, only one had a normal FSH inhibition and another, a normal LH inhibition. Among the 22 patients with NFPA, the EE2 test caused a normal FSH or LH reduction in 10 and 15, respectively, while a normal reduction of both FSH and LH was observed in nine. Conclusions The study demonstrates that estrogen-induced negative feedback of gonadotropin secretion is disrupted in almost all patients with Gn-oma and in half of those with NFPA. This defective feedback is reminiscent of the resistance to thyroid hormones and glucocorticoids observed in patients with thyroid-stimulating hormone- (TSH-) and adrenocorticotropic hormone- (ACTH-)secreting adenomas, respectively. [source] C5a anaphylatoxin as a product of complement activation up-regulates the complement inhibitory factor H in rat Kupffer cellsEUROPEAN JOURNAL OF IMMUNOLOGY, Issue 11 2004Gerald Schlaf Abstract The 155-kDa complement regulator factor H (FH) is the predominant soluble regulatory protein of the complement system. It acts as a cofactor for the factor I-mediated conversion of the component C3b to iC3b, competes with factor B for a binding site on C3b and C3(H2O) and promotes the dissociation of the C3bBb complex. The primary site of synthesis is the liver, i.e. FH-specific mRNA and protein were identified in both hepatocytes (HC) and Kupffer cells (KC). Previous studies in rat primary HC and KC had shown that the proinflammatory cytokine IFN-, influences the balance between activation and inhibition of the complement system through up-regulation of the inhibitory FH. In this study we show that C5a, as a product of complement activation, stimulates the expression of FH-specific mRNA and protein in KC and thus induces a negative feedback. Quantitative-competitive RT-PCR showed an approximate threefold C5a-induced up-regulation of FH. ELISA analyses revealed a corresponding increase in FH protein in the supernatants of KC. The up-regulation of FH was completely inhibited by the C5a-blocking monoclonal antibody 6-9F. Furthermore, an involvement of LPS and IFN-, was excluded, which strongly indicates a direct effect of C5a on the expression of FH in KC. [source] CLINICAL AND IMAGING STUDY: Glucocorticoid negative feedback in methadone-maintained former heroin addicts with ongoing cocaine dependence: dose,response to dexamethasone suppressionADDICTION BIOLOGY, Issue 1 2006Bruno Aouizerate ABSTRACT Combined cocaine and illicit opiate use is common. This study aimed to test the hypothesis that cocaine dependence in former heroin-addicted patients maintained on methadone treatment is associated with enhanced glucocorticoid negative feedback. Multiple dose dexamethasone suppression tests, using a conventional 2.0 mg dose, and two lower doses, 0.5 mg and 0.125 mg, were performed in 10 methadone-maintained former heroin addicts with ongoing cocaine dependence (C-MM), 10 stabilized methadone-maintained former heroin addicts with no ongoing drug or alcohol use (MM), and 22 normal volunteers (NV). At 9 hours, there was no difference in plasma adrenocorticotropin hormone (ACTH) and/or cortisol levels among groups on the baseline day, as well as after the two lower doses of dexamethasone. At 17 hours, C-MM and MM had significantly lower plasma ACTH and/or cortisol levels than NV. However, C-MM did not significantly differ from MM in their hormonal levels. When the hormonal responses to dexamethasone are expressed as magnitude of lowering from baseline, there was no significant difference at any dose among groups. Therefore, C-MM exhibited a normal glucocorticoid negative feedback in the morning. Using the standard interpretation of dexamethasone suppression testing based on the examination of the actual hormonal levels rather than the difference from baseline condition, C-MM appear to have glucocorticoid effects similar to MM, yet were both greater than NV in the late afternoon. Thus, further studies are needed to know whether altered glucocorticoid negative feedback is related to chronic cocaine exposure, or is the result of former heroin addiction and/or its long-term treatment with methadone. [source] Decreased glucocorticoid receptor mRNA and dysfunction of HPA axis in rats after removal of the cholinergic innervation to hippocampus.EUROPEAN JOURNAL OF NEUROSCIENCE, Issue 7 2002Jung-Soo Han Abstract Excess exposure to glucocorticoids can have deleterious effects on physiology and cognition. Glucocorticoids acting via receptors located in hippocampal neurons contribute to negative feedback after stress by terminating the further release of glucocorticoids. The current study investigated the effects of selective immunolesions of septo-hippocampal cholinergic neurons on hippocampal corticosterone receptor mRNA and on hypothalamic-pituitary-adrenal (HPA) activity. As evaluated by in situ hybridization, hippocampal glucocorticoid receptor (GR) mRNA, but not mineralocorticoid receptor (MR) mRNA, was significantly decreased in lesioned rats compared to controls. In a companion study, the peak corticosterone response to one hour of restraint stress did not differ between lesion and control groups but the post-stress decline of corticosterone was more protracted in the lesioned rats. These findings are discussed in terms of their possible relevance to ageing as age-related degeneration of the basal forebrain cholinergic system may contribute to the commonly observed dysfunction of the HPA axis in older animals. Richard Eiser Processes of attitude learning were investigated through a game requiring discrimination between good and bad objects, where feedback about object valence (involving gain or loss) is contingent on approach. Previous research demonstrates a preponderance of false-negative errors, with some good objects (,learning asymmetry') and most novel objects (,generalization asymmetry') being judged as bad, but provides no direct evidence concerning how participants appraise alternative strategies and their own performance. To compare alternative strategies, participants received advice, supposedly from a previous participant, that most objects were bad and should be avoided, or good and should be approached. Learning and generalization asymmetries were replicated, especially among participants who followed the former (risk-averse) advice. Additionally, participants' evaluations of their own game strategy were inversely related to amount of negative feedback (the number of bad objects approached), but unrelated to positive feedback (from good objects approached), pointing to the salience of negative information for self-appraisals. A group of emergency medical service volunteers received negative, positive or no feedback on their volunteer organization and were asked to rate the ingroup and an outgroup on dimensions alternative to the feedback. As predicted, high identifiers showed an increase in ingroup favoritism after negative feedback (i.e. compensation) and a decrease after positive feedback (i.e. modesty effect). In contrast, low identifiers distanced themselves from the ingroup after negative feedback and accentuated self-ingroup similarity after positive feedback. Although such negative regulators are often expressed in a tissue- or cell-type specific manner during development, little is known about the significance of their differential expression patterns and possible interactions. We show the role and interplay of two cell-type specific negative feedback loops during specification of photoreceptor neurons in the Drosophila compound eye, a process that occurs via epidermal growth factor (EGF)-mediated sequential induction through the activation of the Ras/MAPK signaling pathway. Inducing cells secreting EGF express a negative regulator Sprouty (SPRY) that lowers Ras/MAPK signaling activity, and as a consequence reduces the signal-dependent expression of a secreted EGF inhibitor, Argos (AOS). Induced cells in turn express an orphan nuclear receptor Seven-up (SVP), which represses SPRY expression thereby allowing expression and secretion of AOS, preventing further induction. When this intricate system fails, as in spry mutants, sequential induction is no longer constant and the number of photoreceptor neurons becomes variable. Thus, cell-type specific utilization of multiple negative feedback loops not only confers developmental robustness through functional redundancy, but is a key component in generating consistent patterning. Main It is common practice to refer to three independent stages of creep under static loading conditions in the laboratory: namely transient, steady-state, and accelerating. Here we suggest a simple damage mechanics model for the apparently trimodal behaviour of the strain and event rate dependence, by invoking two local mechanisms of positive and negative feedback applied to constitutive rules for time-dependent subcritical crack growth. In both phases, the individual constitutive rule for measured strain , takes the form ,(t),=,,0,[1,+,t/m,]m, where , is the ratio of initial crack length to rupture velocity. For a local hardening mechanism (negative feedback), we find that transient creep dominates, with 0,<,m,<,1. Crack growth in this stage is stable and decelerating. For a local softening mechanism (positive feedback), m,<,0, and crack growth is unstable and accelerating. In this case a quasi-static instability criterion , , , can be defined at a finite failure time, resulting in the localization of damage and the formation of a throughgoing fracture. In the hybrid model, transient creep dominates in the early stages of damage and accelerating creep in the latter stages. At intermediate times the linear superposition of the two mechanisms spontaneously produces an apparent steady-state phase of relatively constant strain rate, with a power-law rheology, as observed in laboratory creep test data. The predicted acoustic emission event rates in the transient and accelerating phases are identical to the modified Omori laws for aftershocks and foreshocks, respectively, and provide a physical meaning for the empirical constants measured. At intermediate times, the event rate tends to a relatively constant background rate. The requirement for a finite event rate at the time of the main shock can be satisfied by modifying the instability criterion to having a finite crack velocity at the dynamic failure time, dx/dt , VR,, where VR is the dynamic rupture velocity. The same hybrid model can be modified to account for dynamic loading (constant stress rate) boundary conditions, and predicts the observed loading rate dependence of the breaking strength. The resulting scaling exponents imply systematically more non-linear behaviour for dynamic loading. [source] Indirect effects of soil moisture reverse soil C sequestration responses of a spring wheat agroecosystem to elevated CO2GLOBAL CHANGE BIOLOGY, Issue 1 2010SVEN MARHAN Abstract Increased plant productivity under elevated atmospheric CO2 concentrations might increase soil carbon (C) inputs and storage, which would constitute an important negative feedback on the ongoing atmospheric CO2 rise. However, elevated CO2 often also leads to increased soil moisture, which could accelerate the decomposition of soil organic matter, thus counteracting the positive effects via C cycling. We investigated soil C sequestration responses to 5 years of elevated CO2 treatment in a temperate spring wheat agroecosystem. The application of 13C-depleted CO2 to the elevated CO2 plots enabled us to partition soil C into recently fixed C (Cnew) and pre-experimental C (Cold) by 13C/12C mass balance. Gross C inputs to soils associated with Cnew accumulation and the decomposition of Cold were then simulated using the Rothamsted C model ,RothC.' We also ran simulations with a modified RothC version that was driven directly by measured soil moisture and temperature data instead of the original water balance equation that required potential evaporation and precipitation as input. The model accurately reproduced the measured Cnew in bulk soil and microbial biomass C. Assuming equal soil moisture in both ambient and elevated CO2, simulation results indicated that elevated CO2 soils accumulated an extra ,40,50 g C m,2 relative to ambient CO2 soils over the 5 year treatment period. However, when accounting for the increased soil moisture under elevated CO2 that we observed, a faster decomposition of Cold resulted; this extra C loss under elevated CO2 resulted in a negative net effect on total soil C of ,30 g C m,2 relative to ambient conditions. The present study therefore demonstrates that positive effects of elevated CO2 on soil C due to extra soil C inputs can be more than compensated by negative effects of elevated CO2 via the hydrological cycle. [source] Combined statistical and dynamical assessment of simulated vegetation,rainfall interactions in North Africa during the mid-Holocene,GLOBAL CHANGE BIOLOGY, Issue 2 2008MICHAEL NOTARO Abstract A negative feedback of vegetation cover on subsequent annual precipitation is simulated for the mid-Holocene over North Africa using a fully coupled general circulation model with dynamic vegetation, FOAM-LPJ (Fast Ocean Atmosphere Model-Lund Potsdam Jena Model). By computing a vegetation feedback parameter based on lagged auto-covariances, the simulated impact of North African vegetation on precipitation is statistically quantified. The feedback is also dynamically assessed through initial value ensemble experiments, in which North African grass cover is initially reduced and the climatic response analyzed. The statistical and dynamical assessments of the negative vegetation feedback agree in sign and relative magnitude for FOAM-LPJ. The negative feedback on annual precipitation largely results from a competition between bare soil evaporation and plant transpiration, with increases in the former outweighing reductions in the latter given reduced grass cover. This negative feedback weakens and eventually reverses sign over time during a transient simulation from the mid-Holocene to present. A similar, but weaker, negative feedback is identified in Community Climate System Model Version 2 (CCSM2) over North Africa for the mid-Holocene. [source] Mediation in a sibling context: the relations of older siblings' mediating behaviour and younger siblings' task performanceINFANT AND CHILD DEVELOPMENT, Issue 4 2002Pnina S. Klein Abstract We investigated the sibling relationship as a context for cognitive development. Forty preschoolers (ages 5,6) and their younger siblings (ages 2,3) were visited at home. Four games were presented to the older siblings and they were asked (a) to estimate how well their younger sibling will perform on each game and (b) to teach the younger sibling how to use the games. The older siblings' mediating behaviours during the teaching session and the younger siblings' performance on the four tasks were coded. The frequency of mediating behaviours,including attention focusing, amplifying affect and providing meaning, fostering a sense of competence, regulating of the learning process, de-contextualization, and negative feedback in the form of mocking and laughing at errors, predicted the younger siblings' task performance. The older sibling's accurate perception of the younger child's competence was uniquely predictive of task performance. The highest amount of mediation was observed in older-brother,younger-brother pairs, in particular the behaviours of negative feedback and amplifying affect. Results contribute to the discussion on the role of siblings as moderators of cognitive development and are discussed in terms of Vygotsky's cultural,historical perspective on apprenticeship. Method: This hypothesis was tested by assessing functional magnetic resonance brain imaging response to a monetary reward task known to activate the anterior ventral striatum (AVS), a region implicated in motivational aspects toward stimuli. To avoid the confounding effects of malnutrition, 10 women who had recovered from bulimia nervosa (BN) were compared with 10 healthy comparison women (CW). Results: For the AVS, CW distinguished positive and negative feedback, whereas recovered BN women had similar responses to both conditions. In addition, these groups had similar patterns of findings for the dorsal caudate. Discussion: We have previously shown that individuals recovered from anorexia nervosa (AN) also had altered striatal responses and difficulties in differentiating positive and negative feedback. Carless The level of agreement (mean differences and correlations) between self, peer and training staff ratings were examined in this study. The sample consisted of 545 participants who were undertaking a Royal Australian Airforce officer training program. Consistent with previous research there was strong agreement between training staff and peers and weak agreement between self-ratings and ratings by others (training staff and peers). Accuracy of ratings was examined by (a) comparing the mean ratings of outstanding, average and below-average performers; and (b) correlating difference scores with a measure of performance. The findings showed that below-average performers have a less accurate view of themselves compared to outstanding performers. Finally, we examined the effects of negative feedback on self-perceptions. The analyses indicated that after receiving negative feedback, average performers adjusted their self-ratings. Various explanations were proposed together with practical implications for training. The managers and subordinates rated 8 written vignettes describing different feedback strategies on a number of variables (perceived effectiveness, perceived risk, level of enactment demand, and congruence of strategy for managers). Feedback strategies that used an invitation for the subordinate to reply to the negative feedback were evaluated as more effective and managerially congruent, and less risky than were the other strategies. However, strategies requiring bidirectional communication (e.g., invitations to reply), as opposed to unidirectional communication (e.g., praise), were generally perceived as more demanding to enact. Zikmund-FisherArticle first published online: 26 NOV 200 Abstract Research on willingness to make marginal investments (e.g., the escalation and sunk cost literatures) has often focused on project completion decisions, such as the "radar-blank plane." This paper discusses a fundamentally different type of marginal investment decision, that of couples deciding whether to continue infertility treatment in the face of repeated failures. Two experiments based on this context show that when people face multiple independent chances to achieve a valued goal but are unsure about chances of success, premature quitting or "de-escalation" is the norm. Repeated negative feedback appears to induce individuals to see each successive failure as more and more diagnostic. As a result, even a short series of failed attempts evokes beliefs that future attempts will also fail. These emergent expectations of failure, generated by causal attribution processes, associative learning, and/or discounting of ambiguous information, appear very compelling and induce people to forgo profitable marginal investments. Webster Abstract On the basis of a domain-specific theory of self-esteem, it was hypothesized that functionally distinct domains of self-esteem would predict aggression differentially. Participants completed self-report measures of self-perceived superiority, mate value, social inclusion, and global self-esteem, as well as of aggression. Self-assessed mate value emerged as a reliable, positive predictor, and social inclusion as a reliable inverse predictor, of self-reported hostility and aggression. In a subsequent laboratory experiment, in which participants had an opportunity to aggress against the source of positive or negative feedback about a personal essay that they had written, mate value again predicted increased aggression, whereas global self-esteem predicted decreased aggression. These main effects were moderated by the feedback manipulation, such that their respective simple effects were only present among participants that received negative feedback. Aggr. Watts Computer-mediated communication (CMC) makes it possible to deliver evaluative feedback, an essential component of learning, over distance. This article presents a theoretical model of the CMC-based evaluative feedback process from the perspective of both senders and receivers of feedback. Hypotheses suggested by the model were tested in a quasi-laboratory experiment with part-time MBA students comparing email with voicemail. Within matched-pair dyads, email senders, but not voicemail senders, viewed their negative feedback as being significantly more negative than did their receivers. Voicemail senders, but not email senders, reported significantly lower comfort levels than did their receivers. No feedback effectiveness differences were found between media conditions, although determinants of feedback effectiveness differed significantly depending on the medium. These results are generally consistent with the theoretical model. Harrison Summary 1.,Our aim was to explore plant,soil feedback in mixed grassland communities and its significance for plant productivity and community composition relative to abiotic factors of soil type and fertility. 2.,We carried out a 4-year, field-based mesocosm experiment to determine the relative effects of soil type, historic management intensity and soil conditioning by a wide range of plant species of mesotrophic grassland on the productivity and evenness of subsequent mixed communities. 3.,The study consisted of an initial soil conditioning phase, whereby soil from two locations each with two levels of management intensity was conditioned with monocultures of nine grassland species, and a subsequent feedback phase, where mixed communities of the nine species were grown in conditioned soil to determine relative effects of experimental factors on the productivity and evenness of mixed communities and individual plant species performance. 4.,In the conditioning phase of the experiment, individual plant species differentially influenced soil microbial communities and nutrient availability. However, these biotic effects were much less important as drivers of soil microbial properties and nutrient availability than were abiotic factors of soil type and fertility. 5.,Significant feedback effects of conditioning were detected during the second phase of the study in terms of individual plant growth in mixed communities. These feedback effects were generally independent of soil type or fertility, and were consistently negative in nature. In most cases, individual plant species performed less well in mixed communities planted in soil that had previously supported their own species. 6.,Synthesis. These findings suggest that despite soil abiotic factors acting as major drivers of soil microbial communities and nutrient availability, biotic interactions in the form of negative feedback play a significant role in regulating individual plant performance in mixed grassland communities across a range of soil conditions. Carter Inter-individual differences in hypothalamic-pituitary-adrenal (HPA) axis activity underlie differential vulnerability to neuropsychiatric and metabolic disorders, although the basis of this variation is poorly understood. 11,-Hydroxysteroid dehydrogenase type 1 (11,-HSD1) has previously been shown to influence HPA axis activity. 129/MF1 mice null for 11,-HSD1 (129/MF1 HSD1,/,) have greatly increased adrenal gland size and altered HPA activity, consistent with reduced glucocorticoid negative feedback. On this background, concentrations of plasma corticosterone and adrenocorticotrophic hormone (ACTH) were elevated in unstressed mice, and showed a delayed return to baseline after stress in HSD1-null mice with reduced sensitivity to exogenous glucocorticoid feedback compared to same-background genetic controls. In the present study, we report that the genetic background can dramatically alter this pattern. By contrast to HSD1,/, mice on a 129/MF1 background, HSD1,/, mice congenic on a C57Bl/6J background have normal basal plasma corticosterone and ACTH concentrations and exhibit normal return to baseline of plasma corticosterone and ACTH concentrations after stress. Furthermore, in contrast to 129/MF1 HSD1,/, mice, C57Bl/6J HSD1,/, mice have increased glucocorticoid receptor expression in areas of the brain involved in glucocorticoid negative feedback (hippocampus and paraventricular nucleus), suggesting this may be a compensatory response to normalise feedback control of the HPA axis. In support of this hypothesis, C57Bl/6J HSD1,/, mice show increased sensitivity to dexamethasone-mediated suppression of peak corticosterone. Thus, although 11,-HSD1 appears to contribute to regulation of the HPA axis, the genetic background is crucial in governing the response to (and hence the consequences of) its loss. Similar variations in plasticity may underpin inter-individual differences in vulnerability to disorders associated with HPA axis dysregulation. They also indicate that 11,-HSD1 inhibition does not inevitably activate the HPA axis. Dudas Abstract The gonadotrophin-releasing hormone (GnRH) represents the final common pathway of a neuronal network that integrates multiple external and internal factors to control fertility. Among the many inputs GnRH neurones receive, oestrogens play the most important role. In females, oestrogen, in addition to the negative feedback, also exhibits a positive feedback influence upon the activity and output of GnRH neurones to generate the preovulatory luteinising hormone surge and ovulation. Until recently, the belief has been that the GnRH neurones do not contain oestrogen receptors and that the action of oestrogen upon GnRH neurones is indirect, involving several, oestrogen-sensitive neurotransmitter and neuromodulator systems that trans -synaptically regulate the activity of the GnRH neurones. Although this concept still holds for humans, recent studies indicate that oestrogen receptor-beta is expressed in GnRH neurones of the rat. This review provides three dimensional stereoscopic images of GnRH-immunoreactive (IR) and some peptidergic (neuropeptide Y-, substance P-, ,-endorphin-, leu-enkaphalin-, corticotrophin hormone-releasing- and galanin-IR) and catecholaminergic neurones and the communication of these potential oestrogen-sensitive neuronal systems with GnRH neurones in the human hypothalamus. Because the post-mortem human tissue does not allow the electron microscopic identification of synapses on GnRH neurones, the data presented here are based on light microscopic immunocytochemical experiments using high magnification with oil immersion, semithin sections or confocal microscopy. Owen Abstract We have previously shown that the foetal guinea-pig hypothalamic-pituitary-adrenal (HPA) axis is activated near the time of parturition and that this is associated with changes in limbic glucocorticoid receptors (GR) and mineralocorticoid receptors. In the present study, we hypothesized that the foetal hypothalamic paraventricular nucleus (PVN) and pituitary contribute significantly to foetal HPA drive but that these areas remain sensitive to negative feedback by circulating glucocorticoids in late gestation. However, we observed decreased corticotrophin-releasing hormone mRNA expression in the PVN and decreased pro-opiomelanocortin (POMC) mRNA levels in the anterior pituitary with advanced gestational age. The reduction in POMC mRNA expression was likely the result of negative feedback via circulating glucocorticoids because GR mRNA was unchanged during development in the foetal pituitary. Furthermore, we found that maternally administered glucocorticoids significantly decreased foetal pituitary POMC mRNA expression in a dose-dependent manner at gestational day (gd) 62 with male foetuses being more sensitive to these effects. These findings show that the foetal HPA axis remains highly sensitive to glucocorticoid feedback even as plasma adrenocorticotropic hormone and cortisol levels are elevated at the end of gestation. Shane High-anxious individuals, low-anxious individuals, and defensive copers completed a computerized go/no-go task, in which they learned when to press or not to press a button, in response to contingent positive and negative feedback. The duration that feedback remained onscreen was self-regulated. Defensive copers showed preferential reflection away from negative feedback, committed more passive-avoidance errors, and were characterized by impaired learning, overall. Further, the ratio of reflection on negative feedback to reflection on positive feedback directly mediated both passive-avoidance errors and overall learning. Defensive coping strategies, therefore, appear to interfere with passive avoidance learning, thereby fostering perseverative, dysfunctional action patterns by reducing knowledge gained from previous mistakes. Implications for the learning of effective socialization strategies, and for psychopathy,which is commonly characterized by similar passive-avoidance deficits,are subsequently considered. [source] Adrenocortical and Pituitary Glucocorticoid Feedback in Abstinent Alcohol-Dependent WomenALCOHOLISM, Issue 5 2010Bryon Adinoff Background:, The long-term ingestion of alcohol diminishes hypothalamic,pituitary,adrenal (HPA) axis reactivity in alcohol-dependent men, potentially altering future relapse risk. Although sex differences in HPA axis functioning are apparent in healthy controls, disruptions in this system have received little attention in alcohol-dependent women. In this study, we assessed the basal secretory profile of adrenocorticotropic hormone (ACTH) and cortisol, adrenocortical sensitivity in both the presence and absence of endogenous corticotropic pituitary activation, and feedback pituitary glucocorticoid sensitivity to dexamethasone. Methods:, Seven women 4- to 8-week abstinent alcohol-only dependent subjects and 10 age-matched female healthy controls were studied. All subjects were between 30 and 50 years old, not taking oral contraceptives, and were studied during the early follicular phase of their menstrual cycle. Circulating concentrations of ACTH and cortisol were measured in blood samples collected at frequent intervals from 2000 to 0800 hour. A submaximal dose of cosyntropin (0.01 ,g/kg), a synthetic ACTH (1,24), was administered at 0800 hour to assess adrenocortical sensitivity. In a separate session, low-dose cosyntropin was also administered following high-dose dexamethasone (8 mg intravenous) to assess adrenocortical sensitivity in the relative absence of endogenous ACTH. In addition, the ACTH response to dexamethasone was measured to determine the pituitary glucocorticoid negative feedback. Sessions were 5 days apart, and blood draws were obtained every 5 to 10 minutes. Results:, Mean concentrations and pulsatile characteristics of ACTH and cortisol over 12 hours were not statistically different between the 2 groups. Healthy controls had a somewhat higher (p < 0.08) net peak, but not net integrated, cortisol response to cosyntropin relative to the alcohol-dependent women. There were no significant group differences in either the ACTH or cortisol response to dexamethasone nor in the net cortisol response to cosyntropin following dexamethasone. Conclusion:, Significant differences in pituitary,adrenal function were not apparent between alcohol-dependent women and matched controls. Despite the small n, it appears that alcohol-dependent women do not show the same disruptions in HPA activity as alcohol-dependent men. These findings may have relevance for gender-specific treatment effectiveness. [source] The cellular level of the recognition factor RssB is rate-limiting for ,S proteolysis: implications for RssB regulation and signal transduction in ,S turnover in Escherichia coliMOLECULAR MICROBIOLOGY, Issue 6 2002Mihaela Pruteanu Summary Degradation of the general stress sigma factor ,S of Escherichia coli is a prime example of regulated proteolysis in prokaryotes. Whereas exponentially growing cells rapidly degrade ,S, various stress conditions result in stabilization and, therefore, rapid accumulation of ,S. Proteolysis of ,S requires the response regulator RssB, a direct recognition factor with phosphorylation-dependent affinity for ,S, which targets ,S to the ClpXP protease. Here, we demonstrate that a sudden increase in ,S synthesis results in ,S stabilization, indicating titration of an essential proteolytic component. Evidence is provided that RssB is the overall rate-limiting factor for ,S proteolysis. As a consequence, the cell has to continuously adjust the expression of RssB to ,S in order to maintain ,S proteolysis in growing cells, despite variations in the rate of ,S synthesis. Such homeostatic feedback-coupling is provided by rssB transcription being dependent on the ,S -controlled rssAB operon promoter. However, strong and rapid increases in ,S synthesis, in re-sponse to acute stress, exceed the compensatory potential of this feedback loop with the result that ,S is stabilized because of RssB titration. We propose that RssB control of ,S proteolysis functions as a genetic switch, in which (i) the ,off' state (low ,S levels caused by proteolysis) is stabilized by a homeostatic negative feedback, and (ii) the threshold for switching to the ,on' state (high levels of stable ,S) is dependent on the cellular level of active, i.e. phosphorylated RssB. [source] Hypoxanthine (HX) inhibition of in vitro meiotic resumption in goat oocytesMOLECULAR REPRODUCTION & DEVELOPMENT, Issue 3 2003Suofeng Ma Abstract To improve in vitro maturation and to understand the mechanism for meiotic resumption of oocytes, meiotic progression, and its control by hypoxanthine (HX) were studied in goat oocytes. Ovaries were obtained from a local abattoir, and cumulus,oocyte complexes (COCs) and follicular fluid were collected from follicles of different surface diameters (SDs). The meiotic competence and progression of oocytes were observed, and the concentration of HX in the follicular fluid and culture media was measured by high-performance liquid chromatography (HPLC). Full meiotic competence of goat oocytes was acquired in follicles of ,1.5 mm in SD with 90% of the oocytes developing to metaphase II (MII) stage after 24 hr in culture. The HX concentration in follicular fluid decreased with follicle development, from the highest level of 1.16 mM in ,0.5 mm follicles to the lowest level of 0.45 mM in ,5 mm follicles. HX inhibited meiotic resumption of goat oocytes in a concentration-related manner but this inhibitory effect declined gradually. When we renewed the medium at 4 hr of HX-199 (TCM-199 supplemented with 4 mM HX) culture, the percentage of oocytes with intact germinal vesicle (GV) did not increase but decreased significantly instead. HPLC measurement of HX in the HX-199 culture drops indicated that the HX concentration declined from 0 hr to 4 hr of culture and after medium renewal at 4 hr of culture. By adding dibutyryl cAMP (db-cAMP) at medium renewal, we found that db-cAMP held up the decline of GV percentages. Together, these results were consistent with the possibility that the decline of HX inhibitory effect was not due to HX depletion but rather due to the negative feedback of the metabolites on its further uptake by oocytes. Goat oocytes were capable of normal nuclear maturation and activation after temporal arrest by HX, but prolonged exposure to HX induced spontaneous activation. Mol. Reprod. [source] The UV properties of E+A galaxies: constraints on feedback-driven quenching of star formationMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 3 2007S. Kaviraj ABSTRACT We present the first large-scale study of E+A galaxies that incorporates photometry in the ultraviolet (UV) wavelengths. E+A galaxies are ,post-starburst' systems, with strong Balmer absorption lines indicating significant recent star formation, but without [O ii] and H, emission lines which are characteristic of ongoing star formation. The starburst that creates the E+A galaxy typically takes place within the last Gyr and creates a high fraction (20,60 per cent) of the stellar mass in the remnant over a short time-scale (<0.1 Gyr). We find a tight correlation between the luminosity of our E+A galaxies and the implied star formation rate (SFR) during the starburst. While low-luminosity E+As [M(z) > ,20] exhibit implied SFRs of less than 50 M, yr,1, their luminous counterparts [M(z) < ,22] show SFRs greater than 300 and as high as 2000 M, yr,1, suggesting that luminous and ultra-luminous infrared galaxies in the low-redshift Universe could be the progenitors of massive nearby E+A galaxies. We perform a comprehensive study of the characteristics of the quenching that truncates the starburst in E+A systems. We find that, for galaxies less massive than 1010 M,, the quenching efficiency decreases as the galaxy mass increases. However, for galaxies more massive than 1010 M,, this trend is reversed and the quenching efficiency increases with galaxy mass. Noting that the mass threshold at which this reversal occurs is in excellent agreement with the mass above which active galactic nuclei (AGN) become significantly more abundant in nearby galaxies, we use simple energetic arguments to show that the bimodal behaviour of the quenching efficiency is consistent with AGN and supernovae (SN) being the principal sources of negative feedback above and below M, 1010 M,, respectively. The arguments assume that quenching occurs through the mechanical ejection or dispersal of the gas reservoir and that, in the high-mass regime (M > 1010 M,), the Eddington ratios in this sample of galaxies scale as M,, where 1 < , < 3. Finally, we use our E+A sample to estimate the time it takes for galaxies to migrate from the blue cloud to the red sequence. We find migration times between 1 and 5 Gyr, with a median value of 1.5 Gyr. [source] |
